Abstract

The International Muon Collider Collaboration (IMCC) [1] was established in 2020 following the recommendations of the European Strategy for Particle Physics (ESPP) and the implementation of the European Strategy for Particle Physics-Accelerator R&D Roadmap by the Laboratory Directors Group [2], hereinafter referred to as the the European LDG roadmap. The Muon Collider Study (MuC) covers the accelerator complex, detectors and physics for a future muon collider. In 2023, European Commission support was obtained for a design study of a muon collider (MuCol) [3]. This project started on 1st March 2023, with work-packages aligned with the overall muon collider studies. In preparation of and during the 2021-22 U.S. Snowmass process, the muon collider project parameters, technical studies and physics performance studies were performed and presented in great detail. Recently, the P5 panel [4] in the U.S. recommended a muon collider R&D, proposed to join the IMCC and envisages that the U.S. should prepare to host a muon collider, calling this their "muon shot". Abstract

Abstract This study investigates the efficacy of NKG2D chimeric antigen receptor (CAR) engineered T cells in targeting and eliminating stress-induced senescent cells in vitro. Cellular senescence contributes to age-related tissue decline and is characterized by permanent cell cycle arrest and the senescence-associated secretory phenotype (SASP). Immunotherapy, particularly CAR-T cell therapy, emerges as a promising approach to selectively eliminate senescent cells. Our focus is on the NKG2D receptor, which binds to ligands (NKG2DLs) upregulated in senescent cells, offering a target for CAR-T cells. Using mouse embryonic fibroblasts (MEFs) and astrocytes (AST) as senescence models, we demonstrate the elevated expression of NKG2DLs in response to genotoxic and oxidative stress. NKG2D-CAR T cells displayed potent cytotoxicity against these senescent cells, with minimal effects on non-senescent cells, suggesting their potential as targeted senolytics. In conclusion, our research presents the first evidence of NKG2D-CAR T cells’ ability to target senescent brain cells, offering a novel approach to manage senescence-associated diseases. The findings pave the way for future investigations into the therapeutic applicability of NKG2D-targeting CAR-T cells in naturally aged organisms and models of aging-associated brain diseases in vivo.

Abstract

Abstract Abnormal intraneuronal accumulation of soluble and insoluble α-synuclein (α-Syn) is one of the main pathological hallmarks of synucleinopathies, such as Parkinson’s disease (PD). It has been well documented that the reversible liquid-liquid phase separation of α-Syn can modulate synaptic vesicle condensates at the presynaptic terminals. However, α-Syn can also form liquid-like droplets that may convert into amyloid-enriched hydrogels or fibrillar polymorphs under stressful conditions. To advance our understanding on the mechanisms underlying α-Syn phase transition, we employed a series of unbiased proteomic analyses and found that actin and actin regulators are part of the α-Syn interactome. We focused on Neural Wiskott-Aldrich syndrome protein (N-WASP) because of its association with a rare early-onset familial form of PD. In cultured cells, we demonstrate that N-WASP undergoes phase separation and can be recruited to synapsin 1 liquid-like droplets, whereas it is excluded from α-Syn/synapsin 1 condensates. Consistently, we provide evidence that wsp-1/WASL loss of function alters the number and dynamics of α-Syn inclusions in the nematode Caenorhabditis elegans. Together, our findings indicate that N-WASP expression may create permissive conditions that promote α-Syn condensates and their potentially deleterious conversion into toxic species.

Abstract

Annually-resolved measurements of the radiocarbon content in tree-rings have revealed rare sharp rises in carbon-14 production. These 'Miyake events' are likely produced by rare increases in cosmic radiation from the Sun or other energetic astrophysical sources. The radiocarbon produced is not only circulated through the Earth's atmosphere and oceans, but also absorbed by the biosphere and locked in the annual growth rings of trees. To interpret high-resolution tree-ring radiocarbon measurements therefore necessitates modelling the entire global carbon cycle. Here, we introduce 'ticktack', the first open-source Python package that connects box models of the carbon cycle with modern Bayesian inference tools. We use this to analyse all public annual 14C tree data, and infer posterior parameters for all six known Miyake events. 10. Internal Carotid Artery Dissection Presenting with Transient or Subclinical Horner Syndrome

Abstract

Introduction: The most frequently encountered symptoms in internal carotid artery dissection (ICAD) are head or neck pain and cerebral ischemia. Ocular symptoms or signs have been reported as the presenting feature in up to 50% of patients, with (painful) Horner syndrome being the most frequently associated. Horner syndrome is part of the classic triad that depicts the characteristic presentation of ICAD and that consists of pain in the ipsilateral neck, head and orbital regions, (partial) Horner syndrome, and cerebral or retinal ischemia. All patients presenting with painful Horner syndrome should therefore require prompt investigations to rule out carotid artery dissection. In patients with confirmed diagnosis, treatment should be started early to prevent permanent ocular or cerebral complications. Case Presentation: Case 1: A 61-year-old woman presented with right temporal headache, an episode of transient visual loss and drooping of the right upper eyelid. Examination revealed anisocoria, which was more important in darkness. Reversal of anisocoria was observed after instilling drops of apraclonidine 0.5%. Neuroimaging demonstrated intrapetrous ICAD. Headaches, eyelid ptosis, and anisocoria all had resolved the next day. Apraclonidine pharmacologic testing a few weeks later was no longer dilating the previously smaller pupil. Case 2: A 48-year-old man presented with drooping of the right upper eyelid and right occipital headache and facial pain that all started one day after an intense yoga workout. Anisocoria was noticed upon examination, with topical cocaine 10% pharmacologic testing confirming a right Horner syndrome. Neuroimaging revealed ICAD. The patient reported resolution of his eyelid ptosis a few days later. Eyelid ptosis and anisocoria had indeed resolved at a follow-up examination a few weeks later. However, cocaine drop testing still produced anisocoria, compatible with subclinical Horner syndrome. Conclusion: Transient or subclinical Horner syndrome can be the presenting feature in ICAD; in such cases, the characteristic eyelid ptosis and anisocoria may be short-lived and resolve in only a few days. If suspected by clinical history, pharmacologic testing may be helpful in identifying subclinical cases.

Abstract

Next-generation plasma-based accelerators can push electron beams to GeV energies within centimetre distances. The plasma, excited by a driver pulse, is indeed able to sustain huge electric fields that can efficiently accelerate a trailing witness bunch, which was experimentally demonstrated on multiple occasions. Thus, the main focus of the current research is being shifted towards achieving a high quality of the beam after the plasma acceleration. In this letter we present beam-driven plasma wakefield acceleration experiment, where initially preformed high-quality witness beam was accelerated inside the plasma and characterized. In this experiment the witness beam quality after the acceleration was maintained on high level, with $0.2\%$ final energy spread and $3.8~\mu m$ resulting normalized transverse emittance after the acceleration. In this article, for the first time to our knowledge, the emittance of the PWFA beam was directly measured.

Abstract

Carbonyl sulfide (COS) is a promising tracer for the estimation of terrestrial ecosystem gross primary production (GPP). However, understanding its non-GPP-related sources and sinks, e.g., anthropogenic sources and soil sources and sinks, is also critical to the success of the approach. Here we infer the regional sources and sinks of COS using continuous in situ mole fraction profile measurements of COS along the 60 m tall Lutjewad tower (1 m a.s.l.; 53∘24′ N, 6∘21′ E) in the Netherlands. To identify potential sources that caused the observed enhancements of COS mole fractions at Lutjewad, both discrete flask samples and in situ measurements in the province of Groningen were made from a mobile van using a quantum cascade laser spectrometer (QCLS). We also simulated the COS mole fractions at Lutjewad using the Stochastic Time-Inverted Lagrangian Transport (STILT) model combined with emission inventories and plant uptake fluxes. We determined the nighttime COS fluxes to be -3.0±2.6 pmol m−2 s−1 using the radon-tracer correlation approach and Lutjewad observations. Furthermore, we identified and quantified several COS sources, including biodigesters, sugar production facilities and silicon carbide production facilities in the province of Groningen. Moreover, the simulation results show that the observed COS enhancements can be partially explained by known industrial sources of COS and CS2, in particular from the Ruhr Valley (51.5∘ N, 7.2∘ E) and Antwerp (51.2∘ N, 4.4∘ E) areas. The contribution of likely missing anthropogenic sources of COS and CS2 in the inventory may be significant. The impact of the identified sources in the province of Groningen is estimated to be negligible in terms of the observed COS enhancements. However, in specific conditions, these sources may influence the measurements in Lutjewad. These results are valuable for improving our understanding of the sources and sinks of COS, contributing to the use of COS as a tracer for GPP.

Abstract

Social media have become an integral part of our lives, expanding our interlinking capabilities to new levels. There is plenty to be said about their positive effects. On the other hand, however, some serious negative implications of social media have been repeatedly highlighted in recent years, pointing at various threats to society and its more vulnerable members, such as teenagers. We thus propose a theoretical framework based on an adaptive "Social Media Virtual Companion" for educating and supporting an entire community, teenage students, to interact in social media environments in order to achieve desirable conditions, defined in terms of a community-specific and participatory designed measure of Collective Well-Being (CWB). This Companion combines automatic processing with expert intervention and guidance. The virtual Companion will be powered by a Recommender System (CWB-RS) that will optimize a CWB metric instead of engagement or platform profit, which currently largely drives recommender systems thereby disregarding any societal collateral effect.We put an emphasis on experts and educators in the educationally managed social media community of the Companion. They play five key roles: (a) use the Companion in classroom-based educational activities; (b) guide the definition of the CWB; (c) provide a hierarchical structure of learning strategies, objectives and activities that will support and contain the adaptive sequencing algorithms of the CWB-RS based on hierarchical reinforcement learning; (d) act as moderators of direct conflicts between the members of the community; and, finally, (e) monitor and address ethical and educational issues that are beyond the intelligent agent's competence and control. Preliminary results on the performance of the Companion's components and studies of the educational and psychological underlying principles are presented.

Abstract

The study of materials for space exploration is one of the most interesting targets of international space agencies. An essential tool for realizing light junctions is epoxy adhesive (EA), which provides an elastic and robust material with a complex mesh of polymeric chains and crosslinks. In this work, a study of the structural and chemical modification of a commercial two-part flexible EA (3M™ Scotch-Weld™ EC-2216 B/A Gray), induced by 60Co gamma radiation, is presented. Combining different spectroscopic techniques, such as the spectroscopic Fourier transform infrared spectroscopy (FTIR), the THz time-domain spectroscopy (TDS), and the electron paramagnetic resonance (EPR), a characterization of the EA response in different regions of the electromagnetic spectrum is performed, providing valuable information about the structural and chemical properties of the polymers before and after irradiation. A simultaneous dissociation of polymeric chain and crosslinking formation is observed.The polymer is not subject to structural modification at an absorbed dose of 10 kGy, in which only transient free radicals are observed. Differently, between 100 and 500 kGy, a gradual chemical degradation of the samples is observed together with a broad and long-living EPR signal appearance. This study also provides a microscopic characterization of the material useful for the mechanism evaluation of system degradation.

Abstract

The plasma, excited by a driver pulse, generates large electric fields that can efficiently accelerate a trailing witness bunch making possible the realization of laboratory-scale applications ranging from high-energy colliders to ultra-bright light sources. So far several experiments have demonstrated a significant acceleration but the resulting beam quality, especially the energy spread, is still far from state of the art conventional accelerators. Here we show the results of a beam-driven plasma acceleration experiment where we used an electron bunch as a driver followed by an ultra-short witness. The experiment demonstrates, for the first time, an innovative method to achieve an ultra-low energy spread of the accelerated witness of about 0.1%. This is an order of magnitude smaller than what has been obtained so far. The result can lead to a major breakthrough toward the optimization of the plasma acceleration process and its implementation in forthcoming compact machines for user-oriented applications.

Abstract

The development of compact accelerator facilities providing high-brightness beams is one of the most challenging tasks in field of next-generation compact and cost affordable particle accelerators, to be used in many fields for industrial, medical and research applications. The ability to shape the beam longitudinal phase-space, in particular, plays a key role to achieve high-peak brightness. Here we present a new approach that allows to tune the longitudinal phase-space of a high-brightness beam by means of a plasma wakefields. The electron beam passing through the plasma drives large wakefields that are used to manipulate the time-energy correlation of particles along the beam itself. We experimentally demonstrate that such solution is highly tunable by simply adjusting the density of the plasma and can be used to imprint or remove any correlation onto the beam. This is a fundamental requirement when dealing with largely time-energy correlated beams coming from future plasma accelerators.

Abstract

BackgroundMR Tractography enables non-invasive preoperative depiction of language subcortical tracts, which is crucial for the presurgical work-up of brain tumors; however, it cannot evaluate the exact function of the fibers.PurposeA systematic pipeline was developed to combine tractography reconstruction of language fiber bundles, based on anatomical landmarks (Anatomical-T), with language fMRI cortical activations. A fMRI-targeted Tractography (fMRI-T) was thus obtained, depicting the subsets of the anatomical tracts whose endpoints are located inside a fMRI activation. We hypothesized that fMRI-T could provide additional functional information regarding the subcortical structures, better reflecting the eloquent white matter structures identified intraoperatively.MethodsBoth Anatomical-T and fMRI-T of language fiber tracts were performed on 16 controls and preoperatively on 16 patients with left-hemisphere brain tumors, using a q-ball residual bootstrap algorithm based on High Angular Resolution Diffusion Imaging (HARDI) datasets (b = 3000 s/mm2; 60 directions); fMRI ROIs were obtained using picture naming, verbal fluency, and auditory verb generation tasks. In healthy controls, normalized MNI atlases of fMRI-T and Anatomical-T were obtained. In patients, the surgical resection of the tumor was pursued by identifying eloquent structures with intraoperative direct electrical stimulation mapping and extending surgery to the functional boundaries. Post-surgical MRI allowed to identify Anatomical-T and fMRI-T non-eloquent portions removed during the procedure.ResultsMNI Atlases showed that fMRI-T is a subset of Anatomical-T, and that different task-specific fMRI-T involve both shared subsets and task-specific subsets - e.g., verbal fluency fMRI-T strongly involves dorsal frontal tracts, consistently with the phonogical-articulatory features of this task. A quantitative analysis in patients revealed that Anatomical-T removed portions of AF-SLF and IFOF were significantly greater than verbal fluency fMRI-T ones, suggesting that fMRI-T is a more specific approach. In addition, qualitative analyses showed that fMRI-T AF-SLF and IFOF predict the exact functional limits of resection with increased specificity when compared to Anatomical-T counterparts, especially the superior frontal portion of IFOF, in a subcohort of patients.ConclusionThese results suggest that performing fMRI-T in addition to the 'classic' Anatomical-T may be useful in a preoperative setting to identify the 'high-risk subsets' that should be spared during the surgical procedure.

Abstract

The aim of the present study is the development and the evaluation of preliminary psychometric characteristics of a short form self-report questionnaire to assess executive functioning (EF). The research was conducted in a sample of 316 Italian children that attended primary or secondary schools, with an age range between 7 and 11. EFA showed that Executive Functioning Self-Report (EF-SR) is composed of eleven items that are adequately represented by a single factor that characterizes the total score of the EFs. Furthermore, preliminary analyses showed an adequate level of skewness and kurtosis, of internal reliabilities, and test-retest reliability. Finally, the EF-SR showed positive moderate correlation with Stroop Congruent Trials, and with the Stroop Control trials, while the correlations with the other measures were not significance (Color Word Stroop task, the Corsi Block Tapping and the Tower of London), demonstrating appropriate discriminant validity. Abstract

Nanoparticles are being increasingly studied to enhance radiation effects. Among them, nanodiamonds (NDs) are taken into great consideration due to their low toxicity, inertness, chemical stability, and the possibility of surface functionalization. The objective of this study is to explore the influence of the chemical/physical properties of NDs on cellular radiosensitivity to combined treatments with radiation beams of different energies. DAOY, a human radioresistant medulloblastoma cell line was treated with NDs—differing for surface modifications [hydrogenated (H-NDs) and oxidized (OX-NDs)], size, and concentration—and analysed for (i) ND internalization and intracellular localization, (ii) clonogenic survival after combined treatment with different radiation beam energies and (iii) DNA damage and apoptosis, to explore the nature of ND–radiation biological interactions. Results show that chemical/physical characteristics of NDs are crucial in determining cell toxicity, with hydrogenated NDs (H-NDs) decreasing either cellular viability when administered alone, or cell survival when combined with radiation, depending on ND size and concentration, while OX-NDs do not. Also, irradiation at high energy (γ-rays at 1.25 MeV), in combination with H-NDs, is more efficient in eliciting radiosensitisation when compared to irradiation at lower energy (X-rays at 250 kVp). Finally, the molecular mechanisms of ND radiosensitisation was addressed, demonstrating that cell killing is mediated by the induction of Caspase-3-dependent apoptosis that is independent to DNA damage. Identifying the optimal combination of ND characteristics and radiation energy has the potential to offer a promising therapeutic strategy for tackling radioresistant cancers using H-NDs in conjunction with high-energy radiation.

Abstract

Beam injection and extraction from a plasma module is still one of the crucial aspects to solve in order to produce high quality electron beams with a plasma accelerator. Proper matching conditions require to focus the incoming high brightness beam down to few microns size and to capture a high divergent beam at the exit without loss of beam quality. Plasma-based lenses have proven to provide focusing gradients of the order of kT/m with radially symmetric focusing thus promising compact and affordable alternative to permanent magnets in the design of transport lines. In this paper an overview of recent experiments and future perspectives of plasma lenses is reported.

Abstract

On the wake of the results obtained so far at the SPARC\_LAB test-facility at the Laboratori Nazionali di Frascati (Italy), we are currently investigating the possibility to design and build a new multi-disciplinary user-facility, equipped with a soft X-ray Free Electron Laser (FEL) driven by a $\sim$1 GeV high brightness linac based on plasma accelerator modules. This design study is performed in synergy with the EuPRAXIA design study. In this paper we report about the recent progresses in the on going design study of the new facility.

Abstract

The current activity of the SPARC_LAB test-facility is focused on the realization of plasma-based acceleration experiments with the aim to provide accelerating field of the order of several GV/m while maintaining the overall quality (in terms of energy spread and emittance) of the accelerated electron bunch. In the following, the current status of such an activity is presented. We also show results related to the usability of plasmas as focusing lenses in view of a complete plasma-based focusing and accelerating system.

Abstract

Plasma wake-field acceleration experiments are performed at the SPARC LAB test facility by using a gas-filled capillary plasma source composed of a dielectric capillary. The electron can reach GeV energy in a few centimeters, with an accelerating gradient orders of magnitude larger than provided by conventional techniques. In this acceleration scheme, wake fields produced by passing electron beams through dielectric structures can determine a strong beam instability that represents an important hurdle towards the capability to focus high-current electron beams in the transverse plane. For these reasons, the estimation of the transverse wakefield amplitudes assumes a fundamental role in the implementation of the plasma wake-field acceleration. In this work, it presented a study to investigate which parameters affect the wake-field formation inside a cylindrical dielectric structure, both the capillary dimensions and the beam parameters, and it is introduced a quantitative evaluation of the longitudinal and transverse electric fields.

Abstract

R\&D activity on Cu photocathodes is under development at the SPARC\_LAB test facility to fully characterize each stage of the photocathode "life" and to have a complete overview of the photoemission properties in high brightness photo-injectors. The nano(n)-machining process presented here consists in diamond milling, and blowing with dry nitrogen. This procedure reduces the roughness of the cathode surface and prevents surface contamination introduced by other techniques, such as polishing with diamond paste or the machining with oil. Both high roughness and surface contamination cause an increase of intrinsic emittance and consequently a reduction of the overall electron beam brightness. To quantify these effects, we have characterized the photocathode surface in terms of roughness measurement, and morphology and chemical composition analysis by means of Scanning Electron Microscopy (SEM), Energy Dispersive Spectroscopy (EDS), and Atomic Force Microscopy (AFM) techniques. The effects of n-machining on the electron beam quality have been also investigated through emittance measurements before and after the surface processing technique. Finally, we present preliminary emittance studies of yttrium thin film on Cu photocathodes.

Abstract

IntroductionState of the art artificial intelligence (AI) models have the potential to become a “one-stop shop” to improve diagnosis and prognosis in several oncological settings. The external validation of AI models on independent cohorts is essential to evaluate their generalization ability, hence their potential utility in clinical practice. In this study we tested on a large, separate cohort a recently proposed state-of-the-art convolutional neural network for the automatic segmentation of intraprostatic cancer lesions on PSMA PET images.MethodsEighty-five biopsy proven prostate cancer patients who underwent 68Ga PSMA PET for staging purposes were enrolled in this study. Images were acquired with either fully hybrid PET/MRI (N = 46) or PET/CT (N = 39); all participants showed at least one intraprostatic pathological finding on PET images that was independently segmented by two Nuclear Medicine physicians. The trained model was available at https://gitlab.com/dejankostyszyn/prostate-gtv-segmentation and data processing has been done in agreement with the reference work.ResultsWhen compared to the manual contouring, the AI model yielded a median dice score = 0.74, therefore showing a moderately good performance. Results were robust to the modality used to acquire images (PET/CT or PET/MRI) and to the ground truth labels (no significant difference between the model’s performance when compared to reader 1 or reader 2 manual contouring).DiscussionIn conclusion, this AI model could be used to automatically segment intraprostatic cancer lesions for research purposes, as instance to define the volume of interest for radiomics or deep learning analysis. However, more robust performance is needed for the generation of AI-based decision support technologies to be proposed in clinical practice.
