Rosemary is one of the most important medicinal plants for natural therapy due to its multiple pharmacological properties, such as antioxidant, anti-inflammatory, neuroprotective, antiproliferative, antitumor, hepato- and nephroprotective, hypolipidemic, hypocholesterolemic, antihypertensive, anti-ischemic, hypoglycemic, radioprotective, antimicrobial, antiviral, antiallergic, and wound healing properties. Our study reports for the first time, over a 12-month period, the identification and quantification of polyphenols and the investigation of the antioxidant and acetylcholinesterase (AChE) inhibitory activities of the Rosmarinus officinalis L. species harvested at flowering from the flora of southwestern Romania (Oltenia Region). Identification and quantification of polyphenolic acids was made by ultra-high-performance liquid chromatography/mass spectrometry (UHPLC/MS). Total phenolic content was determined using the spectrophotometric method. In situ antioxidant and anticholinesterase activities were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and AChE inhibitory assay, respectively, on high-performance thin-layer chromatography (HPTLC) plates. DPPH radical scavenging activity was also assessed spectrophotometrically. The results revealed significant correlations between specific polyphenolic compounds and the measured biological activities, understanding the role of seasonal variations and providing insights into the optimal harvesting times and medicinal benefits of rosemary. Our research brings new information on the phytochemical profile of R. officinalis as a natural source of polyphenols with antioxidant and AChE inhibitory properties. [ABSTRACT FROM AUTHOR]

Background: Heracleum sphondylium, a medicinal plant used in Romanian ethnopharmacology, has been proven to have remarkable biological activity. The escalating concerns surrounding antimicrobial resistance led to a special attention being paid to new efficient antimicrobial agents based on medicinal plants and nanotechnology. We report the preparation of a novel, simple phytocarrier that harnesses the bioactive properties of H. sphondylium and silver nanoparticles (HS-Ag system). Methods: H. sphondylium's low metabolic profile was determined through gas chromatography–mass spectrometry and electrospray ionization–quadrupole time-of-flight–mass spectrometry. The morphostructural properties of the innovative phytocarrier were analyzed by X-ray diffraction, Fourier-transform infrared spectroscopy, Raman spectroscopy, dynamic light scattering, scanning electron microscopy, and energy-dispersive X-ray spectroscopy. The antioxidant activity was evaluated using total phenolic content, ferric reducing antioxidant power, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) in vitro assays. The antimicrobial activity screening against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, and Escherichia coli was conducted using the agar well diffusion method. The 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay estimated the in vitro potential cytotoxicity on normal human dermal fibroblasts (NHDF) and cervical cancer (HeLa) cells. Results: A total of 88 biomolecules were detected, such as terpenoids, flavonoids, phenolic acids, coumarins, phenylpropanoids, iridoids, amino acids, phytosterols, fatty acids. The HS-Ag phytocarrier heightened efficacy in suppressing the growth of all tested bacterial strains compared to H. sphondylium and exhibited a significant inhibition of HeLa cell viability. Conclusions: The new HS-Ag phytocarrier system holds promise for a wide range of medical applications. The development of cost-effective and high-performance technologies for wastewater treatment is essential for achieving a sustainable economy. Among the various methods available for water remediation, adsorption is widely recognized as an effective and straightforward approach for removing a range of pollutants. Gel materials, particularly hydrogels and aerogels, have attracted significant research interest due to their unique properties. Hydrogels, for instance, are noted for their ability to be regenerated and reused, ease of separation and handling, and suitability for large-scale applications. Additionally, their low cost, high water absorption capacity, and contribution to environmental protection are important advantages. Aerogels, on the other hand, are distinguished by their low thermal conductivity, transparency, flexibility, high porosity, mechanical strength, light weight, large surface area, and ultralow dielectric constant. This review provides a comprehensive analysis of the current literature, highlighting gaps in knowledge regarding the classification, preparation, characterization, and key properties of these materials. The potential application of hydrogels and aerogels in water remediation, particularly in removing contaminants such as dyes, heavy metals, and various organic and inorganic pollutants, is also discussed. This review investigates innovative strategies for upcycling agricultural residues into valuable pharmaceutical compounds. The improper disposal of agricultural residues contributes to significant environmental issues, including increased greenhouse gas emissions and ecosystem degradation. Upcycling offers a sustainable solution, transforming these residues into high-value bioproducts (antioxidants, antitumor agents, antidiabetic compounds, anti-inflammatory agents, and antiviral drugs). Nanotechnology and microbial biotechnology have a crucial role in enhancing bioavailability and targeted delivery of bioactive compounds. Advanced techniques like enzymatic hydrolysis, green solvents, microwave processing, pyrolysis, ultrasonic processing, acid and alkaline hydrolysis, ozonolysis, and organosolv processes are explored for their effectiveness in breaking down agricultural waste and extracting valuable compounds. Despite the promising potential, challenges such as variability in residue composition, scalability, and high costs persist. The review emphasizes the need for future research on cost-effective extraction techniques and robust regulatory frameworks to ensure the safety, efficacy, and quality of bioproducts. The upcycling of agricultural residues represents a viable path towards sustainable waste management and production of pharmaceutical compounds, contributing to environmental conservation and public health improvements. This review provides an analysis of the current literature and identifies knowledge gaps, offering recommendations for future studies to optimize the use of agricultural residues in the drug industry. Horseradish is a globally well-known and appreciated medicinal and aromatic plant. The health benefits of this plant have been appreciated in traditional European medicine since ancient times. Various studies have investigated the remarkable phytotherapeutic properties of horseradish and its aromatic profile. However, relatively few studies have been conducted on Romanian horseradish, and they mainly refer to the ethnomedicinal or dietary uses of the plant. This study reports the first complete low-molecular-weight metabolite profile of Romanian wild-grown horseradish. A total of ninety metabolites were identified in mass spectra (MS)-positive mode from nine secondary metabolite categories (glucosilates, fatty acids, isothiocyanates, amino acids, phenolic acids, flavonoids, terpenoids, coumarins, and miscellaneous). In addition, the biological activity of each class of phytoconstituents was discussed. Furthermore, the development of a simple target phyto-carrier system that collectively exploits the bioactive properties of horseradish and kaolinite is reported. An extensive characterization (FT-IR, XRD, DLS, SEM, EDS, and zeta potential) was performed to investigate the morpho-structural properties of this new phyto-carrier system. The antioxidant activity was evaluated using a combination of three in vitro, non-competitive methods (total phenolic assay, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay, and phosphomolybdate (total antioxidant capacity)). The antioxidant assessment indicated the stronger antioxidant properties of the new phyto-carrier system compared with its components (horseradish and kaolinite). The collective results are relevant to the theoretical development of novel antioxidant agent fields with potential applications on antitumoral therapeutic platforms. [ABSTRACT FROM AUTHOR]
