Background: The scarcity and preciousness of plateau characteristic medicinal plants pose a significant challenge in obtaining sufficient quantities of experimental samples for quality evaluation. Insufficient sample sizes often lead to ambiguous and questionable quality assessments and suboptimal performance in pattern recognition. Shilajit, a popular Tibetan medicine, is harvested from high altitudes above 2000 m, making it difficult to obtain. Additionally, the complex geographical environment results in low uniformity of Shilajit quality. Methods: To address these challenges, this study employed a deep learning model, time vector quantization variational auto- encoder (TimeVQVAE), to generate data matrices based on chromatographic and spectral for different grades of Shilajit, thereby increasing in the amount of data. Partial least squares discriminant analysis (PLS-DA) was used to identify three grades of Shilajit samples based on original, generated, and combined data. Results: Compared with the originally generated high performance liquid chromatography (HPLC) and Fourier transform infrared spectroscopy (FTIR) data, the data generated by TimeVQVAE effectively preserved the chemical profile. In the test set, the average matrices for HPLC, FTIR, and combined data increased by 32.2%, 15.9%, and 23.0%, respectively. On the real test data, the PLS-DA model's classification accuracy initially reached a maximum of 0.7905. However, after incorporating TimeVQVAE-generated data, the accuracy significantly improved, reaching 0.9442 in the test set. Additionally, the PLS-DA model trained with the fused data showed enhanced stability. Conclusion: This study offers a novel and effective approach for researching medicinal materials with small sample sizes, and addresses the limitations of improving model performance through data augmentation strategies. Shilajit is a phyto-mineral diffusion and semi-solid matter used as traditional medicine with extraordinary health benefits. This study provides a comprehensive data on Shilajit with emphasis on heavy metal profile, associated toxicities, and metal detoxification mechanisms by humic substances present in Shilajit. Data was searched across papers and traditional books using Google Scholar, PubMed, Science Direct, Medline, SciELO, Web of Science, and Scopus as key scientific databases. Findings showed that Shilajit is distributed in almost 20 regions of the world with uses against 20 health problems as traditional medicine. With various humic substances, almost 11 biological activities were reported in Shilajit. This phyto-mineral diffusion possesses around 65 heavy metals including the toxic heavy metals like Cu, Al, Pb, As, Cd, and Hg. However, humic substances in Shilajit actively detoxify around 12 heavy metals. The recommended levels of heavy metals by WHO and FDA in herbal drugs is 0.20 and 0.30 ppm for Cd, 1 ppm for Hg, 10.00 ppm for As and Pb, 20 ppm for Cu, and 50 ppm for Zn. The levels of reported metals in Shilajit were found to be lower than the permissible limits set by WHO and FDA, except in few studies where exceeded levels were reported. Shilajit consumption without knowing permissible levels of metals is not safe and could pose serious health problems. Although the humic substances and few metals in Shilajit are beneficial in terms of chelating toxic heavy metals, the data on metal detoxification still needs to be clarified. [ABSTRACT FROM AUTHOR]

In this work the physicochemical, antioxidant and antibacterial properties of two varieties of yellow and black shilajit, obtained from Kerman mountains, were studied. The findings indicated notable distinctions between the physicochemical properties of black and yellow shilajit (P < 0.05). Yellow shilajit displayed the greatest and least quantities of iron and calcium, respectively, while black shilajit displayed the opposite pattern. The main functional groups detected in the FTIR spectra of both black and yellow shilajit comprised O-H, C = O, -COO-, C-H, and C-O-C. The results of the colorimetry test revealed a significant difference in the L* color indices between the two types of black and yellow shilajit (p < 0.05). Consequently, the most elevated and most reduced L* color indices were associated with the yellow shilajit and the black sample, respectively. Furthermore, the black and yellow shilajits exhibited the highest and lowest values for the color indexes a* and b*, respectively. Black shilajit showed superior antimicrobial activities compared to yellow shilajit in both disc diffusion method and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Overall, the outcomes of this study indicated that both black and yellow shilajit have the potential to be used in fortified food products after demonstrating their non-cytotoxicity. This is due to the high protein content and strong antioxidant and antimicrobial properties found in black shilajit, as well as the elevated levels of calcium and iron in yellow shilajit. Background: Shilajit is a natural pale-brown to blackish-brown phytocomplex that exudes from mountain rocks in various parts of the world. In recent years, numerous pharmacological effects of shilajit have been explored through extensive experiments. Shilajit is known as an antioxidant and anti-inflammatory agent. Activation of the nuclear factor kappa B (NF-κB) signaling pathway could be an important player in inflammation-driven tumor progression. Therefore, the current study evaluated the anti-inflammatory effect of shilajit on a breast cancer cell line (MCF7). Methods: In this experimental study, the MCF7 cell line was treated with shilajit, and an MTT assay was applied to analyze the half-maximal inhibitory concentration (IC50) after 72 hours of treatment. Following this, the apoptotic rate was assessed using flow cytometry, and p50, RelB, and IKKα/β gene expression were evaluated using real-time PCR assay. Results: Shilajit had potent cytotoxic activity in a dose- and time-dependent manner with an IC50 of 280 μg/mL. Based on the Annexin-PI analysis, the IC50 concentration of this compound induced significant apoptosis in the cells, possibly through suppression of NF-κB (p50, RelB, and IKKα/β)-regulated genes. The real-time PCR results indicated that treating MCF7 cells at the IC50 dose of shilajit for 72 hours could reduce the mRNA expression levels of p50, RelB, and IKKα/β in the cells. Conclusion: The findings of the present experiment showed that shilajit may have promising anti-breast-tumor and antiinflammatory activity through inhibition of the NF-κB signaling pathway. Shilajit is an ancient herbal medicine from India with many uses. It is a viscous material prominently present in the Himalayan and Hindukush rocks of the Indian region. The formation due to plant decompostion with time. Herbs assosciated to the stones reveals about composition of Shilajit. Primary objective is to create GCMS/MS and LC-analysis techniques for separating the phytochemicals and complete profiling of volatile and non-volatile metabolites found in Shilajit. Accordingly, an attempt was undertaken to develop a method for standardisation and, subsequently, certification inorganic content and purity of shilajit in various indian regions S-01, S-02, S-03, S-04, and S-05. Thus, an effort was made to create a fundamental technique for standardization and, consequently, authentication of shilajit that was acquired from various geographical locations.Trace metal concentrations were measured using ICP OES in mg Kg−1 for each sample. These elements included As, Pb, Cd, Ag, Al, B, Ba, Cr, Cu, Fe, Mn, Ni, Sb, Se, Zn, P, Ca, K, Mg, and Na. ICP OES analysis observed the significant concentration of Ca and K, in S-01 (102631 and 26779), S-02 (13529.25 and 36866.35), S-03 (8170.84 and 37989.10), S-04 (9616.37 and 18843.77), and S-05 (4992.10 and 2120.64), appropriately. By measuring the DPPH radical scavenging activity were assessed for their antioxidant activities of the S-01, S-02, S-03, S-04, and S-05 samples: 12.316 ± 0.561 %, 13.144 ± 2.884 %, 15.105 ± 1.421 %, 19.683 ± 2.013 %, and 0.0724 ± 1.045 %, respectively. Concurrently, TPC studies on Shilajit found between 10 and 150 mg g−1 wt Similarly, shilajit’s TFC was analysed with geographically. Background: Medications, including chemotherapeutic drugs, contribute to male infertility as external factors by inducing oxidative stress in testicular cells. Shilajit is a naturally occurring bioactive antioxidant used in Ayurvedic medicine to treat a variety of ailments. Objective: This study examines the potential of Shilajit to counteract the negative effects of the chemotherapeutic drug cyclophosphamide (CPA) on testicular germ cell dynamics. Material and methods: Male Parkes mice received single intraperitoneal CPA injection (200 mg/kg BW) on day one, followed by daily supplementation of Shilajit (100 and 200 mg/kg BW) for one spermatogenic cycle. Results: CPA adversely affected testicular germ cell dynamics by inhibiting the conversion of spermatogonia-to-spermatids, altering testicular histoarchitecture, impairing Sertoli cell function and testicular steroidogenesis, and disturbing the testicular oxido-apoptotic balance. Shilajit supplementation restores testicular germ cell dynamics in CPA-exposed mice, as evidenced by improved histoarchitecture of the testis. Shilajit improves testicular daily production and sperm quality by promoting the conversion of spermatogonia (2C) into spermatids (1C), stimulating germ cell proliferation (PCNA), improving Sertoli cell function (N-Cadherin and β-Catenin), and maintaining the Bax/Bcl2 ratio. Additionally, Shilajit enhances testosterone biosynthesis by activating enzymes like 3β-HSD, and 17β-HSD. Shilajit also reduces testicular oxidative stress by increasing antioxidant enzyme activity (SOD) and decreasing lipid peroxidation (LPO). These effects are mediated by upregulation of the antioxidant protein Nrf-2 and downregulation of Keap-1. Conclusion: The findings underscore the potent androgenic and antioxidant characteristics of Shilajit, as well as its ability to enhance fertility in cases of testicular damage caused by chemotherapeutic drugs.

This study aimed to investigate the effects of adding shilajit to the diet of honey bees on semen freezing. A total of 5 groups were formed in the research, one of which was a control group (SH-0) and the other four were an experimental group (SH-1, SH-2, SH-3, SH-4). A total of 25 study colonies were used, 5 in each group. While the SH-0 group was formed without using any additives, the experimental groups were fed with the addition of shilajit (1/1 sugar/water) in different doses (5, 10, 15, 20 mg/L) to the honey bee diet. The collected semen samples were frozen in liquid nitrogen vapor and then stored in liquid nitrogen at -196°C. Then, semen samples were thawed at 37°C and evaluated to determine spermatological parameters (motility, acrosome integrity, plasma membrane integrity, spermatozoa concentration). It was also examined in terms of total oxidant and total antioxidant. Compared to the control group, it was determined that all shilajit doses significantly increased spermatological parameters such as motility, hypoosmotic swelling test (HOST), acrosome integrity (p<0.001), but did not significantly affect the spermatozoa concentration value (p >0.05). Although there is no statistical difference between the groups in terms of semen TAS (Total Antioxidant Status) and TOS (Total Oxidant Status) values, which are oxidative stress parameters, the numerical increase in TAS values in SH-2 and SH-3 groups; striking. AbstractShilajit, a natural material known for thousands of years, is primarily famous because it was the main constituent used for the mummification of bodies by dynastic Egyptians. However, for millennia, it has also been an important drug and supplement in traditional and modern medicine. A full search of Shilajit in the literature, reveals that its chemical composition is not known with certainty. That’s due to the fact that surprisingly, no comprehensive chemical analysis has ever been performed to find all the chemicals and species present in it. Moreover, its source is not known; some believe that it is formed from dead plant residues or animal excrements, and some others believe it is a mineral secreted from rocks gradually. In this review article, it is tried to gather chemical property information available regarding this soft, mysterious black material to clarify what it is composed of and what formulation commercial supplements and drugs based on Shilajit may have. While there are many differences in the chemical compositions of Shilajit from various regions, it has been discovered that more than 80% of weight of Shilajit is mainly composed of humic substances and almost 20% of its weight consists of calcium, potassium, and magnesium. It also contains amino acids (mainly glycine) and proteins, fatty acids, bioactive compounds such as caffeic acid and gallic acid, and heavy metals including chromium, selenium, and cobalt. Abstract Shilajit, along with Triphala, has been one of the most widely used herbal drug combinations for the symptomatic treatments of hyperthyroidism. However, the high polarity difference among active constituents proposes difficulties while analyzing its quality. In this study, two high-performance thin-layer chromatography methods were developed for chromatographic fingerprinting of the proposed herbal mixture of Triphala and Shilajit. A reliable HPTLC method effectively analyzed bioactive phytoconstituents such as quercetin, myricetin, gallic acid, ellagic acid, tannic acid, ascorbic acid, and humic acid. Method 1 showed good separation of quercetin, myricetin, and tannic acid, but separating gallic and ellagic acid proved difficult. To address this issue, the 2D-TLC approach involves a second mobile phase. Although humic acid and ascorbic acid were not detected due to their highly polar and non-polar nature, so a different HPTLC method 2 was developed. The successful method of development using the Quality by Design approach and validated. The developed method successfully analyzed the bioactive constituents of Shilajit, in combination with Triphala, and validation parameters such as linearity, accuracy, precision, range, specificity, robustness, the limit of detection (LOD), and limit of quantification (LOQ) agreed with given values. Background: Shilajit is mentioned in the “Kama Sutra” as a potent enhancer of sexual desire. This study aimed to investigate the effects of oral Shilajit tablets on sexual function and sexual quality of life among women of reproductive age. Methods: Forty-eight reproductive-aged women participated in a placebo-controlled triple-blind clinical trial. The intervention group took oral Shilajit tablets (200 mg) twice daily for 60 days and the control group took the placebo. Data collection tools were Sexual Quality of Life-Female and Female Sexual Function Index. Data were collected before the intervention, 30, 60, and 90 days after the start of the study. Results: Forty-three women completed the study. The mean score of total sexual function in the intervention group was significantly higher than before the intervention (P < 0.001). The mean score of sexual function was 28.93 after 90 days in the intervention group while it was 22.09 in the control group. This finding was observed in most domains of the sexual function index. The mean score of sexual quality of life increased after 60 days of intervention in both groups; however, the difference was not statistically significant (P = 0.094). Conclusion: The study indicated that Shilajit, as a complementary therapy, may improve sexual function and most of its domains; while there was no effect on improving the quality of sexual life. Natural additives are crucial to maintaining healthy aquaculture practices as they boost fish wellness, reduce related environmental problems, and improve productivity. The current study aims to assess the effect of shilajit on common carp (Cyprinus carpio) metabolites, such as blood parameters, liver function, and growth performance. The fish were fed a basal diet supplemented with 0, 1, 2, and 3 g/kg shilajit for 60 days. The results showed that shilajit supplementation significantly improved the growth performance of the fish with increasing the shilajit concentration in the diet. Using shilajit in experimentally treated fish enhanced hemoglobin levels, red blood cell count, and hematocrit index. On the other hand, the administration of shilajit did not result in any significant modifications to the liver enzymes: aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP), suggesting no effect on liver function. The integration of natural additives in the common carp diet improves growth performance and general fish health, which can lead to increased production and reduced environmental impact. Shilajit is naturally found substance, also termed as shilajatu, salajit, mummiyo, or mimie. It is blackish powder obtained from high mountain rocks, especially in the Himalayans mountains. It has been used by Ayurvedic medicines to cure several disorders and to improve the quality of life. To support its rasayana property, an effort has been made to review shilajit with respect to its origin, variations, physical properties, purification, medicinal properties, and significant biological properties. Shilajit has been demonstrated to offer different health advantages based on the area from where it was harvested. Antioxidant activity and immuno-modulatory activity were highlighted among the many biological characteristics of shilajit since they are strongly related to its rasayana potential. Background and Objective: Liver damage is one of the common complications after ulcerative colitis. The aim of this study is to compare liver protective effects of sulfasalazine and shilajit after ulcerative colitis. Methods: In this experimental intervention study, 49 male Wistar rats weighing 200-250 grams were used in seven groups of 7: sham, ulcerative colitis, oral solution, oral Shilajit, sulfasalazine, rectal solution, and rectal Shilajit. To induce ulcerative colitis, after anesthetizing the animal, 2 cc of acetic acid (4%) was used by rectal administration. Four days after the development of colitis, Shilajit was administered for four days at a dose of 250 mg/kg by gavage and rectal methods. In each group, the levels of liver enzymes (SGPT, SGOT, ALP), direct bilirubin, total, and serum albumin were evaluated eight days after the induction of ulcerative colitis. Findings: Sulfasalazine decreased total bilirubin (0.5967±0.04) and SGOT (113.3±12.7). Administering Shilajit rectally reduced direct bilirubin (0.10±0.02), albumin (3.17±0.2), SGOT (156.2±12.7) and administering Shilajit by gavage reduced SGOT (125.0±12.7), direct bilirubin (0.094±0.02), SGOT (125.0±12.7) and SGPT (93.8±7.5). Conclusion: The results of the study showed that oral administration of Shilajit has more protective effects on liver damage caused by ulcerative colitis than rectal Shilajit, and this result is comparable to sulfasalazine.

Abstract Background Shilajit has been widely used remedy for treating a numerous of illness such as bone defects in Iran traditional folk medicine since hundreds of years ago. The aim of the present study was to explore the effect of Shilajit on the osteogenic differentiation of human adipose-derived mesenchymal stem cells (ASCs) in two- (2D) and three-dimensional (3D) cultures. Materials and methods ASCs were seeded in 3D 1% alginate (Alg) hydrogel with or without Shilajit (500 µg/mL) and compared with 2D cultures. Then, characterization was done using electron microscopy (SEM)/energy-dispersive X-ray spectroscopy (EDX), alkaline phosphatase (ALP) activity, alizarin red staining and Raman confocal microscopy. Results Adding Shilajit had no impact on the Alg scaffold degradability. In the 3D hydrogel and in the presence of osteogenic medium (OM), Shilajit acted as enhancer to increase ALP activity and also showed osteoinductive property in the absence of OM compared to the 2D matched groups at all time points (days 7 and 21 both P = 0.0006, for 14 days P = 0.0006 and P = 0.002, respectively). The present study aims to develop Asphaltum punjabianum (namely Shilajit) coated Polyvinyl alcohol (PVA)/Carboxymethyl cellulose (CMC) hydrogels and examine their structural, morphological, degradation, and biological properties. Hydrogels were produced at two different concentrations: 70:30 PVA/CMC and 90:10 PVA/CMC. Following that, Shilajit was applied to the synthesized hydrogels using electrophoretic deposition for a duration of 3 min at 30 V. The scanning electron microscopy images showed that the hydrogel's surface had a regular distribution of irregular Shilajit particles. Fourier transform infrared spectroscopy (FTIR) analysis demonstrated the presence of hydrogen bonding between PVA and CMC hydrogels and Shilajit, indicating the successful deposition of Shilajit on the hydrogel. The hydrogels coated with Shilajit exhibited strong antimicrobial activity, resulting in an inhibition zone measuring 34 mm against Escherichia coli (E. coli) and 41 mm against Staphylococcus aureus (S. aureus). The hydrogels exhibited a cell viability of 80 % with mesenchymal stem cells (MSCs), and the release of collagen II also increased. Furthermore, the PVA/CMC/Shilajit hydrogel exhibited a lower degradation rate compared to the PVA/CMC hydrogel. The results of the swelling, degradation, and drug release studies indicate that the shilajit coating is appropriate for the long-term process of tissue and cartilage regeneration. This clinical review presents what is known about the antiviral features of humic substances (HS) to the benefit of the clinical healthcare provider using available data in humeomics, the study of the soil humeome. It provides the reader with a working framework of historical studies and includes clinically relevant data with the goal of providing a broad appreciation of the antiviral potential of humic substances while also preparing for a translational leap into the clinical application of humic acid.

Background: Thrombocytopenia is a well-recognized side effect of cytotoxic chemotherapy, and may eventuate in treatment failure. Shilajit is a naturally-occurring exudate traditionally utilized in Ayurvedic medicine for thousands of years. Recent studies have portrayed the antioxidant and anti-inflammatory characteristics of shilajit. Therefore, the aim of this study was to investigate the effect of shilajit on carboplatin-induced thrombocytopenia and oxidative stress in rats. Materials and Methods: This study was conducted on Sprague Dawley rats with a weight range of 200-240 grams. Twenty-four rats were divided into four groups of six, and received 30, 40, or 60 mg/kg carboplatin to establish the most effective regimen to induce thrombocytopenia. Another 24 rats were divided into two control groups (positive and negative), a low-dose (200 mg/kg), and a high-dose (800/mg/kg) shilajit group. After two weeks, blood samples were analyzed regarding platelet count and oxidative stress markers, including intracellular reduced glutathione and (GSH) and malondialdehyde (MDA) concentration, and glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD) activity. Results: Carboplatin at the dose of 60 mg/kg was more prominent in inducing thrombocytopenia. Shilajit exhibited a directly dose-dependent effect in raising platelet count, which failed to show statistical significance. However, a statistically significant dose-dependent effect in increasing intracellular GSH, and GPx, CAT, and SOD activity, and decreasing MDA concentration was documented. Conclusion: Shilajit is capable of significantly reducing carboplatin-induced oxidative stress, but requires further investigation to establish its impact on carboplatin-induced thrombocytopenia. Background: Shilajit has been widely used remedy for treating a numerous of illness such as bone defects in Iran traditional folk medicine since hundreds of years ago. The aim of the present study was to explore the effect of Shilajit on the osteogenic differentiation of human adipose-derived mesenchymal stem cells (ASCs) in two- (2D) and three-dimensional (3D) cultures. Materials and methods: ASCs were seeded in 3D 1% alginate (Alg) hydrogel with or without Shilajit (500 µg/mL) and compared with 2D cultures. Then, characterization was done using electron microscopy (SEM)/energy-dispersive X-ray spectroscopy (EDX), alkaline phosphatase (ALP) activity, alizarin red staining and Raman confocal microscopy. Results: Adding Shilajit had no impact on the Alg scaffold degradability. In the 3D hydrogel and in the presence of osteogenic medium (OM), Shilajit acted as enhancer to increase ALP activity and also showed osteoinductive property in the absence of OM compared to the 2D matched groups at all time points (days 7 and 21 both P = 0.0006, for 14 days P = 0.0006 and P = 0.002, respectively). In addition, calcium deposition was significantly increased in the cultures exposed to Shilajit compared to 2D matched groups on days 14 (P < 0.0001) and 21 (P = 0.0003 and P = 0.003, respectively). In both 3D and 2D conditions, Shilajit induced osteogenic differentiation, but Shilajit/Alg combination starts osteogenic differentiation in a short period of time. Conclusion: As Shilajit accelerates the differentiation of ASCs into the osteoblasts, without changing the physical properties of the Alg hydrogel, this combination may pave the way for more promising remedies considering bone defects. ‏Shilajit is used commonly as Ayurvedic medicine worldwide which is Rasayana herbo-mineral substance and consumed to restore the energetic balance and to prevent diseases like cognitive disorders and Alzheimer. Locally, Shilajit is applied for patients diagnosed with bone fractures. For safety of the patients, the elemental analysis of Shilajit is imperative to evaluate its nutritional quality as well as contamination from heavy metals. The elemental composition of Shilajit was conducted using three advanced analytical techniques (LIBS, ICP, and EDX). For the comparative studies, the two Shilajit kinds mostly sold globally produced in India and Pakistan were collected. Our main focus is to highlight nutritional eminence and contamination of heavy metals to hinge on Shilajit therapeutic potential. In this work, laser-induced breakdown spectroscopy (LIBS) was applied for qualitative and quantitative analysis of the Shilajit. Our LIBS analysis revealed that Shilajit samples composed of several elements like Ca, S, K, Mg, Al, Na, Sr, Fe, P, Si, Mn, Ba, Zn, Ni, B, Cr, Co, Pb, Cu, As, Hg, Se, and Ti. Indian and Pakistani Shilajits were highly enriched with Ca, S, and K nutrients and contained Al, Sr, Mn, Ba, Zn, Ni, B, Cr, Pb, As, and Hg toxins in amounts that exceeded the standard permissible limit. Even though the content of most elements was comparable among both Shilajits, nutrients, and toxins, in general, were accentuated more in Indian Shilajit with the sole detection of Hg and Ti. The elemental quantification was done using self-developed calibration-free laser-induced breakdown spectroscopy (CF-LIBS) method, and LIBS results are in well agreement with the concentrations determined by standard ICP-OES/MS method. To verify our results by LIBS and ICP-OES/MS techniques, EDX spectroscopy was also conducted which confirmed the presence above mentioned elements. “Testosterone boosters” (TB)—are supplements that are claimed to increase testosterone levels in the human body. While the consumption of TB may be popular among athletes, there is insufficient evidence both about the safety and the real efficacy of TB. In our review, we searched MEDLINE/PubMed and Cochrane Library for studies on the effects of 15 substances that are claimed to increase testosterone levels Anacyclus pyrethrum; Bulbine natalensis; Epimedium (horny goat weed); L-arginine; L-carnitine; magnesium; Mucuna pruriens; pantothenic acid; selenium; shilajit Eurycoma longifolia (Tongkat Ali); Serenoa repens (saw palmetto); boron; Withania somnifera (ashwagandha); and Trigonella foenum-graecum (fenugreek) in athletes and healthy adults under 55 years of age. We found such studies regarding 10 out of 15 substances: L-arginine (3 studies); L-carnitine (2); magnesium (1); selenium (2); shilajit (1); Tongkat Ali (2); Serenoa repens (1); boron (3); ashwagandha root (2); and fenugreek (7). Many of them fail to prove the efficacy of these substances to increase testosterone levels. Tongkat Ali, ashwagandha, and fenugreek were the substances with the strongest evidence. The positive effect of magnesium and shilajit on testosterone concentration was shown in single studies. Conflicting data found that L-arginine, L-carnitine, Serenoa repens, selenium and boron do not appear to increase testosterone levels. There are almost no data on the safety profile of various TB components; however, certain TB components may be linked to coagulation, and pancreatic and hepatic disorders. Based on the review, the authors conclude that at present TB cannot be recommended for use by athletes due to insufficient data on their efficacy and safety.

Objective: To evaluate the effect of Shilajit, a medicine of Ayurveda, on the serum changes in cytokines and adipokines caused by non-alcoholic fatty liver disease (NAFLD). Methods: After establishing fatty liver models by feeding a high-fat diet (HFD) for 12 weeks, 35 Wistar male rats were randomly divided into 5 groups, including control (standard diet), Veh (HFD + vehicle), high-dose Shilajit [H-Sh, HFD + 250 mg/(kg·d) Shilajit], low-dose Shilajit [L-Sh, HFD + 150 mg/(kg·d) Shilajit], and pioglitazone [HFD + 10 mg/(kg·d) pioglitazone] groups, 7 rats in each group. After 2-week of gavage administration, serum levels of glucose, insulin, interleukin 1beta (IL-1β), IL-6, IL-10, tumor necrosis factor-alpha (TNF-α), adiponectin, and resistin were measured, and insulin resistance index (HOMA-IR) was calculated. Results: After NAFLD induction, the serum level of IL-10 significantly increased and serum IL-1β, TNF-α levels significantly decreased by injection of both doses of Shilajit and pioglitazone (P<0.05). Increases in serum glucose level and homeostasis model of HOMA-IR were reduced by L-Sh and H-Sh treatment in NAFLD rats (P<0.05). Both doses of Shilajit increased adiponectin and decreased serum resistin levels (P<0.05). Conclusion: The probable protective role of Shilajit in NAFLD model rats may be via modulating the serum levels of IL-1β, TNF-α, IL-10, adipokine and resistin, and reducing of HOMA-IR. Background: The levels of IL-13, IL-4, IL-1B, TNF-α and IL-10 alter in the colon of people with ulcerative colitis. previous studies, it was found that shilajit (asphaltum) was effective in the improvement of ulcerative colitis.we guessed that the shilajit has been able to improve the ulcerative colitis by affecting the amount of cytokines. Objective: The aim of the present study was to investigate the effects of shilajit on IL-13, IL-4, IL-1B, TNF-α and IL-10 in ulcerative colitis. Methods: In this experimental study, 49 white male rats were randomly divided into 7 groups including sham groups, ulcerative colitis, Gavage vehicle, Gavage shilajit, sulfasalazine, Intra Anal Vehicle, Intra Anal shilajit. To produce ulcerative colitis 2 cc acetic acid (4%) was prescribed intra-rectal. 4 days after the induction of colitis, it was given 4 days: shilajit 250 mg / kg was used as a gavage or intra anal. Sulfasalazine was dosed at 250 mg / kg in the form of gavage. Results: The Gavage shilajit group increased IL-10, IL-4and decreased IL-1β, TNF-α compared to Gavage vehicle group. Shilajit has been able to close the levels of IL-4, IL-13, IL-10, TNF-α to normal values of the sham group. Conclusion: Considering that the weight ratio of intestinal rat to the body of rat in the shilajit groups was lower than that of the sulfasalazine group, it can be shown that shilajit has been able to more reduce inflammation by making a more suitable change in cytokines in comparison with sulfasalazine.

Abstract Background Shilajit is a commonly used Tibetan medicine, and its water extract is mainly used for various heat-related syndrome, especially that of stomach, liver and kidney. Shilajit is found to exudate from rocks of cliff at an altitude of 2000–4000 m as a water-soluble mixture of black paste and animal feces of Trodocterus spp. or Ochotona spp. Because it is difficult to reach the exudation points so as to explain the its formation process, the source of Shilajit still remains unclear and controversial, which severely impedes its safety and efficacy in clinical application. Methods In this work, a series of investigations as rock flakes identification, porosity determination, rock mineral analysis, scanning electron microscopy (SEM), and energy dispersive spectrometer (EDS) have been carried out to clarify the source of Shilajit, including the storage condition and exudation process of its organic matter, and to investigate the geological structure of the exudation points as well as physical and chemical characteristics of the mother rocks. Results The Shilajit exudation points were mainly distributed on the steep cliffs, where there were cavities and sections that could not be eroded by rainwater. The fundamental structure of the exudation points was determined by the rock’s bedding planes, joints, fracture surfaces and faults, and developed into micro-topography later. The exudation points were distributed in the Triassic strata and scattered in the Early Mesozoic granitoids. The lithologic features were mainly slate, carbonaceous slate and sandy slate etc. The background rocks were characterized by intergranular pores, dissolved pore, joint and fracture development. Organic matter was widely distributed in these pores and fissures, which had condition for storage and exudation of organic matter. Conclusions Shilajit mainly distributed on sunny steep slopes and cliffs with a slope of 60° or above at altitude of 2000–4000 m. The lithology character of the Shilajit exudation area were mainly various metamorphic rocks of sedimentary rocks that were rich in organic carbon. The organic matter in Shilajit was found to flow out naturally from rocks along pore, structural plane and even accumulate on the surface of rock as a result of storage environment change caused by rock tectonic action.

Abstract A hybrid magnetic material γFe2O3@Sh@cu2O was easily prepared from Shilajit (Sh) decorated Fe3O4 and copper acetate. The prepared magnetic hybrid material was fully characterized using different analysis, including Fourier transform infrared (FT-IR), X-ray diffraction (XRD), inductively coupled plasma (ICP), scanning electron microscopy (SEM), Energy-dispersive X-ray spectroscopy (EDX), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometer (VSM) thermal gravimetric analysis (TGA) and Brunauer–Emmett–Teller (BET). All these analysis revealed that during coating of Fe3O4@Sh using copper salt (II), synchronized redox sorption of CuII to CuI occurs at the same time as the oxidation of Fe3O4 to γFe2O3. This magnetic catalyst exhibited excellent catalytic activity for regioselective synthesis of 1,4-disubstituted-1,2,3-triazoles via one pot three-component click reaction of sodium azide, terminal alkynes and benzyl halides in the absence of any reducing agent. High yields, short reaction time, high turnover number and frequency (TON = 3.5 * 105 and TOF = 1.0 * 106 h−1 respectively), easy separation, and efficient recycling of the catalyst are the strengths of the present method.

Shilajit is a natural substance found in the Himalayan region from Nepal to Pakistan. It is a decomposition product of Royle’s spurge , white clover , and different species of molds. The decomposition takes place over a time span of centuries by the action of microorganism. In the present study, shilajit samples from four different origins including siachen khaplu shilajit (SKS), kharmang pari saspolo shilajit (KPSS), kharmang ghandus shilajit (KGS), and kharmang shilajit center (KSC) of district Skardu, Pakistan were investigated. These samples were characterized using scanning electron microscopy (SEM), Energy-dispersive x-ray spectroscopy (EDS), x-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), and UV-visible spectroscopy (UV/vis). SEM revealed a notable difference in the shape and size of collected samples. All samples were found to possess crystalline nature, which is confirmed from XRD. The presence of multi-components and complex silicates confirmed the presence of humic substances (HS) in shilajit. A slight disparity in physiological properties of four samples were revealed due to geographical variations and ecological conditions, which determine the natural synthesis of shilajit. All samples exhibited antibacterial effects against Gram negative bacteria; Escherichia coli (E.coli) . About 76%, 98%, and 100% of bacteria were killed by SKS, both KPSS and KGS, and KSC samples, respectively. The cell viability analysis revealed that the KPSS (66%) and KGS (53%) were cyto-compatible as compared to the SKS (23%) and KSC (25%) samples. The Chick Chorionic Allantoic Membrane (CAM) assay was used to observe the angiogenic potential for SKS, KSC, and KGS samples. Hence, shilajit sample could be a potential candidate for the medicinal applications and offer a new approach to biomedical applications.

Shilajit is a naturally occurring bioactive antioxidant used in Ayurvedic medicine to treat a variety of ailments., Objective: This study examines the potential of Shilajit to counteract the negative effects of the chemotherapeutic drug cyclophosphamide (CPA) on testicular germ cell dynamics., Material and Methods: Male Parkes mice received single intraperitoneal CPA injection (200 mg/kg BW) on day one, followed by daily supplementation of Shilajit (100 and 200 mg/kg BW) for one spermatogenic cycle., Results: CPA adversely affected testicular germ cell dynamics by inhibiting the conversion of spermatogonia-to-spermatids, altering testicular histoarchitecture, impairing Sertoli cell function and testicular steroidogenesis, and disturbing the testicular oxido-apoptotic balance. Shilajit supplementation restores testicular germ cell dynamics in CPA-exposed mice, as evidenced by improved histoarchitecture of the testis. Shilajit improves testicular daily production and sperm quality by promoting the conversion of spermatogonia (2C) into spermatids (1C), stimulating germ cell proliferation (PCNA), improving Sertoli cell function (N-Cadherin and β-Catenin), and maintaining the Bax/Bcl2 ratio. Additionally, Shilajit enhances testosterone biosynthesis by activating enzymes like 3β-HSD, and 17β-HSD. Shilajit also reduces testicular oxidative stress by increasing antioxidant enzyme activity (SOD) and decreasing lipid peroxidation (LPO). We evaluated macroscopic and histopathological lesions in the stomach, measured the activity of oxidative stress enzymes in gastric tissue homogenates, and assessed serum electrolytes and parameters of kidney and liver function. Forty‐five male rats were allocated to five groups: Normal control, positive control, omeprazole treatment, Shilajit treatment, and Shilajit control. The treatment period lasted for four consecutive days. The size and number of gastric lesions were significantly reduced in the Shilajit and omeprazole groups compared to the positive control group, indicating a reduction in mucosal damage and the severity of edema and leukocyte infiltration in tissue sections. A significant increase was observed in the levels of all oxidative stress parameters, except malondialdehyde, in rats treated with Shilajit and omeprazole compared to those in the positive control group. The effect of the aqueous extract of Shilajit was comparable to that of omeprazole. These results indicated the protective effects of Shilajit against aspirin‐induced gastric lesions.

Objective: This study aims to test the solubility efficiency of Shilajit in vitro for calcium oxalate renal stone. Material & Methods: A small stone was selected for the experiment. The weighed stone was suspended in 25 ml of aqueous extract of Shilajit for 72 hours with the interval of 24 hours. After each 24 hours, the stone was taken out, washed, dried and difference in weight was calculated. The whole procedure was carried out at room temperature. Results: It was found that the weight of the stone was reduced. Conclusion: Shilajit has the ability to dissolve the calcium oxalate renal stone.

Background and purpose: Cytokines regulate a broad range of biological and pathological processes. They play an important role in liver damage. Inflammatory and antioxidant factors are important causes of liver damage. Shilajit is known as an anti-inflammatory and antioxidant agent. Therefore, the main aim of this study was to determine the effect of Shilajit on levels of proinflammatory and anti-inflammatory cytokines in liver injury. Materials and methods: In this experimental study, 80 male Wistar rats were randomly divided into two main groups: A and B. Each group was organized into five subgroups: control, acetaminophen (A) or bile duct obstruction (BDL) (B), low dose, high dose, and vehicle. All subgroups except the controls in group A received 500 mg/ kg acetaminophen via IP injection, while BDL was performed in all subgroups in group B. Then in group A, two hours and in group B seven days after injury, the treatment subgroups with drug and vehicle received 150 mg/kg (low dose) and 250 mg/kg (high dose)shilajit and distilled water as vehicle, for one and six days, respectively. Objectives:Momiai (shilajit, mummy, mumie, or mineral pitch) has been used traditionally in different medical systems for the treatment of a variety of ailments since hundreds of years ago. It is a natural substance found in different rocky parts of the world, formed by plants, mineral, and animal remains gradually. There is also worthwhile evidence supporting its oral use for bone repair in Persian medicine. The aim of this study was to evaluate the efficacy and safety of momiai in tibia fracture healing. Design: This study is a randomized double-blinded controlled trial. Settings/Location: Three different hospitals in Tehran, Iran. Subjects: Patients with age range of 18–60 years admitted due to new tibia fracture were enrolled after meeting the inclusion criteria. Interventions: The patients were divided into two groups randomly and received two 500 mg capsules of momiai or placebo for 28 days. Outcome measures: The process of bone healing was assessed by frequent X-ray radiographies and adverse effects were recorded. Results: Totally, 160 patients participated in the study either in two equal intervention or placebo groups. There was no significant difference between groups in terms of demographic and descriptive data. At the end of the study, the mean time of tibial bone union was 129 days in the experimental group, while it was 153 days in the placebo group (p < 0.049). There was no significant difference in the reported adverse effects between the two groups (p = 0.839). Conclusions: The current study showed that oral consumption of momiai after tibial shaft fracture surgery could be a promising option to reduce the healing time. [ABSTRACT FROM AUTHOR]

Background: Non-alcoholic fatty liver disease (NAFLD) is the main common cause of chronic liver disease. The aim of this study is to evaluate the effect of Shilajit, a medicine of Ayurveda, on the liver damage caused by NAFLD. Materials and methods: Forty male Wistar rats, after being established as fatty liver models by feeding a high-fat diet (HFD, 12 weeks), were divided randomly into five groups as follows: control (standard diet), vehicle (HFD + distilled water), high-dose Shilajit (HFD + 250 mg/kg Shilajit), low-dose Shilajit (HFD + 150 mg/kg Shilajit) and pioglitazone (HFD + 10 mg/kg pioglitazone). The serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglycerides (TG), total cholesterol (TC), low-density lipoprotein (LDL), glucose and liver glutathione peroxidase (GPx), superoxide dismutase (SOD) activity, malondialdehyde (MDA) levels, liver weight, and histopathological manifestation outcomes were measured after the 2-week intervention. Results: Shilajit treatment significantly reduced the values of AST and ALT, TG, TC, LDL, glucose, liver weight, and steatosis, and instead, increased high-density lipoprotein (HDL) compared with the vehicle group (p < 0.05). Further, Shilajit treatment improved the adverse effects of HFD-induced histopathological changes in the liver as compared with the vehicle group (p < 0.001). MDA level and GPx activity increased but SOD activity decreased in the vehicle group compared with the control group (p < 0.05), while treatment with Shilajit restored the antioxidant/oxidant balance toward a significant increase in the antioxidant system in the Shilajit group (p < 0.05). Conclusions: These findings suggest that Shilajit improved the histopathological NAFLD changes in the liver and indicated the potential applicability of Shilajit as a potent agent for NAFLD treatment. The prepared magnetic hybrid material was fully characterized using different analysis, including Fourier transform infrared (FT-IR), X-ray diffraction (XRD), inductively coupled plasma (ICP), scanning electron microscopy (SEM), Energy-dispersive X-ray spectroscopy (EDX), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometer (VSM) thermal gravimetric analysis (TGA) and Brunauer–Emmett–Teller (BET). All these analysis revealed that during coating of Fe3O4@Sh using copper salt (II), synchronized redox sorption of CuII to CuI occurs at the same time as the oxidation of Fe3O4 to γFe2O3. This magnetic catalyst exhibited excellent catalytic activity for regioselective synthesis of 1,4-disubstituted-1,2,3-triazoles via one pot three-component click reaction of sodium azide, terminal alkynes and benzyl halides in the absence of any reducing agent. High yields, short reaction time, high turnover number and frequency (TON = 3.5 * 105 and TOF = 1.0 * 106 h−1 respectively), easy separation, and efficient recycling of the catalyst are the strengths of the present method. Shilajit is a natural material found mostly in the Himalayas (India), formed by the slow decomposition of certain plants by the action of microorganisms. It is an effective and extremely safe dietary supplement, and potentially able to prevent several diseases. Shilajit alcoholic extract was prepared by dissolving 10 grams of it in 200 mL of 95% ethanol. FTIR and other chemical procedures were used to detect the presence of Phenols, Alkaloids, Terpenoids, Tannins, Proteins Carbohydrates and, Steroids. Different concentrations of the extract were prepared and antimicrobial activity against several positive and negative gram stains, anti-protein denaturation and anti-blood cell hemolysis. Investigation results show that the nanoparticles were effective in inhibiting protein denaturation of albumin. Proteins denaturation refers the cause of inflammation, while the maximum inhibition of albumin was 87% observed at 300 μg/ml of Shilajit alcohol extract compared with aspirin at the same concentration. The results of inhibiting hemolysis at 300μg/ ml in comparing with standard drug Diclofenac sodium 300μg/ml resulted in good protection against the damaging effect of heat solution. Background: Shilajit is a nutraceutical exudate found mainly in the Indian Himalayas, which is formed for centuries by the gradual decomposition of certain plants due to microbial action. It has been effectively used as a potent and safe dietary supplement, restoring the energetic balance and potentially able to prevent several diseases. Hence, the aim of the present preliminary in vitro study is to assess the cytotoxicity of shilajit extract and to comparatively evaluate the anti-inflammatory effect of shilajit and diclofenac sodium, a commonly used drug. Materials and Methodology: Shilajit was commercially procured in pure powder form which was dissolved in 10 ml of methanol and boiled for 80 min, followed by centrifugation (2500 rpm) for 10 min. The supernatant fluid thus obtained was used as an experimental solution. 25-mg diclofenac sodium was used as a control drug. The cytotoxicity of shilajit extract was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, whereas the anti-inflammatory property of the test groups was comparatively evaluated using protein denaturation assay. Results: As the dilution of the extracts increased, the amount of cell viability was also increased, thereby showing that the diluted shilajit extract concentrations proved to be least cytotoxic. Different percentages of shilajit exhibited concentration-dependent inhibition of protein denaturation. Although shilajit extract exerted marginally better anti-inflammatory effect than diclofenac sodium, the effect being dose dependent; the protein inhibition values were not statistically significant (P > 0.05). Conclusion: Within the limitations of this in vitro preliminary study, it can be concluded that: (i) shilajit extract was found to be nontoxic when tested on L929 mouse fibroblast cell lines and (ii) the anti-inflammatory effect of shilajit was comparable to that of diclofenac sodium. Introduction: Cholestasis is a clinical condition caused by internal or extrahepatic disorder in bile secretion. Accumulation of bile acids plays an important role in inducing apoptosis, necrosis and increasing oxidative stress in liver. As a strong antioxidant and anti-inflammatory drug, shilajit can probably be effective in reducing some liver complications caused by bile duct obstruction. The aim of this study is to investigate some effects of shilajit on liver damage caused by extrahepatic cholestasis. Methods and Materials: Adult male Wistar rats were randomly assigned to one of five groups: sham, BDL, Vehicle (Salin), BDL+Shi (150 mg/kg), and BDL+Shi (250 mg/kg). On the sixth day after drug administration, blood samples were taken from animals under complete anesthesia for biochemical tests and liver samples were prepared for histopathological examinations. Results: The results showed that BDL induces a significant increase in liver enzymes [(Alkaline phosphatase (ALP), Aspartate aminotransferase (AST), Alanine aminotransferase (ALT), and oxidants levels [(Malondialdehyde (MDA) and Nitric Oxide (NO)] and decreases in antioxidant biomarkers [(superoxide dismutase (SOD), glutathione peroxidase (GPx)] in the plasma and liver tissue. We also found that Shi significantly preserved levels of these biomarkers and enzymatic activity. From the histopathological point of view, the amount of necrosis and liver damage shows a decreasing trend with receiving Shi. Conclusion: In the present study, liver damage was determined by measuring the serum parameters of ALT, AST, GST and its histopathological examination, and the administration of Shilajit with different doses showed its hepatoprotective effects by adjusting the serum parameters and improving the liver lesions caused by BDL. Background Shilajit is a safe, fluvic mineral complex exudate that is common to Ayurvedic medicine and is composed of fulvic acids, dibenzo-α-pyrones, proteins, and minerals. The purpose of this study was to examine the effects of 8 weeks of Shilajit supplementation at 250 mg·d− 1 (low dose) and 500 mg·d− 1 (high dose) versus placebo on maximal voluntary isometric contraction (MVIC) strength, concentric peak torque, fatigue-induced percent decline in strength, and serum hydroxyproline (HYP). Methods Sixty-three recreationally-active men ( \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$ \overline{X} $$\end{document} ± SD: 21.2 ± 2.4 yr.; 179.8 ± 6.3 cm; 83.1 ± 12.7 kg) volunteered to participate in this study. The subjects were randomly assigned to the high dose, low dose, or placebo group (each group: n = 21). During pre-supplementation testing, the subjects performed 2 pretest MVICs, 2 sets of 50 maximal, bilateral, concentric isokinetic leg extensions at 180°·s− 1 separated by 2-min of rest, and 2 posttest MVICs. Following 8 weeks of supplementation, the subjects repeated the pre-supplementation testing procedures. In addition, the groups were dichotomized at the 50th percentile based on pre-supplementation MVIC and baseline HYP. Mixed model ANOVAs and ANCOVAs were used to statistically analyze the dependent variables for the total groups (n = 21 per group) as well as dichotomized groups. Results For the upper 50th percentile group, the post-supplementation adjusted mean percent decline in MVIC was significantly less for the high dose group (8.9 ± 2.3%) than the low dose (17.0 ± 2.4%; p = 0.022) and placebo (16.0 ± 2.4%; p = 0.044) groups. There was no significant (p = 0.774) difference, however, between the low dose and placebo groups. In addition, for the upper 50th percentile group, the adjusted mean post-supplementation baseline HYP for the high dose group (1.5 ± 0.3 μg·mL− 1) was significantly less than both the low dose (2.4 ± 0.3 μg·mL− 1; p = 0.034) and placebo (2.4 ± 0.3 μg·mL− 1, p = 0.024) groups. Conclusions The results of the present study demonstrated that 8 weeks of PrimaVie® Shilajit supplementation at 500 mg·d− 1 promoted the retention of maximal muscular strength following the fatiguing protocol and decreased baseline HYP. Thus, PrimaVie® Shilajit supplementation at 500 mg·d− 1 elicited favorable muscle and connective tissue adaptations.
