Meiosis is a critical process in sexual reproduction, and errors during this cell division can significantly impact fertility. Successful meiosis relies on the coordinated action of numerous genes involved in DNA replication, strand breaks, and subsequent rejoining. DNA topoisomerase enzymes play a vital role by regulating DNA topology, alleviating tension during replication and transcription. To elucidate the specific function of DNA topoisomerase 1α ( A t T O P 1 α ) in male reproductive development of Arabidopsis thaliana , we investigated meiotic cell division in Arabidopsis flower buds. Combining cytological and biochemical techniques, we aimed to reveal the novel contribution of A t T O P 1 α to meiosis. Our results demonstrate that the absence of A t T O P 1 α leads to aberrant chromatin behavior during meiotic division. Specifically, the top1α1 mutant displayed altered heterochromatin distribution and clustered centromere signals at early meiotic stages. Additionally, this mutant exhibited disruptions in the distribution of 45s rDNA signals and a reduced frequency of chiasma formation during metaphase I, a crucial stage for genetic exchange. Furthermore, the atm-2×top1α1 double mutant displayed even more severe meiotic defects, including incomplete synapsis, DNA fragmentation, and the presence of polyads. Trigonella, commonly known as Fenugreek, is among the most promising medicinal herbs consumed worldwide due its protein rich dietary contributions. This study involved induced mutagenesis on two Trigonella species (Trigonella foenum-graecum var. PEB and Trigonella corniculata var. Pusa kasuri) using caffeine and sodium azide as mutagens, resulting in the identification of nine high-yielding mutant lines in the M3 generation. Molecular characterization using SCoT markers revealed a high polymorphism of 28.3% and 46.7% in PEB and Pusa kasuri, respectively, facilitating the investigation of genetic divergence among the control and mutant lines. Similarity correlation analysis indicated a high similarity between mutant A and mutant C (0.97) and between mutant J and mutant O (0.88), while the lowest similarity was observed between mutant B and mutant F (0.74) and between control and mutant L (0.58). Mutant F and Mutant J displayed the highest seed yield and its attributing traits, and seed protein content in PEB and Pusa kasuri, respectively. Physiological parameters, including chlorophyll content (Mutants A and N) and carotenoids (mutant A and J), exhibited improvements. Assessment of stomatal and seed characteristics using scanning electron microscopy may lead to improved physiological processes and distinction at the interspecific level, respectively. Methanolic extracts of the control and the mutant lines of both species were subjected to GC-MS analysis, revealing 24 major phytocompounds known for their pharmacological activities (antioxidant, anti-inflammatory, anticancer, etc.). Statistical methods such as Pearson correlation heatmap and pairwise scatter plot matrix provided insights into the correlations and linear associations among parameters for both PEB and Pusa kasuri. The strong correlation between iron content and seeds per pod in the mutant lines suggests a promising avenue for further research. Mutation breeding is one of the effective techniques used for improving desired traits such as yield quality and quantity in economic crops. The present study aims to develop oil and protein contents in addition to high yield attributes in soybean using gamma rays as a mutagen. Seeds of the soybean genotypes Giza 21, Giza 22, Giza 82, Giza 83 and 117 were treated with gamma rays doses 50, 100, 200 and 300 Gy. Plants were then scored based on morphological parameters correlated with yield quantity including plant height, seed weight and valuable protein and oil contents. Mutant lines exhibiting the highest yield attributes were selected and used as parents for M2 generation. The M2 progeny was further assessed based on their ability to maintain their yield attributes. Twenty mutant lines were selected and used as M3 lines. The yield parameters inferred a positive effect of gamma irradiation on the collected M3 mutant lines compared to their parental genotypes. 100 Gy of gamma rays gave the highest effect on the number of pods, branches and seeds per plant in addition to protein content, while 200 Gy was more effective in increasing plant height, number of pods per plant, and oil content. Six mutant lines scored the highest yield parameters. Further assessment inferred an inverse relationship between oil and protein content in most of the tested cultivars with high agronomic features. However, four mutant lines recorded high content of oil and protein besides their high seed yield as well, which elect them as potential candidates for large-scale evaluation. The correlation among examined parameters was further confirmed via principal component analysis (PCA), which inferred a positive correlation between the number of pods, branches, seeds, and seed weight. Conversely, oil and protein content were inversely correlated in most of yielded mutant lines. Together, those findings introduce novel soybean lines with favorable agronomic traits for the market. Water scarcity due to overuse and growing water pollution has led to the need for upgrading of conventional methods of wastewater treatment. The biological synthesis of zinc oxide nanoparticles (ZnO-NPs) and their photocatalytic capacity to degrade contaminants offer a promising and environment-friendly approach to municipal wastewater treatment. This technique is advantageous due to its cost-effectiveness, sustainability, and reduction in toxic residual substances. In this study, microbial-synthesized ZnO-NPs were used for the treatment of municipal wastewater. The objective of this study was to evaluate the potential of treated wastewater for wheat crop cultivation. Zinc oxide nanoparticles were synthesized from a pre-isolated bacterial strain, namely Shewanela sp., and characterized using UV-VIS, X-ray diffraction (XRD), scanning electron microscopy (SEM), and Fourier transform infrared spectroscopy (FTIR) analyses. The results showed that after the treatment of wastewater, the concentration of total dissolve solids (TDS), the chemical oxygen demand (COD), and sulfate and phosphate levels decreased by 76.5%, 57.1%, 81.1%, and 67.4%, respectively. However, the application of treated wastewater increased chlorophyll, carotenoids, and antioxidants by 45%, 40.8%, and 10.5 to 30.6%, respectively. Further, the application of treated wastewater also significantly decreased oxidative stress induced by hydrogen peroxide (H 2 O 2 ) and malondialdehyde (MDA) by 8.1% and 30.1%, respectively. In conclusion, biosynthesized ZnO-NPs could be an important choice to treat municipal wastewater and to improve wheat productivity.

The rise in global temperature also favors the multiplication of pests and pathogens, which calls into question global food security. Plants have developed special coping mechanisms since they are sessile and lack an immune system. These mechanisms use a variety of secondary metabolites as weapons to avoid obstacles, adapt to their changing environment, and survive in less-than-ideal circumstances. Plant secondary metabolites include phenolic compounds, alkaloids, glycosides, and terpenoids, which are stored in specialized structures such as latex, trichomes, resin ducts, etc. Secondary metabolites help the plants to be safe from biotic stressors, either by repelling them or attracting their enemies, or exerting toxic effects on them. Modern omics technologies enable the elucidation of the structural and functional properties of these metabolites along with their biosynthesis. A better understanding of the enzymatic regulations and molecular mechanisms aids in the exploitation of secondary metabolites in modern pest management approaches such as biopesticides and integrated pest management. The current review provides an overview of the major plant secondary metabolites that play significant roles in enhancing biotic stress tolerance. It examines their involvement in both indirect and direct defense mechanisms, as well as their storage within plant tissues. Additionally, this review explores the importance of metabolomics approaches in elucidating the significance of secondary metabolites in biotic stress tolerance. The application of metabolic engineering in breeding for biotic stress resistance is discussed, along with the exploitation of secondary metabolites for sustainable pest management.

A number of phytochemicals have been identified as promising drug molecules against a variety of diseases using an in-silico approach. The current research uses this approach to identify the phyto-derived drugs from Andrographis paniculata (Burm. f.) Wall. ex Nees (AP) for the treatment of diphtheria. In the present study, 18 bioactive molecules from Andrographis paniculata (obtained from the PubChem database) were docked against the diphtheria toxin using the AutoDock vina tool. Visualization of the top four molecules with the best dockscore, namely bisandrographolide (-10.4), andrographiside (-9.5), isoandrographolide (-9.4), and neoandrographolide (-9.1), helps gain a better understanding of the molecular interactions. Further screening using molecular dynamics simulation studies led to the identification of bisandrographolide and andrographiside as hit compounds. Investigation of pharmacokinetic properties, mainly ADMET, along with Lipinski's rule and binding affinity considerations, narrowed down the search for a potent drug to bisandrographolide, which was the only molecule to be negative for AMES toxicity. Thus, further modification of this compound followed by in vitro and in vivo studies can be used to examine itseffectiveness against diphtheria.

In recent years, the global agricultural system has been unfavorably impacted by adverse environmental changes. These changes in the climate, in turn, have altered the abiotic conditions of plants, affecting plant growth, physiology and production. Abiotic stress in plants is one of the main obstacles to global agricultural production and food security. Therefore, there is a need for the development of novel approaches to overcome these problems and achieve sustainability. Nanotechnology has emerged as one such novel approach to improve crop production, through the utilization of nanoscale products, such as nanofertilizer, nanofungicides, nanoherbicides and nanopesticides. Their ability to cross cellular barriers makes nanoparticles suitable for their application in agriculture. Since they are easily soluble, smaller, and effective for uptake by plants, nanoparticles are widely used as a modern agricultural tool. The implementation of nanoparticles has been found to be effective in improving the qualitative and quantitative aspects of crop production under various biotic and abiotic stress conditions. This review discusses various abiotic stresses to which plants are susceptible and highlights the importance of the application of nanoparticles in combating abiotic stress, in addition to the major physiological, biochemical and molecular-induced changes that can help plants tolerate stress conditions. It also addresses the potential environmental and health impacts as a result of the extensive use of nanoparticles.

Acremonium wilt disease affects grain quality and reduces sorghum yield around the globe. The present study aimed to assess the efficacy of humic acid (HA)-coated Fe 3 O 4 (Fe 3 O 4 /HA) nanoparticles (NPs) in controlling acremonium wilt disease and improving sorghum growth and yields. During the season 2019, twenty-one sorghum genotypes were screened to assess their response to Acremonium striticum via artificial infection under field conditions and each genotype was assigned to one of six groups, ranging from highly susceptible to highly resistant. Subsequently, over the two successive seasons 2020 and 2021, three different concentrations of 10, 40 and 80 mg L -1 of Fe 3 O 4 /HA NPs were tested against A. striticum . The concentrations of 40 and 80 mg L -1 were found to be highly effective in controlling acremonium wilt disease on different sorghum genotypes: LG1 (highly susceptible), Giza-3 (susceptible), and Local 119 (resistant) genotypes. After harvest, the physiological (growth and yield) and biochemical (peroxidase, catalase, and gibberellic acid) attributes of sorghum plants were determined, and the results demonstrated that concentrations of 40 and 80 mg L -1 increased peroxidase and catalase activities in healthy (uninoculated) sorghum genotypes compared to inoculated sorghum genotypes. Additionally, the toxicity of Fe 3 O 4 /HA NPs on male albino rats was investigated via hematological (CBC), chemical (ALT and AST) and histopathological analyses. The concentration 80 mg L -1 of Fe 3 O 4 /HA NPs caused a marked increase in ALT and creatinine level after 51 days of feeding. Severe pathological alterations were also observed in liver and kidney tissues of rats administered with grain sorghums treated with 80 mg L -1 . In comparison with the untreated control plants, a concentration of 40 mg L -1 significantly increased the growth, yield and gibberellic acid levels ( p ≤ 0.05) and was found to be safe in male albino rats. Conclusively, a concentration of 40 mg L -1 of Fe 3 O 4 /HA NPs showed promising results in curtailing A. striticum infections in sorghum, indicating its great potential to substitute harmful fertilizers and fungicides as a smart agriculture strategy.

Acquired immunodeficiency syndrome (AIDS) is a chronic and potentially fatal ailment caused by the human immunodeficiency virus (HIV) and remains a major health problem worldwide. In recent years, the research focus has shifted to a greater emphasis on complementing treatment regimens involving conventional antiretroviral (ARV) drug therapies with novel lead structures isolated from various marine organisms that have the potential to be utilized as therapeutics for the management of HIV-AIDS. The present review summarizes the recent developments regarding bioactive peptides sourced from various marine organisms. This includes a discussion encompassing the potential of these novel marine bioactive peptides with regard to antiretroviral activities against HIV, preparation, purification, and processing techniques, in addition to insight into the future trends with an emphasis on the potential of exploration and evaluation of novel peptides to be developed into effective antiretroviral drugs.

To explore the effects of triacontanol (TR) on drought tolerance of strawberry plants (cv Fertona), two field experiments were carried out to study the effects of three supplementary foliar TR rates (0, 0.5, and 1 ppm) under the following three levels of water irrigation: 11 m 3 /hectare (40% of water holding capacity (WHC) severe as a drought treatment, 22 m 3 /hectare (80% of WHC) as moderate drought stress, and normal irrigation with 27 m 3 /hectare (100% of WHC) server as a control treatment. TR treatments were applied five times after 30 days from transplanting and with 15-day intervals. The results showed that drought stress (40% and 80%) markedly decreased the growth, fruit yield, and chlorophyll reading, as well as the gas exchange parameters (net photosynthetic rate, stomatal conductance, and transpiration rate). Meanwhile, drought stress at a high rate obviously increased antioxidant enzyme activities such as superoxide dismutase (SOD), peroxidase (POX), and catalase (CAT) contents in the leaves of the strawberry plants. The moderate and high drought stress rates enhanced some strawberry fruit quality parameters such as total soluble solids (TSS), vitamin C, and anthocyanin content compared to the control. Additionally, TR increased the activities of SOD, POX, and CAT. TR treatment significantly increased the chlorophyll contents, gas exchange parameters (photosynthetic rate and stomatal conductance), and water use efficiency (WUE). Plant height, fruit weight, and total biomass were increased also via TR application. Total yield per plant was increased 12.7% using 1 ppm of TR compared with the control. In conclusion, our results suggested that TR application could relieve the adverse effects of drought stress on the growth of strawberry plants by enhancing the antioxidant enzymes, photosynthesis rate, and WUE of the leaves.

Determining the appropriate parents for breeding programs is the most important decision that plant breeders must make to maximize the genetic variability and produce excellent recombinant genotypes. Several methods are used to identify genotypes with desirable phenotypic features for breeding experiments. In this study, five kalanchoe genotypes were morphologically characterized by assessing plant height, number of inflorescences, number of flowers, flower length, flower diameter and number of petals. The analysis showed the distinction of yellow kalanchoe in the plant height trait, while the orange kalanchoe was distinguished in the number of inflorescences, the number of flowers and flower length traits, whereas the violet kalanchoe possessed the largest flower diameter and the highest number of petals. The molecular profiling was performed by random amplified polymorphism DNA (RAPD), inter-simple sequence repeats (ISSR) and start codon targeted (SCoT)-polymerase chain reaction (PCR) tools. Genomic DNA was extracted from young leaves and the PCR reactions were performed using ten primers for each SCoT, ISSR and RAPD marker. Only four out of ten primers showed amplicon profiles in all PCR markers. A total of 70 bands were generated by SCoT, ISSR and RAPD-PCR with 35 polymorphic bands and 35 monomorphic bands. The total number of bands of RAPD, ISSR and SCoT was 15, 17 and 38, respectively. The polymorphism percentages achieved by RAPD, ISSR and SCoT were 60.25%, 15% and 57%, respectively. The cluster analysis based on morphological data revealed two clusters. Cluster I consisted of violet and orange kalanchoe, and cluster II comprised red, yellow and purple kalanchoe. Whereas the cluster analysis based on molecular data revealed three clusters. Cluster I included only yellow kalanchoe, cluster II comprised orange and violet kalanchoe while cluster III comprised red, and purple kalanchoe. The study concluded that orange, violet and yellow kalanchoe are distinguished parents for breeding economically valued traits in kalanchoe. Also, the study concluded that SCoT and RAPD markers reproduced reliable banding patterns to assess the genetic polymorphism among kalanchoe genotypes that consider the basis stone for genetic improvements in ornamental plants.

Environmental pollution is one of the most pressing challenges in today's world. The main cause of this pollution is fuel emissions from automobiles and other sources. As industrialization progresses, we will be unable to compromise on the use of energy to power heavy machines and will be forced to seek out the best options. As a consequence, utilizing green fuel, such as biodiesel derived from natural sources, is a realistic option. Jatropha curcas L. (Euphorbiaceae) is recognized as the greatest feedstock for biodiesel production throughout the world, and it has gained a huge market value in the recent years. Conventional cultivation alone will not be sufficient to meet the global need for the plant's biomass for the production of biodiesel. Adoption of plant tissue culture techniques that improve the biomass availability is an immediate need. The present review provides detailed information regarding in-vitro plant propagation (direct and indirect organogenesis), somatic embryogenesis, and acclimatization protocols of plantlets for stabilized production of biomass. The review also focuses on biotechnological approaches such as gene transformation studies, production of haploids, and double haploids for developing elite germplasm for high biomass and improved traits for the production of biodiesel.

Artemisinin is an anti-malarial sesquiterpene lactone derived from Artemisia annua L. (Asteraceae family). One of the most widely used modes of treatment for malaria is an artemisinin-based combination therapy. Artemisinin and its associated compounds have a variety of pharmacological qualities that have helped achieve economic prominence in recent years. So far, research on the biosynthesis of this bioactive metabolite has revealed that it is produced in glandular trichomes and that the genes responsible for its production must be overexpressed in order to meet demand. Using biotechnological applications such as tissue culture, genetic engineering, and bioreactor-based approaches would aid in the upregulation of artemisinin yield, which is needed for the future. The current review focuses on the tissue culture aspects of propagation of A. annua and production of artemisinin from A. annua L. cell and organ cultures. The review also focuses on elicitation strategies in cell and organ cultures, as well as artemisinin biosynthesis and metabolic engineering of biosynthetic genes in Artemisia and plant model systems.

Cucumber production is considered a crucial problem under biotic and abiotic stress, particularly in arid and semi-arid zones. The current study investigated the impact of grafted cucumber plants on five cucurbit rootstocks under infection with Fusarium oxysporum f. sp. cucumerinum alone and in combination with heat stress in two different locations (i.e., Kafr El-Sheikh and Sidi Salem) during the year of 2021. The rootstock of VSS-61 F 1 displayed the highest level of resistance with values 20.8 and 16.6% for wilt incidence and 79.2 and 83.4% for the wilt reduction, respectively for both locations. This rootstock showed the lowest disease severity of fusarium wilt (15.3 and 12%), and high grafting efficiency (85 and 88%), respectively in both locations. Grafting also improved plant vigor and cucumber production under heat stress (40-43 °C). The rootstocks VSS-61 F 1 , Ferro and Super Shintoza significantly increased the total yield of cucumber plants compared to non-grafted cucumber and the rootstock Bottle gourd in both locations. Further studies are needed on grafted plants under multiple stresses in terms of plant biological levels, including physiological, biochemical and genetic attributes.

The production and quality of tomato seedlings needs many growth factors and production requirements besides controlling the phytopathogens. Paclobutrazol (PBZ) has benefit applications in improving crop productivity under biotic stress ( Alternaria solani , the causal agent of early blight disease in tomatoes). In the current study, the foliar application of PBZ, at rates of 25, 50, and 100 mg L -1 , was evaluated against early blight disease in tomatoes under greenhouse conditions. The roles of PBZ to extend tomato seedling lives and handling in nurseries were also investigated by measuring different the biochemical (leaf enzymes, including catalase and peroxidase) and histological attributes of tomato seedlings. Disease assessment confirmed that PBZ enhanced the quality of tomato seedlings and induced resistance to early blight disease post inoculation, at 7, 14, and 21 days. Higher values in chlorophyll content, enzyme activities, and anatomical features of stem (cuticle thickness) and stomata (numbers and thickness) were recorded, due to applied PBZ. This may support the delay of the transplanting of tomato seedlings without damage. The reason for this extending tomato seedling life may be due to the role of PBZ treatment in producing seedlings to be greener, more compact, and have a better root system. The most obvious finding to emerge from this study is that PBZ has a distinguished impact in ameliorating biotic stress, especially of the early blight disease under greenhouse conditions. Further studies, which consider molecular variables, will be conducted to explore the role of PBZ in more detail.

Plant response to salt stress and the mechanism of salt tolerance have received major focus by plant biology researchers. Biotic stresses cause extensive losses in agricultural production globally, but abiotic stress causes significant increase in the methylglyoxal (MG) level of Glyoxalase I ( Gly I ). Identification of salt-tolerant genes when characterizing their phenotypes will help to identify novel genes using polymerase chain reaction (PCR) to amplify the DNA coding region for glyoxalase I . This method is specific, requiring only genomic DNA and two pairs of PCR primers, and involving two successive PCR reactions. This method was used rapidly and easily identified glyoxalase I sequences as salt-tolerant genes from Jojoba ( Simmondsia chinensis (Link) Schneider). In the present study, the glyoxalase I gene was isolated, amplified by PCR using gene-specific primers and sequenced from the jojoba plant, then compared with other glyoxalase I sequences in other plants and glyoxalase I genes like in Brassica napus , ID: KT720495.1; Brassica juncea ID: Y13239.1, Arachis hypogaea ; ID: DQ989209.2; and Arabidopsis thaliana L, ID: AAL84986. The structural gene of glyoxalase I , when sequenced and analyzed, revealed that the uninterrupted open reading frame (ORF) of jojoba Gly I ( Jojo-Gly I ) spans 775 bp, corresponding to 185 amino acid residues, and shares 45.2% amino acid sequence identity to jojoba (Jojo-Gly I). The cloned ORF, in a multicopy constitutive expression plasmid, complemented the Jojo-Gly I , confirming that the encoded Jojo-Gly I in jojoba showed some homology with other known glyoxalase I sequences of plants.

Background and Objectives: Presently, determination of optimum protocol for callus induction of any plant is an important issue in tissue culture technology. Therefore, the main objective of this study was to find out an optimum protocol for callus induction from in vitro cultured jojoba by determining the optimum explant and the best growth regulators mixture for callus induction., Materials and Methods: The study used three variant explants namely the leaf disks, seeds and nodal segments for callus formation. Different culture media containing basic Murashige and Skoog (MS) medium components supplemented with various concentrations of 2,4-dichlorophenoxy acetic acid as an auxin (2,4-D) and Kinetin (Kin) as a cytokinin with various concentrations ranging from 0.0, 0.5, 1.0 and 2.0 mg L-1 were used. The total number of treatments were 16. The callus was induced from all explants on MS medium containing the lowest concentration of 2,4-D 0.5 mg L-1 with any concentration of Kin., Results: The results showed that nodal segments were the best for callus formation followed by the leaf disks (leaves) and seeds, respectively. While, the best concentration of proliferation and development of the used explant was 2.00 followed in descending order by 1.00, 0.5 and 0.0 mg L-1, respectively., Conclusion: The study find out that the best concentration of 2,4-dichlorophenoxy acetic acid as an auxin (2,4-D) and Kinetin (Kin) as a cytokinin was 2.00 followed in descending order by 1.00, 0.5 and 0.0 mg L-1, respectively for callus induction.

Antioxidants present in dates are necessary for all physiological processes of humans and animals. In Saudi Arabia date palm is a national fruit tree, produces millions of tons of dates for consumption and is considered a major source of antioxidants. The main aim of this study was to determine the role of potassium nitrate (KNO3) in the formation of antioxidants from explants collected from date palm cultivars in Al-Ahsa Oasis, Kingdom of Saudi Arabia (KSA) and to monitor the extent of its effect on growth and development of cells during callus formation stage via somatic embryogenesis. The results showed that full concentration of KNO3 was the best for callus formation in general. While, the half concentration of KNO3 played an important role for stimulating the explants to form phenolic compounds and the browning emergence for all the cultivars under investigation. On the other hand, the chemical analysis for measuring the phenolic compounds in the explants showed that all the explants formed antioxidants but with varying degrees. The highest mean of phenolic contents was found in those explants cultured with the half concentration of KNO3 for Shishi cv 2.053 ± 0.010a mg g(-1) and antioxidant activity by ABTS Inhibition and UM Trolox was 80.694 ± 0.439 and 801.575 ± 2.391, respectively.
