Computer vision techniques have become important in agriculture and plant sciences due to their wide variety of applications. In particular, the analysis of seeds can provide meaningful information on their evolution, the history of agriculture, the domestication of plants, and knowledge of diets in ancient times. This work aims to propose an exhaustive comparison of several different types of features in the context of multiclass seed classification, leveraging two public plant seeds data sets to classify their families or species. In detail, we studied possible optimisations of five traditional machine learning classifiers trained with seven different categories of handcrafted features. We also fine-tuned several well-known convolutional neural networks (CNNs) and the recently proposed SeedNet to determine whether and to what extent using their deep features may be advantageous over handcrafted features. The experimental results demonstrated that CNN features are appropriate to the task and representative of the multiclass scenario. In particular, SeedNet achieved a mean F-measure of 96%, at least. Nevertheless, several cases showed satisfactory performance from the handcrafted features to be considered a valid alternative. In detail, we found that the Ensemble strategy combined with all the handcrafted features can achieve 90.93% of mean F-measure, at least, with a considerably lower amount of times. We consider the obtained results an excellent preliminary step towards realising an automatic seeds recognition and classification framework. [ABSTRACT FROM AUTHOR]

Jatropha mollissima is a species of the semiarid region of northeastern Brazil. It is known that individuals of the same species may present differences in seeds viability, thus it is necessary to define methodologies to evaluate the physiological potential of seeds. This study aimed at determining the best pre-moistening time and the concentration of the tetrazolium solution to evaluate the viability of J. mollissima seeds. Three pre-moistening times were tested (12, 14 and 16 hours) and two tetrazolium solution concentrations (0.25 and 0.5 %). As a comparative study, a germination test was performed, which consisted of two treatments: seeds between paper and on vermiculite. The values obtained in the tetrazolium test were similar, with average viable seeds of 21.3% for concentration 0.25 % and 27.3 % for the concentration of 0.5 %. It was observed, in the germination test, that the percentage of germination had averages equal to 22.5 % for the treatment between paper and 13.5 % for the treatment with vermiculite. The best procedure for performing the tetrazolium test in J. mollissima seeds is using the concentration of 0.25 % of tetrazolium solution, pre-moistening time of 16 hours, between paper at 25 ℃ and staining for 4 hours at 40 ℃ in the dark. This study was conducted to evaluate different volumes of distilled water and substrate temperatures on germination and vigor of melon, cucumber variety. The experiment was conducted at the Laboratory of Seed Analysis Unimontes in the period february to april 2012. Sowing was done on paper rolls like “germitest” moistened with volumes (mL) of distilled water equivalent to 1.0; 1.5; 2.0; 2.5 and 3.0 times the weight of dry substrate without further addition of water, with three sheets per roll. The rolls were kept in germination preset constant temperatures of 25 to 30°C and 20-30°C alternate. The experimental design was completely randomized in a factorial 5 x 3 (distilled water volume an temperatures), with four replicates of 50 seeds. We evaluated the germination percentage, germination velocity index, root length and shoot, fresh and dry weight of seedlings. Given the results, it is concluded that combinations of substrate distilled water volume and temperature of exposure that provide better answers on germination and vigor of melon seeds, cucumber variety, are 1.0 times the weight of dry substrate at a temperature of 30ºC and 3.0 times the dry weight of the substrate 25 and 20-30°C. Among the studied temperatures, the temperature of 30°C promotes better responses in all variables, is recommended as an additional temperature to evaluate the physiological quality of melon seeds.

The seeds were sown on blotting paper in transparent plastic boxes and then allowed to germinate in Biochemical Oxygen Demand germinators. The seeds were evaluated over a period of eight days with respect to the following variables: germination percentage, average germination time, shoot and root lengths, shoot and root dry weights and root-shoot ratio. The data were submitted to Tukey’s test (p ≤ 0.05%). The S. hispanica seed (beige colored) germination test can be carried out at constant (25°C) or alternating (25-30°C) temperatures. The germination of the seeds was indifferent to the light conditions evaluated; however, there was increased seedling growth and dry matter accumulation in the presence of light. The duration of the S. hispanica seed germination test can be five days, with the first count on the second day after sowing.

O trabalho teve por objetivo determinar métodos de pré-condicionamento e concentrações da solução de tetrazólio na avaliação da qualidade de sementes de Parkia velutina Benoist. Os tratamentos pré-condicionantes das sementes foram avaliados com três métodos de escarificação: desponte na região oposta ao hilo, punção na região mediana e lixa nos dois lados da semente com posterior embebição em 200 ml de água para cada 25 sementes e permanência em câmara a 30 ºC por 16 horas. As concentrações da solução de tetrazólio testadas foram: 1,0%, 0,5% e 0,1% por duas horas na temperatura de 40 ºC. O tratamento de pré-condicionamento mais eficiente foi a combinação do desponte e lixamento nos dois lados da semente. A concentração de tetrazólio a 0,5% pode ser utilizada para avaliar a viabilidade das sementes de P. velutina como complemento ao teste de germinação. O teste de tetrazólio se mostrou eficiente na caracterização de lesões ocasionadas por insetos e danos mecânicos em sementes de P. velutina.This manuscript aimed to determine methods of daily pre-conditioning and concentrations of the tetrazolium solution for evaluating the quality of Parkia velutina Benoist seeds. The daily pre-condicionanting seed treatments were evaluated using three scarification methods: coating cutting on the opposite side of the hilum region, puncture in the medium region and scarification with sandpaper on both sides of the seed, followed by soaking each of the 25 seeds in 200 ml of wate and permanence in chamber 30 ºC for 16 hours. The concentrations of the tetrazolium solution tested were 1.0%, 0.5% and 0.1% for two hours at 40 ºC. The more efficient daily pre-conditioning treatlment was a combination of the coating cutting and the scarification n both sides of the seed. The 0.5% tetrazolium concentration can be used to evaluate the viability of P. velutina seeds as complement to the germination test. The tetrazolium test showed its efficiency in the characterization of injuries caused by insects and mechanical damages in P. velutina seeds.

The lima beans (Phaseolus lunatus L.) have rusticity and environment adaptability; have high levels of protein and important social aspect. The aim of this study was to evaluation the physical, physiological and morphological characteristics of the external seeds, which been collected in the Montes Claros-MG Municipal Market. The experiment has done at the Laboratory of Seed Analysis ICA/UFMG and has used three batches of four varieties, been studied: the morphology and integument coloration, moisture test, mass of thousand seeds, germination test, germination speed index, a field emergence test, emergence speed index, shoot dry mass. The experimental design has been completely randomized with factorial 4x3 (four seeds varieties vs. three different lots), with 4 repetitions, totaling 48 plots. It has found significant effects in the interaction between variety and lot about the characteristics: thousand seeds weight, germination percentage, germination speed index, emergency and initial moisture. There was variation in physical and physiological quality between studied varieties, as influenced by the environment. The varieties of lima bean called by 'marronzinha', 'leite' and 'anduzinha' demonstrated a high physiological quality. [ABSTRACT FROM AUTHOR]

Objetivando verificar o comportamento das sementes de camu-camu, quanto à germinação, em função da embebição, realizou-se o presente trabalho, utilizando-se sementes extraídas de frutos maduros, provenientes de plantas adultas. Os tratamentos consistiram em seis tempos de pré-embebição: 0 (testemunha), 4, 8, 12, 24 e 36 horas. Após cada período, as sementes foram colocadas em caixas tipo gerbox, tendo como substrato vermiculita textura fina, e mantidas em germinadores BOD à temperatura constante de 25ºC. Para cada tratamento foram realizadas 4 repetições com 25 sementes cada. Realizaram-se avaliações diárias do número de sementes germinadas, sendo os dados agrupados semanalmente e transformados em arc sen x / 100 para fins de análise estatística. As médias foram comparadas pelo teste de Tukey, a 5% de probabilidade. Pelos resultados obtidos, pode-se concluir que a germinação das sementes de camu-camu não é influenciada pela embebição, bem como pelo período deste tratamento.This research was carried out aiming to verify the behavior of camu-camu seeds in their germination, according to imbibition periods. The seeds were extracted from ripe fruits of old plants. The treatments consisted of six preimbibition periods: 0, 4, 8, 12, 24 and 36 hours. After each period, the seeds were placed in gerbox, having vermiculite as substrate. They were maintained in a temperature of 25oC, in a germination chamber (BOD type). For each treatment, 4 replications with 25 seeds in each one were accomplished. Daily evaluations of the number of germinated seeds were done. The data were grouped weekly and they were transformed in arc sen x / 100 . The means were compared by the Tukey test (5% probability). Results showed that the germination of camu-camu seeds is not influenced by the imbibition, as well as by the period of this treatment.

The complexity of plant foods high in dietary fiber poses new challenges to clinical investigators and leads to many study-design dilemmas. There are basic differences in studying purified polymers, highly concentrated but not purified fibers, and diets high in high-fiber whole foods. The fibrils of the plant cell wall are most likely altered when prepared as a pure chemical entity, and when fiber concentrates (eg, wheat bran) are used, the method of preparation may alter the composition of the final product. Whole-plant, high-fiber foods are complex storehouses of a diversity of polymers, including resistant starch, and of bioactive compounds. Furthermore, the addition of a reasonable amount of high-fiber food to the diet not only adds dietary fiber but many digestible, caloric macronutrients that alter the entire diet composition. These problems and dilemmas are reviewed.

An inhibitor (BGIA) against an acidic amino acid-specific endopeptidase of Streptomyces griseus (Glu S. griseus protease) was isolated from seeds of the bitter gourd Momordica charantia L., and its amino acid sequence was determined. The molecular weight of BGIA based on the amino acid sequence was calculated to be 7419. BGIA competitively inhibited Glu S. griseus protease with an inhibition constant (Ki) of 70 nM, and gel filtration analyses suggested that BGIA forms a 1:1 complex with this protease. However, two other acidic amino acid-specific endopeptidases, protease V8 from Staphylococcus aureus and Bacillus subtilis proteinase (Glu B. subtilis protease), were not inhibited by BGIA. BGIA had no inhibitory activity against chymotrypsin, trypsin, porcine pancreatic elastase, and papain, although subtilisin Carlsberg was strongly inhibited. The amino acid sequence of BGIA shows similarity to potato chymotrypsin inhibitor, barley subtilisin-chymotrypsin inhibitor CI-1 and CI-2, and leech eglin C, especially around the reactive site. Although the residue at the putative reactive site of these inhibitors is leucine or methionine, the corresponding amino acid in BGIA is alanine.

Cyclopiazonic acid (CPA), a secondary metabolite produced by several species of Aspergillus and Penicillium, was detected in moldy sunflower seed screenings fed to sows who experienced conception problems and feed refusal. Tandem mass spectrometry confirmed the CPA which was estimated by thin-layer chromatography (TLC) to be present at 10 ppm. A Penicillium species was isolated from the moldy seeds, but in culture it did not produce CPA.

Three fenugreek inhibitors (TFI-A8, TFI-N2, and TFI-B2) were isolated from an inhibitor preparation by anion exchange chromatography and subsequent preparative isoelectric focusing using immobilized pH gradients and the canal technique. The purified inhibitors inhibited the enzymes tested differently: TFI-A8 exhibited a high inhibition of trypsin (8.2 mg human trypsin/mg and 8.1 mg bovine trypsin/mg) and a very low inhibition of chymotrypsin (0.8 mg human chymotrypsin/mg and 1.0 mg bovine chymotrypsin/mg). TFI-N2 inhibited the four enzymes to about the same extent (5.0 mg/mg human and 4.1 mg/mg bovine trypsin; 4.9 mg/mg human and 3.7 mg/mg bovine chymotrypsin). TFI-B2 displayed a high inhibition of trypsin (7.5 mg/mg human and 5.1 mg/mg bovine) and a low inhibition of chymotrypsin (1.8 mg/mg human and 1.9 mg/mg bovine). On average, the human enzymes were inhibited better than the bovine ones by the purified inhibitors. The inhibitors contained high amounts of cystine (five or six disulfide bridges per molecule), aspartic acid, threonine, serine and proline, no valine and methionine and two of them also no tryptophan. Their molecular masses were about 6 kDa. Their inclusion into the Bowman-Birk soybean proteinase inhibitor family is discussed.

Wet heat shock (60 degrees C, 90 s) and caffeine (3.8 X 10(-4) M) afford significant radioprotection against post-irradiation O2-dependent damage which develops in seeds of approximately 3.5% moisture content. The damage was assessed in terms of seedling injury on the eighth day of growth. An increase in seedling injury is clearly seen, associated with a parallel increase in the peroxidase activity. There is a concomitant decrease in the content of total peroxides. Both these post-irradiation treatments potentiate the O2-independent component of seedling injury, irrespective of the seed moisture content. Analysis of the peroxidase activity in the seedlings using non-denaturing polyacrylamide gel electrophoresis reveals that two additional bands appear with the post-irradiation oxic damage. Radioprotection against this damage by caffeine, heat shock and O2-free post-irradiation hydration is accompanied by the disappearance of these two additional bands. However there is no appearance of the two additional bands in the peroxidase family even though the enzyme activity is substantially increased due to the action of caffeine and/or heat shock on the post-irradiation O2-independent pathway of damage. The probable mechanisms of radioprotection are discussed in this paper.

Glycosylation-site-containing peptides were isolated from the proteolytic digests of luffin-a, luffin-b, PAP-S and CNBr-fragments of PAP-S by reverse-phase HPLC, and their amino acid compositions and sequences were analyzed. Six peptides were obtained from luffin-a, and three each from luffin-b and PAP-S. All of these peptides were negative toward the phenol-H2SO4 reaction and gave only N-acetyl-D-glucosamine in gas chromatography after methanolysis and reacetylation. Amounts of N-acetyl-D-glucosamine in these peptides were determined as D-glucosamine to be approximately one mol per peptide by an amino acid analyzer after HCl hydrolysis. Based on these results we concluded that Asn residues at positions of 28, 33, 77, 84, 206, and 227 in luffin-a, of 2, 78, and 85 in luffin-b, and of 10, 44, and 255 in PAP-S were glycosylated with only GlcNAc, and contained one residue per site.

Fenugreek contained proteinase inhibitors inhibiting 5-9 mg human trypsin, 5-7 mg bovine trypsin, 2-6 mg human chymotrypsin, and 1-3 mg bovine chymotrypsin per g seed material. About 30 inhibitors were electrophoretically detected, and 23 of them, inhibiting all the four enzymes, were characterized by means of their isoelectric points: a group of acid inhibitors (TFI-A1 to A10, pI 4.48-5.12), a group of neutral inhibitors (TFI-N1 to -N6, pI 5.91-6.71), and a group of basic inhibitors (TFI-B1 to -B7, pI 7.76-9.77). To eliminate the galactomannans which complicate further purification, coarsely ground seeds were separated by density into two fractions, seed coats + endosperm and cotyledons + embryos (C + E). Isolation of the fenugreek inhibitors by extraction of fraction C + E, followed by ammonium sulfate fractionation and affinity chromatography on anhydrotrypsin-Sepharose, resulted in an about 700-fold enrichment.

Isoelectric focusing performed with immobilized pH gradients was found superior to other commonly used electrophoretic methods for discrimination of 55 European winter and spring barley cultivars. Hordeins, the alcohol-soluble proteins, yielded 32 different patterns, allowing identification of 22 cultivars and classification of the remaining ones into ten groups of two to eight cultivars each. Only 21 different hordein patterns were observed using horizontal sodium dodecyl sulfate-polyacrylamide gel electrophoresis, followed by silver staining. Twelve cultivars exhibited unique hordein patterns, the remaining nine groups contained 2-11 cultivars. Resolution of isoelectric focusing with immobilized pH gradients was further enhanced in some cases when the patterns of urea/dithiothreitol-soluble proteins were used instead of the hordein patterns. However, evaluation was more complicated because of the larger number of protein bands detected.

Two different methods of detecting electroblotted glycoproteins after sodium dodecyl sulfate-polyacrylamide gel electrophoresis of Tris-buffer soluble barley seed proteins were examined for their applicability for barley cultivar discrimination. These are the highly specific, lectin-based concanavalin A/peroxidase method and the more general periodate/danyslhydrazine method. The results of the periodate/dansylhydrazine method enabled us to divide the 20 examined cultivars into three groups, whereas the more sensitive concanavalin A/peroxidase method revealed six different glycoprotein patterns. In comparison, sodium dodecyl sulfate-polyacrylamide gel electrophoresis and silver staining of the alcohol-soluble barley seed proteins (hordeins) gave nine different banding patterns. A combination of hordein electrophoresis together with glycoprotein staining by the concanavalin A/peroxidase method made it possible to classify the cultivars into twelve groups, the largest of which contained four cultivars. The qualitative expression of the glycoprotein patterns seemed to be independent of growth conditions, whereas the band intensities obviously were not. As a whole, glycoprotein blotting is a valuable supplement to sodium dodecyl sulfate-polyacrylamide gel electrophoresis of hordeins in barley cultivar discrimination.

The composition of "group B saponin" in soybean seed was analyzed by HPLC, and six kinds of "group B saponin," named Ba, Bb, Bb', Bc, Bd and Be according to elution order from HPLC, were detected. Of these saponins, Ba, Bb, Bb' and Bc were identified with soyasaponin V, I, III and II, respectively. Bd and Be were novel saponins possessing soyasapogenol E as the aglycone and the same sugar chain as Ba and Bb, respectively. These saponins were very unstable in the isolated state and had a tendency to form Ba and Bb, respectively. From these results, Bd and Be are presumed to be the precursors of Ba and Bb in soybean seed.

The fine structure of a beta-)1----4)-linked glucomannan from Asparagus officinalis has been determined by n.m.r. analysis of the oligosaccharides obtained by acidic and enzymic hydrolyses. Cleavage of the glucomannan with beta-D-mannase from Aspergillus niger and purification by h.p.l.c. gave oligosaccharide fractions that contained Man (mannose), GlcMan (beta-glucopyranosylmannose), Man2, Glc2Man, and Glc3Man as the major components. Simulated digestion of a polymer composed of randomly distributed monomers with the same Glc:Man ratio as glucomannan from A. officinalis led to the same polysaccharides. The random distribution of the monomers of glucomannan from A. officinalis was corroborated by the diffraction diagram of the raw flour, which indicated that the "in situ" glucomannan was amorphous, whereas both cellulose and mannans are crystalline.

Omega-3 Fatty acids (omega-3FAs) are found in seafoods, some plants, and some livestock rations. Fish oils are the only concentrated source of eicosapentaenoic acid (EPA; 20:5 omega-3) and docosahexaenoic acid (DHA; 22:6 omega-3). The major omega-3FA in plants is alpha-linolenic acid (LNA; 18:3 omega-3). LNA must be converted to EPA before it exerts biological effects similar to EPA, such as reduced platelet aggregation. Human beings convert LNA to EPA to a small extent only. LNA may be more readily oxidized than incorporated into tissues. The effects of consuming LNA-rich oils are more modest than the effects of EPA-rich oils. Evidence suggests that omega-3FAs are essential and highly desirable for brain and eye development and heart health. LNA is the only source of omega-3FAs for vegetarians. Because LNA and EPA are not biologically equivalent, food composition data or product claims mentioning total omega-3FA content must clarify the individual omega-3FAs present.

A new type lactose-binding lectin was purified from extracts of Ulex europaeus seeds by affinity chromatography on a column of galactose-Sepharose 4B, followed by gel filtration on Sephacryl S-300. This lectin, designated as Ulex europaeus lectin III (UEA-III), was found to be inhibited by lactose. The dimeric lectin is a glycoprotein with a molecular mass of 70,000 Da; it consists of two apparently identical subunits of a molecular mass of 34,000 Da. Compositional analysis showed that this lectin contains 30% carbohydrate and a large amount of aspartic acid, serine and valine, but no sulfur-containing amino acids. The N-terminal amino-acid sequences of L-fucose-binding Ulex europaeus lectin I (UEA-I) and di-N-acetylchitobiose-binding Ulex europaeus lectin II (UEA-II), both of which we have already purified and characterized, and that of UEA-III were determined and compared.

The effect of an alcoholic extract of seeds of Casimiroa edulis on blood pressure and heart rate was determined in rats anesthetized with pentobarbital and compared with that of histamine. The extract induced hypotension, accompanied at high doses by tachycardia. Hypotension after histamine was more transient and was not accompanied by changes in heart rate. Experiments with a variety of autonomic antagonists revealed that extract-induced hypotension was not mediated by histamine H2, muscarinic, or beta-adrenergic receptors, but involved an H1 mechanism. After H1 blockade, the depressor response was reversed to a pressor effect, mediated by alpha-adrenoceptor stimulation. The increase in heart rate was due in part to H1 and in part to beta-adrenergic receptor activation. It was suggested that imidazole derivatives could be responsible for the depressor effect observed. The pressor response could be caused by these or other components of the extract.

Two anti-H(O) lectins were separated from extracts of Cytisus sessilifolius seeds by successive affinity chromatographies on columns of di-N-acetylchitobiose- and galactose-Sepharose 4B. One was found to be inhibited most by di-N-acetylchitotriose or tri-N-acetylchitotriose [Cytisus-type anti-H(O) lectin designated as Cytisus sessilifolius lectin I (CSA-I)] and the other anti-H(O) lectin was inhibited by galactose or lactose and designated as Cytisus sessilifolius lectin II (CSA-II). These two anti-H(O) lectins were further purified by gel filtration on TSK-Gel G3000SW. These preparations were homogeneous as judged by polyacrylamide gel electrophoresis and gel filtration. The molecular masses of the purified lectins I and II were found to be 95,000 and 68,000 Da, respectively, by gel filtration on TSK-Gel G3000SW. On polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate and 2-mercaptoethanol, both lectins gave a single component of molecular masses of 27,000 +/- 2,000 and 34,000 +/- 2,000 Da, respectively, suggesting that the lectins I and II were composed of four and two apparently identical subunits, respectively. Lectins I and II contain 38% and 13% carbohydrate, respectively, and only very small amounts of cysteine and methionine, but they are rich in aspartic acid, serine and glycine. The N-terminal amino-acid sequences of these two lectins were determined and compared with those of several lectins already published.

The amino acid sequences of the major lectins from the seeds of Dioclea lehmanni and Canavalia maritima were determined by DABITC/PITC microsequence analysis of peptides derived from the proteins by enzymatic digestions with trypsin, chymotrypsin and the protease from S. aureus V8. These sequences were found to be very similar to those of the lectins from Dioclea grandiflora and Canavalia ensiformis (Con A). The D. lehmanni lectin was unusual amongst legume lectins in that it contained a single Cys.

Seeds of the tribal pulse, Mucuna gigantea, were analysed for proximate composition, total (true) seed proteins, seed protein fractionation, amino acid composition, minerals and antinutritional factors. Crude proteins, crude lipids, ash and nitrogen free extractives constituted 30.62%, 9.03%, 5.99% and 42.79%, respectively. The calorific values of 100 g dry matter of seed material is 374.91 kCal. The essential amino acids, leucine + isoleucine, are present in relatively large quantities. The seeds are rich in minerals like K, Ca, Mg and Fe. Antinutritional substances like total free phenols and tannins, trypsin inhibitor activity and haemagglutinating activity also were investigated.

Five new analogues of squash trypsin inhibitor CMTI III were obtained by the solid-phase method, exhibiting antitrypsin, antichymotrypsin or antielastase activity. Modification in the reactive site region changes dramatically the specificity, whereas substitution in non-contact positions facilitates the refolding of the reduced form of the peptides and does not have a significant influence on the association equilibrium constants of the inhibitor analogues with cognate enzymes.

The complete amino acid sequence of luffin-b has been determined. All the twenty-seven tryptic peptides were isolated by reverse-phase HPLC from the tryptic digests of intact luffin-b and one of its CNBr fragments (CB4), and sequenced using the DABITC/PITC double coupling method. The overlap of these peptides was achieved by analyzing the CNBr fragments and their chymotryptic peptides. Luffin-b consists of 250 amino acid residues with a relative molecular mass of 27,275 Da. Investigation for glycosylation sites indicated that Asn at positions 2, 78, and 85 might carry sugars. Sequence comparison with luffin-a showed that amino acid substitution occurred in 55 positions. Luffin-b contains three glycosylation sites instead of the six sites in luffin-a, of which two were found to be conserved.

The effects of various avocado oils on collagen metabolism in skin were studied in growing rats fed diets containing 10% (w/w) of the tested oils. Rats fed the unrefined avocado oil extracted with hexane from the intact fruit, its unsaponifiables or the avocado seed oil, showed significant increases in soluble collagen content in skin, though total collagen content was not affected. The increased soluble collagen content appears to be a consequence of the inhibition of lysyl oxidase activity. The active factor was found to be present in the unrefined avocado oil and probably originated from the avocado seed, since collagen metabolism was affected only by fractions which contained lipids fraction from the seed. In comparison rats fed the refined or unrefined soybean oils showed no effects.

Defatted tomato seed cake was analysed for its nutrient composition and its protein quality was evaluated in 21 day old male rats. The cake is found to be rich not only in protein (40%), minerals like phosphorus, calcium and magnesium but also in crude fiber. Lysine content of the cake protein was in the range of that found in groundnut cake protein. Sulphur containing amino acids were limiting in this protein. However, the protein quality of the cake, as judged by PER and NPU, was relatively lower compared to that of groundnut cake. Its high lysine content (5.1 g%) will be of help to improve the cereal based farm animal feed quality.

An extract of the seed of Sandoricum koetjape has yielded two new limonoids, sandoricin [1] and 6-hydroxysandoricin [2]. Both compounds are effective antifeedants when incorporated into artificial diets and fed to fall armyworm (Spodoptera frugiperda) or European corn borer (Ostrina nubilalis) larvae. Reduced growth rates and increased times to pupation were evident at lower dose levels while significant mortality was noted at higher dose levels. Structures of both compounds were determined by 1H nmr, 13C nmr, and ms and confirmed by X-ray crystallography.

The major inhibitor of trypsin in seeds of Prosopsis juliflora was purified by precipitation with ammonium sulphate, ion-exchange column chromatography on DEAE- and CM-Sepharose and preparative reverse phase HPLC on a Vydac C-18 column. The protein inhibited trypsin in the stoichiometric ratio of 1:1, but had only weak activity against chymotrypsin and did not inhibit human salivary or porcine pancreatic alpha-amylases. SDS-PAGE indicated that the inhibitor has a Mr of ca 20,000, and IEF-PAGE showed that the pI is 8.8. The complete amino acid sequence was determined by automatic degradation, and by DABITC/PITC microsequence analysis of peptides obtained from enzyme digestions of the reduced and S-carboxymethylated protein with trypsin, chymotrypsin, elastase, the Glu-specific protease from S. aureus and the Lys-specific protease from Lysobacter enzymogenes. The inhibitor consisted of two polypeptide chains, of 137 residues (alpha chain) and 38 residues (beta chain) linked together by a single disulphide bond. The amino acid sequence of the protein exhibited homology with a number of Kunitz proteinase inhibitors from other legume seeds, the bifunctional subtilisin/alpha-amylase inhibitors from cereals and the taste-modifying protein miraculin.

The study was aimed at evaluating the effect of sprouting on nitrogenous constituents and mineral composition of cream pigeon pea seeds. After 48 and 96 h of sprouting, there were increases in % moisture, crude protein, ash except during the 96 h; total nitrogen (TN), total non-protein nitrogen (TNPN); protein nitrogen (PN) and true protein nitrogen (TP). Sprouting caused increases in mineral levels except for phosphorus (P). Sprouting for 48 h offers greater advantage over 96 h.

The seeds of lesser-known species - Cucumeropsis mannii, Lagenaria sicceraria varieties 1 and 2 and Telfairia occidentalis, of Cucurbitaceae family were studied. The protein content of C. mannii was 36.1% and the varieties 1 and 2 of L. sicceraria had 32.1% and 33.3% respectively. Telfairia occidentalis had protein content of 33.2%. The fat contents of C. mannii, L. Sicceraria var. 1 and 2 and T. occidentalis were 44.4%, 44.6%, 46.9% and 42.3% respectively. The crude fibre content for each of the two varieties of L. sicceraria was 3.6% and T. occidentalis had 5.5%. Cucumeropsis mannii had the lowest (2.4%) fibre content. The carbohydrate contents for L. Sicceraria var. 2, C. mannii, and T. occidentalis were 12.6%, 13.2% and 14.4% respectively. Lagenaria sicceraria var. 1 had the highest value of carbohydrate (15.8%). The species were relatively rich in potassium and magnesium with range of values of 0.56% to 0.68% and 434 ppm to 444 ppm respectively. Cucumeropsis mannii and Lagenaria sicceraria var. 2 had relatively high contents of calcium (117 ppm) and iron (109 ppm) respectively. The kernels of these species exhibited lipase activity. Telfairia occidentalis showed the highest degree of lipase activity.

The paper reports the experimental result of the simultaneous quantitative determination of strychnine (ST) and brucine(BR) in Semen Strychni by dual wavelength spectrophotometry. The result shows that there are some deviations in the official method, because the wavelength combination might not have been chosen rightly and the correct coefficient is inconsistent with the facts. We have made a study of the thermal stability of ST and BR, too. On this basis a baking principle for Semen strychni is spelt out.

In this report we have compared the lectin-like properties of Pertussis toxin with two plant lectins which are known to possess different specificities towards terminal Neu5Ac Gal linkages on glycoconjugates. The hemagglutinin from elderberry bark (Sambucus nigra) has a binding specificity for terminal Neu5Ac alpha (2-6) Gal sequences and was found to bind a series of glycoconjugates with a similar specificity as Pertussis toxin. The binding specificity of Pertussis toxin was different from that of the leukoagglutinin from the seeds of Maackia amurensis which preferentially binds terminal Neu5Ac alpha (2-3) Gal sequences. These observations confirm the specificity of Pertussis toxin for Neu5Ac alpha (2-6) Gal glycoconjugate sequences.

A method is described for extracting lupin alkaloid (sparteine) and drug metabolites from different matrices (seeds and rat faeces) using microwave energy and for checking the homogeneity of the electric field in the microwave oven. The high-performance liquid chromatographic separation and determination of the extracted compounds showed that the microwave extraction method is more advantageous than other traditional extraction methods with regard to the yield of extraction and the time and cost of the procedure. The potential degradation of the extracted compounds may be considerably reduced.
