Abstract

Research has been conducted on Pseudomonas putida mt-2 archetypal tolyene-xylene catabolic plasmid pWW0. The identification of the insertion sequence ISPpu12 on this plasmid has been carried out, and the details are reported.

Abstract

The role of mucK, a gene found in Acinetobacter calcoaceticus ADP1(BD413), in the growth process of exogenous cis,cis-muconate was analyzed. The study had adopted spontaneous mutation processes to investigate the structural and regulatory genes involved in certain conversion. Results of the study indicated that the mucK gene has the ability to grow on exogenous cic,cis-muconate without significantly affecting its ability to grow on substrates.

Abstract

The study of the Pseudomonas sp. strain IC benzoate catabolism operon and bphE gene reveals that bph, xyl, dmp and nah pathway genes of RNA group I are homologous, colinear and belong to a single evolutionary group. The bphE and bphG genes possess homology with other Pseudomonas strain genes encoding isofunctional proteins. The bph genes are possibly present in the plasmid pWW110, which is more than 200 kb.

Abstract

Plasmids pWW53 and pDK1 contain multiple regions homologous to the xylS regulatory gene of pWW0. Sequence analysis showed that pWW53 contains three copies of xylS with varying levels of homology to the pWW0 xylS gene. On the other hand, pDK1 contains two copies of xylS. However, comparisons of the sequences showed that the pDK1 sequences arose from an internal reciprocal recombination event.

Abstract

Deletion and transposition analysis of two identical IS1246 copies that encode a 1275 bp sequence in Pseudomonas putida reveals that the copies encode xyl genes that mediate P- and M-xylenes and toluene catabolism to form central metabolities. Xylene expression is seen on the TOL catabolic plasmid pWWO. IS1246 encodes a 12 bp terminal inverted repeat that expresses a 280 codon open reading frame, which resembles the ORFs in the bacterial insertion sequences of Escherichia coli, Meisseria and Bacteroides. The IS1246 strain does not have any effect on TOL transposon activity.
