26. Development and characterization of microemulsion based topical gel of essential oil of clove (Syzygium aromaticum)for superficial fungal infections

Abstract

A widespread of superficial fungal infections deals with concerns related to current therapeutic regimen such as drug resistance and adverse events associated with the same which leads exploration of natural oils as an antifungal agent. The aim of present work is to the development of clove oil loaded microemulsion based gel for treatment of superficial fungal infections. The microemulsion based gel was prepared by phase titration method and optimized using D-optimal design considering globule size, drug permeation and drug retention on skin as critical quality attributes. The MIC and zone of inhibition of clove oil was found to be 2.2 mg/ml and 38 mm respectively. A pale to yellowish transparent microemulsion had a globule size, zeta-potential and PDI of 14.41 nm, 0.73 and 0.0113 respectively indicating a stable microemulsion. A clove oil loaded microemulsion based gel (CLMBG) with a pH of 6.27 and viscosity of 12.87 m.pas/sec exhibited a comparable texture profile to marketed preparation. The drug release of the CLMBG with a drug content of 102.6 ± 4% in acetate buffer pH 5.5 was 98.5 ± 0.35% ensuring complete drug release from the formulation. Ex-vivo drug permeation study and skin irritation study dictated the retention of drug at site of action and the formulation to be non-irritant. The antifungal study proved the formulation to have similar efficacy as the marketed product (clobet gel). The stability study indicated the product to be safe, efficacious and stable formulation. From the above results, it can be derived that clove oil loaded microemulsion based gel can be a promising alternative to current antifungal regimens.

Abstract

The objective of present experimental work was to optimize processing parameters of ocular liposomes using artificial neural network (ANN) as an optimization tool. The human eye is easily accessible site for topical dosing of drugs intended for local action or in the interior of the eye. Ganc iclovir (GCV) is a synthetic analogue of 2' - deoxy - guanosine used in treatment and prevention of cytomegalovirus infections and in treatment of ocular herpes infection (orphan disease). The poor bioavailability (5 - 9 %) of this drug has lead development of ocular liposome to improve the bioavailability and enhance therapeutic effectiveness. GCV loaded liposomes were prepared by classic al rotary flask evaporation method using HSPC/DSPG as lipid part and cho lesterol as vesicle stabilizer. Box - B ehnken design was applied on the selected processing parameters i.e. flask speed (X1), hydration volume (X2) and hydration te mperature (X3) whereas the formulation variables were kept constant. The feed forward back propagation neural network (BP NN ) method was optimized using experimental data and validated for accurate prediction of dependent variables. The obtained root mean square value of the tr a ined ANN model was 0.00004 , which indicated that the optimal model was reached. T he predicted data from ANN and the experimental data were compared by chi square test which showed no significant difference. The results conclude that the ANN model provides accurate prediction and should be further explored in optimization of novel pharmaceutical formulations.

Abstract

The objective of this study was to increase the bioavailability of fluvastatin by developing bilayered buccal mucoadhesive compacts. This study also focuses on the mucoadhesive potential of some natural gums for improved mucoadhesion and transmucosal permeation of existing mucoadhesive polymers by certain modifications. Bilayered mucoadhesive compacts with one layer of drug and mucoadhesive polymer, and second, non medicated, non permeable layer of ethylcellulose and magnesium stearate was prepared using direct compression technique. Natural gums like tamrind and xanthan gum were evaluated for its mucoadhesive properties. The mucoadhesion along with permeation character of chitosan was enhanced by immobilization of thiol groups on its surface utilizing 2-iminothiolane (Trauts reagent). The resulting chitosan-4thio-butylamidine conjugate (chitosan 4-thiobutylamidine (TBA) conjugate) was evaluated for its mucoadhesion, permeation and release properties. Experimental data revealed a several fold higher mucoadhesive property of chitosan-TBA conjugate than unmodified chitosan along with good permeation properties. Release studies revealed that the sustained release of fluvastatin over several hours may be obtained by combining the chitosan TBA conjugate with natural gums like xanthan and tamrind gum. Also, the bioavailability studies indicated that bioavailability of fluvastatin was enhanced using the aforementioned drug delivery system. Thus, the potential of the aforementioned drug delivery device is promising and may be considered as a novel tool in order to improve the therapeutic efficacy of various drugs with shorter half life and poor bioavailability. Abstract

Solubility & dissolution rate enhancement from solid oral dosage form is a key issue for current formulation and development. This review discusses, out of several techniques available, liquisolid system to improve dissolution rate of water insoluble drugs and to enhance dissolution rate of water soluble drugs. Different carriers and coating materials like Fujicalin®, Neusilin®, Avicel®, and Aerosil® can be used as carrier materials to prepare liquisolid system. Various non-volatile solvents like propylene glycol, liquid polyethylene glycols, polysorbates, glycerine, N,N-dimethylacetamide and fixed oils can be used to dissolve water insoluble molecules. Liquid drugs can be mixed directly with carriers to produce liquisolid systems. Liquisolid systems can be used to either enhance or retard drug release.
