Spinocerebellar ataxia type 7 (SCA7) is a genetic neurodegenerative disorder caused by a CAG-polyglutamine repeat expansion. Purkinje cells (PCs) are central to the pathology of ataxias, but their low abundance in the cerebellum underrepresents their transcriptomes in sequencing assays. To address this issue, we developed a PC enrichment protocol and sequenced individual nuclei from mice and patients with SCA7. Single-nucleus RNA sequencing in SCA7-266Q mice revealed dysregulation of cell identity genes affecting glia and PCs. Specifically, genes marking zebrin-II PC subtypes accounted for the highest proportion of DEGs in symptomatic SCA7-266Q mice. These transcriptomic changes in SCA7-266Q mice were associated with increased numbers of inhibitory synapses as quantified by immunohistochemistry and reduced spiking of PCs in acute brain slices. Dysregulation of zebrin-II cell subtypes was the predominant signal in PCs of SCA7-266Q mice and was associated with the loss of zebrin-II striping in the cerebellum at motor symptom onset. We furthermore demonstrated zebrin-II stripe degradation in additional mouse models of polyglutamine ataxia and observed decreased zebrin-II expression in the cerebella of patients with SCA7. Our results suggest that a breakdown of zebrin subtype regulation is a shared pathological feature of polyglutamine ataxias.

Policy makers increasingly call on higher education to prepare learners for challenges such as global health emergencies or ecological crises. These can be understood as 'wicked problems', which are unbounded, complex and resist simplistic definition. Wicked problems involve stakeholders with incompatible value positions and attempted solutions can result in unforeseen outcomes. How academics stay committed to teaching about such challenging topics -- despite the many difficulties of contemporary higher education -- is an under-researched area. In this study, we interviewed academics who were deeply engaged with teaching about wicked problems. We drew on the concepts of landscapes of practice, boundary work and academic identities to make sense of the teachers' persistence and practices in this space. We conclude with advice for policy makers on how to support academics in this work.

Purpose: To report a case of a large foveal sub-internal limiting membrane hemorrhage from sickle cell retinopathy in a pediatric patient. Observations: A five-year-old boy with sickle cell disease (SCD) type SS (HbSS) and numerous complications was referred after a failed vision screening and was found to have a large yellow subacute sub-internal limiting membrane hemorrhage overlying the fovea in his right eye. There were several other peripheral salmon patches noted. Optical coherence tomography (OCT) revealed temporal inner retinal macular thinning in both eyes. Serial imaging showed rapid improvement over time of the hemorrhages, though amblyopia persisted. Conclusions and importance: We describe an unusual amblyogenic presentation of non-proliferative sickle cell retinopathy in five-year-old patient with HbSS due to a foveal salmon patch. Numerous vision-threatening complications are possible in SCD, highlighting the need for early vision screening.

The cGAS-STING innate immunity pathway and the SREBP-activated cholesterol and fatty acid synthesis pathway are abnormally co-regulated in neurodegenerative disease. Activation of STING signaling occurs at the endoplasmic reticulum (ER) membrane with STING anchored by INSIG1 along with SREBP and the sterol-bound SREBP cleavage activating protein (SCAP) when sterols are in abundance. When sterols are low, the INSIG-dependent STING pathway is inactivated and the SREBP-SCAP complex is translocated to the Golgi where SREBP is cleaved and translocated to the nucleus to transactivate genes for cholesterol and fatty acid synthesis. Thus, there is inverse activation of STING vs. SREBP: when innate immunity is active, pathways for cholesterol and fatty acid synthesis are suppressed, and vice versa. The STING pathway is stimulated by foreign viral cytoplasmic nucleic acids interacting with the cyclic GMP–AMP synthase (cGAS) DNA sensor or RIG-I and MDA5 dsRNA sensors, but with neurodegeneration innate immunity is also activated by self-DNAs and double-stranded RNAs that accumulate with neuronal death. Downstream, activated STING recruits TBK1 and stimulates the transactivation of interferon stimulated genes and the autophagy pathway, which are both protective. However, chronic activation of innate immunity contributes to microglia activation, neuroinflammation and autophagy failure leading to neurodegeneration. STING is also a proton channel that when activated stimulates proton exit from STING vesicles leading to cell death. Here we review the salient features of the innate immunity and cholesterol and fatty acid synthesis pathways, observations of abnormal STING and SREBP signaling in neurodegenerative disease, and relevant therapeutic approaches.

This paper reports on teachers' perspectives on preparing students for working with 'wicked' problems (Rittel and Webber [1973]. 'Dilemmas in a General Theory of Planning.' "Policy Sciences" 4 (2): 155-169.). These problems are complex, lack clear boundaries, and attempts to solve them -- generally by bringing together multiple stakeholders with contrasting viewpoints -- have unforeseen consequences. Examples include many of the most significant current global challenges. We conducted semi-structured interviews with twenty teachers who focused on wicked problems, and a comparison group of 15 . We used the theoretical lenses 'ways of thinking and practising in the subject area' (Anderson and Hounsell [2007]. 'Knowledge Practices: 'Doing the Subject' in Undergraduate Courses.' "The Curriculum Journal" 18 (4): 463-478. ) and 'figured worlds' (Holland et al. [1998]. "Identity and Agency in Cultural Worlds." Cambridge, MA: Harvard University Press.) to frame our analysis. Our findings elaborate four key aspects of learning for wicked problems.

Theileria orientalis Ikeda, an emerging U.S. bovine hemoparasite, causes anemia, abortion, ill-thrift, and occasionally death. While Haemaphysalis longicornis is the primary vector, it is possible that other U.S. ticks are capable of parasite transmission and may contribute to disease spread. Dermacentor variabilis is highly prevalent in the U.S., exhibits a similar geographical distribution to T. orientalis, and is a competent vector of the related parasite, Theileria equi. Herein, we conducted controlled acquisition and transmission studies using splenectomized calves to assess whether D. variabilis can transstadially transmit T. orientalis. D. variabilis nymphs were applied to an infected, splenectomized calf for parasite acquisition and subsequently incubated to molt into adults. Freshly molted adults were applied to two splenectomized T. orientalis-naïve calves to investigate parasite transmission. Calves were monitored for 59 days, and no evidence of parasite transmission was detected using PCR for the T. orientalis Ikeda major piroplasm surface protein gene, blood smear cytology, complete blood counts, or physical examination. Salivary glands from a subset of D. variabilis adults were assessed for T. orientalis using PCR, and the parasite was not detected. These findings support the conclusion that D. variabilis is not capable of transstadial transmission of the U.S. T. orientalis Ikeda isolate.

The UCSF Mouse Inventory Database Application is an open-source Web App that provides information about the mutant alleles, transgenes, and inbred strains maintained by investigators at the university and facilitates sharing of these resources within the university community. The Application is designed to promote collaboration, decrease the costs associated with obtaining genetically-modified mice, and increase access to mouse lines that are difficult to obtain. An inventory of the genetically-modified mice on campus and the investigators who maintain them is compiled from records of purchases from external sources, transfers from researchers within and outside the university, and from data provided by users. These data are verified and augmented with relevant information harvested from public databases, and stored in a succinct, searchable database secured on the university network. Here we describe this resource and provide information about how to implement and maintain such a mouse inventory database application at other institutions.

Direct air capture (DAC) is an emerging negative CO2 emission technology that aims to introduce a feasible method for CO2 capture from the atmosphere. Unlike carbon capture from point sources, which deals with flue gas at high CO2 concentrations, carbon capture directly from the atmosphere has proved difficult due to the low CO2 concentration in ambient air. Current DAC technologies mainly consider sorbent-based systems; however, membrane technology can be considered a promising DAC approach since it provides several advantages, e.g., lower energy and operational costs, less environmental footprint, and more potential for small-scale ubiquitous installations. Several recent advancements in validating the feasibility of highly permeable gas separation membrane fabrication and system design show that membrane-based direct air capture (m-DAC) could be a complementary approach to sorbent-based DAC, e.g., as part of a hybrid system design that incorporates other DAC technologies (e.g., solvent or sorbent-based DAC). In this article, the ongoing research and DAC application attempts via membrane separation have been reviewed. The reported membrane materials that could potentially be used for m-DAC are summarized. In addition, the future direction of m-DAC development is discussed, which could provide perspective and encourage new researchers’ further work in the field of m-DAC.

This article adds to the corpus of writing that examines collegiality's idealistic yet elusive nature in Higher Education by focusing specifically on how collegiality can be enacted in student--staff partnership work. An innovative initiative, 'Students as Colleagues in the Review of Teaching Practices', provides a case to qualitatively explore the collegial characteristics. Here, students reviewed a paired staff member's teaching practice over one semester, as a peer review exercise. This shift in social roles troubles the understanding of 'peer' and asks if authentic collegiality is possible. The study is conceptually framed by Fielding's notion of 'radical collegiality' and draws on standpoint theory and dialogic education to raise issues of power, mutual support and productive tensions. The findings have implications for how Higher Education institutions can support student and staff to actively engage in authentic collegial partnerships by developing relational and dialogic spaces, rather than just abstract institutional infrastructures.

Recovering species are often limited to much smaller areas than they historically occupied. Conservation planning for the recovering species is often based on this limited range, which may simply be an artifact of where the surviving population persisted. Southern sea otters (Enhydra lutris nereis) were hunted nearly to extinction but recovered from a small remnant population on a remote stretch of the California outer coast, where most of their recovery has occurred. However, studies of recently-recolonized estuaries have revealed that estuaries can provide southern sea otters with high quality habitats featuring shallow waters, high production and ample food, limited predators, and protected haul-out opportunities. Moreover, sea otters can have strong effects on estuarine ecosystems, fostering seagrass resilience through their consumption of invertebrate prey. Using a combination of literature reviews, population modeling, and prey surveys we explored the former estuarine habitats outside the current southern sea otter range to determine if these estuarine habitats can support healthy sea otter populations. We found the majority of studies and conservation efforts have focused on populations in exposed, rocky coastal habitats. Yet historical evidence indicates that sea otters were also formerly ubiquitous in estuaries. Our habitat-specific population growth model for Californias largest estuary-San Francisco Bay-determined that it alone can support about 6,600 sea otters, more than double the 2018 California population. Prey surveys in estuaries currently with (Elkhorn Slough and Morro Bay) and without (San Francisco Bay and Drakes Estero) sea otters indicated that the availability of prey, especially crabs, is sufficient to support healthy sea otter populations. Combining historical evidence with our results, we show that conservation practitioners could consider former estuarine habitats as targets for sea otter and ecosystem restoration. This study reveals the importance of understanding how recovering species interact with all the ecosystems they historically occupied, both for improved conservation of the recovering species and for successful restoration of ecosystem functions and processes.

The genetically heterogeneous spinocerebellar ataxias (SCAs) are caused by Purkinje neuron dysfunction and degeneration, but their underlying pathological mechanisms remain elusive. The Src family of nonreceptor tyrosine kinases (SFK) are essential for nervous system homeostasis and are increasingly implicated in degenerative disease. Here we reveal that the SFK suppressor Missing-in-metastasis (MTSS1) is an ataxia locus that links multiple SCAs. MTSS1 loss results in increased SFK activity, reduced Purkinje neuron arborization, and low basal firing rates, followed by cell death. Surprisingly, mouse models for SCA1, SCA2, and SCA5 show elevated SFK activity, with SCA1 and SCA2 displaying dramatically reduced MTSS1 protein levels through reduced gene expression and protein translation, respectively. Treatment of each SCA model with a clinically approved Src inhibitor corrects Purkinje neuron basal firing and delays ataxia progression in MTSS1 mutants. Our results identify a common SCA therapeutic target and demonstrate a key role for MTSS1/SFK in Purkinje neuron survival and ataxia progression.

Abstract Background There have been ongoing efforts to identify anti-tick vaccine targets to protect cattle from infestation with cattle fever ticks Rhipicephalus (Boophilus) microplus. Two commercial vaccines based on the tick gut protein Bm86 have had variable effectiveness, which has led to poor acceptance, and numerous studies have attempted to identify vaccine antigens that will provide more consistently effective protection. Transcriptomic analysis of R. microplus led to identification of three aquaporin genes annotated to code for transmembrane proteins involved in the transport of water across cell membranes. Previous work showed that vaccination with full-length recombinant aquaporin 1 (RmAQP1) reduced tick burdens on cattle. Targeted silencing of aquaporin 2 (RmAQP2) expression suggested it might also be a good anti-tick vaccination target. Methods Three synthetic peptides from the predicted extracellular domains of RmAQP2 were used to vaccinate cattle. Peptides were conjugated to keyhole limpet hemocyanin (KLH) as an antigenic carrier molecule. We monitored the antibody response with ELISA and challenged vaccinated cattle with R. microplus larvae. Results There was a 25% reduction overall in the numbers of ticks feeding to repletion on the vaccinated cattle. Immune sera from vaccinated cattle recognized native tick proteins on a western blot and reacted to the three individual synthetic peptides in an ELISA. The vaccinated calf with the highest total IgG titer was not the most effective at controlling ticks; ratios of IgG isotypes 1 and 2 differed greatly among the three vaccinated cattle; the calf with the highest IgG1/IgG2 ratio had the fewest ticks. Ticks on vaccinated cattle had significantly greater replete weights compared to ticks on controls, mirroring results seen with RNA silencing of RmAQP2. However, protein data could not confirm that vaccination had any impact on the ability of the tick to concentrate its blood meal by removing water. Conclusions A reduced number of ticks feed successfully on cattle vaccinated to produce antibodies against the extracellular domains of RmAQP2. However, our predicted mechanism, that antibody binding blocks the ability of RmAQP2 to move water out of the blood meal, could not be confirmed. Further study will be required to define the mechanism of action and to determine whether these vaccine targets will be useful components of an anti-tick vaccine cocktail. Graphical Abstract

Accreditation processes for health care professions are designed to ensure that individuals and programs in these fields meet established standards of quality and effectiveness. The accelerating pace of globalization in the health care professions has increased the need for a shared understanding of the vocabulary of evaluation, assessment, and accreditation. The psychometric principles of valid and reliable assessment are commonly accepted, but the terminology is confusing. We believe that all stakeholders – evaluators, faculty, students but also the community – will benefit from a shared language and common set of definitions. We recognize that not all readers will agree with the definitions we propose, but we hope that this guide will help to ensure clarity, consistency, transparency, and fairness, and that it will promote through the stimulation of a debate greater collaboration across national and international boundaries.

Abstract Background Vector-borne diseases pose an increasing threat to global food security. Vaccines, diagnostic tests, and therapeutics are urgently needed for tick-borne diseases that affect livestock. However, the inability to obtain significant quantities of pathogen stages derived from ticks has hindered research. In vitro methods to isolate pathogens from infected tick vectors are paramount to advance transcriptomic, proteomic, and biochemical characterizations of tick-borne pathogens. Methods Nymphs of Rhipicephalus appendiculatus were infected with Theileria parva by feeding on a calf during an acute infection. Isolation of sporozoites was accomplished by feeding infected adult ticks on an in vitro tick feeding system. Sporozoite viability was tested using in vitro bovine lymphocytes. Results We isolated infectious T. parva sporozoites secreted into an in vitro tick feeding system. Infected adult R. appendiculatus ticks attached to and successfully fed on silicone membranes in the in vitro tick feeding system. Bovine blood in the receptacle was replaced with cell-free medium and the ticks were allowed to feed for 3 h to collect secreted T. parva sporozoites. Secreted sporozoites infected in vitro bovine lymphocytes, demonstrating that isolated sporozoites remained viable and infectious. Conclusions This work is the first to report the isolation of mature infectious T. parva sporozoites using an in vitro tick feeding system, which represents a significant step towards the development of a more efficient control strategy for T. parva. Isolation of infectious tick-stage parasites will facilitate the examination of the vector-pathogen interface, thereby accelerating the development of next-generation vaccines and treatment interventions for tick-borne pathogens. Purpose: To describe the clinical characteristics, imaging findings and genetic testing results of a young simplex male with choroideremia. Observations: A 6-year-old Hispanic-Chinese male was referred to the retina clinic for peripheral retinal pigmentary changes observed in both eyes on routine exam. The patient has an unremarkable family history and developmental history. Best corrected visual acuity was 20/25 in both eyes. Optical coherence tomography demonstrated attenuation of the ellipsoid and interdigitation zones. Widefield fundus autofluorescence demonstrated nummular hypo-autofluorescence peripherally in both eyes. Genetic testing revealed a variant originally described as a variant of uncertain significance (VUS) a c. 1775_1814del (p.Glu592Valfs*44) identified in the CHM gene, which was reclassified as pathogenic following segregation analysis. The patient was diagnosed with choroideremia due to a CHM pathogenic variant. Conclusions: The multimodal imaging findings demonstrated here illustrate important clues to the diagnosis of Choroideremia in a simplex male.

Abstract Volunteer monitoring can support conservation of imperiled wildlife, by providing higher resolution data in space and time than those available from professional scientists. However, concerns have been raised that data collected by amateurs are inaccurate or inconsistent and thus do not allow for robust detection of spatial or temporal trends. We evaluated the rigor and value of volunteer monitoring data for one iconic wildlife species, the southern sea otter (Enhydra lutris nereis), in Elkhorn Slough estuary in central California, USA, and explored whether volunteer monitoring could provide added value to complement limited professional surveys. First, we compiled and analyzed sea otter counts taken on daily ecotourist boat trips along the estuary, and then compared temporal patterns to data collected by professional scientists tasked with monitoring this federally listed species. Second, we analyzed data on sea otter abundance, habitat use, and behavior collected by a team of trained volunteers, the Elkhorn Slough Reserve Otter Monitoring Program. Overall, we demonstrated the ability to detect important ecological patterns relevant to sea otter conservation and wetland habitat management using volunteer‐derived datasets. Long‐term trends and inter‐annual variability were similar between professional agency monitoring data and volunteer datasets. Moreover, the much higher frequency of volunteer observations allowed for seasonal and tidal dynamics to be detected that could not be revealed by less frequent professional monitoring. We found higher sea otter abundance in the estuary in spring–summer, indicating seasonality in use of the estuary. We detected differences in habitat use of the estuary between higher and lower tides, and greater frequency of foraging at low tide and in certain areas. Volunteer observations revealed fine‐scale differences in habitat use: eelgrass beds were used much more heavily than adjacent areas only a few meters away. Volunteer data can thus provide critical information about coastal habitat use and behavior that can improve conservation strategies for threatened wildlife species.

In this article, the authors present a collegiate recovery model of service that prioritizes a programmatic approach and intentionally designed services, activities, and mentoring that is structured upon a foundation of what we identify as five "Healing Pillars of Collegiate Recovery." The guiding principle of service is to provide students early in recovery with a healing environment that enables them to maintain and strengthen their paths to recovery and transformation both inside and beyond their College experience.

Students enrolled in two sections of a first-year non-majors biology course at a mid-sized university in the Mid-South were administered an instrument pre- and post-course comprised of the Measure of the Acceptance of the Theory of Evolution (MATE), the Generalized Acceptance of EvolutioN Evaluation (GAENE), 10 items related to pre-existing attitudes towards science and religion, and demographic items. Students in both sections received instruction in evolution, with one section receiving a supplemental presentation on the varying views within creationism. Correlations between MATE and GAENE scores were calculated, and comparisons were made between sections on changes in scores for both measures. We sought to answer these questions: 1) What is the current level of acceptance among first-year students in the Mid-South? 2) Are students amenable to changing their views? 3) Does discussing various creationist views matter? 4) Do demographics or pre-existing attitudes predict students’ acceptance? Students in both sections showed significant increases in acceptance, though no difference in the degree of increase was detected between the supplemented and standard instruction sections. Public high school graduates had higher initial acceptance than non-public school students. Strong correlations between measurement instruments were detected. [ABSTRACT FROM AUTHOR]

Extracellular vesicles have been implicated in the transmission of pathogens from the arthropod to the human host. Here the authors show that tick-derived extracellular vesicles play a role in feeding and modulate the outcome of bacterial infection.
