Cesium (Cs) is a toxic alkaline metal affecting human health. Plant high-affinity K transporters (HAKs) involved in Cs uptake and transport have been identified in several plants. However, the molecular regulatory mechanisms of Cs uptake and transport, and homeostasis between Cs and K by HAKs remain unknown. In this study, TaHAK1 was overexpressed in rice (TaHAK1-OEs) to evaluate Cs absorption capacity and the Cs and K homeostasis mechanisms. Results showed that TaHAK1 promoted seedling growth by fixing Cs in the root cell wall and modifying Cs distribution. Transcriptome and bioinformatics analyses revealed that 37,828 differentially expressed genes (DEGs) were significantly induced in TaHAK1-OEs, of which the pathways involved in cell wall biosynthesis and ion absorption transport were notably affected including genes, XTHs, CSLEs, HAKs, and ABCs. Moreover, under Cs-contaminated soil, TaHAK1-OEs exhibited improved Cs tolerance by decreasing Cs accumulation and increasing K content in different tissues, particularly in the grains, indicating that TaHAK1 acts as a candidate gene for screening genetic modification of Cs phytoremediation and developing low-Cs-accumulation rice varieties. Small chemical molecules are attractive agents for improving the plant processes associated with plant growth and stress tolerance. Recent advances in chemical biology and structure-assisted drug discovery approaches have opened up new avenues in plant biology to discover new drug-like molecules to improve plant processes for sustained food production. Several compounds targeting phytohormone biosynthesis or signalling cascades were designed to alter plant physiological mechanisms. Altering Abscisic acid synthesis and its signalling process can improve drought tolerance, and the processes targeted are reversible. Molecules targeting cytokinin, Auxin, and gibberellic acid regulate plant physiological processes and can potentially improve plant growth, biomass and productivity. The potential of molecules may be exploited as agrochemicals to enhance agricultural productivity. The discovery of small molecules provides new avenues to improve crop production in changing climatic conditions and the nutritional quality of foods. Cadmium is commonly recognized as toxic to plant growth, low-level Cd has promoting effects on growth performance, which is so-called hormesis. Although Cd toxicity in wheat has been widely investigated, knowledge of growth response to a broad range of Cd concentrations, especially extremely low concentrations, is still unknown. In this study, the morphological, physiological, and biochemical performance of wheat seedlings to a wide range of Cd concentrations (0-100 µΜ) were explored. Low Cd treatment (0.1-0.5 µM) improved wheat biomass and root development by enhancing the photosynthetic system and antioxidant system ability. Photosynthetic rate (P n ) was improved by 5.72% under lower Cd treatment (1 µΜ), but inhibited by 6.05-49.85% from 5 to 100 µΜ. Excessive Cd accumulation induced oxidative injury manifesting higher MDA content, resulting in lower photosynthetic efficiency, stunted growth, and reduction of biomass. Further, the contents of ascorbate, glutathione, non-protein thiols, and phytochelatins were improved under 5-100 µΜ Cd treatment. The ascorbate peroxidase activity in the leaf showed a hormetic dose-response characteristic. Correlation analysis and partial least squares (PLS) results indicated that antioxidant enzymes and metabolites were closely correlated with Cd tolerance and accumulation. The results of the element network, correlation analysis, and PLS showed a crucial role for exogenous Cd levels in K, Fe, Cu, and Mn uptake and accumulation. Key Message: Sodium treatment caused the sodium ion accumulation at the milk stage of immature rice grains which in turn triggered the overproduction of reactive oxygen species and oxidative damage. The tolerant cultivar showed an enhanced antioxidative response and induced expressions of OsNHX and OsHKT ion-transporters. Sodium chloride-(NaCl) induced soil salinity is a major constraint hindering global rice production. Amongst its constituent ions, sodium (Na + ) is known to be the main driver of toxicity under salt stress. The present investigation aims to measure the impacts of excess Na + during rice grain filling using two Indica rice cultivars with opposite tolerances to salt (salt tolerant: Panvel-3, salt-sensitive: Sahyadri-3) mainly via oxidative and responsive antioxidative pathways. Plants were treated with Na + -specific treatments and NaCl with equimolar Na + levels (100 mM) at the initiation of the reproductive phase. Stressed and control plants were harvested at three different grain-filling stages- early milk, milk, and dough and assessed for ion accumulation and oxidative damage/antioxidant responses under Na + stress. Na + toxicity triggered reactive oxygen species (ROS) production and upregulated the responsive enzymatic antioxidants. Na + stress also increased the nitric oxide (NO) levels and the activity of nitrate reductase in immature grains. Differential expression levels of OsNHX and OsHKT transporters were observed in response to Na + stress. Mature grains displayed a high accumulation of Na + along with reduced K + content and elevated Na + /K + under high Na + availability. The alterations in mature grains' sugar, starch, and protein content were also observed in response to the Na + stress. Overall, the salt-tolerant cultivar displayed higher antioxidant activities and a lower rate of ROS generation in response to the Na + stress. Fungal plant diseases are a major threat to plants and vegetation worldwide. Recent technological advancements in biotechnological tools and techniques have made it possible to identify and manage fungal plant diseases at an early stage. These techniques include direct methods, such as ELISA, immunofluorescence, PCR, flow cytometry, and in-situ hybridization, as well as indirect methods, such as fluorescence imaging, hyperspectral techniques, thermography, biosensors, nanotechnology, and nano-enthused biosensors. Early detection of fungal plant diseases can help to prevent major losses to plantations. This is because early detection allows for the implementation of control measures, such as the use of fungicides or resistant varieties. Early detection can also help to minimize the spread of the disease to other plants. The techniques discussed in this review provide a valuable resource for researchers and farmers who are working to prevent and manage fungal plant diseases. Abiotic stresses are a major constraint for agricultural productivity and food security in today's era of climate change. Plants can experience different types of abiotic stresses, either individually or in combination. Sometimes, more than one stress event may occur simultaneously or one after another during the lifecycle of the plant. In general, key survival strategies for stress tolerance may differ from one stress to another. However, at the molecular level, evolutionarily conserved protein kinase SUCROSE NONFERMENTING 1 (SNF1)-related protein kinase (SnRK) gene family members, comprising SnRK1, SnRK2, and SnRK3 gene families, play a key role in different types of stress and adaptive responses. SnRK gene family members can act as master regulators and regulate the central metabolism of plants, which determines the energy distribution in either survival or growth/developmental processes. The key mechanism of SnRK-mediated regulation is associated with the phosphorylation of downstream genes, which either induces or dampens the function of target proteins. This may be crucial for maintaining differential morpho-physiological and biochemical processes in plants, including potassium signalling, ROS homeostasis, sugar signalling, and energy homeostasis. Furthermore, phosphorylation sites associated with different targets were also reviewed, which showed that SnRK-mediated phosphorylation of Serine and Threonine residues of the target protein is a site-specific event, where the target consists of specific amino acid sequences, including RXXS/T, Serine-threonine rich regions, or AMPK/SNF1 types. Surface water pollution is a critical and urgent global issue that demands immediate attention. Surface water plays a crucial role in supporting and sustaining life on the earth, but unfortunately, till now, we have less understanding of its spatial and temporal dynamics of discharge and storage variations at a global level. The contamination of surface water arises from various sources, classified into point and non-point sources. Point sources are specific, identifiable origins of pollution that release pollutants directly into water bodies through pipes or channels, allowing for easier identification and management, e.g., industrial discharges, sewage treatment plants, and landfills. However, non-point sources originate from widespread activities across expansive areas and present challenges due to its diffuse nature and multiple pathways of contamination, e.g., agricultural runoff, urban storm water runoff, and atmospheric deposition. Excessive accumulation of heavy metals, persistent organic pollutants, pesticides, chlorination by-products, pharmaceutical products in surface water through different pathways threatens food quality and safety. As a result, there is an urgent need for developing and designing new tools for identifying and quantifying various environmental contaminants. In this context, chemical and biological sensors emerge as fascinating devices well-suited for various environmental applications. Numerous chemical and biological sensors, encompassing electrochemical, magnetic, microfluidic, and biosensors, have recently been invented by hydrological scientists for the detection of water pollutants. Even though researches have shown that biochar can improve soil-health and plant-growth even in harsh environments and get rid of harmful heavy metals and new contaminants, it is still not sustainable, affordable, or effective enough. Therefore, scientists are required to develop nanomaterials in order to preserve numerous aquatic and terrestrial species. The carbonaceous chemical known as nano-biochar (N-BC) can be used to get rid of metal contamination and emerging contaminants. However, techniques to reduce hetero-aggregation and agglomeration of nano-biochar are needed that lead to the emergence of emerging nano-biochar (EN-BC) in order to maximise its capacity for adsorption of nano-biochar. To address concerns in regards to the expanding human population and sustain a healthy community, it is imperative to address the problems associated with toxic heavy metals, emerging contaminants, and other abiotic stressors that are threatening agricultural development. Nano-biochar can provide an effective solution for removal of emerging contaminants, toxic heavy metals, and non-degradable substance. This review provides the detailed functional mechanistic and kinetics of nano-biochar, its effectiveness in promoting plant growth, and soil health under abiotic stress. Climate change poses a substantial threat to agricultural sustainability globally. Agriculture is a vital component of the gross domestic production of developing countries. The socioeconomic status of farmers is particularly vulnerable to climatic extremes with future projections indicating significant increment in ambient air temperatures and unpredictable, intense rainfall patterns. Agriculture has historically relied on the extensive use of synthetic fertilizers, herbicides, and insecticides, combined with advancements in irrigation and biotechnological approaches to boost productivity. It encompasses a range of practices designed to enhance the resilience of agricultural systems, improve productivity, and reduce greenhouse gas emissions. By adopting climate-smart practices, farmers can better adapt to changing climatic conditions, thereby ensuring more sustainable and secure food production. Hypoxia occurs when oxygen levels fall below the levels required for mitochondria to support respiration. Regulated hypoxia is associated with quiescence, particularly in storage organs (seeds) and stem cell niches. In contrast, environmentally induced hypoxia poses significant challenges for metabolically active cells that are adapted to aerobic respiration. The perception of oxygen availability through cysteine oxidases, which function as oxygen-sensing enzymes in plants that control the N-degron pathway, and the regulation of hypoxia-responsive genes and processes is essential to survival. Functioning together with reactive oxygen species (ROS), particularly hydrogen peroxide (H2O2) and reactive nitrogen species (RNS), such as nitric oxide (·NO), nitrogen dioxide (·NO2), S-nitrosothiols (SNOs), and peroxynitrite (ONOO-), hypoxia signaling pathways trigger anatomical adaptations such as formation of aerenchyma, mobilization of sugar reserves for anaerobic germination, formation of aerial adventitious roots, and the hyponastic response. NO and H2O2 participate in local and systemic signaling pathways that facilitate acclimation to changing energetic requirements, controlling glycolytic fermentation, the γ-aminobutyric acid (GABA) shunt, and amino acid synthesis. NO enhances antioxidant capacity and contributes to the recycling of redox equivalents in energy metabolism through the phytoglobin (Pgb)-NO cycle. Microplastics and nanoplastics (MNPs), are minute particles resulting from plastic fragmentation, have raised concerns due to their widespread presence in the environment. This study investigates sources and distribution of MNPs and their impact on plants, elucidating the intricate mechanisms of toxicity. Through a comprehensive analysis, it reveals that these tiny plastic particles infiltrate plant tissues, disrupting vital physiological processes. Micro and nanoplastics impair root development, hinder water and nutrient uptake, photosynthesis, and induce oxidative stress and cyto-genotoxicity leading to stunted growth and diminished crop yields. Moreover, they interfere with plant-microbe interactions essential for nutrient cycling and soil health. The research also explores the translocation of these particles within plants, raising concerns about their potential entry into the food chain and subsequent human health risks. The study underscores the urgency of understanding MNPs toxicity on plants, emphasizing the need for innovative remediation strategies such as bioremediation by algae, fungi, bacteria, and plants and eco-friendly plastic alternatives. The Oryza genus, containing Oryza sativa L., is quintessential to sustain global food security. This genus has a lot of sophisticated molecular mechanisms to cope with environmental stress, particularly during vulnerable stages like flowering. Recent studies have found key involvements and genetic modifications that increase resilience to stress, including exogenous application of melatonin, allantoin, and trehalose as well as OsSAPK3 and OsAAI1 in the genetic realm. Due to climate change and anthropogenic reasons, there is a rise in sea level which raises a concern of salinity stress. It is tackled through osmotic adjustment and ion homeostasis, mediated by genes like P5CS, P5CR, GSH1, GSH2, and SPS, and ion transporters like NHX, NKT, and SKC, respectively. Oxidative damage is reduced by a complex action of antioxidants, scavenging RONS. A complex action of genes mediates cold stress with studies highlighting the roles of OsWRKY71, microRNA2871b, OsDOF1, and OsICE1. There is a need to research the mechanism of action of proteins like OsRbohA in ROS control and the action of regulatory genes in stress response. This is highly relevant due to the changing climate which will raise a lot of environmental changes that will adversely affect production and global food security if certain countermeasures are not taken. Nanotechnology is a new scientific area that promotes unique concepts to comprehend the optimal mechanics of nanoparticles (NPs) in plants under heavy metal stress. The present investigation focuses on effects of synthetic and green synthesized titanium dioxide nanoparticles (TiO 2 NPs and gTiO 2 NPs) against Cr(VI). Green TiO 2 NPs have been produced from plant leaf extract (Ricinus communis L.). Synthesis was confirmed employing an array of optical spectroscopic and electron microscopic techniques. Chromium strongly accelerated H 2 O 2 and MDA productions by 227 % and 266 % at highest chromium concentration (60 mg/kg of soil), respectively, and also caused DNA damage, and decline in photosynthesis. Additionally, anomalies were observed in stomatal cells with gradual increment in chromium concentrations. Conversely, foliar applications of TiO 2 NPs and gTiO 2 NPs considerably mitigated chromium stress. Sunflower plants treated with modest amounts of green TiO 2 NPs had significantly better growth index compared to chemically synthesized ones. Principal component analysis highlighted the variations among photosynthetic attributes, oxidative stress markers, and antioxidant defense systems. Notably, gTiO 2 supplementation to the Cr(VI) strained plants minimized PC 3 production which is a rare report so far. Herein, a polymer-based bioadsorbent was prepared by cross-linking chitosan to filter mud and magnetic pectin (Ch-mPC@FM) for the removal of Bismark Brown R dye (BB-R) from wastewater. Morphological characterization analysis indicated that Ch-mPC@FM had a higher surface area and better pore structure than its components. The Artificial Neuron Network (ANN) and Adaptive Neuro-Fuzzy Inference System (ANFIS) were employed to evaluate the simulation and prediction of the adsorption process based on input variables like temperature, pH, dosage, initial BB-R dye concentration, and contact time. ANFIS and ANN demonstrated significant modeling and predictive accuracy, with R 2  > 0.93 and R 2  > 0.96, and root mean square error < 0.023 and <0.020, respectively. The Langmuir isotherm and the pseudo-second-order kinetic models provided the best fits to the equilibrium and kinetic data. The thermodynamic assessment showed spontaneous and endothermic adsorption with average entropy and enthalpy changes of 119.32 kJ mol -1  K and 403.47 kJ mol -1 , respectively. The study of BB-R dye adsorption on Ch-mPC@FM revealed multiple mechanisms, including electrostatic, complexation, pore filling, cation-π interaction, hydrogen bonding, and π-π interactions. Climate change, food insecurity, and agricultural pollution are all serious challenges in the twenty-first century, impacting plant growth, soil quality, and food security. Innovative techniques are required to mitigate these negative outcomes. Toxic heavy metals (THMs), organic pollutants (OPs), and emerging contaminants (ECs), as well as other biotic and abiotic stressors, can all affect nutrient availability, plant metabolic pathways, agricultural productivity, and soil-fertility. Comprehending the interactions between root exudates, microorganisms, and modified biochar can aid in the fight against environmental problems such as the accumulation of pollutants and the stressful effects of climate change. Microbes can inhibit THMs uptake, degrade organic pollutants, releases biomolecules that regulate crop development under drought, salinity, pathogenic attack and other stresses. However, these microbial abilities are primarily demonstrated in research facilities rather than in contaminated or stressed habitats. Despite not being a perfect solution, biochar can remove THMs, OPs, and ECs from contaminated areas and reduce the impact of climate change on plants. We hypothesized that combining microorganisms with biochar to address the problems of contaminated soil and climate change stress would be effective in the field. The biological synthesis of zinc oxide nanoparticles (ZnO NPs) from plant extracts has emerged as a novel method for producing NPs with great scalability and biocompatibility. The present study is focused on bio-fabricated zinc oxide nanomaterial characterization and investigation of its photocatalytic and antifungal activities. ZnO NPs were biosynthesized using the leaf extract of Polyalthia longifolia without using harmful reducing or capping chemicals, which demonstrated fungicidal activity against Fusarium oxysporum f. sp. ciceris . The results showed that the inhibition of the radial growth of F . oxysporum f. sp. ciceris was enhanced as the concentration increased from 100 ppm to 300 ppm. The effectiveness of the photocatalytic activity of biosynthesized ZnO NPs was analyzed using MB dye degradation in aqueous medium under ultraviolet (UV) radiation and natural sunlight. After four consecutive cycles, the photocatalytic degradation of MB was stable and was 84%, 83%, 83%, and 83%, respectively, during natural sunlight exposure. Under the UV sources, degradation reached 92%, 89%, 88%, and 87%, respectively, in 90 minutes. Key Message: Sodium nitroprusside mediates drought stress responses in tomatoes by modulating nitrosative and oxidative pathways, highlighting the interplay between nitric oxide, hydrogen sulfide, and antioxidant systems for enhanced drought tolerance. While nitric oxide (NO), a signalling molecule, enhances plant tolerance to abiotic stresses, its precise contribution to improving tomato tolerance to drought stress (DS) through modulating oxide-nitrosative processes is not yet fully understood. We aimed to examine the interaction of NO and nitrosative signaling, revealing how sodium nitroprusside (SNP) could mitigate the effects of DS on tomatoes. DS-seedlings endured 12% polyethylene glycol (PEG) in a 10% nutrient solution (NS) for 2 days, then transitioned to half-strength NS for 10 days alongside control plants. DS reduced total plant dry weight, chlorophyll a and b, Fv/Fm, leaf water potential (Ψ I ), and relative water content, but improved hydrogen peroxide (H 2 O 2 ), proline, and NO content. The SNP reduced the DS-induced H 2 O 2 generation by reducing thiol (-SH) and the carbonyl (-CO) groups. SNP increased not only NO but also the activity of L-cysteine desulfhydrase (L-DES), leading to the generation of H 2 S. The mutualistic plant rhizobacteria which improve plant development and productivity are known as plant growth-promoting rhizobacteria (PGPR). It is more significant due to their ability to help the plants in different ways. The main physiological responses, such as malondialdehyde, membrane stability index, relative leaf water content, photosynthetic leaf gas exchange, chlorophyll fluorescence efficiency of photosystem-II, and photosynthetic pigments are observed in plants during unfavorable environmental conditions. Plant rhizobacteria are one of the more crucial chemical messengers that mediate plant development in response to stressed conditions. The interaction of plant rhizobacteria with essential plant nutrition can enhance the agricultural sustainability of various plant genotypes or cultivars. Rhizobacterial inoculated plants induce biochemical variations resulting in increased stress resistance efficiency, defined as induced systemic resistance. Omic strategies revealed plant rhizobacteria inoculation caused the upregulation of stress-responsive genes-numerous recent approaches have been developed to protect plants from unfavorable environmental threats. To the best of our knowledge, there was no prior report providing valuable preliminary data through a demonstration of the quantitative phytochemical and antioxidant activity of Gymnosporia senegalensis. The total contents of phenols, flavonoid, flavanol, tannin, and saponin were evaluated from different fractions extracted from the leaf, stem, and bark of G. senegalensis by using standards such as gallic acid, quercetin, rutin, tannic acid, and saponin quillaja. The antioxidant potential was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydrogen peroxide scavenging (H 2 O 2 ), superoxide anion radical scavenging, metal chelating ferrous ion, ferric reducing antioxidant power (FRAP), and total antioxidant capacity (TAC). Data were subjected to half-inhibitory concentration (IC 50 ) and one-way analysis of variance (ANOVA) at p < 0.05 as a significant value. The total phenol content was found to be highest in the chloroform extract of stem at 97.7 ± 0.02 mg GAE/g. The total flavonoid and flavonol contents in the aqueous extract were 97.1 ± 0.03 mg QE/g and 96.7 ± 0.07 mg RE/g, respectively. The total tannin content in the ethyl acetate extract of leaf was 97.5 ± 0.01 mg TAE/g, and the total saponin content in the methanol extract of stem was 79.1 ± 0.06 mg SQE/g. The antioxidant analysis indicated that IC 50 and percentage (%) inhibition were dose-dependent and showed the highest antioxidant activity (40.9 ± 0.9 µg/mL) in methanol extract of leaf for DPPH, (88.8 ± 1.12 µg/mL) in the chloroform extract of stem for H 2 O 2 , (43.9 ± 0.15 µg/mL) in the aqueous extract of bark for superoxide anion radical scavenging activity, (26.9 ± 0.11 µg/mL) in the chloroform extract of leaf for the metal chelating ferrous ion activity, (7.55 ± 0.10 mg/mL) in the benzene extract of leaf for FRAP, and (2.97 ± 0.01 mg/mL) in the methanol extract of bark for TAC. These results show that G. senegalensis has great potential in antioxidant activities. The isolation and characterization of specific bioactive compounds and the in vivo applicability of such activity await further extensive studies for drug discovery and development.

Sustainable food security and safety are major concerns on a global scale, especially in developed nations. Adverse agroclimatic conditions affect the largest agricultural-producing areas, which reduces the production of crops. Achieving sustainable food safety is challenging because of several factors, such as soil flooding/waterlogging, ultraviolet (UV) rays, acidic/sodic soil, hazardous ions, low and high temperatures, and nutritional imbalances. Plant growth-promoting rhizobacteria (PGPR) are widely employed in in-vitro conditions because they are widely recognized as a more environmentally and sustainably friendly approach to increasing crop yield in contaminated and fertile soil. Conversely, the use of nanoparticles (NPs) as an amendment in the soil has recently been proposed as an economical way to enhance the texture of the soil and improving agricultural yields. Nowadays, various research experiments have combined or individually applied with the PGPR and NPs for balancing soil elements and crop yield in response to control and adverse situations, with the expectation that both additives might perform well together. In recent years, there has been a growing focus on treating textile wastewater due to its escalating threat to aquatic ecosystems and exposed communities. The present study investigates the adsorption efficacy of biopolymer functionalized nanoscale zero-valent iron (CS@nZVI) composite for the treatment of textile wastewater using the RSM-CCD model. The structure and morphology of CS@nZVI were characterized using XRD, FTIR, FESEM, and EDX. CS@nZVI was then evaluated for its adsorption potential in removing COD, color, and other physico-chemical parameters from textile wastewater. The results showed the high efficacy of CS@nZVI for COD and color removal from textile wastewater. Under optimal conditions (pH 6, contact time 60 min, and 1.84 g CS@nZVI), COD removal reached a maximum of 85.53%, and decolorization efficiency was found to be 89.73%. The coefficient of determination R 2 (0.98) and AIC (269.75) values suggested quadratic model as the best-fitted model for optimizing the process parameters for COD removal. Additionally, the physico-chemical parameters were found to be within permissible limits after treatment with CS@nZVI. The influence of coexisting ions on COD removal followed the order PO 4 3-  > SO 4 2-  > Cl - . The kinetics data fitted well with the pseudo-first-order reaction, indicating physisorption as the primary mechanism. The thermodynamic study revealed the endothermic nature of the removal process. Reusability tests demonstrated that great regeneration capacity of spent CS@nZVIafter five consecutive cycles. The increasing global demand for tellurium, driven by its critical role in alloys, photovoltaic devices, and electronics, has raised concerns about its environmental pollution and neurotoxicity. In response, the potential of alginic acid (AA), a renewable, low-cost, and sustainable biopolymer, was explored for the biosynthesis of ultra-small silver nanoparticles (AgNPs) and their application in the detection of tellurium (Te(IV)). The effect of key synthesis parameters on desired physicochemical properties and yield of AgNPs was established to ensure high specificity and sensitivity towards Te(IV). The purified AgNPs with AA surface ligands were utilized to demonstrate a ratiometric absorbance sensor that exhibits excellent linearity and nanomolar-level affinity. This approach achieved a high correlation coefficient of ∼ 0.982, with a low detection limit of about 22 nM. Further investigations into the effect of pH, ionic strength, and organic molecules were conducted to elucidate detection performance and molecular understanding. The detection mechanism relies on the coordination between Te(IV) ions and the carboxylate groups of AA, which initiates aggregation-induced plasmon coupling in adjacent AgNPs. A relative of cultivated rice (Oryza sativa L.), weedy or red rice (Oryza spp.) is currently recognized as the dominant weed, leading to a drastic loss of yield of cultivated rice due to its highly competitive abilities like producing more tillers, panicles, and biomass with better nutrient uptake. Due to its high nutritional value, antioxidant properties (anthocyanin and proanthocyanin), and nutrient absorption ability, weedy rice is gaining immense research attentions to understand its genetic constitution to augment future breeding strategies and to develop nutrition-rich functional foods. Consequently, this review focuses on the unique gene source of weedy rice to enhance the cultivated rice for its crucial features like water use efficiency, abiotic and biotic stress tolerance, early flowering, and the red pericarp of the seed. It explores the debating issues on the origin and evolution of weedy rice, including its high diversity, signalling aspects, quantitative trait loci (QTL) mapping under stress conditions, the intricacy of the mechanism in the expression of the gene flow, and ecological challenges of nutrient removal by weedy rice. Nanotechnology offers the potential to provide innovative solutions for sustainable crop production as plants are exposed to a combination of climate change factors (CO 2 , temperature, UV radiation, ozone), abiotic (heavy metals, salinity, drought), and biotic (virus, bacteria, fungi, nematode, and insects) stresses. The application of particular sizes, shapes, and concentration of nanomaterials (NMs) potentially mitigate the negative impacts in plants by modulation of photosynthetic rate, redox homeostasis, hormonal balance, and nutrient assimilation through upregulation of anti-stress metabolites, antioxidant defense pathways, and genes and genes network. The present review inculcates recent advances in uptake, translocation, and accumulation mechanisms of NMs in plants. Increasing soil cadmium (Cd) contamination is a serious threat to human food health and safety. In order to reduce Cd uptake and Cd toxicity in silage maize, hydroponic tests were conducted to investigate the effect of exogenous Cd on the toxicity of silage maize in this study. In the study, a combination of Cd (5, 20, 50, 80, and 10 μM) treatments was applied in a hydroponic system. With increasing Cd concentration, Cd significantly inhibited the total root length (RL), root surface area (SA), root volume (RV), root tip number (RT), and branching number (RF) of maize seedlings, which were reduced by 28.1 to 71.3%, 20.2 to 64.9%, 11.2 to 56.5%, 43.7 to 63.4%, and 38.2 to 72.6%, respectively. The excessive Cd accumulation inhibited biomass accumulation and reduced silage maize growth, photosynthesis, and chlorophyll content and activated the antioxidant systems, including increasing lipid peroxidation and stimulating catalase (CAT) and peroxidase (POD), but reduced the activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in the root. Besides, selenium (Se) significantly decreased the Cd concentration of the shoot and root by 27.1% and 35.1% under Cd 50 , respectively. Our results reveal that exogenously applied Cd reduced silage maize growth and impaired photosynthesis. Whereas silage maize can tolerate Cd by increasing the concentration of ascorbate and glutathione and activating the antioxidant defense system, the application of exogenous selenium significantly reduced the content of Cd in silage maize., (© 2024. Climate change imposes various environmental stresses which substantially impact plant growth and productivity. Salinity, drought, temperature extremes, heavy metals, and nutritional imbalances are among several abiotic stresses contributing to high yield losses of crops in various parts of the world, resulting in food insecurity. Many interesting strategies are being researched in the attempt to improve plants' environmental stress tolerance. These include the application of nanoparticles, which have been found to improve plant function under stress situations. Nanotechnology will be a key driver in the upcoming agri-tech and pharmaceutical revolution, which promises a more sustainable, efficient, and resilient agricultural and medical system Nano-fertilizers can help plants utilise nutrients more efficiently by releasing nutrients slowly and sustainably. Salt stress is a recognized annihilating abiotic stress that has a significant impact on agricultural and horticulture crop productivity. Plant development faces three distinct dangers as a result of salt stress: oxidative stress, osmotic stress, and ionic toxicity. It has been shown that plants can forecast diurnal patterns using the circadian clock; moreover, they can manage their defensive mechanism for the detoxification of reactive oxygen species (ROS). Circadian rhythmicity in gene expression assembles transcription and translation feedback networks to govern plant shape, physiology, cellular and molecular activities. Both external and internal variables influence the systemic rhythm via input routes. The Malav Jyoti (MJ) and Delhi Green (DG) genotypes of spinach (Spinacia oleracea) were grown in the plant growth chamber. The chamber had an optimized temperature of 25 °C and humidity of 65% containing light emitting diode (LED) having Red: Blue: white (one side) and White fluorescent (other side) under salinity stress. The samples were collected on the basis of 4 h intervals of circadian hours (0 h, 4 h, 8 h and 12 h) during Day-10 and Day-20 of salt treatments. Under salt stress, the circadian and light-emitting diode-based strategy had a substantial influence on spinach's anti-oxidative responses, stomatal movement, CO 2 assimilation, PS-I and II efficiency, phytochrome pigment efficiency, and photosynthesis. Based on the findings of the free radical scavenging enzyme tests, the photoperiodic hours for the proteome analysis were set to 11 am and 3 pm on Day-20. When compared to white fluorescent, this study found that LED has the capacity to influence the entrainment cues of the circadian clock in the cultivation of salt-sensitive spinach genotypes. The evolution and rapid spread of multidrug-resistant (MDR) bacterial pathogens have become a major concern for human health and demand the development of alternative antimicrobial agents to combat this emergent threat. Conventional intracellular methods for producing metal nanoparticles (NPs) using whole-cell microorganisms have limitations, including binding of NPs to cellular components, potential product loss, and environmental contamination. In contrast, this study introduces a green, extracellular, and sustainable methodology for the bio-materialization of silver NPs (AgNPs) using renewable resource cell-free yeast extract. These extracts serve as a sustainable, biogenic route for both reducing the metal precursor and stabilizing the surface of AgNPs. This method offers several advantages such as cost-effectiveness, environment-friendliness, ease of synthesis, and scalability. HR-TEM imaging of the biosynthesized AgNPs revealed an isotropic growth route, resulting in an average size of about ~ 18 nm and shapes ranging from spherical to oval. Further characterization by FTIR and XPS results revealed various functional groups, including carboxyl, hydroxyl, and amide contribute to enhanced colloidal stability. AgNPs exhibited potent antibacterial activity against tested MDR strains, showing particularly high efficacy against Gram-negative bacteria. These findings suggest their potential role in developing alternative treatments to address the growing threat of antimicrobial resistance. Additionally, seed priming experiments demonstrated that pre-sowing treatment with AgNPs improves both the germination rate and survival of Sorghum jowar and Zea mays seedlings. Plant pathogens, including bacteria, fungi, and viruses, pose significant challenges to the farming community due to their extensive diversity, the rapidly evolving phenomenon of multi-drug resistance (MDR), and the limited availability of effective control measures. Amid mounting global pressure, particularly from the World Health Organization, to limit the use of antibiotics in agriculture and livestock management, there is increasing consideration of engineered nanomaterials (ENMs) as promising alternatives for antimicrobial applications. Studies focusing on the application of ENMs in the fight against MDR pathogens are receiving increasing attention, driven by significant losses in agriculture and critical knowledge gaps in this crucial field. In this review, we explore the potential contributions of silver nanoparticles (AgNPs) and their nanocomposites in combating plant diseases, within the emerging interdisciplinary arena of nano-phytopathology. AgNPs and their nanocomposites are increasingly acknowledged as promising countermeasures against plant pathogens, owing to their unique physicochemical characteristics and inherent antimicrobial properties. This review explores recent advancements in engineered nanocomposites, highlights their diverse mechanisms for pathogen control, and draws attention to their potential in antibacterial, antifungal, and antiviral applications. In the discussion, we briefly address three crucial dimensions of combating plant pathogens: green synthesis approaches, toxicity-environmental concerns, and factors influencing antimicrobial efficacy. The rhizosphere consists of a plethora of microbes, interacting with each other as well as with the plants present in proximity. The root exudates consist of a variety of secondary metabolites such as strigolactones and other phenolic compounds such as coumarin that helps in facilitating communication and forming associations with beneficial microbes in the rhizosphere. Among different secondary metabolites flavonoids (natural polyphenolic compounds) continuously increasing attention in scientific fields for showing several slews of biological activities. Flavonoids possess a benzo-γ-pyrone skeleton and several classes of flavonoids have been reported on the basis of their basic structure such as flavanones, flavonols, anthocyanins, etc. The mutualistic association between plant growth-promoting rhizobacteria (PGPR) and plants have been reported to help the host plants in surviving various biotic and abiotic stresses such as low nitrogen and phosphorus, drought and salinity stress, pathogen attack, and herbivory. This review sheds light upon one such component of root exudate known as flavonoids, which is well known for nodulation in legume plants. Drought is undoubtedly a major environmental constraint that negatively affects agricultural yield and productivity throughout the globe. Plants are extremely vulnerable to drought which imposes several physiological, biochemical and molecular perturbations. Increased generation of reactive oxygen species (ROS) and reactive nitrogen species (RNS) in different plant organs is one of the inevitable consequences of drought. ROS and RNS are toxic byproducts of metabolic reactions and poise oxidative stress and nitrosative stress that are detrimental for plants. In spite of toxic effects, these potentially active radicals also play a beneficial role in mediating several signal transduction events that lead to plant acclimation and enhanced survival under harsh environmental conditions. The precise understanding of ROS and RNS signaling and their molecular paradigm with different phytohormones, such as auxin, gibberellin, cytokinin, abscisic acid, ethylene, brassinosteroids, strigolactones, jasmonic acid, salicylic acid and melatonin play a pivotal role for maintaining plant fitness and resilience to counteract drought toxicity. Plastics' unavoidable and rampant usage causes their trash to be extensively dispersed in the atmosphere and land due to its numerous characteristics. Because of extensive plastic usage and increased manufacturing, there is insufficient recycling and a large accumulation of microplastics (MPs) in the environment. In addition to their wide availability in the soil and atmosphere, micro- and nanoplastics are becoming contaminants worldwide. Agro-ecosystem functioning and plant development are being negatively impacted in several ways by the contamination of the environment and farmland soils with MPs (<5 mm) and nanoplastics (<1 µm). The contributions of some recyclable organic waste and plastic film mulching and plastic particle deposition in agroecosystems may be substantial; therefore, it is crucial to understand any potentially hazardous or undesirable impacts of these pollutants on agroecosystems. The dissolution of bioplastics into micro- and nano-particles (MBPs and NBPs) has not been considered in recent studies, which focus primarily on agro-ecosystems. It is essential to properly understand the distribution, concentration, fate, and main source of MPs, NPS, MBPs, and NBPs in agroecosystems. Based on the limited findings, understanding the knowledge gap of environmental impact from micro and nanoplastic in farming systems does not equate to the absence of such evidence. Soil salinity is a global issue that limits plant growth in agricultural fields and contributes to food crisis. Salt stressors impede plant's ionic, osmotic, and oxidative balance, as well as a variety of physiological functions. Exposure to salinity stress manifest considerable ROS clustering, entailing modification in performance of various organelles. To deal with salinity, plants use a variety of coping strategies, such as osmoregulation, ion-homeostasis, increased antioxidant synthesis, and so on. Nitric oxide (NO) is a pivotal signalling molecule that helps facilitate salt stress-induced physiological plant responses. A variety of evidences point to NO being produced under similar stress conditions and with similar kinetics as hydrogen peroxide (H 2 O 2 ). The interplay between H 2 O 2 and NO has important functional implications for modulating plant transduction processes. Besides, NO and calcium (Ca 2+ )-dependent pathways also have some connection in salt stress response mechanisms. Extensive crosstalk between NO and Ca 2+ signalling pathways is investigated, and it suggests that almost every type of Ca 2+ channel is under the tight control of NO, and NO acts as a Ca 2+ mobilising compound and aids in signal reliance. In this article, the impact of an anthropogenically transformed environment on the content of pharmaceutically valuable biologically active compounds in medicinal plants is analyzed. The studied biologically active substances included phenolic compounds (flavonoids, anthocyanins, tannins, and phenolic acids). The number of transmissible forms of heavy metals (HMs), including cadmium, lead, and mercury, were discharged from factories that are present in the soil. Plants uptake these toxic metals from the soil. HM causes changes in the activity of the several enzymes such as phenylalanine ammonia lyase (PAL), chalcone synthase (CHS), chalcone isomerase (CHI) and other enzymes. These enzymes play an important role in biosynthesis of phenolic compounds in medicinal plants. It has been demonstrated that plant materials possess high antioxidant potential due to their high phenolic content. As a result, the present review discusses a thorough investigation of anthropogenically transformed environment effects on the quantity of pharmaceutically valuable phenolic compounds in medicinal plants.

Chromium (VI) is one of the hazardous heavy metal, heavily discharged into the soil and severely hampers the plants yield. The TiO 2 NPs was selected due to its potential to alleviate the heavy metals toxicity. This manuscript unravels the mechanisms for Cr(VI) induced toxicity and how foliar application of TiO 2 NP S potentially ameliorate the toxicity by regulating the photosynthetic attributes, DNA damage, antioxidants defense machinery, and phytochelatins synthesis in Helianthus annuus L. Plants were exposed to Cr(VI) concentrations [0, 15, 30, and 60 mg Cr(VI) kg -1 of soil], and TiO 2 NP S (15 mg L -1 , 25 nm size ) were foliar sprayed thrice to the plants at three days interval. The maximum accumulation of total chromium was recorded in root (12.53 µg g -1 DW) followed by shoot (5.67 µg g -1 DW) at 60 mg Cr(VI) treatment. The presence and localization of TiO 2 NPs inside the plant leaf cells were confirmed by TEM-EDS analysis. The results revealed that Cr(VI) exposure had a dose-dependent inhibitory effects on photosynthetic attributes, structure of guard and epidermal cells, photosynthetic pigments; inducing impacts on H 2 O 2 and MDA productions, DNA damage, AsA-GSH cycle, and most importantly on PC 2 , and PC 3 synthesis which is rarely reported. However, TiO 2 NPs exposure minimized Cr(VI) induced toxicity through reduction of total chromium accumulation, H 2 O 2 and MDA productions, thereby reducing DNA damage reported first time under combined treatment of Cr(VI)+ TiO 2 NPs as evidenced through comet assay. It also positively regulate the photosynthetic pigments, AsA-GSH cycle, and modulates PC 2 and PC 3 synthesis which have crucial impacts on ROS quenching and Cr(VI) detoxification, respectively, and in turn, minimizes Cr(VI) toxicity in H. annuus L. The mitogen-activated protein kinase signaling cascade is conserved across eukaryotes, where it plays a critical role in the regulation of activities including proliferation, differentiation, and stress responses. This pathway propagates external stimuli through a series of phosphorylation events, which allows external signals to influence metabolic and transcriptional activities. Within the cascade, MEK, or MAP2K, enzymes occupy a molecular crossroads immediately upstream to significant signal divergence and cross-talk. One such kinase, MAP2K7, also known as MEK7 and MKK7, is a protein of great interest in the molecular pathophysiology underlying pediatric T cell acute lymphoblastic leukemia (T-ALL). Herein, we describe the rational design, synthesis, evaluation, and optimization of a novel class of irreversible MAP2K7 inhibitors. Novel drugs are needed to increase treatment response in children with high-risk T-cell acute lymphoblastic leukemia (T-ALL). Following up on our previous report on the activation of the MAP2K7-JNK pathway in pediatric T-ALL, here we demonstrate that OTSSP167, recently shown to inhibit MAP2K7, has antileukemic capacity in T-ALL. OTSSP167 exhibited dose-dependent cytotoxicity against a panel of T-ALL cell lines with IC50 in the nanomolar range (10-50 nM). OTSSP167 induces apoptosis and cell cycle arrest in T-ALL cell lines, associated at least partially with the inhibition of MAP2K7 kinase activity and lower activation of its downstream substrate, JNK. Other leukemic T-cell survival pathways, such as mTOR and NOTCH1 were also inhibited. Daily intraperitoneal administration of 10 mg/kg OTSSP167 was well tolerated, with mice showing no hematological toxicity, and effective at reducing the expansion of human T-ALL cells in a cell-based xenograft model. The same dosage of OTSSP167 efficiently controlled the leukemia burden in the blood, bone marrow, and spleen of 3 patient-derived xenografts, which resulted in prolonged survival. OTSSP167 exhibited synergistic interactions when combined with dexamethasone, L-asparaginase, vincristine, and etoposide. Allium cepa L. is an important medicinal and food plant enormously affected by salinity in terms of its growth and quality. This experiment investigates ameliorative potential of NO donor sodium nitroprusside (SNP) on chromosomal aberrations and physiological parameters in A. cepa L. roots exposed to salinity stress. Roots with different concentrations of NaCl (25, 50, and 100 mM) alone, and in combination with 100 µM SNP were analyzed for mitotic aberrations, DNA damage, proline, malondialdehyde (MDA) content, and ascorbate-glutathione (AsA-GSH) cycle after 120 h of salinity treatments. Results revealed that salinity stress increased chromosomal aberrations, MDA, proline accumulation, and severely hampered the AsA-GSH cycle function. The comet assay revealed a significant ( p ≤  0.05) enhancement in tail length (4.35 ± 0.05 µm) and olive tail moment (3.19 ± 0.04 µm) at 100 mM NaCl exposure. However, SNP supplementation decreased total percent abnormalities, while increased the prophase, metaphase, anaphase, and telophase indexes. Moreover, ascorbate peroxidase and glutathione reductase activities increased with AsA/DHA and GSH/GSSG ratios, respectively. Results suggest that SNP supplementation alleviates salinity stress responses by improving AsA-GSH cycle and proline accumulation. Based on present findings, NO supplementation could be recommended as a promising approach for sustainable crop production under salinity stress.

Fetal Alcohol Spectrum Disorders (FASDs) are comprised of developmental, behavioral, and cognitive abnormalities caused by prenatal alcohol exposure, affecting an estimated 2%-5% of children and costing $4 billion annually in the United States. While some behavioral therapies help, the neurobiological mechanisms that underpin FASDs need further elucidation for development of effective pharmacotherapeutics. The role of the tau protein in the hippocampus is likely to be involved. Tau catalyzes microtubule polymerization in developing neurons. However, this function can become disrupted by hyperphosphorylation. Many of the cognitive deficits observed in neurodegenerative tauopathies overlap to some degree with what is observed in juvenile developmental disabilities, such as FASDs (e.g., selective memory, executive dysfunction). Thus, tau protein phosphorylation may be one important mechanism of dysfunction in FASDs. The purpose of this study is to provide an empirical basis for a tauopathic characterization of FASDs. To do so, hippocampal slices were extracted from rats at postnatal day 10 (PND10); hippocampal slices were then exposed to 5 days of 50-mM ethanol between 6 days in vitro (DIV) and 11DIV. Immunoblots were taken for Total and p-Tau (Threonine231) at 12DIV and 24DIV. Immunohistochemical fluorescent images were taken for p-Tau (Threonine231) at 12DIV and 24DIV. Separate p-Tau measures were taken for the cornu ammonis 1 (CA1), CA3, and dentate gyrus (DG). Total Tau protein expression remained unchanged between 12DIV and 24DIV regardless of ethanol condition. In the control group, longer DIV was associated with decreased p-Tau. However, in the ethanol-exposed group, p-Tau was sustained across DIV. This is the first study to show that ethanol exposure sustains tau Threonine231 phosphorylation in the perinatal hippocampus regardless of Total Tau expression. Acute myeloid leukemia (AML) is an aggressive malignancy of the bone marrow with 5-year overall survival of less than 10% in patients over the age of 65. Limited progress has been made in the patient outcome because of the inability to selectively eradicate the leukemic stem cells (LSC) driving the refractory and relapsed disease. Herein, we investigated the role of the reprogramming factor KLF4 in AML because of its critical role in the self-renewal and stemness of embryonic and cancer stem cells. Using a conditional Cre-lox Klf4 deletion system and the MLL-AF9 retroviral mouse model, we demonstrated that loss-of-KLF4 does not significantly affect the induction of leukemia but markedly decreased the frequency of LSCs evaluated in limiting-dose transplantation studies. Loss of KLF4 in leukemic granulocyte-macrophage progenitors (L-GMP), a population enriched for AML LSCs, showed lessened clonogenicity and percentage in the G2/M phase of the cell cycle. RNAseq analysis of purified L-GMPs revealed decreased expression of stemness genes and MLL-target genes and upregulation of the RNA sensing helicase DDX58. However, silencing of DDX58 in KLF4 knockout leukemia indicated that DDX58 is not mediating this phenotype. CRISPR/Cas9 deletion of KLF4 in MOLM13 cell line and AML patient-derived xenograft cells showed impaired expansion in vitro and in vivo associated with a defective G2/M checkpoint. Contamination of agricultural soil by chromium (Cr) is a serious menace to environmental safety and global food security. Although potential of salicylic acid (SA) in mitigating heavy metal (HM) toxicity in plants is well recognized, detailed physiological mechanisms behind such beneficial effects under Cr-stress in tomato ( Solanum lycopersicum L.) plant are far from being completely unravelled. The present study evaluated the efficacy of exogenously applied SA, in alleviating Cr-mediated alterations on photosynthesis and antioxidant defense in tomato exposed to three different concentrations of Cr(VI) [0, 50, and 100 mg Cr(VI) kg -1 soil]. Exposure of tomato plants to Cr resulted in increased Cr-accumulation and oxidative damage, as signposted by high Cr concentration in root as well as shoot, augmented malondialdehyde (MDA) and superoxides levels, and inhibition in enzymes of ascorbate-glutathione (AsA-GSH) cycle. Furthermore, a significant ( P  ≤ 0.05) reduction in photosynthetic pigments and gas exchange parameters was also evident in Cr-stressed tomato plants. Findings of the present study showed that exogenous application of 0.5 mM SA not only promoted plant growth subjected to Cr, but also restored Cr-mediated disturbances in plant physiology. A significant ( P  ≤ 0.05) decrease in Cr acquisition and translocation as evidenced by improved growth and photosynthesis in SA-treated plants was observed. Additionally, exogenous SA application by virtue of its positive effect on efficient antioxidant system ameliorated the Cr-mediated oxidative stress in tomato plants as signposted by lower MDA and superoxide levels and improved AsA-GSH cycle. T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive pediatric leukemia with a worse prognosis than most frequent B-cell ALL due to a high incidence of treatment failures and relapse. Our previous work showed that loss of the pioneer factor KLF4 in a NOTCH1-induced T-ALL mouse model accelerated the development of leukemia through expansion of leukemia-initiating cells and activation of the MAP2K7 pathway. Similarly, epigenetic silencing of the KLF4 gene in children with T-ALL was associated with MAP2K7 activation. Here, we showed the small molecule 5Z-7-oxozeaenol (5Z7O) induces dose-dependent cytotoxicity in a panel of T-ALL cell lines mainly through inhibition of the MAP2K7-JNK pathway, which further validates MAP2K7 as a therapeutic target. Mechanistically, 5Z7O-mediated apoptosis was caused by the downregulation of regulators of the G2/M checkpoint and the inhibition of survival pathways. The anti-leukemic capacity of 5Z7O was evaluated using leukemic cells from two mouse models of T-ALL and patient-derived xenograft cells generated using lymphoblasts from pediatric T-ALL patients. Finally, a combination of 5Z7O with dexamethasone, a drug used in frontline therapy, showed synergistic induction of cytotoxicity. The present study was performed to address how Cr(VI) posed its toxicities on photosynthesis, lipid peroxidation, and its retaliation by antioxidative system of Helianthus annuus L. during Cr(VI) accumulation. For this, a pot experiment was performed wherein three different concentrations viz, 15, 30, and 60 mg Cr(VI) kg -1 soil were applied to Helianthus annuus L. at the time of seeds sowing. The results revealed that Cr(VI) accumulation was two to three folds higher in roots than in shoots which suggests that root is the major site for Cr(VI) accumulation. It was observed that with increasing doses of Cr(VI), growth indices hampered significantly, along with closure of stomata and damaged guard and epidermal cells. Photosynthetic pigments (chlorophyll a , chlorophyll b, carotenoids), leaf gaseous exchange parameters (A, E, GH 2 O), and PSII efficiency (F v /F m ) worsened under Cr(VI) toxicity in dose dependent manner. Cr(VI) accumulation intensified the lipid peroxidation, too by triggering the MDA and H 2 O 2 production, however, the plant responded well against the lipid peroxidation by enhancing the coordinated action of enzymatic (SOD, APX, GR) and non-enzymatic (GSH, AsA) antioxidants. In a nutshell, Helianthus annuus L. could be used as a potential Cr(VI) accumulator because of its good tolerance strategies against Cr(VI) toxicities. Importance: Lung cancer is the second most common cancer and the leading cause of cancer death in the US. In 2020, an estimated 228 820 persons were diagnosed with lung cancer, and 135 720 persons died of the disease. The most important risk factor for lung cancer is smoking. Increasing age is also a risk factor for lung cancer. Lung cancer has a generally poor prognosis, with an overall 5-year survival rate of 20.5%. However, early-stage lung cancer has a better prognosis and is more amenable to treatment., Objective: To update its 2013 recommendation, the US Preventive Services Task Force (USPSTF) commissioned a systematic review on the accuracy of screening for lung cancer with low-dose computed tomography (LDCT) and on the benefits and harms of screening for lung cancer and commissioned a collaborative modeling study to provide information about the optimum age at which to begin and end screening, the optimal screening interval, and the relative benefits and harms of different screening strategies compared with modified versions of multivariate risk prediction models., Population: This recommendation statement applies to adults aged 50 to 80 years who have a 20 pack-year smoking history and currently smoke or have quit within the past 15 years., Evidence Assessment: The USPSTF concludes with moderate certainty that annual screening for lung cancer with LDCT has a moderate net benefit in persons at high risk of lung cancer based on age, total cumulative exposure to tobacco smoke, and years since quitting smoking., Recommendation: The USPSTF recommends annual screening for lung cancer with LDCT in adults aged 50 to 80 years who have a 20 pack-year smoking history and currently smoke or have quit within the past 15 years. Screening should be discontinued once a person has not smoked for 15 years or develops a health problem that substantially limits life expectancy or the ability or willingness to have curative lung surgery. Acute myeloid leukemia (AML) is an aggressive hematological malignancy of the bone marrow that affects mostly elderly adults. Alternative therapies are needed for AML patients because the overall prognosis with current standard of care, high dose chemotherapy and allogeneic transplantation, remains poor due to the emergence of refractory and relapsed disease. Here, we found expression of the transcription factor KLF4 in AML cell lines is not silenced through KLF4 gene methylation nor via proteasomal degradation. The deletion of KLF4 by CRISPR-CAS9 technology reduced cell growth and increased apoptosis in both NB4 and MonoMac-6 cell lines. Chemical induced differentiation of gene edited NB4 and MonoMac6 cells with ATRA and PMA respectively increased apoptosis and altered expression of differentiating markers CD11b and CD14. Transplantation of NB4 and MonoMac-6 cells lacking KLF4 into NSG mice resulted in improved overall survival compared to the transplantation of parental cell lines. Finally, loss-of-KLF4 did not alter sensitivity of leukemic cells to the chemotherapeutic drugs daunorubicin and cytarabine. Background: End-of-life spending and healthcare utilization among older adults with COPD have not been previously described., Methods: We examined data on Medicare beneficiaries aged 65 years or older with chronic obstructive pulmonary disease (COPD) who died during the period of 2013-2014. End-of-life measures were retrospectively reviewed for 2 years prior to death. Hospital referral regions (HRRs) were categorized into quintiles of age-sex-race-adjusted overall spending during the last 2 years of life. Geographic quintile variation in spending and healthcare utilization was examined across the continuum., Results: We investigated data on 146,240 decedents with COPD from 306 HRRs. Age-sex-race-adjusted overall spending per decedent during the last 2 years of life varied significantly nationwide ($61,271±$11,639 per decedent; range: $48,288±$3,665 to $79,453±$9,242). Inpatient care accounted for 40.2% of spending ($24,626±$6,192 per decedent). Overall, 82%±4% of decedents were admitted to the hospital for 13.7±3.1 days, and 55%±11% were admitted to an intensive care unit for 5.4±2.5 days. Compared with HRRs in the lowest spending quintile, HRRs in the highest spending quintile had a 1.5-fold longer hospital length of stay. Skilled nursing facilities accounted for 11.6% of spending ($7101±$2403 per decedent), and these facilities were utilized by 38%±7% of decedents for 18.7±4.9 days. Hospice accounted for 10.3% of spending ($6,307±$2,201 per decedent) and was utilized by 47%±9% of decedents for 39.7±14.8 days. Significant geographic variation in hospice utilization existed nationwide., Conclusions: End-of-life spending and healthcare utilization among older adults with COPD varied substantially nationwide. The present study unravels origin of nitric oxide (NO) and the interaction between 24-Epibrassinolide (EBL) and nitrate reductase (NR) for NO production in Indian mustard (Brassica juncea L.) under salinity stress. Two independent experiments were performed to check whether (i) Nitrate reductase or Nitric oxide synthase takes part in the biosynthesis of endogenous NO and (ii) EBL has any regulatory effect on NR-dependent NO biosynthesis in the alleviation of salinity stress. Results revealed that NR-inhibitor tungstate significantly (P ≤ 0.05) decreased the NR activity and endogenous NO content, while NOS inhibitor l-NAME did not influence NO biosynthesis and plant growth. Under salinity stress, inhibition in NR activity decreased the activities of antioxidant enzymes, increased H 2 O 2 , MDA, protein carbonyl content and caused DNA damage, implying that antioxidant defense might be related to NO signal. EBL supplementation enhanced the NR activity but did not influence NOS activity, suggesting that NR was involved in endogenous NO production. EBL supplementation alleviated the inhibitory effects of salinity stress and improved the plant growth by enhancing nutrients, photosynthetic pigments, compatible osmolytes, and performance of AsA-GSH cycle. It also decreased the superoxide ion accumulation, leaf epidermal damages, cell death, DNA damage, and ABA content. Comet assay revealed significant (P ≤ 0.05) enhancement in tail length and olive tail moment, while flow cytometry did not showed any significant (P ≤ 0.05) changes in genome size and ploidy level under salinity stress. The present experiment unravels how exogenous jasmonic acid regulates photosynthesis, clastogenecity, AsA-GSH cycle and phytochelatins in Brassica juncea L. in response to Pb-subcellular distribution. The plants were evaluated for leaf gas exchange parameters, Fv/Fm, lipid peroxidation, leaf epidermal structures and ABA content. Besides lead accumulation in root, shoot and its subcellular distribution pattern, its role as clastogen and/or aneuploidogen via DNA damage, genome size and ploidy variations, AsA-GSH cycle and quantification of PC 2 and PC 3 were performed as well. Results revealed that Pb inhibited plant growth, disturbed epidermal and guard cells and consequently worsen leaf gas exchange parameters (E, GH 2 O, A), F v /F m and photosynthetic pigments. For clastogenecity, results revealed considerable DNA damage and analysis for genome size showed that differences between unstressed, Pb-stress and JA application were not significant (P ≤ 0.05), however, ploidy ratio analysis proved partial aneuploidogenic role of Pb. The highest Pb exposure affected AsA-GSH cycle negatively but increased PC 2 and PC 3 contents uniformly in roots and leaves.
