17. Optimizing the low-temperature performance of high-wax-content crude oil with emulsion-based wax inhibitors: Mechanisms and efficacy.

- Abstract
Wax deposition in constant-temperature transportation of waxy oil with high pour points poses a significant challenge for the oil industry. The demand for efficient methods to solve wax deposition has gained attention. To elucidate the impact of emulsion-based wax inhibitors on the performance of crude oils with varying wax content at low temperatures, experiments, rheological analyses, and microscopic analyses were conducted to study their pour point regulation, low-temperature flow improvement, wax prevention effectiveness, and wax crystallization behavior. The results indicate that emulsion-based wax inhibitors significantly reduce the pour point of crude oil, especially for high-wax-content oil, while for low-wax-content oil, the pour point only decreased by 3°C. By penetrating and dispersing wax crystals, the inhibitor reduced the viscosity of crude oil at low temperatures, enhancing its flowability. At 28°C, crude oil transitioned from a shear-thinning non-Newtonian fluid to a Newtonian fluid at wax inhibitor dosages of 500-750 ppm. At 1000 ppm, the wax prevention rate for high-wax-content oil reached 87.5%, but the effect plateaued beyond this concentration. Additionally, the wax prevention and removal mechanism of the emulsion-based wax inhibitors is discussed. This study confirms that emulsion-based wax inhibitors significantly enhance the low-temperature processability of crude oil, offering a viable strategy for the conveyance and refinement in cold climates., (© 2024 The New York Academy of Sciences.)

18. Sex differences in the molecular profile of adult diffuse glioma are shaped by IDH status and tumor microenvironment.

- Abstract
Background: Sex differences in adult diffuse glioma (ADG) are well-established clinically, yet the underlying molecular mechanisms remain inadequately understood. Here, we aim to reveal molecular features and cellular compositions unique to each sex in ADG to comprehend the role of sex in disease etiology., Methods: We quantified sex differences in transcriptome of ADG using multiple independent glioma patient datasets. Next, we delved into the single-cell landscape to examine sex differences in gene expression and cellular composition. To explore how sex influences disease progression, we analyzed paired samples from primary and recurrent ADG cases, aiming to identify sex-specific differences in molecular and cellular features., Results: Our analysis revealed that mutations in isocitrate dehydrogenase (IDH) genes and the tumor microenvironment emerged as primary influencers of sex-differential molecular enrichments. In IDHwt tumors, genes in neuronal signaling pathway are found to be enriched in male tumors, while genes in hypoxia and inflammatory response pathways are enriched in female tumors. This pattern was reversed in IDHmut gliomas. We hypothesized that these distinctions could be attributed to heterogeneous cellular composition between sexes. Using single-cell data, we observed distinctive patterns of sex differences in cell states, cell composition and cell-cell interaction in IDHwt and IDHmut tumors separately. Further, by comparing molecular changes in paired primary and recurrent ADG samples, we identified sex-specific differences in molecular characteristics and cellular compositions of recurrent tumors., Conclusion: Our results provide a comprehensive multi-level characterization of sex differences in ADG, such findings provide novel insights into glioma disease progression in each sex., (© The Author(s) 2024. 19. The Influencing Factors and Mechanism of Anionic and Zwitterionic Surfactant on Viscosity Reduction in Heavy O/W Emulsions.

- Abstract
The high viscosity of heavy crude oil has been an obstacle to its safe production and economic transportation. In this work, a screened emulsified viscosity reducer system is conducted. Experimental results demonstrate that the most effective viscosity reducing agent comprises sodium oleate (NaOl) and cocamidopropyl betaine (CAB-35) in a ratio of 1:2, achieving a viscosity reduction rate of 94.65%. Additionally, the interfacial tension between oil and water decreases from 27 to 4 mN/m with 0.1 mass % TEOA and NaOH in a 1:1 ratio. The oil droplet size is uniformly distributed with D
mean is 14 μm and D50 is 11 μm. Droplets flocculate as the salinity increases to 0.2 mol/L, which corresponds to the apparent increase of viscosity. The adsorption of long alkyl chain lipophilic groups on surfactant molecules at the oil-water interface and the water film alters the wettability of pipe steel to water-wet, further enhancing the application of emulsification and viscosity reduction effects. The primary mechanism behind the viscosity reduction in emulsification is attributed to strong electrostatic interactions stemming from molecular electrostatic potential distributions., Competing Interests: The authors declare no competing financial interest., (© 2024 The Authors. 20. A Web Application for Predicting Drug Combination Efficacy Using Monotherapy Data and IDACombo.

- Abstract
We recently reported a computational method (IDACombo) designed to predict the efficacy of cancer drug combinations using monotherapy response data and the assumptions of independent drug action. Given the strong agreement between IDACombo predictions and measured drug combination efficacy in vitro and in clinical trials, we believe IDACombo can be of immediate use to researchers who are working to develop novel drug combinations. While we previously released our method as an R package, we have now created an R Shiny application to allow researchers without programming experience to easily utilize this method. The app provides a graphical interface which enables users to easily generate efficacy predictions with IDACombo using provided data from several high-throughput cell line screens or using custom, user-provided data.

21. Simplicity: web-based visualization and analysis of high-throughput cancer cell line screens.

- Abstract
High-throughput drug screens are a powerful tool for cancer drug development. However, the results of such screens are often made available only as raw data, which is intractable for researchers without informatic skills, or as highly processed summary statistics, which can lack essential information for translating screening results into clinically meaningful discoveries. To improve the usability of these datasets, we developed Simplicity, a robust and user-friendly web interface for visualizing, exploring, and summarizing raw and processed data from high-throughput drug screens. Importantly, Simplicity allows for easy recalculation of summary statistics at user-defined drug concentrations. This allows Simplicity's outputs to be used with methods that rely on statistics being calculated at clinically relevant doses. Simplicity can be freely accessed at https://oncotherapyinformatics.org/simplicity/.
