Abstract In the last few decades, researchers have thoroughly studied the use of plants in Palestine, one of them is Cyclamen persicum Mill. (C. persicum). Cyclamen persicum has been historically cultivated since the 1700s due to its tuber. The tuber is known to stimulate the nasal receptors, thus triggering the sensory neurons. Cyclamen persicum has anti-inflammatory effects, reduces cholesterol levels, treats diabetes, and inhibits tumor growth. In this respect, in-vitro examination of antibacterial and anticancer activities and antioxidative potency of C. persicum ethanolic extract were evaluated. The antioxidative potency of the extracted plant material was determined spectrophotometrically using the DPPH free radical scavenging method and the HPLC–PDA method to evaluate its total phenolic content (TPC) and total flavonoid content (TFC). The experimental results demonstrated limited antibacterial activity of C. persicum extract against both gram negative (E. coli) and gram positive (Streptococcus and S. aureus) bacterial strains, with the observed zones of inhibition measuring less than 8 mm. On the other hand, powerful activity against MCF7 breast cancer as well as HT29 colon cancer cell lines was obtained. The findings also revealed potent inhibition of free radicals and the presence of maximal levels of natural products such as phenolic compounds and flavonoids, which supportits biological activities and powerful ability to scavenge free radicals. HPLC results showed the presence of numerous flavonoid and phenolic compounds such as rutin, chlorogenic acid, kaempferol, trans-cinnamic acid, quercetin, sinapic acid, and p-coumaric acid.

Abstract The utilization of nanotechnology and biotechnology for enhancing the synthesis of plant bioactive chemicals is becoming increasingly common. The hairy root culture technique can be used to increase secondary metabolites such as tropane alkaloids. Agrobacterium was used to induce hairy roots from various explants of Hyoscyamus muticus. The effect of nano-silver particles (AgNPs) at concentrations of 0, 25, 50, 100, and 200 mg/L on tropane alkaloids synthesis, particularly hyoscyamine and scopolamine, was studied in transgenic hairy root cultures. Different types of explants obtained from 10-day-old seedlings of H. muticus were inoculated with two strains of Agrobacterium rhizogenes (15,834 and A4). The antimicrobial activity of an ethanolic extract of AgNPs-induced hairy root cultures of H. muticus was tested. The frequency of hairy roots was higher in hypocotyl, root, leaf, and stem explants treated with A. rhizogenes strain A4 compared to those treated with strain 15,834. In transgenic hairy root cultures, AgNPs application at a concentration of 100 mg/L resulted in the highest total tropane alkaloid production, which exhibited broad-spectrum antimicrobial activity. The study demonstrated the potential of nano-silver as an elicitor for promoting the production of target alkaloids in Hyoscyamus muticus hairy root cultures, which exhibit high biological activity.

Abstract Agriculture, the main water-consuming factor, faces a global water scarcity crisis. Saline water is an alternative water source, while excess NaCl decreases plant growth and productivity of crops. L-cysteine (Cys) is a promising thiol amino acid in plant growth and development. Flax; Linum usitatissimum L. is an economical plant with low salt tolerance. NaCl salt stress at 50 and 100 mM inhibited the growth parameters, the photosynthetic pigments, total soluble sugars, total phenols, and amino nitrogen in flax plants. Salt stress led to a marked rise in proline and lipid peroxidation and altered the protein profile. Foliar application of cysteine at 0.8 and 1.6 mM mitigates the unfriendly effects of NaCl stress on flax plants. Cysteine enhanced the growth traits, photosynthetic pigments, amino nitrogen, total phenols, and new polypeptides in NaCl-stressed plants. However, cysteine declined the total sugars, proline, the activity of peroxidase, and ascorbate peroxidase. The results confirmed that cysteine had reductant properties. Furthermore, it decreased the NaCl oxidative stress and maintained the stability of membranes by lowering lipid peroxidation. Overall, the redox capacity of L-cysteine is the cause behind its potential counteracting of the adverse effects of NaCl toxicity on the growth of flax plants.

Leishmaniasis, one of the most neglected tropical diseases (NTDs), is the third most important vector-borne disease worldwide. This disease has a global impact and severity of the infection and is greatest in the Middle East. The agent of infection is a protozoan parasite of the genus, Leishmania, and is generally transmitted by blood-sucking female sandflies. In humans, there are three clinical forms of infection: (1) cutaneous (CL), (2) mucocutaneous (ML), and (3) visceral leishmaniasis (VL). This review aims to discuss the current epidemiological status of leishmaniasis in Saudi Arabia, Iraq, Syria, and Yemen with a consideration of treatment options. The elevated risk of leishmaniasis is influenced by the transmission of the disease across endemic countries into neighboring non-infected regions.

Background: Crataegus aronia (C. aronia) extracts have been used medicinally since ancient times and are often utilized in traditional Arab medicine. An extensive study has revealed that Crataegus species have antioxidant, antibacterial, anti-inflammatory, and hypotensive properties. Objectives: This work was performed to explore the phytochemical contents of C. aronia extract, as well as its antioxidant and antibacterial properties, and to assess the lipid peroxidation level as an oxidative stress biomarker in erythrocytes. Methods: Chemical constituents in the methanolic extract of C. aronia were identified by gas chromatography-mass spectrometry and their relative concentrations were determined. The antioxidant activity of C. aronia extract was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. The effect of C. aronia on the concentration of malondialdehyde (MDA) in the erythrocyte hemolysates was studied. Also, the crude extract was assessed for its antimicrobial activity through agar diffusion and microbroth dilution assays. Key findings: The DPPH IC50 value of the extract showed that the antioxidants activity was equal to (14.3 μg/mL) and according to FRAP assay, the antioxidant activity was in the range of 33.9 μmol–82.86 μmol Fe+2/g dw. The extract exerts a protective effect against oxidative stress in RBCs and shows a 50% inhibition of malonyldialdehyde (MDA) at 39.48 μg/mL extract. Minimum inhibitory concentrations were found in the range of 800–1000 μg/mL of leave extracts. The phytochemical analysis showed that the total phenols, flavonoids, and flavonols content were 494.071 mg GAE/g extract, 155.251 mg RE/g extract, and 103.2049 mg RE/g extract). C. aronia extract contains alkaloids, flavonoids, terpenoids, and steroids. Crude extract of C. aronia was more potent in inhibiting the growth of B. subtilis, S. aureus and M. luteus with MIC and MBC values of 800,800 and 1000 μg/mL, respectively. According to GC-MS, 20 compounds were identified: dihydro-3-methylene-5-methyl-2-furanone (14.71%), hexanoic acid (6.57%), ethyl 3,5-ditert-butyl-4-hydroxybenzoate (6.4%), N, N-dimethylheptadecan-1-amine (4.91%), methyl 2-oxobutanoate (4.14%), glyceraldehyde (3.98%), and 2-methoxy-1-(2-nitroethenyl)-3-phenylmethoxybenzene (3.16%), were the major constituents. Conclusion: This study may open a window of hope for children with Glucose-6-phosphate dehydrogenase disorder by possible utilization of the active ingredients of C. aronia to minimize both oxidative stress and infection which negatively impact the disease sequelae.According to these in vitro experiments, this plant extract has a significant amount of natural antioxidants, which may aid in the protection of various oxidative stresses. As a result, employing the active components of C. aronia to minimize oxidative stress and infection, both of which have a detrimental impact on disease sequelae, may bring hope to children with Glucose-6-phosphate dehydrogenase disorder.

Nanotechnology is one of the most dynamic research areas and the fastest-growing market. Developing eco-friendly products using available resources to acquire maximum production, better yield, and stability is a great challenge for nanotechnology. In this study, copper nanoparticles (CuNP) were synthesized via the green method using root extract of the medical plant Rhatany (Krameria sp.) as a reducing and capping agent and used to investigate the influence of microorganisms. The maximum production of CuNP was noted at 70 °C after 3 h of reaction time. The formation of nanoparticles was confirmed through UV-spectrophotometer, and the product showed an absorbance peak in the 422–430 nm range. The functional groups were observed using the FTIR technique, such as isocyanic acid attached to stabilize the nanoparticles. The spherical nature and average crystal sizes of the particle (6.16 nm) were determined using Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM), and X-ray diffractometer (XRD) analysis. In tests with a few drug-resistant pathogenic bacteria and fungus species, CuNP showed encouraging antimicrobial efficacy. CuNP had a significant antioxidant capacity of 83.81% at 200 g/m−1. Green synthesized CuNP are cost-effective and nontoxic and can be applied in agriculture, biomedical, and other fields.

Drought stress is a significant environmental variable affecting wheat growth and development. Plant stress tolerance is intimately related to growth regulators of plants as polyamines. The study assessed the impact of drought (50% water irrigation and 100% water irrigation), priming of grains in putrescine (0.25, 0.5, and 1 mM), and their interactions on the growth, yield, and physiological attributes of wheat plants. Drought conditions declined plant height, fresh and dry weights, leaves and tillers numbers, and flag leaf area. However, applying putrescine, especially at (1 mM), enhanced wheat growth performance in normal or water-deficit conditions. Drought stress decreased spike length (28.6%), number of spikelets (15.6%), number of grains (30.3%), the weight of the spike (23.5%), and the weight of the grains/spike (37.5%). In addition, drought decreased the contents of chlorophyll a, chlorophyll b, free amino acids, and total phenols, while applying putrescine enhanced wheat plant growth performance in normal or drought conditions. Putrescine at (1 mM) achieved the highest increase in plant height (38.8%), root length (50%), leaves number (166%), tillers number (80%), flag leaf area (70.3%), shoot fresh weight (99.4%), shoot dry weight (98.4%), root fresh weight (97.8%), root dry weight (210%) compared to the untreated plants. Moreover, pretreatment with putrescine improved chlorophyll a (13.3%), chlorophyll b (70.3%), carotenoids (61.8%), soluble sugars (49.1%), amino acids (42.7%), phenols (52.4%), number of spikelets (59.3%), number of grains (81.1%), and weight of spike (45.4%). Moreover, variations in the protein profile of wheat plants were due to drought conditions and putrescine application. In conclusion, priming wheat grains with putrescine effectively induces protective mechanisms against water stress and improves wheat plants’ physiological attributes and yield components.

Ionizing radiation is abiotic stress limiting the growth and productivity of crop plants. Stigmasterol has positive effects on the plant growth of many crops. The role of stigmasterol in alleviating the effects of ionizing radiation on plant metabolism and development is still unclear. Therefore, the study aimed to investigate the effects of pretreatments with γ-radiation (0, 25, and 50 Gy), foliar application of stigmasterol (0, 100, and 200 ppm), and their interaction on the growth, and biochemical constituents of wheat (Triticum aestivum L., var. Sids 12) plants. Gamma radiation at 25 Gy showed no significant difference in plant height, root length, no. of leaves, shoot fresh weight, root fresh weight, Chl a, ABA, soluble phenols, and MDA compared to the control values. Gamma rays at 50 Gy inhibited shoot and root lengths, flag leaf area, shoot fresh and dry weights, photosynthetic pigments, total soluble sugars, proline, and peroxidase activity. However, it stimulated total phenols, catalase activity, and lipid peroxidation. On the other hand, stigmasterol at 100 ppm showed no significant effects on some of the physiological attributes compared to control plants. Stigmasterol at 200 ppm improved plant growth parameters, photosynthetic pigments, proline, phenols, antioxidant enzyme, gibberellic acid, and indole acetic acid. Correspondingly, it inhibited total soluble sugars, abscisic acid, and lipid peroxidation. Moreover, the application of stigmasterol caused the appearance of new polypeptides and the reappearance of those missed by gamma radiation. Overall, stigmasterol could alleviate the adverse effects of gamma radiation on wheat plants.

Drought is the main limiting abiotic environmental stress worldwide. Water scarcity restricts the growth, development, and productivity of crops. Wheat (Triticum aestivum L.) is a fundamentally cultivated cereal crop. This study aimed to evaluate the effect of grain-priming with arginine (0.25, 0.5, and 1 mM) on growth performance and some physiological aspects of wheat plants under normal or drought-stressed conditions. Morphological growth parameters, photosynthetic pigments, soluble sugars, free amino acids, proline, total phenols, flavonoids, and proteins profiles were determined. Drought stress lowered plant growth parameters and chlorophyll a and b contents while increasing carotenoids, soluble sugars, free amino acids, proline, total phenols, and flavonoids. Soaking wheat grains with arginine (0.25, 0.5, and 1 mM) improves plant growth and mitigates the harmful effects of drought stress. The most effective treatment to alleviate the effects of drought stress on wheat plants was (1 mM) arginine, that increased root length (48.3%), leaves number (136%), shoot fresh weight (110.5%), root fresh weight (110.8%), root dry weight (107.7%), chlorophyll a (11.4%), chlorophyll b (38.7%), and carotenoids content (41.9%) compared to the corresponding control values. Arginine enhanced the synthesis of soluble sugars, proline, free amino acids, phenols, and flavonoids in wheat plants under normal or stressed conditions. Furthermore, the protein profile varies in response to drought stress and arginine pretreatments. Ultimately, pretreatment with arginine had a powerful potential to face the impacts of drought stress on wheat plants by promoting physiological and metabolic aspects.

Different species belonging to the genus Nephthea (Acyonaceae) are a rich resource for bioactive secondary metabolites. The literature reveals that the gastroprotective effects of marine secondary metabolites have not been comprehensively studied in vivo. Hence, the present investigation aimed to examine and determine the anti-ulcer activity of 4α,24-dimethyl-5α-cholest-8β,18-dihydroxy,22E-en-3β-ol (ST-1) isolated from samples of a Nephthea species. This in vivo study was supported by in silico molecular docking and protein–protein interaction techniques. Oral administration of ST-1 reduced rat stomach ulcers with a concurrent increase in gastric mucosa. Molecular docking calculations against the H+/K+-ATPase transporter showed a higher binding affinity of ST-1, with a docking score value of −9.9 kcal/mol and a pKi value of 59.7 nM, compared to ranitidine (a commercial proton pump inhibitor, which gave values of −6.2 kcal/mol and 27.9 µM, respectively). The combined PEA-reactome analysis results revealed promising evidence of ST-1 potency as an anti-ulcer compound through significant modulation of the gene set controlling the PI3K signaling pathway, which subsequently plays a crucial role in signaling regarding epithelialization and tissue regeneration, tissue repairing and tissue remodeling. These results indicate a probable protective role for ST-1 against ethanol-induced gastric ulcers.
