The study of the Red Palm Weevil (RPW), Rhynchophorus ferrugineus (Olivier), as an invasive pest of palm trees. In this study, 36 RPW individuals were collected from 6 different locations in Egypt. The presences of endosymbionts in the RPW individuals were assayed. The phylogenetic analysis of the RPW inhabiting Egypt was conducted using Cytochrome c oxidase sub-unit 1 (CO1) gene. Spiroplasma was found, for the first time, in all individuals, while Rickettsia was found, for the first time, in individuals collected from only 3 of the 6 locations. Endosymbionts harbouring Egyptian RPW were closely related to those harbouring Diptera and\or Trombidiformes associated with palm trees. This may be due to horizontal transmission through palm sap or through ectoparasites living on the RPW. Finally, the phylogenetic analysis of the RPW inhabiting Egypt was conducted. The collected individuals were closely related to Saudi Arabia specimens collected from the eastern region. Thus, Saudi Arabia may be the origin of the RPW which invaded Egypt. Individuals from populations inhabiting the same geographical locations were closely related. This may be due to secondary invasion incidents that may have taken place through transportation of infested date palm trees and offshoots from infected to uninfected locations. This study reports the first incidence for the presence and coexistence of Spiroplasma and Rickettsia in RPW collected from Egypt. In addition, it was found that the collected individuals of RPW were closely related to a Saudi haplotype. Thus, Saudi Arabia may be the origin of infection which invaded Egypt.

Cyclic dipeptides, also know as diketopiperazines (DKP), the simplest cyclic forms of peptides widespread in nature, are unsurpassed in their structural and bio-functional diversity. DKPs, especially those containing proline, due to their unique features such as, inter alia, extra-rigid conformation, high resistance to enzyme degradation, increased cell permeability, and expandable ability to bind a diverse of targets with better affinity, have emerged in the last years as biologically pre-validated platforms for the drug discovery. Recent advances have revealed their enormous potential in the development of next-generation theranostics, smart delivery systems, and biomaterials. Here, we present an updated review on the biological and structural profile of these appealing biomolecules, with a particular emphasis on those with anticancer properties, since cancers are the main cause of death all over the world. Additionally, we provide a consideration on supramolecular structuring and synthons, based on the proline-based DKP privileged scaffold, for inspiration in the design of compound libraries in search of ideal ligands, innovative self-assembled nanomaterials, and bio-functional architectures.

Staphylococcus aureus (S. aureus) is one of the pathogens that may cause a wide range of infections in humans. Clindamycin has been considered as an important medication in treating these infections especially skin and soft tissue infections. Inducible clindamycin resistance (ICR) is a complication in treating S. aureus infections in humans. It is difficult to detect S. aureus strains expressing ICR by using standard susceptibility test methods. Recently, there is an increasing need to find alternatives to antibiotics. Nanoparticles, especially silver nanoparticles, were reported as potential alternatives for traditional antibiotics (or in combination with traditional antibiotics) against the emergence of bacterial multidrug resistance. This research aimed to study the prevalence of ICR among S. aureus clinical isolates and investigate the antibacterial effects of AgNPs solely and in combination with clindamycin against these isolates with evaluation of the acute toxicity of intraperitoneal administrated silver nanoparticles (AgNPs). Out of one hundred isolates of S. aureus studied over a period of one year, 70 isolates were identified as MRSA and 30 isolates were MSSA. Results revealed that the percentage of cMLSB, iMLSB, and MS phenotypes were 40%, 10% and 9% respectively. Overall, 41% isolates of S. aureus showed susceptibility to Erythromycin. Both iMLSB and cMLSB phenotypes were the most predominated among MRSA isolates. AgNPs was found to have strong antibacterial effect with MIC of 1μg/ml and partially synergistic activity with clindamycin towards S. aureus. Intraperitoneal administration of AgNPs was found to be moderately toxic.

Background and study aims Diarrhoea and urinary tract infection (UTI) are common clinical problems. Meanwhile, Escherichia coli (E. coli), is the commonest bacterial pathogen reported in both of them. This study aimed to evaluate the pathogenic E. coli (PEC) in stool of acute diarrhoea and urine of UTI regarding their virulence genes and their influence on the susceptibility to routinely prescribed antibiotics. Patients and methods Twenty two stool and another 22 urine samples of patients with acute diarrhoea and UTI respectively were collected from patients admitted at Kasr Al-Ainy Hospital, Faculty of Medicine, Cairo University, Egypt. E. coli isolation, identification of their phyla; chuA, yjaA, and TspE4.C2, and further identification of 10 virulent genes; fimH, papC, papG//, papG///, papEF, afa, sfa, CNF1, iroN & hlyA was performed. Antibiotic susceptibility was studied against quinolones , gentamicin (GM), and trimethoprim-sulphamethoxazole (TMP-SMX). Results The studied virulence genes were comparably detected in both pathogenic samples. In diarrheogenic E. coli (DEC); phylum A was significantly related to both ciprofloxacin (CIP) and TMP-SMX resistance, and both of the virulence genes fimH and iroN were significantly related to all the studied antibiotics resistance, while afa was significantly related to nalidixic acid (NA) resistance. In uropathogenic E. coli (UEC); phylum D was significantly related to CIP and levofloxacin resistance, and both of the virulence genes fimH and iroN were significantly related to most of the studied antibiotics resistance. Conclusion The isolated PEC was evidently and broadly resistant to the studied antibiotics, with limited influence of their phyla and virulence genes (fimH and iroN).

Introduction Serratia marcescens is a significant hospital-acquired pathogen, and many outbreaks of S. marcescens infection have been reported in neonates. We report a sudden breakout of S. marcescens harboring the bla IMP-4 and bla VIM-2 metallo-β-lactamase (MBL) genes that occurred from March to August 2015 in the neonatal intensive care unit of Cairo University Hospital, Cairo, Egypt. Methods During the study period, 40 nonduplicate clinical isolates of S. marcescens were collected from blood culture samples. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was used to identify each isolate. Then, minimum inhibitory concentrations of different antibiotics were assessed by the Vitek 2 compact system. Screening of the MBL genes bla IMP, bla VIM, bla SIM-1, bla SPM-1, and bla GIM-1 as well as the carbapenemase genes KPC, NDM, OXA-48, SME-1, and SME-2 were evaluated. Pulsed field gel electrophoresis was preformed to detect the genetic relationship of the isolates. Results Analysis showed that 37.5% of the S. marcescens clinical isolates were resistant to meropenem (minimum inhibitory concentrations ≥ 2 µg/mL), and bla IMP-4 and bla VIM-2 were the most prevalent MBL genes (42.5% and 37.5%, respectively). None of the other investigated genes were observed. Pulsed field gel electrophoresis typing revealed two discrete clones; 33/40 (82.5%) were pulsotype A and 7/40 (17.5%) were pulsotype B. Conclusion Here, we report for the first time the detection of MBL-producing S. marcescens isolates, particularly IMP-4 and VIM-2 recovered from inpatients with bacteremias from the intensive care unit at Cairo University Hospital.

Peptides are fragments of proteins that carry out biological functions. They act as signaling entities via all domains of life and interfere with protein-protein interactions, which are indispensable in bio-processes. Short peptides include fundamental molecular information for a prelude to the symphony of life. They have aroused considerable interest due to their unique features and great promise in innovative bio-therapies. This work focusing on the current state-of-the-art short peptide-based therapeutical developments is the first global review written by researchers from all continents, as a celebration of 100 years of peptide therapeutics since the commencement of insulin therapy in the 1920s. Peptide “drugs” initially played only the role of hormone analogs to balance disorders. Nowadays, they achieve numerous biomedical tasks, can cross membranes, or reach intracellular targets. The role of peptides in bio-processes can hardly be mimicked by other chemical substances. The article is divided into independent sections, which are related to either the progress in short peptide-based theranostics or the problems posing challenge to bio-medicine. In particular, the SWOT analysis of short peptides, their relevance in therapies of diverse diseases, improvements in (bio)synthesis platforms, advanced nano-supramolecular technologies, aptamers, altered peptide ligands and in silico methodologies to overcome peptide limitations, modern smart bio-functional materials, vaccines, and drug/gene-targeted delivery systems are discussed.

It is debatable whether an anterior oblique fracture orientation is really a contraindication to anterior odontoid screw fixation. The purpose of this study was to investigate the feasibility of anterior odontoid screw fixation of type II and rostral shallow type III fracture with an anterior oblique fracture orientation.The authors evaluated 16 patients with type II and rostral shallow type III odontoid fracture with an anterior oblique fracture orientation. Of these 16 patients, 8 (group 1) were treated by anterior odontoid screw fixation, and 8 (group 2) by a posterior C1-2 arthrodesis.Of the 8 patients in group 1, seven patients achieved solid bone fusion (87.5%), and one experienced screw back-out of the C-2 body two months after anterior screw fixation. All patients treated by posterior C1-C2 fusion in group 2 achieved successful bone fusion. Mean fracture displacements and fracture gaps were not significantly different in two groups. (p=0.075 and 0.782). However, mean fracture orientation angles were 15.3±3.2 degrees in group 1, and 28.6+8.1 degrees in group 2 (p=0.002), and mean fragment angulations were 3.2±2.1 degrees in group 1, and 14.8±6.7 degrees in group 2 (p=0.001).Even when the fracture lines of type II and rostral shallow type III fractures are oriented in an anterior oblique direction, anterior odontoid screw fixation can be feasible in carefully selected patients with a relatively small fracture orientation angle and relatively small fragment angulation.

COVID-19 is currently considered as a life-threatening pandemic viral infection. Finding an antiviral drug or a vaccine is the only route for humans' survival against it. To date, no specific antiviral treatment has been confirmed. Antimicrobial peptides (AMPs) have been widely regarded as a promising solution to combat harmful microorganisms. They are biologically active molecules produced by different organisms as an essential component of their innate immune response against invading pathogens. Lactoferrin (LF), one of the AMPs, is an iron-binding glycoprotein that is present in several mucosal secretions. The antiviral activity of LF exists against a wide range of human and animal viruses (DNA and RNA). LF was proven to increase the host immunity against viral infection. Since LF is one of the constituents of breast milk and significantly located at the mucosal layers of the human body, it is considered the first line of defense against microbial infection. LF was reported to have antiviral activity against SARS-CoV infection. The significant antiviral activity of LF makes it a potential option as an immunity enhancer, a drug or a drug conjugate with conventional antivirals. The affordability, environmental safety, and efficiency of LFs will make them superior to all other control strategies.

BACKGROUND The increasing incidence of hospital-acquired infections due to extended-spectrum β-lactamase (ESBL)-producing Enterobacteriaceae represent a major health problem because of few therapeutic alternatives. The fecal flora can represent a reservoir for ESBL genes. Integrons are genetic structures capable of capturing gene cassettes that usually encode antibiotic-resistance determinants. OBJECTIVES To investigate the antimicrobial susceptibility of fecal isolates of ESBL-producing Escherichia coli and Klebsiella pneumoniae from hospitalized and nonhospitalized Egyptian patients and to determine the prevalence of class 1 and class 2 integrons together with the most common ESBL-producing genes (bla TEM, SHV, CTX-M, and OXA) among the collected isolates. MATERIALS AND METHODS Ninety-six fecal samples were collected: 48 samples from hospitalized patients admitted at Kasr Al-Ainy University Hospital, Cairo and 48 from outpatient clinics. Samples were inoculated on MacConkey agar and identified. All isolates were tested for their susceptibility to different antimicrobial agents using a standard disk diffusion method. The double-disk synergy test was applied for screening ESBL. All ESBL-producing isolates were confirmed by molecular testing to detect ESBL-encoding genes (SHV, TEM, CTX-M, and OXA). To identify the strains carrying integrons 1 and 2, the conserved regions of integron-encoded integrase gene intI1 and intI2 were amplified. RESULTS E. coli isolates accounted for 52.1% of the isolates collected from hospitalized patients and 60.4% of those collected from outpatient clinics. Results of the double-disk synergy test were positive in all E. coli and K. pneumoniae isolates, indicating the presence of ESBL production. Isolates of both groups showed variably high degrees of resistance to ciprofloxacin and co-trimoxazole. The most predominant ESBL gene in both groups was the bla CTX-M gene (93.8%) and the least prevalent was the bla OXA gene, which was not detected in any of the study isolates. Between the other two genes, the bla TEM gene was more common than the bla SHV gene in the two study groups. Class 1 integron was more prevalent among hospitalized patients, being detected in 64.6% of isolates from this group. Class 1 integron was linked with the bla CTX-M gene (P=0.039). In this study, we report a novel approach for preparing precipitated calcium carbonate for the first time by Bacillus licheniformis using different types of media containing urea as a source of urease enzyme and for the first time using particles of pure calcium. The bacteria used in this study produced urease which catalyzes the hydrolysis of urea (CO(NH 2 ) 2 ) into ammonium (NH 4 ) and carbonate (CO 3 2− ) leading to the precipitation of calcium carbonate. Calcium carbonate precipitation was experimentally studied by spontaneous precipitation at various pH (8–12), temperatures (30, 35, 40, 45, 50 °C), and at different concentrations of urease enzyme (1, 5, 10 mg mL −1 ). The XRD results showed a mixture of calcite and vaterite. The morphology of calcium carbonate particles prepared was studied under SEM after using it as a bioconsolidation of degraded fresco wall paintings.

Bacterial-induced mineralization has been explored for protection and consolidation of degraded limestone, concrete and plaster by precipitation of calcium carbonate. It is the first time that Bacillus sphaericus was used for consolidating the nonsterilized decayed wall paintings samples by immersing them in sterile nutritional media. The B. sphaericus used in this study produced urease, which catalyzes the hydrolysis of urea (CO(NH2)2) into ammonium (NH4) and carbonate (CO3−2) leading to the precipitation of calcium carbonate. The effect of B. sphaericus on wall paintings was determined by recording the evolution of culture media chemistry and examining the treated wall paintings under a scanning electron microscope to show the structural and morphological evolution of calcium carbonate that was investigated in wall paintings models.

Male-killing is 1 of 4 known strategies that inherited parasitic endosymbionts have evolved to manipulate their host's reproduction. In early male-killing, infected male offspring are killed early in embryogenesis. Within the Insecta, male-killing bacteria have been found in a wide range of hosts. The Coccinellidae families of beetles, better known as ladybirds, are particularly prone to male-killer invasion. In samples of the coccinellid, Coccinula crotchi, from Japan, a new male-killing bacterium was revealed by phenotypic assay. Molecular genetic analysis revealed the identity to be a tetracycline-sensitive Flavobacterium that causes female-biased offspring sex ratio. Furthermore, that Flavobacterium strain was found to be closely related to the Flavobacterium causing male-killing in the congeneric Japanese coccinellid, Coccinula sinensis, which was collected from the same region. However, we found slightly different Flavobacterium strains infecting C. sinensis from regions with different environmental conditions. This may be an indication of horizontal transmission of male-killing Flavobacterium between these 2 ladybird spices. Finally, environmental conditions may affect the spread of male-killing bacteria among their hosts.

Male killing bacteria are common in insects and are thought to persist in host populations primarily by indirect fitness benefits to infected females, whereas direct fitness effects are generally assumed to be neutral or deleterious. Here, we estimated the effect of male killer infection on direct fitness (number of eggs laid, as a measure of fecundity, together with survival) and other life-history traits (development time and body size) in seven ladybird host/male killer combinations. Effects of male killers on fecundity ranged, as expected, from costly to neutral; however, we found evidence of reduced development time and increased survival and body size in infected strains. Greater body size in Spiroplasma-infected Harmonia axyridis corresponded to greater ovariole number and therefore higher potential fecundity. To our knowledge, this is the first report of direct benefits of male killer infection after explicitly controlling for indirect fitness effects. Neutral or deleterious fitness effects of male killer infection should not therefore be automatically assumed.

Male-killing bacteria are thought to persist in host populations by vertical transmission and conferring direct and/or indirect fitness benefits to their hosts. Here, we test the role of indirect fitness benefits accrued from resource reallocation in species that engage in sibling egg cannibalism. We found that a single-egg meal significantly increased larval survival in 12 ladybird species, but the value of an egg (to survival) differed substantially between species. Next, we tested the impact of three male-killing bacteria on larval survival in one ladybird species, Adalia bipunctata. Spiroplasma reduced larval survival, whereas Wolbachia and Rickettsia had no effect. However, Spiroplasma-infected larvae showed the greatest response to a single-egg meal. The indirect fitness benefit obtained from a single egg is thus so large that even male-killing bacteria with direct fitness costs can persist in host populations. This study supports the hypothesis that fitness compensation via resource reallocation can explain male-killing bacteria persistence.

A simple and efficient one-pot three-component synthesis of the title compounds (IV) is presented.

A series of trifluoromethyl-substituted hexahydropyrimidine derivatives were efficiently synthesized in excellent yields via one-pot three-component reaction of aromatic aldehydes, ethyl trifluoroacetoacetate and thiourea(urea) in presence of p-toluenesulfonic acid under solvent-free conditions at room temperature by grinding. The present method does not involve any hazardous organic solvent and has proven to be simple, efficient, environmentally benign and cost-effective compared with the classical synthetic methods. These compounds were screened for their antibacterial activities against Escherichia coli and Bacillus thuringiensis and found to exhibit remarkably better antibacterial activities than the control drug.

Ladybirds are a hot-spot for the invasion of male-killing bacteria. These maternally inherited endosymbionts cause the death of male host embryos, to the benefit of female sibling hosts and the bacteria that they contain. Previous studies have shown that high temperatures can eradicate male-killers from ladybirds, leaving the host free from infection. Here we report the discovery of two maternally inherited sex ratio distorters in populations of a coccinellid, Coccinella undecimpunctata, from a hot lowland region of the Middle East. DNA sequence analysis indicates that the male killing is the result of infection by Wolbachia, that the trait is tetracycline sensitive, and that two distinct strains of Wolbachia co-occur within one beetle population. We discuss the implications of these findings for theories of male-killing and suggest avenues for future field-work on this system.
