1. Social sharing and expressive suppression in major depressive disorder and borderline personality disorder: An experience sampling study

Abstract

Major depressive disorder (MDD) and borderline personality disorder (BPD) are characterized by disturbed patterns of emotional and interpersonal functioning, which might imply altered use of emotion regulation in interpersonal contexts. In the current study, we examined how individuals with MDD and/or BPD differ from healthy controls in (1) their overall daily life use of expressive suppression and social sharing and (2) their tendency to adjust the use of these strategies to the emotional context (i.e., preceding negative and positive affect). Thirty-four individuals with MDD, 20 individuals with BPD, 19 individuals with comorbid MDD and BPD, and 40 healthy controls participated in a week of experience sampling during which they reported their use of expressive suppression, social sharing, and experienced negative and positive affect. The results indicated that all clinical groups reported more expressive suppression and social sharing in their daily lives than healthy controls. Group differences remained when controlling for differences in mean experienced affect, except for increased suppression for MDD and increased sharing for BPD and comorbid MDD and BPD, which seemed related to these participants' overall higher levels of negative affect. Additionally, associations between within-person fluctuations in negative or positive affect and subsequent strategy use were equally strong for clinical and control participants, indicating that clinical groups did not differentially adjust the use of suppression and sharing to the emotional context. In conclusion, individuals with MDD and/or BPD showed increased use of suppression and sharing in daily life, which might contribute to, or follow from their emotional and interpersonal difficulties.

Abstract

Traditionally, the cochlea has been considered as an immunological privileged site, as the blood-labyrinth barrier provides isolation from the systemic immune system. However, the presence of immune cells in the cochlea has been recently reported, suggesting that immune-mediated processes can play a crucial role within the auditory system. In this context, both protective and detrimental T-cell functions have been linked to normal hearing and hearing loss, respectively, thereby emphasizing the dual role of T cells in cochlear health. Current knowledge also suggests that a balanced presence and activity of T cells is crucial for tissue homeostasis in the cochlea, avoiding pathologies such as age-related hearing loss and autoimmune inner ear diseases. Although cochlea-resident T cells are scarce under physiological conditions, the number of T cells appear to rise in response to acoustic trauma revealing the active recruitment of peripheral immune cells through cochlear blood vessels. In order to evaluate the induction of a peripheral inflammatory response upon noise trauma, we exposed mice to excessive noise (115 dB) for two hours. Hearing levels were determined 72 h and 1 week after noise-exposure, which revealed a substantial increase in hearing thresholds in all mice. To evaluate if the auditory stimulation would trigger a peripheral immune response we isolated splenocytes and analyzed cytokine production in different immune subsets by high parametric flow cytometry. The analyses revealed increased activation of both CD4+ and CD8+ T cells, illustrated by significant increases in the expression of INF-γ, TNF and CD44. These data suggest a peripheral component in the elevated inflammatory response following acoustic trauma. Future research will be performed to explore the link between noise-induced hearing loss, peripheral inflammation and more specifically antigen-specificity of the observed increase in activated CD4+ and CD8+ T-cell populations. Abstract

The c.151C>T (p.P51S) mutation in COCH is highly prevalent in the Dutch/Belgian population and causes DFNA9 (hearing loss and vestibular dysfunction) in > 1500 individuals. The initial symptoms manifest between the 3rd and 5th decade of life, which leaves ample time for therapeutic intervention. The clear non-haploinsufficiency disease mechanism indicates that blocking or reducing the p.P51S mutant cochlin protein levels may alleviate or prevent the DFNA9 phenotypes. Considering the broad expression of COCH by the fibrocytes of the inner ear, we designed "gapmer" antisense oligonucleotides (ASO) to specifically induce RNase H1-mediated degradation of COCH transcripts containing the c.151C>T mutation. We established several model systems to investigate the molecular efficacy of ASOs targeting the c. 151C>T mutation or low-frequency mutant allele-specific SNPs. Using overexpression models, we identified several ASOs that efficiently induce the degradation of mutant COCH transcripts. By introducing chemical modification to the oligonucleotide bases, we can alter the affinity and selectivity for the mutation transcript. We identified several ASOs with a strong preference for the mutant transcript in overexpression models. To investigate allele-specificity under physiological expression levels, we exposed patient-derived otic progenitor cells (iPCS-OPCs) to different ASOs for 8 days. In parallel, we developed a genetically humanized mouse model for DFNA9 in which human sequence-specific therapeutic strategies can be evaluated. Phenotypic follow-up of mice of all genotypes indicate that the genetic humanization has no adverse effects, and removal of the Cdh23ahl allele is mandatory to observe the late-onset auditory phenotype: the first signs of high-frequency hearing loss emerged at 12 months of age. Studies in iPSC-OPCs indicated that the ASOs identified in overexpression studies also effectively reduce mutant COCH transcript levels in patient-derived cells with physiological expression levels. Unfortunately, variation between replicate wells of OPC differentiation is relatively high, making it difficult to draw conclusions on allele-specificity. We selected a candidate ASO, directed against a rare mutant allele-spe-cific intronic SNP, for subsequent studies in our humanized mouse model. First intracochlear injections will be conducted in May 2024, after which we can collect the first in vivo data on gapmer ASO uptake and efficacy in fibrocytes of the mammalian inner ear. Abstract

Following the global outbreak of COVID-19 in March 2020, individuals report psychological distress associated with the “new normal”—social distancing, financial hardships, and increased responsibilities while working from home. Given the interpersonal nature of stress and coping responses between romantic partners, based on the systemic transactional model this study posits that perceived partner dyadic coping may be an important moderator between experiences of COVID-19 psychological distress and relationship quality. To examine these associations, self-report data from 14,020 people across 27 countries were collected during the early phases of the COVID-19 pandemic (March–July, 2020). It was hypothesized that higher symptoms of psychological distress would be reported post-COVID-19 compared to pre-COVID-19 restrictions (Hypothesis 1), reports of post-COVID-19 psychological distress would be negatively associated with relationship quality (Hypothesis 2), and perceived partner DC would moderate these associations (Hypothesis 3). 7. The current knowledge of spiral ligament fibrocytes in cell culture: a systematic review.

Abstract

The spiral ligament in the cochlea has been suggested to play a significant role in the pathophysiology of sensorineural hearing loss (SNHL). Positioned between the stria vascula-ris and the bony otic capsule, this structure contains spiral ligament fibrocytes (SLFs), categorized into five main types based on their structural characteristics, immunostaining patterns and location within the spiral ligament. Together with the stria vascularis, the SLFs maintain a positive endo-cochlear potential (EP) in the scala media via K+ recycling, which is an essential component in the transduction mechanism of the auditory pathway. It has also been suggested that the SLFs contribute to the cochlear immune response, gluta-mate homeostasis and cochlear blood flow regulation. Spiral ligament damage disrupts K+ recycling, reducing the EP and subsequently causing SNHL. Despite their pivotal roles, a lot remains unknown about the SLFs. Therefore, this systematic review about SLFs in cell culture could give an overview of the current state of the art, providing a basis for future studies trying to investigate those cells in vitro. A literature search was performed using PubMed, Web of Science and Scopus taking into account the PRISMA guidelines. Twenty-five studies were included in this review that report on SLFs in cell culture. The differences in species, sex, age, method of culturing and used antibodies for immunohis-tochemistry are discussed. The majority of these studies cultured spiral ligament fragments onto type I collagen-coated petri dishes; a protocol described by Gratton et al. in 1996, while some recent studies are focussing more on growing these cells in a 3D environment. A better understanding of the currently published methods will help us to optimize fibrocyte culture techniques, thereby allowing to investigate a variety of possibilities that can significantly increase our knowledge about SLFs. Among others, these possibilities include to discover their functional characteristics, such as growth patterns, protein expression profiles and ion channel physiology, to expose the cells to hy-poxia or other toxic conditions and to assess the resulting effects, to genetically modify SLFs, to explore drug repurposing possibilities, to evaluate potential therapies for SNHL and to investigate their capability as a source for transplantable cells. Abstract

Emotions unfold in the context of social interactions and relationships, where they serve social ends. In this review, we discuss recent research on the social functions served by positive emotions. We identify three functions of positive emotions: intimacy, impression, and influence functions. Intimacy functions refer to the benefits individuals acquire in improving their bond with others. Impression and influence functions follow from the information that others derive from positive emotion expressions. Positive emotions can therefore be used in social contexts to achieve goals such as maintaining social standing or enhancing motivation in others to achieve important goals.

Abstract

The topic of work engagement is moving up on the managerial agenda as it sets the stage for numerous beneficial outcomes for both organizations and their employees. It is clear, however, that not all employees are equally engaged in their job. The current study taps into theory on positive self-fulfilling prophecies induced by leaders’ high expectations of followers (i.e., the Pygmalion effect) and examines their potential to facilitate follower work engagement. By integrating literature on implicit followership theories with the Pygmalion model, we investigate the assumption that leaders’ high expectations are universally perceived as and therefore foster the same desirable results for all employees. Abstract

The quality of romantic relationships predicts a variety of health outcomes, ranging from mortality over psychopathology to the speed of wound healing (see e.g., Robles, Slatcher, Trombello, & McGinn, 2014). One key mechanism underlying these associations is the emotions that are experienced in the relationship (Sbarra & Coan, 2018). Romantic partners are thought to influence each other's emotions often, intensely, and in many different ways, and these emotions, in turn, shape the trajectory of the relationship. In this dissertation, we aim to contribute to a better understanding of these interpersonal emotional processes in three different ways. First, we aim to explicitly investigate if and when partners' emotions become interdependent over time, what the underlying mechanisms are, and if this emotional interdependence relates to the couple's functioning (Chapter 1, 2, 3, and 4). Second, we investigate a novel route through which partners may shape each other's emotions over time, namely by means of the perception of each other's emotion (Chapter 5). Third and finally, we examine if and how dynamic aspects of partners' emotions successively predict how they judge their (potential) relationship during defining relational events (Chapter 6 and 7). In Chapter 1, we first offer a structuring framework to capture how partners' emotions can become linked across time. We review existing research on emotional interdependence and its associations with well-being indicators, and speculate about underlying mechanisms. This chapter provides the conceptual basis for the empirical studies conducted in the subsequent chapters. Specifically, in Chapter 2 and 3 we provide a systematic and comprehensive examination of whether and to what extent couples actually show emotional interdependence both in daily life and during lab interactions, using different methods. Additionally, we investigate its stability and moderating factors, and associations with relationship closeness and well-being. To our surprise, the results challenge central notions and definitions in the traditional literature of close relationships. Across methods, types of emotion, timescales or contexts, we find relatively little consistent evidence for the existence of emotional interdependence in partners: the majority of couples in our samples simply does not demonstrate strong signs of emotional interdependence. Further, the couples that demonstrate emotional interdependence, show great differences in their specific interpersonal patterns and are highly unstable, meaning that the degree of observed emotional interdependence does not generalize well across timescales, contexts, or types of emotions. Finally, emotional interdependence is not consistently associated with relationship closeness or well-being. In absence of convincing evidence of direct emotional interdependence, in Chapter 4 we therefore examine whether it is perhaps the perception of emotional interdepence that matters in close relationships. Specifically, we investigate if feeling more emotionally similar to one's partner, and or perceiving oneself to feel more emotionally similar, goes together with increased feelings of closeness to the partner. Supporting the role of perceptions rather than actual experiences, we find that both accurately perceiving and overestimating emotional similarity positively predicts experienced closeness towards a partner. Actual emotional interdependence only predicts closeness through its effect on perception. Chapter 5 further examines the interplay of perceptions and emotions in close relationships, showing indirect ways in which partners might mold each other's emotions over time. Here, we find that how someone thinks their partner feels predicts the partner's actual feelings over time. In Chapter 6 and 7, finally, we investigate how a number of intrapersonal dynamic emotion characteristics during relational events in turn predict how people perceive a romantic partner. Specifically, we report on a speed-dating study that allows us to examine if people's prior emotional states before entering an interaction predict their romantic attraction towards their interaction partner (Chapter 6). We find evidence for contrast effects, meaning that romantic judgments contrast with the emotional states participants are in at the start of a new interaction. Additionally, we examine what aspects of partners' emotional experiences during an important relational event, namely a conflict with their partner, predict how well they recover from the conflict afterwards, in terms of positive and negative feelings towards the partner, and how responsive they perceive their partner to be. In particular, we examine whether the extent of partners' conflict recovery is mainly predicted by their most aversive (negative peak) or pleasant (positive peak) emotional experience during the conflict, or by the emotional tone at the end.
