1. A 3D Printed Toolbox for Opto-Mechanical Components

Abstract

Nowadays is very common to find headlines in the media where it is stated that 3D printing is a technology called to change our lives in the near future. For many authors, we are living in times of a third industrial revolution. Howerver, we are currently in a stage of development where the use of 3D printing is advantageous over other manufacturing technologies only in rare scenarios. Fortunately, scientific research is one of them. Here we present the development of a set of opto-mechanical components that can be built easily using a 3D printer based on Fused Filament Fabrication (FFF) and parts that can be found on any hardware store. The components of the set presented here are highly customizable, low-cost, require a short time to be fabricated and offer a performance that compares favorably with respect to low-end commercial alternatives., Comment: 9 pages, 9 figures

Abstract

We present a proof-of-concept experiment aimed at increasing the sensitivity of temperature sensors implemented with Fiber Bragg gratings by making use of a weak value amplification scheme. The technique requires only linear optics elements for its implementation, and appears as a promising method for extending the range of temperatures changes detectable to increasingly lower values than state-of the-art sensors can currently provide. The device implemented here is able to generate a shift of the centroid of the spectrum of a pulse of $\mathrm{\sim 0.035\,nm/^{\circ}C}$, a nearly fourfold increase in sensitivity over the same Fiber Bragg Grating system interrogated using standard methods., Comment: 4 pages, 4 figures

Abstract

It is controversial what is the true role of entanglement in two-photon virtual-state spectroscopy [Saleh et al, Phys. Rev. Lett. 80, 3483, 1998], a two-photon absorption spectroscopic technique that can retrieve information about the energy level structure of an atom or a molecule. The consideration of closely related techniques, such as multidimensional pump-probe spectroscopy [Roslyak et al, Phys. Rev. A 79, 063409, 2009], might suggest that spectroscopic information retrieved in the two-photon absorption process is the same regardless of the classical or quantum nature of the light source. Here, we solve this debate by making use of a full quantum formalism to show that the ability to obtain information about the energy level structure of a medium requires the existence of temporal (frequency) correlations between the absorbed photons. Moreover, we show that these correlations are not the only requisite for retrieving such information. In fact, it is a combination of both, the presence of frequency correlations and its specific spectral shape, which makes the realization of two-photon virtual-state spectroscopy possible. This result helps clarifying the discussion whether entanglement is needed or not, and also, to specify the type of two-photon source that needs to be used in order to experimentally perform the two-photon virtual-state spectroscopy technique.

4. Therapeutic potential of fetal liver cell transplantation in hemophilia A mice

Abstract

Hemophilia A (HA) cell therapy approaches in pediatric individuals require suitable factor (F)VIII-producing cells for stable engraftment. Liver sinusoidal endothelial cells (LSEC) and hematopoietic stem cells (HSC) have been demonstrated to be suitable for the treatment of adult HA mice. However, after transplantation in busulfan (BU)-conditioned newborn mice, adult LSEC/HSC cannot efficiently engraft, while murine fetal liver (FL) hemato/vascular cells from embryonic day 11-13 of gestation (E11-E13), strongly engraft the hematopoietic and endothelial compartments while also secreting FVIII. Our aim was to investigate the engraftment of FL cells in newborn HA mice to obtain a suitable “proof of concept” for the development of a new HA treatment in neonates. Hence, we transplanted FL E11 or E13 cells and adult bone marrow (BM) cells into newborn HA mice with or without BU preconditioning. Engraftment levels and FVIII activity were assessed starting from 6 weeks after transplantation. FL E11-E13+ BU transplanted newborns reached up to 95% engraftment with stable FVIII activity levels observed for 16 months. FL E13 cells showed engraftment ability even in the absence of BU preconditioning, while FL E11 cells did not. BM BU transplanted newborn HA mice showed high levels of engraftment; nevertheless, in contrast to FL cells, BM cells cannot engraft HA newborns in BU non-conditioning regimen. Finally, none of the transplanted mice developed anti-FVIII antibodies. Overall, this study sheds some light on the therapeutic potential of healthy FL cells in the cure of HA neonatal/pediatric patients.

Trabajo presentado en LXIV Congreso Nacional de la SEHH; XXXVIII Congreso Nacional de la SETH y 38th World Congress of the ISH, celebrado en Barcelona (España) del 06 al 08 de octubre de 2022., El ratón presenta unos niveles de factor V muy altos respecto al humano (1) y pudiera jugar un papel importante en el desarrollo embrionario de la especie. Actualmente no ha sido posible generar un modelo animal murino deficiente en dicha proteína, ya que su déficit a nivel embrionario provoca la interrupción de la embriogénesis y muerte prematura de los neonatos (2). En este trabajo se caracteriza la expresión del gen para el factor V (F5) en distintos estadios del desarrollo embrionario, analizando la evolución de la expresión de F5 desde el día 8 (E8) hasta el día 12 (E12) de gestación.

7. Transplantation of fetal liver hemato/vascular cells in newborn mice for correction of hemophilia A (HA)

We have previously shown that hemato/vascular progenitor cells from the foetal liver (FL) possess higher hematopoietic and vascular endothelial engraftment potential when transplanted to busulfan (BU) conditioned newborn mice compared to adult counterpart cells, constituting likely therapeutic targets for the treatment of congenital hemato/vascular related diseases in neonates. Hemophilia A (HA) is a coagulation disorder cause by a deficient production of factor FVIII by vascular endothelial cells and, to a lesser extent, by hematopoietic cells. Since FL cells are endowed with high hematopoietic and endothelial engraftment, we transplanted FL cells into HA new born BU conditioned mice, showing that engrafted FL-derived cells can improve FVIII production and rescued the HA bleeding phenotype. BU constitutes a mild preconditioning treatment compared to other approaches, including irradiation, still it has several side effects that can compromise the health of HA individuals. Therefore, we also explored the long-term correction of HA in unconditioned newborn mice transplanted with FL cells. Here we present the data showing that infusion of FL cells without preconditioning showed a lower hematopoietic engraftment but long term correction of HA (up to 18 months). Infusion of FL cells without preconditioning was able to move the condition of these mice from severe to moderate/mild.

Abstract

The vascular endothelium has several important functions, including hemostasis. The homeostasis of hemostasis is based on a fine balance between procoagulant and anticoagulant proteins and between fibrinolytic and antifibrinolytic ones. Coagulopathies are characterized by a mutation-induced alteration of the function of certain coagulation factors or by a disturbed balance between the mechanisms responsible for regulating coagulation. Homeostatic therapies consist in replacement and nonreplacement treatments or in the administration of antifibrinolytic agents. Rebalancing products reestablish hemostasis by inhibiting natural anticoagulant pathways. These agents include monoclonal antibodies, such as concizumab and marstacimab, which target the tissue factor pathway inhibitor; interfering RNA therapies, such as fitusiran, which targets antithrombin III; and protease inhibitors, such as serpinPC, which targets active protein C. In cases of thrombophilia (deficiency of protein C, protein S, or factor V Leiden), treatment may consist in direct oral anticoagulants, replacement therapy (plasma or recombinant ADAMTS13) in cases of a congenital deficiency of ADAMTS13, or immunomodulators (prednisone) if the thrombophilia is autoimmune. Monoclonal-antibody-based anti-vWF immunotherapy (caplacizumab) is used in the context of severe thrombophilia, regardless of the cause of the disorder. In cases of disseminated intravascular coagulation, the treatment of choice consists in administration of antifibrinolytics, all-trans-retinoic acid, and recombinant soluble human thrombomodulin.

10. Transplantation of Fetal Liver Cells into Newborn Hemophilic Mice for Hemophilia A Cell Therapy

We previously showed that adult-derived FVIIIproducing cells, i.e. liver sinusoidal endothelial cells (LSEC) and hematopoietic stem cells (HSC), can be used for the treatment of adult hemophilia A (HA) mice. However, after transplantation in busulfan-conditioned newborn mice, adult LSEC/HSC cannot ef¿ciently engraft, while murine fetal liver (FL) hemato/vascular cells from day 11-13 of gestation (E11-E13) strongly reconstitute the hematopoietic compartment and showed multiorgan endothelial reconstitution potential while secreting FVIII. [Aims]: To investigate the engraftment of FL cells in newborn HA mice as a new strategy for the treatment of paediatric HA patients. [Methods]: We transplanted FL cells from E11-E13 GFP+ mice into newborn HA mice pre-treated with busulfan (FLE11-E13+BU). Control groups received FLE11-E13 GFP+ cells without busulfan pretreatment (FLE11-E13noBU) or PBS±BU. The engraftment level as well as the FVIII production and activity was assessed in recipients starting from 4 weeks and followed-up for 18 months after transplantation. Moreover, we evaluated the presence of anti-FVIII antibodies/inhibitors in plasma of transplanted mice. [Results]: In FLE11-E13+BU group we observed >60% engraftment in peripheral blood with concomitant FVIII activity (up to 16%) which remained stable up to 18 months after transplantation, while engraftment and FVIII activity were lower in absence of preconditioning. Additionally, FLE11-E13+BU group showed the presence of up to 4% of GFP+ LSEC 18 months after transplantation, demonstrating that transplanted FL cells in BU-conditioned newborn mice contributed in reconstituting the hemato/vascular compartment. Mice of FLE11-E13+BU and E13noBU groups did not develop anti-FVIII antibodies, not even after FVIII immunization. [Conclusions]: FL cells transplantation may provide a novel and highly promising preclinical model for HA treatment, paving the way for studies aiming at deriving long-term reconstituting ¿fetal-like¿ hemato/vascular progenitors from other sources (e.g. iPSC).

We have previously shown that hemato/vascular progenitor cells from the foetal liver (FL) possess higher hematopoietic and vascular endothelial engraftment potential when transplanted to newborn mice compared with adult-derived counterpart progenitor cells (1), constituting likely therapeutic targets for the treatment of congenital hemato/vascular related diseases in neonates. The hemophilia A (HA, a deficiency of factor FVIII) and other rare coagulation disorders, including deficiency of factor FV, are caused by mutations decreasing the production of coagulation factors by hematopoietic and/or vascular endothelial cells. We have shown that different FL-derived cell populations produce FVIII (2), suggesting its suitability as cell therapeutic targets for the treatment of hemophilia diseases. Here we will present the results on the phenotypic and functional characterization of FL progenitors, focusing in three aspects: 1) Characterization of vascular endothelial stem cells during development; 2) Determination of coagulation factor production; 3) Analysis of chimeric HA mice transplanted with wild type FLGFP-derived cells at day 1 after birth. These research should be instrumental for the establishment of a preclinical model for HA treatment in neonates. Abstract

The development of new strategies based on cell therapy approaches to correct haemophilia A (HA) requires further insights into new cell populations capable of producing coagulation factor VIII (FVIII) and presenting stable engraftment potential. The major producers of FVIII in the adult are liver sinusoidal endothelial cells (LSECs) and in a lesser degree bone marrow-derived cells, both of which have been shown to ameliorate the bleeding phenotype in adult HA mice after transplantation. We have previously shown that cells from the foetal liver (FL) and the aorta-gonads-mesonephros (AGM) haematopoietic locations possess higher LSEC engraftment potential in newborn mice compared with adult-derived LSECs, constituting likely therapeutic targets for the treatment of HA in neonates. However, less is known about the production of FVIII in embryonic locations. Quantitative polymerase chain reaction and Western blot analysis were performed to assess the relative level of FVIII production in different embryonic tissues and at various developmental stages, identifying the FL and AGM region from day 12 (E12) as prominent sources of FVIII. Furthermore, FL-derived VE-cad+CD45-Lyve1+/− endothelial/endothelial progenitor cells, presenting vascular engraftment potential, produced high levels of F8 ribonucleic acid compared with CD45+ blood progenitors or Dlk1+ hepatoblasts. In addition, we show that the E11 AGM explant cultures expanded cells with LSEC repopulation activity, instrumental to further understand signals for in vitro generation of LSECs. Taking into account the capacity for FVIII expression, culture expansion and newborn engraftment potential, these results support the use of cells with foetal characteristics for correction of FVIII deficiency in young individuals.

17. Effect of a Filter Cover on Temperature and Humidity in a Mouse Cage

Abstract

To determine how a filter cap affects the heat and moisture build-up in a mouse cage, and how that build-up is affected by ambient conditions of temperature and relative humidity, 50 adult female mice were housed 10 per cage in polycarbonate cages, which were covered with a fibrous filter and sealed with a neoprene gasket and hold-down rod. The cages were placed in a chamber which controlled ambient temperature and humidity. Observations were made at 1°F intervals of temperature (68–74°F, 19.9–23.3°C) and at three different relative humidities (40, 55, and 70 per cent). Every 24 hours the chamber conditions were changed, and cages, bedding, water, filter caps, and food were replaced. Three sensors suspended just under the filter cover measured temperature and humidity and transmitted the data to a recorder. The first reading was taken after a 3-hour equilibraiion period, then every 3 hours until the next day's change. By measuring the average temperature and humidity, it was possible to study the differences between cage and ambient conditions. At 68°F (19.9°C) and 40 per cent relative humidity, the mean conditions in the cages were 72°F (22.2°C) and 50 per cent; at the upper limit of 74°F (23.3 °C) and 70 per cent, the mean cage conditions were 78°F (25.5°C) and 75 per cent.

Abstract

Rabbits were randomly assigned to two groups. Age and body weight distribution were equal in both groups. All animals were placed on a high cholesterol diet for 9 wk, then returned to a normal diet for 1 wk. At the end of this dietary regimen, one group of animals received subcutaneous injections of psysiological saline 3 times/day, 5 days/wk for 10 wk, and the other group recieved L-ascorbate 2-sulfate (0.37 mmole) according to the same timetable. On alternate weeks, the serum levels of total and free cholesterol were determined. After 10 wk of treatment the animals were killed; the plaques were excised from the aortas and examined for total mass and cholesterol content. We observed that, under these dietary conditions, L-ascorbate 2-sulfate does not mobilize cholesterol or its esters from preformed aortic plaque. However, we did observe that animals showing high cholesterol levels in their sera died prematurely when injected with L-ascorbate 2-sulfate. Gross and histopathological investigations of organs and tissues did not reveal any significant differences among the animals that died prematurely and those that survived to the termination of the experiment.
