Dendritic cell (DC)-based vaccines pulsed with high hydrostatic pressure (HHP)-inactivated tumor cells have been demonstrated to be a promising immunotherapy for solid tumors. We focused on sole injection of tumor cells that were inactivated by HHP and their combination with local radiotherapy (RTx) for in vivo induction of anti-tumor immune responses. HHP-treatment of tumor cells resulted in pre-dominantly necrotic cells with degraded DNA. We confirmed that treatments at 200 MPa or higher completely inhibited the formation of tumor cell colonies in vitro. No tumor growth was seen in vivo after injection of HHP-treated tumor cells. Single vaccination with HHP-killed tumor cells combined with local RTx significantly retarded tumor growth and improved the survival as shown in B16-F10 and CT26 tumor models. In B16-F10 tumors that were irradiated with 2 × 5Gy and vaccinated once with HHP-killed tumor cells, the amount of natural killer (NK) cells, monocytes/macrophages, CD4+ T cells and NKT cells was significantly increased, while the amount of B cells was significantly decreased. In both models, a trend of increased CD8+ T cell infiltration was observed. Generally, in irradiated tumors high amounts of CD4+ and CD8+ T cells expressing PD-1 were found. We conclude that HHP generates inactivated tumor cells that can be used as a tumor vaccine. Moreover, we show for the first time that tumor cell-based vaccine acts synergistically with RTx to significantly retard tumor growth by generating a favorable anti-tumor immune microenvironment. In bisherigen Studien konnte bereits gezeigt werden, dass Tumorzellen, welche durch Hochdruckbehandlung inaktiviert wurden, von dendritischen Zellen aufgenommen und die dendritischen Zellen zu einer Immunantwort stimuliert werden. Somit konnten solche aus dendritischen Zellen bestehenden oder auf dendritischen Zellen basierenden Vakzine als eine weitere Option für eine Immuntherapie bei soliden Tumoren identifiziert werden. Ziel dieser Arbeit war zu untersuchen, dass auch Vakzine, nur bestehend aus hochdruckbehandelten ganzen Tumorzellen, in der Lage sind eine Antitumorantwort auszulösen und insbesondere mit lokaler Bestrahlung von Tumoren kombiniert werden können. Murine B16-F10 Melanomzellen und murine CT26 Zellen eines Kolonkarzinoms wurden mit hydrostatischen Drücken von 100 bis 500 MPa behandelt. Anschließend wurden verschiedene in vitro und in vivo Versuche durchgeführt, in welchen die vollständige Inaktivierung der Tumorzellen überprüft wurde. Das Potential zur Erzeugung einer Antitumorimmunantwort der so generierten Vakzine aus hochdruckbehandelten ganzen Tumorzellen wurde anschließend in vivo untersucht. Hierfür wurden Tumorzellen mit hydrostatischem Hochdruck inaktiviert und ohne Adjuvans tumortragenden Mäusen als Vakzin verabreicht. Der Fokus wurde dann insbesondere auf die Kombination der Vakzine mit lokaler Strahlentherapie gelegt und die immunologische Zusammensetzung des Tumors wurde im Detail analysiert. Eine Hochdruckbehandlung von Tumorzellen mit Drücken von mindestens 200 MPa führte zu einem Zelltod der Tumorzellen durch Nekrose mit degradierter DNA und verhinderte vollständig die Vermehrung von Tumorzellen in vitro. Auch in vivo konnte kein Tumorwachstum nach Injektion von hochdruckbehandelten Tumorzellen festgestellt werden. Eine Vakzinierung mit hochdruckbehandelten Tumorzellen in vivo konnte in Kombination mit einer lokalen Radiotherapie das Tumorwachstum in tumortragenden Mäusen signifikant verzögern und das Überleben verbessern. Im B16-F10 Tumormodell resultierte die kombinierte Therapie aus Vakzinierung und lokaler Radiotherapie in einer signifikant erhöhten Infiltration von natürlichen Killerzellen, Monozyten/Makrophagen, CD4+ T-Zellen und natürlichen Killer T-Zellen sowie einer signifikant verminderten Infiltration von B-Zellen in den Tumor. Im B16-F10 sowie im CT26 Tumormodell wurden eine vermehrte Einwanderung von CD8+ T-Zellen und eine hohe Anzahl von PD-1 exprimierenden CD4+ und CD8+ T-Zellen beobachtet. Zum einen ist die Hochdruckbehandlung geeignet Tumorzellen effektiv zu inaktivieren und somit zur Herstellung von auf ganzen Tumorzellen basierenden Vakzinen einsetzbar. Zum anderen konnte gezeigt werden, dass diese Vakzine in Kombination mit lokaler Radiotherapie das Tumorwachstum signifikant verzögern, indem eine vorteilhafte immunologische Tumormikroumgebung geschaffen wird.

XRR was recorded from the PPyfilm inan aqueous 0.1 M perchloric acid at electric potentials between���0.2 V and +0.5 V vs Ag/AgCl. The PPyfilm shows typicalreversible and linear changes infilm thickness and electron densityarising from the potential-dependent electrolyte incorporation. Byintroducing EC-XRR, a comprehensive analysis combining in situ XRR and EC, the net number of electrons passing through thePPy-electrolyte interface was deduced along with the potential-induced thickness variations, indicating a complex exchangemechanism. Evidently, along with the anion transfer, parallel charge compensation by protons and a volume and electroncompensating counterflow of solvent molecules take place. Using coherent X-ray diffraction imaging (CXDI) as an in situ tool, we determined the shape and strain state of a platinum nanoparticle with ≈160 nm diameter supported by a strontium titanate substrate. The experiment was performed at a temperature of 400 K under continuous gas flow conditions of pure Ar and Ar/CO mixtures. The nanoparticle was preselected by scanning electron microscopy (SEM) and postanalyzed by atomic force microscopy (AFM). We obtain a very good agreement between the overall nanoparticle size, shape, and defect structure as determined by CXDI and AFM. In addition, we compare the strain state in the nanoparticle near surface region and its bulk: For pure Ar flow, we find a slight compressive strain in the nanoparticle bulk compared to an expansion in the near surface region. We ascribe the latter to the presence of high index vicinal surfaces. Our analysis suggests that under mixed Ar/CO flow at 400 K reshaping of the nanoparticle occurred. New high index facets developed, leading to a stronger lattice expansion, also propagating into the nanoparticle bulk. Our high-resolution experiments pave the way for future CXDI experiments under operando catalytic reaction conditions.

Copper oxides, such as CuO and Cu2O, are promising materials for H2S detection because of the reversible reaction with H2S to copper sulfides (CuS, Cu2S). Along with the phase change, the electrical conductance increases by several orders of magnitude. On CuOx films the H2S reaction causes the formation of statistically distributed CuxS islands. Continuous exposition to H2S leads to island growth and eventually to the formation of an electrical highly conductive path traversing the entire system: the so-called percolation path. The associated CuOx ∕ CuxS conversion ratio is referred to as the percolation threshold. This pronounced threshold causes a gas concentration dependent switch-like behaviour of the film conductance. However, to utilize this effect for the preparation of CuO-based H2S sensors, a profound understanding of the operational and morphological parameters influencing the CuS path evolution is needed.Thus, this article is focused on basic features of H2S detection by copper oxide films and the influence of structural parameters on the percolation threshold and switching behaviour. In particular, two important factors, namely the stoichiometry of copper oxides (CuO, Cu2O and Cu4O3) and surface morphology, are investigated in detail. CuOx thin films were synthesized by a radio frequency magnetron sputtering process which allows modification of these parameters. It could be shown that, for instance, the impact on the switching behaviour is dominated by morphology rather than stoichiometry of copper oxide.

Diese Arbeit beschäftigt sich mit der Detektion von Schwefelwasserstoff (H2S) mittels Kupferoxid (CuO)-Strukturen. Die große Selektivität von CuO zur Detektion von H2S zeigte in verschiedenen Arbeiten, dass es unter Exposition einer gewissen Dosis H2S einen Leitwertsprung von mehr als vier Größenordnungen vollführt, weil in einer spezifischen Reaktion halbleitendes CuO zu metallisch leitendem Kupfersulfid (CuS) reagiert. Dieser Sprung wird mit einem Perkolations-Modell beschrieben und als digitales Signal genutzt. Die Zeit bis zum Erreichen dieses Sprungs gibt Information über die durchschnittlich vorliegende Konzentration. Im Fokus dieser Arbeit steht die Aufklärung der Phasen, welche bei diesem Leitwertsprung beteiligt sind, sowie die Synthese von Modellsystemen. Diese wurden in Form von 100 nm dicken Dünnschichten durch Resonanzfrequenz-Magnetron-Sputtern auf Saphir abgeschieden. Durch die Verwendung von H2S in der Synthesekammer ist es möglich, Schwefel in die Dünnschichten einzubauen. Die Zusammensetzung der Schichten wurde mit Röntgen-Photoelektronen-Spektroskopie (engl.: X-ray photoelectron spectroscopy, XPS), Flugzeit-Sekundärionen-Massenspektrometrie(time of flight secondary ion mass spectromety, ToF-SIMS) und Röntgen-Beugung (engl.: X-ray diffraction, XRD) untersucht. Diese Proben werden mit Dünnschichten verglichen, welche unter realen Messbedingungen H2S ausgesetzt wurden. Es zeigt sich, dass die Zusammensetzung von synthetisierten und exponierten Dünnschichten übereinstimmen. In einer weiteren Reihe wurden CuO-Dünnschichten durch thermische Oxidation von Cu hergestellt. The exposition of CuO to a certain dose of H2S results in a change of conductance over four orders of magnitude due to the specific reaction of semiconducting CuO with H2S to metallically conductive CuS. This reaction can be used as highly selective sensor mechanism which exhibits a digital on/off signal. Measuring the time until the conductance jump occurs enables the determination of an average concentration. This thesis concentrates on determining phases contributing to the conductance jump as well as the synthesis of model systems. Therefore, 100 nm thin films were deposited on sapphire by resonant magnetron sputtering. Adding H2S to the sputter gas enables the incorporation of sulfide into the film. The composition of these films was examined by X-ray photoelectron spectroscopy (XPS), time of flight secondary ion mass spectrometry (ToFSIMS) and X-ray diffraction (XRD). Those synthesized samples showed to be comparable to samples which were exposed to H2S under real conditions. In a third series CuO thin films were synthesized by thermal oxidation of Cu films, which exhibiting similar behavior to H2S. CuS particles are obtained on the surface. Even after the conductance reaches the value of CuS they remain separated. Due to that findings, the model for the conversion of CuO thin films under H2S proposed by Hennemann et al. [1] is refined. This work shows that CuS particles inject charge carriers into the CuO film. Furthermore, the choice of contact material reveals to be crucial for a reliable operation of the sensor. Additionally, this work examines the influence of structure on H2S sensing properties. For that, lithographically structured thin CuO films and electrospun CuO nanofibers were synthesized and exposed under real measurement conditions. The behavior of all structures is described by the two dimensional percolation effect. With these findings, a pure CuO nanofiber based sensor was prepared which exhibited percolation based behavior for over 560 cycles indicating that just porosity is crucial to build a reliable and long term stable H2S sensor.

Objective Emergency cervical cerclage is one of the treatment options for the reduction of preterm birth. The aim of this study is to assess neonatal outcome after cerclage with special focus on adverse effects in very low birth weight infants. Study Design Retrospective cohort study. Classification of cerclages in history-indicated (HIC, n = 38), ultrasound-indicated (UIC, n = 29) and emergency/ physical examination-indicated (PEIC, n = 33) cerclage.
