Hair follicles (HF) represent a useful tissue for monitoring the circadian clock in mammals. Irregular light exposure causes circadian disruption and represents a welfare concern for stabled horses. We aimed to evaluate the impact of two stable lighting regimes on circadian clock gene rhythmicity in HF from racehorses. Two groups of five Thoroughbred racehorses in training at a commercial racehorse yard were exposed to standard incandescent light or a customized LED lighting system. The control group received light from incandescent bulbs used according to standard yard practice. The treatment group received timed, blue-enriched white LED light by day and dim red LED light at night. On weeks 0 and 20, mane hairs were collected at 4 h intervals for 24 h. Samples were stored in RNAlater at -20 °C. RNA was isolated and samples interrogated by quantitative PCR for the core clock genes: ARNTL , CRY1 , PER1 , PER2 , NR1D2 , and the clock-controlled gene DBP . Cosinor analyses revealed 24 h rhythmicity for NR1D2 and PER2 and approached significance for CRY1 ( p = 0.013, p = 0.013, and p = 0.051, respectively) in week 20 in the treatment group only. No rhythmicity was detected in week 0 or in week 20 in the HF of control horses. Results suggest that lighting practices in racehorse stables may be improved to better stimulate optimum functioning of the circadian system.

A method to capture three-dimensional (3D) objects image data under extremely low light level conditions, also known as Photon Counting Imaging (PCI), was reported. It is demonstrated that by combining a PCI system with computational integral imaging algorithms, a 3D scene reconstruction and recognition is possible. The resulting reconstructed 3D images often look degraded (due to the limited number of photons detected in a scene) and they, therefore, require the application of superior image restoration techniques to improve object recognition. Recently, Deep Learning (DL) frameworks have been shown to perform well when used for denoising processes. In this paper, for the first time, a fully unsupervised network (i.e., U-Net) is proposed to denoise the photon counted 3D sectional images. In conjunction with classical U-Net architecture, a skip block is used to extract meaningful patterns from the photons counted 3D images. The encoder and decoder blocks in the U-Net are connected with skip blocks in a symmetric manner. A novel (to the best of our knowledge), fast method to measure in-plane object motion in 1D with sub-pixel accuracy which complements the correlation technique is proposed. The method is verified experimentally using both visible and terahertz images. The absolute sum of grey level accumulated change is used to quantify object motion. The method requires calibration for each target, but only addition and subtraction operations. This results in a decrease of two orders of magnitude in the computation time.

This study is novel for several reasons: We used a thin drop cast layer of dry photosensitive materials to study the behaviors of wet photopolymer media using microscopic distances during the Self-Written Waveguide (SWW) process; then, we examined the self-trajectories formed inside the solid material. The results provide a framework for theoretical and experimental examinations by handling the effects of manipulating the alignment of fibers. The other main advantage of these techniques is their lightweight, easy to process, highly flexible, and ultimately low-cost nature. First, the SWW process in wet photopolymer media (liquid solutions) was examined under three cases: single-, counter-, and co-fiber exposure. Then, the SWWs formed inside the solid material were examined along with the effects of manipulating the alignment of the fibers. In all cases, high precision measurements were used to position the fiber optic cables (FOCs) before exposure using a microscope. The self-writing process was indirectly monitored by observing (imaging) the light emerging from the side of the material sample during SWW formation. In this way, we examined the optical waveguide trajectories formed in Acrylamide/Polyvinyl Alcohol (AA/PVA), a photopolymer material (sensitized at 532 nm). First, the transmission of light by this material is characterized. Then, the bending and merging of the waveguides that occur are investigated. The predictions of our model are shown to qualitatively agree with the observed trajectories. The largest index changes taking place at any time during exposure, i.e., during SWW formation, are shown to take place at the positions where the largest exposure light intensity is present. Typically, such maxima exist close to the input face. The first maximum is referred to as the location of the Primary Eye . Other local maxima also appear further along the SWW and are referred to as Secondary Eyes , i.e., eyes deeper within the material.

We present a simple and customizable microneedle mold fabrication technique using a low-cost desktop SLA 3D printer. As opposed to conventional microneedle fabrication methods, this technique neither requires complex and expensive manufacturing facilities nor expertise in microfabrication. While most low-cost 3D-printed microneedles to date display low aspect ratios and poor tip sharpness, we show that by introducing a two-step "Print & Fill" mold fabrication method, it is possible to obtain high-aspect ratio sharp needles that are capable of penetrating tissue. Studying first the effect of varying design input parameters and print settings, it is shown that printed needles are always shorter than specified. With decreasing input height, needles also begin displaying an increasingly greater than specified needle base diameter. Both factors contribute to low aspect ratio needles when attempting to print sub-millimeter height needles. By setting input height tall enough, it is possible to print needles with high-aspect ratios and tip radii of 20-40 µm. This tip sharpness is smaller than the specified printer resolution. Consequently, high-aspect ratio sharp needle arrays are printed in basins which are backfilled and cured in a second step, leaving sub-millimeter microneedles exposed resulting microneedle arrays which can be used as male masters. Silicone female master molds are then formed from the fabricated microneedle arrays. Using the molds, both carboxymethyl cellulose loaded with rhodamine B as well as polylactic acid microneedle arrays are produced and their quality examined. A skin insertion study is performed to demonstrate the functional capabilities of arrays made from the fabricated molds. Unlike coherent imaging techniques, light field imaging uses incoherent (white light) illumination to generate a digital hologram of three-dimensional (3D) objects in real time. Multiple projections (or elemental images) of a 3D object are captured using a microlens array attached to a digital camera. Orthographic projection images (OPIs) can be synthesized from the recorded elemental images. The synthesized intensity-based OPIs are then multiplied by the corresponding phase functions and combined to form a digital hologram (also known as an integral hologram) of a 3D object under illumination. In this study, we analyze the performance of a synthesized integral hologram under low light imaging (photon-counting) conditions. The feasibility of this technique is verified experimentally by capturing the elemental images and subsequently generating orthographic projection images and by varying photon counts to reconstruct the digital holograms.

An experimental and theoretical investigation of the preparation and exposure of multilayer photosensitive materials is presented. It is shown how the recorded change in the refractive index in each layer depends on the dye (photosensitizer) concentrations in each layer. It is also shown how the photosensitive material properties in each layer can be controlled to optimize some recording characteristics for particular applications. To do so, a set of equations, predicting the amplitude of higher harmonics refractive index amplitudes induced in the material layers with depth during exposure, is derived. This results in a technique for varying the dye concentration in each layer of a multilayer, so as to optimize volume diffraction grating performance. In part I of this paper, the 3D nonlocal photopolymerization-driven diffusion (NPDD) model is applied to calculate the resulting combined multilayer absorption and polymerization processes. The NPDD describes the time-varying behaviors taking place during exposure in such photopolymer materials. Simulations are performed for an acrylamide/polyvinyl alcohol-based photopolymer containing erythrosine-B dye. It is predicted that, in general, non-uniform gratings are formed, with the resulting refractive index being distorted both from the ideal sinusoidal cross-sectional spatial distribution and also with depth. This agrees with previous results indicating that increasing the thickness of a single photopolymer layer does not in practice lead to ever-increasing angular selectivity. In part II of this paper, it is confirmed experimentally that a suitably modified multilayer can be used to increase grating angular selectivity, i.e., reduce the width of the off-Bragg replay curve.

In the first part of this study, a 3D nonlocal photopolymerization driven diffusion model was developed and applied to simulate the absorption and polymerization taking place during holographic exposures of a multi-layer. The Beer-Lambert law was used to choose appropriate dye concentrations for each layer, with the objective of improving the resulting volume grating uniformity and thus diffraction characteristics. The predictions made, using previously estimated physical parameter values, indicated that improvements in the uniformity of the recorded modulation were possible. In this paper the results of experiments carried out to explore the validity of these predictions are presented. Improvements in material response are demonstrated experimentally, with improved grating diffraction (narrower angular selectivity) being observed for appropriately sensitized multi-layers.

Airway epithelium structure/function can be altered by local inflammatory/immune signals, and this process is called epithelial remodeling. The mechanism by which this innate response is regulated, which causes mucin/mucus overproduction, is largely unknown. Exosomes are nanovesicles that can be secreted and internalized by cells to transport cellular cargo, such as proteins, lipids, and miRNA. The objective of this study was to understand the role exosomes play in airway remodeling through cell-cell communication. We used two different human airway cell cultures: primary human tracheobronchial (HTBE) cells, and a cultured airway epithelial cell line (Calu-3). After intercellular exosomal transfer, comprehensive proteomic and genomic characterization of cell secretions and exosomes was performed. Quantitative proteomics and exosomal miRNA analysis profiles indicated that the two cell types are fundamentally distinct. HTBE cell secretions were typically dominated by fundamental innate/protective proteins, including mucin MUC5B, and Calu-3 cell secretions were dominated by pathology-associated proteins, including mucin MUC5AC. After exosomal transfer/intake, approximately 20% of proteins, including MUC5AC and MUC5B, were significantly altered in HTBE secretions. After exosome transfer, approximately 90 miRNAs (∼4%) were upregulated in HTBE exosomes, whereas Calu-3 exosomes exhibited a preserved miRNA profile. Together, our data suggest that the transfer of exosomal cargo between airway epithelial cells significantly alters the qualitative and quantitative profiles of airway secretions, including mucin hypersecretion, and the miRNA cargo of exosomes in target cells. This finding indicates that cellular information can be carried between airway epithelial cells via exosomes, which may play an important role in airway biology and epithelial remodeling.

In general, the holographic grating refractive index profiles in photopolymer materials are not identical to the exposing pattern. During exposure, high harmonics of the fundamental refractive index period are generated within the layer volume. A set of equations to calculate the amplitudes of the higher harmonics of refractive index induced in the grating is introduced. Then, an algorithm involving the use of the 3D nonlocal photopolymerization-driven diffusion model is presented and applied to calculate the resulting grating diffraction efficiencies. The experimental observation that the grating diffraction efficiency cannot reach the theoretical maximum value (η max =100%) and that, in the case of over-modulation, the minimum value (η min =0%) is also never achieved, are explained theoretically. The predictions of the simulations are also fit to experimental data for an acrylamide/polyvinyl alcohol photopolymer material with good agreement being achieved.

A method to measure the refractive index of an optically flat, regularly shaped slab of glass using speckle correlation-based techniques is reported. The intensity of the diffraction field of the diffuser is captured by a CCD both with and without the glass present. As the position of the peak correlation coefficient is quantitatively related to the change in optical path length arising due to the presence of the glass, the refractive index of the glass can be evaluated by cross-correlating the two captured images. The theoretical correlation function that describes the effects of such an optical path length change is discussed, and the resulting speckle decorrelation function derived. Two glass samples are measured to demonstrate the accuracy and robustness of the proposed technique.

This feature issue provides a snapshot of some of the applied optics and photonics related research and development activities currently taking place across Ireland. The issue contains some thirty papers, including contributions from universities and institutes of technology research groups, state research laboratories and institutes, and commercial companies.

Self-written waveguide (SWW) trajectories fabricated inside a dry photopolymer bulk material, acrylamide/polyvinyl alcohol (AA/PVA), are studied. Their production using both Gaussian and Laguerre-Gauss exposing (writing) light beams, output from optical fibers, is explored. The formation of the primary and secondary eyes is also discussed. Furthermore, the interactions that take place when two counterpropagating beams pass through the photopolymer material (both Gaussian and Laguerre-Gauss) are examined. In all cases experimental and theoretical results are presented. Good agreement between the predictions of the proposed model and experimental observations are demonstrated.

Photopolymer materials have received a great deal of attention because they are inexpensive, self-processing materials that are extremely versatile, offering many advantages over more traditional materials. To achieve their full potential, there is significant value in understanding the photophysical and photochemical processes taking place within such materials. This paper includes a brief review of recent attempts to more fully understand what is needed to optimize the performance of photopolymer materials for Holographic Data Storage (HDS) and Self-Written Waveguides (SWWs) applications. Specifically, we aim to discuss the evolution of our understanding of what takes place inside these materials and what happens during photopolymerization process, with the objective of further improving the performance of such materials. Starting with a review of the photosensitizer absorptivity, a dye model combining the associated electromagnetics and photochemical kinetics is presented. Thereafter, the optimization of photopolymer materials for HDS and SWWs applications is reviewed. Recently, the vulnerability of the linear canonical transform-based double random phase encryption system to attack has been demonstrated. To alleviate this, we present for the first time, to the best of our knowledge, a method for securing a two-dimensional scene using a quadratic phase encoding system operating in the photon-counted imaging (PCI) regime. Position-phase-shifting digital holography is applied to record the photon-limited encrypted complex samples. The reconstruction of the complex wavefront involves four sparse (undersampled) dataset intensity measurements (interferograms) at two different positions. Computer simulations validate that the photon-limited sparse-encrypted data has adequate information to authenticate the original data set. Finally, security analysis, employing iterative phase retrieval attacks, has been performed.

We report a light-field based method that allows the optical encryption of three-dimensional (3D) volumetric information at the microscopic scale in a single 2D light-field image. The system consists of a microlens array and an array of random phase/amplitude masks. The method utilizes a wave optics model to account for the dominant diffraction effect at this new scale, and the system point-spread function (PSF) serves as the key for encryption and decryption. We successfully developed and demonstrated a deconvolution algorithm to retrieve both spatially multiplexed discrete data and continuous volumetric data from 2D light-field images. Showing that the method is practical for data transmission and storage, we obtained a faithful reconstruction of the 3D volumetric information from a digital copy of the encrypted light-field image. The method represents a new level of optical encryption, paving the way for broad industrial and biomedical applications in processing and securing 3D data at the microscopic scale.

Diversity training is challenging and can evoke strong emotional responses from participants including resistance, shame, confusion, powerlessness, defensiveness, and anger. These responses create complex situations for both presenters and other learners. We observed 3 experienced presenters as they implemented 41 gender bias literacy workshops for 376 faculty from 42 STEMM (science, technology, engineering, mathematics, medicine) departments at one Midwestern university. We recorded questions and answers as well as participants' non-verbal activity during each 2.5-hour workshop. Employing content analysis and critical incident technique, we identified content that elicited heightened activity and challenging dialogues among presenters and faculty. Results from analysis of this observational data found three important findings: (1) presenters continually reinforced the idea that implicit bias is ordinary and pervasive, thus avoiding participant alienation by allowing participants to protect their self-worth and integrity; (2) difficult dialogues were managed calmly without verbal sparring or relinquishing control; (3) the presenters created an environment where individuals were more likely to accept threatening information.

The hybrid input-output algorithm, error reduction algorithm, and combinations of both phase retrieval algorithms are applied to perform ciphertext-only attacks on linear canonical transform (LCT)-based amplitude encoding double-random-phase encryption (DRPE) systems. Special cases of LCT-based DRPE systems, i.e., Fourier-transform-based, fractional-Fourier-transform-based, and Fresnel-transform-based DRPE, can also be successfully attacked using the method proposed. Numerical simulations are performed to demonstrate the efficacy of the proposed attacking method.

The continuous linear canonical transform (LCT) can describe a wide variety of paraxial (quadratic phase) first-order optical systems. Digital algorithms to numerically calculate the LCT are therefore important in modeling the field propagations and are also of interest for many digital signal-processing applications. The continuous LCT is additive (and unitary), but discretization can destroy additivity. In this paper, the general constraint sufficient to ensure the discrete LCTs are additive is derived. Often, we wish to decompose the transform into a series of more computationally efficient steps. Having previously discussed the unitarity of such algorithms, in this paper we consider how our additivity constraint applies to the direct method (DM) and spectral method (SM) algorithms. Examples are presented showing how to correct nonadditive calculations and to appropriately choose parameters.

Photopolymers as recording media are widely used in optical applications. In such materials, changes in the phase of the transmittance function are generated during exposure due to refractive index and thickness modulations. These changes arise primarily as a consequence of photopolymerization and mass transport processes. Characterizing polymers' performance, for example, quantifying the value of monomer diffusion, is therefore very important. Applying index matching, the volume and surface optical effect are separated in an acrylamide/polyvinylalcohol (AA/PVA) material. Using a simplified model that includes the effects of the holes produced during polymerization, both hole and monomer diffusion are analyzed. The analysis presented indicates higher material sensitivity than previously estimated. The results also indicate the possibility of recording sharper diffractive optical elements profiles, like blazed gratings, having diffraction efficiencies higher than 80%.

A novel method for multimode fiber (MMF) laser-beam cleanup is introduced based on the optically induced growth and interaction of self-written waveguides (SWWs) in a photopolymer material. Theoretically, it is predicted that when the light is introduced into a free-radical photopolymerizable system from a MMF, the incident multichannel and structured beam shape can be caused to merge to form a single-channel Gaussian-like beam under specific exposure and material conditions. Experimental validation was carried out using a dry acrylamide/polyvinyl alcohol (AA/PVA) photopolymer sample. This work opens the door to studies involving self-developing laser beam cleanup and also to possible applications in photonic telecommunication systems and integrated optical devices.

We present a method of securing multispectral 3D photon-counted integral imaging (PCII) using classical Hartley Transform (HT) based encryption by employing optical interferometry. This method has the simultaneous advantages of minimizing complexity by eliminating the need for holography recording and addresses the phase sensitivity problem encountered when using digital cameras. These together with single-channel multispectral 3D data compactness, the inherent properties of the classical photon counting detection model, i.e. sparse sensing and the capability for nonlinear transformation, permits better authentication of the retrieved 3D scene at various depth cues. Furthermore, the proposed technique works for both spatially and temporally incoherent illumination. To validate the proposed technique simulations were carried out for both the 2D and 3D cases. Experimental data is processed and the results support the feasibility of the encryption method.

The modified iterative phase retrieval algorithms developed in Part I [Guo et al., Appl. Opt.54, 4698 (2015)] are applied to perform known plaintext and ciphertext attacks on amplitude encoding and phase encoding Fourier-transform-based double random phase encryption (DRPE) systems. It is shown that the new algorithms can retrieve the two random phase keys (RPKs) perfectly. The performances of the algorithms are tested by using the retrieved RPKs to decrypt a set of different ciphertexts encrypted using the same RPKs. Significantly, it is also shown that the DRPE system is, under certain conditions, vulnerable to ciphertext-only attack, i.e., in some cases an attacker can decrypt DRPE data successfully when only the ciphertext is intercepted.

Two modified Gerchberg-Saxton (GS) iterative phase retrieval algorithms are proposed. The first we refer to as the spatial phase perturbation GS algorithm (SPP GSA). The second is a combined GS hybrid input-output algorithm (GS/HIOA). In this paper (Part I), it is demonstrated that the SPP GS and GS/HIO algorithms are both much better at avoiding stagnation during phase retrieval, allowing them to successfully locate superior solutions compared with either the GS or the HIO algorithms. The performances of the SPP GS and GS/HIO algorithms are also compared. Then, the error reduction (ER) algorithm is combined with the HIO algorithm (ER/HIOA) to retrieve the input object image and the phase, given only some knowledge of its extent and the amplitude in the Fourier domain. In Part II, the algorithms developed here are applied to carry out known plaintext and ciphertext attacks on amplitude encoding and phase encoding double random phase encryption systems. Significantly, ER/HIOA is then used to carry out a ciphertext-only attack on AE DRPE systems.

In this Letter, we propose an information encryption technique based on the theory of phase-space optics. We show that encoding the plaintext in phase space provides a higher level of security: first, the key-space is significantly enlarged. Second, it is immune to various known-plaintext (cyphertext) attacks to which the double-random phase encryption (DRPE) is vulnerable. Third, the bilinearity of phase-space distributions offers additional security. Theoretical analysis and numerical calculation results show that the proposed technique has significantly different responses to errors added to the cypheretext and the two phase keys in comparison to the classical DRPE.

We report on the combined far-field measurement of the three involuntary eye movements, drift, microsaccades, and ocular microtremor (OMT), using a noncontact far-field optical method. We review the significance of the smallest and least measured, and thus least understood, of the three, OMT. Using modern digital imaging techniques, we perform detailed analysis, present experimental results, and examine the extracted parameters using a noncontact far-field sensor. The two-dimensional (2D) nonseparable linear canonical transform (NS-LCT) is a unitary, linear integral transform that relates the input and output monochromatic, paraxial scalar wave fields of optical systems characterized by a 4×4 ray tracing matrix. In addition to the obvious generalizations of the 1D LCT (which are referred to as separable), the 2D-NS-LCT can represent a variety of nonaxially symmetric optical systems including the gyrator transform and image rotation. Unlike the 1D LCT, the numerical approximation of the 2D-NS-LCT has not yet received extensive attention in the literature. In this paper, (1) we develop a sampling theorem for the general 2D-NS-LCT which generalizes previously published sampling theorems for the 1D case and (2) we determine which sampling rates may be chosen to ensure that the obvious discrete transform is unitary.

For the first time it is demonstrated that permanent optical waveguides can be self-written in a solid acrylamide/polyvinyl alcohol photopolymer material. The novel (to our knowledge) technique used to prepare the polymeric medium used is described. It is demonstrated that the resulting waveguides formed can be used to guide different wavelengths. A standard theoretical model is used to predict both the evolution of the light intensity distribution and the channel formation inside the material during the exposure. The experimental results and the numerical simulations are compared, and good agreement is obtained.

Chronic obstructive pulmonary disease affects 64 million people and is currently the fourth leading cause of death worldwide. Chronic obstructive pulmonary disease includes both emphysema and chronic bronchitis, and in the case of chronic bronchitis represents an inflammatory response of the airways that is associated with mucus hypersecretion and obstruction of small airways. Recently, it has emerged that exposure to cigarette smoke (CS) leads to an inhibition of the cystic fibrosis transmembrane conductance regulator (CFTR) Cl(-) channel, causing airway surface liquid dehydration, which may play a role in the development of chronic bronchitis. CS rapidly clears CFTR from the plasma membrane and causes it to be deposited into aggresome-like compartments. However, little is known about the mechanism(s) responsible for the internalization of CFTR following CS exposure. Our studies revealed that CS triggered a rise in cytoplasmic Ca(2+) that may have emanated from lysosomes. Furthermore, chelation of cytoplasmic Ca(2+), but not inhibition of protein kinases/phosphatases, prevented CS-induced CFTR internalization. The macrolide antibiotic bafilomycin A1 inhibited CS-induced Ca(2+) release and prevented CFTR clearance from the plasma membrane, further linking cytoplasmic Ca(2+) and CFTR internalization. We hypothesize that CS-induced Ca(2+) release prevents normal sorting/degradation of CFTR and causes internalized CFTR to reroute to aggresomes. Our data provide mechanistic insight into the potentially deleterious effects of CS on airway epithelia and outline a hitherto unrecognized signaling event triggered by CS that may affect the long term transition of the lung into a hyper-inflammatory/dehydrated environment.

Dyes often act as the photoinitiator PI/photosensitizer PS in photopolymer materials and are therefore of significant interest. The properties of the PI/PS used strongly influences grating formation when the material layer is exposed holographically. In this paper, the ability of a recently synthesized dye, D_1, to sensitize an acrylamide/polyvinyl alcohol (AA/PVA) based photopolymer is examined, and the material performance is characterized using an extended nonlocal photopolymerization-driven diffusion model. Electron spin resonance spin-trapping (ESR-ST) experiments are also carried out to characterize the generation of the initiator/primary radical, R(•), during exposure. The results obtained are then compared with those for the corresponding situation when using a xanthene dye, i.e., erythrosine B, under the same experiment conditions. The results indicate that the nonlocal effect is greater when this new photosensitizer is used in the material. Analysis indicates that this is the case because of the dye's (D_1) weak absorptivity and the resulting slow rate of primary radical production.

Speckle suppression in a two-diffuser system is examined. An analytical expression for the speckle space-time correlation function is derived, so that the speckle suppression mechanism can be investigated statistically. The grain size of the speckle field illuminating the second diffuser has a major impact on the speckle contrast after temporal averaging. It is shown that, when both the diffusers are rotating, the one with the lower rotating speed determines the period of the speckle correlation function. The coherent length of the averaged speckle intensity is shown to equal the mean speckle size of the individual speckle pattern before averaging. Numerical and experimental results are presented to verify our analysis in the context of speckle reduction.

Sex plays a significant role in the development of lung diseases including asthma, cancer, chronic bronchitis, and cystic fibrosis. In cystic fibrosis, 17β-estradiol (E2) may inhibit store-operated Ca(2+) entry (SOCE) to impinge upon airway secretions, leaving females at greater risk of contracting lung infections. Stromal interaction molecule 1 (STIM1)-mediated SOCE is essential for cell homeostasis and regulates numerous processes including cell proliferation, smooth muscle contraction, and secretion. E2 can signal nongenomically to modulate Ca(2+) signaling, but little is known of the underlying mechanisms. We found that E2 exposure inhibited STIM1 translocation in airway epithelia, preventing SOCE. This correlated with a decrease in STIM1-STIM1 FRET and STIM1 mobility in E2-exposed HEK293T cells co-expressing estrogen receptor α. We also examined the role of STIM1 phosphorylation in E2-mediated inhibition of STIM1 mobility. STIM1 is basally phosphorylated at serine 575, which is required for SOCE. Exposure to E2 significantly decreased STIM1 serine phosphorylation. Mutating serine 575 to an alanine blocked STIM1 phosphorylation, reduced basal STIM1 mobility, and rendered STIM1 insensitive to E2. These data indicate that E2 can signal nongenomically by inhibiting basal phosphorylation of STIM1, leading to a reduction in SOCE.

The scattering of coherent monochromatic light at an optically rough surface, such as a diffuser, produces a speckle field, which is usually described by reference to its statistical properties. For example, the real and imaginary parts of a fully developed speckle field can be modeled as a random circular Gaussian process. When such a speckle field is used to illuminate a second diffuser, the statistics of the resulting doubly scattered field are in general no longer Gaussian, but rather follow a K distribution. In this paper we determine the space-time correlation function of such a doubly scattered speckle field that has been imaged by a single lens system. A space-time correlation function is derived that contains four separate terms; similar to the Gaussian case it contains an average DC term and a fluctuating AC term. However, in addition there are two terms that are related to contributions from each of the diffusers independently. We examine how our space-time correlation function varies as the diffusers are rotated at different speeds and as the point spread function of the imaging system is changed. A series of numerical simulations are used to confirm different aspects of the theoretical analysis. We then finish with a discussion of our results and some potential applications, including controlling spatial coherence and speckle reduction.

The production of melatonin during night-time hours decodes day length for seasonally breeding animals. The use of artificial light to advance the breeding season in mares is common practice within the equine industry. Four healthy Thoroughbred mares were used to evaluate the minimum intensity of light required to inhibit serum melatonin. Mares were fitted with indwelling jugular catheters and using a crossover design blood samples were collected following 1h exposure to light (barn lighting approximately 200 lux), dark (<0.1 lux), and 3, 10, 50, and 100 lux intensities. The light source was a light-emitting diode (LED; 468 nm) directed at either a single eye or both eyes. All treatments, except the sample collected after 1 h exposure to light, occurred during the dark phase of the 24 h cycle. Serum melatonin levels were determined by radioimmunoassay. Two-way repeated measures ANOVA revealed that there was no difference between the level of melatonin inhibition achieved when light was administered to one or two eyes (P = 0.7028). One-way ANOVA of melatonin levels at light intensities of 10, 50 and 100 lux were significantly different to dark (P < 0.05) and not different to light (P > 0.05) intensities. There was no difference between melatonin levels at 3 lux (P > 0.05) and dark intensities. The threshold level of low wavelength light required to inhibit melatonin production in the horse lies between 3 and 10 lux. Melatonin inhibition can be achieved by exposing a single eye to low wavelength blue light. The numerical approximation of the linear canonical transforms (LCTs) is important in modeling coherent wave field propagation through first-order optical systems and in many digital signal processing applications. The continuous LCTs are unitary, but discretization can destroy this property. We present a sufficient condition on the sampling rates chosen in the discretization to ensure unitarity. We discuss the various subsets of the unitary matrices examined in this paper that have been proposed elsewhere. We offer a proof of the existence of all of the unitary matrices we discuss. We examine the consequences of these results, particularly in relation to the use of discrete transforms in iterative phase retrieval applications.

Imaging systems have numerous applications in industrial, military, consumer, and medical settings. Assembling a complete imaging system requires the integration of optics, sensing, image processing, and display rendering. This issue features original research ranging from fundamental theories to novel imaging modalities and provides a systems perspective to imaging.

Digital in-line holographic microscopy (DIHM) allows access to both intensity and phase information with conventional microscopic lateral resolutions. Such imaging techniques can, however, be used to increase the depth of focus compared to conventional compound microscopes. We present a simple DIHM capable of imaging weakly scattering 10 μm diameter microspheres as well as Hs578T cells over a depth of 1 mm; i.e., we demonstrate an increase by a factor of 100 over the depth of focus of a conventional microscope.

Subpixel methods increase the accuracy and efficiency of image detectors, processing units, and algorithms and provide very cost-effective systems for object tracking. Published methods achieve resolution increases up to three orders of magnitude. In this Letter, we demonstrate that this limit can be theoretically improved by several orders of magnitude, permitting micropixel and submicropixel accuracies. The necessary condition for movement detection is that one single pixel changes its status. We show that an appropriate target design increases the probability of a pixel change for arbitrarily small shifts, thus increasing the detection accuracy of a tracking system. The proposal does not impose severe restriction on the target nor on the sensor, thus allowing easy experimental implementation.

The National Science Foundation and others conclude that institutional transformation is required to ensure equal opportunities for the participation and advancement of men and women in academic science, technology, engineering, mathematics, and medicine (STEMM). Such transformation requires changing the habitual attitudes and behaviors of faculty. Approaching implicit bias as a remediable habit, we present the theoretical basis and conceptual model underpinning an educational intervention to promote bias literacy among university faculty as a step toward institutional transformation regarding gender equity. We describe the development and implementation of a Bias Literacy Workshop in detail so others can replicate or adapt it to their setting. Of the 220 (167 faculty and 53 nonfaculty) attendees from the initial 17 departments/divisions offered this workshop, all 180 who completed a written evaluation found the workshop at least "somewhat useful" and 74% found it "very useful." Over 68% indicated increased knowledge of the workshop material. Of the 186 participants who wrote a commitment to engage in new activities to promote gender equity, 87% incorporated specific workshop content. Twenty-four participants were interviewed 4-6 months after attending the workshop; 75% of these not only demonstrated increased bias awareness, but described plans to change-or had actually changed-behaviors because of the workshop. Based on our sample of faculty from a Midwestern university, we conclude that at least one third of STEMM faculty who are invited will attend a 2.5-hr Bias Literacy Workshop, that nearly all will find it useful, and that most will complete a written commitment to promoting gender equity. These findings suggest that this educational intervention may effectively promote institutional change regarding gender equity.

The statistical properties of speckles in paraxial optical systems depend on the system parameters. In particular, the speckle orientation and the lateral dependence (x and y) of the longitudinal speckle size can vary significantly. For example, the off-axis longitudinal correlation length remains equal to the on-axis size for speckles in a Fourier transform system, while it decreases dramatically as the observation position moves off axis in a Fresnel system. In this paper, we review the speckle correlation function in general linear canonical transform (LCT) systems, clearly demonstrating that speckle properties can be controlled by introducing different optical components, i.e., lenses and sections of free space. Using a series of numerical simulations, we examine how the correlation function changes for some typical LCT systems. The integrating effect of the camera pixel and the impact this has on the measured first- and second-order statistics of the speckle intensities is also examined theoretically. A series of experimental results are then presented to confirm several of these predictions. First, the effect the pixel size has on the measured first-order speckle statistics is demonstrated, and second, the orientation of speckles in a Fourier transform system is measured, showing that the speckles lie parallel to the optical axis.

The development of suitable recording media for applications such as holographic optical elements and holographic data storage are of significant research and commercial interest. In this paper, a photopolymer material developed by Bayer MaterialScience is examined using various optical techniques and then characterised using the Non-local Photo-polymerization Driven Diffusion model. This material demonstrates the capabilities of a new class of photopolymer offering high index modulation, full colour recording, high light sensitivity and environmental stability. One key result of this study is the material's high spatial frequency resolution, indicating a very low non-local effect, thus qualifying it as a very good storage medium.

An compact multispectral digital in-line holographic microscope (DIHM) is developed that emulates Gabor's original holographic principle. Using sources of varying spatial coherence (laser, LED), holographic images of objects, including optical fiber, latex microspheres, and cancer cells, are successfully captured and numerically processed. Quantitative measurement of cell locations and percentage confluence are estimated, and pseudocolor images are also presented. Phase profiles of weakly scattering cells are obtained from the DIHM and are compared to those produced by a commercially available off-axis digital holographic microscope.

An understanding of the photochemical and photo-physical processes, which occur during photopolymerization is of extreme importance when attempting to improve a photopolymer material's performance for a given application. Recent work carried out on the modelling of the mechanisms which occur in photopolymers during- and post-exposure, has led to the development of a tool, which can be used to predict the behaviour of these materials under a wide range of conditions. In this paper, we explore this Non-local Photo-polymerisation Driven Diffusion model, illustrating some of the useful trends, which the model predicts and we analyse their implications on the improvement of photopolymer material performance.

An efficient algorithm for the accurate computation of the linear canonical transform with complex transform parameters and with complex output variable is presented. The product of the spatial and spatial frequency extents of a wave field has proven useful in the analysis of the sampling requirements of numerical simulations. We propose that the ratio of these quantities is also illuminating. We have shown that the distance at which the so-called "direct method" becomes more efficient than the so-called "spectral method" for simulations of Fresnel transforms may be written in terms of this space-bandwidth ratio. We have proposed generalizations of these algorithms for numerical simulations of general ABCD systems and derived expressions for the "transition space-bandwidth ratio," above which the generalization of the spectral method is the more efficient algorithm and below which the generalization of the direct method is preferable.

Members of the Anoctamin (Ano)/TMEM16A family have recently been identified as essential subunits of the Ca(2+)-activated chloride channel (CaCC). For example, Ano1 is highly expressed in multiple tissues including airway epithelia, where it acts as an apical conduit for transepithelial Cl(-) secretion and helps regulate lung liquid homeostasis and mucus clearance. However, little is known about the oligomerization of this protein in the plasma membrane. Thus, utilizing mCherry- and eGFP-tagged Ano1 constructs, we conducted biochemical and Förster resonance energy transfer (FRET)-based experiments to determine the quaternary structure of Ano1. FRET and co-immunoprecipitation studies revealed that tagged Ano1 subunits directly associated before they reached the plasma membrane. This association was not altered by changes in cytosolic Ca(2+), suggesting that this is a fixed interaction. To determine the oligomeric structure of Ano1, we performed chemical cross-linking, non-denaturing PAGE, and electromobility shift assays, which revealed that Ano1 exists as a dimer. These data are the first to probe the quaternary structure of Ano1. Understanding the oligomeric nature of Ano1 is an essential step in the development of therapeutic drugs that could be useful in the treatment of cystic fibrosis.

Access to the spatial derivatives of an optical wave field can be used to enhance edge detection, focusing, and holographic imaging. It was recently shown that, by using digital holographic techniques, the Laplacian of an object field can be extracted. Here it is demonstrated that equivalent results can be found using two holograms captured at either two distances or with two appropriately related wavelengths. Experimental and numerical results confirming the theoretical analyses are presented. The proposed two-wavelength-based system requires no mechanical repositioning of the object and is shown to provide superior performance.

One opportunity to realize the diversity goals of academic health centers comes at the time of hiring new faculty. To improve the effectiveness of search committees in increasing the gender diversity of faculty hires, the authors created and implemented a training workshop for faculty search committees designed to improve the hiring process and increase the diversity of faculty hires at the University of Wisconsin-Madison. They describe the workshops, which they presented in the School of Medicine and Public Health between 2004 and 2007, and they compare the subsequent hiring of women faculty in participating and nonparticipating departments and the self-reported experience of new faculty within the hiring process. Attendance at the workshop correlates with improved hiring of women faculty and with a better hiring experience for faculty recruits, especially women. The authors articulate successful elements of workshop implementation for other medical schools seeking to increase gender diversity on their faculties.
