Über die Entwicklung von Sehnen- und Bändern ist wenig bekannt. Untersuchungen ergaben, dass es bei Überexpression von Smad8ca und BMP2 in der mesenchymalen Vorläuferzelllinie C3H10T1/2 zu einer Differenzierung in Sehnenzellen kam. Zur Untersuchung der Differenzierung modifizierter muriner Stammzellen in vivo wurde ein ektopes Mausmodell zur Sehnenbildung entwickelt. Es zeigte sich, dass BMP2 und Smad8ca die Tendogenese ermöglichen. Es wurde auch das Differenzierungsverhalten von viral-modifizierten C3H10T1/2-Zellen untersucht. Ektope Implantationen mit adenoviral Smad8ca und BMP2 veränderten C3H10T1/2-Zellen zeigten, dass in vivo eine Differenzierung der Zellen sowohl in sehnen-ähnliches als auch in Knochengewebe stattfand. Des Weiteren kam es zur Generierung eines apophysären Knochen-Sehnen-Übergangs. Im Gegensatz dazu bildete sich in C3H10T1/2-Zellen die nur Smad8ca exprimierten, sehnen-ähnliches Gewebe ohne Knochenelemente. Dies zeigt, dass eine Smad8ca-abhängige Signalvermittlung für eine Sehnenzellbildung ausreichend ist. Es wurden auch lentiviral-infizierte humane MSCs ektopisch in Mausgewebe implantiert. Auch hier konnte eine Smad8ca/BMP2-abhängige Differenzierung in Knochen- und sehnen-ähnliches Gewebe beobachtet werden. Mit Hilfe eines Microarrays wurde nach sekretierten Faktoren gesucht. Es zeigte sich, dass das Gen Periostin im Smad8ca/BMP2-Hintergrund hoch reguliert wurde. Bei Implantation von Periostin- und caBMPR-IA-modifizierten murinen als auch humanen MSCs konnte beobachtet werden, dass in beiden Fällen eine stammzellabhängige Differenzierung in sehnen-ähnliches Gewebe stattfand. Bei den modifizierten huMSCs wurde die Bildung des neuen Gewebes allerdings außerhalb der implantierten Trägermatrix beobachtet. Es wird vermutet, dass die huMSCs Periostin in das umliegende Mausgewebe sekretieren und dieses dort die Organisation der extrazellulären Matrix in sehnen-ähnliches Gewebe einleiten kann., Little is known about the development of tendons and ligaments. This means that the presence of Smad8ca is sufficient for the differentiation into tenocytes. Lentiviral-modified human mesenchymal stem cells (huMSCs) were implanted in mice as well. As before, a Smad8ca/BMP2-depending differentiation into bone- and tendon-like tissue could be observed. By the use of microarray analysis it was searched for secreted factors. It appeared that the gene Periostin was up-regulated in the presence of Smad8ca/BMP2. The implantation of modified murine and human MSCs with Periostin and caBMPR-IA resulted in a stem cell-depending differentiation into tendon-like tissue. In the case of modified huMSCs, however, the neotissue was located outside the implanted sponge. It is supposed that Periostin is secreted into the surrounding mouse tissue by huMSCs where it induces the organisation of extracellular matrix into tendon-like tissue.
