1. VLT observations of the Central Compact Object in the Vela Jr. supernova remnant

Abstract

X-ray observations have unveiled the existence of enigmatic point-like sources at the center of young (a few kyrs) supernova remnants. These sources, known as Central Compact Objects (CCOs), are thought to be neutron stars produced by the supernova explosion, although their X-ray phenomenology makes them markedly different from all the other young neutron stars discovered so far.The aim of this work is to search for the optical/IR counterpart of the Vela Junior CCO and to understand the nature of the associated Halpha nebula discovered by Pellizzoni et al. (2002).}{We have used deep optical (R band) and IR (J,H,Ks bands) observations recently performed by our group with the ESO VLT to obtain the first deep, high resolution images of the field with the goal of resolving the nebula structure and pinpointing a point-like source possibly associated with the neutron star.Our R-band image shows that both the nebula's flux and its structure are very similar to the Halpha ones, suggesting that the nebula spectrum is dominated by pure Halpha line emission. However, the nebula is not detected in our IR observations, whick makes it impossible to to constrain its spectrum. A faint point-like object (J>22.6, H~21.6, Ks ~ 21.4) compatible with the neutron star's Chandra X-ray position is detected in our IR images (H and Ks) but not in the optical one (R > 25.6), where it is buried by the nebula background. The nebula is most likely a bow-shock produced by the neutron star motion through the ISM or, alternatively, a photo-ionization nebula powered by UV radiation from a hot neutron star., Comment: 8 pages, 4 figures, A&Aaccepted

Abstract

Macrophages respond to unmethylated CpG motifs present in nonmammalian DNA. Stabilized phosphorothioate-modified oligodeoxynucleotides (PS-ODN) containing CpG motifs form the basis of immunotherapeutic agents. In this study, we show that PS-ODN do not perfectly mimic native DNA in activation of macrophages. CpG-containing PS-ODN were active at 10- to 100-fold lower concentrations than corresponding phosphodiester ODN in maintenance of cell viability in the absence of CSF-1, in induction of NO production, and in activation of the IL-12 promoter. These enhancing effects are attributable to both increased stability and rate of uptake of the PS-ODN. By contrast, PS-ODN were almost inactive in down-modulation of the CSF-1R from primary macrophages and activation of the HIV-1 LTR. Delayed or poor activation of signaling components may contribute to this, as PS-ODN were slower and less effective at inducing phosphorylation of the extracellular signal-related kinases 1 and 2. In addition, at high concentrations, non-CpG PS-ODN specifically inhibited responses to CpG DNA, whereas nonstimulatory phosphodiester ODN had no such effect. Although nonstimulatory PS-ODN caused some inhibition of ODN uptake, this did not adequately explain the levels of inhibition of activity. The results demonstrate that the phosphorothioate backbone has both enhancing and inhibitory effects on macrophage responses to CpG DNA.

11. 305 SPERM CHROMATIN STRUCTURE AND DNA METHYLATION BULLS

Abstract

Mammalian spermatogenesis is a complex process that involves genome-wide deprogramming and reprogramming of DNA methylation at different stages in order to produce sperm that are capable of fertilization and also TO support ongoing embryo development (Webster et al. 2005 PNAS 102, 4068-4073). Embryonic mortality occurs when there is inappropriate expression of developmentally regulated genes of both maternal and paternal origin. DNA methylation is associated with epigenetic regulation of gene expression and it was recently shown that aberrant DNA methylation causes a change in sperm chromatin structure (Webster et al. 2005). Studies, primarily in man, have indicated that sperm chromatin instability, as determined by the sperm chromatin structure assay (SCSA), does not influence fertilization but is linked with early pregnancy failure (Evenson and Jost 2000 Methods Cell Sci. 22, 169-189). The aim of the present study was to investigate the relationship between chromatin structure and DNA methylation in bull sperm. Chromatin structure was determined using the SCSA that involves the exposure of sperm to a low pH (1.2) detergent solution followed by the addition of acridine orange (AO). When exposed to a 488 nm laser, AO fluoresces green when bound to native (double-stranded) DNA and red when bound to denatured (single-stranded) DNA. The SCSA yields a value for the DNA fragmentation index (DFI), and in men a DFI > 27-30% is associated with early embryonic mortality (Larson-Cook et al. 2003 Fertil. Steril. 80, 895-902). Semen was obtained by electroejaculation from Zebu (Bos indicus) bulls (n = 12) in a subtropical environment; a proportion of bulls had a DFI < 15% (n = 4) and the remainder a DFI > 27% (n = 8). The DNA methylation pattern, as determined by the amplified methylation polymorphism (AMP) protocol (Webster et al. 2005), appeared to differ between sperm with a DFI < 15% and sperm with a DFI > 27% when assessed by principle coordinate analysis. Also, sperm with a DFI < 15% had a more consistent methylation pattern compared with apparent differences in methylation among sperm with a DFI > 27%. These preliminary findings could be interpreted to suggest that methylation status can contribute to chromatin structure in sperm of mature bulls. It remains to be determined whether the environment can influence sperm chromatin status in bulls by altering DNA methylation which then impacts on the conformational changes in DNA that occur during spermiogenesis.This work was supported, in part, by Meat and Livestock Australia.

Abstract

Macrophages respond to unmethylated CpG motifs present in nonmammalian DNA. Stabilized phosphorothioate-modified oligodeoxynucleotides (PS-ODN) containing CpG motifs form the basis of immunotherapeutic agents. In this study, we show that PS-ODN do not perfectly mimic native DNA in activation of macrophages. CpG-containing PS-ODN were active at 10- to 100-fold lower concentrations than corresponding phosphodiester ODN in maintenance of cell viability in the absence of CSF-1, in induction of NO production, and in activation of the IL-12 promoter. These enhancing effects are attributable to both increased stability and rate of uptake of the PS-ODN. By contrast, PS-ODN were almost inactive in down-modulation of the CSF-1R from primary macrophages and activation of the HIV-1 LTR. Delayed or poor activation of signaling components may contribute to this, as PS-ODN were slower and less effective at inducing phosphorylation of the extracellular signal-related kinases 1 and 2. In addition, at high concentrations, non-CpG PS-ODN specifically inhibited responses to CpG DNA, whereas nonstimulatory phosphodiester ODN had no such effect. Although nonstimulatory PS-ODN caused some inhibition of ODN uptake, this did not adequately explain the levels of inhibition of activity. The results demonstrate that the phosphorothioate backbone has both enhancing and inhibitory effects on macrophage responses to CpG DNA.

Abstract

Unmethylated CpG motifs within bacterial DNA constitute a pathogen-associated molecular pattern recognized by the innate immune system. Many of the immunomodulatory functions of bacterial DNA can be ascribed to the ability to activate macrophages and dendritic cells. Here we show stimulatory DNA, like LPS, caused growth arrest of murine bone marrow-derived macrophages proliferating in CSF-1. Stimulatory DNA caused selective down-modulation of CSF-1 receptor surface expression. Flow cytometric analysis of CSF-1-deprived bone marrow-derived macrophages revealed that in contrast to the synchronous reduction of CSF-1 receptor upon CSF-1 addition, activating DNA (both bacterial DNA and CpG-containing oligonucleotide) caused rapid removal of receptor from individual cells leading to a bimodal distribution of surface expression at intermediate times or submaximal doses of stimulus. Despite causing growth arrest, both stimulatory DNA and LPS promoted factor-independent survival of bone marrow-derived macrophages, which was associated with phosphorylation of the mitogen-activated protein kinase family members, extracellular-regulated kinase 1 and 2. CSF-1 receptor down-modulation may polarize the professional APC compartment to the more immunostimulatory dendritic cell-like phenotype by suppressing terminal macrophage differentiation mediated by CSF-1.

Abstract

Murine macrophages are able to distinguish bacterial from mammalian DNA. The response is mimicked by single-stranded oligonucleotides containing unmethylated CG dinucleotides ("CpG" motifs) in specific sequence contexts. The dose-response curve for activation is influenced by variation in the sequence flanking the core CpG motif. CpG or bacterial DNA activates several signaling pathways in common with bacterial lipopolysaccharide (LPS), leading to induction of cytokine genes such as tumor necrosis factor alpha. Pretreatment with LPS causes desensitization to subsequent activation by CpG DNA. Both stimuli also cause cell cycle arrest in macrophages proliferating in response to the macrophage growth factor colony-stimulating factor-1 (CSF-1), but prevent apoptosis caused by growth factor removal. In part, cell cycle arrest by CpG DNA and LPS may be linked to rapid down-modulation of the CSF-1 receptor from the cell surface, a response that occurs in an all-or-nothing manner. The response of macrophages to CpG DNA has aspects in common with the DNA damage response in other cell types, which may provide clues to the underlying mechanism.
