Acoustic radiation force optical coherence elastography (ARF-OCE) has been successfully implemented to characterize the biomechanical properties of soft tissues, such as the cornea and the retina, with high resolution using single-element ultrasonic transducers for ARF excitation. Most currently proposed OCE techniques, such as air puff and ARF, have less capability to control the spatiotemporal information of the induced region of deformation, resulting in limited accuracy and low temporal resolution of the shear wave elasticity imaging. In this study, we propose a new method called 2-D ultrasonic array-based OCE imaging, which combines the advantages of 3-D dynamic electronic steering of the 2-D ultrasonic array and high-resolution optical coherence tomography (OCT). The 3-D steering capability of the 2-D array was first validated using a hydrophone. Then, the combined 2-D ultrasonic array OCE system was calibrated using a homogenous phantom, followed by an experiment on ex vivo rabbit corneal tissue. The results demonstrate that our newly developed 2-D ultrasonic array-based OCE system has the capability to map tissue biomechanical properties accurately, and therefore, has the potential to be a vital diagnostic tool in ophthalmology.

While cell-based immunotherapy, especially chimeric antigen receptor (CAR)-expressing T cells, is becoming a paradigm-shifting therapeutic approach for cancer treatment, there is a lack of general methods to remotely and noninvasively regulate genetics in live mammalian cells and animals for cancer immunotherapy within confined local tissue space. To address this limitation, we have identified a mechanically sensitive Piezo1 ion channel (mechanosensor) that is activatable by ultrasound stimulation and integrated it with engineered genetic circuits (genetic transducer) in live HEK293T cells to convert the ultrasound-activated Piezo1 into transcriptional activities. We have further engineered the Jurkat T-cell line and primary T cells (peripheral blood mononuclear cells) to remotely sense the ultrasound wave and transduce it into transcriptional activation for the CAR expression to recognize and eradicate target tumor cells. This approach is modular and can be extended for remote-controlled activation of different cell types with high spatiotemporal precision for therapeutic applications.

This paper reports the fabrication, characterization, and microparticle manipulation capability of an adjustable multi-scale single beam acoustic tweezers (SBAT) that is capable of flexibly changing the size of "tweezers" like ordinary metal tweezers with a single-element ultrahigh frequency (UHF) ultrasonic transducer. The measured resonant frequency of the developed transducer at 526 MHz is the highest frequency of piezoelectric single crystal based ultrasonic transducers ever reported. This focused UHF ultrasonic transducer exhibits a wide bandwidth (95.5% at -10 dB) due to high attenuation of high-frequency ultrasound wave, which allows the SBAT effectively excite with a wide range of excitation frequency from 150 to 400 MHz by using the "piezoelectric actuator" model. Through controlling the excitation frequency, the wavelength of ultrasound emitted from the SBAT can be changed to selectively manipulate a single microparticle of different sizes (3-100 μm) by using only one transducer. This concept of flexibly changing "tweezers" size is firstly introduced into the study of SBAT. At the same time, it was found that this incident ultrasound wavelength play an important role in lateral trapping and manipulation for microparticle of different sizes. Biotechnol. Cardiovascular diseases are the leading cause of fatalities in the United States. Atherosclerotic plaques are one of the primary complications that can lead to strokes and heart attacks if left untreated. It is essential to diagnose the disease early and distinguish vulnerable plaques from harmless ones. Many methods focus on the structural or molecular properties of plaques. Mechanical properties have been shown to change drastically when abnormalities develop in arterial tissue. We report the development of an acoustic radiation force optical coherence elastography (ARF-OCE) system that uses an integrated miniature ultrasound and optical coherence tomography (OCT) probe to map the relative elasticity of vascular tissues. We demonstrate the capability of the miniature probe to map the biomechanical properties in phantom and human cadaver carotid arteries.

We report on a real-time acoustic radiation force optical coherence elastography (ARF-OCE) system to map the relative elasticity of corneal tissue. A modulated ARF is used as excitation to vibrate the cornea while OCE serves as detection of tissue response. To show feasibility of detecting mechanical contrast using this method, we performed tissue-equivalent agarose phantom studies with inclusions of a different stiffness. We obtained 3-D elastograms of a healthy cornea and a highly cross-linked cornea. Finally we induced a stiffness change on a small portion of a cornea and observed the differences in displacement.

In this study, we have developed an acoustic radiation force orthogonal excitation optical coherence elastography (ARFOE-OCE) method for the visualization of the shear wave and the calculation of the shear modulus based on the OCT Doppler variance method. The vibration perpendicular to the OCT detection direction is induced by the remote acoustic radiation force (ARF) and the shear wave propagating along the OCT beam is visualized by the OCT M-scan. The homogeneous agar phantom and two-layer agar phantom are measured using the ARFOE-OCE system. The results show that the ARFOE-OCE system has the ability to measure the shear modulus beyond the OCT imaging depth. The OCT Doppler variance method, instead of the OCT Doppler phase method, is used for vibration detection without the need of high phase stability and phase wrapping correction. An M-scan instead of the B-scan for the visualization of the shear wave also simplifies the data processing.

This paper presents simulation, fabrication, and characterization of single-element ultrahigh frequency (100-300-MHz) needle ultrasonic transducers based on 0-3 composite Pb(Zr0.52Ti0.48)O3 (PZT) films prepared by using composite ceramic sol-gel film and sol-infiltration technique. The center frequency of the developed transducer at 300-MHz was the highest frequency of PbTiO3 ceramic-based ultrasonic transducers ever reported. Furthermore, a brief description of the composite model was followed by the development of a new expression for predicting the longitudinal velocity, the clamped dielectric constant, and the complex electromechanical coupling coefficient kt of these films, which is very important in ultrasonic transducer design. Moreover, these parameters are difficult to obtain by measuring the frequency dependence of impedance and phase angle because of the weak signal of the previous 0-3 composite films transducer (>100 MHz). The modeling results show that the Cubes model with a geometric factor n = 0.05 fits well with the measured data. This model will be helpful for developing the 0-3 composite systems for ultrahigh frequency ultrasonic transducer design.

Intravascular photoacoustic imaging at 1.7 μm spectral band has shown promising capabilities for lipid-rich vulnerable atherosclerotic plaque detection. In this work, we report a high speed catheter-based integrated intravascular photoacoustic/intravascular ultrasound (IVPA/IVUS) imaging system with a 500 Hz optical parametric oscillator laser at 1725 nm. A lipid-mimicking phantom and atherosclerotic rabbit abdominal aorta were imaged at 1 frame per second, which is two orders of magnitude faster than previously reported in IVPA imaging with the same wavelength. Clear photoacoustic signals by the absorption of lipid rich deposition demonstrated the ability of the system for high speed vulnerable atherosclerotic plaques detection.

We report on a novel acoustic radiation force orthogonal excitation optical coherence elastography (ARFOE-OCE) technique for imaging shear wave and quantifying shear modulus under orthogonal acoustic radiation force (ARF) excitation using the optical coherence tomography (OCT) Doppler variance method. The ARF perpendicular to the OCT beam is produced by a remote ultrasonic transducer. A shear wave induced by ARF excitation propagates parallel to the OCT beam. The OCT Doppler variance method, which is sensitive to the transverse vibration, is used to measure the ARF-induced vibration. For analysis of the shear modulus, the Doppler variance method is utilized to visualize shear wave propagation instead of Doppler OCT method, and the propagation velocity of the shear wave is measured at different depths of one location with the M scan. In order to quantify shear modulus beyond the OCT imaging depth, we move ARF to a deeper layer at a known step and measure the time delay of the shear wave propagating to the same OCT imaging depth. We also quantitatively map the shear modulus of a cross-section in a tissue-equivalent phantom after employing the B scan.

Inadequate replacement of lost ventricular myocardium from myocardial infarction leads to heart failure. Investigating the regenerative capacity of mammalian hearts represents an emerging direction for tissue engineering and cell-based therapy. Recent advances in stem cells hold promise to restore cardiac functions. However, embryonic or induced pluripotent stem cell-derived cardiomyocytes lack functional phenotypes of the native myocardium, and transplanted tissues are not fully integrated for synchronized electrical and mechanical coupling with the host. In this context, this review highlights the mechanical and electrical strategies to promote cardiomyocyte maturation and integration, and to assess the functional phenotypes of regenerating myocardium. Simultaneous microelectrocardiogram and high-frequency ultrasound techniques will also be introduced to assess electrical and mechanical coupling for small animal models of heart regeneration.

In this Letter, we present a trimodality imaging system and an intravascular endoscopic probe for the detection of early-stage atherosclerotic plaques. The integrated system is able to acquire optical coherence tomography (OCT), fluorescence, and ultrasound images and simultaneously display them in real time. A trimodality intravascular endoscopic probe of 1.2 mm in diameter and 7 mm in length was fabricated based on a dual-modality optical probe that integrates OCT and fluorescence imaging functions and a miniature ultrasound transducer. The probe is capable of rotating at up to 600 rpm. Ex vivo images from rabbit aorta and human coronary arteries showed that this combined system is capable of providing high resolution, deep penetration depth and specific molecular fluorescence contrast simultaneously.

Lipid deposition inside the arterial wall is a key indicator of plaque vulnerability. An intravascular photoacoustic (IVPA) catheter is considered a promising device for quantifying the amount of lipid inside the arterial wall. Thus far, IVPA systems suffered from slow imaging speed (~50 s per frame) due to the lack of a suitable laser source for high-speed excitation of molecular overtone vibrations. Here, we report an improvement in IVPA imaging speed by two orders of magnitude, to 1.0 s per frame, enabled by a custom-built, 2-kHz master oscillator power amplifier (MOPA)-pumped, barium nitrite [Ba(NO3)2] Raman laser. This advancement narrows the gap in translating the IVPA technology to the clinical setting.

Myocardial infarction results in scar tissue and irreversible loss of ventricular function. Unlike humans, zebrafish has the capacity to remove scar tissue after injury. To assess ventricular function during repair, we synchronized microelectrocardiogram (μECG) signals with a high-frequency ultrasound pulsed-wave (PW) Doppler to interrogate cardiac hemodynamics. μECG signals allowed for identification of PW Doppler signals for passive (early [E]-wave velocity) and active ventricular filling (atrial [A]-wave velocity) during diastole. We designed and developed a confocal acoustic radiation force optical coherence elastography system. A ring ultrasound transducer was used to achieve reflection mode excitation and generate an oscillating acoustic radiation force in order to generate displacements within the tissue, which were detected using the phase-resolved optical coherence elastography method. Both phantom and human tissue tests indicate that this system is able to sense the stiffness difference of samples and quantitatively map the elastic property of materials. Our confocal setup promises a great potential for point by point elastic imaging in vivo and differentiation of diseased tissues from normal tissue.

By placing the OCT ball lens and IVUS 45MHz single element transducer back-to-back at the same axial position, this probe can provide automatically co-registered, co-axial OCT-IVUS imaging. To demonstrate its capability, 3D OCT-IVUS imaging of a pig’s coronary artery in real-time displayed in polar coordinates, as well as images of two major types of advanced plaques in human cadaver coronary segments, was obtained using this probe and our upgraded system. Histology validation is also presented.

For the diagnosis of atherosclerosis, biomedical imaging techniques such as intravascular ultrasound (IVUS) and optical coherence tomography (OCT) have been developed. The combined use of IVUS and OCT is hypothesized to remarkably increase diagnostic accuracy of vulnerable plaques. We have developed an integrated IVUS-OCT imaging apparatus, which includes the integrated catheter, motor drive unit, and imaging system. The dual-function imaging catheter has the same diameter of current clinical standard. The imaging system is capable for simultaneous IVUS and OCT imaging in real time. Ex vivo and in vivo experiments on rabbits with atherosclerosis were conducted to demonstrate the feasibility and superiority of the integrated intravascular imaging modality.

OBJECTIVE: Combined use of optical coherence tomography (OCT) and intravascular ultrasound (IVUS) is a potential method for accurate assessment of plaques characteristics and vulnerability. The aim of this study is to develop and evaluate the feasibility of a fully integrated intracoronary OCT-IVUS imaging technique to visualize plaques in living animals. BACKGROUND: In vivo imaging of plaques by an integrated OCT-IVUS system has not been reported. METHODS: Simultaneous real-time OCT-IVUS imaging is performed using an integrated OCT-IVUS system and a single fully-integrated catheter with a 3.6F outer-diameter same as a commercial IVUS catheter. RESULTS: To verify in vivo imaging capability of this technique, five atherosclerotic-model rabbits and a swine were imaged. Images were obtained in these animals without complications. ObjectivesUltrasound backscatter microscopy (UBM), or ultrasound biomicroscopy, is a noninvasive, label-free, and ionizing radiation-free technique allowing high-resolution 3-dimensional structural imaging. The goal of this study was to evaluate UBM for resolving anatomic features associated with squamous cell carcinoma of the oral cavity.MethodsThe study was conducted in a hamster buccal pouch model. A carcinogen was topically applied to cheeks of 14 golden Syrian hamsters. Six additional hamsters served as healthy controls. A high-frequency (41 MHz, 6-mm focal depth, lateral and axial resolutions of 65 and 37 μm, respectively) UBM system was used for scanning the oral cavity after 14 weeks of carcinogen application. Histologic analyses were conducted on scanned regions.ResultsThe histologic structure of buccal tissue and microvasculature networks could be visualized from the UBM images. Epithelial and mucosal hypertrophy and neoplastic changes were identified in animals subjected to the carcinogen. In animals with invasive squamous cell carcinoma, lesion development and destruction of the structural integrity of tissue layers were noted.ConclusionsIn this pilot study, UBM generated sufficient contrast for morphologic features associated with oral carcinoma compared to healthy tissue. This modality may present a practical technique for detection of oral neoplasms that is potentially translatable to humans.

We report on a resonant acoustic radiation force optical coherence elastography (ARF-OCE) technique that uses mechanical resonant frequency to characterize and identify tissues of different types. The linear dependency of the resonant frequency on the square root of Young's modulus was validated on silicone phantoms. Both the frequency response spectrum and the 3D imaging results from the agar phantoms with hard inclusions confirmed the feasibility of deploying the resonant frequency as a mechanical contrast for tissue imaging. Furthermore, the results of resonant ARF-OCE imaging of a post-mortem human coronary artery with atherosclerosis demonstrate the potential of the resonant ARF-OCE as a non-invasive method for imaging and characterizing vulnerable plaques.

Coronary artery disease remains the leading cause of death and disability in the US. Intravascular ultrasound (IVUS) is one of the standard medical tests routinely performed in hospital to diagnose atherosclerosis by visualizing the structure of affected vessels and identifying the composition of plaques. Optical coherence tomography (OCT) possesses higher resolution to image vulnerable plaque near the lumen boundary, but with a shallow penetration depth. Thus, an integrated IVUS-OCT catheter would potentially capitalize the advantage of sufficient imaging depth of IVUS and the high resolution of OCT while offering improved diagnostic information. Different IVUS-OCT catheter designs have been reported previously by using either co-axial, co-planner or sequential configuration, which suffers from either the large size of catheter or suboptimal image co-registration. By placing the OCT probe and 45MHz single element IVUS transducer back-to-back at the same axial direction, this probe can provide automatically co-registered, co-axial IVUS-OCT imaging. To demonstrate its capability, IVUS-OCT imaging of major types of advanced plaques in human cadaver coronary segments was obtained using this catheter and system. Abstract.The catheter-based intravascular photoacoustic (IVPA) imaging for diagnosing atherosclerosis, which can provide optical absorption contrast of the arterial wall besides acoustic scattering contrast from the conventional intravascular ultrasound (IVUS) imaging, has been intensively researched recently. The resolution of IVPA is determined by the frequency bandwidth of an ultrasonic transducer. Higher resolution can be achieved by increasing the transducer’s working frequency and bandwidth. We introduce IVPA imaging at 35 and 80 MHz by using newly designed integrated IVUS/IVPA probes. This is the first time IVPA has been achieved as high as 80 MHz. Six-micrometer tungsten wires were imaged to evaluate the probes’ spatial resolutions and beam patterns. Healthy rabbit aorta was imaged in vitro. Imaging results show that IVPA has superior contrast over IVUS in identifying the arterial wall, and IVPA at 80 MHz demonstrates extraordinary resolution (35 μm) compared to 35 MHz.

For the diagnosis of atherosclerosis, biomedical imaging techniques such as intravascular ultrasound (IVUS) and optical coherence tomography (OCT) have been developed. The combined use of IVUS and OCT is hypothesized to remarkably increase diagnostic accuracy. We report our progress on the probe design and imaging system for achieving a miniature catheter that is capable for in vivo animal study. The integrated IVUS-OCT catheter is featured by a sequential arrangement of an ultrasound transducer and an OCT probe. The capability of the integrated IVUS-OCT imaging catheter and system is demonstrated by in vitro and in vivo imaging experiments of rabbit abdominal aorta. © 2012 IEEE.

Arterial wall is composed of three layers: intima, media and adventitia. Intima-media thickness (IMT) is an important prognostic indicator of atherosclerotic diseases. Although intravascular ultrasound (IVUS) imaging is a commonly used method for delineation of the layered structures, it is inferior to the optical absorption contrast offered by intravascular photoacoustic (IVPA) imaging. We introduce an integrated miniature probe that combines the capabilities of IVUS and IVPA imaging for the evaluation of arterial wall layered structures. Healthy rabbit aorta was imaged ex vivo. IVPA results showed superior contrast over IVUS in identifying the layered structures of arterial wall. © 2012 SPIE.

A miniature integrated optical coherence tomography (OCT) - ultrasound (US) probing system for real-time intravascular imaging has been developed. The outer diameter of the integrated probe is 0.69 mm, which is small enough for imaging in human coronary arteries. This probe, which has high resolution and deep tissue penetration, is designed to identify vulnerable atherosclerotic plaques in coronary arteries. This study presents a new multimodal imaging approach that includes high-frequency ultrasound, fluorescence intensity, confocal, and spectral imaging to improve the preclinical evaluation of new therapeutics in vivo. Here we use this approach to assess in vivo the therapeutic efficacy of the novel chemotherapy construct, HerDox during and after treatment. HerDox is comprised of doxorubicin non-covalently assembled in a viral-like particle targeted to HER2+ tumor cells, causing tumor cell death at over 10-fold lower dose compared to the untargeted drug, while sparing the heart. Whereas our initial proof-of-principle studies on HerDox used tumor growth/shrinkage rates as a measure of therapeutic efficacy, here we show that multimodal imaging deployed during and after treatment can supplement traditional modes of tumor monitoring to further characterize the particle in tissues of treated mice. Specifically, we show here that tumor cell apoptosis elicited by HerDox can be monitored in vivo during treatment using high frequency ultrasound imaging, while in situ confocal imaging of excised tumors shows that HerDox indeed penetrated tumor tissue and can be detected at the subcellular level, including in the nucleus, via Dox fluorescence. In addition, ratiometric spectral imaging of the same tumor tissue enables quantitative discrimination of HerDox fluorescence from autofluorescence in situ. In contrast to standard approaches of preclinical assessment, this new method provides multiple/complementary information that may shorten the time required for initial evaluation of in vivo efficacy, thus potentially reducing the time and cost for translating new drug molecules into the clinic.

A miniature integrated intravascular optical coherence tomography (OCT) - ultrasound (US) catheter for real-time imaging of atherosclerotic plaques has been developed, providing high resolution and deep tissue penetration at the same time. This catheter, with an outer diameter of 1.18mm, is suitable for imaging in human coronary arteries. The first in vivo 3D imaging of atherosclerotic microstructure in a rabbit abdominal aorta obtained by an integrated OCT-US catheter is presented. In addition, in vitro imaging of cadaver coronary arteries were conducted to demonstrate the imaging capabilities of this integrated catheter to classify different atherosclerotic plaque types.

We report on the synergy of an integrated ultrasound (US) and photoacoustic (PA) probe system for intravascular imaging. The combined dual-modality probe is based on a 39 MHz ring-shaped US transducer which detects both US echoes and laser-generated PA signals. By combining optical fiber, US transducer, and micromirror, we can obtain intravascular cross-sectional B-scan images by internal illumination of the sample. The performance of the probe is evaluated in a phantom study. Moreover, the coaxially designed probe also provides co-registered US and PA images of a normal rabbit aorta, which demonstrates the imaging ability of the dual-functional system, implying future clinical applications.

Detection of atherosclerotic plaque vulnerability has critical clinical implications for avoiding sudden death in patients with high risk of plaque rupture. We report on multimodality imaging of ex-vivo human carotid plaque samples using a system that integrates fluorescence lifetime imaging (FLIM), ultrasonic backscatter microscopy (UBM), and photoacoustic imaging (PAI). Biochemical composition is differentiated with a high temporal resolution and sensitivity at the surface of the plaque by the FLIM subsystem. 3D microanatomy of the whole plaque is reconstructed by the UBM. Functional imaging associated with optical absorption contrast is evaluated from the PAI component. Simultaneous recordings of the optical, ultrasonic, and photoacoustic data present a wealth of complementary information concerning the plaque composition, structure, and function that are related to plaque vulnerability. This approach is expected to improve our ability to study atherosclerotic plaques. The multimodal system presented here can be translated into a catheter based intraluminal system for future clinical studies.

We have developed a miniature integrated optical coherence tomography (OCT) ultrasound (US) probing system for intravascular imaging applications. In the OCT probe, the light coming out of a single mode fiber is focused by a gradient-index lens and then reflected by a right-angle prism from the side of the probe into the sample. It was combined with a 35 MHz PMN-PT side-viewing ultrasound transducer to obtain the ultrasound image as well. The OCT and ultrasound probes were integrated as a single probe to obtain OCT and ultrasound images simultaneously. The integrated probe has an outer diameter of 0.69 mm which, to our knowledge, is the smallest integrated OCT-US probe reported. Fast data acquisition and processing was implemented for real-time imaging. In vitro OCT and US images of human coronary artery with pathology, as well as in vivo images of normal rabbit abdominal aorta, were obtained using the integrated OCT-US probe to demonstrate its capability.

We report on the development of a multimodal optical coherence tomography (OCT) - ultrasound (US) system and miniaturized OCT-US probe for intravascular imaging. Both OCT optical components and a US transducer were integrated into a single probe, enabling both OCT and US imaging at the same time. A miniaturized OCT-US probe using a single element transducer was designed with a maximum outer diameter of 0.8 mm, which is suitable for in vivo intravascular imaging. The integrated OCT-US imaging system adopted a two-channel data acquisition card to digitize both OCT and US signals. Simultaneous OCT and US data processing and image display were also achieved using our home-developed software. In vitro OCT and US imaging of human aortic tissue was performed using this multimodal imaging system, which demonstrated the feasibility of the OCT-US system in intravascular imaging and its potential in detection of atherosclerotic plaques. © 2011 Copyright Society of Photo-Optical Instrumentation Engineers (SPIE).

For in vivo medical applications, endoscopy shows great potential for its minimally invasive manner, flexibility and close-up imaging characteristic. A miniaturized imaging probe combining ultrasound and photoacoustic endoscopy has been developed. The output of a 532-nm pulse laser was coupled into and delivered to the probe by a 200-micron-core multimode fiber. A 40 MHz ring shape ultrasound transducer was fabricated to receive pulse echo ultrasound and photoacoustic signals as well. The light-guiding optical fiber, the ring ultrasound transducer, and a mirror-based reflective material for the coaxial laser beam and ultrasound signal were integrated into the probe with a final packaged diameter of 2.5 mm. The performance of the probe was tested by imaging a graphite rod. The imaging ability of this dual-modality system was demonstrated by imaging the cross section of a rabbit aorta. © 2011 SPIE.
