Alterations in lipid and lipoprotein metabolism are factors that trigger several negative metabolic complications. Hyperlipidemia is the starting point for the development of comorbidities of the cardiovascular system, such as atherosclerosis. The search for compounds that reduce high levels of total cholesterol and triglycerides has been widely reported in several publications in the literature. Phthalimide derivatives have been extensively researched with various biological actions. In this study we evaluated the antihyperlipidemic ability of three phthalimide derivatives (FGT-2, FGT-3 and FGT-4) on a model of obesity and insulin resistance in mice. The animals were submitted to a hyperlipid diet for 60 days. On the thirtieth day they were treated with phthalimides (20 mg/kg). The positive control group was treated with Simvastatin (20 mg/kg) and the negative control received only the carboxymethylcellulose vehicle. Biochemical and histological analyzes of all groups were analyzed. The animals treated with phthalimidic derivatives had a reduction in total cholesterol, low density and very low density lipoproteins (LDL-c and VLDL-c), triglycerides and fasting glycemia when compared to the negative control group. The treated animals also showed good results when analyzing the atherogenic indexes Castelli i and II and the ratio Triglycerides/HDL-c. In the oral glucose tolerance test and in the insulin tolerance test, animals treated with phthalimides were more sensitive to the action of the hormone regulating carbohydrate uptake. In the evaluation of the transaminases (AST/ALT), the animals of the group treated with phthalimides presented a lower elevation than the other groups of the experiment, the same observed with the uric acid evaluation. Histological analyzes were performed on liver, kidney, heart and pancreas samples. The groups treated with the compounds FGT-2 and FGT3 presented discrete alterations in the liver and kidney. FGT-4 did not present histological alterations for both tissues and the three phthalimide derivatives did not cause alterations in the other organs. These results suggest that the phthalimides tested can act as antihyperlipidemic agents and have a pleiotropic action, by acting also reducing glycemia in insulin resistance model mimicking diabetes mellitus type 2. These compounds may appear as a new approach in the treatment of obesity and complications, which are multifaceted.

Introdução: As parasitoses são causas relevantes de agravo à saúde em países subdesenvolvidos e em desenvolvimento, sendo os parasitas intestinais os principais fatores debilitantes da população, associando-se, frequentemente, a quadros de diarreia crônica e desnutrição. Na transmissão dos parasitas intestinais, destacam-se o solo, o ar, a água, as moscas, as mãos e os alimentos (incluindo as verduras e hortaliças) como os principais elementos do ciclo da cadeia epidemiológica. Objetivo: Estudar a ocorrência de contaminação em hortaliças comercializadas em feira livre na Região Metropolitana do Recife, por formas infectantes de parasitas intestinais humanos. Material e métodos: Para o estudo foram selecionadas a alface lisa (Lactuva sativa L.) e o coentro (Coriandrum sativum L.), cinco amostras de cada, cultivadas em hortas comerciais e/ou de uso coletivo e comercializadas em uma feira livre da Região Metropolitana do Recife, localizada no bairro do Cordeiro, no ano de 2019. Foi estabelecido como unidade amostral para a alface o pé e para o coentro o maço, sendo a pesquisa de parasitas realizada nas partes comestíveis (talos e folhas). Logo após a obtenção das amostras, estas foram armazenadas sob refrigeração por, 48 horas. Em seguida, foram realizados os procedimentos laboratoriais, tais como, lavagem das hortaliças, filtração e sedimentação das amostras (método da sedimentação espontânea) e observação em microscópio, no Laboratório de Parasitologia – UNICAP. Resultados: Protozoários e helmintos foram observados nas hortaliças estudadas, sendo verificada a presença de um total de 8 (oito) diferentes tipos de organismos, distribuídos entre Balantidium coli (trofozoíto), Entamoeba coli (cisto), Endolimax nana (cisto), Ascaris lumbricoides (ovo), Enterobius vermicularis (ovo) e Ancylostoma duodenale (ovo e larva), além dos protozoários de vida livre, Paramecium sp. e Vorticella sp. Conclusão: Conclui-se que, as hortaliças podem ser importantes reservatórios de parasitas patogênicos e não patogênicos, tornando-se veículos de contaminação parasitológica para o homem e representando um relevante problema para a saúde pública.

Introdução: Diante do cenário emergencial atual, com a pandemia do COVID-19 e o distanciamento social, como medida preventiva, Instituições de Ensino Superior (IES) interromperam o aprendizado presencial oferecido aos estudantes e adquiriram o Regime Letivo Remoto (RLR). Logo, foi implementado um novo modelo de ensino para melhorar essa forma de aprendizado, através de metodologias ativas de ensino, tal modelo foi adotado na disciplina de Parasitologia, iniciando com o conteúdo sobre o Ascaris lumbricoides. Objetivo: Analisar novos métodos pedagógicos de ensino para a disciplina de Parasitologia, na pandemia do Covid-19, com enfoque na metodologia ativa de ensino. Materiais e métodos: Durante o semestre de 2020.2, as aulas da disciplina de Parasitologia foram reformuladas. Inicialmente, a docente responsável pela disciplina, começava com uma aula teórica de 50 minutos sobre o conteúdo do dia (o foco desse trabalho foi para o A. lumbricoides), em seguida, era aplicada a clínica interativa, destacando o diagnóstico clínico e laboratorial da ascaridíase. Para finalização da aula era aplicada uma atividade na forma de jogo, elaborada pelos monitores, sob a supervisão da docente sobre o parasita. O jogo foi elaborado através da plataforma Google Meet. A turma de Medicina do 3 período da UNICAP, foi dividida em 4 grupos de 13 alunos. A atividade foi intitulada de “MemoriÁscaris” e se baseou no jogo da memória onde os alunos deveriam seguir 3 etapas: selecionar duas imagens iguais, analisá-las e responder uma pergunta sobre o tema. Ao todo foram selecionadas 10 perguntas que abordavam a problematização da ascaridíase, aplicação na vida real, diagnóstico clínico e laboratorial, ciclo do parasita e medidas profiláticas efetivas. Foram utilizadas imagens do atlas virtual de Parasitologia sendo excluídas aquelas que não apresentavam boa resolução ou relevância para o assunto. Resultados: Todos os alunos da referida turma participaram da atividade, obtendo 100% de acerto nas questões presentes na avaliação somativa sobre o assunto e externaram a importância de metodologias ativas no êxito de seus aprendizados. Conclusão: A metodologia ativa aplicada na disciplina de Parasitologia, exposição oral, clínica interativa e atividade na forma de jogo “MemoriÁscaris” mostrouse eficaz no aprendizado dos alunos, além de ter estimulado uma participação ativa na construção do conhecimento.

Phthalimido-alkyl-1H-1,2,3-triazole derivatives 3a-d and 4a-d were efficiently synthesized using 1,3-dipolar cycloaddition reaction. Anti-inflammatory activity and toxicity studies were performed. The results demonstrated that all the tested compounds reduced carrageenan-induced paw edema and indicated no lethality for toxicity against Artemia salina and acute toxicity in vivo (LD50 up to 1 g kg -1). Furthermore, the structure of phthalimide linked to phenyl group proved to be more active than the compounds containing benzothiazole moiety. Structural modifications such as removal of the phthalimide group and subsequent acetylation, to exemplify a non-cyclic amide, demonstrate that the phthalimide and triazole moieties are important for design of potent candidates with anti-inflammatory drug proprieties. Docking into the cyclooxygenase-2 (COX-2) confirms the importance of the phthalimide and triazole groups in the anti-inflammatory activity. The histopathological studies showed that the compounds 3a-d and 4a-d did not cause serious pathological lesions liver or kidneys.

Phthalimide and some N-substituted cyclic imides may be potent hypolipidemic agents, capable of reducing plasma cholesterol, and triglyceride levels. In this work, N-phenylphthalimide (5a), N-(2-nitrophenyl)phthalimide (5b), N-(3-nitrophenyl)phthalimide (5c), N-(4-nitrophenyl)phthalimide (5d), N-(4-bromophenyl)phthalimide (5e), N-(4-amidaphenyl)phthalimide (5f), N-(4-methylthiophenyl)phthalimide (5g), and N-(4-chlorophenyl)phthalimide (5h) were synthesized, and their effects on reducing lipid levels and as acute antiinflammatory agents in 3-month-old male Swiss mice were compared with phthalimide. To access their safety, the drugs were administered to determine the acute toxicity (LD50 value). In addition, the acute anti-inflammatory activity was evaluated via carrageenan-induced edema; and the effective dose was determined for compounds that showed the best anti-inflammatory activity in comparison with ibuprofen. The compound (5g) proved to be a more active hypolipidemic compound than phthalimide, at a dose level of 20 mg kg−1 day−1. The derivative (5g) reduced the plasmatic triglycerides and cholesterol levels by 54 and 41 %, respectively, while the percentages of reduction were 43 and 30 %, respectively, with phthalimide. The hypocholesterolemic and hypotriglyceridemic activities of the (5g) derivative were similar to that obtained with the commercially available drug, pravastatin. The anti-inflammatory activity of compound (5b) was similar to ibuprofen and aspirin at 250 mg kg−1. Taken together, it was demonstrated that the new phthalimide derivatives were able to increase HDL-cholesterol levels, along with reducing the cholesterol and triglyceride levels and carrageenan-induced edema in mice. Therefore, they may offer promise in the future as new hypolipidemic and anti-inflammatory agents.

In this work, the preparation of various glucosyl triazoles from a reaction between 2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl azide and terminal alkynes was developed in moderate to excellent yields (63-99%). Ultrasound energy was applied at each step of the reaction to increase chemical reactivity. In addition, the compounds conjugated with benzoheterocycles moieties revealed potent anti-inflammatory activity.
