Introduction This research aimed to determine whether Class II malocclusion can be treated with clear aligners after completing treatment with the initial set of aligners. Methods A sample of 80 adult patients were divided into Group 1 with Class I molar malocclusions (n = 40 [11 men and 29 women]; 38.70 ± 15.90 years) and Group 2 with Class II molar malocclusions (n = 40 [11 men and 29 women]; 35.25 ± 15.21 years). All patients had finished treatment with the initial set of Invisalign aligners (Align Technology, Santa Jose, Calif) without known centric occlusion–centric relation discrepancies, issues of compliance, or overcorrection. The 7 measurements using the American Board of Orthodontics (ABO) Model Grading System and millimetric measurements for anteroposterior (AP) and vertical dimensions were assessed and compared between the 2 groups at pretreatment, posttreatment ClinCheck (Align Technology) prediction, and posttreatment. Results No improvements were observed in the AP correction. The amount of AP correction in patients with Class II malocclusion was 6.8% of the predicted amount. The amount of overbite correction achieved was 28.8% and 38.9% of the predicted amounts in patients with Class I and Class II malocclusion, respectively. Significant improvements in alignment and interproximal contact scores were observed, with only slight improvements in total ABO scores. An increase in mean occlusal contacts score was observed after treatment. No patient with Class II malocclusions would meet the ABO standards after Invisalign treatment. Conclusions The Invisalign system successfully achieves certain tooth movements but fails to achieve other movements predictably. No significant Class II correction or overjet reduction was observed with elastics for an average of 7-month duration in the adult population. Additional refinements may be necessary to address problems created during treatment, as evidenced by a posterior open bite incidence.

Classroom learning must be addressed in order to provide an inclusive course experience for all students. This study examined the effectiveness of support structures for Deaf and Hard-of-Hearing (DHH) students in the form of a web conferencing system that allowed for a composite screen solution. Quantitative and qualitative data analysis indicated that DHH students rely on access services and accommodations such as sign language interpreting and priority seating in order to have access to content in a mainstream classroom. However, the usage by hearing students was higher than anticipated.

According to the 2016 National Association of Colleges and Employers survey, employers rated ability to work on a team as one of the most sought out attributes in hiring employees. As such, it is incumbent on educators to consider how computing students are being prepared to engage in the workforce as effective team members. Within computing curriculum, teamwork is commonly aligned with collaborative learning. While many studies have examined how students learn in a collaborative environment, little research has been conducted on the faculty practices around collaborative learning in computing education. This paper reports on the findings from an exploratory qualitative case study that examined the faculty perspectives and instructional approaches to collaborative learning in first-year and second-year post-secondary computing education. Findings indicated that collaborative learning was limited to informal instructor-student classroom interactions and unstructured student group work. Furthermore, faculty emphasized building individual student skillsets over collaborative learning or teamwork skills in the computing domain. This suggests that there is space for faculty development around pedagogical knowledge, training and curriculum development specifically related to collaborative learning in computing education. Future research should extend to examine instructional approaches that foster and inform the development of collaborative learning, particularly in the scope of fostering diverse and inclusive computing environments.

Organic dairy products are the second largest sector of the organic food market. Organic dairy products come from United States Department of Agriculture (USDA) certified organic dairy cattle that meet USDA organic standards. Organic dairy cattle in the US cannot be treated with antibiotics for mastitis, one of the costliest diseases of dairy cattle, and thus effective alternatives are needed. When any compound (medication or other non-food product) is used in a food producing animal, a withhold time for that compound that meets US Food and Drug Administration (FDA) standards for food safety must be applied to the animal and its products (like milk). However, there are no US FDA products approved for mastitis that maintain USDA certified organic dairy cattle’s organic status. Thus, we studied the pharmacokinetics of 3 compounds (garlic, thymol and carvacrol) used on organic both healthy and mastitic organic dairy cattle. We also used this information to estimate a milk withhold time using methods consistent with US FDA requirements. For thymol intra-mammary and carvacrol intra-mammary or topical administration, all compounds were partially absorbed into the body from the milk or skin. Thymol and carvacrol are measurable in plasma (at 0.0183 and 0.0202 µg/mL, respectively) after intramammary administration with similar elimination half lives of 1.7 h. Milk concentrations of thymol and carvacrol are much higher at 2.958 and 4.487 µg/mL in healthy cattle, respectively. Concentrations are not significantly different in cows with mastitis as compared to those in healthy cows. Despite these compounds being natural products, they should have a withhold time for milk of at least 24 h after administration. For garlic, levels remained below the limit of detection in milk and plasma and thus no withdrawal time appears to be needed for milk.

A program was written in R to facilitate the implementation of the tolerance limit method (TLM) for establishing regulatory withdrawal times for limiting drug residues in meat, milk, and eggs. The developed computer source code can use pharmacokinetic and regulatory data to calculate the drug withdrawal period according to United States Food and Drug Administration (U.S. FDA) guidelines. The code called the “Withdrawal Time Calculator (WTC)” applied this TLM method to meat samples. The program was tested with the data provided by the U.S. FDA guidance and other published data collected from in vivo studies. Additional algorithm validation data were flunixin and sulfamethazine liver concentration data from peer-reviewed publications generated by our laboratory. This manuscript reports the withdrawal period results from testing the developed WTC code. Moreover, the source code for the WTC contains a data removal algorithm, constructed according to U.S. FDA data elimination recommendations if the user chooses. The power of the WTC is that it bypasses the use of multiple platforms typically required to perform the TLM, including standard commercial spreadsheet software (i.e., Microsoft Excel) and Statistical Analysis System (SAS) while providing speed and usability. This novel program provides a platform to calculate a withdrawal period recommendation for any drug in any class of animal for various regulatory body standards and could be very helpful in cases of extra-label drug use in food animals.

This exploratory case study examined problem solving approaches across three computing education programs: computer science, information sciences and technologies and software engineering. Using an Experiential Learning Theory framework, faculty perceptions and approaches to problem solving and reflection and the nature of social learning in both classroom enactment and laboratory sessions for second-year undergraduate students were examined. Classroom and laboratory observations, in-depth interviews and course artifacts served as the main sources of data. Qualitative data analysis revealed three key findings: a) faculty perceptions and approaches to problem solving centered on modeling problem decomposition to break down larger problems into smaller steps or sub-problems, b) reflection was informally modeled by faculty during instruction, but primarily occurred internally for students and c) faculty noted the importance of collaboration in the workforce, yet perceived collaborative student work to be at odds with developing individual skillsets, leading to the predominance of informal rather than structured social learning. These findings have implications for faculty understanding of computing instruction related to problem solving, intentional strategies that promote student reflections of their learning processes and importantly, how social learning is conceptualized and enacted in computing. In addition, the study also had implications for how computing faculty are currently preparing students for transition to workforce demands.

Thymol and carvacrol may be present in several phytoceutical products but there are no well-defined methods to measure these compounds in meat and milk from treated animals. U.S. regulatory authorities deem their presence as an adulteration of food. A rapid and sensitive HS-SPME-GC-MS/MS method was developed for the detection of thymol and carvacrol in bovine milk, plasma, liver, kidney, and fat. Inter- and intraday precision values were all less than 15.7 and 20.2% for thymol and carvacrol, respectively. The accuracy was in ranges of 69.9-111.8% for thymol and 74.0-119.2% for carvacrol. With the exception of fat tissue, stability studies showed that both compounds are stable over a 2 month period. A pilot pharmacokinetic study was conducted to evaluate the developed analytical method and to provide initial estimates of thymol and carvacrol depletion in plasma, milk, and several tissues. Treatment of lactating dairy cattle with phytoceutical products containing these substances resulted in low but measurable residue levels at 96 h for liver and 36 h for milk with very short apparent plasma and milk half-lives (3.0 h).

This pharmacokinetic study was designed to determine the pharmacokinetics of enrofloxacin at 5 mg/kg when given to sea hares in their hemolymph. Enrofloxacin is a commonly used antimicrobial in veterinary medicine and potentially could be used to treat sea hares exposed to susceptible bacterial species. We individually identified 8 juvenile Aplysia californica and group housed them in an open seawater flow system at 14 to 18 °C; 2 served as untreated controls. The remaining 6 animals were injected into the hemocoel with 0.030 mL of 22.7 mg/mL enrofloxacin (average dose, 5 to 6 mg/kg). At each time point, 300 μL hemolymph was collected from the pedal hemolymph sinus and HPLC-analyzed for enrofloxacin and ciprofloxacin levels. Enrofloxacin was detected in all dosed animals, at an average peak concentration of 3 μg/mL in hemolymph, and remained in the body for 20.3 h with an average clearance of 0.19 μg × h/mL. No ciprofloxacin was detected in any Aplysia in this study. Hemocoel injection appears to be an effective way to administer enrofloxacin to Aplysia and reach clinically relevant concentrations. Enrofloxacin reached therapeutic target concentrations in A. californica when dosed according to the regimen described in the current report.

Mastitis is a costly disease for dairy farmers. Some dairy farmers use herbal products, or phytoceuticals, to treat mastitis. Phytoceuticals have not been approved for this use by the United States Food and Drug Administration, and have not been tested to determine how they impact antibiotic residue detection testing. The current study tested the potential for phytoceuticals to cause positive results on two milk antibiotic residue screening tests, the Delvotest P and Charm SL Beta-lactam test, or to interfere with the detection of antibiotics by these tests. The three phytoceuticals tested were labelled for intramammary, topical or intravulvar administration. Testing was performed in vitro using the products diluted in milk obtained from healthy organic dairy cows. Phytoceuticals were tested at concentrations ranging from 1.5 per cent to 100 per cent. Concentration levels were replicated at least twice on each milk antibiotic residue screening test. The Delvotest P is based on detection of bacterial inhibitors and no positive results were obtained for any product at concentrations less than 50 per cent. The Charm SL Beta-lactam test uses a receptor for the detection of beta-lactam antibiotics and no concentration of phytoceuticals caused an interference with these tests. Based on dilution of the products in bovine milk at physiologically achievable levels, phytoceutical products tested at levels expected after treatment do not cause positive test results for the Delvotest P nor do they interfere with the Charm SL Beta-lactam test in detection of various antibiotics.

Nonantibiotic treatments for mastitis are needed in organic dairy herds. Plant-derived oils may be useful but efficacy and potential mechanisms of action of such oils in mastitis therapy have not been well documented. The objective of the current study was to evaluate the antibacterial activity of the plant-derived oil components of Phyto-Mast (Bovinity Health LLC, Narvon, PA), an herbal intramammary product, against 3 mastitis-causing pathogens: Staphylococcus aureus, Staphylococcus chromogenes, and Streptococcus uberis. Plant-derived oils evaluated were Thymus vulgaris (thyme), Gaultheria procumbens (wintergreen), Glycyrrhiza uralensis (Chinese licorice), Angelica sinensis, and Angelica dahurica. Broth dilution testing according to standard protocol was performed using ultrapasteurized whole milk instead of broth. Controls included milk only (negative control), milk + bacteria (positive control), and milk + bacteria + penicillin-streptomycin (antibiotic control, at 1 and 5% concentrations). Essential oil of thyme was tested by itself and not in combination with other oils because of its known antibacterial activity. The other plant-derived oils were tested alone and in combination for a total of 15 treatments, each replicated 3 times and tested at 0.5, 1, 2, and 4% to simulate concentrations potentially achievable in the milk within the pre-dry-off udder quarter. Thyme oil at concentrations ≥2% completely inhibited bacterial growth in all replications. Other plant-derived oils tested alone or in various combinations were not consistently antibacterial and did not show typical dose-response effects. Only thyme essential oil had consistent antibacterial activity against the 3 mastitis-causing organisms tested in vitro. Further evaluation of physiological effects of thyme oil in various preparations on mammary tissue is recommended to determine potential suitability for mastitis therapy.

Tetracycline is a broad-spectrum antibiotic used to treat infections in swine. The maximum residue levels of tetracycline in pork stomach tissue in Russia, Europe, and the United States are 10, 200, and 2,000 ppb, respectively. This difference in accepted safety levels may be the reason why stomach tissues that the United States exports continue to be residue violators in overseas markets. In this study, 30 pigs at two different stages of production (weanling and finisher) were treated with tetracycline at 22 mg/kg of body weight per day for a total of 5 days via a water medicator. Blood samples were collected at 0, 72, 78, 96, and 102 h after the start of medication. The medication was stopped at 120 h, and blood samples were again collected at 126, 144, 168, 192, and 216 h after exposure. Five animals were slaughtered for stomach tissue 0, 24, 48, 96, and 192 h after the drug was flushed from the water line. All blood and tissue samples were analyzed by high-performance liquid chromatography-UV methods. The tetracycline levels in plasma were below the level of detection after the U.S.-labeled withdrawal time of 4 days. The stomach tissue residues averaged 671.72, 330.31, 297.77, 136.36, and 268.08 ppb on withdrawal days 0, 1, 2, 4, and 8, respectively. Using the U.S. Food and Drug Administration tolerance limit method and a population-based pharmacokinetic model with Monte Carlo simulation, a withdrawal interval was estimated. This study demonstrated that tetracycline residues are still detectable in the stomach tissues after the established United States withdrawal time of 4 days. These residue levels may explain why stomach tissues tested in Russia and Europe show positive residues for tetracycline, even though the meat may pass inspection here in the United States prior to export.

Mastitis is among the most costly concerns for dairy producers whether cattle are managed conventionally or organically. Unfortunately, there are no USFDA-approved mastitis treatments that allow dairy cows in the United States to maintain organic dairy status. We investigated the plasma pharmacokinetics of three organic mastitis products currently used by organic producers and organic dairy veterinarians. Those products include intramammary, topical and intravaginal preparations, each dosed at two levels. Additionally, tissue data were collected for kidney, liver and fat in order to estimate a withholding time for each of the products. The lower limit of quantification (LOQ) and lower limit of detection (LOD) were 0.001 and 0.0005 µg ml–1, respectively, in plasma and all tissues except fat for both thymol and carvacrol. Fat had an LOQ of 0.01 µg ml–1 and an LOD of 0.005 µg ml–1 for thymol and carvacrol. Diallyl disulfide had an LOQ of 0.005 µg ml–1 and LOD of 0.001 µg ml–1 in all tissues. For dial...

This panel will present the efforts of the NSF ADVANCE Program along with specific efforts by university awardees to activate change for STEM faculty. Program elements, milestones, progress and challenges will be described along with strategies on getting started at your own university.

We compared cell number, putative stem cell markers, and clonogenic ability in fresh uncultured human limbal epithelial cells to that obtained from stored organ-cultured tissue.Cell suspensions were formed from fresh and organ culture-stored human limbal epithelium. Expression of putative stem cell markers ΔNp63 and TrkA was performed using immunofluorescent staining before culture. Colony-forming efficiency (CFE) assays were performed at first passage. The effects of tissue storage, age, and postmortem/culture times were analyzed in a general linear model.Limbal tissue from 94 donors (34 fresh and 60 stored) was compared. Three times more cells were obtained per eye from fresh (35.34 × 104; SD, 17.39) than stored (11.24 × 104; SD, 11.57; P0.01) tissue. A higher proportion of cells from fresh tissue were viable (91.9%; SD, 5.7 vs. 85%; SD, 10.8) P0.01. Higher total cell expression of ΔNp63 (20.19 × 104; SD, 15.5 vs. 3.28 104; SD, 4.33) and TrkA (59.24 × 104; SD, 13.21 vs. 7.65 × 104; SD, 1.05) was observed in fresh than stored tissue (P0.01). Colony-forming efficiency was higher for fresh (1.42; SD, 0.12) than stored (0.43; SD, 0.15; P0.01) cells. For stored tissue only, there was a significant inverse relationship between donor age and total number of cells isolated (R2 = 0.27, P0.001).Storage of corneoscleral discs in organ culture medium leads to significant reduction in limbal epithelial cell number, expression of ΔNp63 and TrkA, and viability compared to fresh tissue. There is a smaller basal stem cell population in stored compared to fresh tissue.

On-line thermally assisted hydrolysis and methylation (THM) in the presence of both unlabelled and 13C-labelled tetramethylammonium hydroxide (TMAH) was used to assess the relative contributions of phenolics (lignin, demethylated lignin and non-lignin phenolics) in a peaty gley soil profile beneath an unimproved grassland (LL), from a study site located at Harwood (Northumberland, northeast England, UK). This site also includes an unforested moorland (ML) and a second rotation Sitka spruce stand (SS). The common lignin proxies have been corrected for contributions of non-lignin phenols and demethylated lignin in the LL ecosystem and then compared with those from the ML and SS ecosystems. The phenolic compositions from the contributory vegetation inputs (i.e. grasses, heather and Sitka spruce) to all three soils (LL, ML and SS) were also analysed. By using 13C-labelled TMAH it was possible to show that the chemical composition of soil organic matter (SOM) reflected the different vegetation inputs in each of the L/F layers but these characteristics were lost from the deeper organic and mineral layers. Similar changes in the yield of lignin monomers (Λ) with increasing soil depth were displayed in the LL soil profile as reported previously in the ML soil in that no maxima were observed in these amount-depth profiles. The tannin input to the LL soil is low and as a consequence, unlike the ML and SS soils, there is no progressive decrease in the amounts of these non-lignin phenolics with increasing depth. Finally the methylated carbohydrate derivatives (MC) become more abundant relative to the phenolics with increasing soil depth in all three ecosystems (LL, ML and SS).

Despite the recent growth of the organic dairy industry, organic producers and veterinarians have limited information when choosing mastitis treatments for animals in organic dairy production. Organic producers commonly administer homeopathic or other plant-based products without having research evaluating the efficacy of these products and using estimated or no withholding times to treat mastitis and other health problems in their herds. In this pilot study, we attempted to identify several active ingredients of Phyto-Mast (Penn Dutch Cow Care, Narvon, PA), a plant-based mastitis treatment used on organic dairy farms, and to quantify the product residue in milk and plasma after intramammary administration. We developed an assay to quantify thymol (one of the active ingredients in Phyto-Mast) in milk and plasma using gas chromatography and mass spectrometry (GC-MS). Thymol is a volatile aromatic compound with antiinflammatory properties. As a model for dairy cows, 5 healthy, lactating alpine dairy goats were given 5 mL of Phyto-Mast per udder half. For 10 d following treatment, we analyzed blood and milk samples for thymol residues using GC-MS. The GC-MS assay was very sensitive for thymol detection, to a concentration of 0.01 μg/mL in plasma. Using thymol as a marker, Phyto-Mast was detectable and quantifiable in plasma beginning with the 15-min posttreatment sample, but was no longer detectable in the 4-h posttreatment sample. Thymol residues were only detected in the 12-h posttreatment milk sample. An inflammatory response was not evident in the udder following phytoceutical administration. Although this study provides information about the elimination of thymol, the product contains several other active chemicals, which may have different pharmacokinetic behaviors. Further analysis and additional study animals will help to determine a milk withholding time for Phyto-Mast. Given the recent growth of the organic dairy industry, understanding the pharmacokinetics of therapeutics used in organic production and developing accurate withholding recommendations will help to ensure milk safety.

There have been numerous reports of melamine-related illnesses following oral exposure to this contaminant. These studies have been in monogastrics, but there are few reports of adverse effects and pharmacokinetics of melamine in ruminants. The purpose of this project was to determine how melamine is systemically cleared from the blood and milk in lactating animals. Five lactating goats were given a single oral dose of 40 mg/kg body weight. Milk and blood samples were collected for 144 h and analyzed to determine key pharmacokinetic parameters. The apparent plasma half-life (11.12 h) was 3 times longer in these ruminants than that reported in monogastrics and the apparent volume of distribution was more than 6 times greater than that reported in monogastrics. The milk had an apparent half-life of 9.44 h and less than 0.4% of the melamine dose was eliminated in milk. All milk samples were below the LOQ at 4 days (96 h) after exposure. In summary, the pharmacokinetics of melamine in ruminants is not predictive from monogastrics and milk from similarly exposed animals should be condemned for at least 4 days after the last exposure to avoid violation of proposed MRLs or safe levels for milk.

We previously identified the induction of senescence in melanoma cell lines sensitive to diterpene esters, indicating a therapeutic potential. Here we compared the cytostatic effects of two diterpene esters: the prototypic PKC-activating drug TPA (12-O-tetradecanoylphorbol-13-acetate), and the novel compound PEP008 (20-O-acetyl-ingenol-3-angelate) in cell lines derived from melanoma, breast cancer and colon cancer. The diterpene esters induced permanent growth arrest with characteristics of senescence in a subset of cell lines in all three solid tumor models at 100-1000 ng/ml. Use of the PKC inhibitor bisindolylmaleimide-l demonstrated that activation of PKC was required for growth arrest. Full genome expression profiling identified pivotal genes involved in DNA synthesis and cell cycle control down-regulated by treatment in all three sensitive tumor models. At the protein level, prolonged down-regulation of E2F-1 and proliferating cell nuclear antigen (PCNA), sustained expression of p21(WAF1/CIP1) and dephosphorylation of retinoblastoma (Rb) occurred in the sensitive cells. Additionally, the type II tumor suppressor HRASLS3, which has a role in mitogen-activated protein kinase (MAPK) pathway suppression, was constitutively elevated in cell lines resistant to the senescence effects compared to their sensitive counterparts. Together, these results demonstrate that both TPA and the novel PKC-activating drug PEP008 induce growth arrest with characteristics of senescence in solid tumor cell lines derived from a variety of tissue types, and by a similar mechanism. PKC-activating diterpene esters may therefore have therapeutic potential in a subset of breast cancer, colon cancer and melanoma tumors.

In Britain substantial areas of both deep and shallow peatland have been afforested with conifers since the 1950s. However, information about the effects of afforestation on the properties of soil organic carbon (SOC) is lacking. Investigating the geochemical changes that take place when lignin- and tannin-derived phenols are degraded and incorporated into SOC will provide us with an insight into soil carbon dynamics at the molecular level. Here we compare the phenolic distributions in two different peaty gley soil profiles using on-line thermally assisted hydrolysis and methylation (THM) in the presence of both unlabelled and 13 C-labelled tetramethylammonium hydroxide (TMAH). The two soil profiles were beneath respectively an unforested moorland (ML) and a second rotation Sitka spruce ( Picea sitchensis ) afforested moorland (SS), from Harwood (Northumberland, northeast England, UK). THM of these soils in the presence of 13 C-labelled TMAH enabled us to assess the relative contributions of lignin, demethylated lignin, and non-lignin phenolics. The lignin phenolic distributions differed in both soil profiles reflecting changing source and decay dynamics within and between the sites. A progressive degradation of syringyl (S) and guaiacyl (G) phenolics was observed in the ML soil, compared with an increase of such components in the organic/mineral horizons of the SS soil. A significant tannin input was observed, particularly in the upper horizons of the SS soil. The S/G ratio gradually decreased with increasing burial in the ML soil, whilst a change in vegetation input and land preparation was recorded by this ratio in the SS soil. Overall, this suggests that afforestation influences the phenolic compositional profiles in these peaty gley soils as a result of one or more of the following processes: changing vegetation input, horizon inversion prior to planting, root input or leaching. This highlights the potential of using lignin and tannins as molecular indicators to assess the effects of afforestation on SOC.

A physiologically based pharmacokinetic model with partition coefficients estimated from quantum dot (QD) 705 biodistribution was compared with the biodistribution of other QDs in mice and rats to determine the model's predictive ability across QD types, species, and exposure routes. The model predicted the experimentally observed persistence of QDs in tissues but not early time profiles or different QD biodistribution. Therefore, more complex models will be needed to better predict QD biodistribution in vivo.

This paper reports a laboratory study where multiple variables have been monitored in parallel as a function of white-rot incubation time. A model system with Pleurotus ostreatus growing on unamended wheat ( Triticum aestivum ) straw under solid-state cultivation conditions was used to investigate changes in lignin oxidation, amounts of the fungal biomarker ergosterol and activity of manganese peroxidase (MnP) at regular intervals over 84 days. Lignin oxidation was monitored with thermally assisted hydrolysis and methylation (THM) using tetramethylammonium hydroxide (TMAH) where 3,4-dimethoxybenzoic acid, methyl ester (G6) to 3,4-dimethoxybenzaldehyde (G4) [Ac/Al] G and 3,4,5-trimethoxybenzoic acid, methyl ester (S6) to 3,4,5-trimethoxybenzaldehyde (S4) [Ac/Al] S were used as relative lignin decomposition state proxies. [Ac/Al] G, [Ac/Al] S and ergosterol production show little change during the first 21 days of incubation. MnP activity, however, rises rapidly and peak activity is reached during the same time interval. This is followed by an increase in both [Ac/Al] G , [Ac/Al] S and ergosterol formation where most of the increase in these variables takes place after 21 days. Therefore a rapid early rise in manganese-dependent peroxidase activity precedes significant changes both in lignin oxidation and fungal growth. We have demonstrated that it is possible to analyse lignin oxidation using THM in the presence of TMAH and, in the same system, measure enzyme activity and amounts of fungal biomarkers as a function of incubation time. This approach will be useful when investigating compositional changes in litter and soil layers.

The underrepresentation of women faculty in science, technology, engineering and mathematics (STEM) has been studied for several decades. Many programs have been developed with a bottom-up approach toward addressing this underrepresentation. While these bottom-up approaches are an important component in addressing the underrepresentation, the question arises as to their sufficiency for transforming an institution to address the issue. Another method considers the combined use of a bottom-up and a top-down approach in looking toward the goal of institutional transformation by removing barriers to resources that support career success and creating new interventions and resources. This paper reports on the progress of programming and activities that are underway during the first two years of an institutional transformation project (NSF ADVANCE 1209115) at a large-private technical university that incorporates this combined approach. The transformation is outlined using a multi-frame organizational analysis approach from Bolman and Deal[l], which integrates several aspects of organizational theory, including structural, human resources, political and symbolic perspectives. Data used to inform the top-down and bottom-up programming and activities is presented, as well as a discussion of milestones, accomplishments and lessons learned in the context of the overarching project goals and the framework posed by Bolman and Deal's lenses.

The use of water medications is a common practice in the US swine industry to treat and prevent infections in swine herds with minimal labor and without risk of needle breakage. There are concerns that FDA-approved withdrawal times (WDT) may be inadequate for several water medications when exporting pork products to countries where MRLs (maximum residue limits) are lower than US tolerance levels. In this study, withdrawal intervals (WDI) were estimated for pigs when dosed with tetracycline and sulfamethazine in water. The WDI were calculated using the FDA tolerance method (TLM) and a population-based pharmacokinetic method (PopPK). The estimated WDIs (14–16 days using TLM) were similar to the approved WDT of 15 days for sulfamethazine. However, the PopPK method extended WDIs for both sulfamethazine (19–20 days) and tetracycline (12 days) compared to the currently approved WDTs in the U.S. This study also identified potential differences in WDI between weanling and finisher pigs. In conclusion, the TLM may not always provide adequate WDT for foreign export markets especially when MRLs differ from tolerance levels approved for US markets. However, PopPK methods can provide conservative WDIs in situations with considerable variability in medication exposure such as with administration in water.

Tulathromycin is a triamilide antibiotic that maintains therapeutic concentrations for an extended period of time. The drug is approved for the treatment of respiratory disease in cattle and swine and is occasionally used in goats. To investigate the pharmacokinetics of tulathromycin in meat goats, 10 healthy Boer goats were administered a single 2.5 mg/kg subcutaneous dose of tulathromycin. Plasma concentrations were measured by ultra-high pressure liquid chromatography tandem mass spectrometry (UPLC-MS/MS) detection. Plasma maximal drug concentration (Cmax) was 633 ± 300 ng/ml (0.40 ± 0.26 h post-subcutaneous injection). The half-life of tulathromycin in goats was 110 ± 19.9 h. Tulathromycin was rapidly absorbed and distributed widely after subcutaneous injection 33 ± 6 L/kg. The mean AUC of the group was 12,500 ± 2020 h ng/mL for plasma. In this study, it was determined that the pharmacokinetics of tulathromycin after a single 2.5 mg/kg SC injection in goats were very similar to what has been previously reported in cattle.

Medicating drinking water with tetracycline is commonly used in swine production systems to treat and prevent disease outbreaks. However, little information is known of the pharmacokinetics of this medication in water formulations. Twenty-four barrows, divided into 1 control group (of nontreated animals) and 3 equally sized treatments groups (n = 6/group), were treated with tetracycline water medication for 5 d at 125, 250, and 500 mg/L. Blood samples were collected at 0 (prestudy), 4, 8, 12, 24, 32, 48, 56, 72, 80, 96, and 104 h after exposure. Data analyses consisted of a noncompartmental pharmacokinetic analysis and statistical analysis of steady state concentrations with repeated measures ANOVA and multiple-comparison testing to determine whether plasma concentrations differed among groups. Derived pharmacokinetic parameters were consistent with previously published feed and intravenous data. Plasma tetracycline concentrations at steady state were 0, 0.33, 0.47, and 0.77 microg/mL for 0-, 125-, 250-, and 500-mg/L exposures, respectively. Treatment group steady-state plasma concentrations were significantly different from plasma concentrations in control animals (P < 0.0001); however, whereas the 125- and 250-mg/L groups were significantly different from the 500-mg/L group (P < 0.0001), their mean plasma tetracycline concentrations did not differ from one another. Furthermore, the study showed that tetracycline oral bioavailability is very small. The dose response curve also shows that concentrations of plasma tetracycline increase linearly, yet not in a 1 to 1 ratio, to the direct increase in water medication dose.

Sulfamethazine is often used to treat disease in the swine industry. Sulfamethazine is available as water or feed medication and historically (over the past 40 years) has been associated with residue violations in both the United States and Europe. Despite sulfamethazine's approval for use as a water medication, little research on the pharmacokinetics of the water formulation is available. Therefore, a pilot study was performed to determine the plasma levels of an approved sulfamethazine water medication. Plasma levels in pigs treated with an oral bolus (250 mg/kg), which is equivalent to the total drug consumed within a 24-h period, achieved therapeutic concentrations (50 microg/ml). Noncompartmental-based pharmacokinetic model parameters for clearance, half-life, and volume of distribution were consistent with previously published values in swine. However, the above treatment resulted in exposure of pen mates to sulfamethazine at levels currently above tolerance (0.1 ppm). Using a physiologically based pharmacokinetic model, the treatment dose simulation was compared with observed plasma levels of treated pigs. Flexibility of the physiologically based pharmacokinetic model also allowed simulation of control-pig plasma levels to estimate contamination exposure. A simulated exposure to 0.15 mg/kg twice within approximately 8 h resulted in detectable levels of sulfamethazine in the control pigs. After initial exposure, a much lower dose of 0.059 mg/kg maintained the contamination levels above tolerance for at least 3 days. These results are of concern for producers and veterinarians, because in commercial farms, the entire barn is often treated,and environmental contamination could result in residues of an unknown duration.

Melamine-contaminated pet food was recently added as a supplement to livestock feed. There is little or no information concerning the pharmacokinetics of melamine in livestock, and the aim of this study was to obtain pharmacokinetic parameters for this contaminant in pigs. Melamine was administered intravenously to five weanling pigs at a dose of 6.13 mg/kg and plasma samples were collected over 24 h, extracted for melamine, and then analyzed by HPLC–UV. The data was shown to best fit a one-compartment model with melamine’s half-life of 4.04 (±0.37) h, clearance of 0.11 (±0.01) L/h/kg, and volume of distribution of 0.61 (±0.04) L/kg. These data are comparable to the only mammalian study in rats and suggests that melamine is readily cleared by the kidney and there is unlikely to be significant tissue binding. Further tissue residue studies are required to assess the depletion kinetics of this contaminant in the pig which will determine whether residue levels in the kidney should be of public health concern if pigs were exposed to a similar dose.

In recent years, computer and network security education has become a priority among employers, the government, colleges and universities. Another emerging priority among these institutions is collaborative working environments. Rochester Institute of Technology (RIT) is among the many universities and colleges developing and offering Computer and Network Security curriculum. However, we believe that our innovative approach to this new curriculum is unique because of the collaborative nature of the "ABC Defense Lab Project" recently implemented across course boundaries. The Information Technology Department offers a Bachelor of Science Degree in Applied Networking and Systems Administration (ANSA). Within this degree, the curricula is distinctly segmented between the two areas. The networking curriculum studies the infrastructure that ties all the systems together. Students learn about the protocols and technologies required to design, implement, troubleshoot and maintain a complete local area network. The systems administration curriculum studies what is happening "inside the box". Students once again learn about the design, implementation, troubleshooting and maintenance, but here the focus is on the servers and desktop systems required to run within a company.Within the ANSA Bachelor of Science Degree Program, two courses are offered to undergraduate students: Computer Systems Security and Network Security. These courses serve to provide conceptual and practical instruction in the afore-mentioned areas.The ABC Defense Lab Project was initially introduced in the Systems Administration Security course. Students were assigned membership in one of three teams: Team A, Team B or Team C. Ten to 15 students made up each of the three teams. Each team was provided with a set of equipment to configure, secure and eventually defend against attacks from the other two teams. Since this was a project within the Systems Administration Security course, the focus was on securing the systems and not the network.As the Network Security course was first offered approximately one year after the initial offering of the Systems Administration Security course, a very similar project was assigned. However, the focus was of course on designing, building, securing, and defending the network infrastructure.Although the project was successful in both courses, there were some drawbacks. Namely, the System Administration Security students still needed a network on which to run their systems and the Network Security students still needed systems on their network to be able to provide services. For the students in each course, this detracted from the task at hand; Systems Administration Security or Network Security, because they had to deal with the other side of the issue.While these two courses were originally designed independently of one another, in the Spring Quarter of the 2003 academic year, Professors Sharon Mason and Daryl Johnson took a new approach to teaching this security curriculum. In this new approach students from each of the two classes worked together in one of three teams (Team A, Team B, or Team C) to design, implement and secure a network with computer systems offering services. The teams were then instructed to work with one another across course boundaries; Team A from the Systems Security class worked with Team A from the Network Security class, etc. In the end each of the teams defended their own network and systems while attempting to exploit security flaws in the other teams' network and systems. This is the first time in the ANSA Degree Program that such a project has been attempted at this scope and scale across course boundaries.This paper explores the triumphs and breakdowns of this innovative collaborative class project. The successes and failures of both the students and the instructors are reviewed to provide insights as to the lessons learned from this experience.

Several studies indicate each piglet within a litter is affected differently by weaning and that these individual responses may be altered by the age of the litter at weaning. In the present study, eight litters of individually marked piglets (n=79) were randomly assigned to be weaned at 21 or 35 days (four litters per treatment), and behavioural and physiological measures were used to elucidate the relationship between age and weaning, and the within-litter variation in short-term responses to weaning that is thought to occur. Weights, vocalisations, littermate directed behaviours, suckling related parameters and salivary cortisol concentrations were recorded for individual piglets on both treatments from birth to 37 days of age. The time piglets spent at the udder, massaging or suckling, gradually increased over the preweaning period, as did aggression, nosing and chewing of littermates. Higher levels of aggression were found on the day of weaning relative to the 2 days following, whilst vocalisations did not peak until the day after weaning. Salivary cortisol was found to be lower for D35 piglets (day×treatment effect: F2,116(38)=2.67, P=0.073) and ‘high vocalisations’ twice as high on the day after weaning for D21 piglets compared to D35 (day×treatment effect: F2,146(8)=3.93, P
