Abstract

Background: The effects of extremely low frequency electromagnetic fields (ELF-EMF) on Toxoplasma gondii have not been explained yet. The aim of this study was to assess the possible effects of ELF-EMF on growth, survival time and viability of Toxoplasma gondii. In addition, the life span of Toxoplasma infected animals was investigated. Methods: Sixty adult male BALB/c mice were used for in vivo and in vivo experiments in Laboratory of Biopyhsics and Parasitology of Medical Faculty, Adnan Menderes University, Turkey, in 2010. During in vivo experiments, pulsed and continuous EMFs were applied for 5 d to the infected mice. During in vivo experiments, pulsed and continuous EMF was applied to the tachyzoites within peritoneal exudates for 8 h/d at 4 °C and the tachyzoites were then injected to mice. In both experiments, the number of T. gondii in peritoneal exudates was counted and T. gondii protein bands patterns were investigated with polyacrylamide gel electrophoresis and Western Blotting. Results: Pulsed and continuous EMF exposure reduced the number of T. gondii tachyzoites in comparison to controls. However, no statistically significant differences were observed at the patterns of protein bands among the samples. Conclusion: EMF exposure induces a decrease in the number of T. gondii. Further studies are required to understand the mechanism of EMF on intracellular parasites.

Abstract

Aim: Sonodynamic antitumor therapy is a promising, novel method for the treatment of cancer. To determine the effects of malachite green (MG) in the presence of ultrasound (US), MG was tested in vitro on HL60 cells at different concentrations as a sonodynamic compound. We investigated cell viability, morphology, and the occurrence of endoplasmic reticulum (ER) stress after MG-mediated sonodynamic therapy (SDT) in HL60 cells. Material and Method: Four groups were formed, including a control group, a group subjected to ultrasound (US) only, a group treated with various concentrations of MG, and a group treated with US using the same concentrations. The cells were treated with 1MHz ultrasound at 2 W/cm2 for 3 minutes. The assessment of cell viability was conducted 24 hours post-treatment through the utilization of the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Cell morphology and apoptotic index were determined using Giemsa staining, while GRP78 and PERK expressions were determined through immunocytochemistry staining. Results: The cell cytotoxicity of HL60 cells significantly increased after MG-mediated sonodynamic therapy. After treatment, apoptotic cells with micronuclei were observed morphologically. Significant levels of GRP78 and PERK expression were observed in all groups, except for PERK expression in the US group, compared to the control group. Conclusion: The induction of ER stress, accompanied by intense apoptosis and a marked decrease in cell viability, demonstrates the potential of MG-mediated sonodynamic therapy in cancer treatment. Abstract

Aim: Our objective is to explore the relationship between the anti-leukemic impact of malachite green-mediated photodynamic therapy (PDT) and the induction of endoplasmic reticulum (ER) stress in acute promyelocytic leukemia cells (HL-60). Material and Method: For one hour the cells were incubated with different concentrations (3.125, 1.56, 0.78, 0.39, 0.195, 0.0975, 0.04875 µM) of malachite green and then were exposed to 0.47 mW/cm2 irradiance and 0.84 J/cm2 fluence for 30 minutes. Also, HL-60 cells were exposed to PDT with light only and both in the presence or absence of malachite green. MTT assay was used to determine cell viability, and immunocytochemical staining was used to detect the expression of ER stress markers Protein Kinase R-like ER Kinase (PERK) and Glucose-regulated protein 78 (GRP78). Results: The cell viability of the treatment group (combination of malachite green and light) was significantly decreased compared to the malachite green, control group, and light control. Moreover, immunocytochemical staining scores showed that PERK and GRP78 were significantly upregulated in the treatment group compared with other groups. Conclusion: Our results indicate that ER stress may contribute to the cytotoxicity occurring in HL-60 cancer cells after malachite green-mediated PDT. Future studies will be crucial in shedding light on the molecular mechanisms underlying ER stress that may occur after PDT. These findings lay the foundation for further investigations in this area. Abstract

Photodynamic therapy is a two-step procedure, involving the use of photosensitizing agents followed by selective illumination of the target lesion with visible light. Photodynamic therapy has been described recently as a promising strategy for treatment of leishmaniasis. This study aims to evaluate the in vitro phototoxic, morphological, and apoptotic effect of methylene blue, toluidine blue, chloro-aluminum phthalocyanine, and pheophorbide a-mediated photodynamic therapy on the viability of Leishmania tropica promastigotes.Parasites were treated with methylene blue, toluidine blue, chloro-aluminum phthalocyanine, and pheophorbide a or/and methylene blue, toluidine blue, chloro-aluminum phthalocyanine, and pheophorbide a-mediated photodynamic therapy, and cell proliferation, morphological changes, and apoptosis were evaluated by XTT, giemsa staining, DAPI staining, and DNA fragmentation, respectively.Parasite viability was significantly different in between the groups treated with methylene blue, toluidine blue, and pheophorbide a, with or without irradiation. chloro-aluminum phthalocyanine treatment did not lead to any alterations in cell viability in Leishmania tropica promastigotes with or without irradiation. DAPI staining results indicated that apoptotic bodies and nucleus fragmentation started to be visible in methylene blue, chloro-aluminum phthalocyanine, and pheophorbide a-mediated photodynamic therapy groups. DNA ladder pattern which is used to define apoptosis was observed in irradiated methylene blue, chloro-aluminum phthalocyanine, and pheophorbide a groups.The results revealed that apoptosis-induced cell death was observed in Leishmania tropica promastigotes after the application of photosensitizers in combination with light irradiation.

Abstract

Leishmaniasis is a common zoonotic disease that is transmitted by phlebotomus and causes several clinical conditions, from self healing lesion to deadly internal organ involvement. Photodynamic therapy (PDT) is a treatment method that leads to the generation of cytotoxic species and consequently to cell death and tissue destruction by visible light in the presence of a photosensitizer and oxygen. The aim of this study was to investigate effect of malachite green (MG)-mediated PDT in Leishmania tropica (L. tropica) promastigotes.Parasites were incubated with 0.19, 0.39, 1.56, 3.25 and 6.25 μM of MG for one hour and subjected to 46.4 J/cmMalachite green mediated photodynamic therapy at 1.56 and 3.125 μM decreased the viability of the L. tropica promastigotes and induced changes in the mitochondrial membrane potential. L.tropica promastigotes was bloked in G0/G1 phase. The morphology of the parasite was affected at the 1.56 and 3.125 μM MG+PDT, resulting in rounded cells with loss of flagellum and irregular shape.This study demonstrated that antileishmanial effects through mitochondrial dysfunction, cell cycle arrest, and apoptosis-like cell death to parasites. This work showed PDT with MG effectedparasites. Therefore, MG-mediated PDT may provide a promising approach for L. tropica promastigotes.

Abstract

WOS: 000465377600009 PubMed ID: 31130125 Cutaneous leishmaniasis (CL) is a parasitic disease transmitted by vector sand flies Phlebotomus and Lutzomyia. This disease is characterized by long time non-healing skin lesions, and caused by Leishmania species. CL is the most common infection in Eastern and Southeastern Anatolia in Turkey and L. tropica is known as the main agent of the disease. Number of cases is increasing in our country in time because of malnutrition, migration, travel, low socioeconomic level and ecological changes. For the treatment, the pentavalent antimonials are often used as intralesionally for many years, and it was reported that resistant cases have increased in recent years. New treatment methods and anti-Leishmanial activity of new agents have been investigated because of side effects, resistance development and toxic reactions of the present drugs. These studies are first carried out in vitro and afterwards with in vivo experimental animal models. Reporter gene technology has been used to investigate a variety of purposes like biological events in microorganisms and the efficacy and resistance of drugs in recent years. The major areas that green fluorescent protein (gfp) used are that they can be incorporated into different genes to determine the amount of expression of these genes in different organisms and can be used as markers in living cells. Especially gfp gene, which encodes the green fluorescent protein, is widely used nowadays. Gene-based assays have several advantages like being easy to follow-up, inexpensive and have improved biosecurity. The aim of the present study was to perform the transfection of L. tropica with "enhanced gfp (egfp)" and in vitro usefulness of gfp-transfectants as a drug screening model in comparison to the conventional methods. Promastigotes of L. tropica were transfected with p6.5/egfp by electroporation and selected for tunicamycin-resistance as previously described. L. tropica promastigotes transfected with gfp and in vitro effect of meglumine animoniate was assessed using different methods such as fluorescence microscopy, fluorometer and XTT (2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxyanilide) assay. The use of gfp-transfected Leishmania strains was found more rapid and more sensitive by fluorescent microscopy and fluorometry than conventional assays for the evaluation of potential anti-leishmanial agents. Consequently, stable gfp-transfected Leishmania species will be used in vitro and in vivo for screening of anti-leishmanial drugs and vaccine development as well as for understanding the biology of the host-parasite interactions at the cellular level. As a result ot this study, gfp transfected model using a Turkish L. tropica isolate was established to be used in further studies.

Abstract

Aim: Acute myeloid leukemia (AML) is a disease characterized by relapse and treatment resistance in most patients. Therefore, there is a need for targeted therapies in AML. Photodynamic therapy (PDT) is a promising alternative for the treatment of malignant tumors. Also, PDT has the potential to be used individually or complementally in the treatment of leukemia. In this study, it was aimed to investigate possible the effect of malachite green (MG)-based PDT on acute myeloid leukemia cells. Materials and Methods: Cells were incubated with 0.19, 0.39, 0.78,1.56, 3.125, and 6.25 µM MG for one hour and irradiated with 46.4 J/cm2 of light. The trypan blue test was used to assess the viability of cells, and the change in mitochondrial activity was determined by MTT. Morphological features were determined by Giemsa staining and scanning electron microscopy. Cell cycle and Annexin V/PI assays (measuring fluorescence emitted by staining reagents) were measured by flow cytometry. Results: With the combination of MG and light, HL60 cell viability was found to be significantly reduced compared to the control group. Giemsa staining and SEM results showed that 3.125 μM MG-based PDT induced various morphological changes in cells typical for apoptosis. Late apoptosis was observed in cells treated with 3.125 μM MG combined PDT according to Annexin/PI staining, further showing that it caused an arrest in the subG1 phase of the cell cycle. Conclusion: MG-based PDT has the potential to inactivate HL60 cells. Thus, MG-based PDT may ensure a promising approach for treating acute myeloid leukemia cells.

Abstract

Objective:There is a need for new treatment options for treating Leishmaniasis, since there is no standard treatment scheme with few side effects. Sonodynamic therapy (SDT) is also a candidate to be one of these options. SDT is a treatment method based on the simultaneous combination of low-intensity ultrasound and a sonosensitizer, and the generation of reactive oxygen species in cells in the presence of molecular oxygen. Sonosensitizer, ultrasound, and molecular oxygen individually, these components are not toxic, but when combined form cytotoxic reactive oxygen species In this study, we evaluated the effect of rose bengal (RB)-mediated SDT on Leishmania tropica (L. tropica) promastigotes.Methods:SDT was performed using different concentrations of RB (20, 40, and 80 μM) and ultrasound at a frequency of 1 MHz with an intensity of 1, 1.5, and 2 W/cm2 for 10, 20, and 30 min.Results:Incubation with different RB concentrations applied alone had no effect on L. tropica promastigotes. Ultrasound application time for L. tropica promastigotes alone was determined as 10 min. Ultrasound application intensity showed more significant results at 2 W/cm2. It was determined that the number of promastigotes was lower than that of the control group after 10 min of exposure to ultrasound at 2 W/cm2 at 1 MHz frequency for 10 min with RB (80 μM). Morphologically, round, swollen, atypical forms of the parasite with indistinguishable nuclei are observed, but typical narrow cell body forms have also been detected.Conclusion:These results showed that RB-mediated SDT on L. tropica could be a candidate treatment approach. This approach can be used for both superficial and deeply located lesions. This study emphasized the biophysical mechanisms, ultrasound exposure strategies, reliability and difficulties in the clinical practice of RB-mediated SDT on L. tropica promastigotes.

Abstract

Chronic Obstructive Pulmonary Disease (COPD) is mainly expressed by weight loss with especially fat-free mass (FFM) depletion and a low body weight correlates with increased mortality and a poor prognosis. We investigated whether anthropometric body composition equations could be used for evaluation of the body composition in COPD. Thirty clinically stable patients with COPD and 13 healthy age matched control subjects underwent the skinfolds and circumference measurements in addition to body mass index (BMI) calculations. Body fat mass (BFM) and FFM were determined by using anthropometric equations. The percent BFM, predicted from body density either using Siri's or Brozek's equations was determined lower than that of calculated with equations by using BMI, age and gender. The values of BFM and body weight were reduced in patients with FEV1% predicted compared to other participants. The FFM values also dropped depending on the severity of COPD. BMI was not statistically different among the participants while FFM index (FFMI) reflected the nutritional status of the disorders. Anthropometric equations easily and cost effectively applied for prediction of % BFM, FFM, and FFMI in patients with COPD. In addition, the FFMI can be possibly used for expressing COPD severity.

Abstract

Objective Blood transfusion therapy for Thalassaemia patients may cause transmission of some infectious agents to the recipients. As Toxoplasma gondii is a transfusion-transmitted parasite, we aimed to investigate anti-Toxoplasma antibodies. Methods We arranged two groups: patients and controls. There were 36 Thalassaemia cases in the patient group. The control group also included 36 healthy people (of the same ages and gender) who were selected by the one-to-one mapping method. Mean age was 18.1 ± 8.3 (4-35). Toxoplasma gondii IgG and IgM antibodies were analyzed by the Enzyme-Linked Immunosorbant Assay (ELISA). Results We determined parasite-specific Toxoplasma gondii IgG antibodies in 7 (19.4%) and borderline levels of IgM antibody in 2 Thalassaemia major cases (5.5%). In the control group, parasite-specific IgG antibody was found in 5 cases (14%). Serum samples of patients with borderline levels were analyzed with an avidity test and were found to have high avidity. There were no significant differences between patient and control groups when considering the presence of anti-Toxoplasma IgG antibodies (p=0.752). Conclusion Although there is no significant statistical difference, we suggest that blood transfusion donors should be investigated for infectious agents such as Toxoplasma gondii.

Abstract

Objective:Echinococcus granulosus is the causative helminth of cystic echinococcosis (CE). The parasite is known to form fluid-filled cysts that grow slowly in the internal organs, particularly the liver and/or lungs. This disease is still important in terms of public health and economically in Turkey and other countries where animal husbandry is widespread. The aim of our study was to retrospectively evaluate the cases that were admitted to the Adnan Menderes University, Training and Research Hospital Parasitology laboratory on suspicion of CE between January 2005 and January 2017.Methods:Totally, 3446 sera (from 2019 female and 1427 male) were tested with an in-house ELISA for the presence of E. granulosus specific IgG antibodies at the timeswhen they were sent. Socio-demographic characteristics (age, gender, residence, and dog ownership), positivity titers, and cyst locations of pathologically confirmed CE patients were analyzed retrospectively.Results:The ages of patients varied between 4-87 years. It was found that 1104 (32%) of the 3446 sera were positive, and of them, 642 (58.1%) were female and 462 (41.9%) were male. Patients who had pathologically confirmed CE diagnosis constituted 247 (22.3%) of the total seropositive sera. Liver was the most commonly affected organ (81.8%), followed by lungs (6.1%).Conclusion:CE remains an important public health problem in our city; therefore, it is once again emphasized that preventive studies should be planned.
