11. Optimization of a nanoparticle uptake protocol applied to amniotic-derived cells: unlocking the therapeutic potential.

Abstract

Stem cell-based therapy implementation relies heavily on advancements in cell tracking. The present research has been designed to develop a gold nanorod (AuNR) labeling protocol applied to amniotic epithelial cells (AECs) leveraging the pro-regenerative properties of this placental stem cell source which is widely used for both human and veterinary biomedical regenerative applications, although not yet exploited with tracking technologies. Ovine AECs, in native or induced mesenchymal (mAECs) phenotypes via epithelial–mesenchymal transition (EMT), served as the model. Initially, various uptake methods validated on other sources of mesenchymal stromal cells (MSCs) were assessed on mAECs before optimization for AECs. Furthermore, the protocol was implemented by adopting the biological strategy of MitoCeption to improve endocytosis. The results indicate that the most efficient, affordable, and easy protocol leading to internalization of AuNRs in living mAECs recognized the combination of the one-step uptake condition (cell in suspension), centrifugation-mediated internalization method (G-force) and MitoCeption (mitochondrial isolated from mAECs). This protocol produced labeled vital mAECs within minutes, suitable for preclinical and clinical trials. The optimized protocol has the potential to yield feasible labeled amniotic-derived cells for biomedical purposes: up to 10 million starting from a single amniotic membrane. Similar and even higher efficiency was found when the protocol was applied to ovine and human AECs, thereby demonstrating the transferability of the method to cells of different phenotypes and species-specificity, hence validating its great potential for the development of improved biomedical applications in cell-based therapy and diagnostic imaging. Abstract

Simple Summary: This study reported for the first time in Italy the determination of 4,6-dinitro-ortho-cresol (DNOC) in the baits and gastric contents of poisoned animals. The determination of DNOC was achieved by using gas chromatography coupled to mass spectrometry (GC-MS) with both full-scan and selected ion monitoring (SIM) modes (ion m/z values 198, 121, and 105). This study reports the determination of DNOC in the baits and gastric contents of poisoned dogs and wild canids collected in the Abruzzo region between 2014 and 2022. A total of 663 samples of the baits and carcasses of domestic and wild animals were analyzed for the determination of DNOC through GC-MS. DNOC was found to be present in 58 cases from 663 analyzed samples, with 49 poison baits and nine gastric content samples from dogs and red foxes. This study confirms for the first time that the banned pesticide DNOC still poisons both domestic and wild animals in the Abruzzo region. It should be considered that this study was carried out on a population of animals from a regional geographical area, and more detailed country-wide studies need to be carried out to obtain the incidence of poisoning from this herbicide in Italy. Abstract

BackgroundThe Caserta and Naples areas in Campania Region experience heavy environmental contamination due to illegal waste disposal and burns, thus representing a valuable setting to develop a general model of human contamination with dioxins (PCDDs-PCDFs) and dioxin-like-PCBs (dl-PCBs).Methods94 breastfeeding women (aged 19-32 years; mean age 27.9 ± 3.0) were recruited to determine concentrations of PCDDs-PCDFs and dl-PCBs in their milk. Individual milk samples were collected and analyzed according to standard international procedures. A generalized linear model was used to test potential predictors of pollutant concentration in breast milk: age, exposure to waste fires, cigarette smoking, diet, and residence in high/low risk area (defined at high/low environmental pressure by a specific 2007 WHO report). A Structural Equation Model (SEM) analysis was carried out by taking into account PCDDs-PCDFs and dl-PCBs as endogenous variables and age, waste fires, risk area and smoking as exogenous variables.ResultsAll milk samples were contaminated by PCDDs-PCDFs (8.6 pg WHO-TEQ/98g fat ± 2.7; range 3.8-19) and dl-PCBs (8.0 pg WHO-TEQ/98g fat ± 3.7; range 2.5-24), with their concentrations being associated with age and exposure to waste fires (p < 0.01). Exposure to fires resulted in larger increases of dioxins concentrations in people living in low risk areas than those from high risk areas (p < 0.01).ConclusionsA diffuse human exposure to persistent organic pollutants was observed in the Caserta and Naples areas. Dioxins concentration in women living in areas classified at low environmental pressure in 2007 WHO report was significantly influenced by exposure to burns.

Abstract

In the present study, 334 samples of mussels (Mytilus galloprovincialis) harvested along the coasts of the Central Adriatic Sea during the years 2020–2021 were analyzed for the presence of lipophilic marine biotoxins according to the European Harmonized Standard Operating Procedure. The results showed that 74 (22%) and 84 (25%) samples were positive to okadaic acid and yessotoxin groups, respectively. Among them, only 11 (3.3%) samples resulted as non‐compliant, as they exceeded the maximum limits (160 μg okadaic acid equivalent/kg) established by the Regulation (EC) 853/2004. The method applied in this study was able to detect and quantify lipophilic marine biotoxins concentrations, in order to monitor their presence in molluscs and avoid the risk of consumer exposure. Abstract

Along the Italian coasts, toxins of algal origin in wild and cultivated shellfish have been reported since the 1970s. In this study, we used data gathered by the Veterinary Public Health Institutes and the Italian Environmental Health Protection Agencies from 2006 to 2019 to investigate toxicity events and relate them to the distribution of toxic species. Among detected toxins (OA and analogs, YTXs, PTXs, STXs, DAs, AZAs), OA and YTX were those most frequently reported in cases of seafood contamination. Levels of those toxins exceeding regulatory limits for OA were associated with high abundances of Dinophysis spp. and for YTX with blooms of Gonyaulax spinifera, Lingulodinium polyedra, and Protoceratium reticulatum. Seasonal blooms of Pseudo-nitzschia spp. occurred all along the Italian coast, but DA was only occasionally found in shellfish, at concentrations consistently below the regulatory limit. Alexandrium was recorded in many more areas than STXs, which only rarely exceeded the regulatory limit. Azadinium was sporadically recorded, and AZAs were sometimes detected in low quantities. Among the emerging toxins, PLTX-like toxins often accumulated in wild mussels and sea urchins due to the occurrences of Ostreopsis cf. ovata. Overall, Italian coastal waters harbour numerous potentially toxic species, with a few HAB hotspots related to DSP toxins. Abstract

To implement European Union Directive 78/659/EEC concerning the quality of fresh waters that require protection or improvement in order to support fish life, the Abruzzo Regional Council commissioned the Istituto Zooprofilattico Sperimentale dell'Abruzzo e del Molise 'G. Caporale' (by Regional Law No. 50 of 10 August 1994, to conduct a census of the surface freshwaters in the region, and to classify them. The authors describe the freshwater classifications obtained through monitoring campaigns conducted over a period of twelve years (1996 to 2008). The results indicate slight improvements in the quality of the water over time in terms of contamination by chemicals for which they were monitored; however, 14 stretches do not comply with the requirements of the European Union Directive.
