Abstract

The 2-point correlation function of the galaxy spatial distribution is a major cosmological observable that enables constraints on the dynamics and geometry of the Universe. The Euclid mission aims at performing an extensive spectroscopic survey of approximately 20--30 million H$\alpha$-emitting galaxies up to about redshift two. This ambitious project seeks to elucidate the nature of dark energy by mapping the 3-dimensional clustering of galaxies over a significant portion of the sky. This paper presents the methodology and software developed for estimating the 3-dimensional 2-point correlation function within the Euclid Science Ground Segment. The software is designed to overcome the significant challenges posed by the large and complex Euclid data set, which involves millions of galaxies. Key challenges include efficient pair counting, managing computational resources, and ensuring the accuracy of the correlation function estimation. The software leverages advanced algorithms, including kd-tree, octree, and linked-list data partitioning strategies, to optimise the pair-counting process. The implementation also includes parallel processing capabilities using shared-memory open multi-processing to further enhance performance and reduce computation times. Extensive validation and performance testing of the software are presented. The results indicate that the software is robust and can reliably estimate the 2-point correlation function, which is essential for deriving cosmological parameters with high precision. Abstract

We analyze causality and unitarity constraints in graviton scattering amplitudes, aiming to establish new bounds on theories with $U(1)$-gravitational anomalies, such as axion models or strongly-coupled gauge theories. For this purpose, we show the necessity of coupling these theories to gravity. We obtain a universal scale $\Lambda_{\rm caus}$ at which states with $J\geq 4$ must appear in the theory. We show that this scale can lie below the quantum gravity scale. For axion models, we get $\Lambda_{\rm caus}\sim\sqrt{M_P f_a}$ where $f_a$ is the axion decay constant. In strongly-coupled gauge theories in the large-$N_c$ limit, the presence of glueballs allows to evade these bounds, provided the number of fermions $N_F\ll N_c$ and the 'tHooft coupling is not large. Nevertheless, for models that have a holographic 5D dual (large 'tHooft coupling), $\Lambda_{\rm caus}$ emerges as a new cutoff scale, unless certain conditions on the parameters of the 5D models are satisfied.

Abstract

We introduce OneCovariance, an open-source software designed to accurately compute covariance matrices for an arbitrary set of two-point summary statistics across a variety of large-scale structure tracers. Utilising the halo model, we estimate the statistical properties of matter and biased tracer fields, incorporating all Gaussian, non-Gaussian, and super-sample covariance terms. The flexible configuration permits user-specific parameters, such as the complexity of survey geometry, the halo occupation distribution employed to define each galaxy sample, or the form of the real-space and/or Fourier space statistics to be analysed. We illustrate the capabilities of OneCovariance within the context of a cosmic shear analysis of the final data release of the Kilo-Degree Survey (KiDS-Legacy). Upon comparing our estimated covariance with measurements from mock data and calculations from independent software, we ascertain that OneCovariance achieves accuracy at the per cent level. When assessing the impact of ignoring complex survey geometry in the cosmic shear covariance computation, we discover misestimations at approximately the $10\%$ level for cosmic variance terms. Nonetheless, these discrepancies do not significantly affect the KiDS-Legacy recovery of cosmological parameters. We derive the cross-covariance between real-space correlation functions, bandpowers, and COSEBIs, facilitating future consistency tests among these three cosmic shear statistics. Additionally, we calculate the covariance matrix of photometric-spectroscopic galaxy clustering measurements, validating Jackknife covariance estimates for calibrating KiDS-Legacy redshift distributions. Abstract

We present cosmological parameter forecasts for the Euclid 6x2pt statistics, which include the galaxy clustering and weak lensing main probes together with previously neglected cross-covariance and cross-correlation signals between imaging/photometric and spectroscopic data. The aim is understanding the impact of such terms on the Euclid performance. We produce 6x2pt cosmological forecasts, considering two different techniques: the so-called harmonic and hybrid approaches, respectively. In the first, we treat all the different Euclid probes in the same way, i.e. we consider only angular 2pt-statistics for spectroscopic and photometric clustering, as well as for weak lensing, analysing all their possible cross-covariances and cross-correlations in the spherical harmonic domain. In the second, we do not account for negligible cross-covariances between the 3D and 2D data, but consider the combination of their cross-correlation with the auto-correlation signals. We find that both cross-covariances and cross-correlation signals, have a negligible impact on the cosmological parameter constraints and, therefore, on the Euclid performance. In the case of the hybrid approach, we attribute this result to the effect of the cross-correlation between weak lensing and photometric data, which is dominant with respect to other cross-correlation signals. In the case of the 2D harmonic approach, we attribute this result to two main theoretical limitations of the 2D projected statistics implemented in this work according to the analysis of official Euclid forecasts: the high shot noise and the limited redshift range of the spectroscopic sample, together with the loss of radial information from subleading terms such as redshift-space distortions and lensing magnification. 5. Deriving Cosmological Parameters from the Euclid mission

Abstract

The Euclid mission is a visionary project undertaken by the European Space Agency (ESA) to probe the universe's evolution and geometry by surveying the position and gravitational shape distortion of billions of galaxies. These observations bear the potential to offer unprecedented measurements of the cosmological parameters, thereby advancing our understanding of the cosmos. This work revolves around the central theme of quantifying the constraining power of the upcoming Euclid 3$\times$2pt photometric survey, accounting for several factors which have been neglected to this date in the official forecasts, especially more subtle sources of uncertainty which need to be included in the forecast (and data) analysis due to the precision of the observations. First, we include and study the impact of super-sample covariance, a source of sample variance coming from the incomplete sampling of the density and shear field Fourier modes caused by the limited survey volume. Second, we examine the effect of scale cuts, translating them from Fourier to harmonic space through the use of the BNT transform, which offers an efficient way of separating angular scales for the cosmic shear signal. This analysis allows quantifying and mitigating the bias coming from the uncertainty on our modelling of small scales. These updated forecasts, validated against the reference Euclid ones, provide insights into the expected precision achieved on the cosmological and nuisance parameters, for a variety of survey settings and for the inclusion of different realistic systematics, such as multiplicative shear bias, magnification bias, uncertainty in the mean of the redshift distribution and so on., Comment: 158 pages, 41 figures. arXiv admin note: text overlap with arXiv:2310.15731

Abstract

The current standard model of cosmology successfully describes a variety of measurements, but the nature of its main ingredients, dark matter and dark energy, remains unknown. Euclid is a medium-class mission in the Cosmic Vision 2015-2025 programme of the European Space Agency (ESA) that will provide high-resolution optical imaging, as well as near-infrared imaging and spectroscopy, over about 14,000 deg^2 of extragalactic sky. In addition to accurate weak lensing and clustering measurements that probe structure formation over half of the age of the Universe, its primary probes for cosmology, these exquisite data will enable a wide range of science. This paper provides a high-level overview of the mission, summarising the survey characteristics, the various data-processing steps, and data products. Abstract

Forthcoming large-scale structure (LSS) Stage IV surveys will provide us with unprecedented data to probe the nature of dark matter and dark energy. However, analysing these data with conventional Markov Chain Monte Carlo (MCMC) methods will be challenging, due to the increase in the number of nuisance parameters and the presence of intractable likelihoods. In light of this, we present the first application of Marginal Neural Ratio Estimation (MNRE) (a recent approach in simulation-based inference) to LSS photometric probes: weak lensing, galaxy clustering and the cross-correlation power spectra. In order to analyse the hundreds of spectra simultaneously, we find that a pre-compression of data using principal component analysis, as well as parameter-specific data summaries lead to highly accurate results. Using expected Stage IV experimental noise, we are able to recover the posterior distribution for the cosmological parameters with a speedup factor of $\sim 10-60$ compared to classical MCMC methods. To illustrate that the performance of MNRE is not impeded when posteriors are highly non-Gaussian, we test a scenario of two-body decaying dark matter, finding that Stage IV surveys can improve current bounds on the model by up to one order of magnitude. This result supports that MNRE is a powerful framework to constrain the standard cosmological model and its extensions with next-generation LSS surveys., Comment: 18+9 pages, 9 figures, 3 tables. Abstract

Deviations from Gaussianity in the distribution of the fields probed by large-scale structure surveys generate additional terms in the data covariance matrix, increasing the uncertainties in the measurement of the cosmological parameters. Super-sample covariance (SSC) is among the largest of these non-Gaussian contributions, with the potential to significantly degrade constraints on some of the parameters of the cosmological model under study - especially for weak lensing cosmic shear. We compute and validate the impact of SSC on the forecast uncertainties on the cosmological parameters for the Euclid photometric survey, obtained with a Fisher matrix analysis, both considering the Gaussian covariance alone and adding the SSC term - computed through the public code $\tt{PySSC}$. The photometric probes are considered in isolation and combined in the '3$\times$2pt' analysis. We find the SSC impact to be non-negligible - halving the Figure of Merit of the dark energy parameters $(w_0, w_a)$ in the 3$\times$2pt case and substantially increasing the uncertainties on $\Omega_{{\rm m}, 0}, w_0$, and $\sigma_8$ for cosmic shear; photometric galaxy clustering, on the other hand, is less affected due to the lower probe response. The relative impact of SSC does not show significant changes under variations of the redshift binning scheme, while it is smaller for weak lensing when marginalising over the multiplicative shear bias nuisance parameters, which also leads to poorer constraints on the cosmological parameters. Finally, we explore how the use of prior information on the shear and galaxy bias changes the SSC impact. Improving shear bias priors does not have a significant impact, while galaxy bias must be calibrated to sub-percent level to increase the Figure of Merit by the large amount needed to achieve the value when SSC is not included., Comment: 22 pages, 13 figures

Abstract

The simple analytic structure of meson scattering amplitudes in the large-$N_c$ limit, combined with positivity of the spectral density, provides precise predictions on low-energy observables. Building upon previous studies, we explore the allowed regions of chiral Lagrangian parameters and meson couplings to pions. We reveal a structure of kinks at all orders in the chiral expansion and develop analytical tools to show that kinks always correspond to amplitudes with a single light pole. We build (scalar- and vector-less) deformations of the Lovelace-Shapiro and Coon UV-complete amplitudes, and show that they lie close to the boundaries. Moreover, constraints from crossing-symmetry imply that meson couplings to pions become smaller as their spin increases, providing an explanation for the success of Vector Meson Dominance and holographic QCD. We study how these conclusions depend on assumptions about the high-energy behavior of amplitudes. Abstract

Lapatinib, an approved epidermal growth factor receptor inhibitor, was explored as a starting point for the synthesis of new hits against Trypanosoma brucei, the causative agent of human African trypanosomiasis (HAT). Previous work culminated in 1 (NEU-1953), which was part of a series typically associated with poor aqueous solubility. In this report, we present various medicinal chemistry strategies that were used to increase the aqueous solubility and improve the physicochemical profile without sacrificing antitrypanosomal potency. To rank trypanocidal hits, a new assay (summarized in a cytocidal effective concentration (CEC50)) was established, as part of the lead selection process. Increasing the sp3 carbon content of 1 resulted in 10e (0.19 μM EC50 against T. brucei and 990 μM aqueous solubility). Further chemical exploration of 10e yielded 22a, a trypanocidal quinolinimine (EC50: 0.013 μM; aqueous solubility: 880 μM; and CEC50: 0.18 μM). Compound 22a reduced parasitemia 109 fold in trypanosome-infected mice; it is an advanced lead for HAT drug development.

Abstract

Abstract The spread of Plasmodium falciparum parasites resistant to most first-line antimalarials creates an imperative to enrich the drug discovery pipeline, preferably with curative compounds that can also act prophylactically. We report a phenotypic quantitative high-throughput screen (qHTS), based on concentration–response curves, which was designed to identify compounds active against Plasmodium liver and asexual blood stage parasites. Our qHTS screened over 450,000 compounds, tested across a range of 5 to 11 concentrations, for activity against Plasmodium falciparum asexual blood stages. Active compounds were then filtered for unique structures and drug-like properties and subsequently screened in a P. berghei liver stage assay to identify novel dual-active antiplasmodial chemotypes. Hits from thiadiazine and pyrimidine azepine chemotypes were subsequently prioritized for resistance selection studies, yielding distinct mutations in P. falciparum cytochrome b, a validated antimalarial drug target. The thiadiazine chemotype was subjected to an initial medicinal chemistry campaign, yielding a metabolically stable analog with sub-micromolar potency. Our qHTS methodology and resulting dataset provides a large-scale resource to investigate Plasmodium liver and asexual blood stage parasite biology and inform further research to develop novel chemotypes as causal prophylactic antimalarials.

Abstract

Abstract Background Rodent malaria models are extensively used to predict treatment outcomes in human infections. There is a constant need to improve and refine these models by innovating ways to apply new scientific findings and cutting edge technologies. In addition, and in accordance with the three R’s of animal use in research, in vivo studies should be constantly refined to avoid unnecessary pain and distress to the experimental animals by using preemptive euthanasia as soon as the main scientific study objective has been accomplished. Methods The new methodology described in this manuscript uses the whole-body bioluminescence signal emitted by transgenic, luciferase-expressing Plasmodium berghei parasites to assess the parasite load predicted parasitaemia (PLPP) in drug and control treated female ICR-CD1 mice infected with 1 × 105 luciferase-expressing P. berghei (ANKA strain) infected erythrocytes. This methodology can replace other time-consuming and expensive methods that are routinely used to measure parasitaemia in infected animals, such as Giemsa-stained thin blood smears and flow cytometry. Results There is a good correlation between whole-body bioluminescence signal and parasitaemia measured using Giemsa-stained thin blood smears and flow cytometry respectively in donor and study mice in the modified Thompson test. The algebraic formulas which represent these correlations can be successfully used to assess PLPP in donor and study mice. In addition, the new methodology can pinpoint sick animals 2–8 days before they would have been otherwise diagnosed based on behavioural or any other signs of malaria disease. Conclusions The new method for predicting parasitaemia in the modified Thompson test is simple, precise, objective, and minimizes false positive results that can lead to the premature removal of animals from study. Furthermore, from the animal welfare perspective of replace, reduce, and refine, this new method facilitates early removal of sick animals from study as soon as the study objective has been achieved, in many cases well before the clinical signs of disease are present.

O presente trabalho tem como objetivo apresentar a trajetória de vida e profissional de um professor da Educação Infantil, bem como algumas questões de gênero envolvendo a docência. A pesquisa ocorreu em uma pré-escola municipal. Para a coleta de dados foram utilizadas as estratégias de investigação de abordagem qualitativa, como a observação participante e a realização de entrevista semiestruturada. Apreendemos momentos de interação do professor com a sua turma de alunos e alunas e suas práticas cotidianas. Através de seus relatos, o docente revelou como foi o seu ingresso na profissão, os desafios e os entraves enfrentados. Concluímos que a entrada de professores homens nesta etapa da educação pode contribuir para desconstruir concepções naturalizadas acerca da profissão, bem como romper com a imagem da mulher como educadora de crianças pequenas.

Background and Objectives: Unicompartmental knee arthroplasty (UKA) is a valid alternative to total knee arthroplasties (TKAs) in selected cases. After surgery, patients’ experience and satisfaction were traditionally evaluated by pre- and postsurgical scores and Patient-Reported Outcome Measures (PROMs). Otherwise, a statistically significant change does not necessarily correlate to a clinically meaningful improvement when measured using PROMs. To evaluate the real effect of a specific treatment and understand the difference between groups in a clinical trial, it is necessary to use a meaningful quantum of change on the score assessed. The minimal clinically important difference (MCID) and the Patient Acceptable Symptom State (PASS) can provide this meaningful change. This paper aimed to calculate the MCID and the PASS of the Forgotten Joint Score (FJS-12) after UKA. Materials and Methods: A total of 40 patients with a mean age 72.5 ± 6.4 years undergoing UKA were assessed preoperatively and six months postsurgery using the FJS-12 and the Oxford Knee Score (OKS). The baseline and 6-month postoperative scores were compared using the Wilcoxon signed ranks test. The correlation was calculated with Spearman’s rho. Both distribution-based approaches and anchor approaches were used to estimate MCID for the FJS-12. The 75th percentile and the Receiver operating characteristic (ROC) curve methods were used to calculate the PASS of FJS-12. Results: MCID estimates for normalized FJS-12 for UKA ranged from 5.68 to 19.82. The threshold of the FJS-12 with ROC method was 72.92 (AUC = 0.76). The cut-off value computed with the 75th percentile approach was 92.71. Conclusions: The MCID and PASS represent valid tools to assess the real perception of clinical improvement in patients who underwent UKA. The MCID value of FJS-12 was 12.5 for patients who underwent UKA. The value of the PASS for the FJS-12 in patients who underwent UKA was 72.92.

Slogan projekta Europeana Sounds kaže »Zvukovno nasljeđe Europe pod vašim prstima « i to je, u suštini, glavni cilj Projekta. Ovaj članak daje pregled Europeana Sounds projekta koji je započet u veljači 2014, a završen u siječnju 2017. godine. Projekt čini dostupnim sadržaje sakupljene putem Europeane koja nudi pristup do više od 53,870.000 izvora, uključujući knjige, videosnimke, slike, zvukove, umjetnička djela i još mnogo više iz europskog kulturnog nasljeđa. U prvome dijelu članka objašnjavaju se ciljevi Europeana Sounds, a to su: povećati ukupnu količinu zvučnih izvora koji su dostupni putem portala Europeana, obogatiti metapodatke vezane uz te izvore, kreirati specifičan kanal posvećen zvukovima i ostalim srodnim sadržajima, istražiti u svrhu rješavanja ograničenja domene vezane uz zvukovno nasljeđe, potaknuti kreativno korištenje navedenih resursa te kreirati mrežu interesenata i stručnjaka na polju zvuka. Unutar projekta formirano je nekoliko grupa koje su provele radnje vezane uz konačne ciljeve projekta: od sakupljanja sadržaja do tehničke infrastrukture koja omogućava objavljivanje na Europeani, od obogaćivanja metapodataka vezanih za izvore i crowdsourcinga do pitanja autorskih prava, od realizacije tematskog kanala do složenih aktivnosti vođenja projekta. Za vrijeme projekta svi postignuti ciljevi promovirani su kroz širok spektar aktivnosti, uključujući dva međunarodna skupa, sudjelovanjem na događanjima, dinamičnim blogom s više objava koje su vezane za zvukovno nasljeđe te stranicama na društvenim mrežama. Zaključno, trud uložen u pokretanje tematskoga kanala posvećenoga glazbi. Projekt, koji je u rasponu od tri godine ostvario razne ciljeve, nastavit će svoje aktivnosti zahvaljujući radnoj skupini formiranoj unutar International Association of Sound and Audiovisual Archives (IASA).

Abstract

Deviations from Gaussianity in the distribution of the fields probed by large-scale structure surveys generate additional terms in the data covariance matrix, increasing the uncertainties in the measurement of the cosmological parameters. Super-sample covariance (SSC) is among the largest of these non-Gaussian contributions, with the potential to significantly degrade constraints on some of the parameters of the cosmological model under study -- especially for weak lensing cosmic shear. We compute and validate the impact of SSC on the forecast uncertainties on the cosmological parameters for the Euclid photometric survey, obtained with a Fisher matrix analysis, both considering the Gaussian covariance alone and adding the SSC term -- computed through the public code PySSC. The photometric probes are considered in isolation and combined in the `3$\times$2pt' analysis. We find the SSC impact to be non-negligible -- halving the Figure of Merit of the dark energy parameters ($w_0$, $w_a$) in the 3$\times$2pt case and substantially increasing the uncertainties on $\Omega_{{\rm m},0}, w_0$, and $\sigma_8$ for cosmic shear; photometric galaxy clustering, on the other hand, is less affected due to the lower probe response. The relative impact of SSC does not show significant changes under variations of the redshift binning scheme, while it is smaller for weak lensing when marginalising over the multiplicative shear bias nuisance parameters, which also leads to poorer constraints on the cosmological parameters. Finally, we explore how the use of prior information on the shear and galaxy bias changes the SSC impact. Improving shear bias priors does not have a significant impact, while galaxy bias must be calibrated to sub-percent level to increase the Figure of Merit by the large amount needed to achieve the value when SSC is not included., Comment: 22 pages, 13 figures

Abstract

Antibiotic-resistant nosocomial infections are an emerging public health issue; carbapenem-resistant gram-negative bacteria such as Acinetobacter baumannii are among the pathogens against which new therapeutic agents are desperately needed. Drug repurposing has recently emerged as an alternative approach to rapidly identifying effective drugs and drug combinations to combat drug resistant bacteria. We performed a drug repurposing screen against a highly virulent, multidrug resistant, Acinetobacter baumannii strain AB5075. This strain, isolated from a patient, is resistant to 25 first-line antibiotics for gram-negative bacteria. A compound screen using a bacterial growth assay led to identification and confirmation of 43 active compounds. Among these confirmed compounds, seven are approved drugs or pharmacologically active compounds for non-antimicrobial indications. Three of these drugs, 5-fluorouracil, fluspirilene, and Bay 11-7082 resensitized strain AB5075 to azithromycin and colistin in a two-drug combination format. The approach using a drug repurposing screen with a pathogen sample isolated from a patient and a high throughput bacterial growth assay led to the successful identification of new drug combinations to overcome a multidrug resistant bacterial infection.

Abstract

We recently reported the medicinal chemistry re-optimization of a series of compounds derived from the human tyrosine kinase inhibitor, lapatinib, for activity against Plasmodium falciparum. From this same library of compounds, we now report potent compounds against Trypanosoma brucei brucei (which causes human African trypanosomiasis), T. cruzi (the pathogen that causes Chagas disease), and Leishmania spp. (which cause leishmaniasis). In addition, sub-micromolar compounds were identified that inhibit proliferation of the parasites that cause African animal trypanosomiasis, T. congolense and T. vivax. We have found that this set of compounds display acceptable physicochemical properties and represent progress towards identification of lead compounds to combat several neglected tropical diseases.
