Gamma-aminobutyric acid (GABA), a primary neurotransmitter, accomplishes its activities by binding to different receptor sites in different conformations. It is known to have two major conformers: the closed and open forms. Earlier studies on preferred conformation of GABA in water revealed differing results with some reporting the open form while others inferring the closed form to be more stable. We found the existence of many open forms and only one closed form of GABA in water through ab initio metadynamics simulation. Some of the open conformers are equally or more stable while others are less stable than the closed form. Free energy barriers reveal that different conformers are interconvertible at room temperature in typical experimental time scales. Ab initio molecular dynamics simulations are performed to further investigate the inter-conversion of various conformers of GABA in water and their dipole moments and also to make connections to experiments on the conformation of GABA in water. Local approximations of high-level ab initio methods make superior accuracy in the computation of molecular properties accessible by drastically decreasing computational times. As a consequence, these methods become applicable not only for large systems but also in schemes for which large numbers of calculations are necessary. In this work, we apply a recently developed open-shell implementation of the domain-based pair natural orbital coupled cluster singles doubles (DLPNO-CCSD) approach for the computation of vibrational corrections to the isotropic values of electron paramagnetic resonance (EPR) hyperfine coupling constants. We assess density functional theory (DFT) and DLPNO-CCSD approaches using two common but very different schemes: (1) vibrational perturbation theory based on equilibrium geometries, and (2) explicit canonical ensemble averages using configuration snapshots sampled from revPBE0-D3(0) ab initio molecular dynamics simulations. Both approaches are found to yield very similar results for the spin probe 2,2,3,4,5,5-hexamethylperhydroimidazol-1-oxyl (HMI) and are both feasible for systems of around 30 atoms. However, the numerical stability required for higher derivatives can become a limitation for local correlation methods in the case of vibrational perturbation theory. [ABSTRACT FROM AUTHOR]

The influence of processing variables was experimentally studied for glass fiber reinforced epoxy–clay nanocomposites manufactured using vacuum-assisted wet layup method. The tensile strength, flexural strength, and interlaminar shear strength of these nanocomposites were significantly influenced by the processing variables including the temperature of resin–clay mixture, speed of homogenization, and ultrasonic probe amplitude during premixing of clay minerals in epoxy. The glass transition temperature of glass fiber reinforced composites increased with incorporation of clay minerals in epoxy. Also, the postcuring of the laminates was carried out at three different temperatures, e.g. 100, 130, and 150 °C for 3 h. A decrease in tensile modulus, tensile strength, and flexural strength of nanocomposites postcured at 130 and 150 °C was observed. Also, the use of non-stoichiometric epoxy resin and hardener ratios had an adverse effect on mechanical properties of fiber reinforced epoxy–clay nanocomposites. In fiber reinforced composites incorporating clay minerals, a uniform dispersion of clay minerals besides a strong interfacial adhesion between clay minerals and polymer and optimum conditions of curing of matrix is a crucial aspect for improved performance over conventional fiber reinforced composites. Despite the broadly acknowledged importance of solvation effects on measured UV/Vis spectra in the context of solvatochromism or chemical reactions in solution, it is still an open challenge to calculate UV/Vis spectra with predictive accuracy. This is particularly true when it comes to the impact of nuclear quantum effects on these experimental observables. In the present work, we calculate the UV/Vis absorption spectrum of indole in aqueous solution with a combination of a correlated wavefunction method for computing electronic excitation energies and enhanced path integral simulations for rigorous sampling of nuclear configurations including the quantum effects in solution. After validating our approach based on gas-phase benchmarking, we demonstrate that the lineshape of the spectrum measured in aqueous solution is quantitatively recovered, without the application of any shifting, scaling, or broadening, only after including nuclear quantum effects in addition to thermal fluctuations and solvation at ambient conditions. Our findings demonstrate that nuclear quantum effects are "visible" in UV/Vis spectra of chromophores measured in solution even at room temperature and, therefore, that they must be considered computationally to achieve predictive accuracy., (© 2024 Wiley‐VCH GmbH.)

Free radical species are used as spin labels in electron paramagnetic resonance (EPR) spectroscopy of biomolecular systems in water, for instance in the frame of Overhauser dynamic nuclear polarization (ODNP) relaxometry to probe the local hydration water dynamics close to protein surfaces in aqueous environments. Widely used in this context are nitroxide spin probes such as TEMPO, PROXYL or MTSL derivatives. Here, we study the THz spectroscopy of HMI (2,2,3,4,5,5-HexaMethylImidazolidin-1-oxyl) in water at ambient conditions which has been recently investigated as to how its EPR properties depend on its solvation pattern in water. To enable theoretical THz spectroscopy of molecular radicals in solution, we have generalized well-established methodologies for THz spectral decomposition of closed-shell systems, namely the supermolecular solvation complex (SSC) and cross-correlation analysis (CCA) techniques, to open-shell polyatomic solute species in water. Based on this methodological advance, we have decomposed and assigned the THz response of HMI including its solvation shell by employing the generalized SSC and CCA methods to cope with the open-shell character of this free radical solute, in particular its unpaired electron localized at the nitroxy group. We reveal that the main modulations of the far-IR spectrum of HMI are dominated by the low-frequency intramolecular modes of the spin probe molecule itself while the solvation of its two hydrogen bonding sites contribute much less intensely in this spectral window. Finally, we have computed THz spectra of HMI with its local solvation water with an aim to provide a theoretical analogue of experimental THz difference spectroscopy. Beyond the specific case, our decomposition methodology that now is able to include open shells can be applied in future work to analyze the low-frequency vibrational response of the solvation shell of other free radicals in solution.
