10. Auricular and laryngeal chondritis in nursery and finishing pigs.

Abstract

This work aimed to characterize the clinic-pathological presentation of an outbreak of auricular and laryngeal chondritis in pigs. Visits were made to pig farms, where the clinical history was obtained, and clinical and postmortem examinations were performed. In those farms, 3% to 4% of pigs presented otohematomas, which started in the nursery and extended to the finishing phase. Moreover, some finishing pigs presented with respiratory distress, initially characterized as inspiratory dyspnea, associated by an uncommon respiratory stridor and culminating in death. Grossly, nursery piglets had enlarged ears, and on the cut surface, the cartilage was fragmented and associated with blood clots. In the finishing phase, in addition to auricular lesions, the epiglottis and arytenoid cartilages were thickened and distorted, which partially occluded the lumen. Microscopically, the laryngeal and auricular cartilages were fragmented, displayed a loss of matrix basophilia, and were surrounded by lymphohistiocytic inflammatory infiltrate, with occasional multinucleated giant cells and fibrosis. The lesions exclusively affected elastic cartilages. The disease in finishing pigs led to increased mortality and was a differential diagnosis to respiratory challenges. It was not possible to determine the factor that triggered this condition; however, a nutritional association is suspected. Abstract

Sow mortality is directly related to financial losses and productive efficiency in pig farms. Despite this, diseases associated with the death of sows are poorly understood. This study aimed to determine the main causes of death of sows in Brazilian pig farms. To perform this research, three Brazilian pig farms were visited, and necropsies were performed on all sows that had died spontaneously or were subjected to euthanasia. Tissue fragments were collected for histopathological and bacteriological examination. In addition, the clinical signs, productive stage, parity, and type of death (spontaneous or euthanasia) were recorded. A total of 138 necropsies were performed, and 132 had a conclusive diagnosis. The most frequent productive stages were gestation and lactation (33.3 and 31.9%, respectively), followed by parturient sows (17.4%), cull sows (9.4%), weaning-to-estrus interval (WEI) (4.3%), and unmated gilts (3.6%). The most affected organ systems were reproductive (28%), digestive (25%), locomotor (22%), cardiovascular (9.1%), and hematopoietic (6.1%). The most frequently diagnosed conditions were uterine prolapse (16/132; 12.1%), gastric ulcer (13/132; 9.8%), suppurative arthritis (11/132; 8.3%), liver lobe torsion (11/132; 8.3%), heart failure (9/132; 6.8%), vaginal or vaginal and rectal prolapse (9/132; 6.8%), and pododermatitis (8/132; 6.1%). Although 58.2% of the deaths were due to one of these seven diseases, there was a great variability in diagnoses. RESUMO: A mortalidade de porcas está diretamente relacionada a perdas financeiras e à eficiência produtiva das granjas. Apesar disso, as doenças associadas à morte de porcas são pouco conhecidas. Este estudo teve como objetivo determinar as principais causas de morte de porcas em granjas suinícolas brasileiras. Para a realização desta pesquisa, três granjas brasileiras foram visitadas e necropsias foram realizadas em todas as porcas que morreram espontaneamente ou foram submetidas à eutanásia. Fragmentos de tecidos foram coletados para exame histopatológico e bacteriológico. Além disso, foram registrados os sinais clínicos, estágio reprodutivo, ordem de parto e tipo de morte (espontânea ou eutanásia). Um total de 138 necropsias foram realizadas e 132 tiveram um diagnóstico conclusivo. Os estágios produtivos mais frequentes foram gestação e lactação (33,3 e 31,9%, respectivamente), seguidos por porcas parturientes (17,4%), porcas de descarte (9,4%), intervalo desmame-estro (IDE) (4,3%) e leitoas vazias (3,6%). Os sistemas orgânicos mais afetados foram reprodutor (28%), digestivo (25%), locomotor (22%), cardiovascular (9,1%) e hematopoiético (6,1%). As condições mais frequentemente diagnosticadas foram prolapso uterino (16/132; 12,1%), úlcera gástrica (13/132; 9,8%), artrite supurativa (11/132; 8,3%), torção do lobo hepático (11/132; 8,3%), insuficiência cardíaca (9/132; 6,8%), prolapso vaginal ou prolapso vaginal e retal (9/132; 6,8%) e pododermatite (8/132; 6,1%). Embora 58,2% dos óbitos tenham ocorrido por uma dessas sete doenças, houve grande variabilidade de diagnósticos.

Abstract

This report describes 2 events of degenerative myelopathy in 4- to 27-day-old piglets, with mortality rates reaching 40%. Sows were fed rations containing low levels of pantothenic acid. Piglets presented with severe depression, weakness, ataxia, and paresis, which were more pronounced in the pelvic limbs. No significant gross lesions were observed. Histologically, there were degeneration and necrosis of neurons in the spinal cord, primarily in the thoracic nucleus in the thoracic and lumbar segments, and motor neurons in nucleus IX of the ventral horn in the cervical and lumbar intumescence. Minimal-to-moderate axonal and myelin degeneration was observed in the dorsal funiculus of the spinal cord and in the dorsal and ventral nerve roots. Immunohistochemistry demonstrated depletion of acetylcholine neurotransmitters in motor neurons and accumulation of neurofilaments in the perikaryon of neurons in the thoracic nucleus and motor neurons. Ultrastructurally, the thoracic nucleus neurons and motor neurons showed dissolution of Nissl granulation. The topographical distribution of the lesions indicates damage to the second-order neurons of the spinocerebellar tract, first-order axon cuneocerebellar tract, and dorsal column-medial lemniscus pathway as the cause of the conscious and unconscious proprioceptive deficit, and damage to the alpha motor neuron as the cause of the motor deficit. Clinical signs reversed and no new cases occurred after pantothenic acid levels were corrected in the ration, and piglets received parenteral administration of pantothenic acid. Abstract

Sida carpinifoliais a small subshrub that is distributed throughout Brazil and is responsible for lysosomal storage disease and occasional reproductive problems in cattle, goats, equids, sheep, and deer. We describe herein the clinical, epidemiologic, and pathologic features of hydrallantois in 3 cows naturally poisoned by S. carpinifoliain Rio Grande do Sul State, Brazil. Clinically, all cows had marked abdominal distension and mild ataxia. After natural death or euthanasia, autopsies revealed that the abdominal distension in all 3 cases was caused by severe enlargement of the uterus, which contained 100–120 L of translucent fluid within the allantois, in addition to adventitial placentation. Microscopic evaluation of the placenta revealed marked diffuse edema, sometimes with a myxomatous appearance. Neurons in the cerebellum and obex were swollen, with mild-to-moderate cytoplasmic granular vacuolation. Histochemical examination with lectins ConA, WGA, and sWGA revealed mild-to-marked staining in the cytoplasm of neurons of the cerebellum and medulla at the level of the obex, indicating the occurrence of α-mannosidosis.

Abstract

The aim of this study was to investigate acetylcholinesterase (AChE) in total blood and liver tissue; butyrylcholinesterase (BChE) in serum and liver tissue; adenosine deaminase (ADA) in serum and liver tissue; and pyruvate kinase (PK) in liver tissue of rats experimentally infected by Fasciola hepatica. Animals were divided into two groups with 12 animals each, as follows: group A (uninfected) and group B (infected). Samples were collected at 20 (A1 and B1;n=6 each) and 150 (A2 and B2; n=6 each) days post-infection (PI). Infected animals showed an increase in AChE activity in whole blood and a decrease in AChE activity in liver homogenates (P<0.05) at 20 and 150 days PI. BChE and PK activities were decreased (P<0.05) in serum and liver homogenates of infected animals at 150 days PI. ADA activity was decreased in serum at 20 and 150 days PI, while in liver homogenates it was only decreased at 150 days PI (P<0.05). Aspartate aminotransferase and alanine aminotransferase activities in serum were increased (P<0.05), while concentrations of total protein and albumin were decreased (P<0.05) when compared to control. The histological analysis revealed fibrous perihepatitis and necrosis. Therefore, we conclude that the liver fluke is associated with cholinergic and purinergic dysfunctions, which in turn may influence the pathogenesis of the disease., (Copyright © 2015 Elsevier GmbH. Abstract

The aim of this study was to evaluate the nitric oxide (NO) levels, and oxidative and antioxidant markers of lambs experimentally and naturally infected by Haemonchus contortus, and its relation to lesions in the abomasum. For experimental study, a total of 14 healthy lambs were divided into two groups with seven animals each. Group A represented the uninfected animals (control), and Group B was formed by infected animals with 15,000 larvae of H. contortus. Blood was collected on days 15, 45, and 75 post-infection (PI) to obtain serum for biochemical analysis: lactate dehydrogenase (LDH), nitrite/nitrate (NOx), advanced oxidation protein products (AOPP), and ferric reducing ability of plasma (FRAP). Parasitological stool examination (eggs per gram of feces--EPG) was performed on days 15, 45, and 75 PI to verify the evolution of the infection. On day 15 PI EPG was negative, but on days 45 and 75 PI the EPG was positive for animals from Group B. In the three periods evaluated it was observed an increase of LDH levels in serum of lambs infected with gastrointestinal nematodes, but on the other hand NOx levels were reduced on the same periods in infected animals. The AOPP and FRAP levels did not differ between groups on days 15 and 45 PI, but increased significantly on day 75 PI in infected lambs. The same variables were studied in 10 lambs naturally infected with helminths, where more than 97% corresponded to H. contortus (hematocrit and EPG values were 18.8 ± 2.5% and 7120 ± 2940, respectively). Similar to the experimental study, the levels of NOx reduced, and the levels of LDH, FRAP, and AOPP increased in serum of this animal associated inflammatory infiltrate in the mucosa of the abomasum. Therefore, during the infection by H. contortus it was observed alterations in oxidative markers, indicators of cell lesion confirmed by histological examination of the abomasum, and consequently there were changes in antioxidant levels, with the purpose of cell protection.
