Abstract

Simple Summary: Proteomics can be very useful in identifying proteins, which helps find potential markers for diseases. Managing endometrial cancer can be difficult and finding reliable markers can contribute to an early diagnosis, to manage its evolution, and even predict the response to treatment. This paper reviews the current research on the proteins involved in endometrial cancer. Most studies used tissue, serum, and plasma samples and found potential diagnostic and prognostic markers. Eight studies were examined closely, with three showing strong similarities, sharing forty-five proteins. This review also identified the 10 most commonly reported proteins in these studies. While proteomics shows promise in finding diagnostic and prognostic markers for endometrial cancer, there is still a need for more research on new therapeutic targets. Proteomics can be a robust tool in protein identification and regulation, allowing the discovery of potential biomarkers. In clinical practice, the management of endometrial cancer can be challenging. Thus, identifying promising markers could be beneficial, helping both in diagnosis and prognostic stratification, even predicting the response to therapy. Therefore, this manuscript systematically reviews the existing evidence of the proteomic profile of human endometrial cancer. The literature search was conducted via Medline (through PubMed) and the Web of Science. The inclusion criteria were clinical, in vitro, and in vivo original studies reporting proteomic analysis using all types of samples to map the human endometrial cancer proteome. A total of 55 publications were included in this review. Most of the articles carried out a proteomic analysis on endometrial tissue, serum and plasma samples, which enabled the identification of several potential diagnostic and prognostic biomarkers. In addition, eight articles were analyzed regarding the identified proteins, where three studies showed a strong correlation, sharing forty-five proteins. This analysis also allowed the identification of the 10 most frequently reported proteins in these studies: EGFR, PGRMC1, CSE1L, MYDGF, STMN1, CASP3 ANXA2, YBX1, ANXA1, and MYH11. Proteomics-based approaches pointed out potential diagnostic and prognostic candidates for endometrial cancer. However, there is a lack of studies exploring novel therapeutic targets. Abstract

The increasing cancer incidence has certified oncological management as one of the most critical challenges for the coming decades. New anticancer strategies are still needed, despite the significant advances brought to the forefront in the last decades. The most recent, promising therapeutic approaches have benefitted from the application of human perinatal derivatives (PnD), biological mediators with proven benefits in several fields beyond oncology. To elucidate preclinical results and clinic outcomes achieved in the oncological field, we present a narrative review of the studies resorting to animal models to assess specific outcomes of PnD products. Recent preclinical evidence points to promising anticancer effects offered by PnD mediators isolated from the placenta, amniotic membrane, amniotic fluid, and umbilical cord. Described effects include tumorigenesis prevention, uncontrolled growth or regrowth inhibition, tumor homing ability, and adequate cell-based delivery capacity. Furthermore, PnD treatments have been described as supportive of chemotherapy and radiological therapies, particularly when resistance has been reported. However, opposite effects of PnD products have also been observed, offering support and trophic effect to malignant cells. Such paradoxical and dichotomous roles need to be intensively investigated. Current hypotheses identify as explanatory some critical factors, such as the type of the PnD biological products used or the manufacturing procedure to prepare the tissue/cellular treatment, the experimental design (including human-relevant animal models), and intrinsic pathophysiological characteristics. The effective and safe translation of PnD treatments to clinical practice relies on the collaborative efforts of all researchers working with human-relevant oncological preclinical models. Abstract

Dissertação de Mestrado em Biotecnologia Farmacêutica apresentada à Faculdade de Farmácia Endometrial cancer is the most frequent malignant neoplasm of the female genital tract in Western countries, which affects postmenopausal women and has its peak incidence at the beginning of the sixth decade of life. In general, it presents a favorable prognosis, since the majority of the patients have the diagnosis at an early stage, with disease confined to the uterus. However, up to 20% of endometrial carcinomas present as aggressive neoplasms, such as high-grade lesions or myometrial invasion, with a risk of recurrence. Cancer stem cells (CSC) are characterized as a subpopulation of undifferentiated cancer cells that exhibit self-renewal, differentiation, resistance to conventional therapies and metastatic potential.The aim of this work was the isolation and characterization of an endometrial CSC population and the evaluation of the ALDH pathway inhibition, described as a CSC marker. We intended to study the therapeutic response of CSC to cytostatics used in clinical practice and contribute for clarification of resistance mechanisms. In a translational perspective, using samples from patients with endometrial cancer, it was intended to isolate and evaluate CSC in order to contribute to the development of individualized therapies.The characterization of CSC from endometrial cancer cell lines comprised isolation of sphere-forming populations, evaluation of self-renewal and expression of typical CSC markers (by flow cytometry and Western blot). The influence of ALDH inhibitors on this phenotype was also evaluated. The response to carboplatin, paclitaxel and doxorubicin was evaluated in terms of metabolic activity, expression of proteins involved in resistance to therapy and cell death, as well as DNA damage.The sphere formation protocol was also applied to human tumor samples in order to isolate and characterize CSC population. In parallel, tumor sections were prepared and exposed to the same drugs in order to evaluate the therapy response by immunohistochemistry.The action of the ALDH inhibitor, DEAB, on endometrial CSC populations, in fact, decreased self-renewal and sphere dimensions. RL95-2 spheres submitted to DEAB showed increased expression of CD133. However, other phenotypes associated with CSC, such as CD44+/CD24-, expression of ALDH and P53, were not influenced by the inhibitor.Endometrial CSC populations were more resistant to carboplatin, paclitaxel, and doxorubicin than adherent populations. The parental cell line and the adherent derivatives showed an increase in caspase 3 levels after treatment with carboplatin, accompanied by a decrease in the expression of P53 and ALDH, in adherent populations respectively. Treatment with carboplatin and doxorubicin was significantly more genotoxic in adherent cells than CSC.In human endometrial tumor samples, it was possible to optimize the protocol of spheres isolation and to perform cultures of tumor slices. Expression of CSC markers was most evident in tumors with poor prognosis. From the perspective of defining a personalized response to treatment, there was a variable sensitivity between different tumors. In the future, the prediction of response to therapy, prior to the definition of adjuvant treatment, may become a reality by identifying cytostatics with the best performance in CSC, the minority population responsible for chemoresistance and recurrence. O cancro do endométrio é a neoplasia maligna do trato genital feminino mais frequente nos países ocidentais, que afeta mulheres habitualmente na pós-menopausa e tem o seu pico de incidência no início da sexta década de vida. Na generalidade, apresenta um prognóstico favorável, dado que a maioria das doentes tem o diagnóstico num estádio inicial, com doença confinada ao útero. No entanto, até 20% dos carcinomas endometriais apresentam-se como neoplasias agressivas, como lesões de alto grau ou com invasão miometrial, com risco de recorrência. As células estaminais do cancro (CSC, do inglês cancer stem cells) caracterizam-se como uma subpopulação de células cancerígenas indiferenciadas que apresentam capacidade de autorrenovação, de diferenciação, de resistência às terapêuticas convencionais e potencial metastático.O objetivo principal deste trabalho foi o isolamento e caracterização de uma população de CSC do endométrio e a avaliação da inibição da via da ALDH, descrita como marcador desta população. Pretendeu-se estudar a resposta à terapêutica das CSC aos citostáticos utilizados na prática clínica e contribuir para esclarecer os mecanismos de resistência. Numa perspetiva translacional, com recurso a amostras de doentes com cancro do endométrio, pretendeu-se isolar e avaliar as CSC de modo a contribuir para o desenvolvimento de terapêuticas individualizadas.A caracterização das CSC de linhas celulares de cancro do endométrio, compreendeu o isolamento de populações com capacidade para formar esferas, a avaliação de propriedades como a autorrenovação e a expressão de marcadores típicos de CSC (por citometria de fluxo e por Western blot). Avaliou-se também a influência dos inibidores da ALDH neste fenótipo. Avaliou-se a resposta aos fármacos carboplatina, paclitaxel e doxorrubicina, em termos de atividade metabólica, expressão de proteínas envolvidas na resistência à terapêutica e morte celular, assim como danos no DNA. O protocolo de formação de esferas também foi aplicado a amostras tumorais das doentes de modo a isolar e caracterizar as CSC. Paralelamente, foram preparadas secções tumorais e expostas aos mesmos fármacos de modo a avaliar a resposta à terapêutica por imunohistoquímica.A ação do inibidor da ALDH, DEAB, nas populações de CSC do endométrio, de facto, diminuiu a autorrenovação e as dimensões das esferas. As esferas de RL95-2 submetidas ao DEAB apresentaram aumento na expressão de CD133. No entanto outros fenótipos associados a CSC, como CD44+/CD24-, a expressão da ALDH e P53, não foram influenciados pelo inibidor. As populações de CSC do endométrio apresentaram maior resistência à terapêutica com carboplatina, com paclitaxel e com doxorrubicina do que as populações aderentes. A linha parental e as derivadas aderentes, apresentaram um aumento dos níveis de caspase 3 após o tratamento com carboplatina, acompanhado de diminuição da expressão da P53 nas células RL95-2 e da ALDH nas derivadas aderentes. O tratamento com carboplatina e doxorrubicina foi significativamente mais genotóxico nas populações aderentes em relação às populações de CSC. Nas amostras de tumores endometriais humanos, foi possível otimizar o protocolo de isolamento de esferas e realizar culturas de fatias de tumor. A expressão de marcadores de CSC foi mais evidente em tumores com pior prognóstico. Na perspetiva de definir uma resposta personalizada ao tratamento verificou-se de facto uma sensibilidade variável entre os diferentes tumores. No futuro, a predição de resposta à terapêutica, prévia à definição do tratamento adjuvante, pode tornar-se uma realidade pela identificação dos citostáticos com o melhor desempenho nas CSC, população minoritária responsável pela quimiorresistência e recorrência. Abstract

Background: Head and neck cancer is the sixth most common cancer worldwide, with oral squamous cell carcinoma (OSCC) being the most representative type. OSCC is a public health problem with high morbidity and poor survival rate. Epithelial‐mesenchymal transition is emerging as a hallmark in OSCC. Methods: In this study, we described the role of microRNAs in epithelial‐mesenchymal transition regulation in OSCC based on a PubMed search using articles published in English between January 1, 2010, and January 31, 2018. Results: MicroRNA's regulatory networks seem to be a hallmark of epithelial‐mesenchymal transition in OSCC pathophysiology becoming a growing challenge to design new studies and strategies from biology to clinical applications. Conclusion: Therefore, we propose that targeting therapies to epithelial‐mesenchymal transition‐type cells, namely, coordinating microRNAs and/or hydrophobic drugs, such as conventional therapy, could be a promising strategy to improve the outcomes of patients with OSCC. Abstract

The sealers used for root canal treatment should be biocompatible for the peri-radicular tissues, to evaluate the cytotoxic effects of GuttaFlow ® bioseal sealer and to compare them with AH26 ® epoxy resin. Culture media were conditioned with the GuttaFlow ® bioseal and AH26 ® pellets. MDPC-23 odontoblast cell cultures were treated with conditioned medium and serial dilutions. To evaluate the metabolic activity and cellular viability, the MTT and SRB assays were performed. To determine the production of reactive oxygen species, the DHE and DCF-DA probes were used. Cell cycle and cell-death types were assessed by cytometry, and to evaluate the mineralization capacity, the Alizarin Red S coloration was used. Statistical analysis was performed using analysis of variance (ANOVA) when normality was found and Kruskal-Wallis on the opposite case. For the comparison with normality values, the Student t -test was used. Cells exposed to the GuttaFlow ® bioseal conditioned medium maintained high metabolic activities, except at higher concentrations. Likewise, viability was maintained, but a significant decrease was observed after exposure to the highest concentration ( p < 0.001), associated with cell death by late apoptosis and necrosis. When cell cultures were exposed to AH26 ® , metabolic activity was highly compromised, resulting in cell death. An imbalance in the production of peroxides and superoxide anion was observed. GuttaFlow ® bioseal showed higher biocompatibility than AH26 ® .
