Despite the known benefits of BFR exercise, it remains unclear whether or not the pressurization of blood vessels damages the endothelial cells or increases risk for formation of thrombi. Thus, the effects of BFR exercise on coagulation, fibrinolysis, or hemostasis, remains speculative.Objective: The aim of the present study was to perform a systematic review of the short and long- term effects of BFR exercise on blood hemostasis in healthy individuals and patients with known disease (ie, hypertension, diabetes, obesity, and ischemic heart disease).Data Sources: A systematic review of English and non-English articles was conducted across PubMed, Science Direct, and Google Scholar databases, including reference lists of relevant papers. Study quality assessment was evaluated using the modified version of Downs and Black checklist. Search results were limited to exercise training studies investigating the effects of BFR exercise on blood hemostasis in healthy individuals and patients with disease. Level of evidence was determined according to the criteria described by Oxford Center for Evidence-Based Medicine. Study selection: Only randomized controlled trials (RCTs) and non-randomized controlled trials (NRCTs) that examined the effects of exercise with BFR exercise vs exercises without BFR on blood hemostasis in healthy individuals and patients were included.Data extraction: Nine studies were eligible (RCT =4; NRCT =5).Results: The average score on the Downs and Black checklist was 11.22. All studies were classified as having poor methodological quality wherein the level of evidence provided in all reviewed studies was level IIb only (ie, poor quality RCTs).Conclusion: Considering the limitations in the available evidence, firm recommendations cannot be provided. Methods: A total of 64 elderly obese women (age: 68.35±6.04 years) underwent body composition analysis by dual-energy X-ray absorptiometry. Participants were classified into two groups according to the definition of SO and NSO. Blood samples were collected for total cholesterol, triglycerides, high-density lipoprotein, low-density lipoprotein, uric acid, urea, interleukin-6 (IL-6), glucose, and creatine kinase (CK) measurements. Results: The SO group presented a significantly greater BMI, fat (%), glucose, a marginal trend toward significance for uric acid, and IL-6 compared to the NSO group. In addition, the SO group displayed lower values for muscle strength and lean body mass. From a correlation standpoint, a higher aLM/BMI was positively associated with lean body mass and muscle strength and negatively associated with a lower BMI and percentage body fat. Conclusion: The definition criteria from FNIH and obesity permit the ability to illustrate the prevalence and identify SO in elderly women with low muscle mass, low muscle strength, and impaired markers of inflammation. Methods: Fifteen elderly women (mean age ± standard deviation, 67.1±6.9 years) classified as having hypertension stage 1 or 2 were randomly allocated to complete traditional RT or HVRT; 1 week later, subjects allocated to RT completed the HVRT session and vice-versa. Heart rate, blood pressure, affective response, perceived effort, and blood samples analyzing lactate, nitrate, nitrite, oxidative damage (thiobarbituric acid reactive substances [TBARS]), and 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid equivalent antioxidant capacity (TEAC) collected before and after training sessions were assessed. Nutritional counseling was provided regarding nutrients that could affect cardiovascular and nitrate/nitrite analysis. Results: Systolic blood pressure was not statistically different (p>0.05) between conditions at the beginning and during 30 minutes after sessions. Exercise is a nonpharmacological treatment shown to benefit all patients with hypertension. Objective: This study examined the effects of a 14-week moderate intensity resistance training program (RT) on the maintenance of blood pressure and hand grip strength during an extended detraining period in elderly hypertensive women. Methods: Twelve hypertensive sedentary elderly women completed 14 weeks of whole body RT at a moderate perceived exertion following a detraining period of 14 weeks. Results: Following the training period, participants demonstrated an increase in absolute hand grip strength (P=0.001), relative hand grip strength (P=0.032) and a decrease of systolic (P=0.001), diastolic (P=0.008), and mean blood pressure (P=0.002) when compared to pre-exercise values. In addition, these effects were sustained after 14 weeks of detraining. Conclusion: Resistance training may be a valuable method to improve muscular strength and blood pressure in elderly people with benefits being maintained up to 14 weeks following training cessation. Editorial No decorrer da história do Programa de Pós Graduação em Odontologia (Reabilitação Oral), da Faculdade de Odontologia de Ribeirão Preto, da Universidade de São Paulo, o Curso de Verão vem divulgando as atividades e linhas de pesquisas desenvolvidas pelo Programa e Departamento com o objetivo de incentivar a docência e pesquisa odontológica Brasileira. Contudo, devido a condição sanitária imposta pela pandemia do SARS-CoV-2, com a necessidade de isolamento social e a característica dos participantes do curso serem de diferentes regiões do Brasil e da América Latina, o curso que vinha sendo preparado com formato presencial, foi replanejado de forma que as atividades propostas pudessem ser realizadas de forma remota, à distância. Graças às tecnologias digitais e empenho da Comissão Organizadora da XIII Edição do Curso de Verão do Programa de Pós Graduação em Reabilitação Oral, o curso pode ser oferecido aos seus 80 participantes, de todos os estados do país e de outros países da américa latina, com uma programação científica que contou com a participação de docentes, pós-graduandos egressos do Programa e pós-graduandos da Unidade. O engajamento da comissão organizadora juntamente com a qualidade de material apresentado pelos palestrantes proporcionou uma experiencia única, concretizando ainda mais a existência do curso dentro do programa e aumentando a abrangência das nossas feitorias para a população. Ainda, graças à possibilidade ofertada aos participantes para apresentação de seus trabalhos e ao alto nível dos trabalhos selecionados, garantindo a troca de experiências entre as diversas universidades, apresentamos a terceira edição dos Anais do Curso de Verão. Certos de que o objetivo do Curso foi alcançado, mesmo frente aos desafios impostos pela Pandemia, agradecermos a participação de todos no evento, com muita responsabilidade, dedicação e carinho. Two hundred and seventy-two sedentary women with a mean age of 66.75 ± 5.38 years were recruited for participation in this study. Obesity was determined by both body mass index and dual-energy X-ray absorptiometry (DXA) evaluations. Sarcopenic obesity diagnosis was established from the ratio between fat-free mass and body surface area as obtained by DXA. There was no association of obesity with sarcopenic obesity (P = 0.424). In contrast, sarcopenia was significantly related to sarcopenic obesity (P < 0.001), although most of the elderly women with sarcopenia (n = 171) did not exhibit sarcopenic obesity. These results highlight the importance of diagnosing sarcopenic obesity as elderly women exhibiting sarcopenia could be either eutrophic or obese.Keywords: sarcopenic obesity, aging, obesity, sarcopenia, health

EditorialNo decorrer da história do Programa de Pós Graduação em Odontologia (Reabilitação Oral), o Curso de Verão vem promovendo as pesquisas desenvolvidas no Departamento de Materiais Dentários e Prótese, com o intuito de convidar a comunidade odontológica a fazer parte desse ambiente acadêmico. Sendo assim a comissão organizadora da XII Edição do Curso de Verão do Programa de Pós Graduação em Reabilitação Oral ofereceu aos seus participantes de diversos estados do país e de outros países uma gama de oportunidades dentro da Faculdade de Odontologia de Ribeirão Preto, com a sua grade cientifica voltada para pesquisas laboratoriais e clínicas, das quais diversos docentes da unidade expuseram as suas linhas de pesquisa.Essa edição do curso atingiu proporções inimagináveis, com altos números de inscrições, nunca obtido nas versões anteriores. Além disso o engajamento da comissão organizadora juntamente com a qualidade de material apresentado pelos palestrantes proporcionaram uma experiencia que tem sido avaliada pelos participantes como única e imensurável, concretizando ainda mais a existência do curso dentro do programa e aumentando a abrangência das nossas feitorias para a população.Assim, damos início a série de Anais do Curso de Verão, da qual essa é a segunda edição que possibilitou aos seus participantes apresentarem seus trabalhos, garantindo a troca de experiencias entre as diversas universidades. Pesquisa qualitativa que objetivou analisar sob a ótica dos educadores de escolas públicas do ensino fundamental, como o tema "orientação sexual" vem sendo incorporado nas práticas pedagógicas. Participaram do estudo 23 professores da rede municipal de ensino de Cajazeiras, Paraíba. Elegeu-se o grupo focal como técnica de investigação, e os dados empíricos obtidos foram organizados mediante análise de conteúdo temática. Constatou-se que há um esforço dos atores em privilegiar conteúdos relativos à orientação sexual no ambiente escolar, porém esses debates exigem que preliminarmente seja incentivado um aprofundamento de caráter informativo e subjetivo sobre "sexualidade", propiciando aos educadores um espaço para re-significações de sua internalidade e valores. A informação direcionada ao cuidado de si deve extrapolar os limites preventivistas e higienistas, incorporando metodologias extensivas, inclusivas e reflexivas, que reconheçam os direitos humanos e sociais e favoreçam a construção ética da cidadania.

Background Acute respiratory distress syndrome (ARDS) is associated with high in-hospital mortality. Alveolar recruitment followed by ventilation at optimal titrated PEEP may reduce ventilator-induced lung injury and improve oxygenation in patients with ARDS, but the effects on mortality and other clinical outcomes remain unknown. This article reports the rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART). Methods/Design ART is a pragmatic, multicenter, randomized (concealed), controlled trial, which aims to determine if maximum stepwise alveolar recruitment associated with PEEP titration is able to increase 28-day survival in patients with ARDS compared to conventional treatment (ARDSNet strategy). We will enroll adult patients with ARDS of less than 72 h duration. The intervention group will receive an alveolar recruitment maneuver, with stepwise increases of PEEP achieving 45 cmH2O and peak pressure of 60 cmH2O, followed by ventilation with optimal PEEP titrated according to the static compliance of the respiratory system. In the control group, mechanical ventilation will follow a conventional protocol (ARDSNet). In both groups, we will use controlled volume mode with low tidal volumes (4 to 6 mL/kg of predicted body weight) and targeting plateau pressure ≤30 cmH2O. The primary outcome is 28-day survival, and the secondary outcomes are: length of ICU stay; length of hospital stay; pneumothorax requiring chest tube during first 7 days; barotrauma during first 7 days; mechanical ventilation-free days from days 1 to 28; ICU, in-hospital, and 6-month survival. ART is an event-guided trial planned to last until 520 events (deaths within 28 days) are observed. These events allow detection of a hazard ratio of 0.75, with 90% power and two-tailed type I error of 5%. All analysis will follow the intention-to-treat principle. Discussion If the ART strategy with maximum recruitment and PEEP titration improves 28-day survival, this will represent a notable advance to the care of ARDS patients. Conversely, if the ART strategy is similar or inferior to the current evidence-based strategy (ARDSNet), this should also change current practice as many institutions routinely employ recruitment maneuvers and set PEEP levels according to some titration method. Perfluoroalkyl acids (PFAAs) are persistent compounds that are ubiquitous globally, though some uncertainties remain in the understanding of their long-range transport mechanisms. They are frequently detected in remote locations, where local sources may be unimportant. We collected a 16.5 metre ice core on northern Ellesmere Island, Nunavut, Canada to investigate PFAA deposition trends and transport mechanisms. The dated core represents fifty years of deposition (1967-2016), which accounts for the longest deposition record of perfluoroalkylcarboxylic acids (PFCAs) in the Arctic and the longest record of perfluoroalkylsulfonic acids (PFSAs) globally. PFCAs were detected frequently after the 1990s and have been increasing since. Homologue pair correlations, molar concentration ratios, and model comparisons suggest that PFCAs are primarily formed through oxidation of volatile precursors. PFSAs showed no discernible trend, with concentrations at least an order of magnitude lower than PFCAs. We observed episodic deposition of some PFAAs, notably perfluorooctane sulfonic acid (PFOS) and perfluorobutane sulfonic acid (PFBS) before the 1990s, which may be linked to Arctic military activities. Tracer analysis suggests that marine aerosols and mineral dust are relevant as transport vectors for selected PFAAs during specific time periods. These observations highlight the complex mechanisms responsible for the transport and deposition of PFAAs in the High Arctic.

18. Effects of perfluoroalkyl sulfonic acids on developmental, physiological, and immunological measures in northern leopard frog tadpoles.

The chronic toxicity of short chain perfluoroalkyl sulfonic acids (PFSAs), such as perfluorobutanesulfonic acid (PFBS) and perfluorohexanesulfonic acid (PFHxS), are relatively understudied despite the increasing detection of these compounds in the environment. We investigated the chronic toxicity and bioconcentration of PFBS and PFHxS using northern leopard frog (Rana [Lithobates] pipiens) tadpoles. We exposed Gosner stage (GS) 25 tadpoles to either PFBS or PFHxS at nominal concentrations of 0.1, 1, 10, 100, and 1000 μg/L until metamorphosis (GS42). We then assessed tadpole growth, development, stress, and immune metrics, and measured fatty acid (FA) composition and PFSA concentrations in liver and whole-body tissues. Tadpole growth and development measures were relatively unaffected by PFSA exposure. However, tadpoles exposed to 1000 μg/L PFBS or PFHxS had significantly increased hepatosomatic indexes (HSI) relative to controls. Further, tadpoles from the 1000 μg/L PFHxS treatment had altered FA profiles relative to controls, with increased total FAs, saturated FAs, monounsaturated FAs, and omega-6 polyunsaturated FAs. In addition, tadpoles from the 1000 μg/L PFHxS treatment had a higher probability of waterborne corticosterone detection. These results suggest that PFBS and PFHxS influence the hepatic health of tadpoles, and that PFHxS may alter lipid metabolism in tadpoles. We also observed a higher probability of tadpoles being phenotypically female after exposure to an environmentally relevant concentration (0.1 μg/L) of PFHxS, suggesting that PFHxS may exert endocrine disrupting effects on tadpoles during early development. The measured bioconcentration factors (BCFs) for both compounds were ≤10 L kg
-1 wet weight, suggesting low bioconcentration potential for PFBS and PFHxS in tadpoles. - Abstract
In the present study, we investigated whether curcumin administration would interfere with the main renal features of l-NAME-induced hypertension model. For this purpose, we conducted both in vitro and in vivo experiments to evaluate renal indicators of inflammation, oxidative stress, and metalloproteinases (MMPs) expression/activity. Hypertension was induced by l-NAME (70 mg/kg/day), and Wistar rats from both control and hypertensive groups were treated with curcumin (50 or 100 mg/kg/day; gavage) or vehicle for 14 days. Blood and kidneys were collected to determine serum creatinine levels, histological alterations, oxidative stress, MMPs expression and activity, and ED1 expression. l-NAME increased blood pressure, but both doses of curcumin treatment reduced these values. l-NAME treatment increased creatinine levels, glomeruli area, Bowman's space, kidney MMP-2 activity, as well as MMP-9 and ED1 expression, and reduced the number of glomeruli. Curcumin treatment prevented the increase in creatinine levels, MMP-2 activity, and reduced MMP-2, MMP-9, ED1, and superoxide levels, as well as increased superoxide dismutase activity and partially prevented glomeruli alterations. Moreover, curcumin directly inhibited MMP-2 activity in vitro. Thus, our main findings demonstrate that curcumin reduced l-NAME-induced hypertension and renal glomerular alterations, inhibited MMP-2 and MMP-9 expression/activity, and reduced oxidative stress and inflammatory processes, which may indirectly impact hypertension-induced renal outcomes., (© 2024 John Wiley & Sons Ltd.)

The present study focuses on the elaboration of magnetic nanocomposites by the in situ incorporation of magnetite (Fe
3 O4 ) nanoparticles (NPs) with spherical and nanoflower-like morphologies in graphitic carbon nitride (g-C3 N4 ) sheets using two different synthetic routes. Nanomaterials are characterized by TEM, SEM, XRD, FTIR, BET, zetametry, vibrating sample magnetometry, and UV-vis absorption spectroscopy. The decoration of the carbon nitride matrix with the magnetic NPs enhanced optical and textural properties. The influence of the morphology of the magnetic NPs on the adsorptive and photocatalytic properties of the nanocomposites under different pH conditions (4.5, 6.9, and 10.6) was assessed from batch tests to remove methylene blue (MB) from aqueous solutions. In extreme pH conditions, the nanocomposites exhibited lower or equivalent MB removal capacity compared to the pure g-C3 N4 . However, at neutral medium, the nanocomposite with incorporated Fe3 O4 nanoflowers showed a significantly higher removal efficiency (80.7%) due to the combination of a high adsorption capacity and a good photocatalytic activity in this pH region. The proposed nanocomposite is a promising alternative to remove cationic dyes from water by magnetic assistance, since no pH adjustment of the polluted effluent is required, reducing costs and environmental impact in the dyeing industry., (© 2024. 21. Cholesterol transfer to high-density lipoprotein in obesity and the effects of weight loss after bariatric surgery.

Obesity increases serum triglycerides and decreases high-density lipoprotein cholesterol (HDL-C). The objective is to explore some functions of HDL, cholesterol transfers and antioxidant, in subjects with grade I (G1-OB) and III (G3-OB) obesity and effects of bariatric surgery on G3-OB. Fifteen G3-OB patients (43 ± 6 years, BMI 49 ± 3 kg/m
2 ) were studied before and 1 year after bariatric surgery; 15 G1-OB (32 ± 2 years, 32 ± 2 kg/m2 ) and 15 normal weight (NW) (38 ± 6 years, 22 ± 1 kg/m2 ) were also studied. HDL diameter, cholesterol transfer to HDL and antioxidant capacity of HDL were determined. G3-OB had higher triglycerides and lower HDL-C; G1-OB had higher triglycerides than NW but HDL-C was equal. Compared to NW, HDL size was smaller in G3-OB but equal in G1-OB. One year after bariatric surgery, BMI and triglycerides of G3-OB decreased (p < .0001 and p = .0012, respectively) and HDL-C increased (p < .0001), equalling of NW group. Antioxidant capacity and cholesterol transfers were not different in groups and were unchanged 1 year after bariatric surgery in G3-OB. HDL antioxidant capacity and transfer of cholesterol to HDL were not defective in obesity despite HDL-C reduction and smaller HDL size. Substituted diphenylamine antioxidants (SDPAs) and benzotriazole UV stabilizers (BZT-UVs) are industrial additives of emerging environmental concern. However, little is known about their environmental fate and bioaccumulation. This study investigated the concentrations of SDPAs and BZT-UVs in the water, sediment and biota samples in the freshwater ecosystem and adjacent riparian environment using Hamilton Harbour in the Great Lakes of North America as a study site. The bioaccumulation factors and trophodynamics of these contaminants were studied using field-collected samples. Eight target SDPAs and two BZT-UVs (2-(2 H -benzotriazol-2-yl)-4,6-bis(1-methyl-1-phenylethyl)phenol (UV234) and 2-(2 H -benzotriazol-2-yl)-4,6-di- tert -pentylphenol (UV328)) were frequently detected in the sediment, water and biota samples. UV328 showed significantly greater concentrations in water (0.28-2.8 ng L
-1 ) and sediment (8.3-48 ng g-1 , dry weight) than other target contaminants, implying greater contamination of UV328 in Hamilton Harbour. SDPAs exhibited trophic dilution in species living in the water, whereas UV234 was biomagnified in the same samples. No clear trophodynamic trend was found for UV328 for water-respiring species. Air-breathing invertebrates had higher concentrations of both SDPAs and BZT-UVs than water-respiring invertebrates, and biomagnification was observed particularly for adult dragonflies. These results suggest that the trophodynamics of SDPAs and BZT-UVs vary depending on whether the food web is terrestrial or aquatic. Future research should investigate the occurrence and partitioning of SDPAs and BZT-UVs in the air-water interface and evaluate the toxicities of these contaminants in air-breathing species.- Published

PFAAs (Perfluoroalkyl acids) are a class of bioaccumulative, persistent and ubiquitous environmental contaminants which primarily occupy the hydrosphere and its sediments. Currently, a paucity of toxicological information exists for short chain PFAAs and complex mixtures. In order to address these knowledge gaps, we performed a 3-week, aqueous exposure of rainbow trout to 3 different concentrations of a PFAA mixture (50, 100 and 500 ng/L) modeled after the composition determined in Lake Ontario. We conducted an additional set of exposures to individual PFAAs (25 nM each of PFOS (12,500 ng/L), PFOA (10,300 ng/L), PFBS (7500 ng/L) or PFBA (5300 ng/L) to evaluate differences in biological response across PFAA congeners. Untargeted proteomics and phosphorylated metabolomics were conducted on the blood plasma and head kidney tissue to evaluate biological response. Plasma proteomic responses to the mixtures revealed several unexpected outcomes including Similar proteomic profiles and biological processes as the PFOS exposure regime while being orders of magnitude lower in concentration and an atypical dose response in terms of the number of significantly altered proteins (FDR < 0.1). Biological pathway analysis revealed the low mixture, medium mixture and PFOS to significantly alter (FDR < 0.05) a number of processes including those involved in lipid metabolism, oxidative stress and the nervous system. We implicate plasma increases in PPARD and PPARG as being directly related to these biological processes as they are known to be important regulators in all 3 processes. In contrast to the blood plasma, the high mixture and PFOA exposure regimes caused the greatest change to the head kidney proteome, altering many proteins being involved in lipid metabolism, oxidative stress and inflammation. 24. Modulation of tumor plasticity by senescent cells: Deciphering basic mechanisms and survival pathways to unravel therapeutic options.

- Abstract
Senescence is a cellular state in which the cell loses its proliferative capacity, often irreversibly. Physiologically, it occurs due to a limited capacity of cell division associated with telomere shortening, the so-called replicative senescence. It can also be induced early due to DNA damage, oncogenic activation, oxidative stress, or damage to other cellular components (collectively named induced senescence). Tumor cells acquire the ability to bypass replicative senescence, thus ensuring the replicative immortality, a hallmark of cancer. Many anti-cancer therapies, however, can lead tumor cells to induced senescence. Initially, this response leads to a slowdown in tumor growth. However, the longstanding accumulation of senescent cells (SnCs) in tumors can promote neoplastic progression due to the enrichment of numerous molecules and extracellular vesicles that constitutes the senescence-associated secretory phenotype (SASP). Among other effects, SASP can potentiate or unlock the tumor plasticity and phenotypic transitions, another hallmark of cancer. This review discusses how SnCs can fuel mechanisms that underlie cancer plasticity, like cell differentiation, stemness, reprogramming, and epithelial-mesenchymal transition. We also discuss the main molecular mechanisms that make SnCs resistant to cell death, and potential strategies to target SnCs. At the end, we raise open questions and clinically relevant perspectives in the field.

Oxidative/nitrosative damage takes part in chronic disease development, which generates an urgent need for intervention and better therapies to manage them. The scientific community has demanded easy-to-run, cheap, and reliable methods for cellular antioxidant activity assays. This work standardised and validated an erythrocyte cellular antioxidant activity and membrane protection/injury (HERYCA-P) protocol to study food-derive extracts. The method measures intracellular reactive oxygen species (ROS) generation, lipoperoxidation, and haemolysis induced by 2,2'-azobis(2-amidinopropane) dihydrochloride. Quercetin decreased ROS generation by 50.4% and haemolysis by 2.2%, while ascorbic acid inhibited lipid peroxidation by 40.1%. Total phenolic contents of teas were correlated with decreased ROS generation (r = -0.924), lipoperoxidation (r = -0.951), and haemolysis (r = -0.869). The erythrocyte ROS generation and lipoperoxidation were also associated with CUPRAC (r = -0.925; r = -0.951) and hydroxyl radical scavenging activity (r = -0.936; r = -0.949). The precision rates of antioxidant standards and tea samples were below 15%. Cellular senescence is a state of permanent growth arrest. It can be triggered by telomere shortening (replicative senescence) or prematurely induced by stresses such as DNA damage, oncogene overactivation, loss of tumor suppressor genes, oxidative stress, tissue factors, and others. Advances in techniques and experimental designs have provided new evidence about the biology of senescent cells (SnCs) and their importance in human health and disease. This review aims to describe the main aspects of SnCs phenotype focusing on alterations in subcellular compartments like plasma membrane, cytoskeleton, organelles, and nuclei. We also discuss the heterogeneity, dynamics, and plasticity of SnCs' phenotype, including the SASP, and pro-survival mechanisms. We advance on the multiple layers of phenotypic heterogeneity of SnCs, such as the heterogeneity between inducers, tissues and within a population of SnCs, discussing the relevance of these aspects to human health and disease. We also raise the main challenges as well alternatives to overcome them. Ultimately, we present open questions and perspectives in understanding the phenotype of SnCs from the perspective of basic and applied questions., (© 2024 The Authors. We show that architected materials, which have macroscopic properties that differ from those of their constituent components, can increase wave energy dissipation by more than an order of magnitude over both natural and existing artificial reefs, while providing a biocompatible environment. Short-chain perfluoroalkyl carboxylic acids (PFCAs) have been detected in the environment globally. The presence and persistence of these compounds in the environment may lead to chronic wildlife exposure. We used northern leopard frog (Rana pipiens) tadpoles to investigate the chronic toxicity and the bioconcentration of two short-chain PFCAs, perfluorobutanoic acid (PFBA) and perfluorohexanoic acid (PFHxA). We exposed Gosner stage 25 tadpoles to PFBA and PFHxA (as individual chemicals) at nominal concentrations of 0.1, 1, 10, 100, and 1000 µg/L for 43-46 days. Tadpoles exposed to 0.1 to 100 µg/L of PFBA and PFHxA had significantly higher mean snout-to-vent lengths, mean masses, and scaled mass indexes than control tadpoles. These results indicate that exposure to short-chain PFCAs influences tadpole growth. Further investigation into the mechanism(s) causing the observed changes in tadpole growth is warranted. We observed a significantly higher proportion of males in the PFBA 1 µg/L treatment group, however further histological analyses are required to confirm visual sex identification before making concrete conclusions on the effects of PFCAs on amphibian sex ratios. PFBA concentrations in tissues were higher than PFHxA concentrations; a pattern that contrasts with previously published studies using fish, suggesting potential differences between taxa in PFBA and PFHxA bioconcentration. Bioconcentration factors were <10 L/kg wet weight, indicating low bioconcentration potential in tadpoles. Our results suggest that PFBA and PFHxA may have effects at environmentally-relevant concentrations (0.1-10 µg/L) and further investigation is required before these compounds can be deemed a "safe" alternative to their long-chain counterparts., (© 2024. The concentrations of per- and polyfluoroalkyl substances (PFAS) were determined in raw influent, final effluent, and treated biosolids at Canadian wastewater treatment plants (WWTPs) to evaluate the fate of PFAS through liquid and solids trains of typical treatment process types used in Canada and to assess time trends of PFAS in wastewater between 2009 and 2021. Data for 42 PFAS in samples collected from 27 WWTP across Canada were used to assess current concentrations and 48 WWTPs were included in the time trends analysis. Although regulated and phased-out of production by industry since the early 2000s and late 2000s/early2010s, respectively, perfluorooctanesulfonate (PFOS), perfluorooctanoate (PFOA), and other long-chain PFAS continue to be widely detected in Canadian wastewater and biosolids. Short-chain PFAS that are not currently regulated in Canada were also widely detected. In general, elevated concentrations of several PFAS were observed at WWTPs that receive landfill leachate. Except for PFOS, concentrations of long-chain perfluoroalkyl carboxylates (PFCAs) and perfluoroalkane sulfonates (PFSAs) generally decreased over time in influent, effluent, and biosolids, which is attributable to industrial production phase-outs and regulations. Concentrations of PFOS did not decrease over time in wastewater media. This indicates that regulatory action and industrial phase-outs of PFOS are slow to be reflected in wastewater. Concentrations of short-chain PFCAs in wastewater influent and effluent consistently increased between 2009 and 2021, which reflect the use of short-chain PFAS as replacements for phased-out and regulated longer-chained PFAS. Short-chain PFAS were infrequently detected in biosolids. - Abstract
Colorectal cancer (CRC) is the third most common and deadliest cancer globally. Regimens using 5-fluorouracil (5FU) and Oxaliplatin (OXA) are the first-line treatment for CRC, but tumor recurrence is frequent. It is plausible to hypothesize that differential cellular responses are triggered after treatments depending on the genetic background of CRC cells and that the rational modulation of cell tolerance mechanisms like autophagy may reduce the regrowth of CRC cells. This study proposes investigating the cellular mechanisms triggered by CRC cells exposed to 5FU and OXA using a preclinical experimental design mimicking one cycle of the clinical regimen (i.e., 48 h of treatment repeated every 2 weeks). To test this, we treated CRC human cell lines HCT116 and HT29 with the 5FU and OXA, combined or not, for 48 h, followed by analysis for two additional weeks. Compared to single-drug treatments, the co-treatment reduced tumor cell regrowth, clonogenicity and stemness, phenotypes associated with tumor aggressiveness and poor prognosis in clinics. This effect was exerted by the induction of apoptosis and senescence only in the co-treatment. However, a week after treatment, cells that tolerated the treatment had high levels of autophagy features and restored the proliferative phenotype, resembling tumor recurrence. The pharmacologic suppression of early autophagy during its peak of occurrence, but not concomitant with chemotherapeutics, strongly reduced cell regrowth. Overall, our experimental model provides new insights into the cellular mechanisms that underlie the response and tolerance of CRC cells to 5FU and OXA, suggesting optimized, time-specific autophagy inhibition as a new avenue for improving the efficacy of current treatments., (© 2024 Wiley Periodicals LLC.)

31. Comparison of force variables and dynamic strength index between age groups in elite young Brazilian football goalkeepers.

Introduction: The application of muscle force is a determinant of football success as it is inherent to the motor control and sport. The aims of this study are: (1) to describe force variables Isometric Maximal Force (IMF), Concentric Peak Force (CPPF), and Dynamic Strength Index (DSI) in football goalkeepers from different age groups; (2) to compare these variables' behavior between those groups., Methods: The sample was formed by 19 youth players (15.97 ± 1.55 years old) from a first-division Brazilian football team. The CPPF and IMF variables were obtained through the Countermovement jump and isometric squat tests, respectively. For data collection, a force plate (Cefise, Brazil) was used with an acquisition frequency of 600 Hz and mono-axial. The DSI was calculated using the ratio between CPPF and IMF. For data analysis, the sample was separated into clusters by age. After the grouping, a descriptive analysis of the data and a comparison between the groups with p < 0.05., Results: The sample was grouped into three groups (GA, GB, and GC) and one of the individuals did not enter the group, totaling 18 individuals in the analyzed sample. The comparison between the ages of the groups showed a significant difference and small and moderate effect size (ES), validating the cluster strategy. The CPPF and IMF variables showed increased values according to chronological age. CPPF showed a significant difference between GA-GB, (ES = very large) GA-GC (ES = very large), and GB-GC (ES = moderate). The IMF variable had significant differences between GA-GB (ES = moderate) and GA-GC (ES = very large). However, DSI showed significant differences only between GA GB (ES = small) and GB-GC (ES = very large)., Conclusions: The CPPF and IMF variables had constant increases and distinct values with an increase according to age, and this did not occur for DSI. 2 O4 nanoparticles (NPs), with mean diameter ∼5 nm, were investigated by means of DC magnetization and AC susceptibility over the temperature range of 4-400 K. All NPs present the same CoFe2 O4 core, with different molecular surface coatings, increasing gradually the number of carbon atoms in the coating layer: glycine (C2 H5 NO2 ), alanine (C3 H7 NO2 ), aminobutanoic acid (C4 H9 NO2 ), aminohexanoic acid (C6 H13 NO2 ), and aminododecanoic acid (C12 H25 NO2 ). Samples were intentionally fabricated in order to modulate the core-core magnetic dipolar interaction, as the thickness of the coating layer increases with the number of carbon atoms in the coating molecule. The magnetic data of the uncoated CoFe2 O4 NPs were also collected for comparison. All investigated CoFe2 O4 NPs (coated and uncoated) are in a magnetically blocked state at room temperature as evidenced by ZFC/FC measurements and the presence of hysteresis with ∼700 Oe coercivity. Low temperature magnetization scans show slightly constricted hysteresis loops with coercivity decreasing systematically with a decreasing number of carbon atoms in the coating molecule, possibly resulting from differences in magnetic dipole coupling between NPs. Large thermomagnetic irreversibility, slow monotonic increase in the FC magnetization and non-saturation of the magnetization give evidence for the cluster glass (CG) nature in the CoFe2 O4 NPs. The out of phase part ( χ '') of AC susceptibility for all samples shows a clear frequency dependent hump which was analyzed to distinguish superparamagnetic (SPM), cluster glass (CG) and spin glass (SG) behavior by using Néel-Arrhenius, Vogel-Fulcher, and power law fittings. These analyses rule out the SPM state and suggest the presence of significant inter-cluster dipolar interaction, giving rise to CG cooperative freezing in the high-temperature region. In the low-temperature range, however, the disordered spins on the nanoparticle's surface play an important role in the formation of the SG-like state, as evidenced by Arrott plots and temperature dependency of d M /d H in the initial magnetization curves. In summary, the magnetic measurements showed that undercooling the system evolves from a SPM state of weakly interacting spin clusters, through the CG state induced by strong dipolar interaction, to the SG state resulting from the frustration of the disordered surface spins.- Published

Organophosphate esters (OPEs) have been used as flame retardants, plasticizers, and anti-foaming agents over the past several decades. Of particular interest is the long range transport potential of OPEs given their ubiquitous detection in Arctic marine air. Here we report 19 OPE congeners in ice cores drilled on remote icefields and ice caps in the Canadian high Arctic. A multi-decadal temporal profile was constructed in the sectioned ice cores representing a time scale spanning the 1970s to 2014-16. In the Devon Ice Cap record, the annual total OPE (∑OPEs) depositional flux for all of 2014 was 81 μg m
-2 , with the profile dominated by triphenylphosphate (TPP, 9.4 μg m-2 ) and tris(2-chloroisopropyl) phosphate (TCPP, 42 μg m-2 ). Here, many OPEs displayed an exponentially increasing depositional flux including TCPP which had a doubling time of 4.1 ± 0.44 years. At the more northern site on Mt. Oxford icefield, the OPE fluxes were lower. Here, the annual ∑OPEs flux in 2016 was 5.3 μg m-2 , dominated by TCPP (1.5 μg m-2 ) but also tris(2-butoxyethyl) phosphate (1.5 μg m-2 TBOEP). The temporal trend for halogenated OPEs in the Mt. Oxford icefield is bell-shaped, peaking in the mid-2000s. The observation of OPEs in remote Arctic ice cores demonstrates the cryosphere as a repository for these substances, and supports the potential for long-range transport of OPEs, likely associated with aerosol transport.- Published

34. Effects of Water Deficit on Growth and Productivity in Tomato Crops Irrigated with Water Treated with Very Low-Frequency Electromagnetic Resonance Fields.

The tomato crop is very sensitive to stress conditions. A water deficit is defined as when precipitation is less than the evapotranspiration (ETc) of the crop in a given period, and in this scenario of climate change, it is identified as responsible for global productivity losses. The use of potential technologies for better irrigation management, such as electromagnetically treated water, remains controversial. Thus, the objective of the present work was to investigate the effects of very low-frequency electromagnetic resonance field treatment on water for tomato crops submitted to different irrigation rates. For this, an experiment was carried out under controlled conditions with different types of water: electromagnetically treated water (WTVLF) and untreated water (UNW), as well as four water replacement rates: 40, 60, 80, and 100% ETc. The electromagnetic treatment of the water was carried out using the commercial equipment AQUA4D
® . The experiment was carried out in pots with five replications per treatment. Lower activity of SOD, POD, CAT, and APX enzymes was observed in plants irrigated with water treated with very low-frequency electromagnetic resonance fields (WTVLF), indicating less oxidative stress caused by water deficit. Water deficit reduced chlorophyll content, but the effects were less harmful with WTVLF water. The water deficit resulted in less accumulation of dry matter and less productivity in a linear relationship. However, plants irrigated with WTVLF showed increments of about 20% in dry matter accumulation and 20% in fruit production concerning plants irrigated with untreated water, independent of the irrigation rates. We can conclude that irrigation with WTVLF can be a solution to reduce the damage caused by water deficits and increase the productivity of tomato crops.- Published

35. Soil arthropod community responses to restoration in areas impacted by iron mining tailings deposition after Fundão dam failure.

In 2015, the failure of the Fundão dam in Mariana, Brazil released ~43 million m
3 of iron mining tailings into the environment. Despite restoration initiatives in the following years, few studies-and most focused on revegetation-have evaluated the effectiveness of the restoration process in areas impacted by the disaster. We aimed to evaluate the responses of the arthropod community in areas impacted by iron mining tailings deposition from the Fundão dam that is in the restoration process. We defined sampling units in the riparian zone of the Gualaxo do Norte River, which is under restoration, and in a native not impacted riparian zone. We collected soil arthropods using pitfall traps and sampled environmental variables in the same sites. We used generalize least squares models (GLS) to test if the restored areas already presented values of arthropod diversity and functional group abundance similar to the reference area and to test which environmental variables are influencing arthropod diversity. We also tested how large the differences of arthropod community composition between the study areas and used the index of indicator species (IndVal) to verify which species could be used as an indicator of reference or restoration areas. The diversity of arthropods and the functional groups of detritivores and omnivores were higher in the native riparian zone. Understory density, soil density, organic matter content, and microbial biomass carbon were the environmental variables that significantly explained the diversity and species composition of arthropods. We show that restoration areas still have different soil arthropod diversity values and community composition when compared to reference areas. Evaluating the response of the arthropod community to the restoration process and long-term monitoring are essential to achieve a satisfactory result in this process and achieve a self-sustaining ecosystem., (© 2023. 36. Sedimentary records of contaminant inputs in Frobisher Bay, Nunavut.

- Abstract
Contaminants, such as polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs), heavy metals, and per and polyfluoroalkyl substances (PFASs), primarily reach the Arctic through long-range atmospheric and oceanic transport. However, local sources within the Arctic also contribute to the levels observed in the environment, including legacy sources and new sources that arise from activities associated with increasing commercial and industrial development. The City of Iqaluit in Frobisher Bay, Nunavut (Canada), has seen rapid population growth and associated development during recent decades yet remains a site of interest for ocean protection, where Inuit continue to harvest country food. In the present study, seven dated marine sediment cores collected in Koojesse Inlet near Iqaluit, and from sites in inner and outer Frobisher Bay, respectively, were analyzed for total mercury (THg), major and trace elements, PAHs, PCBs, and PFASs. The sedimentary record in Koojesse Inlet shows a period of Aroclor 1260-like PCB input concurrent with military site presence in the 1950-60s, followed by decades of input of pyrogenic PAHs, averaging about ten times background levels. Near-surface sediments in Koojesse Inlet also show evidence of transient local-source inputs of THg and PFASs, and recycling or continued slow release of PCBs from legacy land-based sources. Differences in PFAS congener composition clearly distinguish the local sources from long-range transport. Outside Koojesse Inlet but still in inner Frobisher Bay, 9.2 km from Iqaluit, sediments showed evidence of both local source (PCB) and long-range transport. In outer Frobisher Bay, an up-core increase in THg and PFASs in sediments may be explained by ongoing inputs of these contaminants from long-range transport. Introduction: Lysosomal storage disorders (LSD) are a group of monogenic rare diseases caused by pathogenic variants in genes that encode proteins related to lysosomal function. These disorders are good candidates for gene therapy for different reasons: they are monogenic, most of lysosomal proteins are enzymes that can be secreted and cross-correct neighboring cells, and small quantities of these proteins are able to produce clinical benefits in many cases. Ex vivo gene therapy allows for autologous transplant of modified cells from different sources, including stem cells and hematopoietic precursors., Areas Covered: Here, we summarize the main gene therapy and genome editing strategies that are currently being used as ex vivo gene therapy approaches for lysosomal disorders, highlighting important characteristics, such as vectors used, strategies, types of cells that are modified and main results in different disorders., Expert Opinion: Clinical trials are already ongoing, and soon approved therapies for LSD based on ex vivo gene therapy approaches should reach the market.

Kefir is a probiotic mixture with anxiolytic and antioxidant properties. Chronic stress can lead to anxiety disorders and increase oxidative damage in organs such as the heart and kidney. In this study, we examined whether kefir ameliorates the anxiety-like behavior of mice submitted to chronic unpredictable stress (CUS) by modulating brain-derived neurotrophic factor (BDNF) and corticosterone levels and whether kefir modifies the oxidative parameters in the heart and kidney of mice. Male Swiss mice received kefir (0.3 mL/100 g/day) or milk for 30 days (gavage). On the 10th day, the mice were submitted to CUS. Behavioral analysis was performed using the elevated plus maze and forced swimming tests. BDNF levels were analyzed in brain tissues. Heart and kidney superoxide dismutase (SOD), catalase, glutathione (GSH), thiobarbituric acid reactive substances (TBARS), 3-nitrotyrosine, metalloproteinase-2 (MMP-2), and plasma corticosterone were evaluated. Kefir reverted the CUS-induced decrease in the time spent in the open arms, the increase in grooming frequency, and decrease in the head dipping frequency, but not the reduced immobility time. CUS decreased the cerebellum BDNF levels and increased corticosterone levels, which were restored by Kefir. Neither catalase and SOD activities nor GSH, TBARS, 3-nitrotyrosine, and MMP-2 were modified by CUS in the heart. In the kidney, CUS increased 3-nitrotyrosine and MMP-2. Kefir increased the antioxidant defense in the heart and kidney of control and CUS mice. These results suggest that kefir ameliorated CUS-induced anxiety-like behavior by modulating brain BDNF and corticosterone levels. Per- and polyfluoroalkyl substances (PFAS) are a class of thousands of highly fluorinated, anthropogenic compounds that are used in a wide variety of consumer applications. Due to their widespread use and high persistence, PFAS are ubiquitous in drinking water, which is of concern due to the threats these compounds pose to human health. Reduction via the hydrated electron is a promising technology for PFAS remediation and has been well-studied. However, since previous work rarely reports fluorine atom balances and often relies on suspect screening, some transformation products are likely unaccounted for. Therefore, we performed non-target analysis using high-resolution mass spectrometry on solutions of perfluorooctanesulfonate (PFOS), perfluorobutanesulfonate (PFBS), perfluorooctanoate (PFOA), and 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy)propanoate (GenX) that had been treated with UV/sulfite to produce hydrated electrons. We determined fluorine atom balances for all compounds studied, finding high fluorine atom balances for PFOS and PFBS. PFOA and GenX had lower overall fluorine atom balances, likely due to the production of volatile or very polar transformation products that were not measured by our methods. Transformation products identified by our analysis were consistent with literature, with a few exceptions. Namely, shorter-chain perfluorosulfonates (PFSA) and their H/F substituted counterparts were also detected from PFOS. This is an unexpected result based on literature, as no documented pathway exists for the formation of shorter-chain PFSA during UV/sulfite treatment. Furthermore, the nontarget approach we employed allowed for identification of novel, unsaturated products from the hydrated electron treatment of perfluorooctanesulfonate (PFOS) that warrant further investigation.

Increased demand for recycling plastic has prompted concerns regarding potential introduction of hazardous chemicals into recycled goods. We present a broad screening of chemicals in 21 plastic flake and pellet samples from Canadian recycling companies. From target analysis, the organophosphorus ester flame retardants and plasticizers exhibited the highest detection frequencies (DFs) (5-100%) and concentrations (
280 chemicals were detected in recycled plastic pellets and flakes, suggesting potential incorporation into recycled goods. Individual concentrations indicate unintentional trace contamination following European Union threshold limits for recycled granules (500 mg/kg) and waste plastic flakes (1,000 mg/kg), although do not reflect toxicological thresholds, if any. Our study highlights that while recycling addresses sustainability goals, additional screening of goods and products made from recycled plastics is needed to fully document potentially hazardous chemicals and exposure. - Published

42. Anterior Upper Teeth Golden Proportion Analysis with Millimetric Templates: An Invention Developed at Londrina State University.

In order to achieve aesthetic and harmonious smile results, the use of anterior upper teeth golden proportion concepts represents reliable and scientific based guidelines. However, measuring, recording and analysing teeth and smiles biometric values proves to be a clinical and laboratory routine chalenge, once it is time consuming and demands additional especific math calculus or formulas. The aim of this paper is present an invention, "anterior upper teeth golden proportion millimetric templates," a set of instruments fabricated in order to achieve precise and fast millimetric measures, once they present predefined geometrical drawings and diagrams. Currently, when planning aesthetic tooth size modifications treatments, tools are used as pachymeters, dry edge compass, or some softwares. Most of times this strategy relies on professional expertise and intuiton, which shows to be limitating factors with the need of trial-and-error training and an accurate critical and artistic sense. Unfortunately, this is not inherent to all professionals, especially undergraduate dental school students. Thus, the templates bring differentials and advantages, being versatile and convenient, allowing countless clinical and laboratory uses, even over a cellphone or a computer screen. Nicotine has been used during pregnancy and lactation as a tobacco harm reduction strategy. However, it is unclear whether nicotine exposure during a critical development period negatively impacts stress responses in adulthood. This study investigated how nicotine, administered via breastfeeding, affects the brain-derived neurotrophic factor (BDNF), synaptic proteins levels, and anxiety-like behavior in adult female mice subjected to stress. Female Swiss mice were exposed to saline or nicotine (8 mg/kg/day) through breastfeeding between their fourth and 17th postnatal days (P) via implanted osmotic mini pumps. The unpredictable chronic mild stress (UCMS) protocol was performed during their adulthood (P65) for 10 consecutive days, followed by the elevated plus maze (EPM) test 1 day after the protocol. Animals were euthanized and their blood, collected for plasma corticosterone measurements and their brain structures, dissected for BDNF and synaptic proteins analyses. We found no significant differences in corticosterone levels between groups (Saline/Non-stress, Nicotine/Non-stress, Saline/Stress, and Nicotine/Stress). The UCMS protocol hindered weight gain. Mice exposed to nicotine through breastfeeding with or without the UCMS protocol in adulthood showed higher grooming and head dipping frequency; decreased BDNF levels in cerebellum and striatum; increased postsynaptic density protein 95 (PSD-95), synapsin I, and synaptophysin levels in cerebellum; and decreased PSD-95 and synapsin I levels in brainstem. Our results indicate that nicotine exposure through breastfeeding leads to long-lasting behavioral effects and synaptic protein changes, most of which were independent of the UCMS protocol, even after a long nicotine-free period, highlighting the importance of further studies on nicotine exposure during development., (© 2022 International Society for Developmental Neuroscience.)

44. Potentially toxic elements in iron mine tailings: Effects of reducing soil pH on available concentrations of toxic elements.

Tailings from iron mining are characterized by high concentrations of iron and manganese oxides, as well as high pH values. With these characteristics, most of the potentially toxic elements (PTE) contained in the tailings are somewhat unavailable. The aim of the present study was to evaluate how a reduction in the pH of iron mine tailings may affect PTE availabilities. The tailings were collected on the banks of the Gualaxo do Norte River (Mariana, MG, Brazil), one of the main areas impacted by the rupture of the Fundão Dam (Barragem de Fundão). A completely randomized experimental design was used, including five pH values (6.4, 5.4, 4.3, 3.7, and 3.4) and five replications. The concentrations of the PTE (Ba, Cr, Cd, Co, Cu, Fe, Mn, Pb, Ni, and Zn) were determined after extraction following different methodologies: USEPA 3051A, DTPA, Mehlich-1, Mehlich-3, and distilled water. A comparison of the available concentrations of the elements in the tailings with those in a soil not impacted by tailings shows that Cr, Cd, Cu, Fe, Mn, Ni, Ba, and Co were higher in the soil impacted by the tailings. The different methods used for evaluating the availability of PTE in the tailings at various pH exhibited the following decreasing order in relation to the quantity extracted: Mehlich-3 > Mehlich-1 > DTPA > distilled water. However, regarding sensitivity to change in pH, the order was DTPA > water > Mehlich-1 > Mehlich-3. The increases in the concentrations of PTE due to the reduction in the pH of the tailings did not lead to concentrations that exceed the limits of Brazilian regulations. Reactive oxygen species (ROS) are bioproducts of cellular metabolism. There is a range of molecules with oxidizing properties known as ROS. Despite those molecules being implied negatively in aging and numerous diseases, their key role in cellular signaling is evident. ROS control several biological processes such as inflammation, proliferation, and cell death. The redox signaling underlying these cellular events is one characteristic of the new generation of scientists aimed at defining the role of ROS in the cellular environment. - Abstract
Partially stabilized zirconia (5Y-PSZ) has been widely used to manufacture indirect monolithic restorations, and the effect of finishing procedures on the optical and mechanical properties of these materials are still unclear. The purpose of this study was to evaluate the effect of staining, polishing and glazing on surface roughness, crystalline phase content, microhardness, fracture toughness, dynamic elastic modulus, three-point flexural strength, strain distribution, color (∆E
00 /∆L/∆a/∆b), and translucency before and after artificial accelerated aging (water spray and ultraviolet) of 5Y-PSZ. Bar-shaped and rectangle-shaped specimens of the 5Y-PSZ were prepared and divided into six groups, according to finishing procedure: GC (control), GS (staining), GG (glazing), GSG (staining and glazing), GP (polishing), GSP (staining and polishing). There was a significant difference between groups for surface roughness (p < 0.05), dynamic elastic modulus (p = 0.007), microhardness (p = < 0.05), ∆E00 (p = 0.010), and ∆a (p = 0.008). GC presented higher cubic phase content, and the stained groups (GS, GSG and GSP) presented higher monoclinic content. The different finishing procedures affected roughness, dynamic elastic modulus, microhardness, and color of 5Y-PSZ; polishing being the finish that provides minors changes to the 5Y- PSZ. - Abstract
In this paper we evaluate the performance of point and interval estimators based on the maximum likelihood(ML) method for the nonlinear simplex regression model. Inferences based on traditional maximum likelihood estimation have good asymptotic properties, but their performance in small samples may not be satisfactory. At out set we consider the maximum likelihood estimation for the parameters of the nonlinear simplex regression model, and so we introduced a bootstrap-based correction for such estimators of this model. We also develop the percentile and bootstrapt confidence intervals for those parameters as competitors to the traditional approximate confidence interval based on the asymptotic normality of the maximum likelihood estimators (MLEs). We then numerically evaluate the performance of these different methods for estimating the simplex regression model. The numerical evidence favors inference based on the bootstrap method, in special the bootstrapt interval, which was decisive in an application to real data., Competing Interests: The authors have declared that no competing interests exist.

- Abstract
Determining health risks associated with per-/polyfluoroalkyl substances (PFASs) is a highly complex problem requiring massive efforts for scientists, risk assessors, and regulators. Among the most poorly understood pressing questions is the relative importance of pre-PFAAs, which are PFASs that degrade to highly persistent perfluoroalkyl acids. How many of the vast number of existing pre-PFAAs are relevant for direct human exposure, and what are the predominant exposure pathways? What evidence of direct exposure to pre-PFAAs is provided by human biomonitoring studies? How important are pre-PFAAs and their biotransformation products for human health risk assessment? This article outlines recent progress and recommendations toward widening the lens on human PFAS exposure to include the pre-PFAA subclass.

49. A Deep Dive into the Complex Chemical Mixture and Toxicity of Tire Wear Particle Leachate in Fathead Minnow.

The ecological impact of tire wear particles in aquatic ecosystems is a growing environmental concern. We combined toxicity testing, using fathead minnow (Pimephales promelas) embryos, with nontarget high-resolution liquid chromatography Orbitrap mass spectrometry to characterize the toxicity and chemical mixture of organic chemicals associated with tire particle leachates. We assessed: 1) exposure to tire particle leachates after leaching for 1-, 3-, and 10-d; and 2) the effect of the presence and absence of small tire particulates in the leachates. We observed a decrease in embryonic heart rates, hatching success, and lengths, as well as an increase in the number of embryos with severe deformities and diminished eye and body pigmentation, after exposure to the leachates. Overall, there was a pattern whereby we observed more toxicity in the 10-d leachates, and greater toxicity in unfiltered leachates. Redundancy analysis showed that several benzothiazoles and aryl-amines were correlated with the toxic effects observed in the embryos. These included benzothiazole, 2-aminobenzothiazole, 2-mercaptobenzothiazole, N,N'-diphenylguanidine, and N,N'-diphenylurea. However, many other chemicals characterized as unknowns are likely to also play a key role in the adverse effects observed. Our study provides insight into the types of chemicals likely to be important toxicological drivers in tire leachates, and improves our understanding of the ecotoxicological impacts of tire wear particles. 50. Initial Muscle Quality Affects Individual Responsiveness of Interleukin-6 and Creatine Kinase following Acute Eccentric Exercise in Sedentary Obese Older Women.

This study aimed to evaluate the time course and responsiveness of plasma interleukin-6 (IL-6) and creatine kinase (CK) levels following acute eccentric resistance exercise in sedentary obese older women with a different muscle quality index (MQI). Eighty-eight participants (69.4 ± 6.06 years) completed an acute eccentric resistance exercise (7 sets of 10 repetitions at 110% of 10-repetition maximum with 3 min rest interval). Participants were divided into two groups: high or low MQI according to 50th percentile cut-off. The responsiveness was based on minimal clinical important difference. There were no differences between groups and time on IL-6 and CK levels (p > 0.05). However, the high MQI group displayed a lower proportion of low responders (1 for laboratory and 2 for field-based vs. 5 and 4) and a higher proportion of high responders for IL-6 (7 for laboratory and 6 for field-based vs. 4 and 5) compared to low MQI group. In addition, the high MQI group showed a higher proportion of high responders for CK (11 for laboratory and 9 for field-based vs. 6 and 6) compared to low MQI. A prior MQI screening can provide feedback to understand the magnitude response. Individual responsiveness should be taken into consideration for maximizing eccentric exercise prescription.
