Abstract A simple, novel, and less cost yellow (Erythrosine) modified pencil graphite electrode (Po-ERY/MGPE) was successfully fabricated via electropolymerization method using cyclic voltammetric techniques. The fabricated Po-ERY/MGPE opted as a sensor for the detection of Adrenaline (ADR) in 0.2 M PBS (7.4 pH). This reported senor displayed excellent electrocatalytic activity, increased sensitivity, high stability, superior electron transfer kinetics in the oxidation of ADR once relative to BGPE. The significance of pH, scan rate, and impact of concentration was assessed at the sensor. As per the pH and scan rate study, redox routes carry the same number of electrons and protons, and electro-oxidation of ADR was adsorption controlled respectively. The LOD of ADR was found to be 0.499 µM. The DPV data indicate that there is a significant peak divergence among ADR and uric acid (UA) which could make it easier to determine them alone and simultaneously on the sensor. The described method has been employed for the determination of ADR in injection sample. Good recovery values indicate the efficacy and applicability of the sensor in detecting ADR.

Marker-assisted backcross breeding enables selective insertion of targeted traits into the genome to improve yield, quality, and stress resistance in wheat. In the current investigation, we transferred four drought tolerance quantitative trait loci (QTLs) controlling traits, viz canopy temperature, normalized difference vegetative index, chlorophyll content, and grain yield from the drought-tolerant donor line, C306, into a popular high-yielding, drought-sensitive variety, HD2733. Marker-assisted selection coupled with stringent phenotypic screening was used to advance each generation. This study resulted in 23 improved lines carrying combinations of four drought tolerance QTLs with a range of 85.35%–95.79% background recovery. The backcross-derived lines gave a higher yield under moisture-deficit stress conditions compared with the recipient parent. They also showed higher phenotypic mean values for physiological traits and stability characteristics of HD2733. A promising genotype, HD3411, derived from this cross was identified for release after national multi-location coordinating trials under the All India Coordinated Wheat Improvement Project. Our study is a prime example of the advantages of precision breeding using integrating markers and phenotypic selection to develop new cultivars with desirable traits like drought tolerance.

Abstract In the present work, Orange CD was chosen as an intriguing modifier for the electropolymerization on the surface of CPE by the CV technique. A novel, sensitive, and cost-effective poly (Orange CD) MCPE (PoOCD/MCPE) sensor was utilized for the selective detection of paracetamol (PA) in 0.2 M phosphate buffer solution (PBS) of pH 7.4. The oxidation peak current of PA was vastly enhanced at the sensor. The scan rate study is suggested that electro-oxidation of PA was adsorption-controlled. The pH study testifies the redox pathways transport with the same quantity of electrons and protons. The detection limit of PA is found to be 2.64 µM. DPV results show that substantial peak separation between PA, folic acid (FA), and dopamine (DA) could be facilitating their individual and simultaneous determination on the sensor. The decorated sensor demonstrates high sensitivity, stability, reproducibility, repeatability and has been successfully exploited for the detection of PA in a tablet with promising results.

Manipulation of flowering time for adaptation through natural or genetic approaches may combat heat-stress damage that occurs at the reproductive stages in production conditions. HD2733, a popular wheat variety of the eastern plains of India, is largely sensitive to heat stress. Therefore, the current study aims to improve heat tolerance of HD2733 by introgression of QTLs associated with early anthesis and high kernel weight linked to markers Xbarc186 and Xgwm190, respectively, through marker-assisted backcross breeding (MABB) from a tolerant donor, WH730. A total of 124 simple sequence repeat (SSR) markers distributed evenly across the genome were used for the background selection. The alleles of Xbarc186 and Xgwm190 were fixed in BC2F1 and BC1F2 generations by selecting individual plants heterozygous for both marker loci and backcrossed with HD2733 and simultaneously selfed to generate BC2F1 and BC1F2 populations, respectively. Furthermore, the selected BC1F2 were selfed to generate the BC1F4 population. By background screening, a total of 39 BC2F3 and 21 BC1F4 families homozygous for the targeted QTLs with 90.9–97.9% and 86.8–88.3% RPG recoveries were selected. The best performing 17 BC2F3 and 10 BC1F4 lines were evaluated for various morpho-physiological traits. Phenotypic evaluation and multi-location trials of the introgressed lines under late sown conditions led to the selection of three promising lines with early anthesis and higher grain yield. The improved lines will serve as an excellent genetic material for functional genomics and expression studies to understand the molecular mechanisms and pathways underlying the stress tolerance.

Abstract In the present work, the ZnO and Al-ZnO nanoflakes (NFs) were synthesized by the co-precipitation process. The synthesized NFs were characterized by X-ray diffraction and field emission scanning electron microscopy. Energy dispersive X-ray spectrometer was explored for the elemental chemical compositions. The prepared NFs were taken for the modification of the electrode and developed the modified electrode for the electrochemical analysis of the dopamine (DOA) at pH 7.4. The Al-ZnO modified carbon paste electrode (MCPE) was electropolymerised by using textile dye red DSBR. The Po-RD/Al-ZnO MCPE exhibited good electrochemical sensor properties towards the electrochemical detection of DOA. Several factors such as the impact of speed rate (υ), pH and concentration of the DOA were analyzed at the modified electrode. The great sensitivity was established to the fast electron-transfer kinetics and surface coverage of the DOA on the electrode. The prepared electrode exhibits satisfactory stability at the ambient conditions. The detection limit of 0.58 μM was achieved for the DOA. The decorated sensor was stable, sensitive, selective, and reproducible and used in the analytical applications.

Abstract The RGO-Y2O3 and RGO-Y2O3: Cr3+ (5 mol %) nanocomposite (NC) synthesized by hydrothermal technique. The structure and morphology of the synthesized NCs were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Y2O3:Cr3+ displays spherical-shaped particles. Conversely, the surface of the RGO displays a wrinkly texture connecting with the existence of flexible and ultrathin graphene sheets. The photoluminescence (PL) emission spectra showed series of sharp peaks at 490, 591, and 687 nm which corresponding to 4F9/2 → 6H15/2, 4F9/2 → 6H13/2, and 4F9/2 → 6H11/2 transitions and lies in the blue, orange, and red region. The prepared NCs were used for the preparation of modified carbon paste electrodes (MCPE) in the electrochemical detection of dopamine (DA) at pH 7.4. Both modified electrodes provide a good current response towards voltammetric detection of DA. Doping is an effective method to improve the conductivity of Y2O3:Cr3+ and developed a method for the sensor used in analytical applications.

Abstract Identification of markers associated with major physiological and yield component traits under moisture deficit stress conditions in preferred donor lines paves the way for marker-assisted selection (MAS). In the present study, a set of 183 backcross inbred lines (BILs) derived from the cross HD2733/2*C306 were genotyped using 35K Axiom genotyping array and SSR markers. The multi-trait, multi-location field phenotyping of BILs was done at three locations covering two major wheat growing zones of India, north-western plains zone (NWPZ) and central zone (CZ) under varying moisture regimes. A linkage map was constructed using 705 SNPs and 86 SSR polymorphic markers. A total of 43 genomic regions and QTL × QTL epistatic interactions were identified for 14 physiological and yield component traits, including NDVI, chlorophyll content, CT, CL, PH, GWPS, TGW and GY. Chromosomes 2A, 5D, 5A and 4B harbors greater number of QTLs for these traits. Seven Stable QTLs were identified across environment for DH (QDh.iari_6D), GWPS (QGWPS.iari_5B), PH (QPh.iari_4B-2, QPh.iari_4B-3) and NDVI (QNdvi1.iari_5D, QNdvi3.iari_5A). Nine genomic regions identified carrying major QTLs for CL, NDVI, RWC, FLA, PH, TGW and biomass explaining 10.32–28.35% of the phenotypic variance. The co-segregation of QTLs of physiological traits with yield component traits indicate the pleiotropic effects and their usefulness in the breeding programme. Our findings will be useful in dissecting genetic nature and marker-assisted selection for moisture deficit stress tolerance in wheat.

The electrochemical sensor for Dopamine (DA) was investigated using poly(amido black) modified carbon paste electrode (MCPE) by the cyclic voltammetric (CV) technique. The film-coated electrode exposed supreme electrocatalytic properties towards electrochemical detection DA and uric acid (UA). The limit of detection (LOD) of DA and UA found to be 2.03 µM and 3.6 µM respectively. Furthermore effectively selective separation of DA and UA in a binary mixture was accomplished. The application of the developed electrode was demonstrated by detecting DA in the injection sample with adequate recoveries. The sensor was stable, sensitive, selective and reproducible.

The carbon paste electrode was modified with poly (benzydamine) by cyclic voltammetric technique (CV) and its electrocatalytic activity towards the oxidation of catechol (CC) and hydroquinone (HQ) was studied. The detailed investigation on the various parameters influencing the activity was also carried out. From the obtained result, it was concluded that the process was adsorption - controlled and the involvement of same number of electron and proton. The simulations resolution of CC and HQ along with excellent selectivity and sensitivity were also exhibited by the studied MCPE. The detection limits 5.79 and 3.78 μM were found for CC and HQ respectively.

Drought stress is well known phenomenon that affects the productivity of wheat (Triticum aestivum L). Knowledge on genetic variation, genotype × environment interaction and association between physiological and yield component traits is crucial for the development of improved varieties having high yield and water use efficiency. The present study consists of 280 backcross inbred lines (BILs) population evaluated for grain yield and morpho-physiological traits for two years at three locations. Combined ANOVA unfolded significant variability among traits in BILs population for yield and morpho-physiological traits.Grain yield showed significant association with normalized difference vegetation index (NDVI), soil plant analysis development (SPAD), thousand grain weight (TGW), and canopy temperature (CT). The genotype, environment and genotype ×environment interaction for yield was highly significant (p< 0.01). ASV (AMMI stability value) was calculated and top 29 genotypes were selected and further analyzed with AMMI and GGE biplot analysis for dissecting out genotype × environment interaction. The results classified genotypes G82, G202, G234, G263, G6, G192 and G77 are most stable and high yielding genotypes.

The Poly (Reactive Blue) modified carbon paste electrode (MCPE) exhibited good stable and sensitive current responses towards the determination of dopamine (DA) in phosphate buffer solution (PBS pH 7.4). The various parameters like the effect of scan rate, pH and concentration of DA studied. The electrode process was found to be adsorption-controlled and the limit of detection (LOD) of DA obtained to be 4.64 μM. The simultaneous determination of DA in the presence of Ascorbic acid (AA) and Uric acid (UA) was investigated by using cyclic voltammetry (CV). The application of the developed electrode was demonstrated by detecting DA in injection sample with satisfactory recoveries. The sensor was stable, sensitive, selective and reproducible.

Electropolymerization of Benzydamine was achieved on carbon paste electrode surface by cyclic voltammetric technique. The poly (benzydamine) MCPE exhibited an excellent electrocatalytic behavior of uric acid (UA). The critical examination was conducted on the electrochemical measurements at poly (benzydamine) MCPE for analysis of UA and dopamine (DA) by CV and DPV techniques. From the scan rate study, it was found that the oxidation of UA was diffusion controlled. Uric acid exhibits good detection limit (11.33 nM) at modified electrode in DPV technique. Hence, the investigated sensor poly (benzydamine) MCPE shows excellent selectivity and sensitivity towards resolution of UA in existence of DA.

Climate change and global warming are the foremost anthropogenically accelerated catastrophes that are already causing world-wide challenges, but threaten to thwart global food, environmental and nutritional security in the future. Climate change affects ecosystem services and interactions between biotic and abiotic factors. The most drastic consequences have been observed in the agricultural and livestock sector, with diminished production and productivity potential. Agriculture and allied sectors contribute markedly to the production of greenhouse gases; however, integrated management practices can be used to curtail greenhouse gas emissions and its adverse impacts. Forage crops and their wild relatives maintain biodiversity and ecosystem services and minimise the drastic effects of climate change. Forage crops adapted to harsh environments have certain unique features such as perenniality, deep root system, high resource-use efficiency (light, nutrients and water), and low production of methane and N2O, making them suitable for future use under climate change. This review highlights the prominent features of various cultivated and rangeland forage crops that may be crucial to understanding impacts of climate change. We discuss the wild relatives of forage crops, which are often adapted for multiple stresses, and highlight their mechanisms for adaptation under climate change. We consider the advanced breeding and biotechnological tools useful for developing climate-smart forage crops. This review provides novel insight into forage crops and their wild relatives in terms of their exploitation in future stress breeding programmes and paths for developing climate-resilient crops. Climate change substantially affects the global food, environmental and nutritional security, as well as biodiversity and ecosystem services. Wild relatives of forage crops and their functional trait diversity can be exploited for future stress breeding programs and development of climate-resilient crops.
