A prominent strengthened Y2O3 stabilized t-ZrO2 (YSTZ)/MgO nanocomposite coating is achieved by the plasma electrolytic oxidation (PEO) process and in-situ synthesized YSTZ reinforced phase with a quantitative control approach. The idea of activating double slip systemic semi-coherent interface dislocations in YSTZ/MgO nanocomposite coating to realize crack self-healing is proposed. High dislocation densities are associated with {101} YSTZ slip and {111} MgO slip system to coordinate interfacial deformation to stop crack initiation and propagation. This crack propagation path can absorb more fracture energy, providing more opportunities for crack deflection and bridge, which closes crack and realizes crack self-healing.

Synchroa medogensis, a new species of Synchroidae Lacordaire, 1859, is described and illustrated, based on a single male collected from Mêdog, Xizang, China. This new species is close to S. elongatula Nikitsky, 1999 and S. melanotoides Lewis, 1895, but could be distinguished by the elongated antennae and elytra, the apically narrowed prosternal process and the stout parameres.The new information of this new species provided in this paper include: description, type information, distribution and comparison amongst S. medogensis sp. nov., S. elongatula and S. melanotoides.

Ward-acquired infections (WAIs) has become a critical concern, particularly for ships with limited resources. An assessment approach combining validated computational fluid dynamics and the novel Wells-Riley assessment method is proposed to evaluate risks in a shipboard ward. Subsequently, the effects of ventilation, bed-side curtains, and non-adiabatic boundaries on substantial aerosol dispersion are analysed, including X/Y directional walkways (activity zones). Results show that patients with severe symptoms can be prioritized for the exhaust zone rather than the supply zone, and the maximum reduction in suspended particles can differ up to 3-folds. Bed-side curtains can decrease the average risk by −26 % and −48 %/−32 %, respectively, for X/Y directional walkways. Moreover, the average risk increases by approximately +21 % and +14 %/+11 % for X/Y directional walkways under a low air change rate (ACH) of 6 h−1. Considering the non-adiabatic boundary, the average risk reduction is approximately −6.2 % for winter and −7.0 % for summer. It supports that bed-side curtains and a recommended ACH of 12 h−1 are effective in reducing risk. The heat transfer through walls has a marginal effect on risks. Overall, this study provides a framework for predicting and analysing spatial-temporal exposure risks. Better strategies can be developed for protecting medical staff from WAIs.

The Paleogene Shahejie Formation in the KL16 oilfield, Bohai bay, is characterized by a thinly interbedded mixed sedimentary system, with complex sedimentary facies, lithologic types and distributions. It is hard for conventional logging methods to identify the lithology therein. In order to solve the difficulty in lithologic identification of mixed sedimentary system, analyses based on graph data base using elemental capture energy spectrum log have been proposed. Due to the different composition for the various minerals, we innovatively established the molar numbers of silicon, calcium, magnesium, and aluminum as characteristic parameters for sandstone, limestone, dolomite, and mudstone, and a graph clustering analysis method was applied to identify lithology. Considering the seismic waveforms corresponding to lithologic impedance of reservoir, three seismic phases were identified by neural network clustering analysis of seismic waveform, and the seismic attributes with high sensitivity to reservoir thickness were then selected to realize the fine description of the mixed carbonate-siliciclastic reservoir. Drilling results confirmed that the sedimentary facies were accurately identified, with reservoir prediction accuracy reaching up to 80%. Under the guidance of reservoir research, the oil-in-place discovered in the oilfield were estimated to be more than 5 million tonnes. This technology provides reference for the exploration and development of oilfields of mixed sedimentary system.

Abstract Background To determine the efficacy and safety of intravitreally injected conbercept, a vascular endothelial growth factor receptor fusion protein, for the treatment of idiopathic choroidal neovascularization (ICNV). Methods This retrospective study analyzed outcomes in 40 patients (40 eyes) with ICNV who received intravitreal injections of conbercept 0.5 mg (0.05 ml) and were followed up for at least 12 months. All patients underwent full ophthalmic examinations, including best-corrected vision acuity (BCVA), intraocular pressure (IOP), slit-lamp examination, color fundus photography, optical coherence tomography angiography, multifocal electroretinogram, and fundus fluorescence angiography, if necessary, at baseline and after 1, 3, 6, and 12 months. BCVA, macular central retinal thickness (CRT), IOP, CNV blood flow area, thickness of the CNV-pigment epithelial detachment complex, thickness of the retinal nerve fiber layer (RNFL), and the first positive peak (P1) amplitude density in ring 1 before and after treatment were compared. Radio propagation environment plays a critical role in the performance of wireless communication systems, and understanding channel characteristics is vital for ensuring reliable communication links and optimizing system performance. Ray tracing is an effective method to investigate propagation characteristics in a complex environment, and how to quickly and accurately obtain environmental information needs to be solved. This paper presents dynamic environment reconstruction and ray tracing simulation in railway tunnel environment based on Simultaneous Localization and Mapping (SLAM) algorithm and Poisson reconstruction algorithm. Accurate channel parameters are obtained and analyzed based on ray tracing simulation. Both straight and curved tunnels are considered and investigated, and the results show the channel characteristics in complex railway tunnel environments.

Background: Tangshen formula (TSF) has an ameliorative effect on hepatic lipid metabolism in non-alcoholic fatty liver disease (NAFLD), but the role played by the gut microbiota in this process is unknown. Method: We conducted three batches of experiments to explore the role played by the gut microbiota: TSF administration, antibiotic treatment, and fecal microbial transplantation. NAFLD mice were induced with a high-fat diet to investigate the ameliorative effects of TSF on NAFLD features and intestinal barrier function. 16S rRNA sequencing and serum untargeted metabolomics were performed to further investigate the modulatory effects of TSF on the gut microbiota and metabolic dysregulation in the body. Results: TSF ameliorated insulin resistance, hypercholesterolemia, lipid metabolism disorders, inflammation, and impairment of intestinal barrier function. 16S rRNA sequencing analysis revealed that TSF regulated the composition of the gut microbiota and increased the abundance of beneficial bacteria. Antibiotic treatment and fecal microbiota transplantation confirmed the importance of the gut microbiota in the treatment of NAFLD with TSF. Subsequently, untargeted metabolomics identified 172 differential metabolites due to the treatment of TSF. Functional predictions suggest that metabolisms of choline, glycerophospholipid, linoleic acid, alpha-linolenic acid, and arachidonic acid are the key metabolic pathways by which TSF ameliorates NAFLD and this may be influenced by the gut microbiota. Conclusion: TSF treats the NAFLD phenotype by remodeling the gut microbiota and improving metabolic profile, suggesting that TSF is a functional gut microbial and metabolic modulator for the treatment of NAFLD.

Abstract Eutrophication usually impacts grassland biodiversity, community composition, and biomass production, but its impact on the stability of these community aspects is unclear. One challenge is that stability has many facets that can be tightly correlated (low dimensionality) or highly disparate (high dimensionality). Using standardized experiments in 55 grassland sites from a globally distributed experiment (NutNet), we quantify the effects of nutrient addition on five facets of stability (temporal invariability, resistance during dry and wet growing seasons, recovery after dry and wet growing seasons), measured on three community aspects (aboveground biomass, community composition, and species richness). Nutrient addition reduces the temporal invariability and resistance of species richness and community composition during dry and wet growing seasons, but does not affect those of biomass. Different stability measures are largely uncorrelated under both ambient and eutrophic conditions, indicating consistently high dimensionality. Harnessing the dimensionality of ecological stability provides insights for predicting grassland responses to global environmental change.

Permanent magnet claw pole machine (PMCPM) is a special kind of transverse flux permanent magnet machine. Compared with other electrical machines, it has the advantages of high torque density and high efficiency for high speed operation. However, because of its complex irregular structure, the manufacturing process using silicon sheets is complicated. Soft magnetic composite material (SMC) is manufactured by powder metallurgy technology, which can produce various shapes of stator core structures, so it is easier to produce various irregular shapes of the stator core. However, the raw SMC material is relatively expensive, and the mechanical strength of SMC is weak. In this paper, a PMCPM with hybrid cores is proposed. With the adoption of hybrid silicon sheet - SMC cores and amorphous alloy - SMC cores, the torque ability of PMCPM can be improved greatly and it can have higher efficiency for more wide operation frequency. Meanwhile, its mechanical strength has been improved and it can be designed for high torque direct drive applications as it is a modular machine. Furthermore, three methods are proposed to reduce the additional eddy current loss which resulted from the employment of hybrid cores in PMCPM.

Abstract C9ORF72 hexanucleotide repeat expansion is the most common genetic cause of both amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). One pathogenic mechanism is the accumulation of toxic dipeptide repeat (DPR) proteins like poly-GA, GP and GR, produced by the noncanonical translation of the expanded RNA repeats. However, how different DPRs are synthesized remains elusive. Here, we use single-molecule imaging techniques to directly measure the translation dynamics of different DPRs. Besides initiation, translation elongation rates vary drastically between different frames, with GP slower than GA and GR the slowest. We directly visualize frameshift events using a two-color single-molecule translation assay. The repeat expansion enhances frameshifting, but the overall frequency is low. There is a higher chance of GR-to-GA shift than in the reversed direction. Finally, the ribosome-associated protein quality control (RQC) factors ZNF598 and Pelota modulate the translation dynamics, and the repeat RNA sequence is important for invoking the RQC pathway. This study reveals that multiple translation steps modulate the final DPR production. Understanding repeat RNA translation is critically important to decipher the DPR-mediated pathogenesis and identify potential therapeutic targets in C9ORF72-ALS/FTD.

Cytokinin (CK) is an important hormone that regulates cell differentiation. The CK content in plants is regulated by cytokinin oxidase (CKX), an important enzyme that participates in hormone-regulated pathways. Additionally, CKXs comprise a large family of enzymes, but little information exists on the CKXs in potato (Solanum tuberosum). In this study, nine CKXs were identified in the potato genome and named StCKX01-09, according to their order on the linkage groups (LGs). They belong to six subfamilies, and the members within the respective subfamilies had similar motifs, a similar gene structure, and similar cis-acting elements. Additionally, the CKXs from four other species, including Arabidopsis, rice (Oryza sativa), soybean (Glycine max), and maize (Zea mays), were also divided into six subfamilies, while members within each subfamily had similar types of motifs. Moreover, the potato StCKXs were shown to influence plant hormones and stress-related factors. StCKXs were collinear, with one CKX in Arabidopsis and five CKXs in Glycine max. Quantitative real-time PCR (qRT-PCR) revealed tissue-specific expression patterns in the potato seedlings and changes in the expression levels in response to stress. Furthermore, the cytokinin content and CKX enzyme activity were shown to be regulated by StCKXs. This study provides detailed information that can help future endeavors in the molecular breeding of potato (Solanum tuberosum).

Abstract RNA translation is tightly controlled in eukaryotic cells to regulate gene expression and maintain proteome homeostasis. RNA binding proteins, translation factors, and cell signaling pathways all modulate the translation process. Defective translation is involved in multiple neurological diseases including amyotrophic lateral sclerosis (ALS). ALS is a progressive neurodegenerative disorder and poses a major public health challenge worldwide. Over the past few years, tremendous advances have been made in the understanding of the genetics and pathogenesis of ALS. Dysfunction of RNA metabolisms, including RNA translation, has been closely associated with ALS. Here, we first introduce the general mechanisms of translational regulation under physiological and stress conditions and review well-known examples of translation defects in neurodegenerative diseases. We then focus on ALS-linked genes and discuss the recent progress on how translation is affected by various mutant genes and the repeat expansion-mediated non-canonical translation in ALS.

Abstract Temperature and biodiversity changes occur in concert, but their joint effects on ecological stability of natural food webs are unknown. Here, we assess these relationships in 19 planktonic food webs. We estimate stability as structural stability (using the volume contraction rate) and temporal stability (using the temporal variation of species abundances). Warmer temperatures were associated with lower structural and temporal stability, while biodiversity had no consistent effects on either stability property. While species richness was associated with lower structural stability and higher temporal stability, Simpson diversity was associated with higher temporal stability. The responses of structural stability were linked to disproportionate contributions from two trophic groups (predators and consumers), while the responses of temporal stability were linked both to synchrony of all species within the food web and distinctive contributions from three trophic groups (predators, consumers, and producers). Our results suggest that, in natural ecosystems, warmer temperatures can erode ecosystem stability, while biodiversity changes may not have consistent effects.

Di-2-ethylhexyl phthalate (DEHP), as a common endocrine disrupting chemicals, can induce toxicity to reproductive system. However, the mechanism remains to be explored. In our study, DEHP exposure induced testicular injury in rats. The high throughput transcriptional sequencing was performed to identify differentially expressed genes (DEGs) between the treatment and control groups. KEGG analysis revealed that DEGs were enriched in apoptosis, PPARα, and ER stress pathway. DEHP up-regulated the expression of PPARα, Bax, Bim, caspase-4. GRP78, PERK, p-PERK, eIF2α, p-eIF2α, ATF4 and CHOP. This view has also been confirmed in TM3 and TM4 cells. In vitro, after pre-treatment with GW6471 (an inhibitor of PPARα) or GSK (an inhibitor of PERK), the apoptosis was inhibited and mitochondrial dysfunction was improved. Moreover, the improvement of mitochondrial dysfunction decreased the expression of PERK pathway by using SS-31(a protective agent for mitochondrial function). Interestingly, ER stress promoted the accumulation of ROS by ERO1L (the downstream of CHOP during ER stress), and the ROS further aggravated the ER stress, thus forming a feedback loop during the apoptosis. In this process, a vicious cycle consisting of PERK, eIF2α, ATF4, CHOP, ERO1L, ROS was involved. Taken together, our results suggested that mitochondrial dysfunction and ER stress-ROS feedback loop caused by PPARα activation played a crucial role in DEHP-induced apoptosis. This work provides insight into the mechanism of DEHP-induced reproductive toxicity.

The insurance hypothesis posits that more diverse communities are more stable through time. Here, the authors show that plant biodiversity reduces the spatial variability of productivity in grassland communities, demonstrating that the insurance hypothesis applies also across space.

Despite being the most widespread blister beetle subfamily, Nemognathinae is unfairly understudied in China. In this study, a new genus and species, Sinostenoria yangi Pan, from northern China is described and illustrated. The antennae, elytra, hind wings, and claws of the new genus form a truly unique set of characteristics never observed in other genera of Nemognathini Laporte de Castelnau, 1840. Three species from China are newly recorded and illustrated: Megatrachelus sibiricus (Tauscher, 1812), Zonitomorpha dollei (Fairmaire 1889), and Stenodera djakonovi Aksentjev, 1978. The genus Oreomeloe Tan, 1981, is transferred from the tribe Meloini Gyllenhal, 1910, to Nemognathini based on an examination of the types. Aiming to test the morphology-based placement of the new genus, we conducted molecular phylogenetic analyses using two mitochondrial (COI, 16S) and three nuclear markers (28S, CAD, ITS2). The results confirm our tribal assignment of the new genus and support a clade that consists of Sinostenoriagen. n., Longizonitis Pan and Bologna, 2018, Stenoria cf. grandiceps, and Ctenopus cf. persicus.

Cr coatings with a thickness of about 19 μm were synthesized on Zr-4 cladding using plasma-enhanced arc ion plating. A Zr-Cr micro-diffusion layer was formed via Cr ion cleaning before deposition to enhance the interface bonding strength. Cr coatings exhibit an obvious columnar crystal structure with an average grain size of 1.26 μm using SEM (scanning electron microscopy) and EBSD (electron backscatter diffraction) with a small amount of nanoscale pores on the surface. A long-term aqueous test at 420 ± 3 °C, 10.3 ± 0.7 MPa and isothermal oxidation tests at 900~1300 °C in air were conducted to evaluate the Cr-coated Zr-4 cladding. All the results showed that the Cr coatings had a significant protective effect to the Zr-4 alloy. However, the corrosion deterioration mechanism is different. A gradual thinning of the Cr coating was observed in a long-term aqueous test, but a cyclic corrosion mechanism of void initiation–propagation–cracking at the oxide film interface is the main corrosion characteristic of the Cr coating in isothermal oxidation. Different corrosion models are constructed to explain the corrosion mechanism.

IntroductionChina’s traditional culture makes rural women and men take on different family responsibilities.MethodsUse “China Family Panel Studies” (CFPS) data and build Logit and propensity score matching models to empirically study the impact of children care and elderly care on rural married women going out to work. And explore the welfare effects of basic education public services in helping rural women take care of their families.ResultsThe results show that caring for children has a significant hindering effect on rural married women’s job hunting. Especially for those in low-income families, the employment inhibition is most significant among women aged 20–30 with multiple children. Contrary to previous cognition, supporting the elderly has a certain weak stimulating effect. The kindergarten public services in rural areas can help women take care of their children and relieve their work pressure. The primary school public services have not played a role in alleviating them.DiscussionThis shows that there are still a large number of female laborers in rural China who are unable to go out to work due to family care. The improvement of rural basic education public services can promote more rural women going out to work. This finding will provide a policy reference for the introduction of a formal care system and the establishment of basic education public services in China.

Positive relationships between biodiversity and temporal stability through species asynchrony are well-documented, but the underlying mechanisms remain debated. Here, the authors show that statistical averaging is the main mechanism of plant diversity effects on community stability.

Introduction Perineural use of dexamethasone is demonstrated to extend the analgesia duration of peripheral nerve blocks (PNB), but its optimal dose remains unclear. This systematic review and meta-analysis aims to determine the optimal dose of perineural dexamethasone in the prolongation of analgesia for PNB.Methods and analysis PubMed, EMBASE, the Cochrane Central Register of Controlled Trials and Web of Science will be searched from their inception to 1 March 2023. Language will be restricted to English. Randomised controlled trials that compared the efficacy and safety of different doses of perineural dexamethasone for PNB in adult patients will be included. Retrospective studies, reviews, meta-analyses, case reports, conference abstracts, comments and studies regarding paediatric surgeries will be excluded. The duration of analgesia will be defined as the primary outcome. Secondary outcomes will include pain scores, the total analgesic requirement over 48 hours and the incidence of adverse effects. Two reviewers will independently perform the study selection, data extraction and quality assessment. RevMan V.5.3 software will be used for data analysis. The quality of evidence will be assessed using the Grading of Recommendation, Assessment, Development and Evaluation (GRADE) approach.Ethics and dissemination No ethical approval is required. The detection and analysis of small molecules, typically defined as molecules under 1000 Da, is of growing interest ranging from the development of small-molecule drugs and inhibitors to the sensing of toxins and biomarkers. However, due to challenges such as their small size and low mass, many biosensing technologies struggle to have the sensitivity and selectivity for the detection of small molecules. Notably, their small size limits the usage of labeled techniques that can change the properties of small-molecule analytes. Furthermore, the capability of real-time detection is highly desired for small-molecule biosensors’ application in diagnostics or screening. This review highlights recent advances in label-free real-time biosensing technologies utilizing different types of transducers to meet the growing demand for small-molecule detection.

Bacterial infections, increasingly resistant to common antibiotics, pose a global health challenge. Traditional diagnostics often depend on slow cell culturing, leading to empirical treatments that accelerate antibiotic resistance. We present a novel large-volume microscopy (LVM) system for rapid, point-of-care bacterial detection. This system, using low magnification (1–2×), visualizes sufficient sample volumes, eliminating the need for culture-based enrichment. Employing deep neural networks, our model demonstrates superior accuracy in detecting uropathogenic Escherichia coli compared to traditional machine learning methods. Future endeavors will focus on enriching our datasets with mixed samples and a broader spectrum of uropathogens, aiming to extend the applicability of our model to clinical samples.

In this paper, a new defective phononic crystal (PC) microbeam model in a thermal environment is developed with the application of modified couple stress theory (MCST). By using Hamilton’s principle, the wave equation and complete boundary conditions of a heated Bernoulli–Euler microbeam are obtained. The band structures of the perfect and defective heated PC microbeams are solved by employing the transfer matrix method and supercell technology. The accuracy of the new model is validated using the finite element model, and the parametric analysis is conducted to examine the influences of size and temperature effects, as well as defect segment length, on the band structures of current microbeams. The results indicate that the size effect induces microstructure hardening, while the increase in temperature has a softening impact, decreasing the band gap frequencies. The inclusion of defect cells leads to the localization of elastic waves. These findings have significant implications for the design of microdevices, including applications in micro-energy harvesters, energy absorbers, and micro-electro-mechanical systems (MEMS).

Two-dimensional (2D) piezoelectric semiconductor materials are garnering significant attention in applications such as intelligent sensing and energy harvesting due to their exceptional physical and chemical properties. Among these, molybdenum disulfide (MoS2), a 2D wide-bandgap semiconductor, exhibits piezoelectricity in odd-layered structures due to the absence of an inversion symmetry center. In this study, we present a straightforward chemical vapor deposition (CVD) technique to synthesize monolayer MoS2 on a Si/SiO2 substrate, achieving a lateral size of approximately 50 µm. Second-harmonic generation (SHG) characterization confirms the non-centrosymmetric crystal structure of the wide-bandgap MoS2, indicative of its piezoelectric properties. We successfully transferred the triangular MoS2 to a polyethylene terephthalate (PET) flexible substrate using a wet-transfer method and developed a wide-bandgap MoS2-based micro-displacement sensor employing maskless lithography and hot evaporation techniques. Our testing revealed a piezoelectric response current of 5.12 nA in the sensor under a strain of 0.003% along the armchair direction of the monolayer MoS2. Furthermore, the sensor exhibited a near-linear relationship between the piezoelectric response current and the strain within a displacement range of 40–100 µm, with a calculated response sensitivity of 1.154 µA/%. This research introduces a novel micro-displacement sensor, offering potential for advanced surface texture sensing in various applications.

Ketones are well-known biomarkers of fat oxidation produced in the liver as a result of lipolysis. These biomarkers include acetoacetic acid and β-hydroxybutyric acid in the blood/urine and acetone in our breath and skin. Monitoring ketone production in the body is essential for people who use caloric intake deficit to reduce body weight or use ketogenic diets for wellness or therapeutic treatments. Current methods to monitor ketones include urine dipsticks, capillary blood monitors, and breath analyzers. However, these existing methods have certain disadvantages that preclude them from being used more widely. In this work, we introduce a novel acetone sensor device that can detect acetone levels in breath and overcome the drawbacks of existing sensing approaches. The critical element of the device is a robust sensor with the capability to measure acetone using a complementary metal oxide semiconductor (CMOS) chip and convenient data analysis from a red, green, and blue deconvolution imaging approach. The acetone sensor device demonstrated sensitivity of detection in the micromolar-concentration range, selectivity for detection of acetone in breath, and a lifetime stability of at least one month. The sensor device utility was probed with real tests on breath samples using an established blood ketone reference method.

Single molecule detection based on evanescent illumination usually require specially designed nanomaterials. Here, the authors show that single molecule detection can be realised on a plain glass surface via interference between the evanescent lights scattered by molecules and the natural roughness of the glass.

PurposeTo investigate the micro-vascular changes in choroidal structures in patients with pre- and early-stage clinical diabetic retinopathy (DR) using wide-field Swept-Source Optical Coherence Tomography Angiography (SS-OCTA).MethodThis observational cross-sectional study included 131 eyes of 68 subjects that were divided into healthy controls (group 1, n = 46), pre-DR (group 2, n = 43), early-stage DR (group 3, n = 42) cohorts. All participants that underwent SS-OCTA examination were inpatients in the department of Ophthalmology and the department of Endocrinology, Qilu Hospital, Shandong University, and Department of Ophthalmology, Aier Eye Hospital, Jinan, from July 11, 2021 to March 17, 2022. The choroidal vascularity index (CVI), choroidal thickness (ChT) and central macular thickness (CMT) in the whole area (diameter of 12 mm) and concentric rings with different ranges (0–3, 3–6, 6–9, and 9–12 mm) were recorded and analyzed from the OCTA image.ResultCompared with healthy eyes, decreases in CVI and ChT were found in the eyes of patients with pre-or early-stage DR. The changes were more significant in the peripheral choroid, with the most prominent abnormalities in the 9-12mm area (P < 0.001). However, there was no obvious difference in the average CMT value. Furthermore, CVI and ChT were significantly correlated with the duration of diabetes in the range of 6–9 and 9–12 mm (Ps < 0.05; Correlation coefficient = -0.549, -0.395, respectively), with the strongest correlation (Ps < 0.01; Correlation coefficient = -0.597, -0.413, respectively) observed at 9–12 mm.ConclusionThe CVI and ChT values of diabetic patients are significantly lower than in healthy controls, especially in patients with early-stage DR. In addition, the peripheral choroidal capillaries are more susceptible to early DM-induced injury than in the central area.

Single-molecule measurements provide statistical distributions of molecular properties, in addition to the ensemble averages. Evanescent detection approaches have been widely used for single-molecule detection because the evanescent field can significantly enhance the light-analyte interaction and reduce the background noise. However, current evanescent single-molecule detection systems mostly require specially designed sensing components. Here, we show that single proteins can be imaged on a plain cover glass surface by detecting the evanescent waves scattered by the target molecules. This allows us to quantify the protein–antibody interactions at the single-molecule level. This protocol describes a label-free single-molecule imaging approach with conventional consumables and may pave the road for detecting single molecules with commercial optical microscopy.

Extending knowledge on ecosystem stability to larger spatial scales is urgently needed because present local-scale studies are generally ineffective in guiding management and conservation decisions of an entire region with diverse plant communities. We investigated stability of plant productivity across spatial scales and hierarchical levels of organization and analyzed impacts of dominant species, species diversity, and climatic factors using a multisite survey of Inner Mongolian grassland. We found that regional stability across distant local communities was related to stability and asynchrony of local communities. Using only dominant instead of all-species dynamics explained regional stability almost equally well. The diversity of all or only dominant species had comparatively weak effects on stability and synchrony, whereas a lower mean and higher variation of precipitation destabilized regional and local communities by reducing population stability and synchronizing species dynamics. We demonstrate that, for semi-arid temperate grassland with highly uneven species abundances, the stability of regional communities is increased by stability and asynchrony of local communities and these are more affected by climate rather than species diversity. Reduced amounts and increased variation of precipitation in the future may compromise the sustainable provision of ecosystem services to human well-being in this region.

To generate and evaluate post-therapeutic optical coherence tomography (OCT) images based on pre-therapeutic images with generative adversarial network (GAN) to predict the short-term response of patients with retinal vein occlusion (RVO) to anti-vascular endothelial growth factor (anti-VEGF) therapy. Real-world imaging data were retrospectively collected from 1 May 2017, to 1 June 2021. A total of 515 pairs of pre-and post-therapeutic OCT images of patients with RVO were included in the training set, while 68 pre-and post-therapeutic OCT images were included in the validation set. A pix2pixHD method was adopted to predict post-therapeutic OCT images in RVO patients after anti-VEGF therapy. The quality and similarity of synthetic OCT images were evaluated by screening and evaluation experiments. We quantitatively and qualitatively assessed the prognostic accuracy of the synthetic post-therapeutic OCT images. The post-therapeutic OCT images generated by the pix2pixHD algorithm were comparable to the actual images in edema resorption response. Retinal specialists found most synthetic images (62/68) difficult to differentiate from the real ones. The mean absolute error (MAE) of the central macular thickness (CMT) between the synthetic and real OCT images was 26.33 ± 15.81 μm. There was no statistical difference in CMT between the synthetic and the real images. In this retrospective study, the application of the pix2pixHD algorithm objectively predicted the short-term response of each patient to anti-VEGF therapy based on OCT images with high accuracy, suggestive of its clinical value, especially for screening patients with relatively poor prognosis and potentially guiding clinical treatment. Importantly, our artificial intelligence-based prediction approach’s non-invasiveness, repeatability, and cost-effectiveness can improve compliance and follow-up management of this patient population.

They demonstrate tunable sensitivity and dynamic range, deep vertical sensing range, also for intracellular signals.

As there is no need of permanent magnet (PM) material and only silicon steel sheet required on the rotor, synchronous reluctance machine (SynRM) can be used for many applications and draws a great research interest. For the SynRM, the torque ripple is a big issue and a great of work could been done on reducing it. In this paper, asymmetrical magnetic flux barriers in the SynRM rotor were studied comprehensively, including angle and width of each layer and each side of the magnetic barrier. The SynRMb with asymmetrical and parallel magnetic flux barrier was found as the best way to design SynRM based on the multi-objective design optimization method. Moreover, each parameter was studied to show the design rule of the asymmetrical magnetic flux barrier. As the average torque will be reduced with the asymmetrical barrier is used, the grain-oriented silicon steel is used on stator teeth of the SynRMb (SynRMbG) was proposed and studied. The analysis results show that the proposed new method can make the SynRM have better performance.

The present work developed an electrochemical genosensor for the detection of virulence outer membrane protein A (ompA, tDNA) gene of Cronobacter sakazakii (C. sakazakii) by exploiting the excellent glucose-oxidase-mimicking activity of copper Metal-organic frameworks (Cu-MOF) doped with gold nanoparticle (AuNPs). The signal nanotags of signal probes (sDNA) that biofunctionalized AuNPs@Cu-MOF (sDNA-AuNPs@Cu-MOF) were designed using an Au-S bond. The biosensor was prepared by immobilization capture probes (cDNA) onto an electrodeposited AuNPs-modified glassy carbon electrode (GCE). AuNPs@Cu-MOF was introduced onto the surface of the GCE via a hybridization reaction between cDNA and tDNA, as well as tDNA and sDNA. Due to the enhanced oxidase-mimicking activity of AuNPs@Cu-MOF to glucose, the biosensor gave a linear range of 1.0 × 10−15 to 1.0 × 10−9 mol L−1 to tDNA with a detection limit (LOD) of 0.42 fmol L−1 under optimized conditions using differential pulse voltammetry measurement (DPV). It can be applied in the direct detection of ompA gene segments in total DNA extracts from C. sakazakii with a broad linear range of 5.4−5.4 × 105 CFU mL−1 and a LOD of 0.35 CFU mL−1. The biosensor showed good selectivity, fabricating reproducibility and storage stability, and can be used for the detection of ompA gene segments in real samples with recovery between 87.5% and 107.3%.

Atomically thin two-dimensional (2D) CN sheets have attracted extensive attention in the field of photocatalysis because of their shorter diffusion path of photogenerated carriers and abundant surface reaction sites than bulk CN. However, 2D CNs still exhibit poor visible-light photocatalytic activity because of a strong quantum size effect. Here, PCN-222/CNs vdWHs were successfully constructed using the electrostatic self-assembly method. The results showed that PCN-222/CNs vdWHs with 1 wt.% PCN-222 enhanced the absorption range of CNs from 420 to 438 nm, which improved the absorption capacity of visible light. Additionally, the hydrogen production rate of 1 wt.% PCN-222/CNs is four times that of the pristine 2D CNs. This study provides a simple and effective strategy for 2D CN-based photocatalysts to promote visible light absorption.

Eutrophication has been shown to weaken diversity-stability relationships in grasslands, but it is unclear whether the effect depends on scale. Analysing a globally distributed network of grassland sites, the authors show a positive role of beta diversity and spatial asynchrony as drivers of stability but find that nitrogen enrichment weakens the diversity-stability relationships at different spatial scales.

Protein identification at the single-molecule level is the ultimate goal for biological research and disease diagnosis. Here, the authors identify the size, charge, mobility, and binding of individual protein molecules by measuring the optical and electrical responses of each protein molecule tethered to a surface.

The performance of traditional flux switching permanent magnet tubular machine (FSPMTM) are improved by using new material and structure in this paper. The existing silicon steel sheet making for all mover cores or part of stator cores are replaced by soft magnetic composite (SMC) cores, and the lamination direction of the silicon steel sheet in stator cores have be changed. The eddy current loss of the machine with hybrid cores will be reduced greatly as the magnetic flux will not pass through the silicon steel sheet vertically. In order to reduce the influence of end effect, the unequal stator width design method is proposed. With the new design, the symmetry of the permanent magnet flux linkage has been improved greatly and the cogging force caused by the end effect has been reduced. Both 2-D and 3-D finite element methods (FEM) are applied for the quantitative analysis.

Canopy structural complexity is a critical emergent forest attribute, and light detection and ranging (lidar)-based fractal dimension has been recognized as its powerful measure at the individual tree level. However, the current lidar-based estimation method is highly sensitive to data characteristics, and its scalability from individual trees to forest stands remains unclear. This study proposed an improved method to estimate fractal dimension from lidar data by considering Shannon entropy, and evaluated its scalability from individual trees to forest stands through mathematical derivations. Moreover, a total of 280 forest stand scenes simulated from the terrestrial lidar data of 115 trees spanning large variability in canopy structural complexity were used to evaluate the robustness of the proposed method and the scalability of fractal dimension. The results show that the proposed method can significantly improve the robustness of lidar-derived fractal dimensions. Both mathematical derivations and experimental analyses demonstrate that the fractal dimension of a forest stand is equal to that of the tree with the largest fractal dimension in it, manifesting its nonscalability from individual trees to forest stands. The nonscalability of fractal dimension reveals its limited capability in canopy structural complexity quantification and indicates that the power-law scaling theory of a forest stand underlying fractal geometry is determined by its dominant tree instead of the entire community. Nevertheless, we believe that fractal dimension is still a useful indicator of canopy structural complexity at the individual tree level and might be used along with other stand-level indexes to reflect the “tree-to-stand” correlation of canopy structural complexity.

In recent years, the earthen ruins’ cultural relics have aroused people’s extensive attention, and the protection of them is more and more urgent. The aim of the paper is to present the essential results of an ongoing research on a reinforced rammed earthen wall in the Suoyang ancient city (Guazhou, China). A variety of methods to combining aerial survey, dynamic test, and numerical model is provided with the purpose of estimating the structural characteristics of the ancient rammed wall. An aerial geometric survey based on high-resolution images made by unmanned aerial vehicle (UAV) has been approached to acquiring a 3D model. An ambient vibration test has been adopted by using vibration pick-ups. The experimental data were dealt with the operational modal analysis (OMA) followed by comparing with the numerical results delivered by the model. Meanwhile, the results showed the maximum displacements and corresponding positions of the structure under the three modes by the finite element model (FEM).

The high-sulfated derivative of Ulva pertusa polysaccharide (HU), with unclear structure, has better anti-hyperlipidmia activity than U pertusa polysaccharide ulvan (U). In this study, we explore the main structure of HU and its therapeutic effect against nonalcoholic fatty liver disease (NAFLD). The main structure of HU was elucidated using FT-IR and NMR (13C, 1H, COSY, HSQC, HMBC). The anti-NAFLD activity of HU was explored using the high-fat diet mouse model to detect indicators of blood lipid and liver function and observe the pathologic changes in epididymal fat and the liver. Results showed that HU had these main structural fragments: →4)-β-D-Glcp(1→4)-α-L-Rhap2,3S(1→; →4)-α-L-Rhap3S(1→4)-β-D-Xylp2,3S(1→; →4)-α-L-Rhap3S(1→4)-β-D-Xylp(1→; →4)-α-L-IdopA3S(1→4)-α-L-Rhap3S(1→; →4)-β-D-GlcpA(1→3)-α-L-Rhap(1→; →4)-α-L-IdopA3S(1→4)-β-D-Glcp3Me(1→; →4)-β-D-Xylp2,3S(1→4)-α-L-IdopA3S(1→; and →4)-β-D-Xylp(1→4)-α-L-IdopA3S(1→. Treatment results indicated that HU markedly decreased levels of TC, LDL-C, TG, and AST. Furthermore, lipid droplets in the liver were reduced, and the abnormal enlargement of epididymal fat cells was suppressed. Thus, HU appears to have a protective effect on the development of NAFLD.

The low temperature solid deposition characteristics of cold spraying technology are conducive to the remanufacturing/repair of oxidation sensitive titanium alloy in the atmosphere. Ti6Al4V alloy deposits prepared by cold spray (CS) usually show porous microstructure due to insufficient plastic deformation of Ti6Al4V alloy particles. The interfacial bonding and the microstructural evolution mechanisms of Ti6Al4V particles during cold spray are still unclear. In this study, the Ti6Al4V deposits were prepared by cold spray with nitrogen as process gas at a temperature of 800 °C and a pressure of 4 MPa. The micron CT results show that the porosity is 8.25%. The deformation ultrafine grain zone and un-deformation martensitic laths zone at the bonding interface between Ti6Al4V particles were observed. The EBSD results show that the grain orientation signal was only detected at the edge of the particles deformation area and the 23% of grain sizes detected by EBSD are less than 500 nm, which indicates that the nanocrystallization induced by dynamic recrystallization occurs at the inter-particle boundaries between cold-sprayed Ti6Al4V particles. The TEM and SADPs demonstrate a gradient structure consisting of equiaxed nanograins zone, crushing dendrites transition zone and original martensitic laths zone from the interface to the core of particles.

Prussian blue is an iron-cyanide-based pigment steadily becoming a widely used electrochemical sensor in detecting hydrogen peroxide at low concentration levels. Prussian blue nanoparticles (PBNPs) have been extensively studied using traditional ensemble methods, which only provide averaged information. Investigating PBNPs at a single entity level is paramount for correlating the electrochemical activities to particle structures and will shed light on the major factors governing the catalyst activity of these nanoparticles. Here we report on using plasmonic electrochemical microscopy (PEM) to study the electrochemistry of PBNPs at the individual nanoparticle level. First, two types of PBNPs were synthesized; type I synthesized with double precursors method and type II synthesized with polyvinylpyrrolidone (PVP) assisted single precursor method. Second, both PBNPs types were compared on their electrochemical reduction to form Prussian white, and the effect from the different particle structures was investigated. Type I PBNPs provided better PEM sensitivity and were used to study the catalytic reduction of hydrogen peroxide. Progressively decreasing plasmonic signals with respect to increasing hydrogen peroxide concentration were observed, demonstrating the capability of sensing hydrogen peroxide at a single nanoparticle level utilizing this optical imaging technique.

Abstract To date, the decadal variability of the Global Monsoon (GM) has been mainly studied using instrumental data collected over the last 50 years, and further discussion has been hampered by the limited data length. Here, we present a coral δ18O record from the northern South China Sea, where the hydrology is related to the seasonal Intertropical Convergence Zone (ITCZ) migration, and we use this record to reconstruct the decadal migration of the ITCZ since 1851 A.D. Combining our record with a synthesis of monsoon records reveals an anti‐phase inter‐hemispheric variability of the GM over the last 150 years, indicating that the decadal‐scale hydrologic variability in the tropics is modulated by the meridional migration of the ITCZ. The plausible relationship observed between the decadal variability of the GM and the Atlantic Multidecadal Oscillation suggests that the decadal variability of tropical hydrological changes is likely linked to the climate perturbations in the North Atlantic.

Urban heat island (UHI) draws more attention as it affects not only the health of residents but also the energy consumption of buildings at the city scale. To achieve carbon neutrality goals, it is crucial to better understand the mechanism of the UHI influences on building energy consumption. The characteristics of urban heat island intensity (UHII) and the relationship between the UHII effect and building electricity and related coal consumption were analyzed, based on the long period of monitoring data with hourly weather data from 1 January to 31 December 2019. Results show that a strong correlation between the annual mean UHII and the median daily mean UHII exists. The synthetic diurnal UHII of most cities presents a U-shaped variation trend. In different building climate zones in China, namely, severe cold region (SCR), cold region (CR), hot summer cold winter region (HSCWR), hot summer and warm winter region (HSWWR), and mild region (MR), the influences of UHII on building energy consumption were analyzed. The existence of UHI reduces building energy consumption in 96.7% of SCR cities and 60.8% of CR cities, while in HSCWR, HSWWR, and MR cities, the percentage of cities where the building energy consumption is increased by UHI is 69.4%, 80%, and 63.6%, respectively. Urban climate strongly influences building energy consumption, indicating that it should be considered and analyzed in detail for making future urban development or carbon emission reduction strategies.

Phage display technology has been a powerful tool in peptide drug development. However, the supremacy of phage display-based peptide drug discovery is plagued by the follow-up process of peptides synthesis, which is costly and time consuming, but is necessary for the accurate measurement of binding kinetics in order to properly triage the best peptide leads during the affinity maturation stages. A sensitive technology is needed for directly measuring the binding kinetics of peptides on phages to reduce the time and cost of the entire process. Here, we show the capability of a charge-sensitive optical detection (CSOD) method for the direct quantification of binding kinetics of phage-displayed peptides to their target protein, using whole phages. We anticipate CSOD will contribute to streamline the process of phage display-based drug discovery.

This paper proposes a new rotary flux switching transverse flux machine with the ability of linear motion (FSTFMaLM), in which both the stator and the rotor cores are made by using soft magnetic composite (SMC) materials. With the special design pattern, for the rotary motion model, the proposed machine can combine both the advantages of the flux switching permanent magnet machine (FSPMM) and the transverse flux machine (TFM). It can output with relatively high torque density, and as there is no windings or the magnets on the rotor cores, the proposed machine can operate in the high speed region to improve the output power. With the adoption of the SMC materials, the manufacturing of this machine can be quite easy. By stacking the rotor core together and prolong it with the determined length in the axial direction, in addition with the special control algorithm, the proposed machine can have the ability of the linear motion. In this paper, the operation principle of this machine has been explained and the design methods are also presented. To seek the better performance, the main dimension of the machine is optimized, and for the performance evaluation, the finite element method (FEM) is adopted. The proposed machine can be used for the electric driving systems, robotic systems or other applications where the linear motion ability is required.
