Lysosome is closely linked to autophagy, which plays a vital role in pancreatic adenocarcinoma (PDAC) tumor biology. This study investigated whether lysosome storage dysfunction (LSD) contributes to PDAC development. Germline putative pathogenic variants (PPVs) in genes involved in lysosome functions were compared between PDAC patients (N=418) and healthy controls (N=845). Furthermore,Galc-knockout mouse pancreas organoids and human PDAC organoids were used to evaluate the consequences of PPV status in PDAC development and establishment. LSD PPVs were enriched in PDAC patients (Log2OR=1.65, P=3.08×10−3). PPV carriers diagnosed with PDAC were younger than non-carriers (61.5 vs. 65.3 years, P=0.031). Hampered autophagolysosome activity with increased autophagy flux and elevated Ki-67 index were observed following GALC downregulation. RNA sequencing of human PDAC organoids revealed upregulation of metabolism related to LSD. Genetically defined lysosome dysfunction is frequently observed in young-age onset PDACs. Lysosome dysfunction might contribute to PDAC development via altered metabolism and impaired autophagolysosome activity.

Myxofibrosarcoma (MFS) is a histologic subtype of malignant fibrous histiocytoma (MFH), with a predominant myxoid component. MFS is characterized by locally aggressive behavior and a high rate of local recurrence, however, with a good prognosis. Head and neck MFS accounts for 3% of all cases of MFS. To date, only two cases of laryngeal MFS have been reported. Owing to the rarity of MFS, the clinical characteristics and optimal treatment options remain controversial. Surgical resection with a clear margin is considered the treatment of choice. Compared to traditional MFS tumors, epithelioid variants have worse prognosis. Other factors associated with a poor prognosis of MFS tumors include inadequate surgical margins, large tumor size, old age, and high-grade tumors. Herein, we report a case of high-grade epithelioid variant MFS located in the false vocal fold, requiring total laryngectomy to obtain an adequate surgical margin. To our knowledge, this is the first case report of epithelioid variant of high-grade MFS presenting in the larynx.

Aminoacyl-tRNA synthetase-interacting multifunctional protein 2 (AIMP2) is a non-enzymatic component required for the multi-tRNA synthetase complex. While exon 2 skipping alternatively spliced variant of AIMP2 (AIMP2-DX2) compromises AIMP2 activity and is associated with carcinogenesis, its clinical potential awaits further validation. Here, we found that AIMP2-DX2/AIMP2 expression ratio is strongly correlated with major cancer signaling pathways and poor prognosis, particularly in acute myeloid leukemia (AML). Analysis of a clinical patient cohort revealed that AIMP2-DX2 positive AML patients show decreased overall survival and progression-free survival. We also developed targeted RNA-sequencing and single-molecule RNA-FISH tools to quantitatively analyze AIMP2-DX2/AIMP2 ratios at the single-cell level. By subclassifying hematologic cancer cells based on their AIMP2-DX2/AIMP2 ratios, we found that downregulating AIMP2-DX2 sensitizes cells to anticancer drugs only for a subgroup of cells while it has adverse effects on others. Collectively, our study establishes AIMP2-DX2 as a potential biomarker and a therapeutic target for hematologic cancer., Ku, Kim et al develop a method to analyse the ratio of the alternatively spliced variant of AIMP2 to full length AIMP via single-molecule RNA-FISH. They can subclassify hematologic cancer based on AIMP2-DX2/AIMP2 ratio and find that cells with high AIMP2-DX2 ratio can be sensitized to chemotherapy drugs by depleting AIMP2-DX2.

The transformation of papillary thyroid carcinoma (PTC) to anaplastic thyroid carcinoma (ATC) is well documented in the literature but is an exceptionally rare occurrence in metastatic foci outside the primary thyroid lesion. Even rarer is the simultaneous occurrence of PTC and ATC in the cervical lymph nodes. We report the case of an 85-year-old man who presented with a rapidly growing neck mass diagnosed as PTC. Following surgery, multiple ATC foci in the metastatic cervical lymph node were found coexisting with PTC, whereas in the thyroid, only PTC was found. This case is of high clinical significance because transformation of PTC to ATC outside the thyroid gland per se is very rare and because it suggests rapidly growing tumors in an elderly patient. The use of core needle biopsies in cases with suspected rapid tumor growth can aid in proper diagnosis, surgical decision making, and patient counselling.

With the unprecedented development of perovskite solar cells (PSCs), various interlayers have been investigated. Interlayers have diverse roles in shifting the energy levels, attracting and extracting charges, and passivating surface traps. These can improve the stability of PSCs, as well as their power conversion efficiency (PCE). In this study, a small molecular self-assembled monolayer (2-[carbamimidoyl(methyl)amino]acetic acid) is introduced on a SnO2 layer. The acetic acid is known as creatine, existing naturally in the human body. It is eatable and dosed by athletes for energy. Due to its structural properties, creatine can be anchored on a SnO2 layer, thus, it can strengthen the charge extraction from a perovskite layer by forming interface dipoles and reducing the work function. Also, it can passivate the defects in the perovskite layer. These properties are comprehensively investigated via various experimental techniques. With the creatine layer as a cathode interfacial layer on the SnO2 layer, the PSC employing a triple cation-based perovskite achieves the highest PCE of 20.8% and retained 90% of it after 50 days. Furthermore, 22.1% of PCE is obtained from the formamidinium-based perovskite. This report represents an interesting application of creatine as an interlayer in PSCs and suggests the possible application of other essential amino acids.

Inborn Errors of Metabolism (IEMs) are a hereditary disorder in which a gene fails to operate correctly as a result of a mutation in a metabolic-related gene passed down from the parents. Majority of IEMs are inherited in autosomal recessive manner. Recent research has been undertaken on mutations in numerous metabolic genes, including IEMs and cancer development. This study used Next Generation Sequencing (NGS) data to confirm the link between metabolic-related genes and cancer development in heterozygous carriers. The PCAWG (Pan-Cancer Analysis of Whole Genomes) cancer patient cohort and the 1000G (1000 Genomes) healthy cohort were used in this work, with the TCGA (The Cancer Genome Atlas) cancer patient cohorts being used for validation analysis. A regression analysis was used to determine the statistical significance of the cancer patient group versus the normal cohort. We analyzed RNA-seq data by separating heterozygous carrier and non-carrier groups of IEM related genes. The proportion of IEM gene heterozygote carriers was higher in cancer patients than in the normal population. We chose genes showing similar patterns of occurrence with statistical significance across several cancer patient cohorts. Accordingly, the most important gene, phenylalanine hydroxylase (PAH), has been established, and the carrier frequency was consistently high in lung squamous cell carcinoma (LUSC). RNA analysis results revealed that the PAH heterozygous carrier group was enriched with various immune response pathways suggesting inflammation related carcinogenesis in PAH related LUSC. Our finding suggest that heterozygote carrier status of IEM genes may lead to generation of aberrant metabolites and the breakdown of metabolic networks, which eventually leads to cancer. Lysosome storage disease (LSD) is a congenital metabolic disorder caused by mutations in genes involved in the function of lysosomes. Defects in the genes encoding hydrolases and transporters and enzyme activators are found in LSD’s, leading to the accumulation of macromolecules in the late endocytic system. And this disruption of lysosomal homeostasis increases the induction of endoplasmic reticulum and oxidative stress, proliferative signal transduction, extracellular matrix degradation, Integrin-β4-mediated cell migration and invasion, and consequently can promote the development of oncogenic cells and malignant tumors. It is known that the function of lysosomes is very important in cancer diseases. Recently, through a preliminary study, we found that the incidence of myeloproliferative neoplasm (MPN) increases statistically significantly when the LSD putative pathogenic germline variant (PPGV) is present. Based on this, we performed single-cell RNA (scRNA) sequencing using MPN patient samples of both LSD PPGV carrier and non-carrier. We tried to determine how the LSD PPGV affects MPN cancer progression at the single-cell level. To confirm functional differences between the two groups, DEG, pathway enrichment, LR-pair interaction, and transcription enrichment analyses were performed. In the analysis, immune cells were composed of T-cell, NK-cell, and Monocyte (in the order of frequency). We are focusing on the germline mutation data to understand the possible cancer-causing gene mutation that has a chance of inheritability to the next generation. Cancer genomics has been already widely studied around the world in a pan-cancer manner. But we found that the majority of the ethnicity of the data sequenced is mainly the US and European origin. For the Korean cancer cohort, it is clear that the lack of sample size was the bottleneck for ethnicity-specific studies. It is necessary that there is a limited number of the study contribute to the knowledge that Korean specifically applicated to explore the possible association between germline mutation and cancer occurrences. The objective of this study is to discover the Korean-specific candidate genes that associate with the tumor.Our study collected around 3000 Korean-specific samples consisting of various cancer types and normal control. We also gathered normal control samples for 354 Super-Normal (tumor-free, age < 65, med-checked) samples, and 1094 Korean normal (U1K) samples. Whole Exome Sequencing has been applied for every sample except U1K cohort that has already been whole genome sequenced. Genome-Wide Association Test (GWAS) has been executed between Korean-specific cancer patients and Korean normal samples. The study has been dichotomously analyzed, common variants are studied by GWAS fashion, and the rare variants are studied by gene-level aggregated fashion. Not only statistically, but also functionally we make use of the knowledge that Nonsense Mediated Decay (NMD) information to break the limitation of the unexplained gene mutation that does not follow the Central Dogma. Clinical validation data is also utilized. In a clinical aspect, we validated the dataset with a Clinvar and decided whether the mutation has a piece of clinical evidence or not.By conducting a series of studies, we have successfully found that some candidate genes which are known for DNA binding genes including PABPC1, PABPC3, and ZNF717 were significantly associated. Indeed, a cell division cycle associated gene like CDC27 also represents a significant association with cancer phenotype. Furthermore, ATXN3, SEPT2, PCMTD1 mutation is observed more in the cancer cohort. We are performing comparison studies to further elucidate which ethnic group represents more gene mutation on different cancer types.In summary, we propose the method that presents the possible germline mutation that is associated with phenotype by various factors, and also proposed Korean-specific candidate genes that associate with cancer. Organic–inorganic perovskites are crystalline light absorbers which are gaining great attraction from the photovoltaic community. Surprisingly, the power conversion efficiencies of these perovskite solar cells have rapidly increased by over 25% in 2019, which is comparable to silicon solar cells. Despite the many advances in efficiency, there are still many areas to be improved to increase the efficiency and stability of commercialization. For commercialization and enhancement of applicability, the development of electron transport layer (ETL) and its interface for low temperature processes and efficient charge transfer are very important. In particular, understanding the ETL and its interface is of utmost importance, and when this understanding has been made enough, excellent research results have been published that can improve the efficiency and stability of the device. Here, we review the progress of perovskite solar cells. Especially we discuss recent important development of perovskite deposition method and its engineering as well as the electron transport layer.

Objectives: The aim of this study was to analyze the risk of malignancy in salivary gland tumors on the basis of the Milan System for Reporting Salivary Gland Cytopathology. Methods: A retrospective review was performed of the charts of patients with salivary gland tumors in whom the final diagnosis was confirmed by surgical excision. Preoperative fine needle aspiration results were categorized according to the Milan System for Reporting Salivary Gland Cytopathology: non-diagnostic (category I), nonneoplastic (category II), atypia of undetermined significance (category III), neoplasm (category IV), suspicious for malignancy (category V), and malignant (category VI). Fine needle aspiration and final diagnosis were compared, and the risk of malignancy and operative/oncological outcomes were analyzed. Results: A total of 288 patients were enrolled in this study. Postoperative histopathologic salivary gland malignancies were found in 30 (10.4%) patients. Risk of malignancy was 7.1%, 0%, 48.0%, 4.8%, 88.7%, and 100% in categories I, II, III, IV, V, and VI, respectively. The most common malignant tumor in category III was salivary duct carcinoma (37.5%), followed by acinic cell carcinoma (25.0%), mucoepidermoid carcinoma (25.0%), and squamous cell carcinoma (12.5%). The 5-year survival rate of patients with malignant tumors showed no statistical difference between category III and category V/VI ( P = .140). Risk of malignancy was 88.9% and 100% in category V and VI, respectively. Conclusions: A half of atypia of undetermined significance (category III) cases were malignant. Once diagnosed, the prognosis of malignant tumor in category III was similar with that in category V/VI.

Background/aim Using a syngeneic tongue cancer mouse model, the effect of CD40 agonist and PD-1 antagonist combination therapy for local recurrence after surgery was evaluated in a partially depleted CD4 model. Materials and methods C3H/HeN mice were injected with 0.05 mg of the anti-mouse CD4 clone GK1.5, causing partial depletion of CD4 cells. Tongue cancer was induced by injecting the squamous cell carcinoma (SCC) VII cell line, the tumor was resected by partial glossectomy, and CD40 agonist and/or PD-1 antagonist therapy was administered postoperatively. Results Partial depletion of CD4 cells resulted in faster growth of a recurring tumor in the tongue, faster loss of body weight, and decreased number of CD8a-positive cells in the tumor. Postoperative adjuvant therapy with a combination of CD40 agonist and PD-1 antagonist resulted in a significant increase in survival compared to the CD40 agonist single treatment. Conclusion CD40 agonist and PD-1 antagonist combination therapy could be an effective postoperative adjuvant treatment, especially in cases with decreased CD4 T cell activity.

In addition to having proper energy levels and high hole mobility (μh) without the use of dopants, hole-transporting materials (HTMs) used in n-i-p-type perovskite solar cells (PSCs) should be processed using green solvents to enable environmentally friendly device fabrication. Although many HTMs have been assessed, due to the limited solubility of HTMs in green solvents, no green-solvent-processable HTM has been reported to date. Here, we report on a green-solvent-processable HTM, an asymmetric D–A polymer (asy-PBTBDT) that exhibits superior solubility even in the green solvent, 2-methylanisole, which is a known food additive. The new HTM is well matched with perovskites in terms of energy levels and attains a high μh (1.13 × 10–3 cm2/(V s)) even without the use of dopants. We report a simple post annealing-free (PAF) method for making uniform, large grain-sized, and highly crystalline triple cation perovskite films. In this simple process, a perovskite precursor solution was spin-coated onto a TiO 2 mesoporous substrate and as an antisolvent, diethyl ether (DEE) was subsequently dripped onto the film during spinning to produce an intermediate phase (IP) of perovskite film. This IP was immediately immersed into a DEE bath at room temperature for only 1 min in replacement of the conventional post annealing (PA) treatment. The as-prepared PAF film was characterized by X-ray powder diffraction, UV–vis absorption spectroscopy, fourier transform infrared spectroscopy, scanning electron microscopy, atomic-force microscopy, and photoluminescence spectroscopy. The overall power conversion efficiency (PCE) of the PAF devices was in the range of 18.8–19.5%, which is comparable with the reported PA devices (17.2–18.1%), mainly due to the J SC and the FF values, caused by the high absorption ability and the large crystal size with better surface smoothness in the PAF film. This efficiency is the highest reported for perovskite deposition by PAF methods. This new method enables reducing the device fabrication time at room temperature, which will reduce the cost of manufacturing efficient perovskite solar cells.

To evaluate the prognostic value of the posterior cricoarytenoid (PCA) muscle atrophy observed on neck computed tomography (CT) in patients with unilateral vocal fold paralysis. CT images of 87 subjects with unilateral vocal fold paralysis (UVFP) were evaluated to analyze the PCA muscle atrophy and to measure the severity of the PCA muscle atrophy in semi-quantitative manner. The grading of the PCA muscle atrophy was compared with the recruitment pattern of laryngeal electromyography (LEMG) and restoration of vocal fold movement. The PCA muscle was identifiable on CT in 73 subjects. Using the PCA muscle atrophy as an indicator of UVFP, we correctly predicted the paralysis in 69 (94.5%). Grade of the PCA muscle atrophy is significantly correlated with recruitment pattern of LEMG. If the positive result is defined as the PCA muscle showed moderate to severe degree of atrophy, we could predict the persistent UVFP in 88% of patients. PCA muscle atrophy identified on CT scan in patients with UVFP, is associated with low rates of return of mobility in the affected vocal fold.

BACKGROUND/AIM We have yet to understand why JAK2-positive myeloproliferative neoplasms (MPN) patients manifest different phenotypes despite harboring JAK2 mutations and what drives secondary transformations. PATIENTS AND METHODS Using targeted sequencing, we analyzed mutational status of 17 polycythemia vera (PV), 16 essential thrombocythemia (ET), 8 primary myelofibrosis (PMF) patients who tested positive for JAK by polymerase chain reaction. RESULTS The somatic mutations in JAK2 influence the clinical behavior of the disease. We found that ASXL1 or EZH2 mutation acquisition after JAK2 leads to PV, while ASXL1 mutation acquisition before JAK2 leads to ET or PMF. Mutations in TP53, ASXL1, and splicing genes are associated with the prognosis of MPN. PMF was more frequently associated with splicing mutations, while PV was more closely related to mutations in chromatin modifiers. The presence of these mutations influenced hemogram at MPN diagnosis. CONCLUSION Each subtype of MPN harbors distinct patterns of somatic mutations and acquisition order, while mutations in TP53, ASXL1, and splicing genes may be associated with the prognosis of MPN.

Image processing and communications networking technologies are evolving at a rapid pace. Users are demanding high-quality and realistic services to enjoy cultural contents such as sports and concerts. Free-viewpoint video (FVV) streaming is an active field of research that enables realistic services. FVV streaming service augments the user experience by providing different viewpoints of the same scene. The user can navigate freely and continuously to any viewpoint. In order to provide FVV streaming services, the transmission method of various on-demand user viewpoints is important. In this paper, we propose and implement a DASH-based FVV streaming client to reduce the view switching latency by receiving a number of adjacent images. We verify the performance of the proposed client and analyze the switching time and transmission data rate depending on the number of simultaneously received videos.

We introduce 1 step pin-hole free CH3NH3PbI3−xClx perovskite layers by using heated airflow during the nucleation stage of the perovskite. Upon employing heated air, we stimulate uniformly distributed nuclei growth, resulting in a pin-hole free planar perovskite layer. We find an optimized heated airflow of 100 °C as the optimized condition. The resulting planar device employing a conventional TiO2 electron transporting layer exhibits 17.6% average power conversion efficiency with 14.3% maximum powerpoint (MPP) efficiency. In addition, our method gives a very reproducible perovskite layer. Thus, our pin-hole free perovskite layer allows for 14.9% efficiency in a larger area device (0.71 cm2) that is generally prone to shunting paths.

Hybrid perovskites form an extremely attractive class of materials for large scale, low-cost photovoltaic applications. Fullerene-based charge extraction layers have emerged as a viable n-type charge collection layer, and in “inverted” p–i–n device architectures the solar cells are approaching efficiencies of 20%. However, the regular n–i–p devices employing fullerenes still lag behind in performance. Here, we show that partial solubility of fullerene derivatives in the aprotic solvents used for the perovskites makes it challenging to retain integral films in multilayer solution processing. To overcome this issue we introduce cross-linkable fullerene derivatives as charge collection layers in n–i–p planar junction perovskite solar cells. The cross-linked fullerene layers are insolubilized and deliver improved performance in solar cells enabled by a controllable film thickness.

Simple Summary The recent advancement in high-throughput sequencing has become indispensable for immune-genomics and profiling the T- and B-cell receptor repertoires. Immune repertoire sequencing (IR-seq) and whole transcriptome sequencing (WTS) can be implemented to investigate and quantitatively characterize the complex pattern of the CDR3 region. We conducted T-cell diversity analysis result comparisons of these sequencing methods and suggest an intuitive approach to discriminate reliable TCR sequences and clonotype patterns from capturing errors. Although bulk-RNA sequencing is commonly used for cancer analysis, we confirmed capturing highly enriched TCR transcripts with IR-seq is more reliable for accurate immune repertoire discovery, and singleton read filtering criteria should be applied to capture true clonotypes from error-prone sequencing data. The use of such well-established data and analytical methodologies can broaden understanding of antigen specificity in immunity and enabling efficient therapeutic antibody finding. Abstract Analysis of the T-cell receptor (TCR) repertoire is essential to characterize the extensive collections of T-cell populations with recognizing antigens in cancer research, and whole transcriptome sequencing (WTS) and immune repertoire sequencing (IR-seq) are commonly used for this measure. To date, no standard read filtering method for IR measurement has been presented. We assessed the diversity of the TCR repertoire results from the paired WTS and IR-seq data of 31 multiple myeloma (MM) patients. To invent an adequate read filtering strategy for IR analysis, we conducted comparisons with WTS results. First, our analyses for determining an optimal threshold for selecting clonotypes showed that the clonotypes supported by a single read largely affected the shared clonotypes and manifested distinct patterns of mapping qualities, unlike clonotypes with multiple reads. Second, although IR-seq could reflect a wider TCR region with a higher capture rate than WTS, an adequate comparison with the removal of unwanted bias from potential sequencing errors was possible only after applying our read filtering strategy. As a result, we suggest that TCR repertoire analysis be carried out through IR-seq to produce reliable and accurate results, along with the removal of single-read clonotypes, to conduct immune research in cancer using high-throughput sequencing.
