The Perseverance rover landed in Jezero crater, Mars, in February 2021. We used the Scanning Habitable Environments with Raman and Luminescence for Organics and Chemicals (SHERLOC) instrument to perform deep-ultraviolet Raman and fluorescence spectroscopy of three rocks within the crater. We identify evidence for two distinct ancient aqueous environments at different times. Reactions with liquid water formed carbonates in an olivine-rich igneous rock. A sulfate-perchlorate mixture is present in the rocks, which probably formed by later modifications of the rocks by brine. Fluorescence signatures consistent with aromatic organic compounds occur throughout these rocks and are preserved in minerals related to both aqueous environments.

Advances in Raman instrumentation have led to the implementation of a remote dispersive Raman spectrometer on the Perseverance rover on Mars, which is used for remote sensing. For remote applications, dispersive spectrometers suffer from a few setbacks such as relatively larger sizes, low light throughput, limited spectral ranges, relatively low resolutions for small devices, and high sensitivity to misalignment. A spatial heterodyne Raman spectrometer (SHRS), which is a fixed grating interferometer, helps overcome some of these problems. Most SHRS devices that have been described use two fixed diffraction gratings, but a variance of the SHRS called the one-grating SHRS (1g-SHRS) replaces one of the gratings with a mirror, which makes it more compact. In a recent paper we described monolithic two-gratings SHRS, and in this paper, we investigate a single-grating monolithic SHRS (1g-mSHRS), which combines the 1g-SHRS with a monolithic setup previously tested at the University of South Carolina. This setup integrates the beamsplitter, grating, and mirror into a single monolithic device. This reduces the number of adjustable components, allows for easier alignment, and reduces the footprint of the device (35 × 35 × 25 mm with a weight of 80 g). This instrument provides a high spectral resolution (∼9 cm−1) and large spectral range (7327 cm−1) while decreasing the sensitivity to alignment with a field of view of 5.61 mm at 3m. We discuss the characteristics of the 1g-mSHRS by measuring the time-resolved remote Raman spectra of a few inorganic salts, organics, and minerals at 3 m. The 1g-mSHRS makes a good candidate for planetary exploration because of its large spectral range, greater sensitivity, competitively higher spectral resolution, low alignment sensitivity, and high light throughput in a compact easily aligned system with no moving parts.

This study investigates the interfacial behavior of the proteinase K enzyme at air-water interface. Adsorption of enzyme on the surface was induced using saline subphase. The surface packing and stability of the enzyme was investigated using of surface pressure-area (π-A) and surface potential-area (ΔV-A) isotherms. Proteinase K enzyme forms film at air-aqueous interface and demonstrates good stability as shown through compression-decompression cycle experiments. To characterize the surface assembly morphology of the interfacial enzymes UV-vis and fluorescence spectroscopic techniques were used. The data revealed that the enzyme Langmuir monolayer has good homogeneity with no evidence of aggregates during compression. The secondary structure of the enzyme at interface was determined to be α-helix using p-polarized infrared-reflection absorption spectroscopy. This was confirmed through Circular dichroism spectra of the enzyme Langmuir-Blodgett (LB) film which showed that the major conformation present were α-helices.

A critical role of protein modifications such as phosphorylation and acetylation in synaptic plasticity and memory is well documented. Tyrosine sulfation plays important roles in several biological processes. However, its role in synaptic plasticity and memory is not well understood. Here, we show that sulfation contributes to long-term potentiation (LTP) in the hippocampal slices. In addition, inhibition of sulfation impairs long-term memory in a spatial memory task without affecting acquisition or short-term memory. Furthermore, LTP-inducing stimulus enhances protein tyrosine sulfation. These results suggest an important role for tyrosine sulfation in LTP and memory.

This article investigates the main aspects of the surface chemistry properties of the lactate oxidase (LacOx) enzyme monolayer at the air-subphase interface. Surface chemistry study determined the important properties like the surface packing and stability of the formed layer, whereas the spectroscopic experiments provided information regarding its secondary structure conformation of the enzyme. We have demonstrated that the LacOx in the monolayer form remained active for extended time period. In accordance to the data obtained from the isotherm it was also found that LacOx forms a stable monolayer that does not aggregate at the air-subphase interface. The stability of the monolayer at the air-subphase interface was studied by using compression-decompression cycles which revealed the stability with no significant evidence of aggregates or irreversible domains. This was further confirmed by UV-vis absorption and fluorescence measurements. Spectra from circular dichroism (CD) showed that the LB film retains the characteristic of an α-helix conformation.

Raman scattering (RS) by graphitic phases BCxN (g-BCxN), obtained by different methods, was studied using an excitation laser operating in the near-IR (NIR) range. With such excitation the most characteristic peaks for g-BCxN are 1300 cm–1 (D-peak) and 1505 cm–1 (G-peak). It is shown that the D- and G-peaks are associated with disordered graphite: the D-peak is observed because the selection rules change, and its intensity is correlated with the size of the graphite cluster sp2 in a polycrystalline graphite; the G-band is associated with thegraphite mode that is RS-active in the E2g plane. The shift of the D-peak in g-BCxN (1300 cm–1 ) as compared with its position in graphite (1355 cm–1 ) can be explained by the positional dispersion of the D-peak with excitation wavelength. The RS line centered at 1260 cm–1, which could be associated with a RS-active mode, is observed only for samples with high crystallinity and for RS spectra obtained at high pressures and high temperatures.

Clostridioides difficile infection (CDI) is the major identifiable cause of antibiotic-associated diarrhea. The emergence of hypervirulent C. difficile strains has led to increases in both hospital- and community-acquired CDI. Furthermore, the rate of CDI relapse from hypervirulent strains can reach up to 25%. Thus, standard treatments are rendered less effective, making new methods of prevention and treatment more critical. Previously, the bile salt analog CamSA (cholic acid substituted with m-aminosulfonic acid) was shown to inhibit spore germination in vitro and protect mice and hamsters from C. difficile strain 630. Here, we show that CamSA was less active in preventing spore germination by other C. difficile ribotypes, including the hypervirulent strain R20291. The strain-specific in vitro germination activity of CamSA correlated with its ability to prevent CDI in mice. Additional bile salt analogs were screened for in vitro germination inhibition activity against strain R20291, and the most active compounds were tested against other strains. An aniline-substituted bile salt analog, CaPA (cholic acid substituted with phenylamine), was found to be a better antigerminant than CamSA against eight different C. difficile strains. In addition, CaPA was capable of reducing, delaying, or preventing murine CDI signs with all strains tested. CaPA-treated mice showed no obvious toxicity and showed minor effects on their gut microbiome. CaPA’s efficacy was further confirmed by its ability to prevent CDI in hamsters infected with strain 630. These data suggest that C. difficile spores respond to germination inhibitors in a strain-dependent manner. However, careful screening can identify antigerminants with broad CDI prophylaxis activity.

This work investigates the physicochemical properties of mixed stearic acid (HSt)/phenylalanine dehydrogenase enzyme (PheDH) Langmuir films and their immobilization onto solid supports as Langmuir-Blodgett (LB) films. PheDH from the aqueous subphase enters the surfactant matrix up to an exclusion surface pressure of 25.3 mN/m, leading to the formation of stable and highly condensed mixed Langmuir monolayers. Hydrophobic interactions between the enzyme and HSt nonpolar groups tuned the secondary structure of PheDH, evidenced by the presence of β-sheet structures as demonstrated by infrared and circular dichroism spectra. The floating monolayers were successfully transferred to solid quartz supports, yielding Y-type LB films, and then characterized employing fluorescence, circular dichroism, and microscopic techniques, which indicated that PheDH was co-immobilized with HSt proportionally to the number of transferred layers. The enzyme fluidized the HSt monolayers, reducing their maximum dipoles when condensed to their maximum, and disorganized the alkyl chains of the fatty acid, as detected with infrared spectroscopy. The stability of the mixed floating monolayers enabled their transfer to solid supports as LB films, which is important for producing optical and electrochemical sensors for phenylalanine whose molecular architecture can be controlled with precision.

Carbon dots (CDs) are a novel class of nanoparticles with excellent properties. The development of CDs involves versatile synthesis, characterization, and various applications. However, the importance of surface chemistry of CDs, especially in applications, is often underestimated. In fact, the study of the surface chemistry of CDs is of great significance in the explanation of the unique properties of CDs as well as the pursuit of potential applications. In this feature article, we do not only introduce the development of CDs in our group but also highlight their applications where surface chemistry plays a critical role.

Carbon dots (CDs) are zero-dimensional carbon-based spherical nanoparticles with diameters less than 10 nm. Here, we report for the first time CDs forming stable Langmuir monolayers at the air-subphase interface. Langmuir monolayers are of great interest both fundamentally to study the interactions at the interfaces and for many applications such as the development of sensors. However, CDs usually do not form Langmuir monolayers because of their highly hydrophilic nature. In this study, amphiphilic CDs were prepared through hydrothermal carbonization using saccharides as the precursors. The surface chemistry behavior and optical properties of CDs at the air-subphase interface were studied. CDs derived from saccharides consistently formed stable Langmuir monolayers which show all essential phases, namely, gas, liquid-expanded, liquid-condensed, and solid phases. The compression-decompression cycle method showed minimum hysteresis (4.3%), confirming the retaining capacity of the CDs as a monolayer. Limiting CD areas from surface pressure-area isotherm at the air-subphase interface were used to calculate the average diameter of the CDs at the air-subphase interface. UV/vis absorption spectra of CDs dispersed in water and in Langmuir monolayers had the same bands in the UV region. The intensity of the UV/vis absorption increases with increasing surface pressure at the air-subphase interface. Interestingly, photoluminescence (PL) of the Langmuir monolayer of CDs was excitation-independent, whereas the same CDs had excitation-dependent PL when dispersed in water.

To develop the next generation of hypergolic, ionic-liquid-based fuels, it is important to understand the fundamental reaction mechanisms for the oxidation of ionic liquids (ILs). We consequently studied the oxidation of a levitated 1-butyl-3-methylimidazolium dicyanoborate ([BMIM][DCBH]) droplet by nitrogen dioxide (NO2). The properties of [BMIM][DCBH], including short ignition-delay times, low viscosities, and a wide liquid temperature range, make the ionic liquid especially suitable as a component of a hypergolic fuel. The chemical modifications were monitored with Fourier-transform infrared (FTIR), Raman, and ultraviolet–visible spectroscopies. To identify changes induced by the oxidation, it was first necessary to assign vibrational modes to the FTIR and Raman spectra of unreacted [BMIM][DCBH]. OrganiCam is a laser-induced luminescence imager and spectrometer designed for standoff organic and biosignature detection on planetary bodies. OrganiCam uses a diffused laser beam (12° cone) to cover a large area at several meters distance and records luminescence on half of its intensified detector. The diffuser can be removed to record Raman and fluorescence spectra from a small spot from 2 m standoff distance. OrganiCam’s small size and light weight makes it ideal for surveying organics on planetary surfaces. We have designed and built a brassboard version of the OrganiCam instrument and performed initial tests of the system.

On the NASA 2020 rover mission to Jezero crater, the remote determination of the texture, mineralogy and chemistry of rocks is essential to quickly and thoroughly characterize an area and to optimize the selection of samples for return to Earth. As part of the Perseverance payload, SuperCam is a suite of five techniques that provide critical and complementary observations via Laser-Induced Breakdown Spectroscopy (LIBS), Time-Resolved Raman and Luminescence (TRR/L), visible and near-infrared spectroscopy (VISIR), high-resolution color imaging (RMI), and acoustic recording (MIC). SuperCam operates at remote distances, primarily 2-7 m, while providing data at sub-mm to mm scales. We report on SuperCam's science objectives in the context of the Mars 2020 mission goals and ways the different techniques can address these questions. The instrument is made up of three separate subsystems: the Mast Unit is designed and built in France; the Body Unit is provided by the United States; the calibration target holder is contributed by Spain, and the targets themselves by the entire science team. This publication focuses on the design, development, and tests of the Mast Unit; companion papers describe the other units. The goal of this work is to provide an understanding of the technical choices made, the constraints that were imposed, and ultimately the validated performance of the flight model as it leaves Earth, and it will serve as the foundation for Mars operations and future processing of the data. In February 2021, NASA’s Perseverance rover will begin its exploration of Jezero crater near a putative ancient delta. Orbital mineralogy indicates the presence of carbonates and clay minerals in the landing site, which will be key targets for study. The SuperCam instrument provides an important tool for remotely surveying for these and other minerals using multiple techniques: Laser-Induced Breakdown Spectroscopy (LIBS), Time-Resolved Raman (TRR) and Luminescence (TRL) spectroscopies, Visible-Near Infrared (VisIR) spectroscopy, micro-imaging, and acoustics. TRR and TRL use a pulsed 532 nm laser with an adjustable gate width, from 100 ns to several ms. The time at which the gate opens is also adjustable, from coincident with the laser pulse to obtain Raman and fast luminescence out to 10 ms or more to capture the lifetimes of luminescence signals. These techniques will operate at distances up to 7 m from the rover mast and will be most effective if LIBS first removes dust from the targets and chemistry is subsequently obtained at the same location. Early lab results show that TRR is effective for detecting certain carbonates (magnesite, hydromagnesite, siderite, ankerite, calcite, and dolomite), sulfates (gypsum, anhydrite, barite, epsomite, and coquimbite), phosphates (apatite), and silicates (e.g., quartz, feldspar, forsteritic olivine, topaz, and diopside). Many of these minerals are high-priority targets for astrobiology studies because they represent habitable environments and have high biosignature preservation potential in terrestrial rocks. Raman signal strength is significantly decreased in fine-grained materials, however, and clay minerals will be a challenge to detect, as will opaque minerals such as Fe-oxides. TRL will be useful for identifying rare earth elements in phosphates and zircon, Fe3+ in silicates such as feldspar, Mn2+ in carbonates, and Cr3+ in Al-oxides and some silicates. We describe the fabrication of an underwater time-gated standoff Raman sensor, consisting of a custom Raman spectrometer, custom scanner, and commercial diode-pumped pulsed 532 nm laser all located inside a pressure housing. The Raman sensor was tested in the laboratory with samples in air, a tank containing tap water and seawater, and in the coastal Hawaiian harbor. We demonstrate our new system by presenting standoff Raman spectra of some of the chemicals used in homemade explosive devices and improvised explosive devices, including sulfur, nitrates, chlorates, and perchlorates up to a distance of ∼6 m in seawater and tap water. Finally, the Raman spectra of these hazardous chemicals sealed inside plastic containers submersed in the Hawaiian Harbor water are also presented.

The SuperCam instrument suite provides the Mars 2020 rover, Perseverance, with a number of versatile remote-sensing techniques that can be used at long distance as well as within the robotic-arm workspace. These include laser-induced breakdown spectroscopy (LIBS), remote time-resolved Raman and luminescence spectroscopies, and visible and infrared (VISIR; separately referred to as VIS and IR) reflectance spectroscopy. A remote micro-imager (RMI) provides high-resolution color context imaging, and a microphone can be used as a stand-alone tool for environmental studies or to determine physical properties of rocks and soils from shock waves of laser-produced plasmas. SuperCam is built in three parts: The mast unit (MU), consisting of the laser, telescope, RMI, IR spectrometer, and associated electronics, is described in a companion paper. The on-board calibration targets are described in another companion paper. Here we describe SuperCam's body unit (BU) and testing of the integrated instrument. The BU, mounted inside the rover body, receives light from the MU via a 5.8 m optical fiber. The light is split into three wavelength bands by a demultiplexer, and is routed via fiber bundles to three optical spectrometers, two of which (UV and violet; 245-340 and 385-465 nm) are crossed Czerny-Turner reflection spectrometers, nearly identical to their counterparts on ChemCam. The third is a high-efficiency transmission spectrometer containing an optical intensifier capable of gating exposures to 100 ns or longer, with variable delay times relative to the laser pulse. This spectrometer covers 535-853 nm ( 105 - 7070 cm − 1 Raman shift relative to the 532 nm green laser beam) with 12 cm − 1 full-width at half-maximum peak resolution in the Raman fingerprint region. The BU electronics boards interface with the rover and control the instrument, returning data to the rover. Thermal systems maintain a warm temperature during cruise to Mars to avoid contamination on the optics, and cool the detectors during operations on Mars. Results obtained with the integrated instrument demonstrate its capabilities for LIBS, for which a library of 332 standards was developed. Examples of Raman and VISIR spectroscopy are shown, demonstrating clear mineral identification with both techniques. Luminescence spectra demonstrate the utility of having both spectral and temporal dimensions. The application of β-galactosidase enzyme ranges from industrial use as probiotics to medically important application such as cancer detection. The irregular activities of β-galactosidase enzyme are directly related to the development of cancers. Identifying the location and expression levels of enzymes in cancer cells have considerable importance in early-stage cancer diagnosis and monitoring the efficacy of therapies. Most importantly, the knowledge of the efficient method of detection of β-galactosidase enzyme will help in the early-stage treatment of the disease. In this review paper, we provide an overview of recent advances in the detection methods of β-galactosidase enzyme in the living cells, including the detection strategies, and approaches in human beings, plants, and microorganisms such as bacteria. Further, we emphasized on the challenges and opportunities in this rapidly developing field of development of different biomarkers and fluorescent probes based on β-galactosidase enzyme. We found that previously used chromo-fluorogenic methods have been mostly replaced by the new molecular probes, although they have certain drawbacks. Upon comparing the different methods, it was found that near-infrared fluorescent probes are dominating the other detection methods.

Spatial heterodyne spectrometers are members of the static Fourier transform class of spectrometers, well-regarded for their ability to acquire high-resolution, high wavelength precision emission spectra in compact, light footprint packages. In a spatial heterodyne spectrometer experiment, a Fizeau fringe is generated for every spectral feature in a given spectrum, and spatial heterodyne spectrometer records the superposition of all Fizeau fringes in the spectrum on a detector. Hence, the sensitivity of spatial heterodyne spectrometers is constrained by uncorrelated, multiplicative photon noise that limits the detection of spectral features to those that are more luminous than the square root of the total incident flux onto the detector. In essence, powerful spectral features create a rising floor of noise that wash out less luminous features. In the present work, we introduce a novel spectrometer coupling, that being an Amici prism spectrometer in series with spatial heterodyne spectrometer, that correlates photon shot noise along one axis of a detector that in turn suppresses multiplicative photon noise within each row of the interferogram image. We demonstrate that this spectrometer pairing facilitates the measurement of weak Raman spectral features that, in a traditional spatial heterodyne spectrometer measurement, would be washed out by multiplicative photon noise.

Raman spectroscopy is a technique that can detect and characterize a range of molecular compounds such as water, water ice, water-bearing minerals, and organics of particular interest to planetary science. The detection and characterization of these molecular compounds, which are indications of habitability on planetary bodies, have become an important goal for planetary exploration missions spanning the solar system. Using a compact portable remote Raman system consisting of a 532 nm neodymium-doped yttrium aluminum garnet- (Nd:YAG-) pulsed laser, a 3-in. (7.62 cm) diameter mirror lens and a compact spectrograph with a miniature intensified charge coupled device (mini-ICCD), we were able to detect water (H2O), water ice (H2O-ice), CO2-ice, hydrous minerals, organics, nitrates, and an amino acid from a remote distance of 122 m in natural lighting conditions. To the best of our knowledge, this is the longest remote Raman detection using a compact system. The development of this uniquely compact portable remote Raman system is applicable to a range of solar system exploration missions including stationary landers for ocean worlds and lunar exploration, as they provide unambiguous detection of compounds indicative of life as well as resources necessary for further human exploration.

The World Health Organization has declared the COVID-19 outbreak a pandemic. The causative agent for COVID-19 is an RNA virus of the Coronaviridae family. In addition to the respiratory and other complications, a significant proportion of COVID-19 patients show neurological manifestations. Dementia is a neurocognitive disorder, the prevalence of which is projected to increase in the coming decades. This review provides an overview of the effects of COVID-19 on dementia patients.

Amyloid beta is a major constituent of the plaques found in the brains of patients suffering from Alzheimer’s disease (AD). A growing body of research work suggests that neuroinflammation plays important roles in the development of AD. Thus, considerable efforts are directed towards identification of compounds that can reduce or inhibit neuroinflammation. Here, we show that sinomenine, a compound present in a Chinese medicinal plant, Sinomenium acutum, inhibits oligomeric amyloid beta-induced production of reactive oxygen species (ROS), nitric oxide (NO) and inflammation-related molecules from astrocytic cells. The conditioned medium from oligomeric amyloid beta-treated astrocytic cells induces cell death in the hippocampal neuronal cells. Importantly, sinomenine inhibits this cell death. In addition, this compound has inhibitory effects on the production of ROS, NO and inflammation-related factors from oligomeric amyloid-beta treated human astrocytes. Finally, the conditioned medium from oligomeric amyloid beta-treated human astrocytes induces cell death in the primary culture of human neurons, which is inhibited by sinomenine. Thus, sinomenine inhibits amyloid beta-induced production of toxic factors from astrocytes, and confers protection to hippocampal neuronal cells as well as human neurons against indirect toxicity. The results suggest that this compound could provide beneficial effects in AD and other neurodegenerative conditions by reducing inflammation and neuronal cell death.

We report the remote Raman spectra of natural igneous, metamorphic, and sedimentary rock samples at a standoff distance of 5 m. High-quality remote Raman spectra of unprepared rocks are necessary for accurate and realistic analysis of future Raman measurements on planetary surfaces such as Mars. Our results display the ability of a portable compact remote Raman system (CRRS) to effectively detect and isolate various light- and dark-colored mineral phases in natural rocks. The CRRS easily detected plagioclase and potassium feldspar end members, quartz, and calcite in rocks with high fluorescence backgrounds. Intermediate feldspars and quartz, when found in rocks with complex mineralogies, exhibited band shifts and broadening in the ${504{-}510}\,\,{{\rm cm}^{ - 1}}$504−510cm−1 and ${600{-}1200}\,\,{{\rm cm}^{ - 1}}$600−1200cm−1 regions. A good approximation of intermediate plagioclase feldspars was possible by using overall Raman spectral shape and assigning other minor Raman peaks in addition to the $ 504{-}510\,\,{{\rm cm}^{ - 1}}$504−510cm−1 peaks. Detection of olivine and pyroxene in mafic rocks allowed for compositional characterization.

A spatial heterodyne Raman spectrometer (SHRS) is a variant of a Michelson interferometer in which the mirrors of a Michelson are replaced with two stationary diffraction gratings. When light enters the SHRS, it is reflected off of diffraction gratings at frequency-dependent angles that produce crossed wavefronts in space that can be imaged using a plane array detector. The crossed wavefronts, which represent a superposition of interference fringes, are converted to a Raman spectrum upon applying a Fourier transform. In this work, a new approach to intensity calibration is discussed that originates from modeling the shot noise produced by the SHRS and converting the real noise to idealized white noise as predicted by theory. This procedure has two effects. First, the technique produces Raman spectra with white noise. Second, when the mean of the noise is normalized to one, the technique produces Raman spectra where the intensity axis is equivalent to signal-to-noise ratio. The data reduction technique is then applied to the measurement of materials of interest to the planetary science community, including minerals and inorganic salts, at a distance of 5 m from the collecting optic.

Chenodeoxycholic acid (CDCA) is a natural germination inhibitor for C. difficile spores. In our previous study (J. Med. Chem., 2018, 61, 6759–6778), we identified N-phenyl-3α,7α,12α-trihydroxy-5β-cholan-24-amide as an inhibitor of C. difficile strain R20291 with an IC(50) of 1.8 μM. Studies of bile salts on spore germination have shown that chenodeoxycholate, ursodeoxycholate and lithocholate are more potent inhibitors of germination compared to cholate. Given this, we created amide analogs of chenodeoxycholic, deoxycholic, lithocholic and ursodeoxycholic acids using amines identified from our previous studies. We found that chenodeoxy- and deoxycholate derivatives were active with potencies equivalent to those for cholanamides. This indicates that only 2 out of the 3 hydroxyl groups are needed for activity and that the alpha stereochemistry at position 7 is required for inhibition of spore germination.

Many industries are striving for the development of more reliable and robust β-galactosidase biosensors that exhibit high response rate, increased detection limit and enriched useful lifetime. In a newfangled technological atmosphere, a trivial advantage or disadvantage of the developed biosensor may escort to the survival and extinction of the industry. Several alternative strategies to immobilize β-galactosidase enzyme for their utilization in biosensors have been developed in recent years in the quest of maximum utility by controlling the defects seen in the previous biosensors. The overwhelming call for on-line measurement of different sample constituents has directed science and industry to search for best practical solutions and biosensors are witnessed as the best prospect. The main objective of this paper is to serve as a narrow footbridge by comparing the literary works on the β-galactosidase biosensors, critically analyze their use in the construction of best biosensor by showing the pros and cons of the predicted methods for the practical use of biosensors.

Raman spectroscopy is ideally suited for planetary exploration because of its ability to unambiguously identify minerals, organic compounds, and biomarkers. Traditionally, Raman spectra were acquired with grating-based dispersive spectrometers that require tens of micrometer-sized entrance slits and thus limited light throughput. Recently, we have evaluated a new type of Fourier transform Raman spectrometer, the spatial heterodyne Raman spectrometer that provides high spectral resolution in a compact system without limiting light throughput. In this work, we present time-resolved Raman spectra of carbonate, sulfate, and silicate minerals, including low Raman scattering efficiency olivine and feldspar minerals, in the 100–1260 cm−1 Raman fingerprint region with spatial heterodyne Raman spectrometer using 1.5-cm-diameter pulsed 532.078-nm Nd:YAG laser beam. Highly photoluminescent gel-like carbon dots (G-CDs) were successfully synthesized for the first time by a rapid one-step solvothermal synthesis approach with citric acid and 1,2-ethylenediamine as the precursors. Their gel-like nature was revealed by the Tyndall and coagulation effects, which were elucidated by a negative ζ potential value. The influences of temperature on the properties and sizes of these G-CDs were analyzed, and the best method for a maximum quantum yield was identified. The resulting products emitted blue photoluminescence under UV light (λ=365 nm) and a gradient of color under regular light. In addition, the UV/Vis absorption and fluorescence emission spectra of the G-CDs indicated that those synthesized at 160 °C exhibited the highest fluorescence quantum yield (33 %). Atomic force microscopy and transmission electron microscopy measurements were performed, and a higher temperature of formation resulted in smaller G-CDs. Furthermore, band shifts in the UV/Vis and fluorescence spectra and sequential changes in the quantum yield values and ζ potentials in addition to elemental compositional changes as determined by X-ray photoelectron spectroscopy were monitored throughout the formation process of the G-CDs. As to applications, G-CDs were prepared as an invisible ink for printers, which exhibited the applicability of G-CDs in daily life and military activities.

This study investigates the surface chemistry properties of the tyrosinase enzyme Langmuir monolayer at air-aqueous interface using sodium chloride in the subphase to induce the surface activity of the enzyme. Investigation of surface packing and stability of the tyrosinase Langmuir monolayer were performed using surface chemistry experiments while spectroscopic analysis was done to study enzyme conformation. It was found that the tyrosinase enzyme forms a fluid film at air-aqueous interface with good stability as shown by compression-decompression cycles experiments and stability measurements at various surface pressures. UV-vis absorption and fluorescence measurements at different surface pressures revealed that the Langmuir monolayer has good homogeneity with no evidence of aggregates during compression. To gain insight on the conformation of tyrosinase Langmuir monolayer p-polarized infrared-reflection absorption spectroscopy was used. It was found that at high surface pressures the predominant secondary structures were β-sheets while at lower surface pressure both α -helices and β-sheets were present. The circular dichroism spectra were obtained by transferring the Langmuir monolayer at 10 mN.m−1 to a solid quartz support (Langmuir-Blodgett film, LB film), which showed that the major conformation present were α-helices. Images from the immobilized LB films were obtained using atomic force microscopy which showed homogenous and regular deposition with a mean thickness ranging from 3 to 4 nm.

Nanoparticles have been conjugated to biological systems for numerous applications such as self-assembly, sensing, imaging, and therapy. Development of more reliable and robust biosensors that exhibit high response rate, increased detection limit, and enhanced useful lifetime is in high demand. We have developed a sensing platform by the conjugation of &beta, galactosidase, a crucial enzyme, with lab-synthesized gel-like carbon dots (CDs) which have high luminescence, photostability, and easy surface functionalization. We found that the conjugated enzyme exhibited higher stability towards temperature and pH changes in comparison to the native enzyme. This enriched property of the enzyme was distinctly used to develop a stable, reliable, robust biosensor. The detection limit of the biosensor was found to be 2.9 &times, 10&minus, 4 M, whereas its sensitivity was 0.81 &micro, A&middot, mmol&minus, 1&middot, cm&minus, 2. Further, we used the Langmuir monolayer technique to understand the surface properties of the conjugated enzyme. It was found that the conjugate was highly stable at the air/subphase interface which additionally reinforces the suitability of the use of the conjugated enzyme for the biosensing applications.

Down syndrome (DS) is a developmental disorder that results from the trisomy of chromosome 21. DS patients show several abnormalities including cognitive deficits. Here, we show enhanced activation of the extracellular signal-regulated kinase (ERK), a kinase that critically regulates synaptic plasticity and memory, in a hippocampal cell line derived from trisomy 16 mouse foetus. In addition, these cells show enhanced activation of p38 mitogen-activated protein kinase (p38 MAPK). The hyper-activation of ERK and p38 MAPK is significantly reduced by a small peptide, Gly-Pro-Glu (GPE), derived from insulin-like growth factor-1. In addition, the trisomic cells show reduced level of inhibitory phosphorylation of glycogen synthase kinase-3β (GSK-3β), which is enhanced by GPE. Furthermore, the trisomic cells do not show ERK activation in response to KCl depolarization or forskolin treatment. Importantly, ERK activation by these stimuli is observed after GPE treatment of the cells. These results suggest that GPE may help reduce aberrant signalling in the trisomic neurons by affecting MAPK and GSK-3β activation.

Various techniques have been developed to determine protein’s structure to understand how proteins work. Compared with X-ray crystallography requiring proteins to form single crystal structure and NMR which usually needs long time measurement, surface FT-IR techniques are able to quickly determine the tilt angle (the key information to determine whether the α-helix is transmembrane) of peptides/proteins in a monolayer at the interface (e.g. membranes). Specifically, for α-helical peptides/proteins in membrane, the tilt angle of the axis is one of the key information. In this paper, Multiple Angle Incidence Resolution Spectroscopy (MAIRS), a recently developed surface FTIR technique, was applied for the first time to quantitatively determine the tilt angle of the axis of α-helical model peptide related to α-synuclein (α-syn). α-Syn is a 140-amino-acid presynaptic protein whose aggregation is the hallmark of Parkinson’s disease (PD). It is difficult for α-syn to form a single crystal structure and the primary structure of α-syn constitutes three domains: the N-terminus containing residues 1–60; the nonamyloid component (NAC) which spans residues 61–95 and is highly prone to aggregation; and C-terminus with residues 96–140. Here, the NAC part (i.e., α-syn(61–95)) responsible for the aggregation was found to change its unstructured conformation in aqueous solution to α-helix at the air-water interface by circular dichroism and MAIRS. In addition, the instinct power of MAIRS to quantitatively measure the tilt angle of the axis of α-helical α-syn(61–95) in monolayer was fully exhibited. Therefore, MAIRS is a potential supplemental technique to X-ray crystallography and NMR to determine the structure of membrane peptides/proteins.

Chemical analysis using laser-induced breakdown spectroscopy (LIBS) is well suited for field applications and was applied here for shipboard characterization of a large sample set during the RR1310 rock dredging expedition to the Tuvalu Seamounts. Although recently the most common data treatment for LIBS has consisted of a partial least squares approach to define sample groupings, we show that quantitative data of useful quality can be obtained with a univariate approach. Here, our analysis goal was a quantitative comparison of the total alkali (Na2O + K2O) versus silica (SiO2) contents of 586 representative dredge samples with known ranges in common rock types. Out of those samples, >400 form a single large group of alkalic basalts with minor basanites/tephrites (SiO2: 43–48 wt%, Na2O + K2O: 3–5 wt%), similar to known shield-stage compositions of the Rurutu and Samoa hotspots in the sampling area. In contrast, several dredge hauls contain samples with compositions that do not overlap with the majority of samples. This includes three dredges performed on the northern boundary of the Lau Basin that contain similar SiO2 compositions, but slightly higher total alkali (Na2O+K2O) content. Despite this difference, they classify as basanite/tephrite, similar to a subset of the main group. More importantly, similar compositions were previously reported from the same tectonic boundary, ascribed to hotspot mantle source material mixed into the Lau Basin back-arc. Although the quality of the compositional data suffices to enable sample selection for time-intensive analyses, higher precision is required for more in-depth petrogenetic interpretation. Error analysis based on repeat standard measurements suggests averaging 100 spectra per sample is optimal here, while use of a higher resolution spectrometer, together with better laser control, would improve results and interpretations.

Fumonisin B1 (FB1), the most abundant component of the fumonisin family, is highly responsible for fungal infections. In this paper, our main aim is to study the surface chemistry and spectroscopic properties of the FB1 molecule and observe the impact of green LED light on the FB1 Langmuir monolayer. From the surface chemistry and spectroscopic studies, we found that the FB1 molecule forms a self-assembled Langmuir monolayer which is sufficient to mimic its interaction with the corneal tissues. The irradiation of green LED light on the FB1 Langmuir monolayer showed the degradation of the FB1 when compared to that in the absence of light. This observation reveals that FB1 molecules lose their tendency to stay as a Langmuir monolayer. The degradation observed on the interface was compared with the bulk phase of FB1. The bulk phase observation also indicated the degradation tendency which reinforced the observed interfacial property of FB1.
