Abstract

Human activity has the great negative impact on water quality. As a consequence, deterioration of freshwater ecosystems has the effect on whole wildlife. Some of the pollutants which are realised into water could interact with DNA molecule and lead to DNA damage. The Velika Morava River is one of the larger tributaries of the Danube River. The major ecological problem of the Velika Morava River basin is discharging of municipal and industrial wastewaters. The aim of our study was the evaluation of genotoxic potential of the Velika Morava River basin. For this purpose, we employed battery of in vitro and in situ bioassays. From 9 selected sites the water samples and blood samples of Alburnus alburnus were collected. Within in vitro approach, the genotoxicity of water samples was tested using the SOS/umuC test on Salmonella thyphimurium TA1535/pSK1002 and alkaline comet assay on the HepG2 cell line. On the other hand, in situ approach included the alkaline and Fpg modified comet assay and the micronucleus test on bleak (A. alburnus) erythrocytes. The obtained results indicated differences of in vitro and in situ tests to evaluate genotoxic potential. The effects of genotoxicity were evident only in tests performed in situ, which is not surprising considering constant exposure of collected organisms to environmental stress. Differential sensitivity was found also within in situ tests where the alkaline comet assay showed the highest potential in distinguishing between sites. So, in accordance with results, we could conclude that application of the battery of in vitro and in situ bioassays is appropriate for assessment of genotoxic potential.

Abstract

Development of a feasible model for transport within complex vasculature network and tissue remains a challenge. Such a model is particularly important when considering drug transport within tumor environment. A drug used to cure the cancer is first transported through blood vessels, then it attaches to the vessel endothelium and faces biological barriers in the vessel wall to reach cancerous cells. We have developed a model for convective-diffusive drug transport which is simple and computationally efficient. One of the challenges was to couple fluid domain within blood vessels and solid domain of the tumor microenvironment. We have introduced fictitious 1D finite elements which appropriately take into account transport characteristics of the vessel walls. These characteristics include leakage and permeability of the walls. In evaluating wall permeability of a drug, we implemented our hierarchical multiscale methodology which couples molecular dynamics (MD) and continuum FE model. A numerical homogenization procedure was employed to obtain equivalent continuum transport parameters which account for interaction on molecular level between drug and solid components of the wall microstructure. Also, a possibility of using equivalent continuum transport models for capillary beds is investigated in order to further simplify and increase efficiency for the overall model of tumor. As a numerical example, we calculate transport through a capillary bed to illustrate applicability of our methodology.

Abstract

Research of the population of crayfish of the family Astacidae on the territory of Montenegro and Serbia by using COI gene mDNA as a genetic marker, have shown that all populations of the species Austropotamobius torrentium are homogenous and belonging to the haplogroup “Southern Balkan”. Population of Austropotamobius italicus, recorded only in Montenegro in river Zeta’s upper flow, belongs to the (haplogroup), subspecies meridionalis. The findings of Austropotamobius italicus meridionalis are new and an expansion of the southern range border of this subspecies in the Balkan peninsula. For the populations of Astacus astacus, two haplogroups have been detected ; the one from Serbia belongs to a new haplotype and is considered to be evolutionally older than the ones from Montenegro and Croatia. Using COI gene analysis, the highest value of nucleotide diversity (π) was noted for the species Austropotamobius torrentium, π = 4.6% (0.046±0.006) ; the lowest one was for the species Astacus astacus, π = 3.2% (0.032±0.006). The achieved results implicate urgent measures for conservation of populations Astacus astacus on the territory of Serbia and Austropotamobius italicus meridionalis on the territory of Montenegro, accompanied by the use of stricter measures in the management of populations of Astacus astacus in Montenegro.

Abstract

We have recently introduced a composite smeared finite element (CSFE) to model gradient-driven mass transport in biological tissue. The transport from capillary system is smeared in a way to transform 1D transport to a continuum, while the tissue is considered as a continuum. Coupling between the smeared pressure and concentration field is achieved through 1D connectivity elements assigned at each FE node. Here we extend our smeared model to include the lymphatic system. The lymphatic vessels are treated in a way analogous to the capillaries, by introducing the corresponding Darcy and diffusion tensors. Additional connectivity elements are added. In the numerical examples we demonstrate accuracy of the smeared model and the effects of the lymph on the pressure and concentration within the extracellular space, assuming that there is no transport to the cell space. Abstract

Liquid thermolysis products of various types of immature kerogen from sedimentary lacustrine rocks from the Valjevo-Mionica basin in Serbia were studied to evaluate the generation potential of kerogen contained in the organic matter (OM) of the rocks, determine the composition of the biomarkers and alkylaromatics in the liquid thermolysis products, and elucidate the effect of Pt4+ and Ru3+ ions (which were added in the form of inorganic salts) on the yield and hydrocarbon composition of the liquid thermolysis products. For this purpose, representative bitumen-free samples A and B of the sedimentary rocks were subjected to thermolysis under various conditions. Rock A contains high amount of immature organic matter, which is dominated by kerogen type I/II and was generated under strongly reduced sedimentation conditions at a high salinity. Sample B is poorer in immature OM than sample A, and the OM of the former contains kerogen type II/III and was generated predominantly in a reduced environment. The content of the liquid products and the concentrations of hydrocarbons obtained in the course of thermolysis of bitumen-free sample A and the typical oil distribution of the biomarkers and alkylaromatics in the thermolysis products confirm a high generation potential of OM in this rock. In all of our experiments on the thermolysis of bitumen-free sample B, the yield of liquid products and hydrocarbons is low. According to the kerogen type, the thermolysis of this rock generates much gases. The Pt4+ and Ru3+ ions (added in the form of simple inorganic salts) increased the yield of liquid (kerogen type I/II) and gaseous (kerogen type II/III) products. During the thermolysis of various type of immature kerogen in the lacustrine sedimentary rocks at a temperature of 400A degrees C, the OM attained maturation corresponding to the early catagenesis level. Abstract

In this paper, we introduce an incremental formulation for large strains which is based on the nodal forces increments. It uses the linear strain increments at the current configuration as the fundamental strain measure, and consequently the stress increments which produce the nodal force increments. It does not assume any total strain and stress calculation according to total values of (large) displacements. It rather employs summation of internal finite element nodal forces during the incremental-iterative computational procedures. Solutions according to this new formulation are compared with a standard updated Lagrangian formulation, with the Green- Lagrangian strains evaluated using the total displacement derivatives with respect to the current reference configurations. The presented incremental-force based formulation can be considered as a generalization of the use of logarithmic strains. [ABSTRACT FROM AUTHOR]

Abstract

The aim of this study was to investigate the release performance of an electrospun composite drug loaded nanofiber mat. Electrospun nanofiber mats are promising as drug carriers which offer site-specific delivery of drugs to a target in the human body and may be used for cancer therapy. The authors have formulated a simple radial 1D finite element, which is used to model diffusion within fibers releasing a drug to the surrounding medium discretized by continuum 3D finite elements. The numerical model includes degradation effects and hydrophobicity at the fibers/surroundings interface. For the purpose of experimental investigation, a poly(D,L-lacticco-glycolic acid) (PLGA) implant has been created at the Faculty of Technology and Metallurgy, University of Belgrade. The radial 1D element provides accurate predictions of the diffusion process and serves as an efficient tool for describing transport inside the polymer fiber and surrounding porous medium; which is illustrated through numerical examples.
