Gryllotalpa gryllotalpa (Orthoptera: Gryllotalpidae)’nın bakteriyel mikrobiyotası, araştırılmıştır ve G. gryllotalpa’nın nimflerinden 10 farklı bakteriyel aerobik tür izole edilmiştir. G. gryllotalpa en önemli tarım zararlılarından biridir. Bu çalışmada, bakterilerin morfolojik, fizyolojik ve biyokimyasal özelliklerini ve API 20E ve API 50CH panel test sistemleri sayesinde metabolik enzim profillerini belirledik. Ayrıca, izolatların moleküler seviyede belirlenmesi için 16S rRNA geni dizi analizi ve Hiper değişken (HV) analizleri uygulanmıştır. Bakteriyel türler, Bacillus weihenstephanensis (Gg1), Serratia ureilytica (Gg2), Bacillus simplex (Gg3), Bacillus nanhaiensis (Gg5), Bacillus sp. (Gg6), Bacillus gibsonii (Gg7), Citrobacter braakii (Gg8), Paenibacillus sp. (Gg9), Enterobacter sp. (Gg10), and Providencia rettgeri (Gg11) olarak belirlenmiştir. Bütün bakteriyel türler, on gün boyunca 3.6×109 hücre/ml dozda G. gryllotalpa’nın nimflerine karşı test edilmiştir. Bu sonuçlar, özellikle B. weihenstephanensis (Gg1) ve Bacillus nanhaiensis (Gg5) bakterilerinin, G. gryllotalpa’nın mikrobiyal mücadelesinde değerli bir etken olabileceğini göstermiştir., The bacterial microbiota of the Gryllotalpa gryllotalpa (Orthoptera: Gryllotalpidae) was investigated and 10 different aerobic bacterial strains were isolated from the nymphs of G. gryllotalpa. It is one of the most important agricultural pests. We determined the morphological, physiological, and biochemical properties of the bacteria and metabolic enzyme profiles by API 20E and API 50CH panel test systems. Additionally 16S rRNA gene sequence and Hyper variable (HV) analyses were performed to determine the strains at the molecular level. They were identified as Bacillus weihenstephanensis (Gg1), Serratia ureilytica (Gg2), Bacillus simplex (Gg3), Bacillus nanhaiensis (Gg5), Bacillus sp. (Gg6), Bacillus gibsonii (Gg7), Citrobacter braakii (Gg8), Paenibacillus sp. (Gg9), Enterobacter sp. (Gg10), and Providencia rettgeri (Gg11), according to results. All these bacterial strains were tested against the nymphs of G. gryllotalpa at 3.6×109 cfu/ml dose, within ten days. Our results indicate that especially B. weihenstephanensis (Gg1) and Bacillus nanhaiensis (Gg5) may be valuable as a microbial control agent for G. gryllotalpa.

Objectives: Lipolytic genes have been investigated in several viral genomes, and some of them show enzyme activity which can be used for various functions including the production of DNA replication metabolites, rescue from endosomes, and membrane fusion. Amsacta moorei entomopoxvirus (AMEV) replicates in nearly the entire insect body, especially in the adipose tissue. One of the open reading frames (ORFs) in the AMEV genome, amv133, encodes a putative lipase enzyme. In this study, we therefore investigate the enzyme activity of amv133. Methods: amv133 was aligned with known lipase genes and their homologs in entomopoxviruses. Expressed proteins were partially purified and assayed for lipase, esterase and protease. Results: We found that amv133 contains all the domains required for a functional lipase enzyme and that it shows a significant similarity with homologs in other entomopoxviruses. Since there is a similarity of the catalytic triad between lipases and serine proteases, we also investigated the protease activity of amv133. Lipase, esterase and protease assays showed that amv133 encodes a functional esterase enzyme with protease activity. Conclusion: The current data show that amv133 is a conserved gene in all entomopoxvirus genomes sequenced so far and might contribute greatly to degrading the lipids or proteins and hence improve the virus infection. ÖZET Kızılağaç yaprak böceği, Agelastica alni (L.) (Coleoptera: Chrysomelidae), gün dönümü böceği, Amphimallon solstitiale (L.) (Coleoptera: Scarabaeidae), fındık yaprak deleni, Anoplus roboris (Sufr.) (Coleoptera: Curculionidae), fındık kurdu, Balaninus nucum (L.) (Coleoptera: Curculionidae) ve adi mayıs böceği, Melolontha melolontha (L.) (Coleoptera: Scarabaeidae), Coleoptera takımına ait önemli fındık zararlılarıdır. Şu ana kadar bu zararlılar ile mücadele büyük oranda kimyasal ilaçlar ile yapılmaktadır. Kimyasal ilaçların yan etkilerinin keşfedilmesi, bilim adamlarım daha etkili ve daha güvenli bir mücadele yöntemi geliştirmeye sevk etmiştir. Bu çalışmada, Coleoptera takımına ait olan A. alni, A. solstitiale, A. roboris, B. nucum ve M. melolontha' 'da patojenik virüslerin tespiti, izolasyonu, karakterizasyonu, histopatolojisi, infeksiyon özelliği, konak hassasiyeti ve horizontal enfeksiyonu araştırıldı. Adi mayıs böceği larvalarında bir viral enfeksiyon belirlendi. Yapılan ışık mikroskobu, elektron mikroskobu ve moleküler çalışmalar sonucunda tespit edilen virüsün entomopoxviridae familyasından Melolontha melolontha entomopoxvirüsün yeni bir izolatı olduğu belirlendi (MmEPV-TR). Işık ve elektron mikroskobu sonuçlarına göre, virüse ait sferoidlerin uzunluğu 5,45+2,34 um, genişliği ise 2,89±1,13 um olarak tespit edildi. Mikroskobik çalışmalara ilave olarak virüsün varlığı ayrıca fusolin geni için özel prob kullanarak DNA-DNA hibridizasyon yöntemi ile moleküler olarak da tespit edildi. Yapılan bioassay çalışmaları MmEPV'nin ikinci-üçüncü evre M. melolontha larvaları üzerinde çok etkili olduğunu gösterdi. En yüksek insektisidal etki 7,5x1 06 sferoid/ml konsantrasyonla % 93,3 olarak belirlendi. Yapılan çalışmalar, dünyada ikinci Türkiye için ilk ve tek izolat olan M. melolontha entomopoxvirüsün, M. melolontha larvaları ile biyolojik mücadelede etkili bir ajan olarak kullanılabileceği sonucunu ortaya koymuştur. (Coleoptera: Chrysomelidae), summer chafer, Amphimallon solstitiale (L.) (Coleoptera: Scarabeidae), hazelnut leave holer, Anoplus roboris (Sufr.) (Coleoptera: Curculionidae), hazelnut weevil, Balaninus nucum (L.) (Coleoptera: Curculionidae) and common cockchafer, Melolontha melolontha (L.) (Coleoptera: Scarabaeidae) are very important hazelnut pests belong to Coleoptera order. Up to now, the chemical pesticides have been mostly utilized to control these pests. However, recent concern on the hazardous effects of chemical pesticides made scientists consider findining more effective and safer control agents. In this study, the isolation, characterization, histopathology, infection, host spectrum and horizontal infection of pathogenic viruses were investigated on the coleopteran hazelnut pests, A. alni, A. solstitiale, A. roboris, B. nucum and M. melolontha. The viral infection was observed only in the larvae of common cockchafer. Morphological, microscopical and molecular studies of the virus showed that the virus is an entomopoxvirus called MmEPV-TR. This study is the first record of M melolontha entomopoxvirus in Turkey. It is also the second isolate in all over the world. Based on the results of light and electron microscopy studies, the lenght of spheroids was 5,45+2,34 um, the width of spheroids was 2,89±1,13 urn. The presence of virus was also demonstrated by DNA-DNA hybridization assay using fusolin gene specific probe of Choristoneura fumiferana (Lepidoptera: Tortricidae) entomopoxvirus. The bioassays carried out to determine the infectivity of the MmEPV showed that it is very effective on the second-third instar larvae of M. melolontha. The highest insecticidal activity determined on M melolontha larvae was 93,3 % with 7,5x1 06 spheroids/ml dosage. Our results showed that Turkish isolate of M. melolontha entomopoxvirus will provide a means of obtaining new control opportunities more proper than with conventional techniques of insecticides applications. ÖZET Fındık kurdu {Balaninus nucum L., Coleoptera, Curculionidae) dünyada ve Türkiye'de fındığa önemli ölçüde zarar veren bir böcektir. Türkiye'de her yıl yaklaşık %30-40 oranında ekonomik zarar oluşturmaktadır. Fındık kurduna karşı etkin bir biyolojik mücadele ajanı geliştirmek amacıyla düzenlenen bu çalışmada bu böceğin biyolojisinin aydınlatılması, mikrobiyal florasının belirlenmesi ve ayrıca, gerek yeni izole edilen bakterilerin ve gerekse literatürde mevcut olan biyolojik materyallerin böcek üzerindeki zararlı etkilerinin araştırılması amaçlanmaktadır. Bu amaçla, 1995-1996 yıllarında Trabzon yöresinden toplanan ergin ve larvalar laboratuvarda incelenmiştir. Böceğin biyolojik özellikleri, mikrobiyal florası ve çeşitli biyolojik materyallerin bu böceğe karşı olan insektisid etkileri test edilmiştir. Sonuç olarak, dişi ve erkek böceğin kanat uzunlukları (10±0.8 mm ve 9.5±0.3 mm), hortum uzunlukları (6±0.4 mm ve 3.8±0.2 mm), cinsiyet oranı (1/1), yumurta boyutları (0.795±0.14 mm ve 0.525±0.11 mm), larval dönemlere ait baş kapsül genişlikleri (512.5±15 pim, 800±8 pim, 1005±10 pim ve 1295±6 pim), ergin ve larva başına düşen toplam bakteri sayısı (1.41x1ü11 bakteri/ergin ve 1.8xl09 bakteri/larva) tespit edildi. Larvalardan izole edilen 5 bakteriyal izolatın morfolojik, fizyolojik ve biyokimyasal karakterleri belirlendi. Bu özelliklerine göre bakterilerin Bacillus sp., Escherichia sp., Micrococcus sp., Serratia sp. ve Pseudomonas sp. cinslerine ait oldukları tespit edildi. Ayrıca bu izolatlardan olan Serratia sp.'nin B. nucum larvaları üzerinde 3 günlük sürede %100'lük bir ölüm meydana getirdiği belirlendi. Diğer yandan insektisidal etkileri test edilen bazı materyallerin fındık kurdunu öldürücü etkilere sahip oldukları bulundu. 3 günlük sürede B. nucum üzerinde tespit edilen insektisidal etkiler, LdNPV %90, BTS-1 %90, PHE4-ADR %66, PHE4-AR %60, PHE4-AD %60, PHE4-A %56, AcNPV %60 ve HD-1 %70'dir. Hazelnut beetle {Balaninus nucum L., Coleoptera, Curculionidae) is the most effective damager of hazelnut fruits in all over the world and in Turkey. It causes approximately 30-40% of economical damage per year in Turkey. Investigation of the biology and microbial flora of larvae and adults of B. nucum. were purposed in this study. Also, insecticidal activities of bacteria isolated from B. nucum and various effective biological agents recorded in the literature were tested against B. nucum. For this purpose, we studied the larvae and adults collected from the vicinity of Trabzon in laboratory in 1995-1996. The biological features and bacterial flora of this insect were investigated. Also, insecticide effects of various biological agents and isolated bacteria against hazelnut beetle were tested. As conclusion, we determined the lengths of forewing of adult female and male (10±0.8 mm and 9.5±0.3 mm), the lengths of rostrum (6±0.4 mm and 3.8±0.2 mm), sexual index (1/1), dimensions of egg (0.795±0.14 x 0.525+0.11 mm), dimension of head capsule of each instars (512.5±15 um, 800±8 um, 1005±10 /*m and 1295±6 /*m) and the numbers of total bacteria in larvae (1.8xl09 bacteria/larva) and adults (1.41x1ü11 bacteria/adult). The morphological, physiological and biochemical characteristics of five isolated bacteria were also identified. According to determined features, the identified isolates placed into the genera of Bacillus sp., Escherichia sp., Micrococcus sp., Serratia sp. and Pseudomonas sp. Among five bacterial isolates, it is determined that Serratia sp. causes 100% mortality on B. nucum larvae. It is also found that tested biological agents have mortality effects on B. nucum larvae. The insecticidal effects determined on B. nucum within three days are 90% with LdNPV and BTS-1, 66% with PHE4-ADR, 60% with PHE4-AR, PHE4-AD, 56% with PHE4-A, 60% with AcNPV and 70% with HD-1.
