The tumor immunosuppressive microenvironment (IME) significantly affects tumor occurrence, progression, and prognosis, but the underlying molecular mechanisms remain to make known. We investigated the prognostic significance of PDPN and its role in IME in glioma. Weighted gene co-expression network analysis (WGCNA) found PDPN closely related to IDH wildtype status and higher immune score. Correlation analysis suggested PDPN was highly positively relevant to immune checkpoints expression and immune checkpoints block responding status. Correlation analysis together with verification in vitro suggested PDPN highly positively relevant tumor-associated neutrophils (TANs) and tumor-associated macrophages (TAMs). Least absolute shrinkage and selection operator (LASSO) regression employed to develop the prediction model with TANs and TAMs markers showed that high risk scores predicted worse prognosis. We highlight that PDPN overexpression is an independent prognostic indicator, and promotes macrophage M2 polarization and neutrophil degranulation, ultimately devotes to the formation of an immunosuppressive tumor microenvironment. Our findings contribute to re-recognizing the role of PDPN in IDH wildtype gliomas and implicate promising target therapy combined with immunotherapy for this highly malignant tumor.

Modern engine is a typical multi-objective control system. This paper proposed a multi-objective Bayesian optimization strategy to deal with the performance optimization for diesel engines. Since the objective functions of diesel engines are complicated and computationally expensive, Gaussian processes(GPs) are constructed by using the data collected from the diesel engine to approximate the real objective functions. Non-dominated sorting genetic algorithm II(NSGAII) leverages the Gaussian process to generate the Pareto-optimal solutions. The Gaussian process will be updated iteratively by Bayesian posterior information, which increases the reliability of the models. The acquisition function Expected Hyper Volume Improvement(EHVI), which can balance the trade-off between exploration and exploitation throughout the optimization process, is used to select the solutions for real computationally expensive multi-objective evaluation. The proposed algorithm is applied on a diesel engine, which shows its reliability and high efficiency. The metrics hypervolume(HV) and the control results demonstrate that the proposed algorithm has outstanding effects for performance optimization of diesel engine airpath.

In this research, all the efforts, based on a series of molecular dynamics simulations on the interfacial process between VOC-contaminated air and acidic sulfate, were made to find how the structures and properties of VOCs are related to the formation of SOAs. The experimental fractional aerosol coefficients (FACs) were used to quantify the SOA formation and 14 VOC species were chosen based on the atmosphere inventory and the FAC magnitude. Finally, the quantitative relationship (QR) was found through the FAC as a function of the two variables the total valid interactions (Tg) and the diffusion coefficient (D), with R square 0.94. Meanwhile, the effect of water was explored and the QR was proved to be rational and reliable. The QR not only explained the SOA formation capacity of VOCs, but could also predict the SOA formation of new molecules.

This article aims to design an intelligent mobile charging control mechanism for electric vehicles (EVs), by promoting charging reservations (including service start time, expected charging time, and charging location). EV mobile charging could be implemented as an alternative recharging solution, wherein charge replenishment is provided by economically mobile plug-in chargers, capable of providing on-site charging services. With intelligent charging management, readily available mobile chargers are predictable and could be efficiently scheduled toward EVs with charging demand, based on updated context collected from across the charging network. The context can include critical information relating to charging sessions and charging demand. Furthermore, with reservations introduced, accurate estimations on charging demand for a future moment are achievable, and correspondingly, optimal mobile chargers selection can be obtained. Therefore, charging demands across the network can be efficiently and effectively satisfied, with the support of intelligent system-level decisions. In order to evaluate critical performance attributes, we further carry out extensive simulation experiments with practical concerns to verify our insights observed from the theoretical analysis. Results show great performance gains by promoting the reservation-based mobile charger selection, especially for mobile chargers equipped with suffice power capacity. Note to Practitioners —The convenience of charging service is one major concern for EVs, especially when an urgent charging is required while none charging points are reachable. Recently, a Chinese EV company (NIO, Inc., Shanghai, China) is promoting its mobile charger (ES8 model) to Tesla. Driven by such market trend, this article proposes an efficient approach toward intelligent scheduling of mobile chargers toward parked EVs. Different from fixed charging stations focusing on the problem of long waiting times, the proposed solution is applicable to charging-on-demand with precharging appointment at mobile chargers. Preliminary experiments show great charging efficiency achieved by concerning the issue of where to reserve, i.e., the consideration of optimal selection on mobile chargers. Such mobile charging services can coexist with the governmental or pilots’ initiated charging station deployment. However, future research will need to evaluate the holistic service platform.

Herein, using global TF target gene sets, we comprehensively investigated their relationship with prognosis and potential biological effect in lower-grade glioma (LGG). We aimed to develop a less-biased prognostic model and provide new insight for personalized management of this disease.Methods: TF target gene sets were collected from MSigDB and GRID database followed by ssGSEA calculating normalized enrichment score. Comprehensive survival analysis was combined with Kaplan–Meier and Cox algorithms. Consensus cluster and lasso regression were performed to develop prognostic signatures with validation of ROC and independent external cohort. Approaches of xCell/CIBERSORT/TIMER were involved in analyzing the immune microenvironment. We also correlated identified prognostic signatures with tumor mutational burden (TMB) and m6A genes.Results: Fourteen TFs were significantly screened based on survival. Patients were classified into 2 prognosis-related clusters based on 14-TFs features. The function of differentially expressed TF target genes between Cluster1/2 was enriched mostly on glioma invasion/migration. The prognostic model was trained by 6 out of 14-TFs followed by generating risk-score as an independent prognostic indicator. We found differences between the high/low-risk group in TMB and the immune microenvironment, where the high-risk group represented “hot-tumor”. Besides, 6-TFs were correlated with m6A regulation genes.Conclusion: Our findings suggested that the 6-TFs model could be used to predict prognosis and predict the status of the immune microenvironment in LGG.Keywords: transcription factor target gene set, lower-grade glioma, prognostic model, tumor immune microenvironment, risk signature

Background Intracranial aneurysm (IA) is a serious disease. Analyze and review the cases of anterior circulation ruptured IA by supraorbital lateral keyhole approach, and summarize the experiences of this approach. Methods Retrospective analysis of 16 cases of ruptured anterior circulation IA in our department from January 2019 to June 2020, CT angiography (CTA) was performed before operation. Analyzing the IA’s parameters by 3D-CT reconstruction. The IA was clipped by supraorbital lateral keyhole approach combined with the 3D-skull reconstruction. Extraventricular drainage was performed before craniotomy. Intraoperative neurophysiological monitoring was performed during the operation. After operation, fluorescein angiography and vascular ultrasound were performed to check the clipping effect. Intracranial pressure monitor was performed postoperatively. CTA was reexamined one week after operation. The modified Rankin Scale (MRS) was performed 6 months after operation. Results There were 7 males (43.8%) and 9 females (56.2%), and the average age is 52.31 ± 11.12 years old. Among them, 11 patients (68.8%) were anterior communicating artery aneurysms and 5 (31.2%) were middle cerebral artery aneurysms. All patients were out of hospital within 10 days without any death, without cerebral infarction, cerebrospinal fluid leakage and neurological impairments. About mRS score, after 6 months follow-up, 8 cases (50%) had 0 point, 4 cases (25%) had 1 point, and 4 cases (25%) had 2 points. Conclusions For ruptured anterior circulation IA, the supraorbital lateral keyhole approach combined with ventricular drainage, intraoperative electrophysiological monitoring, and intraoperative vascular ultrasound is a safe and minimally invasive treatment. The application of reconstruction clipping can reconstruct the diameter of parent vessel and reduce the recurrence rate of IA.

BACKGROUND Dysplastic gangliocytoma of the cerebellum (Lhermitte-Duclos disease) is an extremely rare, slow-growing hereditary mass lesion that is mainly characterized by both specific neuroradiological features and secondary hydrocephalus. Patients may present with symptoms of cerebellar mass lesion and increased intracranial pressure. As an important part of Cowden syndrome, Lhermitte-Duclos disease in adults is typically marked by PTEN gene mutation. OBSERVATIONS The clinical management of a 31-year-old woman who suffered Lhermitte-Duclos disease was introduced in this case report. Subtotal resection was performed with the assistance of intraoperative sonography to relieve obstructive hydrocephalus, and prophylactic C1 laminectomy was performed to prevent possible postoperative progression of the residual lesion. Perioperative care and surgical process were clearly revealed in an accompanying video. Intraoperative sonography of Lhermitte-Duclos disease presents hyperechoic distorted thickening cortices surrounded by hypoechoic edema belt. The patient did not report any significant neurological complications or sequelae after the lesion resection. LESSONS The authors first reported the use of intraoperative sonography in resection of adult-onset Lhermitte-Duclos disease. Hopefully, the educative case report can provide a feasible experience in the diagnosis and treatment of Lhermitte-Duclos disease.

ACT001, which is derived from an ancient anti-inflammatory drug, has been shown to cross the blood-brain barrier in preclinical studies and has demonstrated anti-glioblastoma (GBM) activity in clinical trials. However, its pharmacological potential for anti-GBM immune response modulation remains unclear. The chemical structure of ACT001 indicates that it may bind to STAT3 and thus modulate antitumor immune response. Methods: Bioinformatics and immunohistochemistry (IHC) were used to assess STAT3 and PD-L1 expression in gliomas. Western blotting, RT-PCR and immunofluorescence were used to detect PD-L1 and p-STAT3 expression in glioma cells exposed to ACT001. Chromatin immunoprecipitation, an ACT001-Biotin probe, and a dual-luciferase reporter assay were used to detect direct modulation. The in vivo efficacy of ACT001 was evaluated in GL261 murine glioma model. Survival analyses were conducted using the log-rank (Mantel-Cox) test. Results: Bioinformatic analysis of 1,837 samples from 4 public glioma datasets showed that STAT3 mRNA expression was correlated with the degree of malignancy and therapeutic resistance and that STAT3 mRNA expression was related to immunosuppression, leukocyte infiltration, and PD-L1 expression. IHC staining of 53 tissue samples confirmed that relatively high phosphorylated STAT3 and PD-L1 protein expression was associated with a relatively advanced World Health Organization (WHO) glioma grade. Next, we confirmed that ACT001 treatment reduced PD-L1 expression and STAT3 phosphorylation. An ACT001-biotin probe was used to verify that ACT001 bound to STAT3. We also demonstrated that STAT3 bound to the PD-L1 promoter. The inhibition of PD-L1 expression and STAT3 phosphorylation by ACT001 could be rescued by STAT3 overexpression. Additionally, ACT001 inhibited GBM growth and decreased PD-L1 expression in vivo. The expression of the M2 markers CD206 and CD163 was decreased, while that of the antitumor immune markers iNOS and IFNγ was increased by ACT001 in vivo. Conclusion: Our results demonstrate that STAT3 plays a key role in immunosuppression of glioma and is inhibited by ACT001. ACT001 inhibits PD-L1 transcription and modulates anti-tumor immune response in glioma bearing mice. These findings will help us to understand the mechanism of ACT001 in GBM therapy.

In recent years, research on glioma immunotherapy have grown rapidly. However, the autoimmune‐like side effects that are caused by blocking immunological checkpoints hinder their clinical application in gliomas currently. Galectin‐9, a ligand for T‐cell immunoglobulin mucin 3, has shed a new light on the treatment of malignant glioma. However, the potential mechanism of Galectin‐9 is still under discussion. In this study, first, we methodically gathered 1,027 glioma patients with RNA‐seq and 986 patients with survival data to explore the role and mechanism of Galectin‐9 in gliomas. Second, we analyzed glioma samples from 50 patients in the Department of Neurosurgery, Tianjin Medical University General Hospital. Finally, we found that Galectin‐9 was strongly upregulated in glioblastoma multiforme compared with normal brain tissues and lower‐grade glioma. Patients with Galectin‐9 overexpression had a significantly shorter overall survival. Moreover, the tissue microarray data displayed that the expression of Galectin‐9 in the core of tumor is higher than that in the border and was correlated with the shorter survival in glioma patients. Galectin‐9 is more highly expressed in the mesenchymal subtype of glioblastoma multiforme than in the other subtypes. Simultaneously, Galectin‐9 was closely associated with the immune response and lymphocyte activation, especially T‐cell activation. To further determine the underlying role of Galectin‐9 in the immune response, we selected seven immune metagenes. Through cluster analysis and correlation analysis, we discovered that Galectin‐9 was highly correlated with immune checkpoint molecules and M2 tumor‐associated macrophages. 2,3-Indolinedione derivatives have been identified as a novel class of promising agents for cancer treatment. In this study, eighteen 2,3-indolinedione derivatives were designed and synthesized, and their anticancer activities against mantle cell lymphoma (MCL) cells were evaluated. Most of them exhibited significant antiproliferative activity against the tested cell lines, and compound K5 was the most potent (MCL cellular IC50 = 0.4–0.7 μM). Further, compound K5 could induce cell apoptosis and cell cycle arrest in G2/M phase. Additionally, the results of drug-likeness analysis demonstrated that these novel 2,3-indolinedione derivatives could have potential as novel treatment strategies for MCL.

Glioblastoma is the most common primary intracranial malignant tumor in adults and has high morbidity and high mortality. TMEM158 has been reported to promote the progression of solid tumors. However, its potential role in glioma is still unclear. Here, we found that TMEM158 expression in human glioma cells in the tumor core was significantly higher than that in noncancerous cells at the tumor edge using bioinformatics analysis. Cancer cells in patients with primary GBMs harbored significantly higher expression of TMEM158 than those in patients with WHO grade II or III gliomas. Interestingly, regardless of tumor grading, human glioma samples that were IDH1-wild-type (IDH1-WT) exhibited higher expression of TMEM158 than those with IDH1-mutant (IDH1-Mut). We also illustrated that TMEM158 mRNA expression was correlated with poor overall survival in glioma patients. Furthermore, we demonstrated that silencing TMEM158 inhibited the proliferation of glioma cells and that TMEM158 overexpression promoted the migration and invasion of glioma cells by stimulating the EMT process. We found that the underlying mechanism involves STAT3 activation mediating TMEM158-driven glioma progression. In vivo results further confirmed the inhibitory effect of the TMEM158 downregulation on glioma growth. Collectively, these findings further our understanding of the oncogenic function of TMEM158 in gliomas, which represents a potential therapeutic target, especially for GBMs.

Background. Glioma is the most common primary intracranial tumor and is associated with poor prognosis. Identifying effective biomarkers for glioma is particularly important. MXRA5, a secreted glycoprotein, is involved in cell adhesion and extracellular matrix remodeling and has been reported to be expressed in many cancers. However, the role and mechanism of action of MXRA5 in gliomas remain unclear. This study was aimed at investigating the role of MXRA5 at the transcriptome level and its clinical prognostic value. Methods. In this study, RNA microarray data of 301 glioma patients from the Chinese Glioma Genome Atlas (CGGA) were collected as a training cohort and RNA-seq data of 702 glioma samples from The Cancer Genome Atlas (TCGA) were used for validation. We analyzed the clinical and molecular characteristics as well as the prognostic value of MXRA5 in glioma. In addition, the expression level of MXRA was evaluated in 28 glioma tissue samples. Results. We found that MXRA5 expression was significantly upregulated in high-grade gliomas and IDH wild-type gliomas compared to controls. Receiver operating characteristic (ROC) analysis showed that MXRA5 is a potential marker of the mesenchymal subtype of glioblastoma multiforme (GBM). We found that MXRA5 expression is highly correlated with immune checkpoint molecule expression levels and tumor-associated macrophage infiltration. High MXRA5 expression could be used as an independent indicator of poor prognosis in glioma patients. Conclusion. Our study suggests that MXRA5 expression is associated with the clinicopathologic features and poor prognosis of gliomas. MXRA5 may play an important role in the immunosuppressive microenvironment of glioma. As a secreted glycoprotein, MXRA5 is a potential circulating biomarker for glioma, deserving further investigation.

Gliomas are the most common primary brain cancer. While it has been known that calcium-related genes correlate with gliomagenesis, the relationship between calcium-related genes and glioma prognosis remains unclear. We assessed TCGA datasets of mRNA expressions with differentially expressed genes (DEGs) and enrichment analysis to specifically screen for genes that regulate or are affected by calcium levels. We then correlated the identified calcium-related genes with unsupervised/supervised learning to classify glioma patients into 2 risk groups. We also correlated our identified genes with immune signatures. As a result, we discovered 460 calcium genes and 35 calcium key genes that were associated with OS. There were 13 DEGs between Clusters 1 and 2 with different OS. At the same time, 10 calcium hub genes (CHGs) signature model were constructed using supervised learning, and the prognostic risk scores of the 3 cohorts of samples were calculated. The risk score was confirmed as an independent predictor of prognosis. Immune enrichment analysis revealed an immunosuppressive tumor microenvironment with upregulation of checkpoint markers in the high-risk group. Finally, a nomogram was generated with risk scores and other clinical prognostic independent indicators to quantify prognosis. Our findings suggest that calcium-related gene expression patterns could be applicable to predict prognosis and predict levels of immunosuppression.

The secondary effluent of the wastewater treatment plant is considered as one of the reused water sources and needs advanced treatment to meet increasingly stringent water treatment standards. Ultrafiltration, as one of the most widely used advanced treatment technologies, is limited due to membrane fouling, and coagulation and pre-oxidation have received extensive attention as pretreatment methods to alleviate membrane fouling. This research proposes a new method of Fe(II)-activated peroxymonosulfate (PMS) coagulation and a pre-oxidation system coupled with ultrafiltration (UF) to treat secondary effluent from sewage plants, separately evaluating the treatment effect under different molar ratios of Fe(II)/PMS. The Fe(II)/PMS decontamination mechanism and membrane fouling control effect were elucidated through pollutant removal efficiency, membrane morphology, membrane flux trend, and membrane fouling resistance distribution. According to the experimental results, the optimal effect of organic matter removal and membrane fouling mitigation was achieved at the Fe(II)/PMS dosage of 60/60 μM/μM (molar ratio 1:1). The efficiency of pretreatment methods in removing organics and fluorescent components and mitigating membrane fouling followed the order of Fe(II)/PMS > Fe(III) > inactivated PMS. Fe(II)/PMS could produce a synergistic effect in a high concentration state (60 μM), relying on the dual effects of coagulation and oxidation to alleviate membrane fouling. Coagulation and pre-oxidation by Fe(II)/PMS significantly reduced the clogging of membrane pores and the proportion of irreversible resistance, effectively controlling membrane fouling and improving effluent quality. SEM images further confirmed its effectiveness, and EPR results unequivocally indicated that its synergistic mechanism was mediated by •OH and SO4•−. The research results can provide ideas for advanced wastewater treatment and secondary effluent reuse.

The production planning of bearing cap directly affects whether the production rhythm of the process is coordinated or not. Based on the analysis of production scheduling objectives and constraints of flexible automatic production line for bearing cap, a mathematical model for production planning of bearing cap is established, and the mathematical model is solved. On the premise of defining the production date and corresponding production volume of production orders on each production line, the method of production planning for on-site day is improved based on heuristic method. The production planning optimization algorithm divides the production order and determines the order of the production order. According to the sorting result, the delivery time can be calculated to determine whether the work order is produced or not. The validity of the mathematical model and the optimization algorithm are proved by experiments.

Inhibitor of nuclear factor kappa‐B kinase subunit epsilon (IKBKE) plays critical roles in the proliferation, invasion, and epithelial‐mesenchymal transition (EMT) of glioblastoma (GBM). However, as an immune response factor, few studies have focused on the role of IKBKE in the glioblastoma‐mediated immunosuppressive microenvironment. Here, we found a higher IKBKE expression level in gliomas corresponding to higher malignancy of the tumor. The highest level of IKBKE expression was examined in the core region of GBM tissues as well as the mesenchymal subtype, which are featured with necrosis, immunocyte infiltration, and immunosuppression. Further in silico analysis demonstrated that the JAK/STAT as the signaling pathway most associated with IKBKE in mesenchymal GBM. The co‐expression patterns of IKBKE, pSTAT3, and PD‐L1 were detected within GBM tissues. Mechanistically, IKBKE could interact with STAT3 and thus enhancing the phosphorylation level of STAT3 and its nuclear translocation. In addition, pSTAT3 could transcriptionally regulate the expression of PD‐L1 through binding to its promoter. In vivo results further confirmed the inhibitory effect of the IKBKE downregulation on tumor growth. Collectively, our findings suggest IKBKE as the central node in the crosstalk between NF‐κB and STAT3 signaling within mesenchymal GBM. Targeting GBM through inhibiting IKBKE could restrain tumor growth and tumor‐mediated immunosuppressive environment., 1. Higher IKBKE expression was detected in higher grades of glioma. 2. IKBKE is enriched in mesenchymal GBM, which confers immunosuppressive signature. 3. IKBKE correlates with JAK/STAT signaling in MES GBM. 4. Background/Aims: Jagged1 is the ligands of the Notch signaling and has been shown to promote glioma-initiating cells (GICs) in glioblastoma. The role of Jagged1 in GICs invasion and underlying molecular mechanisms remain unclear. Methods: Survival data from R2 genomics analysis, the Cancer Genome Atlas (TCGA), the Chinese Glioma Genome Atlas (CGGA) and visualization platform database were used to evaluate the effects of Jagged1 on overall patient survival. we investigated Jagged1 induced the GICs cells’ invasion by matrix degradation assays and Transwell cell invasion assays in vitro, then we further explored the underlying molecular mechanisms using Co-immunoprecipitation (co-IP) analysis. Results: High expression of Jagged1 in human glioma was associated with poor survival. Clinical data analysis showed that the Jagged1 was positively correlated with NF-κB(p65). Jagged1-induced invasion of GICs cells through activation of NF-κB(p65) pathway. In vivo, knockdown of Jagged1 could suppress the tumorigenicity of GICs cells through NF-κB(p65) signaling. Conclusion: Insights gained from these findings suggest that Jagged1 plays an important oncogenic role in GICs malignancy by activation of NF-κB(p65) signaling, and Jagged1 could be employed as an effective therapeutic target for GICs.

Glioblastoma is the most common and lethal primary intracranial tumor. As the key regulator of tumor cell volume, sodium‐potassium‐chloride cotransporter 1 (NKCC1) expression increases along with the malignancy of the glioma, and NKCC1 has been implicated in glioblastoma invasion. However, little is known about the role of NKCC1 in the epithelial‐mesenchymal transition‐like process in gliomas. We noticed that aberrantly elevated expression of NKCC1 leads to changes in the shape, polarity, and adhesion of cells in glioma. Here, we investigated whether NKCC1 promotes an epithelial–mesenchymal transition (EMT)‐like process in gliomas via the RhoA and Rac1 signaling pathways. Pharmacological inhibition and knockdown of NKCC1 both decrease the expressions of mesenchymal markers, such as N‐cadherin, vimentin, and snail, whereas these treatments increase the expression of the epithelial marker E‐cadherin. These findings indicate that NKCC1 promotes an EMT‐like process in gliomas. The underlying mechanism is the facilitation of the binding of Rac1 and RhoA to GTP by NKCC1, which results in a significant enhancement of the EMT‐like process. Specific inhibition or knockdown of NKCC1 both attenuate activated Rac1 and RhoA, and the pharmacological inhibitions of Rac1 and RhoA both impair the invasion and migration abilities of gliomas. Furthermore, we illustrated that NKCC1 knockdown abolished the dissemination and spread of glioma cells in a nude mouse intracranial model. These findings suggest that elevated NKCC1 activity acts in the regulation of an EMT‐like process in gliomas, and thus provides a novel therapeutic strategy for targeting the invasiveness of gliomas, which might help to inhibit the spread of malignant intracranial tumors.

Gravity-driven membrane filtration (GDM) is promising for decentralized rainwater recycling, owing to low maintenance and energy consumption. However, the organic removal by GDM process is sometimes undesirable and the quality of the permeate cannot meet the standard of water reuse. To improve this, granular activate carbon (GAC) was added as a particle layer on the membrane surface of GDM system. Additionally, a system with sand addition and a system with no particle addition were trialed as comparisons, to study the combined effects of particle hindering and adsorption on the removal efficacy of organics and the development of permeate flux. Results showed that GDM with a GAC layer improved removal efficiency of organics by 25%, and that GAC enhanced removal of florescent compounds (e.g., aromatic proteins, tryptophan proteins and humics), compared with the other two systems. Additionally, the permeate flux in three systems stabilized after Day 25, and kept stable until the end of the operation. However, the presence of GAC layer decreased the level of stable flux (3.2 L/m2h) compared with the control system (4.5 L/m2h). The factors responsible for the lower flux and severe membrane fouling in GAC layer assisted system were the combined effects of particle and adsorption which led to a denser bio-fouling layer with higher amount of biomass and extracellular polymeric substances contents (proteins and polysaccharides). Resistance distribution analyses revealed that GAC layer mainly increased hydraulically reversible resistance (occupied 93%) of the total resistance, indicating that the flux could be recovered easily by simple physical cleaning.

The investigation and development of recently introduced agents or radiological measurements caused emergent misunderstandings to the response assessment of glioma. To date, the classical Macdonald criteria and the response assessment of neuro-oncology (RANO) criteria have been used successively for the evaluation of glioma outcome. However, ongoing efforts on complementary assessments are necessary to combat this malignancy. In this review, we highlight the shortcomings of the current criteria and introduce the initiative effort of RANO guideline and its offspring. We also discuss some future barriers for accurate assessment of treatment response in glioma.

Glioblastoma (GBM) invasion and migration are key biological behaviors leading to refractoriness to current therapies and infiltration into the non-tumor brain parenchyma. GBM cell migration is strongly dependent on tumor architecture in vivo, which is absent in traditional two-dimensional (2D) monolayer culture. The present study applied a three-dimensional (3D) hydrogel model to rebuild the tumor architecture in vitro. Treatment with NSC23766, a specific inhibitor of Ras-related C3 botulinum toxin substrate 1 (Rac1), inhibited the mesenchymal invasiveness however triggered the amoeboid motility called mesenchymal-amoeboid transition (MAT). Notably, NSC23766 stimulated U87 GBM cell migration in the 3D hydrogel. However, this compound inhibited cell motility in 2D monolayer culture without tumor architecture for MAT, suggesting the advantage of 3D hydrogel to investigate tumor cell invasion. Due to the inverse interaction of Rac1 and Ras homolog family member A (RhoA) signaling in the transition between mesenchymal and amoeboid morphology, simultaneous treatment of NSC23766 and Y27632 (selective Rho associated coiled-coil containing protein kinase 1 inhibitor), abolished U87 GBM cell migration through inhibiting MAT and amoeboid-mesenchymal transition. In addition, Y27632 induced integrin expression which gave rise to the focal adhesion to facilitate the mesenchymal invasion. The results of the present study demonstrated that the 3D hydrogel was a preferable model in vitro to study tumor cell invasion and migration. The combined inhibition of Rac1 and RhoA signaling would be a promising strategy to suppress GBM invasion.
