Abstract

Asthma is one of the main non-communicable chronic diseases and affects a huge portion of the population. It is a multifactorial disease, classified into several phenotypes, being the allergic the most frequent. The pathophysiological mechanism of asthma involves a Th2-type immune response, with high concentrations of allergen-specific immunoglobulin E, eosinophilia, hyperreactivity and airway remodeling. These mechanisms are orchestrated by intracellular signaling from effector cells, such as lymphocytes and eosinophils. Ion channels play a fundamental role in maintaining the inflammatory response on asthma. In particular, transient receptor potential (TRP), stock-operated Ca2+ channels (SOCs), Ca2+-activated K+ channels (IKCa and BKCa), calcium-activated chloride channel (TMEM16A), cystic fibrosis transmembrane conductance regulator (CFTR), piezo-type mechanosensitive ion channel component 1 (PIEZO1) and purinergic P2X receptor (P2X). The recognition of the participation of these channels in the pathological process of asthma is important, as they become pharmacological targets for the discovery of new drugs and/or pharmacological tools that effectively help the pharmacotherapeutic follow-up of this disease, as well as the more specific mechanisms involved in worsening asthma. Abstract

Spirulina platensis has shown effectiveness in the treatment of allergic rhinitis in rats, but its action in tracheal reactivity or on markers of relaxation and antioxidant profile has not yet been possible to determine. In this paper, the animals were divided into the groups healthy (SG) and supplemented with S. platensis at doses of 50 (SG50), 150 (SG150), and 500 mg/kg (SG500). We also evaluated nitrite levels, lipid peroxidation, and antioxidant activity through biochemical analysis. For contractile reactivity, only SG500 (pEC50 = 5.2 ± 0.06 showed reduction in carbachol contractile potency. Indomethacin caused a higher contractile response to carbachol in SG150 and SG500. For relaxation, curves for SG150 (pEC50 = 5.0 ± 0.05) and SG500 (pEC50 = 7:3 ± 0:02) were shifted to the left, more so in SG500. We observed an increase in nitrite in the trachea only with supplementation of 500 mg/kg (54.0 ± 8.0 µM), also when compared to SG50 (37.0 ± 10.0 µM) and SG150 (38.0 ± 7.0 µM). We observed a decrease in lipid peroxidation in the plasma and an increase in oxidation inhibition for the trachea and lung in SG150 and SG500, suggesting enhanced antioxidant activity. S. platensis (150/500 mg/kg) decreased the contractile response and increased relaxation by increasing antioxidant activity and nitrite levels and modulating the inflammatory response.

Abstract

Lippia species (Verbenaceae) are widely used on folk medicine to treat respiratory and gastrointestinal disorders. In this work, we evaluated the relaxant effect of the essential oil from Lippia microphylla Cham. (LM‑EO) and its major components on guinea pig ileum as well as elucidate the involved mechanisms. LM-EO inhibited phasic contractions as well as relaxed tonic contractions induced by several contractile agents on guinea pig ileum. Similar results were observed in tests using thymol and carvacrol, the major constituents of LM-EO. Voltage-dependent calcium channel (CaV) blockade was suggested since LM-EO shifted to the right non parallelly CaCl2-induced cumulative contractions and confirmed when LM-EO relaxed the ileum pre contracted by S-(-)-Bay K8644, a selective CaV agonist. On circular layer, LM-EO inhibited phasic contractions, being equipotent to the effect observed in the intact ileum. In the myocytes from ileum longitudinal layer, LM-EO reduced fluorescence intensity induced by histamine, similarly to verapamil (CaV blocker), indicating a decrease in cytosolic concentration of Ca2+ ([Ca2+]c). In conclusion, LM-EO presents a spasmolytic activity on guinea pig ileum by CaV blockade and [Ca2+]c reduction. These results present a rationale for the use of L. microphylla as a potential product to intestinal disorders. Las especies del género Lippia (Verbenaceae) se utilizan ampliamente en la medicina tradicional para tratar desórdenes respiratorios y gastrointestinales. En este estudio, el efecto relajante del aceite esencial obtenido de Lippia microphylla Cham. (LM-EO) y sus componentes principales en el íleon de cobaya, así como los mecanismos implicados. LM-EO inhibió las contracciones fásicas y relajó la tonicidad inducida por varios agentes contráctiles en el íleon de cobaya. Se observaron resultados similares en los protocolos que utilizan timol y carvacrol, los componentes principales de LM-EO. Se sugirió el bloqueo de los canales de calcio dependientes de voltaje (CaV) ya que LM-EO se desplazó hacia la derecha y de manera no paralela las contracciones acumulativas inducidas por CaCl2 y se confirmó cuando LM-EO relajó el íleon precontraído por S‑(‑)‑Bay K8644, un agonista selectivo de CaV. En la capa muscular circular del íleon, LM-EO inhibió las contracciones fásicas, siendo equipotente en relación a los efectos sobre el íleon intacto. En miocitos aislados de la capa de íleon longitudinal, LM-EO redujo la intensidad de la fluorescencia inducida por histamina, de manera similar al verapamilo (bloqueador de CaV), lo que indica una reducción en la concentración de calcio citosólico ([Ca2+]c). Por tanto, LM-EO presenta actividad espasmolítica en el íleo de cobaya al bloquear el CaV y la consiguiente reducción de [Ca2+]c. Estos resultados se muestran como una justificación para el uso de L. microphylla como producto terapéutico en trastornos intestinales. As espécies do gênero Lippia (Verbenaceae) são amplamente usadas na medicina tradicional no tratamento de desordens respiratórias e gastrointestinais. Neste estudo, avaliou-se o efeito relaxante do óleo essencial obtido de Lippia microphylla Cham. (LM‑EO) e seus componentes majoritários em íleo isolado de cobaias bem como elucidou‑se os mecanismos envolvidos. LM-EO inibiu as contrações fásicas e relaxou as tônicas induzidas por vários agentes contráteis em íleo de cobaia. Resultados semelhantes foram observados nos protocolos utilizando-se timol e carvacrol, os componentes majoritários de LM-EO. O bloqueio dos canais de cálcio dependentes de voltagem (CaV) foi sugerido uma vez que LM-EO deslocou para a direita e de maneira não paralela as contrações cumulativas induzidas por CaCl2 e confirmou-se quando LM-EO relaxou o íleo pré-contraído por S‑(‑)‑Bay K8644, um agonista seletivo dos CaV. Na camada muscular circular do íleo, LM-EO inibiu as contrações fásicas, sendo equipotente em relação aos efeitos no íleo íntegro. No miócitos isolados da camada longitudinal do íleo, LM-EO reduziu a intensidade da fluorescência induzida por histamina, semelhantemente ao verapamil (bloqueador CaV), indicando uma redução na concentração citosólica de cálcio ([Ca2+]c). Dessa forma, LM-EO apresenta uma atividade espasmolítica em íleo de cobaia pelo bloqueio dos CaV e consequente redução da [Ca2+]c. Esses resultados demostram-se como uma justificativa para o uso de L. microphylla como um produto terapêutico nas desordens intestinais.

Abstract

Asthma is a chronic inflammatory disease of the airways related to epithelial damage, bronchial hyperresponsiveness to contractile agents, tissue remodeling, and luminal narrowing. Currently, there are many data about the pathophysiology of asthma; however, a new aspect has emerged related to the influence of reactive oxygen and nitrogen species (ROS and RNS) on the origin of this disease. Several studies have shown that an imbalance between the production of ROS and RNS and the antioxidant enzymatic and nonenzymatic systems plays an important role in the pathogenesis of this disease. Considering this aspect, this study is aimed at gathering data from the scientific literature on the role of oxidative distress in the development of inflammatory airway and lung diseases, especially bronchial asthma. For that, articles related to these themes were selected from scientific databases, including human and animal studies. The main findings of this work showed that the respiratory system works as a highly propitious place for the formation of ROS and RNS, especially superoxide anion, hydrogen peroxide, and peroxynitrite, and the epithelial damage is reflected in an important loss of antioxidant defenses that, in turn, culminates in an imbalance and formation of inflammatory and contractile mediators, such as isoprostanes, changes in the activity of protein kinases, and activation of cell proliferation signalling pathways, such as the MAP kinase pathway. Thus, the oxidative imbalance appears as a promising path for future investigations as a therapeutic target for the treatment of asthmatic patients, especially those resistant to currently available therapies.

Abstract

Erectile dysfunction is increasingly affecting men, from the elderly to young adults, being a sexual disorder related to the inability to generate or maintain a penile erection. This disorder is related to psychosocial factors such as anxiety, depression, and low self-esteem, to organic factors such as the presence of preexisting conditions like hypertension, diabetes and dyslipidemia. The pathophysiology of the disease is related to changes in the neurotransmission of the autonomic or the non-cholinergic non-adrenergic nervous system, as well as the release of local mediators, such as thromboxane A2 and endothelin, and hormonal action. These changes lead to impaired relaxation of cavernous smooth muscle, which reduces local blood flow and impairs penile erection. Currently, therapy is based on oral vasodilation, such as sildenafil, tadalafil, vardenafil and iodenafil, or by direct administration of these agents into the corpus cavernosum or by intraurethral route, such as alprostadil and papaverine. Despite this, studies that consolidate the understanding of its pathophysiological process contribute to the discovery of new more efficient drugs for the treatment of erectile dysfunction. In this sense, in the present work an extensive survey was carried out of the mechanisms already consolidated and the most recent ones related to the development of erectile dysfunction. Abstract

The essential oil of Lippia microphylla (LM-OE) presents several pharmacological activities. This work evaluates the tocolytic effect of LM-OE on rats. LM-OE inhibited phasic contractions and relaxed tonic contractions on rat uterus. Considering that nitric oxide (NO) pathway regulates uterine contraction, LM-OE potency was attenuated in the presence of NO synthase (NOS) inhibitor and this reduction was reversed in the presence of a NOS substrate. Similarly, the relaxant potency of LM-OE was reduced in the presence of soluble guanylyl cyclase (sGC) and protein kinase G (PKG) inhibitors. LM-OE also demonstrates a positive modulation of large and small conductance calcium-activated, voltage-gated and adenosine triphosphate-sensitive potassium channels and inhibited curves to CaCl2 as well as relaxed the uterus pre-contracted by S-(-)-Bay K8644, suggesting voltage-gated calcium channels type-1 (CaV1) blockade. Thus, the tocolytic effect of LM-OE on rat involves positive modulation of NO/NOS/sGC/PKG/K+-channels pathway and Ca2+ influx blockade through CaV1. [ABSTRACT FROM AUTHOR]
