We carry out a detailed superspace analysis of the OPE of two N=2 stress-tensor multiplets. Knowledge of the multiplets appearing in the expansion, together with the two-dimensional chiral algebra description of N=2 SCFTs, imply an analytic bound on the central charge c. This bound is valid for any N=2 SCFT regardless of its matter content and flavor symmetries, and is saturated by the simplest Argyres-Douglas fixed point. In this review, we consider the case where electrons, magnetic monopoles, and dyons become massless. Here we consider the ${\cal N} = 2$ supersymmetric Yang-Mills (SYM) theories with classical gauge groups with a rank r, SU(r+1), SO(2r), Sp(2r), and SO(2r+1). which are studied by Riemann surfaces called Seiberg-Witten curves. We discuss physical singularity associated with massless particles, which can be studied by geometric singularity of vanishing 1-cycles in Riemann surfaces in hyperelliptic form. We pay particular attention to the cases where mutually non-local states become massless (Argyres-Douglas theories), which corresponds to Riemann surfaces degenerating into cusps. We discuss non-trivial topology on the moduli space of the theory, which is reflected as monodromy associated to vanishing 1-cycles. Gravity duals for little string theories --- which give rise to four-dimensional theories that undergo permanent confinement in the infrared --- have not been studied in great detail. We address this question in the framework of heterotic SO(32) and E_8 x E_8 string theory, constructing these backgrounds by wrapping heterotic five-branes on calibrated two-cycles of non-Kahler resolved conifolds. Related to deformations of the underlying little string theories, we find numerous analytic solutions preserving N = 1 supersymmetry in four-dimensions. These theories all have non-abelian global symmetries that generally arise from both the heterotic vector bundle and from certain orbifold states. In the decoupling limit, we argue that the gravity duals are given by non-Kahler manifolds that have both blown-up two-cycles and three-cycles at the origin. We argue this following certain duality sequences that include M-theory torsional manifolds at an intermediate step, which help us to construct new type I' gauge/gravity duality pairs. Considering SO(32) heterotic string theory compactified on a torus of dimension 4 and less, stability of non-supersymmetric states is studied. A non-supersymmetric state with robust stability is constructed, and its exact stability is proven in a large region of moduli space against all the possible decay mechanisms allowed by charge conservation. Using various T-duality transform matrices, we translate various selection rules about conserved charges into simpler problems resembling partition and parity of integers. For heterotic string on T^4, we give a complete list of BPS atoms with elementary excitations, and we study BPS and non-BPS molecules with various binding energies. Seiberg-Witten theory can be embedded in F-theory using D3 branes probing an orientifold geometry. The non-perturbative corrections in the orientifold picture map directly to the instanton corrections in the corresponding gauge theory that convert the classical moduli space to the quantum one. In this short review we argue that the recently proposed class of conformal Gaiotto models may also be embedded in F-theory. The F-theory constructions help us not only to understand the Gaiotto dualities but also to extend to the non-conformal cases with and without cascading behaviors. N=2 Seiberg-Witten theories allow an interesting interplay between the Argyres-Douglas loci, singularity structures and wall-crossing formulae. In this paper we investigate this connection by first studying the singularity structures of hyper-elliptic Seiberg-Witten curves for pure N=2 gauge theories with SU(r+1) and Sp(2r) gauge groups, and propose new methods to locate the Argyres-Douglas loci in the moduli space, where multiple mutually non-local BPS states become massless. In a region of the moduli space, we compute dyon charges for all 2r+2 and 2r+1 massless dyons for SU(r+1) and Sp(2r) gauge groups respectively for rank r>1. From here we elucidate the connection to the wall-crossing phenomena for pure Sp(4) Seiberg-Witten theory near the Argyres-Douglas loci, despite our emphasis being only at the massless sector of the BPS spectra. The spectra of supergravity modes in anti de Sitter (AdS) space on a five-sphere endowed with the round metric (which is the simplest 5d Sasaki-Einstein space) has been studied in detail in the past. However for the more general class of cohomogeneity one Sasaki-Einstein metrics on S^2 x S^3, given by the Y^{p, q} class, a complete study of the spectra has not been attempted. Earlier studies on scalar spectrum were restricted to only the first few eigenstates. In this paper we take a step in this direction by analysing the full scalar spectrum on these spaces. However it turns out that finding the exact solution of the corresponding eigenvalue problem in closed form is not feasible since the computation of the eigenvalues of the Laplacian boils down to the analysis of a one-dimensional operator of Heun type, whose spectrum cannot be computed in closed form. However, despite this analytical obstacle, we manage to get both lower and upper bounds on the eigenvalues of the scalar spectrum by comparing the eigenvalue problem with a simpler, solvable system. F-theoretic constructions can alternatively be understood as consequences of certain N = 2 Seiberg-Witten theories via type IIB r D3s probing the quantum corrected orientifold backgrounds. We present four models that come out from such consideration. In Model 1, the 7-branes wrap the flat R^4 directions, leading to the well known Sp(2r) theories. We study singularity structure of moduli space of Seiberg-Witten curve, such as maximal Argyres-Douglas loci, in order to construct 1-1 map between moduli spaces. In Model 2, the 7-branes are wrapped on Taub-NUT and multi Taub-NUT spaces instead of R^4. These configurations may explain many of the Gaiotto-type constructions including possible extensions to non-conformal models with cascading behaviors. In this model the UV is described by the probe D3s decomposed into D5-anti D5 pairs wrapped on multi Taub-NUT space, while the IR remains a 4d theory. For certain arrangements of the 7-branes, this model may be dualized to the brane networks of Benini, Benvenuti and Tachikawa. The Gaiotto dualities in Model 2 are explained by chiral anomaly cancellations, anti-GSO projections and brane transmutations. Model 3 is described by seven-branes wrapped on a K3 manifold and D3 and anti-D3 probes whose number may differ at most by 24. These constructions could lead to new N = 2 models with possible dualities to both type IIB and heterotic theories on non-Kahler manifolds. In the limit where the number of probes becomes very large, the physics is captured by M(atrix) theory on K3 x K3 manifold. Finally, Model 4 is described by k D3s probing intersecting seven-brane backgrounds with Sp(2k) x Sp(2k) gauge group. These constructions could produce new N =1 heterotic dual on a non-Kahler K3 manifold that is no longer conformally Calabi-Yau. This thesis studies meta- and exactly stable supersymmetry breaking mechanisms in heterotic and type IIB string theories and constructs an F-theory Grand Unified Theory model for neutrino physics in which neutrino mass is determined by the supersymmetry breaking mechanism. Focussing attention on heterotic string theory compactified on a 4-torus, stability of non-supersymmetric states is studied. A non-supersymmetric state with robust stability is constructed, and its exact stability is proven in a large region of moduli space of T^4 against all the possible decay mechanisms allowed by charge conservation. Using string-string duality, the results are interpreted in terms of Dirichlet-branes in type IIA string theory compactified on an orbifold limit of a K3 surface. In type IIB string theory, metastable and exactly stable non-supersymmetric systems are constructed using D-branes and Calabi-Yau geometry. Branes and anti-branes wrap rigid and separate 2-spheres inside a non-compact Calabi-Yau three-fold: supersymmetry is spontaneously broken. These metastable vacua are analyzed in a holographic dual picture on a complex-deformed CY3 where 2-spheres have been replaced by 3-spheres with flux through them. By computing bosonic masses, we identify location and mode of instability. The moduli space of this complex-deformed CY3 is studied, and methods for studying the global phase structure of supersymmetric and non-supersymmetric flux vacua are proposed. By turning on a varying Neveu-Schwarz flux inside the CY3, we build meta- and exactly stable non-supersymmetric configurations with D-branes but with no anti-D-branes. Finally, a scenario for Dirac neutrinos in an F-theory SU(5) GUT model is proposed. Supersymmetry breaking leads to an F-term for Higgs field which induces a Dirac mass. We study minimal implementations of Majorana and Dirac neutrino scenarios in F-theory GUT models. In both cases the mass scale of the neutrinos m_nu ~ (M_weak)^2/M_UV arises from integrating out Kaluza-Klein modes, where M_UV is close to the GUT scale. The participation of non-holomorphic Kaluza-Klein mode wave functions dilutes the mass hierarchy in comparison to the quark and charged lepton sectors, in agreement with experimentally measured mass splittings. The neutrinos are predicted to exhibit a "normal" mass hierarchy, with masses m_3,m_2,m_1 ~ .05*(1,(alpha_GUT)^(1/2),alpha_GUT) eV. When the interactions of the neutrino and charged lepton sectors geometrically unify, the neutrino mixing matrix exhibits a mild hierarchical structure such that the mixing angles theta_23 and theta_12 are large and comparable, while theta_13 is expected to be smaller and close to the Cabibbo angle: theta_13 ~ theta_C ~ (alpha_GUT)^(1/2) ~ 0.2. We study N=1 supersymmetric U(N) gauge theories coupled to an adjoint chiral field with superpotential. We consider the full supersymmetric moduli space of these theories obtained by adding all allowed chiral operators. These include higher-dimensional operators that introduce a field-dependence for the gauge coupling. We show how Feynman diagram/matrix model/string theoretic techniques can all be used to compute the IR glueball superpotential. Moreover, in the limit of turning off the superpotential, this leads to a deformation of N=2 Seiberg-Witten theory. In the case where the superpotential drives the squared gauge coupling to a negative value, we find that supersymmetry is spontaneously broken, which can be viewed as a novel mechanism for breaking supersymmetry. We further analyze a class of recently studied metastable string vacua obtained by wrapping D5-branes and anti-D5-branes over rigid homologous S^2's of a non-compact Calabi-Yau threefold. The large N dual description is characterized by a potential for the glueball fields which is determined by an auxiliary matrix model. The higher order corrections to this potential produce a suprisingly rich phase structure. In particular, at sufficiently large 't Hooft coupling the metastable vacua present at weak coupling cease to exist. This instability can already be seen by an open string two loop contribution to the glueball potential. The glueball potential also lifts some of the degeneracy in the vacua characterized by the phases of the glueball fields.
