The response to the COVID-19 pandemic in Timor-Leste offers lessons that may be useful for incorporating into future responses to infectious disease outbreaks in similar resource-limited settings. In this paper, we identify nine key areas for learning from Timor-Leste’s experience of the COVID-19 pandemic: (1) the importance of prior preparation for health emergencies, (2) the establishment of effective leadership and governance structures, (3) the protective impact of early border restrictions, (4) the rapid expansion of diagnostic laboratory capacity, (5) the impact of effective health communications in supporting the vaccine roll-out, (6) the opportunity to build capacity for clinical care, (7) the use of public health interventions that were found to have limited public health impact, (8) the broader effects of the pandemic and the public health response and (9) translation of lessons from COVID-19 to other public health priorities.

En las economías emergentes, la implementación de la Gestión de Infraestructura del Transporte es una necesidad imperante. Con el propósito de utilizar de la forma más eficiente posible los limitados recursos disponibles, la tecnología surge como un aliado estratégico en dicho objetivo. Con el fin de preservar los pavimentos y puentes, diferentes metodologías han sido desarrolladas para la detección de vehículos con sobrepeso. En el presente artículo se presenta la implementación y validación del pesaje de vehículos en movimiento mediante la instrumentación de un puente en servicio ubicado en Costa Rica, con una cantidad reducida de sensores. Se utilizó el enfoque basado en áreas de influencia de ondas de respuesta de deformaciones unitarias. Dada la distribución de los sensores, minimizando los requisitos de instrumentación, se utilizaron dos tipos de deformación unitaria: horizontal y cortante. Dos camiones de calibración fueron utilizados como referencia y luego se procedió a estimar el peso del tráfico aleatorio, para confrontar dichos resultados con los datos de la estación de pesaje estática más cercana.

Existe literatura que demuestra que el pesaje en movimiento con puentes, conocido por sus siglas en inglés como BWIM (Bridge Weigh-In-Motion) es confiable para obtener información acerca de las características de los camiones que transitan por las carreteras. El mejoramiento continuo de estos sistemas presenta oportunidades para incrementar su uso. Los métodos BWIM tradicionales basados en la flexión en vigas enfrentan distintos retos, los cuales han propiciado la aparición de otras metodologías que emplean deformaciones unitarias cortantes. Sin embargo, las técnicas conocidas que utilizan deformaciones unitarias cortantes, asumen o miden líneas de influencia para el cálculo de los pesos brutos vehiculares. En este artículo, se propone una metodología BWIM alternativa, que es independiente de las líneas de influencia, no requiere mediciones de la velocidad de los camiones y se basa en la discontinuidad que se observa en el registro de deformaciones unitarias cortantes, la cual ocurre cuando un camión cruza sobre el lugar en donde se coloca el sensor. A partir de una serie de experimentos realizados en campo, se demuestra que los niveles de error obtenidos con el método propuesto son similares a los obtenidos con otros métodos BWIM más complejos, por lo que se considera que existe potencial para poder ser utilizado para obtener los pesos brutos vehiculares de camiones que transitan por puentes continuos o simplemente apoyados, en una forma consitente y confiable.

Los sistemas de pesaje en movimiento para puentes, en inglés Bridge-Weigh-In-Motion (BWIM), han sido utilizados con éxito para identificar algunas de las propiedades de los vehículos pesados que transitan sobre una vía. No obstante, las fuentes de incertidumbre no son siempre fáciles de controlar y por lo tanto podrían afectar la confiabilidad de los resultados. En esta investigación, se estudió la influencia de algunas de las dificultades experimentales que se presentan al utilizar el método BWIM basado en deformaciones unitarias. En particular, se analizaron tres efectos: el nivel de ruido presente en la señal, la ubicación de los sensores en el puente y la tasa de muestreo utilizada para la recolección de datos. Estos efectos se simularon numéricamente al añadir ruido generado sintéticamente a una onda de respuesta idealizada. En este trabajo se discuten los resultados con énfasis en posibles consecuencias sobre las estimaciones de velocidad y peso de vehículos pesados en pruebas de campo.

We present the analysis of an intronic polymorphism of the nephrin gene and its relationship to the development of diabetic nephropathy in a study of diabetes type 1 and type 2 patients. The frequency of the single nucleotide polymorphism rs#466452 in the nephrin gene was determined in 231 patients and control subjects. The C/T status of the polymorphism was assessed using restriction enzyme digestions and the nephrin transcript from a kidney biopsy was examined. Association between the polymorphism and clinical parameters was evaluated using multivaríate correspondence analysis. A bioinformatics analysis of the single nucleotide polymorphism rs#466452 suggested the appearance of a splicing enhancer sequence in intron 24 of the nephrin gene and a modification of proteins that bind to this sequence. However, no change in the splicing of a nephrin transcript from a renal biopsy was found. No association was found between the polymorphism and diabetes or degree of renal damage in diabetes type 1 or 2 patients. The single nucleotide polymorphism rs#466452 of the nephrin gene seems to be neutral in relation to diabetes and the development of diabetic nephropathy, and does not affect the splicing of a nephrin transcript, in spite of a splicing enhancer site.

Diabetic nephropathy (DN) is one of the major complications of type 2 diabetes and is associated with coronary disease. Nephrin, a protein mainly expressed in glomeruli, is decreased in DN and other kidney diseases. Since insulin levels are misregulated in type 2 diabetes, a possible connection between DN and its decreased nephrin expression could be the presence of regulatory elements responsive to insulin in the nephrin gene (NPHS1) promoter region. In this work, using bioinformatic tools, we identified a purine-rich GAGA element in the nephrin gene promoter and conducted a genomic study in search of the presence of polymorphisms in this element and its possible association with DN in type 2 diabetic patients. We amplified and sequenced a 514 bp promoter region of 100 individuals and found no genetic variants in the purine-rich GAGA-box of the nephrin gene promoter between groups of patients with diabetes type 2 with and without renal and coronary complications, control patients without diabetes and healthy controls

In this report we describe the isolation and characterization of a gene encoding the transcription factor Acel (Activation protein of cup 1 Expression) in the white rot fungus Phanerochaete chrysosporium. Pc-acel encodes a predicted protein of 633 amino acids containing the copper-fist DNA binding domain typically found in fungal transcription factors such as Acel, Macl and Haal from Saccharomyces cerevisiae. The Pc-acel gene is localized in Scaffold 5, between coordinates 220841 and 222983. A S. cerevisiae acel null mutant strain unable to grow in high-copper medium was fully complemented by transformation with the cDNA of Pc-acel. Moreover, Northern blot hybridization studies indicated that Pc-acel cDNA restores copper inducibility of the yeast cup 1 gene, which encodes the metal-binding protein metallothionein implicated in copper resistance. To our knowledge, this is first report describing an Acel transcription factor in basidiomycetes

BACKGROUND: Paper mulberry has been used for thousands of years in Asia and Oceania for making paper and bark-cloth, respectively. Museums around the world hold valuable collections of Polynesian bark-cloth. Genetic analysis of the plant fibers from which the textiles were made may answer a number of questions of interest related to provenance, authenticity or species used in the manufacture of these textiles. Recovery of nucleic acids from paper mulberry bark-cloth has not been reported before. METHODOLOGY: We describe a simple method for the extraction of PCR-amplifiable DNA from small samples of contemporary Polynesian bark-cloth (tapa) using two types of nuclear markers. We report the amplification of about 300 bp sequences of the ITS1 region and of a microsatellite marker. CONCLUSIONS: Sufficient DNA was retrieved from all bark-cloth samples to permit successful PCR amplification. This method shows a means of obtaining useful genetic information from modern bark-cloth samples and opens perspectives for the analyses of small fragments derived from ethnographic materials.

A Ordem Dominicana implantou-se no Brasil a partir de 1881,em resposta ao apelo formulado pelo então bispo de Goiás, D. Cláudio José Gonçalves Ponce de León. Isto condicionou fortemente a presença dominicana no Brasil em sua etapa inicial, restringindo-a aos limites da Diocese de Goiás, vasto território escassamente habitado, estendendo-se por todo o sertão goiano (Planalto Central brasileiro), que incluía, além da então província de Goiás, o chamado Triângulo Mineiro, hoje pertencente a Minas Gerais. Isso permite caracterizar como "fase sertaneja" essa etapa inicial da presença dominicana no Brasil.
