Tiller number and grain size are important agronomic traits that determine grain yield in rice. Here, we demonstrate that DEFECTIVE TILLER GROWTH 1 (DTG1), a member of the casein kinase 1 protein family, exerts a co-regulatory effect on tiller number and grain size. We identified a single amino acid substitution in DTG1 (I357K) that caused a decrease in tiller number and an increase in grain size in NIL-dtg1. Genetic analyses revealed that DTG1 plays a pivotal role in regulation of tillering and grain size. The DTG1I357K allelic variant exhibited robust functionality in suppressing tillering. We show that DTG1 is preferentially expressed in tiller buds and young panicles, and negatively regulates grain size by restricting cell proliferation in spikelet hulls. We further confirm that DTG1 functioned in grain size regulation by directly interacting with Grain Width 2 (GW2), a critical grain size regulator in rice. The CRISPR/Cas9-mediated elimination of DTG1 significantly enhanced tiller number and grain size, thereby increasing rice grain yield under field conditions, thus highlighting potential value of DTG1 in rice breeding.

The utilization of heavy-panicle hybrid rice exemplifies the successful integration of architectural enhancement and heterosis, which has been widely adopted in the southwest rice-producing area of China. Iterative improvement in disease resistance and grain quality of heavy-panicle hybrid rice varieties is crucial to promote their sustainable utilization. Here, we performed a molecular design breeding strategy to introgress beneficial alleles of broad-spectrum disease resistance and grain quality into a heavy-panicle hybrid backbone restorer line Shuhui 600 (R600). We successfully developed introgression lines through marker-assisted selection to pyramid major genes (Wxb + ALKA-GC + Pigm + Xa23) derived from three parents (Huanghuazhan, I135, I488), which significantly enhance grain quality and confer resistance to rice blast and bacterial blight (BB). The improved parental R600 line (iR600) exhibited superior grain quality and elevated disease resistance while maintaining the heavy-panicle architecture and high-yield capacity of R600. Moreover, the iR600 was crossed with male sterility line 608A to obtain a new heavy-panicle hybrid rice variety with excellent eating and cooking quality (ECQ) and high yield potential. This study presents an effective breeding strategy for rice breeders to expedite the improvement of grain quality and disease resistance in heavy-panicle hybrid rice.

Calcium sulphoaluminate cement (CSA) is considered a sustainable alternative to traditional Portland cement to reduce CO2 emissions. To further improve the hardening properties and water resistance of CSA and advance its extensive special engineering application, nanoparticles including nano-SiO2 (NS), nano-MgO (NM) and nano-Al2O3 (NA) were used as nano-modifiers for CSA. The effects of nanoparticles dosage on the setting time, compressive strength and water resistance of nano-modified CSA were experimentally evaluated, and the micro-mechanisms were also clarified by conducting X-ray diffraction (XRD), scanning electron microscopy (SEM) and thermogravimetric analysis (TG) tests. The results showed that incorporating nanoparticles into CSA obviously shortened the initial and final setting times. The appropriate addition of nanoparticles effectively enhanced the compressive strength of CSA, and the optimum dosages of NS, NM, and NA were respectively determined to be 0.5 %, 1.0 %, and 0.5 %. Water immersion decreased the bonding strength of CSA, and adding nanoparticles can effectively weaken this water-induced deterioration effect. Under water immersion environment, the optimum dosages of nanoparticles would be increased to obtain the highest residual water immersion strength and minimum strength loss rate. The generation of ettringite was mainly responsible for the compressive strength and water resistance improvements of nano-modified CSA. The hydration process of nano-modified CSA was clarified and the corresponding micro-mechanism model was proposed. Furthermore, the benefits and challenges of using nano-modified CSA were also analyzed, advancing its widespread application in rapid repair of special engineering.

The state of health (SOH) and remaining useful life (RUL) of lithium-ion batteries are crucial for health management and diagnosis. However, most data-driven estimation methods heavily rely on scarce labeled data, while traditional transfer learning faces challenges in handling domain shifts across various battery types. This paper proposes an enhanced vision-transformer integrating with semi-supervised transfer learning for SOH and RUL estimation of lithium-ion batteries. A depth-wise separable convolutional vision-transformer is developed to extract local aging details with depth-wise convolutions and establishes global dependencies between aging information using multi-head attention. Maximum mean discrepancy is employed to initially reduce the distribution difference between the source and target domains, providing a superior starting point for fine-tuning the target domain model. Subsequently, the abundant aging data of the same type as the target battery are labeled through semi-supervised learning, compensating for the source model's limitations in capturing target battery aging characteristics. Consistency regularization incorporates the cross-entropy between predictions with and without adversarial perturbations into the gradient backpropagation of the overall model. In particular, across the experimental groups 13–15 for different types of batteries, the root mean square error of SOH estimation was less than 0.66 %, and the mean relative error of RUL estimation was 3.86 %. Leveraging extensive unlabeled aging data, the proposed method could achieve accurate estimation of SOH and RUL.

Objective: The present study aims to evaluate the impact of Astilbin (AST) on cortical neuron survival in vitro under conditions of oxygen-glucose deprivation and reoxygenation (OGD/R) and determine the role of NF-E2-related factor 2 (Nrf2) in this process. Methods: Primary neurons were pre-treated with various concentrations of AST for 8 h before OGD induction. Cell viability and lactate dehydrogenase (LDH) leakage were assessed to determine the optimal concentration. Biomarkers related to oxidative stress, antioxidant enzyme activities, and apoptosis were evaluated at 24 h post-OGD/R. To investigate the involvement of Nrf2 in AST-mediated neuroprotection, we conducted molecular docking and microscale thermophoresis analyses, as well as examined the expression levels of Nrf2 and its regulatory genes including heme oxygenase-1(HO-1), (NAD(P)H: quinone oxidoreductase 1 (NQO-1), and peroxiredoxin 1 (Prdx1). Additionally, lentivirus-mediated knockdown of Nrf2 and overexpression of Nrf2 with L-sulforaphane (SFN) were performed, followed by an assessment of cell viability, oxidative stress, antioxidant enzyme activities and apoptosis. Results: Pre-treatment with AST reduced oxidative stress levels while increasing antioxidant enzyme activities and mitigating neuronal apoptosis. After OGD/R exposure, AST upregulated nuclear Nrf2 expression and increased the expression of HO-1, NQO-1 and Prdx1 in the cytoplasm. However, the knockdown of Nrf2 abolished the antioxidative and protective effects exerted by AST treatment. Conversely, combining AST with the Nrf2 agonist SFN demonstrated an enhancement in the protective effects provided by AST. These results demonstrate that Nrf2-dependent antioxidant responses contribute to AST-induced tolerance against neuronal injury caused by OGD/R injury. Conclusions: Overall findings support the ability of AST to protect primary neurons from OGD/R-induced damage through activation of Nrf2-dependent antioxidant responses.

Abstract To reveal the mechanism of the influence of the curing temperature on the strength of lime activated fly ash-GGBS cured silt soil, the curing of dredged silt was carried out by using fly ash and GGBS as the curing agent and lime as the activator. Unconfined compressive strength (UCS) experiments were carried out, and the micro-analysis of the cured silt was carried out by experimental methods including scanning electron microscope (SEM) tests, X-ray diffraction (XRD), etc. to reveal the mechanism of the curing temperature on the dredged silt. According to the test results, the hydration reaction and pozzolanic reaction between lime-fly ash-GGBS and silt soil were promoted with the increase of the curing temperature. when the curing temperature of the sample reached 40 ℃, a large amount of gel products such as hydrated calcium aluminate (C–A–H) and hydrated calcium silicate (C–S–H) were generated, which enhanced the bonding force between soil particles and filled up the inter-particle pore space, thereby improving the UCS of the sample. The results of SEM confirmed that C–A–H and C–S–H were the main substances for the construction of cured silt skeleton. C–S–H and C–A–H were detected by XRD. The results of the study fill the gap in the effect of curing temperature on the direction of lime-activated fly ash-GGBS cured silt soil.

Glycophagy has evolved from an alternative glycogen degradation pathway into a multifaceted pivot to regulate cellular metabolic hemostasis in peripheral tissues. However, the pattern of glycophagy in the brain and its potential therapeutic impact on ischemic stroke remain unknown. Here, we observed that the dysfunction of astrocytic glycophagy was caused by the downregulation of the GABA type A receptor-associated protein like 1 (GABARAPL1) during reperfusion in ischemic stroke patients and mice. PI3K-Akt pathway activation is involved in driving GABARAPL1 downregulation during cerebral reperfusion. Moreover, glycophagy dysfunction-induced glucosamine deficiency suppresses the nuclear translocation of specificity protein 1 and TATA binding protein, the transcription factors for GABARAPL1, by decreasing their O-GlcNAcylation levels, and accordingly feedback inhibits GABARAPL1 in astrocytes during reperfusion. Restoring astrocytic glycophagy by overexpressing GABARAPL1 decreases DNA damage and oxidative injury in astrocytes and improves the survival of surrounding neurons during reperfusion. In addition, a hypocaloric diet in the acute phase after cerebral reperfusion can enhance astrocytic glycophagic flux and accelerate neurological recovery. In summary, glycophagy in the brain links autophagy, metabolism, and epigenetics together, and glycophagy dysfunction exacerbates reperfusion injury after ischemic stroke.

Abstract Background Lipid metabolism has a crucial role in neural repair in neurodegenerative diseases. We recently revealed that lipogenesis-mediated interleukin-33 (IL-33) upregulation lead to blood–brain barrier (BBB) repair after ischemic stroke. However, manipulating the key enzyme fatty acid synthase (FASN) to enhance lipogenesis was very challenging. Glyceryl triacetate (GTA) was used as a donor of acetate and precursor of acetyl coenzyme A, the key substrate for de novo lipogenesis catalyzed by FASN. Therefore, we hypothesized that GTA would promote lipogenesis the peri-infarct after ischemic stroke and contribute to the BBB repair through IL-33. Methods Middle cerebral artery occlusion (MCAO) was performed on C57BL mice and GTA was gavage administrated (4 g/kg) on day 2 and 4 after MCAO. Lipogenesis was evaluated by assessment of the protein level of FASN, lipid droplets, and fatty acid products through liquid chromatography-mass spectrometry in the peri-infarct area on day 3 after MCAO, respectively. BBB permeability was determined by extravasation of Evans blue, IgG and dextran, and levels of tight junction proteins in the peri-infarct area on day 7 after MCAO, respectively. Infarct size and neurological defects were assessed on day 7 after MCAO. Brain atrophy on day 30 and long-term sensorimotor abilities after MCAO were analyzed as well. The inhibitor of FASN, C75 and the virus-delivered FASN shRNA were used to evaluate the role of FASN-driven lipogenesis in GTA-improved BBB repair. Finally, the therapeutic potential of recombinant IL-33 on BBB repair and neurological recovery was evaluated. Results We found that treatment with GTA increased the lipogenesis as evidenced by lipid droplets level and lauric acid content, but not the FASN protein level. Treatment with GTA increased the IL-33 level in the peri-infarct area and decreased the BBB permeability after MCAO. However, infarct size and neurological defect score were unchanged on day 7 after MCAO, while the long-term recovery of sensorimotor function and brain atrophy were improved by GTA. Inhibition of lipogenesis using C75 or FASN shRNA reversed the beneficial effect of GTA. Finally, exogenous IL-33 improved BBB repair and long-term functional recovery after stroke. Conclusion Collectively, we concluded that treatment with GTA improved the BBB repair and functional recovery after ischemic stroke, probably by the enhancement of lipogenesis and IL-33 expression. Cement-based solidification is a widely used sustainable technology for transforming waste dredged sludge (DS) into backfill materials. This study developed a novel solidification/oxidization synergistic (SOS) method, using Portland cement as a binder and sodium persulfate (SP) as an oxidant, for treating the organic DS with different fulvic acid (FA) content. The results demonstrated that adding SP could significantly improve the UCS of the cement-solidified DS doped with FA. When the FA content was 4%, the 28-day UCS of the sample with 25% cement content was only 160.5 kPa. The addition of 6% SP increased the UCS to 2225.5 kPa, which was 15.1 times that of the sample without SP. However, excessive SP hydrolyzed to produce a large amount of sulfuric acid, leading to a low pH value of the pore solution. This in turn hindered further persulfate activation and cement hydration, which was detrimental to the strength of cement-solidified DS.

Shank3, a key molecule related to the development and deterioration of autism, has recently been found to downregulate in the murine brain after ischemia/reperfusion (I/R). Despite this discovery, however, its effects on neuronal injury and the mechanism underlying the effects remain to be clarified. To address this, in this study, based on genetically modified mice models, we revealed that the expression of Shank3 showed a time-dependent change in murine hippocampal neurons after I/R, and that conditional knockout (cko) of Shank3 in neurons resulted in aggravated neuronal injuries. The protective effects of Shank3 against oxidative stress and inflammation after I/R were achieved through direct binding STIM1 and subsequent proteasome-mediated degradation of STIM1. The STIM1 downregulation induced the phosphorylation of downstream Nrf2 Ser40, which subsequently translocated to the nucleus, and further increased the expression of antioxidant genes such as NQO1 and HO-1 in HT22 cells. In vivo, the study has further confirmed that double knockout of Shank3 and Stim1 alleviated oxidative stress and inflammation after I/R in Shank3cko mice. In conclusion, the present study has demonstrated that Shank3 interacts with STIM1 and inhibits post-I/R neuronal oxidative stress and inflammatory response via the Nrf2 pathway. This interaction can potentially contribute to the development of a promising method for I/R treatment.

A composite material was developed using carbide slag, water glass, slag, and micron silicon to facilitate the use of industrial waste resources. The mechanical properties of dredge sludge (DS) were analyzed, considering different proportions of cement, organic debris, and carbide slag. The composition and microstructure of the hydration products were analyzed using the X-ray diffractometer (XRD), scanning electron microscopy (SEM), and thermogravimetric (TG) analysis. The results indicate that with a precursor content of 20%, a water glass content of 3%, and an increase in carbide slag content from 4% to 12%, the strength of the sample initially increases and subsequently drops at each age. With a carbide slag level of 8%, the combination of CaO in the slag and water glass stimulated the slag and micron silica, leading to the formation of gel substances such C-S-H and C-A-S-H. The soil particles exhibited increased density as a result of the cohesive properties of the gel products. Following a maintenance period of 28 days, the sample's compressive strength rose to 2280 kPa. When the carbide slag level exceeds 8%, the presence of Ca(OH)2 in the mixture leads to the formation of carbonates, such as calcite, during the carbonization process. The organic matter subsequently undergoes a reaction with the Ca(OH)2 produced during the hydration of the mixture, leading to the formation of a highly soluble complex. As a result, only a limited quantity of calcium ions in the pore solution participate in the pozzolanic reaction, hence reducing the formation of gel reaction products such C-S-H.

The rapid development of modern communication systems propels the demand of high performance and miniaturization of RF/microwave components. Power dividers and filters are indispensable circuit components in amplifiers and antenna feed-networks which occupy large footprint of circuit broads, particularly at low frequency. The purpose of this study is to initiate a new type of planar transmission line with unique dual signal-path characteristics and ultimately achieve circuit miniaturization. In this paper, a novel technique of consolidating microstrip lines (MLs) and coplanar waveguide (CPW) lines on a single dielectric substrate, designated as the composite microstrip/CPW line (CM/CPW line), is proposed. With reference to the average line length of ML and CPW, it warrants a physical length reduction of 48.3% and further achieves a reduction of 80.8% after zigzagging CM/CPW line. The proof-of-concept design example of a two-way Wilkinson power divider (WPD) has demonstrated the effectiveness of the miniaturization technique. The simulation, in combination with the experimental results, validates that a size reduction factor of 86.9% when compared to conventional one has been achieved while maintaining a fractional bandwidth of 57.7% for the WPD.

Abstract In this study, a novel frequency beam‐scanning (FBS) antenna based on composite substrate integrated waveguide/spoof surface plasmon polariton (SIW/SSPP) waveguide is proposed. The composite SIW/SSPP slow‐wave waveguide is designed by embedding a series of periodic subwavelength H‐shaped grooves on the top surface of the SIW. Its transmission characteristics own both the features from SIW and SSPP, and can be flexibly mansplained by tuning the key structural parameters. To convert the propagating composite slow‐wave mode into the radiation mode effectively, a new flexible digital modulation method with staggered metallic vias is proposed and used. The presented beam‐scanning antenna has a simple feeding structure, a single dielectric layer, and a compact volume of 0.75 × 8.55 × 0.015 λ03. A prototype of the antenna is fabricated and tested to verify its performance. The experimental results demonstrate that the proposed antenna can realise a continuous beam‐scanning angle of 49° in an operating bandwidth of 8.7–9.2 GHz with a high frequency‐scanning sensitivity (FSS) of 9.8 °/100 MHz−1. Moreover, the measured results indicate that the antenna has high peak gain with an average level of 12.7 dBi, high radiation efficiency with an average level of 87%, and stable unidirectional radiation pattern in the working band.

Abstract Background Astrocytic glycogen works as an essential energy reserve for surrounding neurons and is reported to accumulate excessively during cerebral ischemia/reperfusion (I/R) injury. Our previous study found that accumulated glycogen mobilization exhibits a neuroprotective effect against I/R damage. In addition, ischemia could transform astrocytes into A1-like (toxic) and A2-like (protective) subtypes. However, the underlying mechanism behind accumulated glycogen mobilization-mediated neuroprotection in cerebral reperfusion injury and its relationship with the astrocytic A1/A2 paradigm is unknown. Methods Astrocytic glycogen phosphorylase, the rate-limiting enzyme in glycogen mobilization, was specifically overexpressed and knocked down in mice and in cultured astrocytes. The I/R injury was imitated using a middle cerebral artery occlusion/reperfusion model in mice and an oxygen–glucose deprivation/reoxygenation model in cultured cells. Alterations in A1-like and A2-like astrocytes and the expression of phosphorylated nuclear transcription factor-κB (NF-κB) and phosphorylated signal transducer and activator of transcription 3 (STAT3) were determined by RNA sequencing, immunofluorescence and immunoblotting. Metabolites, including glycogen, NADPH, glutathione and reactive oxygen species (ROS), were analyzed by biochemical analysis. Results Here, we observed that astrocytic glycogen mobilization inhibited A1-like astrocytes and enhanced A2-like astrocytes after reperfusion in an experimental ischemic stroke model in vivo and in vitro. In addition, glycogen mobilization could enhance the production of NADPH and glutathione by the pentose phosphate pathway (PPP) and reduce ROS levels during reperfusion. NF-κB inhibition and STAT3 activation caused by a decrease in ROS levels were responsible for glycogen mobilization-induced A1-like and A2-like astrocyte transformation after I/R. The astrocytic A1/A2 paradigm is closely correlated with glycogen mobilization-mediated neuroprotection in cerebral reperfusion injury. Conclusions Our data suggest that ROS-mediated NF-κB inhibition and STAT3 activation are the key pathways for glycogen mobilization-induced neuroprotection and provide a promising metabolic target for brain reperfusion injury in ischemic stroke.

Abstract Background Sudden cardiac death (SCD) remains a great health threat and diagnostic challenge, especially those cases without positive autopsy findings. Molecular biomarkers have been urgently needed for the diagnosis of SCD displaying negative autopsy results. Due to their nature of stability, microRNAs (miRNAs) have emerged as promising diagnostic biomarkers for cardiovascular diseases. Methods This study investigated whether specific cardio-miRNAs (miR-3113-5p, miR-223-3p, miR-499a-5p, and miR-133a-3p) could serve as potential biomarkers for the diagnosis of SCD. Thirty-four SCD cases were selected, 18 categorized as SCD with negative autopsy (SCD-negative autopsy) findings and 16 as SCD with positive autopsy (SCD-positive autopsy) findings such as coronary atherosclerosis and gross myocardial scar. Carbon monoxide (CO) intoxication (n = 14) and fatal injury death (n = 14) that displayed no pathological changes of myocardium were selected as control group, respectively. Histological analyses were performed to reveal the pathological changes and real-time quantitative polymerase chain reaction (RT-qPCR) was used to determine the expression of those miRNAs. Results It showed that heart samples from the SCD-negative autopsy group displayed no remarkable difference with regard to the expression of cleaved-caspase3, CD31, and CD68 and the extent of fibrotic tissue accumulation when compared with control samples. The four cardio-miRNAs were significantly up-regulated in the SCD samples as compared with control. When discriminating SCD from controls, receiver operating characteristic (ROC) curve analysis revealed that the areas under the curve (AUC) of these 4 miRNAs were from 0.7839 to 0.9043 with sensitivity of 64.71–97.06% and specificity of 70–100%. Moreover, when discriminating the specific causes of SCD, the four miRNA expressions increased in the heart from the SCD-negative autopsy group as relative to that from the SCD-positive autopsy group, and a combination of two miRNAs presented higher diagnostic value (AUC = 0.7407–0.8667). Conclusion miR-3113-5p, miR-223-3p, miR-499a-5p, and miR-133a-3p may serve as independent diagnostic biomarkers for SCD, and a combination of two of these miRNAs could further discriminate detailed causes of SCD.

Abstract Background Rice (Oryza sativa) bacterial leaf blight (BLB), caused by the hemibiotrophic Xanthomonas oryzae pv. oryzae (Xoo), is one of the most devastating diseases affecting the production of rice worldwide. The development and use of resistant rice varieties or genes is currently the most effective strategy to control BLB. Results Here, we used 259 rice accessions, which are genotyped with 2 888 332 high-confidence single nucleotide polymorphisms (SNPs). Combining resistance variation data of 259 rice lines for two Xoo races observed in 2 years, we conducted a genome-wide association study (GWAS) to identify quantitative trait loci (QTL) conferring plant resistance against BLB. The expression levels of genes, which contains in GWAS results were also identified between the resistant and susceptible rice lines by transcriptome analysis at four time points after pathogen inoculation. From that 109 candidate resistance genes showing significant differential expression between resistant and susceptible rice lines were uncovered. Furthermore, the haplotype block structure analysis predicted 58 candidate genes for BLB resistance based on Chr. 7_707158 with a minimum P-value (–log 10 P = 9.72). Among them, two NLR protein-encoding genes, LOC_Os07g02560 and LOC_Os07g02570, exhibited significantly high expression in the resistant line, but had low expression in the susceptible line of rice. Conclusions Together, our results reveal novel BLB resistance gene resources, and provide important genetic basis for BLB resistance breeding of rice crops.

Background: Stroke is a major health concern and a leading cause of mortality and morbidity. We and other groups have documented that hyperbaric oxygen preconditioning could significantly alleviate neuronal damage in ischemia‒reperfusion models through various mechanisms. However, we found that some of the subjects did not benefit from preconditioning with hyperbaric oxygen. The preconditioning phenomenon is similar to vaccination, in which the endogenous survival system is activated to fight against further injuries. However, with vaccine inoculations, we could test for specific antibodies against the pathogens to determine if the vaccination was successful. Likewise, this experiment was carried out to explore a biomarker that can reveal the effectiveness of the preconditioning before neuronal injury occurs. Methods: Middle cerebral artery occlusion (MCAO) was used to induce focal cerebral ischemia-reperfusion injury. 2D-DIGE-MALDI-TOF-MS/MS proteomic technique was employed to screen the differentially expressed proteins in the serum of rats among the control (Con) group (MCAO model without hyperbaric oxygen (HBO) preconditioning), hyperbaric oxygen protective (HBOP) group (in which the infarct volume decreased after HBO preconditioning vs. Con), and hyperbaric oxygen nonprotective (HBOU) group (in which the infarct volume remained the same or even larger after HBO preconditioning vs. Con). Candidate biomarkers were confirmed by western blot and enzyme linked immunosorbent assay (ELISA), and the relationship between the biomarkers and the prognosis of cerebral injury was further validated. Results: Among the 15 differentially expressed protein spots detected in the HBOP group by Two-dimensional fluorescence difference gel electrophoresis (2D-DIGE), 3 spots corresponding to 3 different proteins (haptoglobin, serum albumin, and haemopexin) products were identified by MALDI-TOF-MS/MS. Serum albumin and haemopexin were upregulated, and haptoglobin was downregulated in the HBOP group (p < 0.05 vs. Con and HBOU groups). After the western blot study, only the changes in haemopexin were validated and exhibited similar changes in subjects from the HBOP group in accordance with MALDI-TOF-MS/MS proteomic analysis and enzyme linked immunosorbent assay (ELISA) analysis. The serum level of the hemopexin (HPX) at 2 h after HBO preconditioning was correlated with the infarct volume ratio after MCAO. Conclusions: Haemopexin may be developed as a predictive biomarker that indicated the effectiveness of a preconditioning strategy against cerebral ischaemic injury.

Schizophrenia is a severe mental disorder that is often comorbid with heart dysfunction and even sudden cardiac death (SCD). Clinical studies of SCD in schizophrenia have been largely reported, while there are limited autopsy studies that directly showed whole-scale information of such events. In this study, we present nine autopsy-based SCD cases in schizophrenia patients who died suddenly during hospitalization. Their medical records before and during hospitalization, and postmortem autopsy findings were summarized. These decedents had an average duration of schizophrenia for 6.83 ± 3.75 years with a male/female ratio of 4:5. They were all on intermittent antipsychotics medication before hospitalization and died within 15 days after hospitalization. Seven of the nine cases (77.8%) died of organic heart diseases such as severe coronary artery atherosclerosis (n = 4), myocarditis (n = 1), cardiomyopathy (n = 1), and pulmonary thromboembolism (n = 1). Two cases remained unexplained after systemic autopsy and toxicological examinations. Postmortem autopsy identified hepatic steatosis (n = 6) and respiratory inflammation (n = 3) as the most common associate extra-cardiac lesions. Our data provided autopsy-based data of SCD cases in schizophrenia and highlighted an intensive care of such patients during hospitalization.

Pollen fertility is an agronomic trait that strongly influences rice yield. Recent studies have revealed that the development of the pollen wall is required for pollen fertility and is regulated by several genes. However, the mechanisms underlying pollen and pollen wall development in rice remain largely unknown. In the present study, a point mutation in a gene on chromosome 1 was identified that resulted in the production of less and shrunken pollen (LSP) and led to defects in pollen wall formation. This gene was named LSP1 and was found to encode a member of the adenosine triphosphate-binding cassette (ABC) transporter G subfamily, OsABCG3. Two other loss-of-function mutants of LSP1/OsABCG3, generated using CRISPR/Cas9 technology, showed the same male sterile phenotype. The LSP1/OsABCG3 gene showed a spatio-temporal expression pattern in the developing anthers, and is an ortholog of the Arabidopsis genes AtABCG1 and AtABCG16, which play an important role in pollen wall development. Mutation of LSP1/OsABCG3 affected the expression of several genes involved in pollen and pollen wall formation. These results suggest that LSP1/OsABCG3 is critical for normal pollen fertility and shed light on the molecular mechanisms underlying rice pollen wall development.

In this manuscript, we reconsider the amplitude and phase combination of the plane-wave generator (PWG) from an electromagnetic field inverse radiation perspective and further propose a new method based on the Gauss-Newton inversion (GNI) to perform the inverse design of PWG. Furthermore, the wideband antenna pattern is applied in the transfer function and makes the inversion results closer to the engineering implementation. Compared with the traditional least-squares method (LSM) and singular value decomposition (SVD) method, the proposed method owns the merits of less amplitude range of the input source and stronger robustness.

In this study, a triple-band microstrip antenna with compact size and simple structure is proposed for WLAN and 5G applications. The radiating element consists of a circular patch, a Y-shaped patch, and a monopole impedance converter. In order to obtain the desired operating bands, the monopole impedance converter is inserted between the circular patch and Y-shaped patch. The proposed antenna can work in the frequency range of 2.38∼2.53 GHz, 3.29∼4.11 GHz, and 4.72∼5.01 GHz, with the corresponding peak gains of 4.09 dBi (2.4 GHz), 2.95 dBi (3.5 GHz), and 4.01 dBi (4.9 GHz), respectively. The measures’ results are in approximate agreement with the simulated values, which shows that the proposed compact antenna can offer omnidirectional radiation, appropriate gains, and sufficient bandwidths.

Hot dry rock (HDR) geothermal energy has many advantages, such as being renewable, clean, widely distributed, and without time and weather limitations. Hydraulic fracturing is usually needed for the exploitation of HDR geothermal energy. It has many hidden faults in the reservoir/caprock sequences. Injecting fluid into underground formations during hydraulic fracturing often induces fault slip and leads to earthquakes. Therefore, to well understand the induced fault slip and earthquakes is important for the applications and development of HDR geothermal exploitation. In this study, we investigated the hazardous injection area of the induced earthquakes during hydraulic fracturing. The study was based on a hydraulic fracturing test in Qiabuqia geothermal field in China. According to the field, a fault-surrounding rock-fracturing region system was developed to study the influences of fluid injection on the stability of the specific fault. A total of 60 hydraulic fracturing regions and 180 numerical experiments were designed. The results revealed that the hazardous injection regions that threaten the fault’s stability were near to the fault and concentrated on the following four areas: (a) above the top of the fault in underlying strata; (b) above the top of the hanging wall of the fault in underlying strata; (c) near to the fault planes in both footwall and hanging wall; (d) at the bottom of the footwall of the fault in underlying strata. The hazardous injection area can be controlled effectively by adjusting the injection pressure.

Hot dry rock (HDR) geothermal energy has become promising resources for relieving the energy crisis and global warming. The exploitation of HDR geothermal energy usually needs an enhanced geothermal system (EGS) with artificial fracture networks by hydraulic fracturing. Fault reactivation and seismicity induced by hydraulic fracturing raise a great challenge. In this paper, we investigated the characteristics of fault slip and seismicity by numerical simulation. The study was based on a hydraulic fracturing project in the geothermal field of Yishu fault zone in China. It revealed that fluid injection during hydraulic fracturing can cause the faults that exist beyond the fluid-pressurized region to slip and can even induce large seismic event. It was easier to cause felt earthquakes when hydraulic fracturing was carried out in different layers simultaneously. We also examined the effects of the location, permeability, and area of the fracturing region on fault slip and magnitude of the resulting events. The results of the study can provide some useful references for establishing HDR EGS in Yishu fault zone.

Postmenopausal women have a higher incidence of stroke compared to the age-matched males, and the estrogen was thought to be the main cause of such difference. However, estrogen replacement therapy for the prevention of postmenopausal stroke shows controversial results and is widely disputed because of its serious side effects after chronic administration. Genistein (Gen), a natural phytestrogen with fewer side effects, has a protective effect against cerebral ischemia damage. However, whether Gen could effectively prevent postmenopausal stroke has not been elucidated. In the current study, reproductively senescent mice were treated with Gen (10 mg/kg) for 2 weeks before having transient cerebral ischemia insults. Neurological scores, infarct volumes, and cell apoptosis were evaluated 24 h after reperfusion. The levels of inflammatory factors and nod-like receptor protein 3 (NLRP3) inflammasome-related proteins were also examined. The results showed that Gen treatment reduced infarct volumes, improved neurological scores, attenuated apoptosis, and decreased inflammatory factor release. The expression of NLRP3 inflammasome-related proteins in microglia was downregulated by Gen. However, the overexpression of NLRP3 in microglia abrogated the Gen-induced inhibition of inflammatory factor release and reversed the neuroprotective effect of Gen. Taken together, the results suggest that Gen treatment could attenuate the acute injury induced by cerebral ischemia in reproductively senescent mice via the inhibition of the NLRP3 inflammasome in microglia, indicating that Gen could be a candidate drug for the treatment of stroke in postmenopausal women.

Summary: Astrocytic glycogen is an important energy reserve in the brain and is believed to supply fuel during energy crisis. However, the pattern of glycogen metabolism in ischemic stroke and its potential therapeutic impact on neurological outcomes are still unknown. Here, we found extensive brain glycogen accumulation after reperfusion in ischemic stroke patients and primates. Glycogenolytic dysfunction in astrocytes is responsible for glycogen accumulation, caused by inactivation of the protein kinase A (PKA)-glycogen phosphorylase kinase (PhK)-glycogen phosphorylase (GP) cascade accompanied by the activation of glycogen synthase kinase-3β (GSK3β). Genetic or pharmacological augmentation of astrocytic GP could promote astrocyte and neuron survival and improve neurological behaviors. In addition, we found that insulin exerted a neuroprotective effect, at least in part by rescuing the PKA-PhK-GP cascade to maintain homeostasis of glycogen metabolism during reperfusion. Together, our findings suggest a promising intervention for undesirable outcomes in ischemic stroke.

Abstract Background Sevoflurane preconditioning induces brain ischemic tolerance, but the mechanism remains poorly elucidated. Nitration is an important form of post-translational modification in pathological signaling. This study was to investigate the role of thioredoxin-1 (Trx-1) nitration in neuroprotection effect induced by sevoflurane preconditioning in a transient stroke model in rats. Methods Adult male Sprague–Dawley rats were preconditioned with 2% sevoflurane or vehicle oxygen exposure, 1 h per day, for 5 consecutive days. At 24 h after the last exposure, rats were subjected to focal brain ischemia induced by middle cerebral artery occlusion (MCAO) for 90 min, followed by 72-h reperfusion. Trx-1 expression and activity, as well as the content of nitrotyrosine at penumbra were detected at 24 h after preconditioning and 2, 8, 24, 72 h after MCAO. Nitrated Trx-1 was examined by immunoprecipitation at 8 h after MCAO. The role of Trx-1 nitration in ischemic tolerance was assessed by administration of nitrated human-Trx-1 prior to MCAO. Neurological scores, brain infarct volumes and TUNEL staining were evaluated at 24 h after reperfusion. Results Ischemic stroke decreased Trx-1 activity but not the expression in penumbra tissue. The content of nitrotyrosine was elevated after MCAO. Preconditioning with sevoflurane increased Trx-1 activity and reduced its nitration at 8 h after MCAO in comparison with vehicle preconditioning. The decrement of Trx-1 activity was correlated with its nitration level. Exogenous administration of nitrated human-Trx-1 reversed the brain ischemic tolerance of sevoflurane preconditioning, exacerbating brain infarct volume, neurobehavioral defects and apoptosis, while administration of human-Trx-1 had no effect on the sevoflurane preconditioning-induced neuroprotection. Conclusion Ischemic stroke reduces Trx-1 activity via post-translational nitrative modulation in rats. Sevoflurane preconditioning induces brain ischemic tolerance and anti-apoptosis by partially preserving Trx-1 activity via inhibiting nitration.

Abstract Background The steroid hormone estrogen (17-β-estradiol, E2) provides neuroprotection against cerebral ischemic injury by activating estrogen receptors. The novel estrogen receptor G protein-coupled receptor 30 (GPR30) is highly expressed in the brain and provides acute neuroprotection against stroke. However, the underlying mechanisms remain unclear. Methods In this study, ovariectomized female mice were subjected to middle cerebral artery occlusion (MCAO), and E2, G1, and ICI182780 were administered immediately upon reperfusion. The infarction volume, neurological scores, and neuronal injuries were examined. Primary microglial cells were subjected to oxygen-glucose deprivation (OGD), and the drugs were administered immediately upon reintroduction. The pro-inflammatory cytokines TNF-α, IL-1β, and IL-6 in penumbra and microglia were assessed by ELISA. The cell viability and lactose dehydrogenase (LDH) release of neurons co-cultured with microglia were analyzed using cell counting kit-8 (CCK8) and LDH release assays. Microglial activation as well as GPR30, Iba1, and Toll-like receptor 4 (TLR4) protein expression and TLR4 mRNA expression were detected. Additionally, NF-κB activity was detected in lipopolysaccharide (LPS)-activated microglia after the activation of GPR30. Results GPR30 was highly expressed in microglia and significantly increased after ischemic injury. The activation of GPR30 significantly reduced the infarction volume, improved the neurological deficit, and alleviated neuronal injuries. Moreover, GPR30 activation significantly reduced the release of TNF-α, IL-1β, and IL-6 from ischemic penumbra and microglia subjected to OGD and alleviated neuronal injury as assessed using the CCK8 and LDH assays. Finally, the activation of GPR30 relieved microglial activation, reduced Iba1 and TLR4 protein expression and TLR4 mRNA levels, and inhibited NF-κB activity. Conclusions Microglial GPR30 exerts acute neuroprotective effects by inhibiting TLR4-mediated microglial inflammation, which indicates that GPR30 may be a potential target for the treatment of ischemic stroke.

Abstract Background Male fertility is crucial for rice yield, and the improvement of rice yield requires hybrid production that depends on male sterile lines. Although recent studies have revealed several important genes in male reproductive development, our understanding of the mechanisms of rice pollen development remains unclear. Results We identified a rice mutant oslap6 with complete male sterile phenotype caused by defects in pollen exine formation. By using the MutMap method, we found that a single nucleotide polymorphism (SNP) variation located in the second exon of OsLAP6/OsPKS1 was responsible for the mutant phenotype. OsLAP6/OsPKS1 is an orthologous gene of Arabidopsis PKSA/LAP6, which functions in sporopollenin metabolism. Several other loss-of-function mutants of OsLAP6/OsPKS1 generated by the CRISPR/Cas9 genomic editing tool also exhibited the same phenotype of male sterility. Our cellular analysis suggested that OsLAP6/OsPKS1 might regulate pollen exine formation by affecting bacula elongation. Expression examination indicated that OsLAP6/OsPKS1 is specifically expressed in tapetum, and its product is localized to the endoplasmic reticulum (ER). Protein sequence analysis indicated that OsLAP6/OsPKS1 is conserved in land plants. Conclusions OsLAP6/OsPKS1 is a critical molecular switch for rice male fertility by participating in a conserved sporopollenin precursor biosynthetic pathway in land plants. Manipulation of OsLAP6/OsPKS1 has potential for application in hybrid rice breeding.

Abstract Strictosidine synthase (STR) plays an important role in the biosynthesis of terpenoid indole alkaloids (TIAs) and is expressed in a range of active meristematic tissues of higher plants. STR proteins are involved in different physiological and biochemical pathways. However, the function of STR proteins in rice development remains poorly understood. In this study, we identified 21 possible STR-like (OsSTRL) family members in rice genome and found that only one gene, OsSTRL2, exhibited a pre-emergency specific florescence expression pattern. Tissue-specific expression profile analysis, β-glucuronidase histochemical (GUS) staining and RNA in situ hybridization confirmed that OsSTRL2 was highly expressed in tapetal cells and microspores. Comparative protein sequence analysis indicated that OsSTRL2 lacked the key catalytic residue found in a typical STR (STR1), although it possessed conserved β-propellers and α-helices formed the basic structure of STR1. OsSTRL2 knockout mutant resulted to male sterility because of the defects in anther development and pollen wall formation. Subcellular localization of OsSTRL2-YFP revealed that the OsSTRL2 protein was primarily localized in the endoplasmic reticulum (ER). Therefore, OsSTRL2 is an atypical strictosidine synthase that plays crucial roles in regulating anther development and pollen wall formation in rice.

This study developed a new cable-less seismograph system, which can transmit seismic data in real-time and automatically perform high-precision differential self-positioning. Combining the ZigBee technology with the high-precision differential positioning module, this new seismograph system utilized the wireless personal area network (WPAN) and real-time kinematic (RTK) technologies to improve its on-site performances and to make the field quality control (QC) and self-positioning possible. With the advantages of low-cost, good scalability, and good compatibility, the proposed new cable-less seismograph system can improve the field working efficiency and data processing capability. It has potential applications in noise seismology and mobile seismic monitoring.

Growth-regulating factor (GRF) interacting factors (GIFs) are involved in several developmental processes in Arabidopsis. We previously showed that upregulation of OsGIF1 expression improves rice grain size. However, whether OsGIF1 is involved in other developmental processes remains unclear. Here, we report pleiotropic effects of OsGIF1 on rice organ size regulation. Overexpression and functional knock-out via a CRISPR/Cas9 strategy revealed that OsGIF1 not only positively regulates the sizes of rice leaf, stem, and grain but also influences rice reproduction. Expression profiles based on both qRT-PCR and GUS (β-glucuronidase) histochemical staining suggested that OsGIF1 is differentially expressed across various rice tissues, consistent with its roles in regulating the development of multiple rice organs. Additionally, we found that OsGIF1-GFP localized preferentially in the nucleus, which supports its proposed role as a transcriptional cofactor. Further histological analysis suggested that OsGIF1 affected rice organ size possibly by regulating cell size. Our results suggest that OsGIF1 plays important roles in vegetative and reproductive developmental processes, with important implications for rice breeding.

Rice sheath blight, caused by Rhizoctonia solani, is one of the most devastating diseases for stable rice production in most rice-growing regions of the world. Currently, studies of the molecular mechanism of rice sheath blight resistance are scarce. Here, we used an RNA-seq approach to analyze the gene expression changes induced by the AG1 IA strain of R. solani in rice at 12, 24, 36, 48, and 72 h. By comparing the transcriptomes of TeQing (a moderately resistant cultivar) and Lemont (a susceptible cultivar) leaves, variable transcriptional responses under control and infection conditions were revealed. From these data, 4,802 differentially expressed genes (DEGs) were identified. Gene ontology and pathway enrichment analyses suggested that most DEGs and related metabolic pathways in both rice genotypes were common and spanned most biological activities after AG1 IA inoculation. The main difference between the resistant and susceptible plants was a difference in the timing of the response to AG1 IA infection. Photosynthesis, photorespiration, and jasmonic acid and phenylpropanoid metabolism play important roles in disease resistance, and the relative response of disease resistance-related pathways in TeQing leaves was more rapid than that of Lemont leaves at 12 h. Here, the transcription data include the most comprehensive list of genes and pathway candidates induced by AG1 IA that is available for rice and will serve as a resource for future studies into the molecular mechanisms of the responses of rice to AG1 IA.

This paper presents a computational model and design optimization strategy for shape memory alloy (SMA) flexural actuators. These actuators consist of curved SMA wires embedded within elastic structures; one potential application is positioning microcatheters inside blood vessels during clinical treatments. Each SMA wire is shape-set to an initial curvature and inserted along the neutral axis of a straight elastic member (cast polydimethylsiloxane, PDMS). The elastic structure preloads the SMA, reducing the equilibrium curvature of the composite actuator. Temperature-induced phase transformations in the SMA are achieved via Joule heating, enabling strain recovery and increased bending (increased curvature) in the actuator. Actuator behavior is modeled using the homogenized energy framework, and the effects of two critical design parameters (initial SMA curvature and flexural rigidity of the elastic sleeve) on activation curvature are investigated. Finally, a multi-objective genetic algorithm is utilized to optimize actuator performance and generate a Pareto frontier, which is subsequently experimentally validated.

A flexible robotic spine actuated by shape memory alloy (SMA) can achieve both bending motion and impact absorption, which will allow robots to realize a variety of postures. In this paper, the robotic spine is designed and simplified into a multi-segment dynamic model based on several verified assumptions. The SMA wire is modelled using the Seelecke-Muller-Acenbach theory. An iterative algorithm is developed to address the external forces distributed along the spine and compute the spine's bending angle. Based on the dynamic model, we improve the simulation structure and search algorithm to achieve good efficiency and stable solutions. Experiments are conducted to verify the simulation and the results fit the simulation prediction well, with error of less than five degrees. Design optimization with our simulation tool based on several parameters is also discussed in this paper.

Rice restorer lines play an important role in three-line hybrid rice production. Previous research based on molecular tagging has suggested that the restorer lines used widely today have narrow genetic backgrounds. However, patterns of genetic variation at a genome-wide scale in these restorer lines remain largely unknown. The present study performed re-sequencing and genome-wide variation analysis of three important representative restorer lines, namely, IR24, MH63, and SH527, using the Solexa sequencing technology. With the genomic sequence of the Indica cultivar 9311 as the reference, the following genetic features were identified: 267,383 single-nucleotide polymorphisms (SNPs), 52,847 insertion/deletion polymorphisms (InDels), and 3,286 structural variations (SVs) in the genome of IR24; 288,764 SNPs, 59,658 InDels, and 3,226 SVs in MH63; and 259,862 SNPs, 55,500 InDels, and 3,127 SVs in SH527. Variations between samples were also determined by comparative analysis of authentic collections of SNPs, InDels, and SVs, and were functionally annotated. Furthermore, variations in several important genes were also surveyed by alignment analysis in these lines. Our results suggest that genetic variations among these lines, although far lower than those reported in the landrace population, are greater than expected, indicating a complicated genetic basis for the phenotypic diversity of the restorer lines. Identification of genome-wide variation and pattern analysis among the restorer lines will facilitate future genetic studies and the molecular improvement of hybrid rice.
