2. Progeny Selection and Genetic Diversity in a Pinus taeda Clonal Seed Orchard.

Abstract

The present article describes the development of an improved Pinus taeda clonal seed orchard adapted to the edaphoclimatic conditions of Uruguay. Initially, 2068 hectares distributed in nine companies were prospected, and 124 plus trees were identified based on growth, straightness, and health traits. These trees were clonally propagated via grafting to establish a clonal seed orchard. For the genetic evaluation of the orchard, two progeny tests were carried out in the Rivera and Paysandú municipalities. Quantitative genetic analyses allowed us to identify a simple genotype–environment interaction and an expected genetic gain for volumes of 17%, 13%, and 8% for selection intensities of 12%, 25%, and 50%, respectively. Moreover, the genetic diversity of the 124 clones of the orchard was assessed using 10 microsatellite markers. The fingerprinting profiles allowed us to identify a total of 224 alleles. The polymorphism information content of the different markers was in the range of 0.594 to 0.895. The combined probability of identity and probability of identity among siblings had a discrimination power of 8.26 × 10–14 and 5.91 × 10–5, respectively. Abstract

We investigated the utility of nuclear and cytoplasmic single nucleotide polymorphism (SNP) markers for timber tracking of the intensively logged and commercialized Amazonian tree Jacaranda copaia. Eight hundred and thirty-two trees were sampled (cambium or leaves) from 38 sampling sites in Bolivia, Brazil, French Guiana, and Peru. A total of 128 SNP markers (113 nuclear, 11 chloroplastic, and 4 mitochondrial) were used for genotyping the samples. Bayesian cluster analyses were carried out to group individuals into homogeneous genetic groups for tests to self-assign groups of individuals or individuals to their population of origin. Cluster analysis based on all the SNP markers detected seven main genetic groups. Genetic differentiation was high among populations (0.484) and among genetic groups (0.415), and populations showed a strong isolation-by-distance pattern. Self-assignment testing of the groups of individuals for all loci was able to determine the population origin of all the samples (accuracy = 100%). Self-assignment tests of individuals were able to assign the origin of 94.5%–100% of individuals (accuracy: 91.7%–100%). Our results show that the use of the 128 SNP markers is suitable to correctly determine the origin of J. copaia timber, and they should be considered a useful tool for customs and local and international police.

31. Genetic diversity and mating system of Euterpe precatoriain three localities along the lower Solimões River in Central Amazonia

Abstract

Euterpe precatoriais a Neotropical palm from South and Central America and is hyperdominant in Amazonia, where it is increasingly important in the Brazilian market for açaí. Genetic diversity, population structure and mating system of E. precatoriawere characterized with 10 microsatellite markers in three localities (Codajás, Manacapuru, and Manaquiri) along the lower Solimões River, Amazonas, Brazil. Leaves of 63 mature palms were collected, as well as fruits from 30 of these to analyze 20 seedlings per progeny. Genetic differentiation across localities was substantial (GS′= 0.304) for mature palms, suggesting that gene flow is restricted between localities due to isolation by distance. Inbreeding was significant in progenies from all populations (ranging from 0.059–0.076), but not for mature palms, suggesting selection for heterozygosity during maturation to the adult stage. The outcrossing rate was high (0.917–1.0), confirming that the species is predominantly allogamous. Matings were not random due to the occurrence of biparental inbreeding (0.021–0.079) and correlated matings (0.059–0.424), so open-pollinated progenies present mixtures of self-sibs, half-sibs, full-sibs, and self-half-sibs, resulting in mean effective population size within progenies (2.23–3.06) lower than expected for a random mating population. The mating system’s characteristics are those expected in a predominantly allogamous hyper-abundant palm and suggest that human management of these often-anthropogenic populations is unlikely to reduce genetic diversity in the short term if done in situ, as practiced by Indigenous Peoples and local communities. Intensification will require careful attention to maintain genetic diversity.

Abstract

Euterpe precatoria (Açaí-do-Amazonas) produces fruits of which the fresh pulp is consumed. It is almost exclusively collected by extractivist farmers, because no selected genotypes are available for the establishment of plantations. For the domestication and breeding of the species, mating system studies are needed for strategy formulation. This study evaluated the mating system of a natural population of E. precatoria. Thirteen progenies were genotyped with 13 microsatellite loci by capillary electrophoresis in an automated DNA sequencer. Estimates of single-locus and multilocus outcrossing rates were 1.0, and paternity correlation was low ( r ^ p ( m )>= 0.293). Euterpe precatoria families consist mainly of half-sibs and the reproductive strategy of the species is allogamy.

Abstract

Guazuma crinitais a fast-growing tree with potential for use in agroforestry systems, due to its rapid wood production, which can contribute significantly to the livelihoods of small-scale farmers in the Peruvian Amazon. However, indiscriminate logging due to high demand is leading to the disappearance of natural forests. As such, the International Council for Research in Agroforestry (ICRAF) began a domestication program to reduce pressure on natural populations of the species. The objective of the present study was to use analyses of genetic parameters, adaptability (PRVG), productivity (MHPRVG), and stability (MHVG), to select G. crinitagenotypes from a three-year-old progeny test established in the Aguaytía River Basin, in Ucayali, Peru. The test was established in three different sites, with three blocks, 200 progeny per block, and two individuals per plot. The measured traits were diameter at breast height (DBH), total height (H), and total aerial biomass (B). Significant differences in traits between progenies were detected, but with no genotype x environment interaction (GxE). However, the genotypic correlation among sites was important (> 0.702), suggesting that genetic improvement is possible by selecting the same progeny across sites. The mean heritability among progenies was moderate for all traits (0.34–0.369) and selective precision through combined site analysis was relatively high (0.583–0.608). Based on selection for DBH through combined analysis, MHVG, PRVG, and MHPRVG, 50 superior progenies (25.9 %) were identified for all environments. These should be prioritized in breeding programs as they can offer stable genetic variability for future selection cycles.
