Drought is an important environmental factor limiting the productivity of various crops worldwide. The development of crop cultivars with improved adaptation to drought is a major goal in many crop breeding programs. In addition to classical breeding approaches, genetic transformation to introduce candidate genes into plants for better tolerance to water deficit has been successfully developed. Pathogenesis-related proteins (PR proteins) are defined as plant proteins induced in response to pathogen attack. However, it is known that these proteins may also be involved in response to abiotic stresses. The objective of this study was to transform tobacco plants (as plant model for subsequent analysis of cultivated plant such as citrus) with a PR-4b protein from Theobroma cacao (TcPR-4b) and to select and test the tolerance of such transformed plants to water/osmotic stress. First, an in silico analysis of the TcPR-4b using the BLAST, Pfam,InterProScan, ORF-Finder programs, as well as a search on Cocoa GenDB databank were performed. The TcPR-4b belongs to a small family of PR-4 proteins whose members were mainly located on the chromosomes 5 (five genes) and 10 (one gene). The complete TcPR-4b sequence is 802 bp in length and contains two exons (171 and 258 bp), and one intron (82 bp); the corresponding protein is 142 amino acids in length. For plant transformation experiment, the TcPR-4b cDNA (from cacao-M. perniciosa interaction library) was cloned into the pGem- T Easy vector then subcloned on the pCambia binary vector 1390. Then, Agrobacterium tumefaciens strain EHA 105 was transformed with the35S::TcPR-4b::pCambia construction, and the transformed A. tumefaciens used for Nicotiana tabacum cv.Havana transformation by co-cultivation of leaf segments in selection medium. Transformed shoots from three transformation events are under selection for subsequent in vitro water/osmotic stress, using, among others, mannitol and NaCl. RESUMO - Com objetivo de estudar diversidade genética e identificar marcadores associados à resistência ao míldio (Plasmopara viticola) e ao oídio (Uncinula necator), foram analisadas as cultivares de videira A 1976, CG 87746, CNPUV 154-27, Crimson Seedless, Gota de Ouro, Itália, Seyve Villard 12327 e Seyve Villard 12375. As análises de polimorfismo de comprimento de fragmento amplificado (AFLP) seguiram as etapas de digestão, ligação, pré-amplificação e amplificação. Efetuou-se a separação dos fragmentos amplificados em gel de 7% de poliacrilamida desnaturante (uréia 7M) e coloração com nitrato de prata. A similaridade foi estimada com base no coeficiente de Jaccard, e a agregação, por UPGMA. Foi gerada uma matriz de similaridade com bom ajustamento da agregação (r = 0,84). Os agrupamentos obtidos corresponderam à origem e classificação botânica das cultivares. Foram identificadas 15 marcas dissimilares associadas à resistência, sendo oito para míldio e sete para oídio. ABSTRACT - The grapevine cultivars A 1976, CG 87746, CNPUV 154-27, Crimson Seedless, Gota de Ouro, Itália, Seyve Villard 12327 and Seyve Villard 12375 were analyzed to establish the genetic relationship and to identify markers associated with resistance to downy mildew (Plasmopara viticola) and powdery mildew (Uncinula necator). The amplified fragment lenght polymorphism (AFLP) analysis comprise the steps of digestion, ligation, preamplification and amplification. The amplified fragments were separeted on a 7% denaturing polyacrylamide gel (7M urea) and silver stained. The similarity was estimated using Jaccard?s coefficient and the clustering was estimated by UPGMA. A similarity matrix was generated with a good fit for aggregation (r = 0,84). The clusters corresponded to origin and botany classification of the cultivars. Fifteen markers were associated with resistance, eight for downy mildew and seven for powdery mildew. Abstract Background The cultivar Micro-Tom (MT) is regarded as a model system for tomato genetics due to its short life cycle and miniature size. However, efforts to improve tomato genetic transformation have led to protocols dependent on the costly hormone zeatin, combined with an excessive number of steps. Results Here we report the development of a MT near-isogenic genotype harboring the allele Rg1 (MT-Rg1), which greatly improves tomato in vitro regeneration. Regeneration was further improved in MT by including a two-day incubation of cotyledonary explants onto medium containing 0.4 μM 1-naphthaleneacetic acid (NAA) before cytokinin treatment. Both strategies allowed the use of 5 μM 6-benzylaminopurine (BAP), a cytokinin 100 times less expensive than zeatin. The use of MT-Rg1 and NAA pre-incubation, followed by BAP regeneration, resulted in high transformation frequencies (near 40%), in a shorter protocol with fewer steps, spanning approximately 40 days from Agrobacterium infection to transgenic plant acclimatization. Conclusions The genetic resource and the protocol presented here represent invaluable tools for routine gene expression manipulation and high throughput functional genomics by insertional mutagenesis in tomato.

Key Message: Transgenic sugarcane expressing V-ATPase subunit E dsRNA affects growth and survival of Sphenophorus levis. Plants being sessile organisms are constantly confronted with several biotic and abiotic stresses. Sugarcane (Saccharum spp) is a major tropical crop widely cultivated for its sugar and other by-products. In Brazil, sugarcane plantations account for significant production losses due to Sphenophorus levis (sugarcane weevil) infestations. With the existing control measures being less effective, there arises a necessity for advanced strategies. Our bioassay injection experiments with V-ATPase E dsRNA in S. levis larvae showed significant mortality and reduction in transcription levels. Furthermore, we down-regulated the V-ATPase E gene of S. levis in transgenic sugarcane using an RNAi approach. The resultant RNAi transgenic lines exhibited reduction in larval growth and survival, without compromising plant performance under controlled environment. Our results illustrate that RNAi-mediated down-regulation of key genes is a promising approach in imparting resistance to sugarcane weevil.

- Abstract
Background: Globally, osteoarthritis (OA) is the third condition associated with disability. There is still poor treatment in OA but science holds the key to finding better treatments and a cure. It is essential to learn what's important to patients from them to implement the most effective OA management. The OA Patients Task Force, conducted the Global OA Patient Perception Survey (GOAPPS)-the first global survey made by patients to analize the quality of life (QoL) & patient perceptions of care. The goal was to collect data on OA patients' perception of OA to understand patients' needs and expectations to improve OA management., Methods: Observational, cross-sectional study by online survey data collection from six countries, translated into three languages. The questionnaire was comprised of 3 sections: patient demographics and clinical symptomology characteristics; relationship with physicians: perception of attention, treatment, and information provided; and OA impact on daily activity and QoL. The results of the survey were evaluated using the Limited Data Set. The survey results were analyzed using descriptive statistics to characterize the patients' answers. Additionally, Cronbach's alpha was calculated to determine internal consistency validity., Results: A total of 1512 surveys were completed in 6 countries. 84.2% of respondents reported pain/tenderness and 91.1% experienced limitations to physical activities. 42.3% of patients were not satisfied with their current OA treatment. 86% had comorbidities, especially hypertension, and obesity. 51.3 and 78% would like access to additional drug or additional non-drug/non-surgical treatments respectively. 48.2% of patients perceived their QoL to be affected by OA. The Cronbach's alpha was 0.61., Conclusions: OA has a significant impact on patients' daily activities and their desire to play an active role in managing this disease. Patients are seeking additional treatments, especially no pharmacological/no surgical treatments stressing the need for investing in clinical research, implementing OA preventive measures, and managing interventions to improve the healthcare value chain in OA.

Background: Knee osteoarthritis (KOA) is a prevalent form of chronic joint disease associated with functional restrictions and pain. Activity limitations negatively impact social connectedness and psychological well-being, reducing the quality of life (QoL) of patients. The purpose of this review is to summarize the existing information on QoL in KOA patients and share the reported individual factors, which may influence it., Methods: We conducted a systematic review examining the literature up to JAN/2017 available at MEDLINE, EMBASE, Cochrane, and PsycINFO using KOA and QOL related keywords. Inclusion criteria were QOL compared to at least one demographic factor (e.g., age, gender), lifestyle factor (e.g., functional independence), or comorbidity factor (e.g., diabetes, obesity) and a control group. Analytical methods were not considered as part of the original design., Results: A total of 610 articles were reviewed, of which 62 met inclusion criteria. Instruments used to measure QoL included: SF-36, EQ-5D, KOOS, WHOQOL, HAS, AIMS, NHP and JKOM. All studies reported worse QoL in KOA patients when compared to a control group. When females were compared to males, females reported worse QOL. Obesity as well as lower level of physical activity were reported with lower QoL scores. Knee self-management programs delivered by healthcare professionals improved QoL in patients with KOA. Educational level and higher total mindfulness were reported to improve QoL whereas poverty, psychological distress, depression and lacking familial relationships reduce it. Surgical KOA interventions resulted in good to excellent outcomes generally; although, results varied by age, weight, and depression., Conclusion: KOA has a substantial impact on QoL. In KOA patients, QoL is also influenced by specific individual factors including gender, body weight, physical activity, mental health, and education. Importantly, education and management programs designed to support KOA patients report improved QoL. QoL data is a valuable tool providing health care professionals with a better comprehension of KOA disease to aid implementation of the most effective management plan.

Rising costs and slower economic growth are resulting in progressive stress on the current structure of health care finance in Australia. As in other developed countries, managed competition offers a potential solution, by increasing system efficiency. The case for restructuring current financing arrangements is even stronger than in other countries because of inefficiencies arising from inconsistencies and overlaps between Commonwealth, State and private sector functions. This paper outlines an illustrative model for managed competition in Australia. In addition, it discusses in more detail two specific issues which have received some attention in the literature: whether to have multiple payers, in the form of competing public and private budget holders; and whether 'catastrophic risks' such as long-term institutional care should be incorporated in a managed competition program.
