Concerned about reports of a 15% decline in HIV testing among high-risk youth in an earlier study in Pittsburgh, this study was initiated to explore reasons why young people are not getting tested for HIV, while gathering data on their respective level of risk taking behaviors. A total of 580 surveys were collected from youth aged between 14 and 24. Overall, 17% of youth were found to be at high to some risk for HIV infection, while many of them agreed they were not at-risk for HIV. Young MSM reported being afraid of testing HIV positive. Young people continue to engage in high-risk behaviors and avoid HIV testing, and efforts are needed to identify these youth and equip them with the resources they need to avoid infection. We describe the efforts of a 4-city campaign to recruit Black and Hispanic men who have sex with men into an established HIV epidemiological study. The campaign used community organizing principles and a social marketing model that focused on personnel, location, product, costs and benefits, and promotion. The campaign was developed at the community, group, and individual levels to both increase trust and reduce barriers. The proportion of Hispanic men recruited during the 2002-2003 campaign doubled compared with the 1987 campaign, and the proportion and number of White men decreased by 20%. The proportion of Black men decreased because of the large increase in Hispanic men, although the number of Black men increased by 56%. Successful recruitment included training recruitment specialists, involving knowledgeable minority community members during planning, and having an accessible site with convenient hours. The Centers for Disease Control reported in October 1991 that many people at risk for human immunodeficiency virus (HIV) infection had not been tested for antibodies to HIV. This study identifies differences among 110 gay and bisexual men in three small cities in Pennsylvania who decided whether to be tested for antibodies to HIV and, if so, whether to return for results. These men were given self-administered questionnaires and were offered free and confidential HIV antibody tests. Fifty percent of the men refused testing. Of those tested, only 35 percent returned to obtain test results. Contrary to other health prevention data, education was significantly and inversely related to being tested and to returning for results. Men who most often participated in the institutionalized gay community were least likely to be tested. The findings suggest that gay men who are most aware of the potential psychosocial problems associated with HIV antibody testing are more likely to avoid testing. [ABSTRACT FROM AUTHOR]

Background: We investigated sexual-orientation differences in typologies of self-reported familial and non-familial warmth in childhood (before age 11) and adolescence (ages 11-17); and tested whether warmth explained sexual minority emerging adults' (ages 18-25) heightened odds of having heavier alcohol use trajectories (AUTs) and heightened risk for past-year alcohol use disorder (AUD) compared to completely heterosexuals., Methods: Using self-reported data from the U.S.-based Growing Up Today Study cohort, latent class analyses identified typologies of familial and non-familial warmth during childhood and adolescence. Multivariable regression models tested our objectives., Results: Six warmth classes emerged, including: High-High (i.e., high familial and high non-familial warmth, respectively); High-Moderate; Moderate-Moderate; Moderate-Occasional; Occasional-Occasional; and Low-Low. Among women, sexual minorities had higher odds than completely heterosexuals of being in the Moderate-Moderate, Moderate-Occasional, and Occasional-Occasional versus the High-High warmth class. There were not significant associations between sexual orientation and warmth classes for men. Lower warmth classes were generally associated with greater past-year AUD, and mediated heightened disparities in AUD for sexual minority women versus completely heterosexual women (4.3% mediated), but not among men. Objectives: We sought to calculate HIV incidence in a retrospective cohort of young (13-29 years old) black men who have sex with men (YBMSM) accessing repeated HIV-antibody testing in a mid-size city in the USA., Methods: We aggregated site-specific HIV-antibody testing results from the project's inception among YBMSM who received an initial negative result and accessed at least one additional HIV-antibody test. From these data, we assessed number of seroconversions and person-years and calculated HIV incidence using a mid-P exact test to estimate 95% CIs., Results: Five seroconversions were documented over 42.3 person-years (the mean age at first onsite test: 19.7 years), resulting in an HIV incidence rate of 11.8% (95% CI 4.3% to 26.2%). The mean age at seroconversion was 20.4 (±3.0) years., Conclusions: Even in mid-size cities with low HIV prevalence rates in the general population, HIV incidence among YBMSM may be high. Community-based HIV-antibody testing organisations serving YBMSM should be encouraged and trained to track repeated HIV testing and calculate HIV incidence rates. Aims: We estimated sexual-orientation differences in alcohol use trajectories during emerging adulthood, and tested whether alcohol use trajectories mediated sexual-orientation differences in alcohol use disorders (AUDs)., Design: Longitudinal self-reported survey data from the Growing Up Today Study., Setting: United States., Participants: A total of 12 493 participants aged 18-25 during the 2003, 2005, 2007 or 2010 surveys., Measurements: Stratified by gender, longitudinal latent class analyses estimated alcohol use trajectories (using past-year frequency, quantity and binge drinking from 2003 to 2010). Multinomial logistic regression tested differences in trajectory class memberships by sexual orientation [comparing completely heterosexual (CH) participants with sexual-minority subgroups: mainly heterosexual (MH), bisexual (BI) and gay/lesbian (GL) participants]. Modified Poisson regression and mediation analyses tested whether trajectories explained sexual-orientation differences in AUDs (past-year DSM-IV abuse/dependence in 2010)., Findings: Six alcohol use trajectory classes emerged for women and five for men: these included heavy (23.5/36.9% of women/men), moderate (31.8/26.4% of women/men), escalation to moderately heavy (9.7/12.0% of women/men), light (17.0% for women only), legal (drinking onset at age 21; 11.1/15.7% of women/men) and non-drinkers (7.0/9.1% of women/men). Compared with CH women, MH and BI women had higher odds of being heavy, moderate, escalation to moderately heavy and light drinkers versus non-drinkers (odds ratios = 2.02-3.42; P-values < 0.01-0.04). Compared with CH men, MH men had higher odds of being heavy, moderate and legal drinkers versus non-drinkers (odds ratios = 2.24-3.34; P-values < 0.01-0.01). MH men and women, BI women and GLs had higher risk of AUDs in 2010 than their same-gender CH counterparts (risk ratios = 1.34-2.17; P-values < 0.01). Alcohol use trajectories mediated sexual-orientation differences in AUDs for MH and GL women (proportion of effect mediated = 30.8-31.1%; P-values < 0.01-0.02), but not for men., Conclusions: In the United States, throughout emerging adulthood, several sexual-minority subgroups appear to have higher odds of belonging to heavier alcohol use trajectories than completely heterosexuals. The volatile terpenic and norisoprenoids profile from elderflowers (Sambucus nigra L.) was established for two cultivars by multidimensional gas chromatography. From 47 monoterpenic, 13 sesquiterpenes and 5 norisoprenoids components, 38 are reported for the first time on elderflowers. Elderflower seasonality implies proper handling and storage conditions, for further processing, thus the impact of freezing, freeze-drying, air drying and vacuum packing, was evaluated on these potential aroma metabolites. The most suitable preservation methods, regarding the total metabolites content, were vacuum packing and freezing for intermediary storage times (24-32weeks) with a reported overall decrease of the volatile terpenic and norisoprenoids of up to 58.6%; and freezing, for longer period (52weeks), with a decrease of up to 47.4% (compared to fresh elderflowers). The Strecker aldehydes formed during the reaction between α-amino acids (phenylalanine or methionine) and either gallic acid, caffeic acid or (+)-catechin ortho-quinones were evaluated in wine-model systems. It was demonstrated that phenylacetaldehyde was formed by quinone intermediates at wine pH. The highest amounts of phenylacetaldehyde during the 10days of experiment (69±5µg/L/day; 7x>Control) were obtained from (+) catechin, followed by gallic acid (61±4µg/L/day; 6x>Control) and caffeic acid (41±4µg/L/day; 4x>Control). Bisexual men experience significant health disparities likely related to biphobia. Biphobia presents via several preconceptions, including that bisexuality is transitory, and that bisexual men act as viral bridges between men who have sex with men and heterosexual populations. We analyzed data from a prospective cohort of gay and bisexual men, the Multicenter AIDS Cohort Study, to test these preconceptions. Men reporting both male and female sexual partners (MSMW) between 2002 and 2009 (n = 111) were classified as behaviorally bisexual. We assessed five hypotheses over two domains (transience of bisexual behavior and viral bridging). No evidence was found supporting the transitory nature of bisexuality. Trajectories of bisexual behavior were not transient over time. We found little evidence to support substantial viral bridging behavior. Notably, HIV-positive MSMW reported lower proportions of female partners than HIV-negative MSMW. Our results provide no empirical support for bisexual transience and scant support for viral bridging hypotheses. Our results provide key data showing that male bisexual behavior may be stable over long time periods and that behaviorally bisexual men's risk to female sexual partners may be lower than expected.

Though functional social support has been shown to serve as a protective factor for HIV viral load suppression in other populations, scant research has examined this relationship among men who have sex with men (MSM) in the United States. We assessed characteristics of social support, effects of social support on HIV viral load, and moderation by social support of the relationship between psychosocial indicators of a synergistic epidemic (syndemic) and HIV viral load. We analyzed longitudinal data from HIV-positive MSM using antiretroviral therapy who were enrolled in the Multicenter AIDS Cohort Study between 2002 and 2009 (n = 712). First, we conducted reliability assessments of a one-item social support measure. Then, we conducted a series of generalized longitudinal mixed models to assess our research questions. Moderation was assessed using an interaction term. A three-level (low/medium/high) social support variable demonstrated high reliability (intraclass correlation coefficients  = 0.72; 95% CI: 0.70, 0.75). Black and Hispanic MSM reported lower social support than their White counterparts (p < .0001). Recent sero-conversion was associated with higher social support (p < .05). Higher numbers of concomitant syndemic indicators (depression, polysubstance use, and condomless anal sex) were associated with lower social support (p < .0001). Medium and high social support levels were associated with greater viral load suppression and lower viral load means (p < .0001). Social support moderated the relationships between syndemic and HIV viral load (p < .05). HIV-positive MSM, particularly those of color, may benefit greatly from interventions that can successfully boost functional social support. Creating strengths-based interventions may also have particularly high impact among HIV-positive MSM with the highest psychosocial burdens.

Introduction: The design of efficient therapeutic delivery devices has become a tremendously active area of research with a strong contribution from the layer-by-layer (LbL) technology. The application of this simple yet firmly established technique for the design of drug reservoirs originates a multitude of multilayered systems of tailored architecture and with a high level of control of drug administration. Areas covered: This review will focus on the most recent and original research on LbL assemblies based on biopolymers including polysaccharides, polypeptides and proteins, with potential use in drug delivery. Herein, drug reservoirs consisting of multilayered planar films and capsules will be examined with emphasis on the ones benefiting from the non-cytotoxic and biocompatible nature of biopolymers, which are suitable to load, protect and release a high payload of toxic and fragile drugs. Expert opinion: The combination of biopolymers with LbL technology has undergone extensive research, still, there is a multitude of R&D opportunities for the design of smart drug delivery systems with distinct multilayered morphologies, low immunological response, non-invasive drug release devices, as well as the design of theranostic systems combining diagnostics and therapeutic features. Further developments in terms of scaling towards mass production in the pharmaceutical industry are expected in the long-term.

Besides being traditionally used to relieve hepatobiliary disorders, Cynara cardunculus L. has evidenced anticancer potential on triple-negative breast cancer (TNBC). This study highlights the antiproliferative effects of lipophilic extracts from C. cardunculus L. var. altilis (DC) leaves and florets, and of their major compounds, namely cynaropicrin and taraxasteryl acetate, against MDA-MB-231 cells. Our results demonstrated that MDA-MB-231 cells were much less resistant to leaves extract (IC 50 10.39 µg/mL) than to florets extract (IC 50 315.22 µg/mL), during 48 h. Moreover, leaves extract and cynaropicrin (IC 50 6.19 µg/mL) suppressed MDA-MB-231 cells colonies formation, via an anchorage-independent growth assay. Leaves extract and cynaropicrin were also assessed regarding their regulation on caspase-3 activity, by using a spectrophotometric assay, and expression levels of G2/mitosis checkpoint and Akt signaling pathway proteins, by Western blotting. Leaves extract increased caspase-3 activity, while cynaropicrin did not affect it. Additionally, they caused p21 Waf1/Cip1 upregulation, as well as cyclin B1 and phospho(Tyr15)-CDK1 accumulation, which may be related to G2 cell cycle arrest. They also downregulated phospho(Ser473)-Akt, without changing total Akt1 level. Cynaropicrin probably contributed to leaves extract antiproliferative action. These promising insights suggest that cultivated cardoon leaves lipophilic extract and cynaropicrin may be considered toward a natural-based therapeutic approach on TNBC., Competing Interests: The authors declare no conflict of interest.

Elderberry ( Sambucus nigra L.) lipophilic and polar extract dietary supplementation effects were evaluated according to diabetes management indices, using an in vivo model. A research pipeline was constructed, that ranged from extract preparation, partial chemical characterization and toxicity evaluation, to examining the elderberry extract dietary supplementation effects on biofluid and tissues. Extracts toxicity was screened using an Aliivibrio fischeri bioluminescence model. A concentration of up to 60 mg/L was selected, and rat doses for oral supplementation were computed applying the interspecies correlation between A. fischeri and rats. Wistar type 2 diabetic rats, induced by streptozotocin (STZ), were fed a high-fat diet and supplemented for 4 weeks at doses of 190 and 350 mg/kg body weight/day of lipophilic and polar extract, respectively. As far as we know, lipophilic elderberry extract supplementation was assessed for the first time, while polar extract was administrated at higher doses and for a shorter period compared to previous studies, aiming to evaluate subacute supplementation effects. The polar extract modulated glucose metabolism by correcting hyperglycemia, while the lipophilic extract lowered insulin secretion. Both extracts lowered insulin resistance, without remarkable alterations to hematological indices, sera lipids and sera and tissular trace element homeostasis. The main purpose of this study was to investigate the potential of suberin (a naturally occurring aromatic-aliphatic polyester ubiquitous to the vegetable realm) as a renewable source of chemicals and, in particular, to assess their physical properties. A comparison between cork and birch suberin fragments obtained by conventional depolymerisation processes (hydrolysis or methanolysis) is provided, focusing essentially on their thermal and crystallinity properties. It was found that suberin fragments obtained by the hydrolysis depolymerisation of birch had a high degree of crystallinity, as indicated by their thermal analysis and corroborated by the corresponding XRD diffractions, as opposed to hydrolysis-depolymerised cork suberin counterparts, which were essentially amorphous., (Copyright © 2016 Elsevier B.V. The lipophilic and phenolic extractives of the ripe mesocarp of four cherimoya cultivars ('Perry Vidal', 'Mateus I', 'Mateus III' and 'Funchal') from Madeira Island, were studied for the first time. The predominant lipophilic compounds are kaurene diterpenes (42.2-59.6%), fatty acids (18.0-35.6%) and sterols (9.6-23.7%). Kaur-16-en-19-oic acid is the major lipophilic component of all cultivars accounting between 554 and 1350mgkg(-1) of dry material. The studied fruits also contain a high variety of flavan-3-ols, including galloylated and non-galloylated compounds. Five phenolic compounds were identified for the first time: catechin, (epi)catechin-(epi)gallocatechin, (epi)gallocatechin, (epi)afzelechin-(epi)catechin and procyanidin tetramer. 'Mateus I' and 'Mateus III' cultivars present the highest content of phenolic compounds (6299 and 9603mgkg(-1) of dry weight, respectively). Insulin amyloid fibrils with enhanced aspect ratio, were prepared using a timesaving microwave assisted (MW) methodology, reducing the incubation time from 13 to 2h. The fibrillation process was followed indirectly by Thioflavin T Fluorescence and UV-vis analysis, by measuring the amount of β-sheets formed and the insulin present in solution, respectively. TEM and AFM analysis revealed that the insulin fibrils obtained through the MW method, have very similar lengths but are much thinner than the ones obtained using the conventional method (CM). Additionally, it was verified that the nature of the peptides present in the final insulin fibrils was not affected by microwave irradiation. Lipoxygenases (LOXs) are iron- or manganese-containing oxidative enzymes found in plants, animals, bacteria and fungi. LOXs catalyze the oxidation of polyunsaturated fatty acids to the corresponding highly reactive hydroperoxides. Production of hydroperoxides by LOX can be exploited in different applications such as in bleaching of colored components, modification of lipids originating from different raw materials, production of lipid derived chemicals and production of aroma compounds. Most application research has been carried out using soybean LOX, but currently the use of microbial LOXs has also been reported. Development of LOX composition with high activity by heterologous expression in suitable production hosts would enable full exploitation of the potential of LOX derived reactions in different applications. Here, we review the biological role of LOXs, their heterologous production, as well as potential use in different applications. LOXs may fulfill an important role in the design of processes that are far more environmental friendly than currently used chemical reactions. Difficulties in screening for the optimal enzymes and producing LOX enzymes in sufficient amounts prevent large-scale application so far. With this review, we summarize current knowledge of LOX enzymes and the way in which they can be produced and applied.

With the aim of producing materials with enhanced optical and photocatalytic properties, titanate nanotubes (TNTs) modified by cobalt doping (Co-TNT) and by Na(+)→ Co ion-exchange (TNT/Co) were successfully prepared by a hydrothermal method. The influence of the doping level and of the cobalt position in the TNT crystalline structure was studied. Although no perceptible influence of the cobalt ion position on the morphology of the prepared titanate nanotubes was observed, the optical behaviour of the cobalt modified samples is clearly dependent on the cobalt ions either substituting the Ti(4+) ions in the TiO6 octahedra building blocks of the TNT structure (doped samples) or replacing the Na(+) ions between the TiO6 interlayers (ion-exchange samples). The catalytic ability of these materials on pollutant photodegradation was investigated. First, the evaluation of hydroxyl radical formation using the terephthalic acid as a probe was performed. Afterwards, phenol, naphthol yellow S and brilliant green were used as model pollutants. Anticipating real world situations, photocatalytic experiments were performed using solutions combining these pollutants. The results show that the Co modified TNT materials (Co-TNT and TNT/Co) are good catalysts, the photocatalytic performance being dependent on the Co/Ti ratio and on the structural metal location. The Co(1%)-TNT doped sample was the best photocatalyst for all the degradation processes studied.

The integration of plant metabolomics to support preharvest fruit development studies can provide important insights into the biochemical mechanisms involved and lately support producers on harvesting management. A metabolomic-based strategy for fingerprinting of volatile terpenoids and norisoprenoids from Sambucus nigra L. berries from three cultivars, through ripening, was established. From 42 monoterpenic, 20 sesquiterpenic, and 14 norisoprenoid compounds, 48 compounds are reported for the first time as S. nigra berries components. Chemometric tools revealed that ripening was the factor that influenced more the volatile fraction profile and physicochemical parameters (pH, TS, and TSS), followed by cultivar. For the unripe stages, a higher overall content of the studied metabolites was observed, which gradually decreased over the ripening stages, being consistent for the three cultivars. These trends were mainly ruled by limonene, p-cymene, aromadendrene, β-caryophyllene, and dihydroedulan, which might therefore be used by producers as an additional simple decision making tool in conjunction with physicochemical parameters.

Transparent corrole grafted-chitosan films were prepared by chemical modification of chitosan with a corrole macrocycle, namely, 5,10,15-tris(pentafluorophenyl)corrole (TPFC), followed by solvent casting. The obtained films were characterized in terms of absorption spectra (UV-vis), FLIM (fluorescence lifetime imaging microscopy), structure (FTIR, XPS), thermal stability (TGA), thermomechanical properties (DMA), and antibacterial activity. The results showed that the chemical grafting of chitosan with corrole units did not affect its film-forming ability and that the grafting yield increased with the reaction time. The obtained transparent films presented fluorescence which increases with the amount of grafted corrole units. Additionally, all films showed bacteriostatic effect against S. aureus, as well as good thermomechanical properties and thermal stability. Considering these features, promising applications may be envisaged for these corrole-chitosan films, such as biosensors, bioimaging agents, and bioactive optical devices.

A series of bacterial cellulose (BC)/polycaprolactone (PCL) nanocomposite films were successfully prepared by supplementation of the BC culture medium with variable amounts of PCL powder followed by hot-pressing of the BC/PCL mixtures obtained after incubation. PCL powder was fully incorporated into the BC network during its production and did not change the BC network morphology. The obtained films showed a homogenous distribution of PCL throughout the BC network, as well as good thermal stability (up to 200 °C) and improved mechanical properties, when compared to pristine PCL. In addition, the intrinsic biodegradability and biocompatibility of the nanocellulose fibers and PCL opens the possibility of using this novel nanocomposite in the biomedical field and food packaging. Arabinose from arabinogalactan side chains was hypothesized as a possible binding site for chlorogenic acids in coffee melanoidins. To investigate this hypothesis, a mixture of 5-O-caffeoylquinic acid (5-CQA), the most abundant chlorogenic acid in green coffee beans, and (α1 → 5)-L-arabinotriose, structurally related to arabinogalactan side chains, was submitted to dry thermal treatments. The compounds formed during thermal processing were identified by electrospray ionization mass spectrometry (ESI-MS) and characterized by tandem MS (ESI-MS(n)). Compounds composed by one or two CQAs covalently linked with pentose (Pent) residues (1-12) were identified, along with compounds bearing a sugar moiety but composed exclusively by the quinic or caffeic acid moiety of CQAs. The presence of isomers was demonstrated by liquid chromatography online coupled to ESI-MS and ESI-MS(n). Pent1-2CQA were identified in coffee samples. A detailed study of the lipophilic composition of Codium tomentosum, Ulva lactuca, Gracilaria vermiculophylla and Chondrus crispus macroalgae cultivated in the Portuguese coast was carried out by gas chromatography-mass spectrometry before and after alkaline hydrolysis. Their long-chain aliphatic alcohols and monoglycerides compositions are reported for the first time. Additionally, other new compounds were also identified: phytol and neophytadiene in C. tomentosum, U. lactuca and G. vermiculophylla and stigmasterol, α-tocopherol and 24-methylenecholesterol in C. tomentosum. The lipophilic fraction of the studied macroalgae are mainly constituted by fatty acids (110.1-1030.5mgkg(-1) of dry material) and sterols (14.8-1309.1mgkg(-1) of dry material). C. tomentosum showed to be a valuable source of stigmasterol (1229.0mgkg(-1) of dry material) and α-tocopherol (21.8mgkg(-1) of dry material). The lipophilic composition of wild Arbutus unedo L. berries, collected from six locations in Penacova (center of Portugal), as well as some general chemical parameters, namely total soluble solids, pH, titratable acidity, total phenolic content and antioxidant activity was studied in detail to better understand its potential as a source of bioactive compounds. The chemical composition of the lipophilic extracts, focused on the fatty acids, triterpenoids, sterols, long chain aliphatic alcohols and tocopherols, was investigated by gas chromatography-mass spectrometry (GC-MS) analysis of the dichloromethane extracts. The lipophilic extractives of the ripe A. unedo berries ranged from 0.72% to 1.66% (w/w of dry weight), and consisted mainly of triterpenoids, fatty acids and sterols. Minor amounts of long chain aliphatic alcohols and tocopherols were also identified. Forty-one compounds were identified and among these, ursolic acid, lupeol, α-amyrin, linoleic and α-linolenic acids, and β-sitosterol were highlighted as the major components. To the best of our knowledge the current research study provides the most detailed phytochemical repository for the lipophilic composition of A. unedo, and offers valuable information for future valuation and exploitation of these berries.

Antimicrobial bacterial cellulose/poly(2-aminoethyl methacrylate) (BC/PAEM) nanocomposites were prepared by in situ radical polymerization of 2-aminoethyl methacrylate, using variable amounts of N,N-methylenebis(acrylamide) (MBA) as cross-linker. The obtained nanocomposites were characterized in terms of their structure, morphology, thermal stability, mechanical properties and antibacterial activity. The ensuing composite membranes were significantly more transparent than those of pure BC and showed improved thermal and mechanical properties. The antibacterial activity of the obtained nanocomposites was assessed towards a recombinant bioluminescent Escherichia coli and only the non-crosslinked nanocomposite (BC/PAEM) proved to have antibacterial activity., (Copyright © 2015 Elsevier Ltd. Objectives: The objective of this study is to determine associations between intertwining epidemics (syndemics) and HIV medication adherence and viral load levels among HIV-positive MSM and to test whether adherence mediates the relationship between syndemics and viral load., Design: We analysed participant data collected between 2003 and 2009 from the Multicenter AIDS Cohort Study, a prospective HIV/AIDS cohort study in four U.S. cities., Methods: We conducted longitudinal analyses (repeated measures mixed models) to assess whether differences in viral load levels, undetectable viral load and self-reported HIV medication adherence were associated with count of syndemic conditions (substance use, depression symptoms and sexual risk behaviour, range 0-3), adjusting for race/ethnicity, age and income. Mediation analyses were conducted using structural equation modelling and the SAS %mediate macro., Results: Syndemics count was associated with higher viral loads (P < 0.0001) and lower adherence (P < 0.0001). Increased counts of concomitant syndemics were associated with viral load (P < 0.01), detectable viral load (P < 0.05) and adherence (P < 0.001). Black MSM experienced worse outcomes across domains than white MSM (P < 0.0001) and experienced higher overall rates of syndemics (P < 0.01). Adherence significantly mediated the relationship between syndemics and viral load, accounting for an estimated 32.3% of the effect (P < 0.05)., Conclusion: Effectively lowering viral load levels among MSM has implications for both HIV/AIDS prevention and care. Our findings suggest that integrating substance use interventions, mental healthcare and sexual risk prevention into standard HIV care may be necessary to optimize treatment and Treatment as Prevention (TasP) models.

The chemical composition of the lipophilic extract of ripe pulp of banana fruit from several banana cultivars belonging to the Musa acuminata and Musa balbisiana species (namely 'Chinese Cavendish', 'Giant Cavendish', 'Dwarf Red', 'Grand Nain', 'Eilon', 'Gruesa', 'Silver', 'Ricasa', 'Williams' and 'Zelig') was studied by gas chromatography-mass spectrometry for the first time. The banana cultivars showed similar amounts of lipophilic extractives (ca. 0.4% of dry material weight) as well as qualitative chemical compositions. The major groups of compounds identified in these fractions were fatty acids and sterols making up 68.6-84.3% and 11.1-28.0%, respectively, of the total amount of lipophilic components. Smaller amounts of long chain aliphatic alcohols and α-tocopherol were also identified. These results are a relevant contribution for the valorisation of these banana cultivars as sources of valuable phytochemicals (ω-3 and ω-6 fatty acids, and sterols) with well-established beneficial nutritional and health effects., (Copyright © 2014 Elsevier Ltd. Nanobiotechnology has emerged as a fundamental domain in modern science, and metallic nanoparticles (NPs) are one of the largest classes of NPs studied because of their wide spectrum of possible applications in several fields. The use of plant extracts as reducing and stabilizing agents in their synthesis is an interesting and reliable alternative to conventional methodologies. However, the role of the different components of such extracts in the reduction/stabilization of metal ions has not yet been understood clearly. Here we studied the behavior of the main components of a Eucalyptus globulus Labill. bark aqueous extract during metal-ion reduction followed by advanced chromatographic techniques, which allowed us to establish their specific role in the process. The obtained results showed that phenolic compounds, particularly galloyl derivatives, are mainly responsible for the metal-ion reduction, whereas sugars are essentially involved in the stabilization of the NPs., (© 2014 WILEY-VCH Verlag GmbH & Co. The aim of this work was to examine the effect of Cu(II) on the degradation of thiram (Thi) in aqueous solutions, since the literature focused on this effect is scarce and copper based fungicides can be applied together with thiram or during the same season to agricultural crops. The effect of Cu(II) on the degradation of thiram was followed by both UV-vis and HPLC-MS/MS. When thiram is dissolved in pure water its degradation occurs very slowly, being negligible during the first 7 days. However, the presence of Cu(II) has a strong influence on the thiram degradation in aqueous solutions along time. In the presence of an excess of Cu(II), a [CuThi](2+) complex is initially formed which degrades into a complex formed between the dimethyldithiocarbamate anion (DMDTC) and Cu(II) ion, [Cu(DMDTC)](+). This complex further degrades leading to other copper complexes which were identified for the first time, by MS(n). The results obtained in the present work also demonstrated that a redox reaction involving DMDTC anions and Cu(II) ions gives rise to the formation of a Thi-Cu(I) complex. Finally, some of the complexes resulting from the degradation of [CuThi](2+) are quite persistent in solution for long periods of time (>1 month)., (Copyright © 2014 Elsevier B.V. Introduction: Bacterial cellulose (BC) is an extremely pure form of cellulose, which, due to its unique properties, such as high purity, water-holding capacity, three-dimensional nanofibrilar network, mechanical strength, biodegradability and biocompatibility, shows a high potential as nanomaterial in a wide range of high-tech domains including biomedical applications, and most notably in controlled drug-delivery systems., Areas Covered: This appraisal is intended to cover the major characteristics of BC, followed by the key aspects of BC production both in static and agitated conditions, and a glance of the major applications of BC, giving some emphasis to biomedical applications. Finally, a detailed discussion of the different applications of BC in controlled drug-delivery systems will be put forward, with focus on topical and oral drug-delivery systems, using either native BC or composite materials thereof., Expert Opinion: The limited number of studies carried out so far demonstrated that BC, or materials prepared from it, are interesting materials for drug-delivery systems. There is, however, a large field of systematic research ahead to develop new and more selectively responsive materials and eventually to conjugate them with other biomedical applications of BC under development.

Bacterial cellulose (BC) membranes were explored as novel nanostructured transdermal delivery systems for diclofenac sodium salt (a typical non-steroidal anti-inflammatory drug). Diclofenac sodium salt loaded BC membranes were prepared through a simple methodology, using glycerol as plasticizer, and characterized in terms of structure, morphology and swelling behavior. The membranes were very homogeneous, quite flexible and presented a considerably higher swelling behavior when compared with pure BC. In vitro diffusion studies with Franz cells, were conducted using human epidermal membranes, and showed that the incorporation of diclofenac in BC membranes provided similar permeation rates to those obtained with commercial patches and substantially lower than those observed with a commercial gel. This release profile together with the ease of application and the simple preparation and assembly of the drug-loaded membranes clearly indicates the enormous potentialities of using BC membranes for transdermal administration of diclofenac., (Copyright © 2014 Elsevier Ltd. Introduction: Men who have sex with men and women (MSMW) have been shown in cross-sectional studies to suffer HIV-related health disparities above and beyond those found among men who have sex with men only (MSMO). We conducted a secondary data analysis over a 7-year time frame of participants in the Multicenter AIDS Cohort Study, a long-standing prospective cohort study, to examine whether MSMW had persistently higher rates of depression symptoms, polydrug use, and (among HIV-positive men who have sex with men) HIV viral load levels compared with MSMO., Methods: Men were behaviorally defined as bisexual if they reported sexual activity with at least 1 male and 1 female partner between study waves 38 and 50. We used generalized mixed modeling with repeated measures to test differences in CES-D score, polydrug use, and viral load between sexually active MSMO (n = 1514) and MSMW (n = 111), adjusting for age, income, race/ethnicity, and recent seroconversion., Results: MSMW were significantly more likely than MSMO to have higher CES-D scores, polydrug use, and viral load levels (all P < 0.01). Outcome trajectories did not differ significantly over time between these groups. Black and Hispanic HIV-positive MSMW had higher viral load levels relative to white HIV-positive MSMW (P < 0.01)., Discussion: Compared with MSMO, MSMW in the Multicenter AIDS Cohort Study suffer from profound and persistent HIV-related health disparities across biological, behavioral, and psychosocial domains. Further qualitative and quantitative research contextualizing the pathways underlying these disparities is recommended for intervention development targeting MSMW at risk for HIV acquisition and transmission.

The present study reports the development of a new generation of bio-based nanocomposite proton exchange membranes based on bacterial cellulose (BC) and poly(4-styrene sulfonic acid) (PSSA), produced by in situ free radical polymerization of sodium 4-styrenesulfonate using poly(ethylene glycol) diacrylate (PEGDA) as cross-linker, followed by conversion of the ensuing polymer into the acidic form. The BC nanofibrilar network endows the composite membranes with excellent mechanical properties at least up to 140 °C, a temperature where either pure PSSA or Nafion are soft, as shown by dynamic mechanical analysis. The large concentration of sulfonic acid groups in PSSA is responsible for the high ionic exchange capacity of the composite membranes, reaching 2.25 mmol g(-1) for a composite with 83 wt % PSSA/PEGDA. The through-plane protonic conductivity of the best membrane is in excess of 0.1 S cm(-1) at 94 °C and 98% relative humidity (RH), decreasing to 0.042 S cm(-1) at 60% RH. These values are comparable or even higher than those of ionomers such as Nafion or polyelectrolytes such as PSSA. This combination of electric and viscoelastic properties with low cost underlines the potential of these nanocomposites as a bio-based alternative to other polymer membranes for application in fuel cells, redox flow batteries, or other devices requiring functional proton conducting elements, such as sensors and actuators.

Biopolymers often have unique properties of considerable interest as a basis for new materials. It is however not evident how to extract them from plants without destroying their chemical skeleton and inherent properties. Here we report the ex situ reconstitution of the biopolyester suberin as a new waterproof and antimicrobial material. In plant cell walls, suberin, a cross-linked network of aromatic and aliphatic monomers, builds up a hydrophobic protective and antimicrobial barrier. Recently we succeeded in extracting suberin from the plant cell wall using the ionic liquid cholinium hexanoate. During extraction the native three-dimensional structure of suberin was partially preserved. In this study, we demonstrate that this preservation is the key for its ex situ reconstitution. Without any chemical additives or purification, the suberin composing macromolecules undergo self-association on the casting surface forming a film. Suberin films obtained show barrier properties similar to those of the suberin barrier in plants, including a potentially broad bactericidal effect.

Bacterial cellulose (BC) is a highly pure form of cellulose, produced in the form of a swollen membrane by several bacteria that demonstrated to be able to modulate the skin release of model drugs. In the present study, the skin irritation potential of BC was evaluated in human subjects. BC membranes with and without glycerin (acting as plasticizer) were tested. No significant differences were observed for transepidermal water loss (TEWL) measurements in comparison with negative control, 2 and 24 h after patch removal, which is an indicator of an absence of barrier disruption. Similar results were found for erythema. Clinical scores were zero at both times for all volunteers, with the exception of five volunteers that exhibited weak reactions. BC with glycerin provided a skin moisturizing effect statistically higher than the negative control (p=0.044), which was not observed for BC alone. The good skin tolerance found after a single application under occlusion reinforces the putative interest of BC membranes as supports for drug topical delivery. Besides modifying the mechanical properties, the inclusion of glycerin results in a skin moisturizing effect which could be clinically relevant for the treatment for skin diseases characterized by dryness, such as psoriasis and atopic dermatitis., (Copyright © 2013 Elsevier B.V. Objectives: To estimate the number of men who have sex with men and women who are HIV-positive in the United States, and to compare HIV prevalence rates between men who have sex with men and women, men who have sex with men only, and men who have sex with women exclusively., Methods: Following PRISMA guidelines, we conducted a systematic review and meta-analysis of reports referencing HIV prevalence and men who have sex with men and women. We searched PubMed and Ovid PsycINFO for peer-reviewed, U.S.-based articles reporting on HIV prevalence among men who have sex with men and women. We conducted event rate, effect size, moderation and sensitivity analyses., Results: We estimate that 1.0% of U.S. males are bisexually-behaving, and that 121,800 bisexually-behaving men are HIV-positive. Men who have sex with men and women are less than half as likely to be HIV-positive as men who have sex with men only (16.9% vs. 33.3%; OR = 0.41, 95% CI: 0.31, 0.54), but more than five times as likely to be HIV-positive as men who have sex with women exclusively (18.3% vs. 3.5%; OR = 5.71, 95% CI: 3.47, 9.39). They are less likely to engage in unprotected receptive anal intercourse than men who have sex with men only (15.9% vs. 35.0%; OR = 0.36, 95% CI: 0.28, 0.46). Men who have sex with men and women in samples with high racial/ethnic minority proportions had significantly higher HIV prevalence than their counterparts in low racial/ethnic minority samples., Conclusions: This represents the first meta-analysis of HIV prevalence in the U.S. between men who have sex with men and women and men who have sex with men only. Data collection, research, and HIV prevention and care delivery specifically tailored to men who have sex with men and women are necessary to better quantify and ameliorate this population's HIV burden.

Compared with men who have sex with men only (MSMO), men who have sex with men and women (MSMW) consistently report higher rates of two HIV risk behaviors: transactional sex (TS) and concurrent substance use and sex (CSS). Within MSMW, little is known about how synergistic epidemics ("syndemics") affect TS and CSS. Using a sample of substance-using MSM (n = 515) in South Florida, we compared TS and CSS among MSMO and MSMW; examined whether, within MSMW (n = 86), TS and CSS predict unprotected anal intercourse with partners of serodiscordant/unknown HIV status (SU-UAI); and tested whether syndemics predict TS and CSS. MSMW reported higher rates of engaging in both TS and CSS (AOR = 1.7; 95 % CI 1.0-3.0). Within MSMW, engagement in both TS and CSS predicted SU-UAI (AOR = 3.3; 95 % CI 1.2-9.6); and syndemics predicted TS and CSS involvement (p < 0.01). Substance-using MSMW may benefit from interventions targeting TS, CSS, and background syndemics.

Ultra-high performance liquid chromatography (UHPLC) was applied for the first time in the analysis of wood extracts. The potential of this technique coupled to ion trap mass spectrometry in the rapid and effective detection and identification of bioactive components in complex vegetal samples was demonstrated. Several dozens of compounds were detected in less than 30min of analysis time, corresponding to more than 3-fold reduction in time, when compared to conventional HPLC analysis of similar extracts. The phenolic chemical composition of Eucalyptus grandis, Eucalyptus urograndis (E. grandis×E. urophylla) and Eucalyptus maidenii wood extracts was assessed for the first time, with the identification of 51 phenolic compounds in the three wood extracts. Twenty of these compounds are reported for the first time as Eucalyptus genus components. Ellagic acid and ellagic acid-pentoside are the major components in all extracts, followed by gallic and quinic acids in E. grandis and E. urograndis and ellagic acid-pentoside isomer, isorhamnetin-hexoside and gallic acid in E. maidenii. The antioxidant scavenging activity of the extracts was evaluated, with E. grandis wood extract showing the lowest IC50 value. Moreover, the antioxidant activity of these extracts was higher than that of the commercial antioxidant BHT and of those of the corresponding bark extracts. Lipophilic extracts of Cynara cardunculus L. var. altilis (DC) from the south of Portugal (Baixo Alentejo) were studied by gas chromatography-mass spectrometry. One sesquiterpene lactone, four pentacyclic triterpenes, and four sterols were reported for the first time as cultivated cardoon components, namely, deacylcynaropicrin, β- and α-amyrin, lupenyl and ψ-taraxasteryl acetates, stigmasterol, 24-methylenecholesterol, campesterol, and Δ(5)-avenasterol. In addition, other new compounds were identified: ten fatty acids, eight long-chain aliphatic alcohols, and six aromatic compounds. Four triterpenyl fatty acid esters were also detected. Sesquiterpene lactones and pentacyclic triterpenes were the major lipophilic families, representing respectively 2-46% and 10-89% of the detected compounds. Cynaropicrin was the most abundant sesquiterpene lactone, while taraxasteryl acetate was the main pentacyclic triterpene. Fatty acids and sterols, mainly hexadecanoic acid and β-sitosterol, were present at lower amounts (1-20% and 1-11% of the detected compounds). Long-chain aliphatic alcohols and aromatic compounds were detected at reduced abundances (1-6% of the detected compounds).

Novel nanostructured composite materials based on bacterial cellulose membranes (BC) and acrylate polymers were prepared by in situ atom transfer radical polymerization (ATRP). BC membranes were functionalized with initiating sites, by reaction with 2-bromoisobutyryl bromide (BiBBr), followed by atom transfer radical polymerization of methyl methacrylate (MMA) and n-butyl acrylate (BA), catalyzed by copper(I) bromide and N,N,N',N″,N″-pentamethyldiethylenetriamine (PMDETA), using two distinct initiator amounts and monomer feeds. The living characteristic of the system was proven by the growth of PBA block from the BC-g-PMMA membrane. The BC nanofiber sleeving was clearly demonstrated by SEM imaging, and its extent can be tuned by controlling the amount of initiating sites and the monomer feed. The ensuing nanocomposites showed high hydrophobicity (contact angles with water up to 134°), good thermal stability (initial degradation temperature in the range 241-275 °C), and were more flexible that the unmodified BC membranes.

In this study, the relevance of photodegradation processes on the persistence of the fungicide thiram in waters was investigated. The photodegradation of thiram in Milli-Q water and in aqueous solutions of humic and fulvic acids, as well as the photodegradation in spiked river water were studied. Both pure thiram and one of its commercial formulations were used to prepare the solutions which were irradiated in a solar light simulator. In general, thiram photodegradation follows pseudo-first order kinetics. The half-life time of thiram 2mgL(-1) in Milli-Q water was 28min. However, the degradation rate of thiram was significantly increased (p=0.02) by the inert components of the thiram commercial formulation as well as by commercial humic acids and by fulvic acids isolated from river water (p<0.004). Thus, the half-life time of thiram decreased to 24min in the presence of the inert formulation components, while, in the presence of both humic and fulvic acids (10mgL(-1)) it decreased to 22min. Furthermore, thiram photodegradation in natural river water showed that there is a significant enhancement of the degradation rate constant of thiram relatively to Milli-Q water, corresponding to a decrease of about 38% in its half-life time. This increase of the degradation rate in river water seems to be higher than that observed in the presence of FA, suggesting that beyond organic matter, other natural river components can increase the thiram photodegradation rate. These results allow us to conclude that photodegradation by solar radiation can be an important degradation pathway of thiram in natural waters. HPLC-MS/MS allowed to identify, for the first time, three products of the photodegradation of thiram in aqueous solution. Three compounds were identified and their structure was corroborated by the MS(n) spectra fragmentation profile. There has been a great deal of interest in the use of nanostructured bacterial cellulose membranes for biomedical applications, including tissue implants, wound healing, and drug delivery. However, as bacterial cellulose does not intrinsically present antimicrobial properties, in the present study, antimicrobial bacterial cellulose membranes were obtained by chemical grafting of aminoalkyl groups onto the surface of its nanofibrillar network. This approach intends to mimic intrinsic antimicrobial properties of chitosan. Interestingly, these novel grafted bacterial cellulose membranes (BC-NH2) are simultaneously lethal against S. aureus and E. coli and nontoxic to human adipose-derived mesenchymal stem cells (ADSCs) and thus may be useful for biomedical applications. In addition to these biological properties, the bioactive nanostructured BC-NH2 membranes also present improved mechanical and thermal properties.

Silver has been mainly investigated as an antibacterial agent and less as a fungicide in which concerns antimicrobial properties. In this research, the antifungal activity of composite films of pullulan and Ag nanoparticles (NP) against Aspergillus niger was evaluated using standard protocols. These new materials were prepared as transparent cast films (66-74 μm thickness) from Ag hydrosols containing the polysaccharide. Fungal growth inhibition was observed in the presence of such silver nanocomposite films. Moreover, disruption of the spores cells of A. niger was probed for the first time by means of scanning electron microscopy (SEM). This effect occurred in the presence of the nanocomposites due to Ag NP dispersed as fillers in pullulan. This polysaccharide was used here as a biocompatible matrix, hence making these nanocomposites beneficial for the development of antifungal packaging materials., (Copyright © 2012 Elsevier B.V. A series of bacterial cellulose-poly(2-hydroxyethyl methacrylate) nanocomposite films was prepared by in situ radical polymerization of 2-hydroxyethyl methacrylate (HEMA), using variable amounts of poly(ethylene glycol) diacrylate (PEGDA) as cross-linker. Thin films were obtained, and their physical, chemical, thermal, and mechanical properties were evaluated. The films showed improved translucency compared to BC and enhanced thermal stability and mechanical performance when compared to poly(2-hydroxyethyl methacrylate) (PHEMA). Finally, BC/PHEMA nanocomposites proved to be nontoxic to human adipose-derived mesenchymal stem cells (ADSCs) and thus are pointed as potential dry dressings for biomedical applications.

Too many people with HIV have left the job market permanently and those with reduced work capacity have been unable to keep their jobs. There is a need to examine the health effects of labor force participation in people with HIV. This study presents longitudinal data from 1,415 HIV-positive men who have sex with men taking part in the Multicenter AIDS Cohort Study. Generalized Estimating Equations show that employment is associated with better physical and mental health quality of life and suggests that there may be an adaptation process to the experience of unemployment. Post hoc analyses also suggest that people who are more physically vulnerable may undergo steeper health declines due to job loss than those who are generally healthier. However, this may also be the result of a selection effect whereby poor physical health contributes to unemployment. Policies that promote labor force participation may not only increase employment rates but also improve the health of people living with HIV.

Bacterial cellulose (BC) is a biomaterial with unique physical and mechanical properties that triggered considerable interest, but there are few studies addressing the use of such membranes for drug loading and controlled release. This study aimed to investigate the applicability of BC membranes in topical or transdermal drug delivery systems. To assess its therapeutic feasibility, the permeation through human epidermis of two model drugs (lidocaine hydrochloride and ibuprofen) in BC and other formulation systems was compared in vitro. A uniform distribution of both drugs in the BC membranes was achieved. Diffusion studies with Franz cells showed that the incorporation of lidocaine hydrochloride in BC membranes provided lower permeation rates than those obtained with the conventional formulations. However, the results obtained with the lipophilic drug were quite different, since permeation of ibuprofen in BC was almost three times higher than that of the drug in the gel or in a PEG400 solution. These results indicate that this technology can be successfully applied to modulate the bioavailability of drugs for percutaneous administration, which could be particularly advantageous in the design of delivery systems that have, simultaneously, the ability to absorb exudates and to adhere to irregular skin surfaces., (Copyright © 2012 Elsevier B.V. This paper reports on the ferromagnetic properties of aged Ti(1-x)CoxO(2-delta) anatase nanopowders with different Co contents (0.03 < or = x < or = 0.10). It is shown for the first time that aged Co:TiO2 anatase samples retain rather high values of magnetization (M), remanence (Mr) and coercivity (Hc) which provide strong evidence for a preserved long-range ferromagnetic order. Room temperature M, Mr and Hc values were measured in the ranges of [0.05, 0.79] microB/Co, [0.044, 0.096] microB/Co and [366.7,494.8]Oe, respectively, which are in the same range as in general reported either for newly prepared thin films or nanoparticles.
