With the continuous progress of the society, the new media network has proposed new conditions and provided new means for implicit ideological and political education in private colleges and universities. This article discusses the launching of the e-class platform for private college counselors to develop the feasibility of implicit ideological and political education for private colleges. The involved methods of education are deeply explored, in order to improve the implicit ideological and political education work ability of private college counselors.

College students from five private colleges and universities in Guangdong Province were selected as the research subjects. By using entrepreneurship education as a variable in the theory of planned behavior (TPB) model, a relationship model was constructed based on the influencing factors of intention of participating in online entrepreneurship among private college and university students. Then, the mechanism of entrepreneurial attitude, subjective norm, perceived behavior control, and entrepreneurial education on online entrepreneurial willingness were analyzed. Results show that the standardized path coefficients of entrepreneurial attitude, subjective norm, perceived behavior control, entrepreneurial education and online entrepreneurial intention are as follows: 0.251, 0.239, 0.163 and 0.244, respectively, showing a significant positive impact on online entrepreneurial willingness.

This year, the number college graduates in China surpassed tens of millions for the first time. The employment rate of fresh graduates has been concerning. In view of this, network entrepreneurship has become an important channel and platform to solve the problem of college students’ employment. The government, society and universities has been actively encouraging college students' network entrepreneurship as the value of college students' network entrepreneurship is significant. This paper analyzes the current situation of college students' network entrepreneurship and the problems faced through the survey data and thus suggests corresponding countermeasures.

Bietti crystalline corneoretinal dystrophy (BCD) is an autosomal recessive retinal degenerative disease characterized by yellow-white crystal deposits in the posterior pole, degeneration of the retinal pigment epithelium (RPE), and sclerosis of the choroid. Mutations in the cytochrome P450 4V2 gene (CYP4V2) cause BCD, which is associated with lipid metabolic disruption. The use of gene-replacement therapy in BCD has been hampered by the lack of disease models. To advance CYP4V2 gene-replacement therapy, we generated BCD patient-specific induced pluripotent stem cell (iPSC)–RPE cells and Cyp4v3 knockout (KO) mice as disease models and AAV2/8-CAG-CYP4V2 as treatment vectors. We demonstrated that after adeno-associated virus (AAV)-mediated CYP4V2 gene-replacement therapy BCD–iPSC–RPE cells presented restored cell survival and reduced lipid droplets accumulation; restoration of vision in Cyp4v3 KO mice was revealed by elevated electroretinogram amplitude and ameliorated RPE degeneration. These results suggest that AAV-mediated gene-replacement therapy in BCD patients is a promising strategy.

Current guidelines recommend adjuvant chemotherapy (ACT) for stage II colorectal cancer (CRC) patients by using clinical high risk factors, with which circulating tumor cells (CTCs) were not considered. Here, an assessment to detect CTCs based on subtraction enrichment mediated by magnetic beads conjugated with CD45, immunofluorescence staining of CK, and fluorescence in situ hybridization of CEP8 is established. Neutralizing monoclonal antibodies (NAbs) with prophylactic and therapeutic efficacy have demonstrated fundamental importance in the control of SARS-CoV-2 transmission. However, their wide application has been largely limited by high cost and inconvenience in administration. Here, we developed an intranasal spray containing two synergetic human NAbs that could broadly neutralize the emerging Omicron variants in vitro. A unique synergetic neutralizing mechanism was identified that the two NAbs bound to exclusive epitopes on the RBD and structurally compensate each other in blocking the Spike-ACE2 interaction. Importantly, when given at low dosages for three consecutive days through the intranasal mucosal route, this cocktail showed significant improvement in the emergency preventive and therapeutic effects in hamsters challenged with authentic Omicron BA.1. Further, we performed an investigator-initiated trail in healthy volunteers (ChiCTR2200066525) to study the safety and pharmacokinetics of the antibody cocktail administrated as nasal spray. The nasal spray is generally safe and well tolerated without treatment related severe abnormal effects. The antibody cocktail nasal spray demonstrated nasal concentrations higher than the IC90of neutralization activity against Omicron BA.4/5 even at 24 hours post dosing. This study aimed to investigate the effects of Tilapia skin peptides on cyclophosphamide-induced premature ovarian failure in mice and to further elucidate the potential mechanisms.

Students from private undergraduate universities were included in this study as research subjects. Questionnaires were distributed to the students to collect information in order to keep abreast of students’ motivation, preference, preparation and plan, encountered difficulties and adjustments made, demands, as well as other aspects about their postgraduate entrance examination. The results showed that students from private universities have clear motivation for their postgraduate entrance examination, but there are some problems, such as the lack of scientific choices, long preparation time, more challenges, and lack of school support. In view of these problems, this study offers countermeasures and suggestions for these students and relevant schools.

Poor intrinsic conductivity and the presence of irreversible lithiation phase affect the electrochemical performance of silicon oxycarbide anode materials. Even though it can be improved by increasing free carbon content or composition, scarification of reversible capacity and initial Coulombic efficiency (ICE) remain as challenge. Here, polycarbosilane (PCS) with alternating distribution of silicon and carbon atoms is employed as precursor of SiOC ceramics. Air oxidation cross-linking is used to regulate the content of oxygen and carbon elements in PCS at atom level, so as to explore a solution to improve the intrinsic conductivity and reversible lithium phase content of SiOC ceramics. This strategy provides extremely excellent rate capability, areal/volumetric capacity, and ICE. This is also the first concept for feasible precursor structure design to control the SiOC glass phase and regulate the growth of C nanoribbon that can improve the intrinsic conductivity and reversible capacity of SiOC ceramic anode materials.

Adipocyte tissue inflammation orchestrates the occurrence and development of diseases induced by obesity. Here, the effects of Lagerstroemia speciosa (L.) Pers total flavonoids (LTF) on TNF-α-induced insulin resistance (IR) and inflammatory response in 3T3-L1 adipocytes and underlying mechanisms were studied. Cell glucose uptake test confirmed that TNF-α could induce IR in 3T3-L1 adipocytes. The sensitivity of 3T3-L1 adipocytes to insulin was increased by LTF treatment. The phosphorylation of IRS1 and Akt in TNF-α-treated 3T3-L1 adipocytes was markedly restored by LTF treatment. The results revealed that interleukin-6 (IL-6) and monocyte chemoattractant protein-1 (MCP-1) mRNA expression was elevated in TNF-α-administered 3T3-L1 adipocytes, whereas the mRNA expression of adiponectin (ADPN) was considerably decreased. When treated with 50 μg/mL LTF, the IL-6 and MCP-1 mRNA expression levels were drastically reduced, and the ADPN mRNA expression level was significantly raised. Moreover, the concentration of IL-6 in the supernatant of the medium was decreased after LTF treatment compared with the model group. Western blotting results showed that the phosphorylation of p38, JNK, and ERK of mitogen-activated protein kinase (MAPK) signaling pathways and IкB of NF-кB signaling pathway were inhibited. It suggested that LTF improves TNF-α-induced IR and inflammatory response in 3T3-L1 adipocytes via inhibiting MAPK and NF-кB signaling pathways.

Silicon is considered as the desirable anode material for lithium‐ion batteries due to its suitable discharge potential, abundant reserve, and ultra‐high specific capacity. However, poor conductivity and unstable battery performance caused by large volume change during cycling limit the further development of silicon anode. In order to avoid the unstable solid electrolyte interface layer caused by the direct contact between silicon and electrolyte, and to eliminate the structural collapse caused by the volume change during cycling, dimension size reduction, reserved voids, and novel structural framework are usually adopted. Although these methods can effectively improve the defects, a new problem is introduced and cannot be ignored: contact engineering between the coating layer and silicon core. Herein, contact engineering is classified into three categories: face to face (F2F), line to line (L2L), and point to point (P2P) according to the contact modes between the coating and core. There is utilizability of the structure categories of different contact modes and their influence on electrochemical performance. Therefore, in the future research of silicon anode, contact engineering is a non‐negligible aspect in the structural design process. Finally, feasible strategies based on contact engineering have been indicated.

Improving the electromagnetic wave (EMW) absorption performance of continuous SiC fibers is of great significance for the development of structure-function integrated composites. Doping magnetic metal elements into SiC fibers can effectively improve their dielectric/magnetic loss, which is expected to produce SiC fibers with wide frequency EMW absorption. At present, introducing metal into continuous SiC fibers generally has the disadvantages of (1) low metal content and (2) weak EMW absorption ability. In this paper, vinylferrocene (VF) with monofunctional groups was selected as the iron source to introduce Fe atoms into polycarbosilane (PCS) through chemical modification without changes in the molecular framework of PCS, which is beneficial for the melt spinning of the precursors. A series of continuous Fe-SiC fibers were prepared by polymer-derived ceramic approach, and their composition, structure and EMW properties were systematically studied. Besides SiC crystallization, the turbostratic carbon, core–shell structures of Fe5Si3@C and Fe3Si@C were identified for the continuous Fe-SiC fibers. It is worth noting that in the present work, (1) the introduction of Fe catalyzes the formation of turbostratic carbon, which can be observed at a low temperature of 900 °C and (2) the highest Fe content (7.70 wt%) was obtained among all the reported continuous Fe-SiC fibers. The unique microstructure of final Fe-SiC fibers demonstrated outstanding EMW absorbing performance. For the Fe-SiC-2-900 °C fiber, the minimum reflection loss (RLmin) reached −54.63 dB at 10.2 GHz. By adjusting the thickness, the effective absorption bandwidth of Fe-SiC-3-1200 °C can almost cover the X-band. The results indicate that the presented continuous Fe-SiC fibers exhibit excellent EMW absorbing performance.

Neutralizing antibodies (NAbs) can prevent and treat infections caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). However, continuously emerging variants, such as Omicron, have significantly reduced the potency of most known NAbs. The selection of NAbs with broad neutralizing activities and the identification of conserved critical epitopes are still urgently needed. Here, we identified an extremely potent antibody (55A8) by single B-cell sorting from convalescent SARS-CoV-2-infected patients that recognized the receptor-binding domain (RBD) in the SARS-CoV-2 spike (S) protein. 55A8 could bind to wild-type SARS-CoV-2, Omicron BA.1 and Omicron BA.2 simultaneously with 58G6, a NAb previously identified by our group. Importantly, an antibody cocktail containing 55A8 and 58G6 (2-cocktail) showed synergetic neutralizing activity with a half-maximal inhibitory concentration (IC50) in the picomolar range in vitro and prophylactic efficacy in hamsters challenged with Omicron (BA.1) through intranasal delivery at an extraordinarily low dosage (25 μg of each antibody daily) at 3 days post-infection. Structural analysis by cryo-electron microscopy (cryo-EM) revealed that 55A8 is a Class III NAb that recognizes a highly conserved epitope. It could block angiotensin-converting enzyme 2 (ACE2) binding to the RBD in the S protein trimer via steric hindrance. The epitopes in the RBD recognized by 55A8 and 58G6 were found to be different and complementary, which could explain the synergetic mechanism of these two NAbs. Our findings not only provide a potential antibody cocktail for clinical use against infection with current SARS-CoV-2 strains and future variants but also identify critical epitope information for the development of better antiviral agents.

The quality of graduates is the key factor in evaluating the cultivation effect of colleges and universities. Quantification of whether the graduates qualify for their working post in companies and industries provides conduction for further college cultivation reform enhancement. In this work, we proposed an adaptive multivariate neural network architecture for fusion evaluation of college student cultivation. Specifically, we designed a questionnaire to collect data on the current working status of 1231 graduates and recorded 32 in-school training items categorized into four different modules. For quantitative evaluation, 10 indices of career-require competence were set to describe the graduates’ job abilities. The fused contribution of the in-school training items to the career-required competence was predicted by the multivariate network model with the linking weights adaptively trained. A comprehensive contribution matrix was generated by discrete PCA multivariate transforming to provide a digital reference for the network training. A 7-level scoring system was designed for quantifying the contribution matrix. For model optimization, the network structure was tuned by testing a different number of hidden nodes. The model was trained and optimized to reveal the direct correlation between college cultivation and job-required abilities. Experimental results indicated that the methodology we proposed is feasible to evaluate the cultivation mode in colleges and universities, theoretically and technically providing positive directions for colleges and universities to make their cultivation reforming, as to enhance the quality of their graduates.

Glycyrrhiza and sargassum are among the 18 incompatible medicaments according to traditional Chinese medicine (TCM) theory. Although it contains glycyrrhiza and sargassum, Haizao Yuhu decoction (HYD) is a classic prescription widely used as TCM to treat goiter. According to the Chinese Pharmacopoeia, glycyrrhiza is divided into three varieties: Glycyrrhiza uralensis Fish., Glycyrrhiza glabra L., and Glycyrrhiza inflata Bat. Whether the three varieties of glycyrrhiza have different efficacy or toxicity when applied in the HYD is unknown.To explore whether the HYDs comprising three varieties of glycyrrhiza have different efficacy or toxicity when used to treat goiter in rats and the underlying mechanisms of these HYDs.For two weeks, the goiter model was replicated by intragastric propylthiouracil (PTU) administration. Samples were divided into the control group, model group, euthyrox group, HYD with glycyrrhiza uralensis (HYD-U) group, HYD with glycyrrhiza glabra (HYD-G) group, and HYD with glycyrrhiza inflata (HYD-I) group. After four weeks of treatment, body weight, rectal temperature, thyroid/liver/kidney coefficient, thyroid/liver/kidney function, thyroid/liver/kidney histomorphology, and thyroid ultrastructure were evaluated. Then, real-time quantitative reverse-transcription polymerase chain reaction (RTqPCR), Western blot, and immunofluorescence analyses were performed to detect genes and proteins affecting autophagy and apoptosis in thyroid cells in the AMP-activated Protein Kinases (AMPK)/Mammalian target of rapamycin (mTOR) pathway.All three HYDs increased thyroid hormones (THs) levels, relieved thyroid pathological tissue and ultrastructure, and activated vital proteins and genes in the AMPK/mTOR pathway. Comparisons among the efficacy of the three HYDs indicated that HYD-U restored the THs most effectively; however, no difference in the anti-goiter effect was observed. Moreover, the three HYDs resulted in no toxicity and promoted the recovery of impaired liver and kidney function caused by PTU. Comparisons among the recovery effects of the three HYDs on the liver and kidney were the same.Our experiments demonstrated that the three HYDs had outstanding anti-goiter effects and protected liver and kidney function. Their anti-goiter effects were attributed to AMPK/mTOR pathway-induced autophagy and apoptosis. HYD-U resulted in the best THs recovery. It was further indicated that in our present study, glycyrrhiza and sargassum were compatible in the three HYDs, thereby suggesting their safety of compounding in HYD and providing a basis for the research of the 18 incompatible medicaments.

SARS-CoV-2 variants with adaptive mutations have continued to emerge, causing fresh waves of infection even amongst vaccinated population. The development of broad-spectrum antivirals is thus urgently needed. We previously developed two hetero-bivalent nanobodies (Nbs), aRBD-2-5 and aRBD-2-7, with potent neutralization activity against the wild-type (WT) Wuhan isolated SARS-CoV-2, by fusing aRBD-2 with aRBD-5 and aRBD-7, respectively. Here, we resolved the crystal structures of these Nbs in complex with the receptor-binding domain (RBD) of the spike protein, and found that aRBD-2 contacts with highly-conserved RBD residues and retains binding to the RBD of the Alpha, Beta, Gamma, Delta, Delta plus, Kappa, Lambda, Omicron BA.1, and BA.2 variants. In contrast, aRBD-5 and aRBD-7 bind to less-conserved RBD epitopes non-overlapping with the epitope of aRBD-2, and do not show apparent binding to the RBD of some variants. However, when fused with aRBD-2, they effectively enhance the overall binding affinity. Consistently, aRBD-2-5-Fc and aRBD-2-7-Fc potently neutralized all of the tested authentic or pseudotyped viruses, including WT, Alpha, Beta, Gamma, Delta, and Omicron BA.1, BA.1.1 and BA.2. Furthermore, aRBD-2-5-Fc provided prophylactic protection against the WT and mouse-adapted SARS-CoV-2 in mice, and conferred protection against the Omicron BA.1 variant in hamsters prophylactically and therapeutically, indicating that aRBD-2-5-Fc could potentially benefit the prevention and treatment of COVID-19 caused by the emerging variants of concern. Our strategy provides new solutions in the development of broad-spectrum therapeutic antibodies for COVID-19.

Circular RNAs (circRNA) have been reported as regulators involved in hepatocellular carcinoma (HCC), but their mechanism of activity remains unknown. This study performed quantitative reverse-transcription polymerase chain reaction to determine if circNFATC3 was downregulated in 46 paired HCC tissues and cell lines. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, apoptotic, and transwell assay proved that circNFATC3 can inhibit hepatoma cell proliferation, apoptosis, and migration/invasion in vitro. Mouse xenograft assay demonstrated that circNFATC3 suppressed tumor size and weight and reduced lung metastasis in vivo, and vice versa. The RNA-seq results showed that NFATC3 itself was the most significantly differentially expressed gene when circNFATC3 was manipulated, and bioinformatics and luciferase reporter assays verified circNFATC3 regulated the expression of NFATC3 by interacting with the hsa-miR-548I. Additionally, it was also indicated that the level of NFATC3 was downregulated in HCC patients also and was significantly correlated with the staging and prognosis of HCC. Moreover, both circNFATC3 and NFATC3 were shown to inhibit the phosphorylation of JNK, c-Jun, AKT, and mTOR signaling pathways. Overall, the circNFATC3 can sponge miR-548I to protect NFATC3 itself, then it regulates hepatoma cell function via the JNK, c-Jun, AKT, and mTOR signaling pathways, and the circNFATC3 can be a tumor-repressor on HCC.

Objective: The Flower of Lobed Kudzuvine [Pueraria lobata (Willd.)Ohwi; Gehua in Chinese; GH] and Japanese Raisin Tree Seed (Hovenia dulcis Thunnb.; Zhijuzi in Chinese; ZJZ) are herbs that have been used in China for the treatment of alcohol intoxication and liver diseases. We aimed to evaluate the hepatoprotective potential of a combination of these in mice with acute alcohol-induced liver injury, and to elucidate the mechanisms involved. Methods: Male ICR mice were randomly allocated to six groups: a control group, an alcohol-administered group, and groups that were administered alcohol and one of silibinin, the GH, the ZJZ or a GH-ZJZ combination (at a ratio of 2:1). Animals were orally administered 56% alcohol (Er Guo-tou white spirit, 0.12 mL/10 g/d) for 10 days and at the end of this period, hepatic biochemical indicators, antioxidant parameters, alcohol metabolic enzymes, and histopathologic changes were evaluated. Moreover, the expression of the signaling molecules KEAP1, NRF2, and AQP9 were measured by qRT-PCR and western blotting. Results: Compared with the model group, GH-ZJZ (2:1) had lower serum ALT (12.15 ± 0.39, P = .003), AST (104.07 ± 1.03, P = .001), and ALP (148.09 ± 2.55, P = .010) activities, and lower TC (1.97 ± 0.05, P = .001) and TG (1.54 ± 0.07, P = .002) concentrations. During the two-year pandemic of coronavirus disease 2019 (COVID-19), its causative agent, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has been evolving. SARS-CoV-2 Delta, a variant of concern, has become the dominant circulating strain worldwide within just a few months. Here, we performed a comprehensive analysis of a new B.1.617.2 Delta strain (Delta630) compared with the early WIV04 strain (WIV04) in vitro and in vivo, in terms of replication, infectivity, pathogenicity, and transmission in hamsters. Fusion with host cell membrane is the main mechanism of infection of SARS-CoV-2. Here, we propose a new strategy to double block SARS-CoV-2 membrane fusion by using Harringtonine (HT), a small-molecule antagonist. By using cell membrane chromatography (CMC), we found that HT specifically targeted the SARS-CoV-2 S protein and host cell TMPRSS2, and then confirmed that HT can inhibit pseudotyped virus membrane fusion. Furthermore, HT successfully blocked SARS-CoV-2 infection, especially in the delta and Omicron mutant. Since HT is a small-molecule antagonist, it is minimally affected by the continuous variation of SARS-CoV-2. Our findings show that HT is a potential small-molecule antagonist with a new mechanism of action against SARS-CoV-2 infection, and thus HT mainly targets the S protein, and thus, greatly reduces the damage of the S protein’s autotoxicity to the organ system, has promising advantages in the clinical treatment of COVID-19.

Optimal human resources allocation asks to employ a person to work in the position corresponding to his/her ability. Employment competence is the key feedback to the cultivation of college students’ working ability. The data relationship needs to analyze between the in-school cultivation items and the working abilities required by the companies. Machine learning framework is introduced to study the companies’ responses to the cultivation of college students. In this work, a dual-network architecture is built up for statistical modeling evaluation of college graduates’ working ability in consistence with their job position and remuneration. A requirement network and a cultivation network are constructed for extracting features from the original working ability data required by companies and cultivated ever in school. The networks are fully trained by adaptively tuning the linking weights. The extracted features are fused together to estimate the working competence of each target sample/person. To evaluate the dual-network model, a modeling index system is designed, including proposing a total evaluation index calculus for the dual-network model, and a variable importance index from the original data. The samples are consequently ranked by the model predicted index and by the variable importance index, respectively. The ranking difference is used to evaluate the prediction efficiency of the dual-network model. Experimental results show that the dual network architecture is feasible to establish statistical models for the evaluation of college graduates’ in-school cultivated working ability in consistence with the company’s required working ability at their job position and their deserved remuneration.
