17. Detection of Apoptosis in Ovarian Cells In Vitro and In Vivo Using the Annexin V-Affinity Assay.

Abstract

The ability of a cell to undergo apoptosis is crucial during development, tissue homeostasis, and in the pathogenesis and treatment of disease (1). To study apoptosis, it is important to be able to detect apoptotic cells reliably. Here we describe a method to detect apoptosis in vitro and in vivo on basis of the changes in phospholipid distribution in the plasma membrane that occur during this process. In healthy cells, phosphatidylserine (PS) is maintained predominantly in the inner plasma membrane surface by an aminophospholipid translocase (2). However, early during apoptosis, PS is translocated from the inner to the outer membrane surface and serves as a trigger for adjacent phagocytes to remove the dying cell (3-5). Exposure of PS can be detected in vitro and in vivo with fluorochrome- or biotin-labeled annexin V, a protein that binds to negatively charged phospholipids in the presence of calcium ions (6,7). In cells that are cultured in suspension, detection of apoptosis on the basis of PS exposure is relatively easy (8). However, sample handling of adherent cell lines, such as the ovarian cell line PA-1, might interfere with reliable detection of PS exposure. Therefore, we developed a method to detect PS exposure in adherent cell lines by labeling cells in a monolayer with annexin V and harvesting the cells afterwards by mechanical scraping (9) (Figs. 1 and 2). Fixation of annexin V-labeled cells also allows the study of the relationship between PS exposure and expression of intracellular antigens (10). We also present a method to detect apoptosis in vivo during follicular maturation in the mouse Fig. 3). 38. Butylated hydroxyanisole-induced alterations parameters in rat forestomach in relation to cytochrome P-450-mediated metabolism.

Abstract

Four groups of six male Wistar rats (85 ± 7g)were fed a diet containing 0% (control) or 2% of the carcinogenic food antioxidant butylated hydroxyanisole (BHA) for 2 weeks. In this experiment, feeding 2% BHA is equivalent to a dose of 2.1 ± 0.3 g BHA/kg/day. One 0% BHA and one 2% BHA fed group of rats were daily injected i.p. with the cytochrome P-450 inducer phenobarbital (PB; 60 mg/kg) in saline. These two groups were encoded 0PB and 2PB respectively. Simultaneously, two control groups of rats were injected i.p. with saline only (0 and 2 respectively). PB administration increased relative weight, cytochrome P-450 content and ethoxycoumarin--deethylase activity of livers as compared to control rats, In addition, cytochrome P-450-medlated oxidative demethylatlon of BHA into tert-butyl-hydroquinone (TBHQ), monitored as urinary TBHQ excretion, was significantly increased in PB-induced rats as compared to non-induced rats (0.59 ± 0.19 versus 0.37 ± 0.09%; < 0.05). The mean labelling index (LI) and potential doubling time (T) in rat forestomach were significantly (P < 0.01) altered in groups of rats fed 2% BHA as compared to their appropriate control groups. No differences In cell kinetic parameters between either the two control groups (0, 0PB) or between the 2% BHA-fed groups (2, 2PB) was observed. Thus, although an increase in oxidative demethylation of BHA as a response to PB administration is evident, biotrans formation of BHA into TBHQ is not correlated to changes in cell kinetic parameters in rat forestomach. Moreover, in rats oxidative cytochrome P450-mediated demethylation of BHA into TBHQ appears not to be related to the oral dose of BHA. This indicates that oxidative cytochrome P450-mediated biotransformation of BHA does not contribute to the tumorigenicity of BHA in rat forestomach. [ABSTRACT FROM PUBLISHER]

39. Annexin V-Affinity assay: A review on an apoptosis detection system based on phosphatidylserine exposure

Abstract

Apoptosis is a programmed, physiological mode of cell death that plays an important role in tissue homeostasis. Understanding of the basic mechanisms that underlie apoptosis will point to potentially new targets of therapeutic treatment of diseases that show an imbalance between cell proliferation and cell loss. In order to conduct such research, techniques and tools to reliably identify and enumerate death by apoptosis are essential. This review focuses on a novel technique to detect apoptosis by targeting for the loss of phospholipid asymmetry of the plasma membrane. It was recently shown that loss of plasma membrane asymmetry is an early event in apoptosis, independent of the cell type, resulting in the exposure of phosphatidylserine (PS) residues at the outer plasma membrane leaflet. Annexin V was shown to interact strongly and specifically with PS and can be used to detect apoptosis by targeting for the loss of plasma membrane asymmetry. Labeled annexin V can be applied both in flow cytometry and in light microscopy in both vital and fixed material by using appropriate protocols. The annexin V method is an extension to the current available methods. This review describes the basic mechanisms underlying the loss of membrane asymmetry during apoptosis and discusses the novel annexin V-binding assay. Cytometry 31:1–9, 1998. © 1998 Wiley-Liss, Inc.

Abstract

Groups of five male Wistar rats weighing 306 ± 17 g were fed a diet containing 2% 2-(3)-tert-butyl-4-hydroxyanisole (BHA) or basal diet (control group) for 2 weeks. Subsequently, rats received an i.p. injection of 25 mg/kg bromodeoxyuridine (BrdU), a thymidine analogue, and were killed after 4 h. The gastro-intestinal tract was removed and fixed in 70% ethanol. After pepsin digestion of the fixed tissues, labelled cell nuclei were visualized by means of a monoclonal anti-BrdU antibody technique. Cell kinetic parameters were determined by means of bivariate BrdU/DNA analysis using flow cytometry. Mean labelling index and potential doubling time of forestomach cells were significantly increased (P < 0.002) in the 2% BHA group as compared to control rats. Cell kinetic parameters in other organs—glandular stomach, ileum, caecum and colon—were not affected by short-term consumption of BHA. [ABSTRACT FROM PUBLISHER]

45. Cell kinetics during regeneration in the sponge Halisarca caerulea: how local is the response to tissue damage?

Abstract

Sponges have a remarkable capacity to rapidly regenerate in response to wound infliction. In addition, sponges rapidly renew their filter systems (choanocytes) to maintain a healthy population of cells. This study describes the cell kinetics of choanocytes in the encrusting reef sponge Halisarca caerulea during early regeneration (0-8 h) following experimental wound infliction. Subsequently, we investigated the spatial relationship between regeneration and cell proliferation over a six-day period directly adjacent to the wound, 1 cm, and 3 cm from the wound. Cell proliferation was determined by the incorporation of 5-bromo-2'-deoxyuridine (BrdU). We demonstrate that during early regeneration, the growth fraction of the choanocytes (i.e., the percentage of proliferative cells) adjacent to the wound is reduced (7.0 ± 2.5%) compared to steady-state, undamaged tissue (46.6 ± 2.6%), while the length of the cell cycle remained short (5.6 ± 3.4 h). The percentage of proliferative choanocytes increased over time in all areas and after six days of regeneration choanocyte proliferation rates were comparable to steady-state tissue. Tissue areas farther from the wound had higher rates of choanocyte proliferation than areas closer to the wound, indicating that more resources are demanded from tissue in the immediate vicinity of the wound. There was no difference in the number of proliferative mesohyl cells in regenerative sponges compared to steady-state sponges. Our data suggest that the production of collagen-rich wound tissue is a key process in tissue regeneration for H. caerulea, and helps to rapidly occupy the bare substratum exposed by the wound. Regeneration and choanocyte renewal are competing and negatively correlated life-history traits, both essential to the survival of sponges. The efficient allocation of limited resources to these life-history traits has enabled the ecological success and diversification of sponges.
