Abstract Background The latitudinal diversity gradient (LDG), characterized by an increase in species richness from the poles to the equator, is one of the most pervasive biological patterns. However, inverse LDGs, in which species richness peaks in extratropical regions, are also found in some lineages and their causes remain unclear. Here, we test the roles of evolutionary time, diversification rates, and niche conservatism in explaining the inverse LDG of Potentilla (ca. 500 species). We compiled the global distributions of ~ 90% of Potentilla species, and reconstructed a robust phylogenetic framework based on whole-plastome sequences. Next, we analyzed the divergence time, ancestral area, diversification rate, and ancestral niche to investigate the macroevolutionary history of Potentilla. Results The genus originated in the Qinghai-Tibet Plateau during the late Eocene and gradually spread to other regions of the Northern Hemisphere posterior to the late Miocene. Rapid cooling after the late Pliocene promoted the radiating diversification of Potentilla. The polyploidization, as well as some cold-adaptive morphological innovations, enhanced the adaptation of Potentilla species to the cold environment. Ancestral niche reconstruction suggests that Potentilla likely originated in a relatively cool environment. The species richness peaks at approximately 45 °N, a region characterized by high diversification rates, and the environmental conditions are similar to the ancestral climate niche. Evolutionary time was not significantly correlated with species richness in the latitudinal gradient. Conclusions Our results suggest that the elevated diversification rates in middle latitude regions and the conservatism in thermal niches jointly determined the inverse LDG in Potentilla. This study highlights the importance of integrating evolutionary and ecological approaches to explain the diversity pattern of biological groups on a global scale.

Due to the rapid loss of biodiversity and financial and human resource limitations, there is an urgent need to prioritize biodiversity conservation. Setting conservation priorities is also an important part of conservation biology. Species abundance, richness and International Union for Conservation of Nature (IUCN) ranks are the most commonly used methods for setting conservation priorities, but with the development of bioinformatics, many studies have also demonstrated the utility of incorporating phylogenetic diversity into species prioritization, with evolutionary distinctiveness (ED) and evolutionarily distinct and globally endangered (EDGE) being widely used to determine the prioritization of species for conservation, where ED measures the contribution of species to the overall evolutionary history, and EDGE considers both evolutionary distinctiveness and IUCN ranks. In this study, based on the vascular plant distribution data collected on the Qinghai-Tibet Plateau (QTP), we determined the conservation priorities of plants on the QTP according to ED and EDGE2 considering four extinction risk transformations, investigated the relationship between IUCN ranks and ED, evaluated the effects of unassessed (NA) data deficiency (DD) species on EDGE rankings, and determined the conservation efficiency of the QTP conservation network in areas with evolutionary history and explored the possibility of incorporating evolutionary history into species conservation priorities. The results showed that there was no significant relationship between the ED of vascular plants and the IUCN ranks on the QTP; the results of EDGE2 showed that the top 25 species, with 14 species common to all four transformations, possessed both high ED and IUCN ranks; when EDGE2 containing DD and NA species showed that DD and NA species accounted for a large percentage of the top 25 species for all four transformations, suggesting that they may have significant potential for evolutionary history conservation; that the conservation priority areas for endemic seed plants of the QTP are located in the Hengduan Mountains, the eastern Yarlung Zangbo River, the southern Qionglai Mountains, the western Gangdise Mountains and the southern Kunlun Mountains; and that the QTP conservation networks protect the only 38.2 % of the areas rich in evolutionary history. These results emphasize the need to incorporate evolutionary information into conservation biology, but it should also be considered in conjunction with existing taxonomic diversity methods as a complementary conservation tool to provide a new perspective on the conservation of QTP plant diversity.

Abstract Background Impatiens sect. Impatiens is distributed across the Northern Hemisphere and has diversified considerably, particularly within the Hengduan Mountains (HDM) in southwest China. Yet, the infra-sectional phylogenetic relationships are not well resolved, largely due to limited taxon sampling and an insufficient number of molecular markers. The evolutionary history of its diversification is also poorly understood. In this study, plastome data and the most complete sampling to date were used to reconstruct a robust phylogenetic framework for this section. The phylogeny was then used to investigate its biogeographical history and diversification patterns, specifically with the aim of understanding the role played by the HDM and past climatic changes in its diversification. Results A stable phylogeny was reconstructed that strongly supported both the monophyly of the section and its division into seven major clades (Clades I-VII). Molecular dating and ancestral area reconstruction suggest that sect. Impatiens originated in the HDM and Southeast China around 11.76 Ma, after which different lineages dispersed to Northwest China, temperate Eurasia, and North America, mainly during the Pliocene and Pleistocene. An intercontinental dispersal event from East Asia to western North America may have occurred via the Bering Land Bridge or Aleutian Islands. The diversification rate was high during its early history, especially with the HDM, but gradually decreased over time both within and outside the HDM. Multiple linear regression analysis showed that the distribution pattern of species richness was strongly associated with elevation range, elevation, and mean annual temperature. Finally, ancestral niche analysis indicated that sect. Impatiens originated in a relatively cool, middle-elevation area. Conclusions We inferred the evolutionary history of sect. Impatiens based on a solid phylogenetic framework. The HDM was the primary source or pump of its diversity in the Northern Hemisphere. Orogeny and climate change may have also shaped its diversification rates, as a steady decrease in the diversification rate coincided with the uplift of the HDM and climate cooling. These findings provide insights into the distribution pattern of sect. Impatiens and other plants in the Northern Hemisphere.

Wild plants are an important part of the natural ecological system, and China is one of the countries with the most abundant wild plant species. It is of great significance to study the distribution characteristics, protection status and potential distribution areas of National Key Protected Wild Plants for supporting and formulating biodiversity conservation strategies. Based on 1 032 species (belonging to 315 genera, 129 families) of National Key Protected Wild Plants, this study used the top 5% richness algorithm to identify hotspots, and superimposed with nature reserves to evaluate the conservation effectiveness and determine conservation gaps of current nature reserves. Furthermore, this study used maximum entropy (MaxEnt) model to predict the potential distribution and range shifts of National Key Protected Wild Plants. The results were as follows: (1) The areas with the highest species richness of the National Key Protected Wild Plants mainly confined to the south and southwest of China, especially in central Sichuan, southern and southeastern Yunnan, northern Guangxi, northern Guangdong and Hainan. (2) The conservation effectiveness of hotspot grids showed that 171 (85.50%) hotspot grids were efficient protection (including 80.50% of total species), and 29 hotspot grids (14.50%) were not effectively protected (including 51.20% of total species). (3) By comparing the potential distribution area of National Key Protected Wild Plants under current and future climate scenarios, it is found that potential distribution area would expand to southeastern of Xizang, southwestern Guangxi, southern Guangdong, and southern Fujian in the future, while those around Sichuan Basin, southern Yunnan and southern Guizhou would shrink. Therefore, it is necessary to strengthen the dynamic monitoring of biodiversity and pay continuous attention to the impact of climate change on National Key Protected Wild Plants in the region. Based on the analysis results of hotspot grids, conservation effectiveness and potential distribution areas identified in this study, strong data support and reference can be provided for the identification of diversity priority protected areas for National Key Protected Wild Plants and the formulation of conservation policies.

Abstract Cellular senescence describes a state of permanent proliferative arrest in cells. Studies have demonstrated that diabetes promotes the pathological accumulation of senescent cells, which in turn impairs cell movement and proliferation. Historically, senescence has been perceived to be a detrimental consequence of chronic wound healing. However, the underlying mechanism that causes senescent cells to remain in diabetic wounds is yet to be elucidated. Ferroptosis and ferritinophagy observed in diabetes are due to iron metabolism disorders, which are directly associated with the initiation and progression of diabetes. Herein, we reveal that senescent fibroblasts in diabetic wounds are resistant to ferroptosis and that impaired ferritinophagy may be a contributing cause. Further, the expression of NCOA4, a key factor that influences ferritinophagy, is decreased in both diabetic wound tissue and high glucose-induced senescent fibroblasts. Moreover, NCOA4 overexpression could render senescent fibroblasts more vulnerable to ferroptosis. A faster wound healing process was also linked to the induction of ferroptosis. Thus, resistance to ferroptosis impedes the removal of senescent fibroblasts; promoting ferritinophagy could reverse this process, which may have significant implications for the management of diabetic wounds.

Abstract Background Medicinal plants have always played an important role in the history of human health. However, the populations and sustainable use of medicinal plants have been severely affected by human activities and climate change. Little is known about the current conservation status and distribution pattern of medicinal plants. In this study, based on accurate geographical distribution information of 9756 medicinal plants, we identified diversity hotspots and conservation gaps, evaluated conservation effectiveness of nature reserves, and predicted suitable habitat areas for medicinal plants in China to provide scientific guidance for their long-term conservation and sustainable use. Results A total of 150 diversity hotspot grid cells, mainly concentrated in central and southern China, were identified. These only accounted for 5% of the total distribution area but contained 96% of the medicinal plants of the country. The hotspot grid cells included all traditional hotspot areas, but we also detected three new hotspots, namely Mufu-Lushan Mountains, Tianshan-Altai Mountains, and Changbai Mountains. The current national and provincial nature reserves protect 125 hotspot grid cells, which harbor 94% of all medicinal plants. However, 25 hotspot grid cells, distributed in the Tianshan-Altai Mountains and Hengduan Mountains, are located outside the national and provincial nature reserves. An analysis of the predicted effects of climate change indicated that the suitable habitat areas will shift from southern to northern China, and that southern China will face a considerable loss of suitable habitat areas, while the east and west parts of China will encompass remarkably more suitable habitat areas in the future. Conclusions The current conservation networks have achieved high conservation effectiveness with regard to medicinal plants; however, the conservation gaps we identified should not be neglected, and conservation planning needs to take into account the predicted shifts of some hotspots of medicinal plants due to climate change.

China is known for its abundant plant resources, but biodiversity conservation faces unprecedented challenges. To provide feasible suggestions for sustainable conservation, we used the species richness algorithm and complementary algorithm to study distribution patterns of 34,082 seed plants based on 1,007,196 county-level distribution records. We reconstructed a phylogenetic tree for 95.35% of species and estimated the spatial phylogenetics, followed by correlation analyses between different distribution patterns. We identified 264 counties concentrated in southern and south-western mountainous areas as hotspots which covered 10% of the land area of China and harbored 85.22% of the Chinese seed plant species. The biodiversity conservation priorities we identified were highly representative as we have considered multiple conservation indicators. We evaluated the conservation effectiveness and gaps in the network of nature reserves and identified 31.44, 32.95, and 9.47%, respectively, of the hotspot counties as gaps in the national nature reserves, provincial nature reserves and both together, with respectively 55.77, 61.53, and 28.94% of the species. Analysis of the species composition showed there were a large number of threatened and endemic species occurring in the nature reserves’ gaps. The conservation gaps need to be filled by establishing new nature reserves or national parks, especially in south-western China, and more attentions should be paid to strengthen the conservation of specific plant taxa due to the apparent mismatches between different distribution patterns.

Nanofiber scaffolds have emerged as a revolutionary drug delivery platform for promoting wound healing, due to their unique properties, including high surface area, interconnected porosity, excellent breathability, and moisture absorption, as well as their spatial structure which mimics the extracellular matrix. However, the use of nanofibers to achieve controlled drug loading and release still presents many challenges, with ongoing research still exploring how to load drugs onto nanofiber scaffolds without loss of activity and how to control their release in a specific spatiotemporal manner. This comprehensive study systematically reviews the applications and recent advances related to drug-laden nanofiber scaffolds for skin-wound management. First, we introduce commonly used methods for nanofiber preparation, including electrostatic spinning, sol–gel, molecular self-assembly, thermally induced phase separation, and 3D-printing techniques. Next, we summarize the polymers used in the preparation of nanofibers and drug delivery methods utilizing nanofiber scaffolds. We then review the application of drug-loaded nanofiber scaffolds for wound healing, considering the different stages of wound healing in which the drug acts. Finally, we briefly describe stimulus-responsive drug delivery schemes for nanofiber scaffolds, as well as other exciting drug delivery systems.

Abstract Large river valleys (LRVs) are heterogeneous in habitat and rich in biodiversity, but they are largely overlooked in policies that prioritize conservation. Here, we aimed to identify plant diversity hotspots along LRVs based on species richness and spatial phylogenetics, evaluate current conservation effectiveness, determine gaps in the conservation networks, and offer suggestions for prioritizing conservation. We divided the study region into 50 km × 50 km grid cells and determined the distribution patterns of seed plants by studying 124,927 occurrence points belonging to 14,481 species, using different algorithms. We generated phylogenies for the plants using the “V. PhyloMaker” R package, determined spatial phylogenetics, and conducted correlation analyses between different distribution patterns and spatial phylogenetics. We evaluated the effectiveness of current conservation practices and discovered gaps of hotspots within the conservation networks. In the process, we identified 36 grid cells as hotspots (covering 10% of the total area) that contained 83.4% of the species. Fifty‐eight percent of the hotspot area falls under the protection of national nature reserves (NNRs) and 83% falls under national and provincial nature reserves (NRs), with 42% of the area identified as conservation gaps of NNRs and 17% of the area as gaps of NRs. The hotspots contained high proportions of endemic and threatened species, as did conservation gaps. Therefore, it is necessary to optimize the layout of current conservation networks, establish micro‐nature reserves, conduct targeted conservation priority planning focused on specific plant groups, and promote conservation awareness. Our results show that the conservation of three hotspots in Southwest China, in particular, is likely to positively affect the protection of biodiversity in the LRVs, especially with the participation of the neighboring countries, India, Myanmar, and Laos.

Abstract The Yangtze River Basin (YRB), a key biodiversity area and a major economic zone in China, the biodiversity of the area is confronted with severe challenges. In this paper, we analyzed species distribution patterns based on a dataset with 18,538 seed plant species to identify hotspots and evaluate conservation effectiveness and gaps of the YRB. We calculated distribution patterns of species richness with the top 5% richness algorithm and complementarity algorithm, and reconstructed a phylogenetic tree and elucidated spatial phylogenetics. There were 214 hotspot grid cells covering only 6.8% of the study region, but containing 88% seed plant species. Then, we conducted correlation analysis on distribution patterns of different algorithms and species categories, which showed moderate or weak correlations between the top 5% richness algorithm and complementary algorithm. Conservation effectiveness analysis indicated there were 116 hotspot grid cells (accounting for 73.6% of the total species) protected by current conservation networks and there are many conservation gaps remaining. Finally, we used the maximum entropy model (MaxEnt) to predict suitable habitat areas of threatened species under current and future climate scenarios. Prediction analysis results indicated future expansion of suitable habitat to the southeast, and reduction in the central and western portions of the study area. When we considered anthropogenic areas, the suitable habitat was severely decreased, indicating the importance of optimizing the layout of conservation networks and comprehensive biodiversity conservation planning in the YRB.

The Qinghai-Tibetan Plateau (QTP) harbors abundant and diverse plant life owing to its high habitat heterogeneity. However, the distribution pattern of biodiversity hotspots and their conservation status remain unclear. Based on 148,283 high-resolution occurrence coordinates of 13,450 seed plants, we identified hotspots by integrating data from species richness, species complementarity and spatial phylogenetics. Nine hotspot areas were identified that contained 89% of species but covered only 7% of the total land area of the QTP. Four of nine hotspots were identified firstly, including west Nyainqentanglha Mountains, the middle reaches of Lancang and Jinsha Rivers, the upper reaches of Yellow River and Qilian Mountains. Analysis of conservation efficiency indicated national nature reserves (NNRs) covered 55% of the hotspots, whereas NNRs and provincial nature reserves (PNRs) together protected 73% of the hotspots. Conservation efforts, such as establishing new protected areas and upgrading the level of existing nature reserves, should be strengthened in the conservation gaps. Targeted conservation should be carried out for species endemic to QTP due to their narrow distribution range and low conservation effectiveness. Niche modeling for 336 threatened plants indicated there were apparent range shifts of suitable habitat areas from the eastern edge to the center of the plateau under future climate scenarios, and conservation priority should be focused on the southern QTP for where have stable habitats.

Advanced glycation end products (AGEs), produced by the non-enzymatic glycation of proteins and lipids under hyperglycemia or oxidative stress conditions, has been implicated to be pivotal in the development of diabetic vascular complications, including diabetic retinopathy. We previously demonstrated that Src kinase played a causative role in AGE-induced hyper-permeability and barrier dysfunction in human umbilical vein endothelial cells (HUVECs). While the increase of vascular permeability is the early event of angiogenesis, the effect of Src in AGE-induced angiogenesis and the mechanism has not been completely revealed. Here, we investigated the impact of Src on AGE-induced HUVECs proliferation, migration, and tubulogenesis. Inhibition of Src with inhibitor PP2 or siRNA decreased AGE-induced migration and tubulogenesis of HUVECs. The inactivation of Src with pcDNA3/flag-SrcK298M also restrained AGE-induced HUVECs proliferation, migration, and tube formation, while the activation of Src with pcDNA3/flag-SrcY530F enhanced HUVECs angiogenesis alone and exacerbated AGE-induced angiogenesis. AGE-enhanced HUVECs angiogenesis in vitro was accompanied with the phosphorylation of ERK in HUVECs. The inhibition of ERK with its inhibitor PD98059 decreased AGE-induced HUVECs angiogenesis. Furthermore, the inhibition and silencing of Src suppressed the AGE-induced ERK activation. And the silencing of AGEs receptor (RAGE) inhibited the AGE-induced ERK activation and angiogenesis as well. In conclusions, this study demonstrated that Src plays a pivotal role in AGE-promoted HUVECs angiogenesis by phosphorylating ERK, and very likely through RAGE-Src-ERK pathway.
