The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has caused a global pandemic with dramatic health and socioeconomic consequences. The Coronavirus Disease 2019 (COVID-19) challenges health systems to quickly respond by developing new diagnostic strategies that contribute to identify infected individuals, monitor infections, perform contact-tracing, and limit the spread of the virus. In this brief report, we developed a highly sensitive, specific, and precise “In-House” ELISA to correctly discriminate previously SARS-CoV-2-infected and non-infected individuals and study population seroprevalence. Among 758 individuals evaluated for anti-SARS-CoV-2 serology in the province of Tucumán, Argentina, we found a weak correlation between antibodies elicited against the RBD, the receptor-binding domain of the Spike protein, and the nucleocapsid (N) antigens of this virus. Additionally, we detected mild levels of anti-RBD IgG antibodies in 33.6% of individuals diagnosed with COVID-19, while only 19% showed sufficient antibody titers to be considered as plasma donors. No differences in IgG anti-RBD titers were found between women and men, neither in between different age groups ranging from 18 to 60. Surprisingly, individuals from a high altitude village displayed elevated and longer lasting anti-RBD titers compared to those from a lower altitude city. To our knowledge, this is the first report correlating altitude with increased humoral immune response against SARS-CoV-2 infection.

The Coronavirus Disease 2019 (COVID-19) associated represents a challenge for health systems that had to quickly respond developing new diagnostic and therapeutic strategies. In the present work, we developed an “In House” ELISA with high sensitivity (92.2 %), specificity (100%) and precision (93.9%), with an area under the ROC curve (AUC) of 0.991, rendering the assay as an excellent serological test to correctly discriminate between SARS-COv-2 infected and non-infected individuals and study population seroprevalence. Among 758 patients evaluated for SARS-CoV-2 diagnosis in the province of Tucumán, Argentina, we found a Pearson correlation coefficient of 0.5048 between antibodies elicited against the RBD and the nucleocapsid (N) antigen. Additionally, 33.6% of individuals diagnosed with COVID-19 displayed mild levels of RBD-IgG antibodies, while 19% of the patients showed high antibody titers. Interestingly, patients with SARS-COV-2 infection over 60 years old elicited significantly higher levels of IgG antibodies against RBD compared to younger ones, while no difference was found between women and men. Surprisingly, individuals from a high altitude village displayed statistically significant higher and longer lasting anti-RBD antibodies compared to those from a city at a lower altitude, suggesting that a hypobaric hypoxia-adapted mechanism may act as a protective factor for COVID-19. To our knowledge, this is the first report correlating altitude with increased humoral immune response against SARS-Cov-2 infection.

The search for new biomaterials with superior mechanical properties is the focus in the area of materials science. A promising pathway is drawing inspiration from nature to design and develop materials with enhanced properties. In this work, a novel strategy to produce functionalized supramolecular bionanomaterials from the microbial biofilm is reported. Tuneable biofilms with specific characteristics were obtained by controlling the culture condition of the microorganism. When the exopolysaccharide (EPS) production was desired the tryptone was the best nutritional component for the EPS production into the biofilm. However, for the expression of a high amount of amyloid protein the combination of peptone and glucose was the best nutritional choice. Each biofilm obtained showed its owner rheology properties. These properties were altered by the addition of extracellular DNA, which increased the viscosity of the biofilm and induced a viscoelastic hydrogel behavior. Besides, as a proof of concept of bionanomaterial, a novel supramolecular polymeric hybrid EPS-Amyloid protein (EPAP) was obtained from the biofilm and it was tested as a new natural functionalized support for enzyme immobilization. The results suggest that this technology could be used as a new concept to obtain biomaterials from biofilms by controlling the nutritional conditions of a microorganism. Understanding environmental factors affecting biofilm formation will help the development of methods for controlling biofilm production and therefore obtaining new biomaterials.

Here we report a novel strategy for the immobilization of invertase using amyloid-like fibrils as a support. Optimal conditions to get Tyr-Tyr covalent binding between invertase and the support were determined using a photocrosslinking approach. The biological fibrils with invertase activity turn into microstructured catalysts according to electron microscopy outcomes. Thermal and storage stability as well as optimal pH and temperature of the enzyme were conserved. Moreover, the immobilized enzyme recovered by low g-force centrifugation retained 83% of its initial enzymatic activity after 15 reuse cycles. Considering that enzyme cost is the most significant part of the overall fee of enzymatic biomass conversion, the highly efficient recovery/reuse strategy described herein becomes relevant. Nanotechnology is a wide developing area of the biotechnology since the important applications of nanoparticles (NPs) in different technologies. The NPs produced by green technologies have many advantages such as greater surface area, high catalytic activity, in addition to providing a suitable contact between the metal salt and enzyme. Fungi secrete proteins, enzymes and reducing agents which can be used for the synthesis of metal NPs from metal salts. The biosynthesis of metal NPs by fungi has been explored in recent years, evaluating the extracellular and intracellular chemistry of formation. Emphasis has been given to the potential of metal NPs as an antimicrobial agent to inhibit the growth of pathogenic bacteria and fungi and other potential applications such as their cytotoxic activity against cancer cell lines. Inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis, are important problems in industrialized countries. The complete aetiology of both diseases is still unknown but likely involves genetic, environmental and immunological factors. The aim of this work is to study the anti-inflammatory mechanisms reported for yoghurt in a colon cancer model in order to evaluate its usefulness in the treatment of intestinal inflammation such as Crohn's disease. A trinitrobenzenesulfonic acid (TNBS)-induced colitis model was used. The influence of yoghurt feeding was studied before and after TNBS inoculation. The effect on the intestinal microbiota and on the host immune response was evaluated. IgA-producing cells and cells positive for specific cytokines (IL-12, IL-17, IFNγ and IL-10) were analyzed. Yoghurt administration diminished the severity of inflammation in the TNBS-inoculated mice. This effect occurred mainly through IL-10, which was increased in the intestinal tissues throughout the study, and by the decrease observed in IL-17 and IL-12 levels. In addition to this immunomodulatory capacity, another mechanism by which yoghurt could exert the anti-inflammatory activity observed in our model would be through beneficial changes in the intestinal microbiota (increases in the bifidobacteria and lactobacilli populations). These changes in the intestinal microbiota could also be considered one of the causes of the intestinal inflammation reduction. These results show that yoghurt administration modulated the immune response, inducing down regulation of the inflammatory cytokines produced by the immune cells involved in the inflammatory process. The close relationship between protein aggregation and neurodegenerative diseases has been the driving force behind the renewed interest in a field where biophysics, neurobiology and nanotechnology converge in the study of the aggregate state. On one hand, knowledge of the molecular principles that govern the processes of protein aggregation has a direct impact on the design of new drugs for high-incidence pathologies that currently can only be treated palliatively. On the other hand, exploiting the benefits of protein aggregation in the design of new nanomaterials could have a strong impact on biotechnology. Here we review the contributions of our research group on novel neuroprotective strategies developed using a purely biophysical approach. First, we examine how doxycycline, a well-known and innocuous antibiotic, can reshape α-synuclein oligomers into non-toxic high-molecular-weight species with decreased ability to destabilize biological membranes, affect cell viability and form additional toxic species. This mechanism can be exploited to diminish the toxicity of α-synuclein oligomers in Parkinson’s disease. Second, we discuss a novel function in proteostasis for extracellular glyceraldehyde 3-phosphate dehydrogenase (GAPDH) in combination with a specific glycosaminoglycan (GAG) present in the extracellular matrix. GAPDH, by changing its quaternary structure from a tetramer to protofibrillar assembly, can kidnap toxic species of α-synuclein, and thereby interfere with the spreading of the disease. Finally, we review a brighter side of protein aggregation, that of exploiting the physicochemical advantages of amyloid aggregates as nanomaterials. For this, we designed a new generation of insoluble biocatalysts based on the binding of photo-immobilized enzymes onto hybrid protein:GAG amyloid nanofibrils. Biocatalysis for industrial application is based on the use of enzymes to perform complex transformations. However, these systems have some disadvantage related to the costs of the biocatalyst. In this work, an alternative strategy for producing green immobilized biocatalysts based on biofilm was developed.A study of the rheological behavior of the biofilm from Bacillus sp. Mcn4, as well as the determination of its composition, was carried out. The dynamic rheological measurements, viscosity (G") and elasticity (G') module, showed that the biofilm presents appreciable elastic components, which is a recognized property for enzymes immobilization. After the partial purification, the exopolysaccharidewas identified as a levan with a non-Newtonian behavior. Extracellular DNA with fragments between 10,000 and 1000bp was detected also in the biofilm, and amyloid protein in the extracellular matrix using a fluorescence technique was identified. Bacillus sp. Mcn4 biofilms were developed on different surfaces, being the most stable those developed on hydrophilic supports. The biofilm showed lipase activity suggesting the presence of constitutive lipases entrapped into the biofilm. Indeed, two enzymes with lipase activity were identified in native PAGE. These were used as biocatalysts, whose reuse showed a residual lipase activity after more than one cycle of catalysis. The components identified in the biofilm could be the main contributors of the rheological characteristic of this material, giving an exceptional environment to the lipase enzyme. Based on these findings, the current study proposes green and natural biopolymers matrix as support for the enzyme immobilization for industrial applications.

Herein, we report the preparation and characterization of a new biocatalyst based on the photo-immobilization of a lipase onto hybrid protein : sugar nanofibrils. The nanofibrils are obtained through the aggregation of hen white egg lysozyme induced by the highly sulfated glycosaminoglycan heparin. The new hybrid nanomaterial could be easily functionalized using photochemical reaction to attach lipases through dityrosine covalent bonds. Compared to the free enzyme, the photo-immobilized lipase has better thermostability and enhanced resistance to non-conventional environment. Structural and morphological characterization of the nanofibrils shows that they are compatible with amyloid-like aggregates. In addition, the supramolecular arrangement of heparin and lysozyme within the building unit of the nanofibrils is also proposed. The procedure reported herein could be useful to design a new generation of insoluble biocatalyst by a single photo-click step which is definitely cleaner and faster than conventional chemical cross-linked procedures. Lactic acid bacteria (LAB) found in numerous fermented products can interact with the gut associated lymphoid tissue increasing antibody (principally secretory IgA) production. IgA secreting cells can repopulate not only the lamina propria but also they can go to other distant sites such as bronchus, urogenital tract and mammary glands a phenomenon known as the IgA cycle. Later studies have shown that both B cells of other isotypes and T cells from Peyer's patches also exhibit gut-seeking properties. The aim of this study was to study the effect of different feeding periods of L. casei CRL 431 on the interaction with the immune cells in Peyer's patches studying the migration of not only the IgA+ cells, but also other immune cells (T lymphocytes) to other mucosal sites such as bronchus and mammary glands. BALB/c mice were fed with L. casei CRL 431 during 2, 5 or 7 days. At the end of the feeding period, the mice were killed and the small intestine, the lung and the mammary glands were removed. IgA+ cells and CD4+ and CD8+ T lymphocytes were counted in tissue slices using direct immunofluorescence. IgA+ cells increased in the intestine samples taken after 7 days of LAB feeding. In mammary gland and lung tissues, IgA+ cells increased after five days of feeding. CD4+ and CD8+ T lymphocytes were not able to migrate to sites distant from the intestine and their number did not increase in the lamina propria of the small intestine. L. casei CRL 431 was able to stimulate the IgA cycle without proliferation of T population. These results allow us to suggest that this LAB could be used as oral adjuvant to protect mucosal surfaces from intestinal and respiratory diseases, and would also be useful as an adjuvant to protect against mammary glands pathologies such as cancer.

Crohn's disease and ulcerative colitis, two forms of inflammatory bowel disease, are important problems in industrialized countries. The complete etiology of these two diseases is still unknown but likely involves genetic, environmental, and immunological factors. The aim of the present work was to determine whether the anti-inflammatory effects reported for yoghurt in acute trinitrobenzene sulfonic acid-induced intestinal inflammation in mice also could prevent or attenuate the recurrent intestinal inflammation, thus maintaining remission. The innate response also was evaluated through participation of Toll-like receptors (TLRs) and the analysis of T-cell populations to determine the effects of yoghurt in an acute inflammatory bowel disease model. Yoghurt exerted a beneficial effect on acute intestinal inflammation by regulating T-cell expansion and modulating the expression of TLRs, with decrease of TLR4+ and increase of TLR9z cells. The anti-inflammatory effect of yoghurt also was demonstrated in a recurrent inflammation model. Yoghurt administration during the remission phase prevented the recurrence of inflammation without producing undesirable side effects. Prebiotics are non-digestible but fermentable oligosaccharides that can influence the composition and the activity of some intestinal bacteria to promote the health of the host. Smallanthus sonchifolius (yacon) contains beta-1,2-oligofructans as the main saccharides and its roots are consumed in South American countries. The aim of the study was to evaluate the prebiotic property of yacon root flour. Its influence on the intestinal microbiota and gut immune system were evaluated using a mice model. The results show the prebiotic effects of yacon root flour, stimulating the growth of bifidobacteria and lactobacilli and the intestinal immune system with increases in IgA and different cytokines. Cells from the innate response were mainly involved in the effect of yacon root flour. T cells were also activated and able to induce IL-10 and IFNg production. The long term administration of yacon root flour maintained the intestinal homeostasis without inflammatory effect regulated mainly through IL-10 and IL-4 regulatory cytokines. The effect of the long-term administration of commercial fermented milk containing probiotic bacteria in the mucosal immune response and peritoneal macrophages was analyzed. BALB/c mice were fed with fermented milk for 98 consecutive days. Small and large intestines were removed for histology; IgA, CD4, CD8 cells and cytokines-producing cells were counted. The influence on the immune cells associated with bronchus and mammary glands as well as on peritoneal macrophages was also analyzed. Continuous oral administration of fermented milk increased IgA+ cells in both parts of the intestine (small and large intestine). IL-10, a regulatory cytokine, increased in the intestinal cells in most samples. TNFα, IFNγ and IL-2 producing cells were also enhanced. Values for CD4 and CD8+ cell populations in lamina propria of the intestine were increased in relation to the control throughout the assay. No modifications in the histology of intestines were observed. Long-term consumption of fermented milk enhanced intestinal mucosa immunity, mediated by IgA+ cells and by cytokine production. This improvement of gut immunity was maintained and down-regulated by cytokines such as IL-10, preventing gut inflammatory immune response. The effect of this fermented milk on mucosal sites distant to the gut, such as bronchus and mammary glands, showed that in both tissues the increase in IgA+ cells was only observed at the beginning of the continuous consumption and no modifications in the number of cytokine positive cells were found. Similar observations were found when phagocytic activity of peritoneal macrophages was measured. It was demonstrated that the most evident effect of long-term consumption of fermented milk was observed in the intestine. Immunodulatory effects and the maintenance of intestinal homeostasis without secondary effects after long-term administration of fermented milk were also observed. Fil: de Moreno, Maria Alejandra. The aim of this work was to study the effect of the long-term cyclic administration of the probiotic strain of Lactobacillus (L.) casei CRL 431 as a mucosal immunomodulator of the immune cells associated with the lamina propria of the small intestine, bronchus and other immune cells not associated with mucosal tissues, such as peritoneal macrophages. BALB/c mice were orally administered with a suspension of Lc 109 cfu/ day/animal in non-fat milk (NFM) 10% for two consecutive days, the optimal dose selected in previous studies to reach protective immunity. This administration was repeated cyclically every 5 days for 98 days. Mice in the control group received only NFM 10%. Samples were taken after two days of L. casei administration and every 14 days until day 98. The small intestine and lungs were removed for histological slices preparation. Haematoxilin-eosin stains were made for histological studies of the small intestine. The number of IgA producing cells in the lamina propria of the small intestine and in bronchus was determined by immunofluorescence assays. Regulatory (IL-4, IL-10) and proinflammatory (TNF-α, INF-γ) cytokines were measured in the gut. Peritoneal macrophages were collected during the same periods for phagocytosis assays. We determined an increase in the number of IgA+ cells in the lamina propria of the small intestine in all the periods assayed and in BALT only until day 28. The cytokines studied (IL-10, IL-4, TNF-α and INF-γ) increased in most of the periods assayed, the effect being more remarkable for the anti-inflammatory cytokines such as IL-4 and IL-10. INF-γ was also increased but no modifications in the histological studies of the small intestine were observed, suggesting other roles for this cytokine The phagocytic activity of PM increased for most of the periods assayed. We demonstrated that long-term cyclic oral L. casei administration favors mucosal immunity and modulates the immune response to maintain the homeostasis at the mucosal level.
