The microstructure and materials performance of Laser Powder Bed Fusion-fabricated (LPBFed) NiTi shape memory alloys (SMAs) varies depending on the laser parameters and building directions. This study initially optimized the laser processing parameters (laser power and scanning speed) for LPBFed NiTi SMAs to enhance their printing quality and mechanical properties. We determined that with fixed hatch spacing of 80 μm and layer thickness of 30 μm, the laser power of 105 W, and scanning speed of 600 mm/s produced the best results. Subsequently, the corrosion behavior of LPBFed NiTi SMAs built in three different building directions (0°, 45°, 90°) was investigated in 0.9 wt% NaCl solution at 37 °C. The electrochemical data revealed that the corrosion current density (Icorr) decreased with increasing building direction angle (Icorr-90° < Icorr-45° < Icorr-0°). The corrosion resistance followed the order of 0° sample < 45° sample < 90° sample. These findings were attributed to the difference in the grain size and boundary density. Higher boundary densities enhanced the electron activity capacity and the element diffusion rates, facilitating the rapid formation of a protective film and thus improving the corrosion resistance. This new insight into the anisotropy of corrosion behavior relative to building directions can inform the design of NiTi-SMA products and improve their reliability in tissue engineering applications.

The purpose of the article: Polyarteritis nodosa (PAN) is a rare systemic necrotizing vasculitis preferentially targeting medium-sized arteries. PAN has two clinical entities: systemic PAN (sPAN) and cutaneous PAN (cPAN). cPAN is a skin-limited vasculitis, while ulcerative cPAN often predicts a higher risk of recurrence and a worse prognosis. However, there is still little experience and no consensus on cPAN treatment. Janus kinase-signal transducer and activator of transcription (JAK-STAT) inhibition with baricitinib (JAK1/JAK2) inhibitor in patients with inflammatory skin diseases has demonstrated good therapeutic outcomes in several clinical studies. However, the use of baricitinib in patients with cPAN has not been reported to date.Materials and methods: We report the first case of cPAN successfully treated with baricitinib. The patient had previously undergone various treatments, including corticosteroids and immunosuppressants, but these were continually switched due to inadequate efficacy or significant side effects.Results: Following treatment with baricitinib, the patient’s ulcers healed, subcutaneous nodules resolved, and livedo reticularis alleviated, resulting in a marked improvement in quality of life. No severe adverse reactions were observed during the treatment period. Over 1 year of follow-up, there was no recurrence of the rash.Conclusions: Our results support the evidence that baricitinib is a promising therapy for cPAN.

Abstract Wnt/β-catenin signaling is a critical pathway that influences development and therapeutic response of non-small cell lung cancer (NSCLC). In recent years, many Wnt regulators, including proteins, miRNAs, lncRNAs, and circRNAs, have been found to promote or inhibit signaling by acting on Wnt proteins, receptors, signal transducers and transcriptional effectors. The identification of these regulators and their underlying molecular mechanisms provides important implications for how to target this pathway therapeutically. In this review, we summarize recent studies of Wnt regulators in the development and therapeutic response of NSCLC.

In this work, wall-shaped NiTi components were fabricated by cold metal transfer (CMT) based wire arc additive manufacturing (WAAM) with optimized depositing speed. The microstructure, mechanical properties, and wear performance of the as-fabricated specimens along the building direction, together with their correlations were investigated in-depth. Coarse columnar grains were refined gradually with the increase of wall height. An equiaxed microstructure was obtained in the upper region. The Ni4Ti3 precipitates were distributed in the matrix. The microhardness, critical stress, and elongation increase monotonously with the increase of wall height. Plastic deformation, together with wear-induced work hardening is the main form of wear, which is mainly hindered by the superelasticity of NiTi alloys. Good homogeneity of the microstructure, mechanical properties, and wear resistance is obtained due to the CMT process with optimized depositing speed, indicating that this technique provides great potential to make novel NiTi flexible structures.

The cloud manufacturing system can provide consumers with on-demand manufacturing services, which significantly improve the utilization rate of distributed manufacturing resources and the response speed of personalized product needs. In the cloud manufacturing platform, the successful implementation of various industrial applications relies on the uploading and streaming of related field-level manufacturing data. For example, the realization of manufacturing service composition application should match the manufacturing tasks with distributed manufacturing resources according to their working state data and performance measurement data. Therefore, this paper proposes a data integration and analysis framework of a cloud manufacturing system based on cloud–edge collaboration and the Industrial Internet of Things (IIoT). A service-oriented information model is established to uniformly describe the related operational data and functional attributes of heterogeneous manufacturing resources. Secondly, a real-time transmission and integration method of high-volume operational field and sensor data based on message middleware is proposed to realize the remote monitoring of distributed manufacturing resources and efficient distribution of related data. Finally, a cloud–edge collaboration mechanism is put forward to train and update the parameters of various artificial intelligence models deployed at edge gateways. In the experiment, taking the computer numerical control (CNC) lathe as an example, the effectiveness of the proposed manufacturing resource access method is verified. Taking the fault diagnosis model of the CNC lathe as an example, the efficiency of the proposed cloud–edge collaboration mechanism for model updating is verified.

The pentose phosphate pathway (PPP) branches from glucose 6-phosphate (G6P), produces NADPH and ribose 5-phosphate (R5P), and shunts carbons back to the glycolytic or gluconeogenic pathway. The PPP has been demonstrated to be a major regulator for cellular reduction-oxidation (redox) homeostasis and biosynthesis. Enzymes in the PPP are reported to play important roles in many human diseases. In this review, we will discuss the role of the PPP in type 2 diabetes and cancer.

Vibrio alginolyticus is a major cause of Vibriosis in farmed marine aquatic animals and has caused large economic losses to the Asian aquaculture industry in recent years. Therefore, it is necessary to control V. alginolyticus effectively. The virulence mechanism of V. alginolyticus, the Type III secretion system (T3SS), is closely related to its pathogenicity. In this study, the T3SS gene tyeA was cloned from V. alginolyticus wild-type strain HY9901 and the results showed that the deduced amino acid sequence of V. alginolyticus tyeA shared 75–83% homology with other Vibrio spp. The mutant strain HY9901ΔtyeA was constructed by Overlap-PCR and homologous recombination techniques. The HY9901ΔtyeA mutant exhibited an attenuated swarming phenotype and an ~40-fold reduction in virulence to zebrafish. However, the HY9901ΔtyeA mutant showed no difference in growth, biofilm formation and ECPase activity. Antibiotic susceptibility test was observed that wild and mutant strains were extremely susceptible to Amikacin, Minocycline, Gentamicin, Cefperazone; and resistant to oxacillin, clindamycin, ceftazidime. In contrast wild strains are sensitive to tetracycline, chloramphenicol, kanamycin, doxycycline, while mutant strains are resistant to them. qRT-PCR was employed to analyze the transcription levels of T3SS-related genes, the results showed that compared with HY9901 wild type, ΔtyeA had increased expression of vscL, vscK, vscO, vopS, vopN, vscN, and hop. Following vaccination with the mutant strain, zebrafish had significantly higher survival than controls following infection with the wild-type HY9901 (71.2% relative percent survival; RPS). Analysis of immune gene expression by qPCR showed that vaccination with HY9901ΔtyeA increased the expression of IgM, IL-1β, IL-6, and TNF-α in zebrafish. This study provides evidence of protective efficacy of a live attenuated vaccine targeting the T3SS of V. alginolyticus which may be facilitated by up-regulated pro-inflammatory and immunoglobulin-related genes.

[Objective] To clone araC gene of Vibrio alginolyticus HY9901 strain, and analyze bioinformatics. [Methods] the whole genome sequence of Vibrio alginolyticus on GenBank was used to design specific primers. According to the principle of PCR amplification sequence, the target gene araC was amplified, and then the sequence was further analyzed by bioinformatics method to establish the phylogenetic tree of araC gene and its corresponding subunit three-dimensional structure model. Results] Sequence analysis revealed araC gene is 711 bp and encodes. a putative protein of 236 amino acids. The predicted molecular mass of AraC was 26. 92 ku. Using Signal P 4.0 and TMHMM Server 2.0 soft ware for analysis, it was predicted that the AraC protein did not contain a signal peptide or a transmembranous region. The AraC protein had two cAMP and CGMP dependent protein kinase phosphorylation site, five protein kinase C phosphorylation sites, three casein kinase Il phos phorylation sites, one prenyl group binding site (CAAX box) and five microbodies C-terminal targeting signal. The predicted results of protein subcellular localization showed that AraC was located in the mitochondria, nucleus and cytoplasm. Its protein is unstable and hydrophilic. The AraC protein is a transcriptional regulatory protein which belongs to HTH 18 superfamily. According to the prediction, secondary structure: a-helix (Alpha helix) accounted for 52. 12%, random coil (31. 78%), extended strand (11.02%), b-fold (Beta turn) accounted for 5.08%. V. alginolyticus, Vibrio parahaemolyticus and Vibrio palustris were clustered together, which implies that the genetic relationship be tween these three species was the closest. [Objectives] The purpose was to investigate the function and mechanism of exsA gene in type III secretion system of Vibrio alginolyticus. [Methods] The full length of exsA was cloned using molecular biology techniques to analyze its biological information. Fluorescence quantitative PCR technology was used to analyze the expression of exsA after different media stress. [Results] The exsA gene contains an open reading frame (ORF) of 861 bp, encoding 286 amino acids. The physico-chemical analysis shows that the molecular structural formula is C1442H2267N393O441S12, the theoretical molecular weight is 32.549 kD, the theoretical pl value is 6. 0, and the protein is non-hydrophilic and unstable. The gene does not contain a transmembrane region, and there is no obvious signal peptide. The prediction result of protein subcellular localization shows that the protein is inside the cell. The deduced amino acid sequence and constructed phylogenetic tree show that V. alginolyticus has a close relationship with Vibrio antiquarius. The qPCR results show that the expression level of exsA in different media is different, highest in TSB medium containing bile salts, followed by DMEM medium, and lowest in ordinary TSB medium. Objectives: To clone the pepck gene of Vibrio alginolyticus strain HY9901 and analyze its sequence by bioinformatics. Methods: According to the complete gene sequence of V alginolyticus on GenBank, specific primer were designed to amplify the target gene ppppC by PCR. The sequence of the pepck gene was analyzed using bioinformatics. The phylogenic tree of pepck gene and the corresponding single -subunit three-dimensional structure were constructed. & Results: The pepck gene of V alginolyticus strain HY9901 has a full length of 1 629 bp, with theoretical molecular weight of 60.12 kD. The prediction results show that there is no signal peptide or transmembrane region at the N-terminus of the sequence, the amino acid sequence contains 11 phosphorylation sites of casein kinase II. The prediction results of protein subcellular localization indicate that PEPEK protein is localized in the cytoplasm. The protein is stable and hydrophobic. The tertiary structure of the PEPCK protein of V alginolyticus is similar to that of Vibbo perahaemolgtcus. It is predicted that PEPCK has a major functional domain PEPCK_ATP. In the secondary structure, alpha helix, random coil, and extended strand accounted for 21. 96%, 52. 03% and 26.01%, respectively. The PEPCK homology between V. alginolyticus ad Vibrio diabbllcus is as high as 99%. The tertiary structure of Tye A protein of V. alginolyticus is similar to that of Yersinia sp. According to prediction, Tye A has a major functional domain Pfam. In terms of secondary structure, alpha helix, random coil, extended strand and beta turn account for 85.11%, 7.45%, 4.26% and 3.19%, respectively. The homology of Tye A between V. alginolyticus and Vibrio parahaemolyticus is up to 83%, so they are classified into one cluster. [Objectives] This study aimed to analyze the biological activity and immunogenicity of extracellular products from Streptococcus iniae. [Methods] S. iniae was incubated with brain heart infusion agar medium (BHIA + 4% calf serum) for 60 h. The bacterial liquid was rinsed with PBS, centrifuged, and filtered through microporous filtering film to collect extracellular products (ECPs). [Results]Extracellular proteinase (ECPase) of S. iniae exhibited amylase, protease, lecitinase, gelatinase, lipase activities and hemolytic activity but had no urease activity. EDTA, DTT and PMSF could reduce ECPase activity to 72.4%, 77.6% and 72.4%, respectively. Cu2+, Ca2+, K+ and Mg2+ exhibited an inhibitory effect on ECPase activity, whereas Fe+, Co2+ and Mn2+ could activate ECPase activity. ECPs had good heat stability and exhibited relatively high activities under alkaline conditions. The optimal temperature for ECPs was 55t. Two-dimensional electrophoresis was performed to analyze the main protein of ECPs. The results indicated that there are 12 main bands of ECPs, and the molecular weights mainly ranged between 28 - 68 kDa. About 120 protein spots were detected, and the molecular weights mainly ranged between 26 - 95 kDa. The mouse anti-S. iniae was used for Western-blot analysis of ECPs, and the results showed that there were four proteins, with molecular weights of 26, 37, 95, and 97 kDa, respectively. The pathogenicity assay indicated that ECPs of S. iniae were highly pathogenic to tilapia. The mortality rate of tilapia was enhanced as the concentration of ECPs increased. The article presents a study which examines the neuroprotective effect from the exertion of the Eleutheroside B against 1-methyl-4- phenylpyridinium ion (MPP+) apoptosis in PC12 cells. The viability of PC12 cells and apoptosis were monitored by flow cytometry, where the release of lactate dehydrogenase and malondialdehyde were determined by ultraviolet spectrophotometry. Result shows that the anti-oxidative stress capacity of cells has increased and intracellular calcium has reduced.
