Abstract

Development of effective therapies against SARS-CoV-2 infections relies on mechanistic knowledge of virus-host interface. Abundant physical interactions between viral and host proteins have been identified, but few have been functionally characterized. Harnessing the power of fly genetics, we develop a comprehensive Drosophila COVID-19 resource (DCR) consisting of publicly available strains for conditional tissue-specific expression of all SARS-CoV-2 encoded proteins, UAS-human cDNA transgenic lines encoding established host-viral interacting factors, and GAL4 insertion lines disrupting fly homologs of SARS-CoV-2 human interacting proteins. We demonstrate the utility of the DCR to functionally assess SARS-CoV-2 genes and candidate human binding partners. We show that NSP8 engages in strong genetic interactions with several human candidates, most prominently with the ATE1 arginyltransferase to induce actin arginylation and cytoskeletal disorganization, and that two ATE1 inhibitors can reverse NSP8 phenotypes. The DCR enables parallel global-scale functional analysis of SARS-CoV-2 components in a prime genetic model system.

Abstract

Big data analysis has become an active area of study with the growth of machine learning techniques. To properly analyze data, it is important to maintain high-quality data. Thus, research on data cleaning is also important. It is difficult to automatically detect and correct inconsistent values for data requiring expert knowledge or data created by many contributors, such as integrated data from heterogeneous data sources. An example of such data is metadata for scientific datasets, which should be confirmed by data managers while handling the data. To support the efficient cleaning of data by data managers, we propose a data cleaning architecture in which data managers interactively browse and correct portions of data through views. Abstract

Views are known mechanisms for controlling access of data and for sharing data of different schemas. Despite long and intensive research on views in both the database community and the programming language community, we are facing difficulties to use views in practice. The main reason is that we lack ways to directly describe view update strategies to deal with the inherent ambiguity of view updating. This paper aims to provide a new language-based approach to controlling and sharing distributed data based on views, and establish a software foundation for systematic construction of such data management systems. Our key observation is that a view should be defined through a view update strategy rather than a view definition. We show that Datalog can be used for specifying view update strategies whose unique view definition can be automatically derived, present a novel P2P-based programmable architecture for distributed data management where updatable views are fully utilized for controlling and sharing distributed data, and demonstrate its usefulness through the development of a privacy-preserving ride-sharing alliance system., Comment: 6 pages. arXiv admin note: text overlap with arXiv:1803.06674

Abstract

The view and the view update are known mechanism for controlling access of data and for integrating data of different schemas. Despite intensive and long research on them in both the database community and the programming language community, we are facing difficulties to use them in practice. The main reason is that we are lacking of control over the view update strategy to deal with inherited ambiguity of view update for a given view. This vision paper aims to provide a new language-based approach to controlling and integrating decentralized data based on the view, and establish a software foundation for systematic construction of such data management systems. Our key observation is that a view should be defined through a view update strategy rather than a query. In other words, the view definition should be extracted from the view update strategy, which is in sharp contrast to the traditional approaches where the view update strategy is derived from the view definition. In this paper, we present the first programmable architecture with a declarative language for specifying update strategies over views, whose unique view definition can be automatically derived, and show how it can be effectively used to control data access, integrate data generally allowing coexistence of GAV (global as view) and LAV (local as view), and perform both analysis and updates on the integrated data. Abstract

Introduction: The left dorsolateral prefrontal cortex (lDLPFC) is a commonly targeted brain region for repetitive transcranial magnetic stimulation (rTMS) for depression. The lDLPFC has been identified using the "5-cm rule." However, identification of the lDLPFC may deviate from the ideal stimulation site localized by neuronavigation. Therefore, we aimed to compare this method with other methods and examine the relationship between deviation from the ideal stimulation site and treatment effects. While most existing studies have focused on participants of European descent, this study focused on Japanese participants. Methods: The study participants were 16 patients who underwent rTMS and had the stimulus location identified using the 5-cm method. The lDLPFC was identified by the F3 electrode position and neuronavigation in addition to the 5-cm rule, and these locations were compared. We then performed a correlation analysis of the distance between the sites identified by the 5-cm method and by neuronavigation, as well as changes in scores on the 17-item Hamilton Depression Scale (HAMD-17). Results: The lDLPFC identified by the F3 site and neuronavigation was approximately 3 cm more anterolateral than that identified by the 5-cm method. A significant correlation was found between the distance between the sites identified by the 5-cm method and neuronavigation and the rate of change in HAMD-17 scores. Conclusion: The ideal stimulation site may be approximately 3 cm anterior to the site identified by the 5-cm method, and stimulation of the F3 site may be a valid alternative to the 5-cm method. Coastal cities with low elevation and mild slopes tend to be more vulnerable to the threat of floods and inundations. Expansion of impermeable areas caused by land conversion greatly diminishes a city’s ability to cope with the threat. Urban drainage systems play a crucial role in controlling excess surface water and reducing the threat of flood and inundation. To maintain an urban drainage system within an optimumcondition and to reduce inundation risk, various types of drainage maintenance activities have been implemented in Tegal. This research was conducted in five inundationproneareas in Tegal to analyze the impact of those maintenance activities to urban drainage conditions andin reducing inundation. GIS and statistical analysis revealed that fromthe numerousurban drainage maintenance activities that have been performed, drainage sediment cleanuphas the most obvious impact on drainage conditions and in reducing inundation.

Abstract

Aims: A meta‐analysis of short‐term studies revealed no significant differences between the doses of asenapine, 10 and 20 mg/day, in the acute treatment of schizophrenia. However, it should be noted that many patients from clinical practice were excluded, and the dose–response to asenapine in a real‐world setting is still unclear. Additionally, the dose–response in the maintenance phase is not clear. This study aimed to evaluate the differences in the efficacy of different asenapine doses in patients with maintenance phase of schizophrenia in a real‐world setting. Methods: This study conducted post‐marketing surveillance of asenapine in clinical settings in Japan. It followed patients diagnosed with schizophrenia who received asenapine for the first time for a maximum of 52 weeks. These patients were divided into two categories based on their average daily asenapine dosage: ≤10 mg/day and >10 mg/day. Asenapine efficacy was assessed by adjusting for patient demographics using multivariate logistic regression analysis, employing the Clinical Global Impression‐Global Improvement (CGI‐I) scale, which has seven categories. Results: A total of 2774 patients were included in the analysis. Of these, 1689 and 1085 patients were treated with asenapine ≤10 mg/day and >10 mg/day, respectively. The CGI‐I improvement rate was significantly higher in the asenapine >10 group (p = 0.012) after adjusting for patient background factors. Conclusion: These results suggest that asenapine doses >10 mg/day may be more effective than 10 mg/day in the treatment of schizophrenia; however, further studies are needed to confirm these findings.
