Current trends in the application of bioindication methods are related to the use of submersible tools that perform real‐time measurements directly in the studied aquatic environment. The methods based on the registration of changes in the behavioral responses of zooplankton, in particular Crustaceans, which make up the vast majority of the biomass in water areas, seem quite promising. However, the multispecies composition of natural planktonic biocenoses poses the need to consider the potential difference in the sensitivity of organisms to pollutants. This paper describes laboratory studies of the phototropic response of plankton to attracting light. The studies were carried out on a model natural community that in equal amounts includes Daphnia magna, Daphnia pulex, and Cyclops vicinus, as well as on the monoculture groups of these species. The phototropic response was initiated by the attracting light with a wavelength of 532 nm close to the local maximum of the reflection spectrum of chlorella microalgae. Standard potassium bichromate was used as the model pollutant. The largest phototropic response value is registered in the assemblage. The concentration growth rate of crustaceans in the illuminated volume was 4.5 ± 0.3 ind (L min)−1. Of the studied species, the phototropic response was mostly expressed in Daphnia magna (3.7 ± 0.4 ind (L min)−1), while in Daphnia pulex, it was reduced to 2.4 ± 0.2 ind (L min)−1, and in Cyclops vicinus, it was very small—0.16 ± 0.02 ind (L min)−1. This is caused by peculiar trophic behavior of phyto‐ and zoophages. The addition of a pollutant, namely potassium bichromate, caused a decrease in the concentration rate of crustaceans in the attracting light zone, while a dose‐dependent change in phototropic responses was observed in a group of species and the Daphnia magna assemblage. The results of laboratory studies showed high potential of using the phototropic response of zooplankton to monitor the quality of its habitat thus ensuring the early diagnostics of water pollution. Besides, the paper shows the possibility of quantifying the phototropic response of zooplankton using submersible digital holographic cameras (DHC)., This paper describes laboratory studies of the phototropic response of plankton to attracting light. Ecological monitoring requires sensitive and effective approaches to detecting polluting substances. One such approach involves tracking the behavioral responses of plankton that is initially adapted to normal habitat conditions. Zooplankton such as Daphnia are filter feeders that are highly sensitive to pollution. We describe how a submersible holographic camera with additional attracting lighting (at a wavelength of 532 nm) can be used to track changes of Daphnia response to light for a number of pollutants, including potassium dichromate and crude oil. Our laboratory experiments showed that the sensitivity of this method of bioanalysis is comparable to standard laboratory methods, but has the additional advantage that it can be used in real time in lakes and other water areas, and hence is more versatile than standard methods.

To better understand freezing - thawing cycles operating in peat soils of permafrost landscapes, we experimentally modelled bi-directional freezing and thawing of the three sections of 90-cm long peat core collected from a discontinuous permafrost zone in western Siberia. We measured translocation of microorganisms and changes in porewater chemistry (pH, UV absorbance, dissolved organic carbon (DOC), and major and trace element concentrations) after thawing and two-way freezing of peat cores. We demonstrate that bi-directional freezing and thawing of a peat core is capable of strongly modifying the vertical pattern of bacteria, DOC, nutrients, and trace element concentrations. Sizeable enrichment (a factor of 2 to 5) of DOC, macro- (P, K, Ca) and micro-nutrients (Ni, Mn, Co, Rb, B) and some low-mobile trace elements in several horizons of ice and peat porewater after freeze/thaw experiment may stem from physical disintegration of peat particles, leaching of peat constituents and opening of isolated (non-connected) pores during freezing front migration. However, due to the appearance of multiple maxima of element concentration after a freeze-thaw event, the use of peat ice chemical composition as environmental archive for paleo-reconstructions is unwarranted.

This study shows that the use of a submersible digital holographic camera as part of a multifunctional hardware and software complex allows carrying out in situ measurements of plankton, automating the process of obtaining data on plankton, as well as classifying plankton species up to an order within the specified taxonomic groups. Such automation ensures monitoring expeditionary or stationary research of species diversity and spatial and temporal organization of zooplankton in conjunction with the hydrophysical parameters of the medium. This paper presents the full-scale results of vertical profiles and daily measurements of plankton made with the use of the submersible digital holographic camera as well as the classification of plankton in laboratory and field conditions in the automatic mode. It is shown that, within the accomplished version, the classification algorithm using the morphological parameter makes it possible to solve the problem quickly (the time required to obtain the result is less than 1 s and depends on the number of plankton particles and the frame size of a restored image); however, the classification accuracy by orders varies within 50–60%.

Biological effects and ecotoxicity of NPs CeO2 with particle size Δ50 =16 nm and hydrobionts tolerance was measured in simulation based on test-reactions of a group of organisms representing major trophic levels of aquatic ecosystem. Biological activity of NPs CeO2 was noted against bacterial biosensor Ekolum, Сhlorella v. B.,Parameciumc., Daphniam. S. and Danior. Toxicity and hazard level of NPs CeO2 were measured using biotesting and hydrobionts. Toxicity of NPs CeO2 against luminescent bacteria Ekolum and Parameciumc. and Danior. was not established (L(E)С50 > 100 mg/l). In accordance with SGS and EC 93/67/EEC, NPs CeO2 is considered highly toxic (acute toxicity Level 1). Biomarkers and NPs CeO2 tolerant hydrobionts were determined. Range of tolerance of hydrobionts to NPs was established. The upper limit of tolerance was determined for certain hydrobionts by values of L(E)С50: 0.009 mg/l (by volume of Chl a) for Сhlorella v. B. and 46.15 mg/l (by mobility) for Daphniam. S.

The research studies concentration effects and influence of nCeO2, nZnO, nNi and nPt on photosynthetic pigments in Chlorella v. B. in conditions of homeostated lab cultivation. It was shown that dependency of the content of chlorophylls and carotenoids in chlorella cells on concentrations of nCeO2, nZnO and nPt has non-linear dual-phase character. Growing concentrations of nNi in the culture solution caused monotone decrease of all photosynthetic pigments in chlorella cells. Adding 0.1 mg/L nZnO in the suspension caused a statistically significant increase of the chlorophyll a and b content (27.1 % and 64.2 % respectively) in comparison with the control sample. NPs Pt at 1 mg/L induced stimulating effect on chlorophylls a (17.4 % compared to the control) and b (23.6 % compared to the control) in chlorella cells. The number of carotenoids had a statistically significant decrease in chlorella cells by 25-70 % at high concentrations (1-10 mg/L) for all tested NPs.

The paper is devoted to studying the stability of of dispersed systems (DS) of nanoparticles (NP) of nickel (nNi), platinum (nPt), titanium dioxide (nTiO2), zinc oxide, multicomponent nanopowder (MNP) and multi-walled carbon nanotubes (MWCNT), formed basing on the growth media used in bioassay with application of aquatics: bacterial test-cultures Ecolum, Photobacterium phosphoreum and Escherihia coli, unicellular algae Chlorella vulgaris Beijer, infusoria Paramecium caudatum, cladocera Daphnia magna Straus and Ceriodaphnia affinis, fish Danio rerio. In the process of establishing a test-system the increase in the initial nanosubstance concentration of more than 5 mg/l results in the increase in the speed of aggregation of NP into DS and the decrease in their final concentration. We suggest the variants of estimation of effective concentration of NP in long-term biotesting.

The microbial activity in the soils of the permafrost-affected zones is assumed to be one of the major factors that modify the organic carbon and nitrogen cycle under current climate change. In contrast to the extensive research centered on bacterial abundance, diversity, and metabolic activity in permanently and seasonally frozen mineral soils from high latitudes, frozen peat (organic) environments remain poorly characterized in terms of the physiological diversity and metabolic potential of bacteria. The evolution of soil heterotroph microbial number and metabolic activity across the “seasonally thawed (active)—permanently frozen layer” boundary was studied on 100-cm-thick cores from frozen peat mounds located in the discontinuous permafrost zone in western Siberia. There was a systematic decrease of metabolic activity in the upper 40 cm of the peat core from the surface layers of the mosses and lichens towards the beginning of the frozen horizon, followed by an abrupt increase in bacterial metabolism exactly at the border between the thawed layer and the permafrost table. The aerobic viable cell count and total bacterial number from the active layer were similar to those from the permafrost peat layer. The highest metabolic activity was observed at the beginning of the frozen peat layer and might correspond to the highest availability of amino substrates, which were depleted in the active layer but preserved in the deeper frozen horizons. The enhanced microbial activity at the frozen peat-active layer boundary in western Siberia may persist for another 50–100 years based on the current rate of increase in active layer thickness.

The biological effects of the poorly soluble nanoparticles (NPs) of different chemical nature and structural characteristics were evaluated. It was established that the Zebrafish test response to contamination of aqueous medium with nickel NPs (nNi), platinum (nPt), zinc oxide (nZnO) and cerium oxide (nCeO2) depends on the physicochemical properties of the NPs and embryo development stage. The concentrations of NPs not causing disruptions in embryonic development of Zebrafish were determined. The smallest impact on embryogenesis was exerted by nCeO2: coagulation of a small number of embryos was observed only at C = 20.0 mg/L. The same effect was observed when exposed to lower concentrations of nPt (C = 5.0 mg/L) and nNi (C = 0.1 mg/L). The greatest number of coagulated embryos was observed when grown in the DS nZnO: 37.5% of embryos died at the DS concentration of C = 0.1 mg/L. Zebrafish cultivation in the DS with low concentrations (C ≤ LC10) of nNi and nZnO caused distortions in the development of embryos: development of scoliosis, malformation of somites, inhibited mobility.

An assessment was performed to estimate ecological and biological effects of metallic and binary NPs of various chemical nature and structural characteristics. Application of rapid toxicity tests using hydrobionts of various trophic levels (E. coli, Сhlorella v. B., Paramecium c., Daphnia m. S., Danio r.) allowed to determine resistance range to NPs Ni, Pt, ZnO and CeO2. It was established that test reaction to contamination of water with NPs and development of adverse effects to phyto- and zooplankton is contingent on NPs physicochemical properties and sensitivity of hydrobionts. Concentrations of NPs not causing observed change of test reactions were determined. Chlorella v. B. was established to be the most vulnerable to contamination of medium with NPs; photosynthetic pigment quantity was determined to be the most sensitive parameter: nCeO2 L(E)С10 = 0.0007 mg/l, nNi L(E)С10 = 0.0015 mg/l, nZnO L(E)С10 = 0.0048 mg/l, nPt L(E)С10 = 0.033 mg/l. The highest resistance (L(E)С10 > 100 mg/l) to contamination shown by: E. Coli, Daphnia m. S., Danio r. tonPt; Danio r. to NNi and nZnO, E. Coli, Paramecium c. to nCeO2; and Danior r. by the parameter ‘embryotoxicity’. The most vulnerable chains of trophic structure of communities were revealed as well as ways of disrupting food pyramid of an aquatic system. Range of resistance or range of concentrations within which biota retains resistance was shown to be an integral characteristic of NPs effects on ecosystems.

Environmental toxicity of aqueous disperse systems of nanoparticles of binary compounds of titanium dioxides (with particle size Δ50=5 nm, Δ50=50 nm, Δ50=90 nm), aluminum oxide alpha-forms (Δ50=7 nm and Δ50=70 nm) and macro forms (TiO2 Δ50=350 nm, Al2O3 A50=4000 nm) were studied using biological testing methods. The bioassay was performed using a set of test organisms representing the major trophic levels. We found the dependence of the toxic effect concentration degree of nTiO2 and nAl2O3 on the fluorescence of the bacterial biosensor "Ekolyum", the chemotactic response of ciliates Paramecium caudatum, the growth of unicellular algae Chlorella vulgaris Beijer and mortality of entomostracans Daphnia magna Straus. We revealed the selective dependence of nTiO2 and nAl2O3 toxicity on the size, concentration and chemical nature of nanoparticles. The minimal concentration causing an organism's response on nTiO2 and nAl2O3 effect depends on the type of the test- organism and the test reaction under study. We specified L(E)C50 and acute toxicity categories for all the studied nanoparticles. We determined that nTiO2 (Δ50=5 nm) belong to the category «Acute toxicity 1», nTiO2 (A50=90 nm) and nAl2O3 (Δ50=70 nm) – to the category «Acute toxicity 2», nAl2O3 (Δ50=7 nm) – to the category «Acute toxicity 3». No acute toxicity was registered for nTiO2 (Δ50=50 nm) and macro form TiO2.

The effect of nanoparticles with the particle size Δ50=5 nm on the test function of aquatic organisms was analyzed by means of biotesting methods with the use of a complex of test-organisms representing general trophic levels. The dependence of an infusoria Paramecium caudatum chemoattractant-elicited response, unicellular algae Chlorella vulgaris Beijer growth rate, Daphnia magna Straus mortality and trophic activity and Danio rerio fish kill due to nNi disperse system concentration, is estimated. It is determined that the release of chlorella into cultivated environment including nNi as a feed for daphnias raises the death rate of entomostracans. The minimal concentration, whereby an organism response to the effect of nNi is registered, depends on the type of test organism and the analysed test function. L(E)C20 is determined for all the organisms used in bioassays. L(E)C50 is estimated for Paramecium caudatum (L(E)C50 = 0.0049 mg/l), for Chlorella vulgaris Beijer (L(E)C50 = 0.529 mg/l), for Daphnia m. S (L(E)C50 > 100 mg/l) and for fish Danio rerio (L(E)C50 > 100 mg/l). According to the Globally Harmonized System hazard substance evaluation criteria and Commission Directive 93/67/EEC, nNi belongs to the "acute toxicity 1" category of toxic substances.

The toxicity of zinc oxide nanoparticles (nZnO) with particle size Δ50 = 20 nm was evaluated according to the degree of toxicity of the aqueous disperse system (DS) with biological testing methods using a set of test organisms representing the major trophic levels.We observed the influence of the concentration degree of nZnO on toxic effects level on the fluorescence of the bacterial biosensor "Ekolyum-13", the chemotactic response of ciliates Paramecium caudatum, the rate of growth of unicellular algae Chlorella vulgaris Bayer, mortality of entomostracans Daphnia magna Straus and fish Danio rerio. The detected values of L(E)C50 are: for biosensor "Ekolyum-13" – 0.30 mg/L, for ciliates Paramecium caudatum – 0.14 mg/L, for Chlorella vulgaris Bayer – 0.17 mg/L and for Daphnia magna Straus – 0.52 mg/L. No toxicity of nZnO was detected in relation to fish Danio rerio, L(E)C50 > 100 mg/L. In assessing the maximum effect of nZnO according to GHS and EU Directive 93/67/ EEC, it is assigned to dangerous substances with a high degree of toxicity "Acute toxicity 1".
