Local chemical short-range ordering (SRO) and spatial fluctuations of planar fault energy are important features of multi-element and metastable complex concentrated alloys (CCAs). Arising from them, dislocations in such alloys are distinctively wavy in both static and migrating conditions; yet, such effects on strength have remained unknown. In this work, molecular dynamics simulations are used to show that the wavy configurations of dislocations and their jumpy motion in a prototypic CCA of NiCoCr are due to the local fluctuations of the energy of SRO shear-faulting that accompanies dislocation motion, with the dislocation getting pinned at sites of hard atomic motifs (HAMs) associated with high local shear-fault energies. Unlike the global averaged shear-fault energy which in general will subdue on successive dislocation passes, the local fluctuations in the fault energy always remain in a CCA, thus offering a strength contribution that is unique in such alloys. Analysis of the magnitude of this form of dislocation resistance shows that this is dominating over contributions due to elastic misfit of alloying elements and is in good agreement with strengths predicted from molecular dynamics simulations and experiments. This work has unfolded the physical basis of strength in CCAs, which is important for the development of these alloys into useful structural materials.

HIV gene expression is modulated by the combinatorial activity of the HIV transcriptional activator, Tat, host transcription factors, and chromatin remodeling complexes. To identify host factors regulating HIV transcription, we used specific single-guide RNAs and endonuclease-deficient Cas9 to perform chromatin affinity purification of the integrated HIV promoter followed by mass spectrometry. The scaffold protein, p32, also called ASF/SF2 splicing factor-associated protein, was identified among the top enriched factors present in actively transcribing HIV promoters but absent in silenced ones. Chromatin immunoprecipitation analysis confirmed the presence of p32 on active HIV promoters and its enhanced recruitment by Tat. HIV uses Tat to efficiently recruit positive transcription elongation factor b (p-TEFb) (CDK9/CCNT1) to TAR, an RNA secondary structure that forms from the first 59 bp of HIV transcripts, to enhance RNAPII transcriptional elongation. The RNA interference of p32 significantly reduced HIV transcription in primary CD4 + T cells and in HIV chronically infected cells, independently of either HIV splicing or p32 anti-splicing activity. Conversely, overexpression of p32 specifically increased Tat-dependent HIV transcription. p32 was found to directly interact with Tat’s basic domain enhancing Tat stability and half-life. Conversely, p32 associates with Tat via N- and C-terminal domains. Likely due its scaffold properties, p32 also promoted Tat association with TAR, p-TEFb, and RNAPII enhancing Tat-dependent HIV transcription. In sum, we identified p32 as a host factor that interacts with and stabilizes Tat protein, promotes Tat-dependent transcriptional regulation, and may be explored for HIV-targeted transcriptional inhibition.

Background: The epidemiological data of Streptococcus pneumoniae isolates are important for the practice of treatment and prevention. This research aimed to explore the epidemiological characteristics of pediatric S. pneumoniae isolated from outpatients and inpatients. Methods: S. pneumoniae were isolated from unsterile samples of inpatients and outpatients younger than five years old between March 2013 and February 2014. The serotypes were determined by diagnostic pneumococcal antisera, and resistance against 13 antibiotics was tested by either the E-test or the disc diffusion method. The sequence types (STs) were analyzed with multilocus sequence typing (MLST). Results: The five dominant serotypes obtained from inpatients were 19F(32.9%), 19A(20.7%), 23F(10.7%), 6A(10.0%), and 14 (8.6%), while those in the outpatients were 19F (13.6%), 23F (12.9%), 6A (10.0%), 6B (10.0%), and 19A (7.9%). The coverage rates of the 7-, 10- and 13-valent pneumococcal vaccine formulations were high. The non-susceptibility to penicillin, cefuroxime, imipenem, erythromycin, and trimethoprim-sulfamethoxazole among the inpatient isolates were 7.1%, 92.8%, 65.7%, 100%, and 85.0%, respectively, while those among the outpatient isolates were 0.7%, 50.0%, 38.6%, 96.4%, and 65.7%, respectively. There were 45 and 81 STs detected from the pneumococci isolated from inpatients and outpatients, respectively. CC271 was more prevalent in inpatients. Conclusions: The pneumococcal vaccine related serotypes were still prevalent either in inpatient department or in outpatient department, which with serious antibiotic resistance. These results might be helpful forunderstanding the epidemiology of S. pneumoniae in Beijing. Therefore, universal immunization of PCVs should be implemented to prevent the spread of vaccine related serotypes of S. pneumoniae.

Background: Kechuangping mixture (KCPM) based on Maxingshigan decoction has a significant clinical effect in the treatment of pediatric pneumonia, especially for asthmatic pneumonia. This study explored the effects of KCPM on lipopolysaccharide (LPS)-induced macrophage immune response and the molecular mechanism involved. Methods: CCK-8 assays was used to detect the effect of KCPM on cell viability. The RAW 264.7 macrophages were divided into four groups: blank, LPS, KCPM, LPS+KCPM. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) was used to examine the secretion of inflammatory factors and the expression of TLR4/MyD88/NF-κB signal pathways with or without KCPM stimulation. Results: KCPM inhibited the differentiation of RAW264.7 macrophages induced by LPS. The secretion of pro-inflammatory cytokines IL-6 and TNF-α induced by LPS was decreased by pre-treatment with KCPM. While, treatment with KCPM advanced, the anti-inflammatory cytokines IL-10 and TGF-β increased significantly. Additionally, KCPM exhibited a potent inhibitory effect on the expression of iNOS and a promotive effect on the expression of Arg-1. Moreover, KCPM clearly suppressed the transcription level of TLR4, MyD88 and NF-κB signal pathways.Conclusion: KCPM promoted the macrophages polarization toward to M2 phenotype, as a result, limited the macrophage immune response. TLR4/MyD88/NF-κB signal pathways may play an important role in this process.

While legal scholarship seeks mainly to assess the impact of climate change litigation (CCL) on the regulatory state and on climate change policy in common law countries, the potential influence of government climate policy on the judicial practices of jurisdictions with different legal traditions attracts much less attention. This article fills the gaps by exploring how courts in China, an authoritarian country with a civil law tradition, react to government climate policies and how this judicial response might affect relevant legal rules and eventually contribute to climate regulation. An empirical analysis of 177 Chinese judicial cases reveals that CCL in China consists mostly of contract-based civil actions steered by the government's low-carbon policies. Moreover, although the prospects of CCL against public authorities in China remain very bleak, there is scope for the emergence of tort-based CCL, backed by government policies. In this respect, recent tort-based public interest litigation on air pollution in China may serve as a substitute or, more promisingly, a gateway to the emergence of a tort-based branch of Chinese CCL.

Non-heme iron and α-ketoglutarate-dependent (Fe/αKG) oxygenases catalyze various oxidative biotransformations. Due to their catalytic flexibility and high efficiency, Fe/αKG oxygenases have attracted keen attention for their application as biocatalysts. Here, we report the biochemical and structural characterizations of the unusually promiscuous and catalytically versatile Fe/αKG oxygenase SptF, involved in the biosynthesis of fungal meroterpenoid emervaridones. The in vitro analysis reveals that SptF catalyzes four continuous oxidation reactions, including hydroxylation, desaturation, epoxidation, and skeletal rearrangement. SptF exhibits extremely broad substrate specificity toward various meroterpenoids, and efficiently produced novel cyclopropane-ring-fused 5/3/5/5/6/6 and 5/3/6/6/6 scaffolds from terretonins. Moreover, SptF also hydroxylates steroids, including androsterone, testosterone, and progesterone, with different regiospecificities. Crystallographic and structure-based mutagenesis studies of SptF reveal the molecular basis of the enzyme reactions, and suggest that the malleability of the loop region contributes to the remarkable substrate promiscuity. SptF exhibits great potential as a promising biocatalyst for oxidation reactions.

Shallow water hydrothermal vents are a source of heavy metals leading to their accumulation in marine organisms that manage to live under extreme environmental conditions. This is the case at Kueishantao (KST) shallow-sea vents system offshore northeast Taiwan, where the heavy metal distribution in vent fluids and ambient seawater is poorly understood. This shallow vent is an excellent natural laboratory to understand how heavy and volatile metals behave in the nearby water column and ecosystem. Here, we investigated the submarine venting of heavy metals from KST field and its impact on ambient surface seawater. The total heavy metal concentrations in the vent fluids and vertical plumes were 1–3 orders of magnitude higher than the overlying seawater values. When compared with deep-sea hydrothermal systems, the estimated KST end-member fluids exhibited much lower concentrations of transition metals (e.g., Fe and Mn) but comparable concentrations of toxic metals such as Pb and As. This may be attributed to the lower temperature of the KST reaction zone and transporting fluids. Most of the heavy metals (Fe, Mn, As, Y, and Ba) in the plumes and seawater mainly originated from hydrothermal venting, while Cd and Pb were largely contributed by external sources such as contaminated waters (anthropogenic origin). The spatial distribution of heavy metals in the surface seawater indicated that seafloor venting impacts ambient seawater. The measurable influence of KST hydrothermal activity, however, was quite localized and limited to an area of < 1 km². The estimated annual fluxes of heavy metals emanating from the yellow KST hydrothermal vent were: 430–2600 kg Fe, 24–145 kg Mn, 5–32 kg Ba, 10–60 kg As, 0.3–1.9 kg Cd, and 2–10 kg Pb. This study provides important data on heavy metals from a shallow-sea hydrothermal field, and it helps to better understand the environmental impact of submarine shallow hydrothermal venting.

The spread of carbapenem-resistant Klebsiella pneumoniae (CRKp) has become a significant problem worldwide; however, relevant data in children are limited. We performed a retrospective study to better understand the epidemiology of CRKp bloodstream infections at Beijing Children's Hospital. A total of 164 K. pneumoniae strains were collected from blood cultures between January 2011 and September 2014, of which 52 (31.7%) were CRKp strains. All 52 CRKp strains were multidrug resistant; 46 (88.5%) and 49 (94.2%) isolates were resistant to meropenem and imipenem, respectively. Low rates of resistance to amikacin (5.8%), levofloxacin (7.7%), and ciprofloxacin (15.4%) were observed. All isolates were susceptible to colistin. Among the tested carbapenem resistance genes, the predominant gene was blaNDM-1, detected in 28 (53.8%) isolates, followed by blaIMP-4 (19, 36.5%) and blaKPC-2 (4, 7.7%). The chemical compositions of gas discharges from the Kueishantao (KST) hydrothermal field changed dramatically from 2000 to 2014. In this study, we established a gas mixing model for the KST gases. The N2, Ar, and CO2 contents were mixed from a magmatic endmember with CO2 of about 990 mmol/mol, a hydrothermal and an atmospheric endmember enriched in N2 and Ar. More than 71% KST gas components were mantle-derived/magmatic. The calculated endmember N2/Ar ratio and Ar contents of the hydrothermal endmember (percolated fluid) are about 140 and 5.28–5.52 mmol/mol, respectively. This relatively elevated N2/Ar ratio was probably caused by the thermogenic addition of N2. The log(CH4/CO2) values of the KST gas samples correlate well with the mixing temperature that estimated from the mixing ratio between the percolated fluid and the magmatic endmember. It is indicated that the KST CH4 and CO2 may have attained chemical equilibrium. The temporal variations of the KST gas compositions are determined by the mixing ratio, which is dependent on the magmatic activity underneath the KST field. With the decreasing of magmatic activity since 2005, the proportion of the hydrothermal endmember increased, along with the increasing of N2, Ar, and CH4 contents. This study proposed an effective model to quantitatively assess the sources of gas components discharged from submarine hydrothermal vents. In addition, it is suggested that the mixing between a magmatic and a hydrothermal endmember may play an important role in the concentrations of CO2 and CH4 in hydrothermal gas discharges.

Luffa sponge is an agricultural product with large global production. In this study, we studied the effect of ashing temperature and atmosphere on the physicochemical characteristics of luffa sponge. All of the ashed samples are amorphous materials with porous structures. The luffa sponge ashed in air (LSA) and luffa sponge ashed in nitrogen (LSN) show analogous elemental compositions. However, the oxygen in the air can promote the incineration and combustion of luffa sponge, while nitrogen atmosphere can hinder the decomposition of organic compounds due to carbonization. Their pore characteristics, therefore, vary with temperatures and atmospheres. The BET surface area, total pore volume, and mesopore volume generally increase with ashing temperature, due to the thermal destruction of organic matter in luffa sponge. LSA samples exhibit relatively higher surface area and total pore volume than LSN samples. Their mesopore volumes, however, are comparable, attributed to the preservation of original pores from enlargement under nitrogen atmosphere. With characteristics of low cost, low density, and comparable pore properties with traditional adsorbents, luffa sponge is a potential adsorbent for organic pollutants and a carrier for catalysts.

Background Streptococcus pneumoniae 7-valent conjugate vaccine (PCV7) was made available in China in the private sector in September 2008. Methods This study investigated the serotype distribution, antibiotic resistance, and molecular characteristics of S. pneumoniae in hospitalized pediatric patients. Pneumococcal isolates were collected from hospitalized children younger than 14 years. Their serotypes were determined using Quellung reactions with antisera; antibiotic resistance against 13 antimicrobials was tested using the E-test method or disc diffusion. The sequence types (STs) were analyzed with multilocus sequence typing. Results A total of 187 pneumococcal specimens were collected, including 21 invasive and 166 noninvasive isolates. The prevailing serotypes were 19F (31.6%), 19A (19.8%), 23F (11.2%), 6A (9.1%), 14 (9.1%) and 15B (5.9%). The coverage rates of PCV7, PCV10 and PCV13 were 56.2% (105/187), 56.7% (106/187) and 86.1% (161/187), respectively. The overall nonsusceptibility rate against penicillin was 8.0%; however, this rate would have been 91.5% if based on an oral breakpoint. All but one of the isolates were highly resistant to erythromycin. Multidrug resistance was exhibited by 177 (94.7%) isolates. The 5 predominant multilocus sequence typings for all pneumococci were ST271 (24.1%), ST320 (18.2%), ST81 (7.5%), ST876 (7%) and ST3397 (5.3%), which were primarily related to serotypes 19F, 19A, 23F, 14 and 15B, respectively. CC271 was the most frequent antibiotic-resistant complex clone. Conclusions The coverage rates of PCVs were high, and the antibiotic resistance rates were of serious concern among hospitalized children. Universal immunization using PCVs would likely prevent episodes of S. pneumoniae diseases and the spread of antibiotic resistance in Beijing.

Nanostructures loaded on halloysite nanotubes (HNTs) have attracted global interest, because the nanotubular HNTs could extend the range of their potential applications. In this study, we fabricated a novel nanocomposite with hollow iron nanoparticles loaded on the surface of HNTs. The structure of the iron nanoparticles can be adjusted by ageing time. Owing to the increased remnant magnetization and coercivity values, the nanocomposites loaded with hollow iron nanoparticles showed better electromagnetic performance than that with solid iron nanoparticles. This study opens a new pathway to fabricate halloysite nanotubular nanocomposites that may gain applications in the catalytic degradation of organic pollutants and electromagnetic wave absorption.

In order to study the heavy metal accumulation and distribution in the roots, stems, and leaves of Spartina alterniflora, we collected S. alterniflora samples and the associated sediments along three transects at the Andong tidal flat, Hangzhou Bay. Co, Ni, Cd, Pb, Cu, and Zn were mainly accumulated in the aerial parts (stems and leaves) of the plants, and their distributions depended on their mobility and their roles during the metabolism processes of S. alterniflora. The concentrations of Cu, Zn, Cd, Hg, and Pb were significantly enhanced with the increasing of heavy metal concentrations in the sediments, while those of Co and Ni remained relatively constant. Bioaccumulation factors results showed that the serious heavy metal contamination in the sediments from the transect A may overwhelm the accumulation capability of the plants. In addition, the physicochemical properties of the sediments and the pore water therein also play a role in the heavy metal concentrations and accumulations in the plants, because they can influence the behaviors and bioavailabilities of heavy metals during nutrition and bioaccumulation processes of the plants. The sediments with vegetation did not show significantly decreased heavy metal concentration with respect to the unvegetated sediments, although the plants did absorb heavy metals from the sediments. Principal component analysis and correlation analyses indicated that Co–Ni, Cu–Cd–Hg behaved coherently during accumulation, which may be ascribed to their similar accumulation mechanisms. This work provided essential information on the heavy metal accumulation by plants in a tidal flat, which will be useful for the environmental control through phytoremediation at estuaries.
